Abstract

Rove beetle, Paederus fuscipes Curtis is a natural predator of several crop pests in the agriculture ecosystem, however, their high intrusion into human settlements caused them to become public health concern due to Paedarus dermatitis infection among humans. The entomopathogenic effectiveness of Metarhizium anisopliae Mechnikov was tested as biological control towards adults Paederus fuscipes by inoculating on soils. The mortality of P. fuscipes was observed and data were subjected to analysis using ANOVA and Kaplan-Meier method. Results show that P. fuscipes tested with the highest concentration at 1.3 × 1010 conidia/mL exhibited the shortest mean mortality time at 11.0 ± 2.5 days and survival time of 7.6 ± 0.7 days, yet the secondhighest concentration exhibited at 2.2 × 109 showed mean mortality of 18.4 ± 4.2 days and survival time of 11.9 ± 0.8 days. Log Rank (Mantel-Cox) pairwise comparison indicated the significant differences between the highest concentration of 1.3 × 1010 with the control (χ2 = 62.3, df=1 p<0.0005). Both mean mortality time and survival time of P. fuscipes showed inconsistent trends from the highest concentration of M. anisopliae towards the lowest. Pathogenicity was observed at the concentrations of 106, 109, and 1010 after performing Koch's postulates. The results were unexpected but could indicate that M. anisopliae has the potential to be a biocontrol agent at a higher concentration. [ABSTRACT FROM AUTHOR]

Abstract

The rove beetle, Paederus fuscipes Curtis has caused tremendous dermatitis once unintentionally crushed against human skin due to their potent toxic released called paederin produce by the endosymbiont bacteria. The genus Pseudomonas was hypothesized to be the endosymbiont bacteria involved in the production of paederin in Paederus. However, no extensive studies have been performed to investigate the existence of other bacteria, and the relationship between the endosymbiont counts amongst male and female P. fuscipes beetles. We found that P. fuscipes harbours surprising numbers of bacteria diversity with a total of 20 different bacterial species identified. Bacillus, Serratia, Staphylococcus, and Pseudomonas bacteria were the dominated genus found in all collected beetles from all light trap locations. However, only Pseudomonas aeruginosa Migula is the medically important species and is commonly known to cause severe dermatitis lesions to humans. Our results revealed the P. aeruginosa counts cultured on Pseudomonas medium was found higher total mean bacteria counts in P. fuscipes females (2.58 x 105 ± 2.73 x 104 cell) compared to male beetles (5.68 x 104 ± 3.70 x 10³ cells). We demonstrated that a large diversity of bacterial community has been isolated from both male and female P. fuscipes, however P. aeruginosa colonies were frequently detected in females compared to male beetles. These results can be used in future studies to investigate the possible impact of these bacterial counts on the concentration of paederin produced in P. fuscipes. [ABSTRACT FROM AUTHOR]

Abstract

Background: Mating is a physiological process of crucial importance underlying the size and maintenance of mosquito populations. In sterile and incompatible insect technologies (SIT and IIT), mating is essential for mass production, persistence, and success of released individuals, and is a central parameter for judging the effectiveness of SIT/IIT programs. Some mosquitoes have an enormous reproductive potential for both themselves and pathogens and mating may contribute to persistence of infection in nature. As Aedes albopictus can transmit flaviviruses both sexually and horizontally, and as infected insects are usually derived from laboratory colonies, we investigated the implications of mating between a long-term laboratory colony of Ae. albopictus and wild populations. Methods: Through a series of mating experiments, we examined the reproductive outcomes of sexual cross-affinity between laboratory-raised and wild adults of Ae. albopictus. Results: The results indicated appreciable mating compatibility between laboratory-reared and wild adults, and equivalent levels of egg production among reciprocal crosses. We also observed comparable larval eclosion in lab females mated with wild males, and increased adult longevity in female offspring from wild females|×|laboratory males crosses. Conclusions: Taken together, these data suggest that Ae. albopictus can preserve its reproductive fitness over a long period of time in the laboratory environment and has valuable attributes for SIT application. These observations together with the ability to successfully inseminate heterospecific females indicate the potential of Ae. albopictus to act as an ecological barrier if non-sterilized males are massively released in areas occupied by Aedes aegypti. The observed substantial reproductive fitness combined with the capability to reproduce both, itself and viruses illustrates the potential of Ae. albopictus to pose a serious threat if infected and released accidentally.

AbstractPlants produce a rich source of chemicals, drugs and secondary metabolites that can act as insect killer. The larvicidal activity of various part of Ipomoea cairicaextracts was evaluated against Culex quinquefasciatus. The highest percentage of yield extract was given by Soxhlet with acetone solvent for all of the different plant parts. While, maceration technique only provided 1/3 of the yield compared to Soxhlet technique. Using 24 h larvicidal bioassay technique, third instar larvae of Cx. quinquefasciatuswere exposed to concentrations ranging from 10 to 600 ppm of the acetone and methanol extracts of I. cairicafrom leaves, stem and flower bud. Acethonilic extract of I. cairicaleaves was extracted using a Soxhlet apparatus which exhibited the most promising larvicidal activity against Cx. quinquefasciatuslarvae with significantly lower LC50value of 110.65 ppm, meanwhile methanolic leaves extract exhibited LC50value of 143.47 ppm. Larvae exposed to the I. cairicaextracts for 24 h showed morphological malformation that included a detachment of thorax, disruption of the digestive tract and pigmentation of cuticle on their death. Thus, suggested that I. cairicaplant extract, especially acethonilic leaves extract can be considered as one of plant-derived insecticides for control of Cx. quinquefasciatus,a vector of filariasis.

Abstract

To understand the effects of fish predator's kairomones on Aedes mosquitoes' oviposition, we established an experiment using gravid Aedes females. Kairomones concentrations were established using Hampala macrolepidota. One individual fish was placed inside containers with varying water levels (1 L, 5 L, and 10 L of water). The fish were kept in the containers for 24 hours and were removed immediately at the start of each trial in order to have the kairomones remnants. Twenty gravid adult females of Aedes aegypti and Aedes albopictus were allowed to lay eggs on oviposition site with various treatments: (1) control without any kairomones; (2) kairomone remnant in 1 L of water; (3) kairomone remnant in 5 L of water; and (4) kairomone remnant in 10 L of water. There are significant differences between the numbers of eggs laid by both Aedes species for each different treatment (F = 9.131, df = 16, p<0.001). However, fewer eggs were laid by Ae. albopictus compared to Ae. aegypti in the presence of kairomone remnants. This suggested that Ae. albopictus are significantly affected by the kairomones itself and have ability to detect the residual kairomone presence from H. macrolepidota. [ABSTRACT FROM AUTHOR]

Abstract

The interaction between plants and insects is dynamic, and may favour either the plant or the insect. Plant chemicals are deeply implicated in this relationship and influence insect behaviour. Here, we investigated the oviposition behaviour response of Culex quinquefasciatus mosquitoes based on the colour cues produced by Ipomoea cairica leaves extract. In this study, two sets of oviposition choice experiments were conducted: (1) single solution in a cage; and (2) multiple concentration solutions in a cage. In the single solution experiment, only 1 available oviposition site was offered to 5 gravid females and in the multiple concentration tests, 4 available oviposition sites were offered to 20 gravid females. The tested concentrations were set up at 100 mL of: (1) control (distilled water only); (2) 50 ppm; (3) 150 ppm; and (4) 300 ppm of I. cairica plant extracts. The highest concentration of 300 ppm appeared to show the highest intensity with the darkest colour followed by 150 ppm and 50 ppm concentrations. More gravid females were found drowned in the highest concentration, 300 ppm of acethonilic leaves extract, compared to 150 ppm and 50 ppm of the tested extract. No eggs were found in all tested solutions. The studied extract was found to effectively attract gravid Cx. quinquefasciatus females and subsequently cause mortality and inhibit egg deposition. The interference caused by the acethonilic extract of I. cairica on the oviposition activity of Cx. quinquefasciatus can result in better control of the vector insect. [ABSTRACT FROM AUTHOR]
