In recent decades, there has been a dramatic increase in fundamental microbial disease pervasiveness. The evolution of multi-drug resistant organisms (MDROs) is attributed to the broad usage of commercial antibiotics, which indicates that the previously active antibiotics are now limited in their efficacy. Therefore, this work was designed to examine the antibacterial and antifungal potential of ellagic acid from Xylaria acuta of Millingtonia hortensis L.f. The extraction of the bioactive compound from the extracellular filtrate of the endophytic fungi was performed utilizing the solvent extraction process. The obtained bioactive compound was confirmed as ellagic acid by spectroscopic and chromatographic techniques. The antimicrobial assay was performed using the agar well diffusion method, and the MIC and MBC were performed by the broth microplate dilution technique. The purified compound was characterized as ellagic acid, which showed significant antimicrobial activity against the tested bacterial and fungal pathogens. The MIC of ellagic acid was 62.5 µg/ml against Staphylococcus aureus and Escherichia coli. Further, based on the in silico docking studies, molecular interactions and inhibitory properties of ellagic acid were studied using Autodock Vina. Docking results of ellagic acid predicted the lowest binding free energy against the bacterial target of S. aureus and the fungal target of Cladosporium cladosporioides, to be − 10.3 and − 8.7 kcal/mol, respectively. These results show that ellagic acid is an effective bioactive compound for inhibiting specific bacterial targets and a moderate antifungal agent. Collectively, ellagic acid reported in this study serves as a multi-target bioactive compound and can be used for the development of effective drug with potential health benefits. Molecularly Imprinted Microspheres (MIMs) or Microsphere Molecularly Imprinted Polymers represent an innovative design for the selective extraction of active compounds from natural products, showcasing effectiveness and cost-efficiency. MIMs, crosslinked polymers with specific binding sites for template molecules, overcome irregularities observed in traditional Molecularly Imprinted Polymers (MIPs). Their adaptability to the shape and size of target molecules allows for the capture of compounds from complex mixtures. This review article delves into exploring the potential practical applications of MIMs, particularly in the extraction of active compounds from natural products. Additionally, it provides insights into the broader development of MIM technology for the purification of active compounds. The synthesis of MIMs encompasses various methods, including precipitation polymerization, suspension polymerization, Pickering emulsion polymerization, and Controlled/Living Radical Precipitation Polymerization. These methods enable the formation of MIPs with controlled particle sizes suitable for diverse analytical applications. Control over the template-to-monomer ratio, solvent type, reaction temperature, and polymerization time is crucial to ensure the successful synthesis of MIPs effective in isolating active compounds from natural products. MIMs have been utilized to isolate various active compounds from natural products, such as aristolochic acids from Aristolochia manshuriensis and flavonoids from Rhododendron species, among others. Based on the review, suspension polymerization deposition, which is one of the techniques used in creating MIPs, can be classified under the MIM method. This is due to its ability to produce polymers that are more homogeneous and exhibit better selectivity compared to traditional MIP techniques. Additionally, this method can achieve recovery rates ranging from 94.91% to 113.53% and purities between 86.3% and 122%. The suspension polymerization process is relatively straightforward, allowing for the effective control of viscosity and temperature. Tuberculosis (TB) is the foremost cause of infectious fatality globally. The primary global challenge in combatting TB lies in addressing the emergence of drug-resistant variants of the disease. However, the number of newly approved agents for treating TB has remained remarkably low over recent decades. Hence, research endeavors for discovering novel anti-TB agents are always needed. In the present study, we screened over 1,500 culture extracts from actinomycetes isolated in Indonesia for their inhibitory activity against Mycobacterium smegmatis used as a surrogate in the primary screening. The initial screening yielded approximately 6.2 % hit extracts, with a selection criterion of >80 % growth inhibition. The confirmed hit extracts were subsequently subjected to growth inhibition assay against Mycobacterium bovis and Mycobacterium tuberculosis. Approximately 20 % of the hit extracts that showed growth inhibition also exhibited efficacy against M. bovis BCG and M. tuberculosis H37Rv pathogenic strain. An active compound was successfully purified from a large-scale culture of the most potent representative extract by high-performance liquid chromatography and thin-layer chromatography. The structure of the active compound was elucidated by mass spectrometry and nuclear magnetic resonance. This compound displayed structural similarities to actinomycin group and exhibited robust inhibition, with IC50 values of 0.74, 0.02, and 0.07 μg/mL against M. smegmatis, M. bovis, and M. tuberculosis, respectively. The Actinomycetes strain A612, which produced the active compound, was taxonomically classified by phylogenetic analysis of 16s rRNA gene and whole genome sequencing data as Streptomyces parvus. Computational genome analysis utilizing anti-SMASH 7.0 unveiled that S. parvus A612 strain harbors 40 biosynthetic gene clusters with the potential to produce 16 known (with >70 % similarity) and 24 unknown compounds. A non-ribosomal peptide synthesis (NRPS) gene cluster associated with actinomycin D biosynthesis was also identified, boasting an 85 % similarity. Molecular docking analysis of actinomycin D and 21 potential M. tuberculosis targets revealed possible interactions with multiple targets. The purified active compound inhibited recombinant M. tuberculosis shikimate kinase (MtSK), which validated the results obtained from the docking analysis.

Soilborne diseases cause significant economic losses in agricultural production around the world. They are difficult to control because a host plant is invaded by multiple pathogens, and chemical control often does not work well. In this study, we isolated and identified an endophytic Streptomyces sp. NEAU-DD186 from moss, which showed broad-spectrum antifungal activity against 17 soilborne phytopathogenic fungi, with Bipolaris sorokiniana being the most prominent. The strain also exhibited strong antibacterial activity against soilborne phytopathogenic bacteria Ralstonia solanacearum. To evaluate its biocontrol potential, the strain was prepared into biofertilizer by solid-state fermentation. Response surface methodology was employed to optimize the fermentation conditions for maximizing spore production and revealed that the 1:1 ratio of vermicompost to wheat bran, a temperature of 28°C, and 50% water content with an inoculation amount of 15% represented the optimal parameters. Pot experiments showed that the application of biofertilizer with a spore concentration of 108 CFU/g soil could effectively suppress the occurrence of tomato bacterial wilt caused by R. solanacearum and wheat root rot caused by B. sorokiniana, and the biocontrol efficacy was 81.2 and 72.2%, respectively. Chemical analysis of strain NEAU-DD186 extracts using nuclear magnetic resonance spectrometry and mass analysis indicated that 25-O-malonylguanidylfungin A and 23-O-malonylguanidylfungin A were the main active constituents, which showed high activity against R. solanacearum (EC50 of 2.46 and 2.58 µg ml-1) and B. sorokiniana (EC50 of 3.92 and 3.95 µg ml-1). In conclusion, this study demonstrates that Streptomyces sp. NEAU-DD186 can be developed as biofertilizer to control soilborne diseases. [ABSTRACT FROM AUTHOR]

The development of rice active compounds is conducive to improving the added value of rice. This paper focused on the types and effects of active compounds in rice. Furthermore, it summarized the effect of rice storage and processing technology on rice active compounds. We conclude the following: Rice contains a large number of active compounds that are beneficial to humans. At present, the research on the action mechanism of rice active compounds on the human body is not deep enough, and the ability to deeply process rice is insufficient, greatly limiting the development of the rice active compound industry. To maximize the added value of rice, it is necessary to establish a dedicated preservation and processing technology system based on the physicochemical properties of the required active compounds. Additionally, attention should be paid to the development and application of composite technologies during the development of the rice active compound industry.

Endophytic fungi can be found in various types of plants, especially medicinal plants, such as Syzygium zeylanicum, one of the many medicinal plants found in Indonesia. This study aimed to obtain isolates of endophytic fungi from the bark of Syzygium zeylanicum, L., with antibacterial and antioxidant activities and identify their bioactive compounds. Characterization of endophytic fungi was carried out morphologically and fungi that showed high bioactivity were characterized molecularly. Antibacterial activity was carried out by disc diffusion method and antioxidant activity by DPPH method. Isolation of bioactive compounds was carried out by chromatographic techniques and determination of the structure of pure chemical compounds by spectroscopic analysis. In this study, 3 isolates of endophytic fungi were obtained. Based on their morphological characterization, the endophytic fungi belong to the genera Trichoderma (Code SZT2 and SZT6), and Sclerotium (Code SZT8). Trichoderma mushroom extract (SZT2) gave high antibacterial activity (74.0 % against E. coli; 78.2 % against S. aureus; 80.5 % against S. tyhpi and 74.0 % against B. subtilis). The antioxidant activity was very strong SZT2 (IC50 = 5.83 µg/mL) SZT2 potential endophytic fungus based on molecular analysis is Trichoderma koningi. SZT2 mushroom extract has an active compound in the form of compound 3-hydroxy-4(hydroxy(4-hydroxyphenyl)methyl)-ɣ-butyrolactone based on the results of compound isolation using NMR and HMR. These endophytic fungal compounds can be developed into new sources of antibacterial and antioxidant bioactive compounds through a series of further studies. [ABSTRACT FROM AUTHOR]

The system of traditional Balinese medicine is known as Usada. One type of plant used in Usada is basil (Ocimum basilicum L). Antioxidant activity and phytochemical of ethanol extracts of two varieties of basil, Ocimum basilicum L (var.) Bali (OcB) and Ocimum basilicum L (var.) Lombok (OcL) was investigated. Total phenol was determined by folin-cioccalteu phenol reagent, total flavonoid by quarsetine reagent, tannin by Folin-Denis reagent, and antioxidant activity was determined using the 2,2-diphenyl-1-picrylhydrazyl radical scavenging method and is expressed as IC50. All of the parameters were measured by spectrophotometer. Phytochemicals were determined by Gas chromatography and mass spectrometry (GC-MS) Analysis. Morphological characters (stems, leaves, and flowers) are also observed. The results showed, in OcB obtained 92 compounds, 10 of which are important compounds that have an antioxidant effect, while in OcL 139 compound components were found, 15 of which are important compounds. Levels of phenols, flavonoids, tannins, as well as IC50 in OcB are; 3526.65 mg GAE/100g, 15841.6795 mg QE/100g, 2402.80 mg/100 g, and 24.9410 mg/L, respectively, while in OcL respectively are; 3297.71 mg GAE/100g, 13242.30 mg QE/100g, 850.71 mg/100, and 33.1105 mg/L. Conclusions, morphologically OcB and OcL showed no difference except leaf width and petiole length. The antioxidant activity of OcB is better, as well as higher levels of phenols, flavonoids, and tannins compared to OcL. [ABSTRACT FROM AUTHOR]

Five heat treatment methods including hot-air drying at 60 ℃, pasteurization at 80 ℃, four kinds of baking (110~140 ℃), steaming at 100 ℃ and boiling at 100 ℃ were used to treat Ziziphus jujuba cv. Huizao. The effect of different heating temperatures and heat treatment methods on total phenol content, total flavonoid content, total procyanidin content, phenolic compounds content, triterpenic acids content and antioxidant capacities of jujube were analyzed. The results showed that the active compound contents and antioxidant activities of jujube decreased after hot-air drying at 60 ℃, pasteurization at 80 ℃, steaming at 100 ℃ and boiling at 100 ℃. And the active compound contents in baked jujube at 110~140 ℃ showed an upward trend with the increasing of temperature. After baking at 140 ℃, the contents of total phenol, total flavonoid, total procyanidin and total triterpenic acids reached 341.3, 473.8, 659.3 and 560.4 mg UAE/100 g md. Baking treatment enhanced the antioxidant activities of jujube. After baking at 130 ℃, the DPPH· scavenging capacity of jujube reached 62.97 mmol TE/100 g md, and the ABTS+· scavenging capacity reached 47.96 mmol TE/100 g md. After baking at 130 and 140 ℃, the contents of active compound in jujube were higher than other groups and the antioxidant activities were stronger than other groups, so baking would be a suitable heat treatment methods for Ziziphus jujuba cv. Huizao processing.

Erwinia amylovora is a causative pathogen of fire blight disease, affecting apple, pear, and other rosaceous plants. Currently, management of fire blight relies on cultural and chemical practices, whereas it has been known that few biological resources exhibit disease control efficacy against the fire blight. In the current study, we found that an SFC20201208-M01 fungal isolate exhibits antibacterial activity against E. amylovora TS3128, and the isolate was identified as a Penicillium brasilianum based on the β-tubulin (BenA) gene sequence. To identify active compounds from the P. brasilianum culture, the culture filtrate was partitioned with ethyl acetate and n-butanol sequentially. From the ethyl acetate layer, we identified two new compounds (compounds 3-4) and two known compounds (compounds 1-2) based on spectroscopic analyses and comparison with literature data. Of these active compounds, penicillic acid (1) exhibited promising antibacterial activity against E. amylovora TS3128 with a minimal inhibitory concentration value of 25 μg/ml. When culture filtrate and penicillic acid (125 μg/ml) were applied onto Chinese pearleaf crab apple seedlings prior to inoculation of E. amylovora TS3128, the development of fire blight disease was effectively suppressed in the treated plants. Our results provide new insight into the biocontrol potential of P. brasilianum SFC20201208-M01 with an active ingredient to control fire blight.

Abstract Background Zishen Yutai (ZSYT) pill, a patent Chinese medicine, has been widely used in the treatment of infertility, abortion, and adjunctive treatment of in vitro fertilization (IVF) for decades. Recently, the results of clinical observations showed that premature ovarian failure (POF) patients exhibited improved expression of steroids and clinical symptoms associated with hormone disorders after treatment with Zishen Yutai pills. However, the pharmacological mechanism of action of these pills remains unclear. Methods The compounds of Zishen Yutai pills found in blood circulation were identified via ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF-MS) technique in the serum of POF mice after oral administration of Zishen Yutai pills. The potential targets of compounds were screened using Traditional Chinese Medicine Systems Pharmacology Database, Traditional Chinese Medicine Database@Taiwan, Drugbank Database, PubChem, HIT, Pharmapper, and Swiss Target Prediction. The target genes associated with POF were collected from Online Mendelian Inheritance in Man Database, PharmGkb, Genecards, Therapeutic Target Database, and Genetic Association Database. The overlapping genes between the potential targets of Zishen Yutai pills’ compounds and the target genes associated with POF were clarified via protein-protein interaction (PPI), pathway, and network analysis. Results Nineteen compounds in Zishen Yutai pills were detected in the serum of POF mice after oral administration. A total of 695 Zishen Yutai (ZSYT) pill-related targets were screened, and 344 POF-related targets were collected. From the results of Zishen Yutai (ZSYT) pill-POF PPI analysis, CYP19A1, AKR1C3, ESR1, AR, and SRD5A2 were identified as key targets via network analysis, indicating their core role in the treatment of POF with Zishen Yutai pills. Moreover, the pathway enrichment results suggested that Zishen Yutai pills treated POF primarily by regulating neuroactive ligand-receptor interaction, steroid hormone biosynthesis, and ovarian steroidogenesis. Conclusions Via virtual screening, we found that regulation of neuroactive ligand-receptor interaction, steroid hormone biosynthesis, and ovarian steroidogenesis was the potential therapeutic mechanism of Zishen Yutai pills in treating POF. Our study suggested that combining the analysis of Zishen Yutai pills’ compounds in blood in vivo in the POF model and network pharmacology prediction might offer a tool to characterize the mechanism of Zishen Yutai pills in the POF.

Citrus cultivars have remarkable health benefits, but only the anti-inflammatory activities of the major varieties have been studied. This study investigated the anti-inflammatory effects of various citrus cultivars and their active anti-inflammatory components. The essential oils of 21 citrus peels were extracted via hydrodistillation using a Clevenger-type apparatus, and the chemical compositions of the essential oils were analyzed. D-Limonene was the most abundant constituent. To evaluate the anti-inflammatory effects of the citrus cultivars, the gene expression levels of an inflammatory mediator and proinflammatory cytokines were investigated. Among the 21 essential oils, those extracted from C. japonica and C. maxima exhibited superior anti-inflammatory activities, being able to inhibit the expression of the inflammatory mediators and proinflammatory cytokines in lipopolysaccharide-stimulated RAW 264.7 cells. The essential oils of C. japonica and C. maxima were distinguished into seven distinct constituents, α-pinene, myrcene, D-limonene, β-ocimene, linalool, linalool oxide, and α-terpineol, compared with other essential oils. The anti-inflammatory activities of the seven single compounds significantly inhibited the levels of inflammation-related factors. In particular, α-terpineol exhibited a superior anti-inflammatory effect. This study showed that the essential oils from C. japonica and C. maxima exhibit high anti-inflammatory activity. Inflammatory disease in gastric tract called Inflammatory Bowel Disease (IBD), particularly colon, in general, results from the use of non-steroid anti-inflammatory drug, such as indomethacine. The use of natural material, such as mas ngur oyster (Atactodea striata) as traditional medicine has been long known by people in Kei Islands–Southeast Mallucas. This study is aimed at measuring the ability of active compounds of this oyster extract in reducing NF-κB and TNF-α expression and showing the histological image of indomethacine-induced rat ileum with IBD after treatment with mas ngur oyster extract. It used 8-12 week old male rats (Rattus norvegicus) of 150 - 200 g BW. The rats were divided into 3 groups, healthy group, sick group (induced with 15 mg/kg BW of indomethacine), and treatment group (orally induced with indomethacine at a dose of 15 mg/kg BW then treated with mas ngur oyster extract at the dose of 100, 400, 700 mg/kg BW). Indomethacine induction at the dose of 15 mg/kg BW and mas ngur oyster extract therapy were orally administered. NF-κB and TNF-α expressions were measured using immunohistochemicals, and the histological image used Hematoksilin-Eosin staining. Results showed that extract therapy gave significant effect (P<0.05) at the effective dose of 400 mg/kg BW that could reduce 86.421% of NF-κB expression and 60.972% of TNF-α expression in the rat ileum and result in tissue recovery of the IBD rat’s ileum after the therapy. In the present study, the variation patterns of leaf shape in different populations of individual Semiliquidambar cathayensis plants were analyzed to investigate the relationship among leaf shape variation, photosynthetic properties, and active compounds to understand the genetic characteristics of S. cathayensis and screen elite germplasms. The leaf shape of 18 offspring from three natural S. cathayensis populations was analyzed to investigate the level of diversity and variation patterns of leaf shape. Furthermore, photosynthetic pigment content, physiological parameters of photosynthesis, and the active compounds in leaves of different shapes were determined. Statistical analysis showed that the leaf shape variation in S. cathayensis indicated a high level of genetic diversity among and within the populations. Cluster analysis showed that the three natural populations formed two clusters, one whose offspring was dominated by entire leaves and another characterized by palmately trifoliate leaves. The differences in photosynthetic characteristics and active compounds of leaves of three different shapes were comprehensively evaluated using principal component analysis. Two principal components with a cumulative contribution rate of 92.768% were extracted, of which the highest comprehensive score was for asymmetrically lobed leaves. The leaf shape in different S. cathayensis germplasms exhibited distinct patterns, and there were some correlations between the photosynthetic properties and active compounds in leaves of different shapes. Thus, the leaf shape can be used to predict active compound content, and in turn, select varieties based on that purpose; it also provides a simple and effective method to classify S. cathayensis germplasms. Various active compounds are easily damaged, so they need protection and must be easily absorbed and targeted. This problem can be overcome by encapsulating in the form of solid lipid nanoparticles (SLNs). Initially, SLNs were widely used to encapsulate hydrophobic (non-polar) active compounds because of their matched affinity and interactions. Currently, SLNs are being widely used for the encapsulation of hydrophilic (polar) and semipolar active compounds, but there are challenges, including increasing their entrapment efficiency. This review provides information on current research on SLNs for encapsulation and delivery systems for active and antioxidant compounds, which includes various synthesis methods and applications of SLNs in various fields of utilization. SLNs can be developed starting from the selection of solid lipid matrices, emulsifiers/surfactants, types of active compounds or antioxidants, synthesis methods, and their applications or utilization. The type of lipid used determines crystal formation, control of active compound release, and encapsulation efficiency. Various methods can be used in the SLN fabrication of active compounds and hydrophilic/hydrophobic antioxidants, which have advantages and disadvantages. Fabrication design, which includes the selection of lipid matrices, surfactants, and fabrication methods, determines the characteristics of SLNs. High-shear homogenization combined with ultrasonication is the recommended method and has been widely used because of the ease of preparation and good results. PC-2 is one of the active compounds isolated from Indonesian red betel leaf. The structure of the compound is named 2-allyl-4-(1'-hydroxy-1'-(3",4",5"-trimethoxyphenyl)propan-2'-yl)-3,5-dimethoxycyclohexa-3,5-dienone and included in the neolignan's structure category. This compound was reported to have very good biological activity in giving antiproliferative effects on human breast (T47D) cells. From 2.12 g of its leaf methanolic extract, only 12.1 mg of Pc-2 was collected. This amount is too small to do more studies about its biological activities. The synthetic work should be applied to get a sufficient amount. This paper is aimed to explain the synthetic study and design of the Pc-2 compound. The first approach is by involving the aldol reaction in the synthetic pathway through the disconnection approach. The result showed that this attempt did not work very well. The second approach is to design the new synthetic pathway by using the synthia Organic Retrosynthesis Software by Merck. This software gave a reasonable result to be tried. The use of natural dyes as textile dyes is increasing due to environmentally friendly standards and does not require the use of synthetic dyes. By banning the use of synthetic dyes containing azo groups, this is the right moment to reintroduce natural dyes that have been abandoned for a long time. This study aims to identify the active compound of tantrum leaves (Indigofera tinctoria) as a natural textile dye by maceration extraction method with variations in maceration time and material treatment on wet and dry tantrum leaves. This experimental research method was carried out using several stages of analysis, namely the phytochemical test, FTIR (Fourier Transform Infrared Spectroscopy), and UV-Vis (Ultraviolet-Visible Light) spectrophotometer. The results of the phytochemical tests showed that the compounds contained in the Indigofera tinctoria plant were indigo, flavonoids, steroids, and alkaloids. The FTIR test indicated that the dye content in the extract was from the indigo group. The results of the UV-Vis spectrum showed that there was maximum absorption in wet tantrum leaf extract, dry tantrum leaf extract, and alkaline extract at wavelengths of 406.50 nm, 365.50 nm, and 664.50 nm, respectively. Background: Zishen Yutai (ZSYT) pill, a patent Chinese medicine, has been widely used in the treatment of infertility, abortion, and adjunctive treatment of in vitro fertilization (IVF) for decades. Recently, the results of clinical observations showed that premature ovarian failure (POF) patients exhibited improved expression of steroids and clinical symptoms associated with hormone disorders after treatment with Zishen Yutai pills. However, the pharmacological mechanism of action of these pills remains unclear. Methods: The compounds of Zishen Yutai pills found in blood circulation were identified via ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF-MS) technique in the serum of POF mice after oral administration of Zishen Yutai pills. The potential targets of compounds were screened using Traditional Chinese Medicine Systems Pharmacology Database, Traditional Chinese Medicine Database@Taiwan, Drugbank Database, PubChem, HIT, Pharmapper, and Swiss Target Prediction. The target genes associated with POF were collected from Online Mendelian Inheritance in Man Database, PharmGkb, Genecards, Therapeutic Target Database, and Genetic Association Database. The overlapping genes between the potential targets of Zishen Yutai pills' compounds and the target genes associated with POF were clarified via protein-protein interaction (PPI), pathway, and network analysis. Results: Nineteen compounds in Zishen Yutai pills were detected in the serum of POF mice after oral administration. A total of 695 Zishen Yutai (ZSYT) pill-related targets were screened, and 344 POF-related targets were collected. From the results of Zishen Yutai (ZSYT) pill-POF PPI analysis, CYP19A1, AKR1C3, ESR1, AR, and SRD5A2 were identified as key targets via network analysis, indicating their core role in the treatment of POF with Zishen Yutai pills. Moreover, the pathway enrichment results suggested that Zishen Yutai pills treated POF primarily by regulating neuroactive ligand-receptor interaction, steroid hormone biosynthesis, and ovarian steroidogenesis. Conclusions: Via virtual screening, we found that regulation of neuroactive ligand-receptor interaction, steroid hormone biosynthesis, and ovarian steroidogenesis was the potential therapeutic mechanism of Zishen Yutai pills in treating POF. Our study suggested that combining the analysis of Zishen Yutai pills' compounds in blood in vivo in the POF model and network pharmacology prediction might offer a tool to characterize the mechanism of Zishen Yutai pills in the POF. Studies have shown that approximately two-thirds of the plant species in the world have some medicinal value. Artocarpus lakoocha is a synonym for Artocarpus lacucha and is a plant that can be found in Indonesia. This medicinal plant has been used to treat many diseases. (1) Objective: This article discusses the scientific investigations carried out on A. lacucha, namely the plant's chemical content, pharmacological activity, and active compounds. (2) Methods: The design of this study was based on an article that was a review of previous research. A search for relevant publications over the past ten years (2012–2022) using data from Pubmed, Proquest, Ebsco, ScienceDirect, and Google Scholar resulted in the discovery of 369 articles. (3) Results: Fifty relevant articles investigate A. lacucha's substances and their applications in the health field. The presence of secondary metabolites and bioactive compounds has been reported, which is evidence that A. lacucha possesses antidiarrheal, immunostimulant, anticholesterol, and hepatoprotective agents. (4) Conclusions: Mobe (A. lacucha) is a plant native to North Sumatra, Indonesia. This plant is efficacious as an antioxidant, antibacterial, antidiarrheal, anti-inflammatory, analgesic, antinociceptive, schistosomicidal, hepatoprotective, neuroprotective, cytotoxic, antiglycation, and anticholesterol, and can also be used for anti-aging and wound healing. The Eupatorium plant has been well used in medication and as a decorative plant. Some studies have reported that this herb has biochemical compounds, such as sesquiterpenes, phenolics, polysaccharides, and pyrrolizidine alkaloids. Thus, it has pharmacological effects, including antifungal, antibacterial, cytotoxic, and antinociceptive properties, that can be utilized for cosmetic purposes. However, only a few published works have summarized the active compounds and the application of Eupatorium plants as cosmetic agents. Therefore, this article aims to review the application of Eupatorium plants as a potential cosmetic agent. The active compounds of Eupatorium are contained in the whole plant, as well as the stems, leaves, roots, and aerial parts (flower, fruit, and seeds). In terms of cosmetic applications, the activities of Eupathorium are antioxidant, anti-tyrosinase, anti-melanin/melanogenesis, anti-acne, and anti-inflammatory. Diabetes mellitus is a metabolic disorder disease which is the fourth leading cause of death in Indonesia. The number of people with diabetes in Indonesia shows an increase from year to year, in 2007 people with diabetes reached 14.7% of the population and it is predicted that in 2030 the number of people with diabetes in Indonesia will reach 21.3 million people. Globally, Indonesia ranks fourth in the number of diabetics after India, China and the United States. Herbal medicines from spices have the opportunity to be developed into antidiabetic agents, with considerations of safety, minimal side effects and cheap. Several types of spices have been reported to be used as antidiabetic agents. Cabe jamu, one of the famous spices in Madura, has been reported to have many health benefits. This study will identify the active compounds in cabe jamu that have the ability as antidiabetics by using a molecular docking approach. The results of the study found that cabe jamu has 22 active compounds that have the potential as herbal medicines. Of the 22 active compounds, 10 of them were successfully docked with diabetes aldose reductase protein. The results of this study indicate that the active compounds contained in cabe jamu have the ability as an antidiabetic agent. Based on the strength of the hidrogen bond, and the absence of toxic potential, 3 active compounds from cabe jamu that have strong antidiabetic abilities and no toxic potential were obtained, namely piperine, retrofractamide C, and guineensine.

Laccase enzymes have been used widely in industrial fields such as textile, pulp, paper, food, cosmetic, and pharmaceutical industries. Laccase is used in toothpaste, mouthwash, deodorants, and soaps in personal care products. Previously, laccase enzymes had never been used for formulating hand sanitizer gel. This study aimed to determine the effect of the laccase enzyme on the physicochemical properties and the antibacterial potential of the hand sanitizer gel against pathogenic bacteria. Laccase enzyme was produced through fermentation using the fungus Trametes hirsuta EDN 082 with an activity of 0.032 U/mL. Hand sanitizer gel was made with the addition of laccase enzyme with varying concentrations of 4%, 7%, and 10% (v/v). The physicochemical test included organoleptic tests, pH evaluation, gel spreadability, and viscosity. The antibacterial was tested by the palm swab method. The gel physicochemical characteristics showed that the more laccase enzyme added, the more yellow the color produced, the less thick the shape, the wider the gel spreadability, and the lower the viscosity. The obtained pH ranged from 7.4 to 7.6. The best formulation of the hand sanitizer gel was achieved with the addition of a 7% (v/v) laccase enzyme. This formulation can reduce the number of bacteria colonies of Staphylococcus aureus and Escherichia coli on the palms with effectiveness above 95%. The laccase enzyme can be used as an active ingredient and antibacterial agent in the formulation of hand sanitizers.

Colon-targeted drug delivery systems have been exploited to treat local diseases in the colon, systemic delivery of protein and peptide, and chronotherapeutic drugs. However, the upper gastrointestinal tract restricts the effective delivery. Therefore, several strategies are needed for drugs targeted directly to the colon, such as pH-sensitive and enzyme-sensitive polymers, bacterially degradable polysaccharides, time-dependent polymers, as well as particulate systems. Variable physiological conditions of the gastrointestinal tract in individuals cause the combinations of these strategies to ensure colonic delivery of drugs. This review presents and discusses several potential drugs and the approaches used to design and develop colon-targeted drug delivery systems for medications with particular characteristics. Type 2 diabetes mellitus (T2DM) and Parkinson's disease (PD) are chronic conditions that affect the aging population, with increasing prevalence globally. The rising prevalence of comorbidity between these conditions, driven by demographic shifts, severely impacts the quality of life of patients, posing a significant burden on healthcare resources. Chinese herbal medicine has been used to treat T2DM and PD for millennia. Pharmacological studies have demonstrated that medicinal herbs effectively lower blood glucose levels and exert neuroprotective effects, suggesting their potential as adjunctive therapy for concurrent management of T2DM and PD. To elucidate the shared mechanisms underlying T2DM and PD, particularly focusing on the potential mechanisms by which medicinal herbs (including herbal formulas, single herbs, and active compounds) may treat these diseases, to provide valuable insights for developing therapeutics targeting comorbid T2DM and PD. Studies exploring the mechanisms underlying T2DM and PD, as well as the treatment of these conditions with medicinal herbs, were extracted from several electronic databases, including PubMed, Web of Science, Google Scholar, and China National Knowledge Infrastructure (CNKI). Numerous studies have shown that inflammation, oxidative stress, insulin resistance, impaired autophagy, gut microbiota dysbiosis, and ferroptosis are shared mechanisms underlying T2DM and PD mediated through the NLRP3 inflammasome, NF-κB, MAPK, Keap1/Nrf2/ARE, PI3K/AKT, AMPK/SIRT1, and System XC−-GSH-GPX4 signaling pathways. Thirty-four medicinal herbs, including 2 herbal formulas, 4 single herbs, and 28 active compounds, have been reported to potentially exert anti-T2DM and anti-PD effects by targeting these shared mechanisms. Traditional Chinese medicine effectively combats T2DM and PD through shared pathological mechanisms, highlighting their potential for application in treating these comorbid conditions. Tianshu capsule (TSC) is a traditional Chinese medicine (TCM) preparation and has significant clinical effect on migraine. The composition characteristics of TSC formula are worth exploring for the treatment of migraine. Identify the compounds in vivo after oral administration of TSC, Gastrodin (GAS), ferulic acid, senkyunolide G and senkyunolide I in the blood of healthy and migraine rats were used as representative compounds for time-dependent processes investigation. And we focus on explaining the characteristics of TSC treatment on migraine from a pharmacokinetic perspective. In this study, Ultra-high performance liquid chromatography with Orbitrap tandem mass spectrometry (UPLC-Orbitrap-MS/MS) system were used to detect the compounds in rat plasma and brain after oral administration of TSC. A sensitive, selective and reliable ultra-performance liquid chromatography coupled with triple quadrupole mass spectrometry (UPLC-TQ-MS/MS) system method was established for the simultaneous quantitative analysis of multi-components and evaluate the pharmacokinetic behavior of four main compounds. A total of 46 compounds were significantly identified in vivo of rat, including 35 compounds in plasma, 7 compounds only in brain and 4 compounds both in plasma and brain. The quantitative method has been confirmed to be feasible for multi-components content determination, pharmacokinetic parameters indicated that the four compounds absorbed into blood rapidly, and senkyunolide I also cross the blood-brain barrier (BBB) quickly into brain. GAS has relatively high concentrations in plasma, and the parameters AUC 0-t , CL z/F displayed significant differences between the normal and migraine rats. And the AUC 0-t and C max of GAS and ferulic acid exhibited dose-dependent way. we added compounds for the pharmacokinetic study of TSC, providing powerful help for clinical medication in the treatment of migraine. Warm and humid climate creates ideal conditions for mosquito breeding. The ability of these vectors to spread a number of diseases to humans causes millions of deaths every year. Indiscriminate use of synthetic insecticides leads to the development of resistance in vector mosquitoes and along with this, these pesticides cause biological magnification of toxic components and affects adversely the non-target organisms including human being. Commercially available, chemically manufactured mosquito repellent fast cards are convenient to use and quite effective but burning of such card generates a lot of smoke and might be hazardous to human health in the long run. Thus, alternative approaches are to be adopted to control the population load of vector mosquito. Like that, the present study also reveals the larvicidal effect of Duranta leaf extract against Culex mosquito. In the present study, mosquito repellent fast card has been developed by Duranta-algal mixture which has shown better result than commercially available fast card on the basis of mosquito mortality as well as the amount of gases emitted. Again, the ethanolic crude extract of Duranta leaves leads to 100% mortality of all instars (Culex pipiens) larvae at both 1000 ppm and 500 ppm concentration. Therefore, the active component of Duranta has also been investigated. In Duranta, highest area percentage and peak have been shown by propionic acid in the retention time 18.086 by GC–MS. So, it can be confirmed that the major active ingredient is propionic acid in Duranta which is responsible for the mosquitocidal properties. Bioactive compounds in Eucheuma cottonii include phenols or polyphenols with antioxidant, antibacterial, and antiviral properties. Seaweed quality, including phenolic content, is highly determined by several factors, including the growth location, cultivation technique, and harvest age. This study aimed to determine the effect of harvest age and solvent type on the phenolic content of E.cottonii. The effects of harvest age on water content and of simplicia on the extraction process were also determined. Phenolic active ingredients were extracted at three harvest ages (35, 40, and 45 days) and using four solvents (ethanol, ethyl acetate, hexane, and water). Analysis of variance was performed to determine the effects of both factors and their interactions. The older the seaweed, the lesser the water content. The powder produced at all ages is a good simplicia for materials extraction. Results showed that high phenolic content was obtained from the E. cottonii extract at the age of 35 days with ethyl acetate as the solvent.

Fermentation is considered a promising way to increase the bioactive substance content as well as the curing efficacy of foods and herbal medicines. However, in the vast majority of cases, it remains unknown what bioactive ingredient and physiological activity are altered in traditional herbs after fermentation. Here, in order to find the effect of fermentation on the bioactive compound content and health efficacy of Cornus officinalis fruit, an important Chinese traditional medicine, its total phenolic content, total flavonoid content, antioxidant activity and anti-tyrosinase activity were estimated during Bifidobacterium bifidum fermentation process. The results showed that, during COF (Cornus officinalis fruit)’s fermentation, the total phenolic contents were significantly increased, however, the total flavonoid contents weren’t obviously changed, and that the antioxidant activity as well as the anti-tyrosinase activity was enhanced. In addition, the total phenolic content was positively related not only to the antioxidant activity but also to the anti-tyrosinase activity. This research can help us better understand the alterations of ingredients and physiological activities of fermented traditional medicines that influence curing efficacy.

Kabau (Archidendron bubalinum) is one of the plants that is often used as food ingredients and has the potential to be developed in the health sector. This study aims to determine the antioxidant potential of kabau seeds using 2,2-diphenyl-1-picril hidrazil (DPPH) and ferric reducing ability power (FRAP) methods and to identify active compounds of kabau seeds using GC-MS. Kabau seeds were macerated using 70% ethanol and 99.9% ethanol. The results showed that 99.9% ethanol extract of kabau seed coat had the highest antioxidant activity with IC50 values of 26.75 ppm for the DPPH method and 121.55 ppm for the FRAP method, respectively. Identification of active compounds using GC-MS showed that 99.9% ethanol extract of kabau seed coat contained the most 9,12-octadecadienoic acid compounds (linoleic acid) so that it could be developed as an antioxidant.

Various active compounds are easily damaged, so they need protection and must be easily absorbed and targeted. This problem can be overcome by encapsulating in the form of solid lipid nanoparticles (SLNs). Initially, SLNs were widely used to encapsulate hydrophobic (non-polar) active compounds because of their matched affinity and interactions. Currently, SLNs are being widely used for the encapsulation of hydrophilic (polar) and semipolar active compounds, but there are challenges, including increasing their entrapment efficiency. This review provides information on current research on SLNs for encapsulation and delivery systems for active and antioxidant compounds, which includes various synthesis methods and applications of SLNs in various fields of utilization. SLNs can be developed starting from the selection of solid lipid matrices, emulsifiers/surfactants, types of active compounds or antioxidants, synthesis methods, and their applications or utilization. The type of lipid used determines crystal formation, control of active compound release, and encapsulation efficiency. Tuberculosis (TB) is the foremost cause of infectious fatality globally. The primary global challenge in combatting TB lies in addressing the emergence of drug-resistant variants of the disease. However, the number of newly approved agents for treating TB has remained remarkably low over recent decades. Hence, research endeavors for discovering novel anti-TB agents are always needed. In the present study, we screened over 1,500 culture extracts from actinomycetes isolated in Indonesia for their inhibitory activity against Mycobacterium smegmatis used as a surrogate in the primary screening. The initial screening yielded approximately 6.2 % hit extracts, with a selection criterion of >80 % growth inhibition. The confirmed hit extracts were subsequently subjected to growth inhibition assay against Mycobacterium bovis and Mycobacterium tuberculosis. Approximately 20 % of the hit extracts that showed growth inhibition also exhibited efficacy against M. bovis BCG and M. tuberculosis H37Rv pathogenic strain . An active compound was successfully purified from a large-scale culture of the most potent representative extract by high-performance liquid chromatography and thin-layer chromatography. The structure of the active compound was elucidated by mass spectrometry and nuclear magnetic resonance. This compound displayed structural similarities to actinomycin group and exhibited robust inhibition, with IC 50 values of 0.74, 0.02, and 0.07 μg/mL against M. smegmatis , M. bovis, and M. tuberculosis , respectively. The Actinomycetes strain A612, which produced the active compound, was taxonomically classified by phylogenetic analysis of 16s rRNA gene and whole genome sequencing data as Streptomyces parvus . Computational genome analysis utilizing anti-SMASH 7.0 unveiled that S. parvus A612 strain harbors 40 biosynthetic gene clusters with the potential to produce 16 known (with >70 % similarity) and 24 unknown compounds. A non-ribosomal peptide synthesis (NRPS) gene cluster associated with actinomycin D biosynthesis was also identified, boasting an 85 % similarity. Molecular docking analysis of actinomycin D and 21 potential M. tuberculosis targets revealed possible interactions with multiple targets. Molecular pharmacognosy as an emerging interdisciplinary subject based on molecular biology and Chinese materia medica aims to study the synthesis and molecular regulation of secondary metabolites in medicinal plants. Andrographis Herba, the dried aboveground part of Andrographis paniculata, has liver-protecting, bile secretion-promoting, heat-clearing, toxin-removing, antimicrobial, and anti-inflammatory effects. The quality instability caused by plant varieties, environment, and technology in the production of A. paniculata is a limiting factor for the sustainable development of this industry. Based on the research methods of molecular pharmacognosy and omics, the regulation of secondary metabolites of A. paniculata has become the key solution to the quality problems of A. paniculata. This paper summarized the recent research achievements in the molecular pharmacognosy of A. paniculata, including molecular identification of the resources, genetic diversity, multi-omics, biosynthesis of active compounds, and germplasm resource innovation, and prospected the future development trend in this field. In-depth research of molecular pharmacognosy of A. paniculata will provide more scientific and effective technical support for the development of its medicinal value, give new insights into the cultivation of new A. paniculata varieties, and promote the high-quality sustainable development of this industry.

Xiaoxuming decoction (XXMD) has been traditionally used to manage stroke though debates on its clinical efficacy were present in the history. Till nowadays, it is still one of the most commonly used herbal recipes for stroke. One of the reasons is that a decent proportion of ischemic stroke patients still have residue symptoms even after thrombolysis with rt-PA or endovascular thrombectomy. Numerous clinical studies have shown that XXMD is an effective alternative therapy not only at the acute stage, but also at the chronic sequelae stage of ischemic stroke. Modern techniques have isolated groups of compounds from XXMD which have shown therapeutic effects, such as dilating blood vessels, inhibiting thrombosis, suppressing oxidative stress, attenuating nitric oxide induced damage, protecting the blood brain barrier and the neurovascular unit. However, which of the active compounds is responsible for its therapeutic effects is still unknown. Emerging studies have screened and tested these active compounds aiming to find individual compounds that can be used as drugs to treat stroke. Smoke-derived seed germination is an important trait for plants to colonize postfire habitats. The well-characterized smoke-derived chemicals of karrikins germinate seeds of species not known to occur after fires in nature. Hence, the ecologically relevant germination cues in smoke remain to be explored for native postfire plants. With the fire-chaser, Nicotiana attenuata, we revisit a bioassay-driven fractionation of liquid smoke to identify ecologically relevant germination cues. By combining bioassay-guided fractionation and comparative unbiased metabolomics, we developed a robust and efficient method to identify germination cues in smoke. Syringaldehyde (SAL) was re-identified as a germination cue in fractions of liquid smoke that promote seed germination. SAL was found to be produced during wildfires in the plant's native habitat, efficiently adsorbed to N. attenuata seeds from aqueous solutions and not readily leached from soil and accurately predicted the boundaries of natural fire events that reflect the occurrence of native postfire N. attenuata populations. Xiaoxuming decoction (XXMD) has been traditionally used to manage stroke though debates on its clinical efficacy were present in the history. Till nowadays, it is still one of the most commonly used herbal recipes for stroke. One of the reasons is that a decent proportion of ischemic stroke patients still have residue symptoms even after thrombolysis with rt-PA or endovascular thrombectomy. Numerous clinical studies have shown that XXMD is an effective alternative therapy not only at the acute stage, but also at the chronic sequelae stage of ischemic stroke. Modern techniques have isolated groups of compounds from XXMD which have shown therapeutic effects, such as dilating blood vessels, inhibiting thrombosis, suppressing oxidative stress, attenuating nitric oxide induced damage, protecting the blood brain barrier and the neurovascular unit. However, which of the active compounds is responsible for its therapeutic effects is still unknown. Emerging studies have screened and tested these active compounds aiming to find individual compounds that can be used as drugs to treat stroke. The Eupatorium plant has been well used in medication and as a decorative plant. Some studies have reported that this herb has biochemical compounds, such as sesquiterpenes, phenolics, polysaccharides, and pyrrolizidine alkaloids. Thus, it has pharmacological effects, including antifungal, antibacterial, cytotoxic, and antinociceptive properties, that can be utilized for cosmetic purposes. However, only a few published works have summarized the active compounds and the application of Eupatorium plants as cosmetic agents. Therefore, this article aims to review the application of Eupatorium plants as a potential cosmetic agent. The active compounds of Eupatorium are contained in the whole plant, as well as the stems, leaves, roots, and aerial parts (flower, fruit, and seeds). In terms of cosmetic applications, the activities of Eupathorium are antioxidant, anti-tyrosinase, anti-melanin/melanogenesis, anti-acne, and anti-inflammatory. This review aims to contribute to a better understanding for expanding the utilization of this plant for cosmetic purposes by using these active compounds.

Studies have shown that approximately two-thirds of the plant species in the world have some medicinal value. Artocarpus lakoocha is a synonym for Artocarpus lacucha and is a plant that can be found in Indonesia. This medicinal plant has been used to treat many diseases. (1) Objective: This article discusses the scientific investigations carried out on A. lacucha, namely the plant’s chemical content, pharmacological activity, and active compounds. (2) Methods: The design of this study was based on an article that was a review of previous research. A search for relevant publications over the past ten years (2012–2022) using data from Pubmed, Proquest, Ebsco, ScienceDirect, and Google Scholar resulted in the discovery of 369 articles. (3) Results: Fifty relevant articles investigate A. lacucha’s substances and their applications in the health field. The presence of secondary metabolites and bioactive compounds has been reported, which is evidence that A. lacucha possesses antidiarrheal, immunostimulant, anticholesterol, and hepatoprotective agents. (4) Conclusions: Mobe (A. lacucha) is a plant native to North Sumatra, Indonesia. This plant is efficacious as an antioxidant, antibacterial, antidiarrheal, anti-inflammatory, analgesic, antinociceptive, schistosomicidal, hepatoprotective, neuroprotective, cytotoxic, antiglycation, and anticholesterol, and can also be used for anti-aging and wound healing. In addition to its various benefits, it turns out that this plant also has many active compounds that are useful to the health sector, especially the pharmaceutical field.

Toona sinensis is a traditional vegetable with rich nutrition and wide biological activity. In our previous study, an active composition against α-amylase was screened from T. sinensis by ligand fishing. To investigate the effect of T. sinensis on other targets of antidiabetes, in this work, α-glucosidase was immobilized on polydopamine-coated magnetic nanoparticles to further screen the compounds against α-glucosidase by ligand fishing. The enzyme activity test showed that the 70% ethanol extract of T. sinensis (crude sample) had weak α-glucosidase inhibitory activity, but after extraction with different solvents, the ethyl acetate extract showed the highest α-glucosidase inhibitory activity. After immobilization of α-glucosidase on polydopamine (PDA)-coated magnetic nanoparticles, the properties of the enzyme were characterized and determined, and then quercetin-3-O-α-L-rhamnopyranoside was screened by ligand fishing and isolated by high-speed counter-current chromatography using the two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (1:5:1:5, v/v). The results of enzymes assay showed that the screened compound (IC50 value of 374.94 ± 4.35 μg/mL) possessed moderate α-glucosidase inhibitory activity. The presented results not only proved that ligand fishing based on polydopamine-coated magnetic α-glucosidase was an effective method for the rapid discovery of natural active substances, but also further strengthened the evidence of antidiabetic effect of T. sinensis. Green nanoparticles are receiving great attention due to their broad fields of application. Plant extracts are promising sources, since the synergizing process is easy and cost-effective. Green synergism offers safer nanomaterials for both human health and the environment. The antimicrobial application of prepared nanoparticles is due to special capabilities at the nanoscale size. Green zinc oxide nanoparticles (ZnONPs) from lemongrass and turmeric revealed good antibacterial properties. Antibacterial active compounds were extracted from lemongrass and turmeric using methanol as a solvent. The extracts were introduced to zinc oxide solution to create synergized green ZnONPs and tested for their antibacterial properties. The samples were characterized by means of XRD, SEM, FTIR, and GC-MS. The zones of inhibition of synergized green ZnONPs were successfully measured using the disc diffusion method. The findings using gram-negative bacteria Escherichia coli. showed a higher antibacterial inhibition zone, with a diameter of 1.6 cm. The gram-positive bacteria, Staphylococcus aureus., showed low antibacterial inhibition. The addition of ZnO had positively revealed greater inhibition.
