The relevant social and economic costs associated with aging and neurodegenerative diseases, particularly Alzheimer’s disease (AD), entail considerable efforts to develop effective preventive and therapeutic strategies. The search for natural compounds, whose intake through diet can help prevent the main biochemical mechanisms responsible for AD onset, led us to screen hops, one of the main ingredients of beer. To explore the chemical variability of hops, we characterized four hop varieties, i.e., Cascade, Saaz, Tettnang, and Summit. We investigated the potential multitarget hop activity, in particular its ability to hinder Aβ1− 42 peptide aggregation and cytotoxicity, its antioxidant properties, and its ability to enhance autophagy, promoting the clearance of misfolded and aggregated proteins in a human neuroblastoma SH-SY5Y cell line. Moreover, we provided evidence of in vivo hop efficacy using the transgenic CL2006Caenorhabditis elegans strain expressing the Aβ3−42 peptide. By combining cell-free and in vitro assays with nuclear magnetic resonance (NMR) and MS-based metabolomics, NMR molecular recognition studies, and atomic force microscopy, we identified feruloyl and p-coumaroylquinic acids flavan-3-ol glycosides and procyanidins as the main anti-Aβ components of hop.

- Abstract
Genetic discoveries of Alzheimer’s disease are the drivers of our understanding, and together with polygenetic risk stratification can contribute towards planning of feasible and efficient preventive and curative clinical trials. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer’s disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer’s disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer’s disease.

- Abstract
Introduction Multicentre preclinical randomised controlled trials (pRCT) are emerging as a necessary step to confirm efficacy and improve translation into the clinic. The aim of this project is to perform two multicentre pRCTs (one in rats and one in mice) to investigate the efficacy of remote ischaemic conditioning (RIC) in an experimental model of severe ischaemic stroke. Methods and analysis Seven research laboratories within the Italian Stroke Organization (ISO) Basic Science network will participate in the study. Transient endovascular occlusion of the proximal right middle cerebral artery will be performed in two species (rats and mice) and in both sexes. Animals will be randomised to receive RIC by transient surgical occlusion of the right femoral artery, or sham surgery, after reperfusion. Blinded outcome assessment will be performed for dichotomised functional neuroscore (primary endpoint) and infarct volume (secondary endpoint) at 48 hours. A sample size of 80 animals per species will yield 82% power to detect a significant difference of 30% in the primary outcome in both pRCTs. Analyses will be performed in a blind status and according to an intention-to-treat paradigm. The results of this study will provide robust, translationally oriented, high-quality evidence on the efficacy of RIC in multiple species of rodents with large ischaemic stroke. Ethics and dissemination This is approved by the Animal Welfare Regulatory Body of the University of Milano Bicocca, under project license from the Italian Ministry of Health. Trial results will be subject to publication according to the definition of the outcome presented in this protocol.

Aims: Mitochondria are very dynamic organelles. Mitochondrial dynamics (alternation of fission/fusion cycles and their axonal transport) and biogenesis are crucial for neuronal homeostasis and synaptic function; moreover, alteration of the balance between fission and fusion can determine the fate of neuronal cells. Fission processes require the intervention of Fis1 and the active form of Drp1, whereas fusion is primarily regulated by Opa1 and Mfn1/2. We here focused on correlation between oxidative stress and mitochondrial dynamics and biogenesis. Materials and Methods: We compared the effect of hydrogen peroxide and Rotenone (a neurotoxin that inhibits the Complex-I of electrons transport chain), on primary cortical neurons or models of neurodegeneration based on neuroblastoma or PC12 cells. Protein levels and distribution (cytoplasm, mitochondria or nuclei) were assessed by western blots and immunofluorescence imaging. Results: Exposure of neurons or neuroblastoma to Rotenone determined a dose-dependent (200-400-800 nM) decrease of P-Drp1 and a biphasic trend of Opa1 and Mfn2 in a time-dependent manner (1-3-6-24h). In PC12 cells, alterations of mitochondrial proteins in response to hydrogen peroxide was paralleled by differential distribution (cytoplasm/nuclei) of oxidative-stress sensors (DJ-1, Nrf2) and proteins regulating mitochondrial biogenesis (mtTFAM and PGC1a), while determining a dose- and time-dependent increase of ROS and ATP depletion. Interestingly, human fibroblasts showed similar changes in response to rotenone but a different trend in response to Amyloid-beta oligomers. Discussion/Conclusions: These studies suggest 1) the existence of a complex cross-talk underlying mitochondrial dynamics and biogenesis, 2) alteration of mitochondrial dynamic proteins as biomarkers of mitochondrial dysfunction in neurodegenerative processes.

Objective: Abeta leads to neurodegeneration and memory impairment via critical signal transduction processes, both at central and peripheral level. Studying the oligomeric-Abeta toxicity could helpful to clarify these phenomena, and to develop therapeutic and effective prevention technologies. Nanoparticles (NPs) biocompatibility and their ability to modulate Abeta toxicity were investigated in primary cultures from human fibroblasts. Methods: Control fibroblasts were incubated with Abeta oligomers and NP, functionalized or not with a ApoE monomer (mApo). By MTT assay we evaluate energetic metabolism and cell viability. Western Blot and p-Elisa were performed to evidence any modulation of MAPK and Tau hyperphosphorylation. By uptake studies and confocal images, we investigated the involvement of NPs in modulating the Abeta-induced excitotoxicity and their interaction with oligomers. Amyloid-beta (Abeta) affects cellular functions leading to neurodegeneration and memory impairment via critical signal transduction processes, including also the ERK1/2 pathway. We used fibroblasts from Alzheimer’s disease (AD) patients and age-related controls to investigate possible alterations of MAPK-pathway associated to the various stages of disease that might be helpful to clarify any molecular mechanisms involved in this systemic pathology. Phosphorylated p38- and JNK-MAPK were observed in fibroblasts from MCI (mild cognitive impairment) subjects, AD patients and in oligomeric-Abeta treated cells from healthy subjects, without any correlation with disease severity. On the other hand, ERK1/2 phosphorylation was reduced in fibroblasts from mild and moderate AD, compared to severe AD patients. Also in Abeta-treated fibroblasts from control subjects, a similar decrease was shown. In MCI subjects, a trend to phospho-ERK1/2 decrease was observed too. A specific correlation was demonstrated between ERK1/2 phosphorylation and disease severity.When the four converter-MCI cases were considered, the statistical significance is improved and correlation is confirmed. Moreover, ERK phosphorylation has been involved in hyper-phosphorylation of tau, which is evident before deposition of Abeta fibrils, through the activation of p70S6-kinase. In addition, we show that the ERK pathway is specifically involved in EAAT1 molecular translation, modulating its promoter and thus impairing glutamate transport, as described in fibroblasts from AD patients. These results suggest that the ERK signaling could be of great importance for the study of the pathogenesis and the disease severity, and to investigate new treatments in AD

- Abstract
Objective: Abeta leads to neurodegeneration and memory impairment via critical signal transduction processes, both at central and peripheral level. Studying the oligomeric-Abeta toxicity could helpful to clarify these phenomena, and to develop therapeutic and effective prevention technologies. Nanoparticles (NPs) biocompatibility and their ability to modulate Abeta toxicity were investigated in primary cultures from human fibroblasts. Methods: Control fibroblasts were incubated with Abeta oligomers and NP, functionalized or not with a ApoE monomer (mApo). By MTT assay we evaluate energetic metabolism and cell viability. Western Blot and p-Elisa were performed to evidence any modulation of MAPK and Tau hyperphosphorylation. By uptake studies and confocal images, we investigated the involvement of NPs in modulating the Abeta-induced excitotoxicity and their interaction with oligomers. By commercial kit, malondialdehyde levels were tested. Results: NPs do not affect cell viability, they prevent the Abeta-induced deficit of mitochondrial activity (p<0.001) and MAPK phosphorylation (p<0.001). mApo cross cell membrane, bind oligomers and modulate glutamate uptake in a dose dependent manner (p<0.01); they counteract the increase of Abeta–induced lipid peroxidation (p<0.002) and Tau-hyperphosphorylation (p<0.0001). Conclusion: These results suggest that NPs could represent a useful tool to antagonize Abeta oligomers toxicity, counteracting the cellular stress, protein epigenetic regulation and glutamate cytotoxicity. The modulations of MAPK pathways, altered by Abeta oligomers, might clarify any molecular mechanisms that mirror those involved in the early AD phases, and NPs may be useful to study new potential therapeutic strategies.

- Abstract
Amyloid-beta (Abeta) affects cellular functions leading to neurodegeneration and memory impairment via critical signal transduction processes, including also the ERK1/2 pathway. We used fibroblasts from Alzheimer’s disease (AD) patients and age-related controls to investigate possible alterations of MAPK-pathway associated to the various stages of disease that might be helpful to clarify any molecular mechanisms involved in this systemic pathology. Phosphorylated p38- and JNK-MAPK were observed in fibroblasts from MCI (mild cognitive impairment) subjects, AD patients and in oligomeric-Abeta treated cells from healthy subjects, without any correlation with disease severity. On the other hand, ERK1/2 phosphorylation was reduced in fibroblasts from mild and moderate AD, compared to severe AD patients. Also in Abeta-treated fibroblasts from control subjects, a similar decrease was shown. In MCI subjects, a trend to phospho-ERK1/2 decrease was observed too. A specific correlation was demonstrated between ERK1/2 phosphorylation and disease severity.When the four converter-MCI cases were considered, the statistical significance is improved and correlation is confirmed. Moreover, ERK phosphorylation has been involved in hyper-phosphorylation of tau, which is evident before deposition of Abeta fibrils, through the activation of p70S6-kinase. In addition, we show that the ERK pathway is specifically involved in EAAT1 molecular translation, modulating its promoter and thus impairing glutamate transport, as described in fibroblasts from AD patients. These results suggest that the ERK signaling could be of great importance for the study of the pathogenesis and the disease severity, and to investigate new treatments in AD

Imatinib Mesylate is a competitive tyrosine-kinase inhibitor used in the treatment of multiple types of leukaemia, including Ph+ chronic myelogenous leukaemia (CML). The recent report that Imatinib inhibits gamma-secretase activating protein (gSAP), reducing beta-amyloid- (Abeta) production without affecting Notch-1 cleavage has led to the proposal of a possible application of this drug in AD therapeutics. In fact, one major drawback of Semagacestat, a direct inhibitor of gamma-secretase, was identified in the low Abeta/Notch therapeutic index, with the consequent failure of the phase III trial. Imatinib peripheral administration (20 mg/kg/day) was able to reduce Abeta content in mouse brain. Usually, leukaemia patients receive an average oral dose of 400 mg/day (∼5–6 mg/kg/day). In this longitudinal study we investigated if Imatinib administration in 10 leukemic patients could reduce plasma Abeta1–40 levels. Patients’ blood was sampled prior to treatment (time T0) and after 15, 30, and 60-to-90 days of treatment. Abeta1–40 plasma levels were assessed by ELISA as previously described. No significant reduction in plasma Abeta1–40 was observed at any time point. In fact, ratios to T0 of Abeta1–40 T15-, T30- and T90 were: 1.07 ± 0.39, 1.37 ± 0.59, and 1.48 ± 0.61, respectively, indicating no decrease with Imatinib. While these results are negative at standard dosing of Imatinib for CML, they do not exclude the possibility that Abeta lowering effects might be present at higher doses of this drug or with second generation tyrosine-kinase inhibitors, such as Dasatinib or Nilotinib, which can cross the blood brain barrier more easily. Further exploration of this issue might lead to advances in AD therapeutics

Donepezil (DNPZ), an acetyl-cholinesterase inhibitor (AChEI), is widely used as a symptomatic drug in AD, given the involvement of cholinergic damage in the onset of memory loss. DNPZ, besides being able to improve acetylcholine (ACh) transmission, might have a neuroprotective effect reducing toxic Abeta fibrils and regulating the immune response. Considering that we previously showed a specific increase of plasma anti-Abeta1–42 antibodies in AD patients treated with AChEI, we evaluated in this study the role of DNPZ in favouring a Th2 phenotype, involved in modulating the immune humoral response. Moreover we hypothesized that this mechanism might be mediated by the a7-nicotinic ACh receptor expressed in lymphocytes. 60 patients with mild or moderate AD, either treated (n=22) or not (n=38) with DNPZ, and 30 controls were enrolled. AD DNPZ+ showed significantly higher plasma levels of anti-Abeta antibodies than DNPZ− (+40%) and lower levels of Abeta 1–42 than controls (−50%). In a subgroup of subjects, GATA-3, a transcription factor involved in Th2 differentiation, and a7nAChR expression was evaluated. No differences were found in GATA-3 mRNA in AD DNPZ+ when compared to DNPZ− and controls. On the opposite, we found by chromatine immunoprecipitation a significant three-fold increase of the association of GATA-3 with the IL-5-promoter in DNPZ+ patients, which also showed a significant twofold increase in a7-nAChR mRNA, with respect to DNPZ-ones. In vitro analyses demonstrated that the capacity of DNPZ to modulate GATA-3 expression is mediated by a7nAChR, since MLA, a specific antagonist, prevents it. Further studies are needed to better understand the role of DNPZ in modulating the immune response against Abeta, possibly ameliorating therapeutic strategies for AD

Background: Background There is huge body of evidences concerning the detrimental role of glutamate excess in Multiple Sclerosis (MS). Excessive neuronal stimulation, due to deficient removal of glutamate and\or excessive release by activated Tcell and microglia, may trigger an enzymatic cascade leading to cell death. For MS, however, is still not cleared-up whether this imbalance relies to a glutamate overproduction or to its reduced clearance. Our group previously reported an impairment of glutamate transporter (EAAT) in neurodegenerative diseases at platelets (P) level, mirroring that present at central nervous system. Design/Methods: Design/Methods: We investigated P-EAAT activity as Glutamate-uptake marker, in primary progressive (PP=17), secondary (SP=16), relapsing remitting (RR=25) and benign (BB=12) MS population (=70) and 60 Healthy controls (HC). Sodium/energy-dependent-glutamate-uptake was studied in platelets measuring tritiated-Glu by beta-counter, following the original method of Mangano and Schwarcz, and activity and affinity were expressed by Vm and Km. Glutamate-uptake (GLU-up) impairment and an associated excitotoxic neuronal damage are widely documented in neurodegenerative diseases, as Multiple Sclerosis (MS) can be considered. Several factors negatively affect GLU-up, among them is the oxidative stress (OXS). Considering the suggested involvement of both GLU-up and OXS in neurodegenerative mechanisms of MS, we investigated a possible relationship between these factors. Fifty MS patients (age 33–64 years) with different disease course and 55 controls (HC, age 33–60 years) were studied for several serum markers of OXS and antioxidant factors, namely: coenzyme-Q10, total, oxidized and reduced forms of glutathione, malondialdehyde, reactive-oxygen-species, anti-oxidized-low-density lipoproteins antibodies (Anti-oxLDL), and anti-oxidant-power. Activity and affinity of glutamate platelet transporters were also studied. Coenzyme-Q10 and malondialdehyde were determined by an isocratic HPLC with UV detection, glutathione by HPLC with fluorescence detection, reactive-oxygen-species were photometrically quantified, anti-oxLDL were detected by ELISA, anti-oxidant-power was measured by the Cu++ reduction, subsequently detected by a chromogenic reaction. Sodium/energy-dependent-glutamate-uptake was studied in platelets measuring tritiated-Glu by b-counter and normalized to proteins’ amount. Reduced GLU-up values was found in MS compared to HC (19 ± 1.8; 28 ± 2.4 pmoles/mg prot/30 min ± SEM; p B 0.003), even if in benign MS subgroup no uptake decrease was observed. Coenzyme-Q10, one of the most powerful antioxidant was lower in MS (517 ± 35.7 lg/L) compared to HC (626 ± 27.7 lg/L p.02). While oxidated- glutathione levels were higher in SM (62 ± 3.7 U/ml) versus HC (48 ± 4.3U/ml, p = 0.01). A trend for higher antioxLDL in MS patients (35 ± 2.5 U/ml) versus HC (31 ± 4.7 U/ml) was found as well. We described a reduced GLU-up activity in MS patients compared to HC; Vm was significantly decreased (p.003), whereas Km was unaffected. Hence, we might suggest that the uptake impairment is associated to a reduced GLU- transporter expression or to a structural damage. OXS maybe one of the mechanisms involved in both these processes. Interestingly, we found a higher antioxidant ‘‘protection’’ in benign MS, where GLU-up was increased versus RR and SP SM (p.05). Correlation analyses between all OXS markers measured and GLU-up as well as across different courses of MS patients will be discussed

- Abstract
The neuroepigenome, i.e., the epigenome of the nervous system, has become interesting for therapeutics in the last years due to widespread availability of dedicated drugs. A pivotal role for neuroepigenetics is certainly implied, both in physiology and pathology, by the highly dynamic structural and functional rearrangements that constantly occur into the nervous system, globally known as plasticity. Moreover, the idea that the pathophysiology of several neuropsychiatric disorders might involve epigenetic mechanisms is increasingly taking place due to accumulating experimental data and by the evidence of a synergistic interaction between genes and environment beneath most sporadic forms of these diseases. In this paper we will review the available evidence on the use of epigenome-modifying drugs in the field of neuropsychiatry, shortly describing for each disease the underlying assumptions of an epigenetic dysregulation. © 2014 Bentham Science Publishers.
