Context: The population's use of halophyte plants in traditional medicine is a fundamental component of the exploration of natural remedies. Aims: To analyze a list of halophyte plants and their ethnobotanical characteristics, with a focus on those most commonly used in traditional medicine in the southeast of Algeria (Biskra). Methods: The ethnobotanical surveys were conducted between 2017 and 2019 by interviewing 120 informants using a questionnaire and applying a semi-structured interview methodology. The data was analyzed using ethnobotanical statistical tools, including the informant consensus factor (ICF), citation frequency (CF), use value (UV), family importance value (FIV), relative frequency of citation (RFC), plant part value (PPV), and relative importance (RI), as well as statistics analysis (ANOVA) to compare the use of medicinal plants according to age, gender, and education level. Results: The results of the survey enabled us to identify 12 species distributed across eight different families. The most common family was that of the Amaranthaceae (four species), while the most frequently utilized parts of plants were leaves (PPV = 0.4). Furthermore, decoctions were the most commonly prepared form (33.39%). The highest ICF values were observed for the endocrinological and digestive disease groups (ICF = 0.97). The statistical analysis (ANOVA) revealed a significant difference in the knowledge of the use of halophyte plants in traditional medicine according to age and level of education. Conclusions: The findings of this research suggest that the halophyte flora in our arid study area may offer a promising source for the discovery of new drugs, a prospect that requires further investigation and exploration.

Vaginal cancer, rarest form of gynaecological cancer (0.6%) has been found to be associated with long term use of vaginal ring pessary, which have long been used in alternative management of pelvic organ prolapse (POP) other than surgery. A survey was done in remote areas of Cox's bazar, refugee camp in 2019 where out of 52 cases of pelvic organ prolapse, 7 cases found to use vaginal ring pessary for long and out of 7, vaginal cancer developed in 4 cases. All of them were aged women above 60 and had defaulted follow up presented with vaginal bleeding, varying discharge and pain of varying intensity. One patient had only local lesion -surgical removal followed by radiotherapy and 2nd one, 68 years, received neo-adjuvant therapy and died before planned surgery. The 3rd one, aged 79, did not come for follow up once diagnosis vaginal cancer done and advised for definitive surgical treatment, family refused operation and she passed away after 2 years at 81 years. The 4th one declined further investigations and any form of treatment when diagnosed with vaginal cancer. So, all patients of pelvic organ prolapse should be informed thoroughly (both verbal and written informed consent) before inserting pessary for potential long term serious consequences where strict compliance must be ensured for subsequent regular follow up and biopsy should be mandatory for any suspicious chronic vaginal ulcers among them. Objective: To test the sensitivity of locally available strips for the detection of Plasmodium Vivax and Plasmodium Falciparum species at a tertiary care hospital. Study Design: Cross-sectional study. Place and Duration of Study: Armed Forces Institute of Transfusion, Rawalpindi Pakistan, done over one week during Nov 2021. Methodology: Four different commercially available brands of Malaria Rapid Diagnostic Tests were taken. They were tested against venous blood sample taken from only one patient who had lab detected Malaria. Light microscopy showed trophozoites of both Plasmodium Vivax and Falciparum. Nine serial dilutions of different strengths were made from blood sample. These dilutions were tested against commercially available four different Rapid Diagnostic Tests strips. Results: Malaria Rapid Diagnostic Tests kits from all the four brands which were commercially available showed positive results for Plasmodium vivax up to 1:512 dilutions. However, only one brand Rapid Diagnostic Tests kits showed positive results with Plasmodium falciparum in 1:2 dilutions, in addition to Plasmodium vivax. Conclusion: The commercially available Rapid Diagnostic Tests Immunochromatographic Technique have high sensitivity in diagnosing Malaria, but are not resolute when it comes to speciation, particularly for Plasmodium falciparum.

In the phosphate industry, the use of certain flotation reagents can cause severe water pollution and have a negative effect on the environment and human health. Therefore, the development and utilization of eco-friendly flotation reagents are essential for the sustainable production of phosphate ores and phosphate fertilizers. In this work, an economical and environmentally friendly mixed collector based on soybean and sunflower oils soaps was used to improve the grade and recovery of phosphate flotation. The results obtained from the flotation showed that the best composition of the mixed collector was 1:1 of the mass ratio of soybean and sunflower oils. In addition, the results showed that a low collector mass, medium solid content combined with an acidic medium resulted in a better relative floatability. Multi-response optimization was applied to optimize the process variables for both responses simultaneously using the Face Central Composite Design (FCCD). The effect of three independent variables such pH, collector mass, and solid content on the % P2O5 grade, and % recovery was studied. The results revealed that pH and collector mass had the most significant effect of the % grade response, while collector mass had a significant effect only on % recovery. It was found that more than 29.57% for grade content and 98.82% for the recovery can be achieved at the optimum conditions of low pH (pH 4) and low collector mass (0.13 g) and solid content (17.97 wt%). X-ray diffraction of the resulting solids confirms that the phosphate phase remains in the sink and that the gangue flows into the floated part.

This study presents the first evidence of the mycorrhizal compatibility between the edible desert truffle Tirmania nivea and the valuable fruit tree Argania spinosa. Seed germination trials demonstrated that soaking pre-treatment of argan seeds in hydrogen peroxide (9%) for five days combined with the application of a fungicide treatment on an inert sowing material maximized the seed germination of this tree species. The mycorrhizal synthesis was conducted under greenhouse conditions by inoculating, in vivo, the host plant seedlings with spores of T. nivea. The growth and mycorrhizal status of A. spinosa was assessed 15.5 months after inoculation. The desert truffle mycorrhization significantly promoted all the investigated morphological parameters of growth and improved the physiological performances of the host plant through enhancing plant water status and chlorophyll concentration. The mycorrhizal symbiosis led to the formation of typical desert truffle endomycorrhizae with intracellular coils. The resistance of A. spinosa to the harsh environmental conditions of desert habitats makes it a potential candidate for cultivation of desert truffles.

This study reports the thermal analysis and species transport to manifest non-Newtonian materials flowing over linear stretch sheets. The heat transfer phenomenon is presented by the Cattaneo–Christov definition of heat flux. Mass transportation is modeled using traditional Fick’s second law. In addition, the contribution of Joule heating and radiation to thermal transmission is also considered. Thermo-diffusion and diffusion-thermo are significant contributions involved in thermal transmission and species. The physical depiction of the scenario under consideration is modeled through the boundary layer approach. Similar analysis has been made to convert the PDE model system into the respective ODE. Then, the transformed physical expressions are calculated for momentum, thermal, and species transport within the boundary layer. The reported study is a novel contribution due to the combined comportment of thermal relaxation time, radiation, Joule heating, and thermo-diffusion, which are not yet explored. Several engineering systems are based on their applications and utilization.

The phytoremedial potential of three halophytes and the role of arbuscular mycorrhizal fungi (AMF) isolated from heavy metal contaminated soil under cadmium stress were studied. The plants were treated with different Cd concentrations (0, 50, 200, and 400 µM) with and without AMF inoculation. On the plant growth, AMF inoculation to all species resulted in increased biomass, shoots and roots length, and leave number compared to control (non-inoculated) plants, especially in Limbarda crithmoides, where no mortality was detected neither in controls nor inoculants. Furthermore, L. crithmoides and Atriplex canescens recorded higher values of relative mycorrhizal dependency and water content. The arbuscular mycorrhizal association was observed in all host plants and was not affected by cadmium. Our results indicate that studied halophytes can tolerate high Cd concentrations due to the support of AMF, particularly in Atriplex halimus association with AMF in pot experiment which was recorded for the first time.

The species concept within the genus Picoa Vittad. is here revisited in light of new molecular and ecological data obtained from samples collected throughout the Mediterranean basin. Two highly diverse widespread clades and four additional minor lineages were significantly supported by three genes dataset (ITS, 28s LSU and RPB2) inferences for 70 specimens. The two widespread clades occur in very different geographical and ecological areas associated with exclusive host plants in the genus Helianthemum. SEM study of spore surface morphology in these lineages revealed the existence of smooth ascospores in the majority of these clades. However the most frequent lineage in Europe and coastal North Africa displayed either smooth or verrucose spores. Hence this morphological criterion cannot be reliably used to discriminate between the different clades. In addition, SEM observations made on ascospores from several original collections of P. juniperi and P. lefebvrei supported the hypothesis that ornamentation depends on the degree of maturity in some of these lineages. Geographical and ecological, rather than morphological data are here suggested as the most useful characters to separate the different lineages in Picoa. Further studies focusing on these features are needed before the names P. juniperi and P. lefebvrei can be unambiguously linked with the genetic lineages observed.

Barley Net Blotch caused by Pyrenophora teres f. teres (Ptt) is one of the most important foliar diseases in north western Algeria. The disease could cause appreciable yield losses under favorable environmental conditions. Studies on pathogen variability and distribution of virulence in Ptt are essential to identify effective sources of resistance to net blotch of barley. In this study, pathogenic variability in 48 isolates of Ptt, collected from different barley-growing areas of north-western Algeria were evaluated using 22 barley genotypes as differential hosts. Trials carried out under controlled conditions confirmed the large variation in the virulence of this fungus. The genotypes exhibited variability in response ranging from complete resistance to high susceptibility. This is the first report of pathogenic variability in Ptt in north western Algeria. The 48 isolates tested were further differentiated into 12 pathotypes according to their reaction on the host. Eight differential cultivars were resistant to all isolates tested, but others were susceptible to one or more of the isolates. The variability in the pathogen and the resistance identified in some genotypes used in this study are being investigated further to develop superior, adapted germplasm for use in barley breeding programs in north western Algeria.

Peganum harmala seeds crude hydro-methanolic extract and their fractions (obtained with ethyl acetate and butan-1-ol) were analyzed and compared for their chemical profiles of alkaloids and polyphenols content. Moreover, their antioxidant, a-glucosidase, acetylcholinesterase, and butyrylcholinesterase inhibitory activities were evaluated. The butan-1-ol fraction revealed the highest total phenolic content and exhibited the highest antioxidant capacity. From the inhibitory enzyme evaluations, it should be highlighted the butan-1-ol fraction inhibitory potential of ɑ-glucosidase (the IC50= 141.18±4μg/mL), which was better than the acarbose inhibitory effect (IC50= 203.41±1.07 μg/mL). The extracts' chemical profile analysis revealed several compounds, in which quercetin dimethyl ether, harmine and harmaline emerged as the major compounds. The different solvents used impacted Peganum harmala seed contents and biological responses. Statistical analysis showed a significant correlation between bioactive compounds and biological activities. Thus, Peganum harmala seeds could be a promising natural source of bioactive compounds at the crossroads of many human diseases, and its cultivation may be encouraged., (© 2024 Wiley‐VCH GmbH.)

The literature of human and other host-associated microbiome studies is expanding rapidly, but systematic comparisons among published results of host-associated microbiome signatures of differential abundance remain difficult. We present BugSigDB, a community-editable database of manually curated microbial signatures from published differential abundance studies accompanied by information on study geography, health outcomes, host body site and experimental, epidemiological and statistical methods using controlled vocabulary. The initial release of the database contains >2,500 manually curated signatures from >600 published studies on three host species, enabling high-throughput analysis of signature similarity, taxon enrichment, co-occurrence and coexclusion and consensus signatures. These data allow assessment of microbiome differential abundance within and across experimental conditions, environments or body sites. Database-wide analysis reveals experimental conditions with the highest level of consistency in signatures reported by independent studies and identifies commonalities among disease-associated signatures, including frequent introgression of oral pathobionts into the gut., (© 2023. Background: In the summer of 2022, the United States faced a nationwide outbreak of mpox, with cases concentrated in sexual and gender minorities who have sex with men. Understanding rates of mpox vaccine uptake and concomitant behavioral change is essential to guide the implementation of targeted public health responses to the potential reemergence of mpox., Methods: Between August 2022 and November 2022, 8551 individuals recruited via geosocial networking apps completed a brief survey that assessed mpox vaccine uptake, intentions to get a mpox vaccine, and behavioral change., Results: In August, 17.4% of participants reported having received at least 1 dose of the mpox vaccine. By November, this prevalence estimate was 35.0%. Black participants were significantly less likely to be vaccinated, and vaccine hesitancy increased among Black participants over time. Among those who had not yet received a vaccination, the intention to get vaccinated decreased over time. We observed trends that coincided with the evolving outbreak, such as decreased worry about mpox and less engagement in risk reduction behaviors over time., Conclusions: Despite a 2-fold increase in mpox vaccine uptake between August 2022 and November 2022 in sexual and gender minorities who have sex with men, disparities in vaccine uptake were observed among Black participants. The extant data suggest that LGBT communities were disproportionately impacted by the economic ramifications of the pandemic and were more likely to report being uninsured throughout the first two years of the pandemic. Additionally, these groups are at heightened vulnerability for several health conditions that require insurance to manage or prevent. Thus, there is a need to assess changes in pandemic-era insurance coverage among these populations. This study uses data collected as part of the Together 5,000 study, a U.S. national, internet-based cohort study of cisgender men, trans men, and trans women who have sex with men. We analyze insurance data across three different assessments between 2019 and 2021, exploring changes in insurance coverage and type. Among our sample, 6.4% lost their insurance in 2020 because of the pandemic. Insurance loss was associated with living in a state that had not expanded Medicaid, race/ethnicity, employment status, and income. Among those who lost their insurance in early 2020, most (59.2%) reported gaining insurance by 2021, with those living in non-expanded states less likely to gain insurance. Finally, those who were uninsured prior to the pandemic were less likely to report gaining insurance by 2021, when compared to those uninsured as a result of the pandemic. This suggests that there are uninsured cisgender gay and bisexual men and transgender individuals that continue to go unreached by policies to assuage uninsurance. The COVID-19 pandemic affects individuals' mental health that can result in fear of getting COVID-19 infection and depression. As there is no prior study available, we evaluated these mental health outcomes and associated factors among the general population and healthcare professionals (HCPs) in Bangladesh. This nationwide cross-sectional study comprised 3388 individuals including 834 HCPs. The measures included socio-demographics, healthcare, and patient-care related information, the Bangla Patient Health Questionnaire, and the Bangla Fear of COVID-19 Scale. Multiple linear regression analyses were performed to identify risk factors. Just over one-quarter of the participants were depressed, and was significantly associated with COVID-19 fear. Regression analyses showed that, both in general population and HCPs, depression and fear of COVID-19 were strongly predicted by being female; however, depression was inversely associated with being married. Particularly, among the HCPs, being restless while examining a patient with flu-like symptoms and while examining a patient returning from abroad was found to be significant predictor for both depression and fear of COVID-19. HCPs who were using single protective equipment for a week had greater depression and those who felt insecure due to the pandemic had a high level of COVID-19 fear. Aim: To assess the prevalence of intestinal helminth infections and associated risk factors among people living in urban slums in Bangladesh., Methods: A cross-sectional study was conducted across three clusters: Dhaka, Sylhet and Chattogram. In total, 360 individuals divided into two groups (school-aged children and adults) provided stool samples and completed a semi-structured questionnaire. Parasitological assessment was performed using the formol-ether concentration technique., Results: Overall, 31.7% (114/360) of participants had helminthiasis, with 13.3% (48/360) having mixed infections. Among the infected participants, school-aged children had a higher rate of infection (41.7%, 75/180) compared with adults (21.7%, 39/180). Ascaris lumbricoides was the predominant parasite, followed by hookworms, Trichuris trichiura, Hymenolepis nana, Enterobius vermicularis and Rhabditis sp. Parasitic infections were significantly associated with type of latrine used, direct exposure to soil, open defaecation, and presence of free-roaming animals., Conclusion: Despite continuous efforts to control helminthiasis, a substantial proportion of the study participants were infected with intestinal helminths. Ignorance of the roles of the environment and animals was influential, and had a negative impact on existing control interventions. Since the emergence of the COVID-19 pandemic, there has been an increasing body of research focused on the effects that measures like stay-at-home orders and social distancing are having on other aspects of health, including mental health and sexual health. Currently, there are limited extant data on the effects of the pandemic on sexual and gender minorities. Between April 15, 2020, and May 15, 2020, we invited participants in an ongoing U.S. national cohort study (Together 5000) to complete a cross-sectional online survey about the pandemic, and its effects on mental and sexual health and well-being (n = 3991). Nearly all (97.7%) were living in an area where they were told they should only leave their homes for essentials. Most (70.1%) reported reducing their number of sex partners as a result of the pandemic. Among the 789 participants prescribed HIV pre-exposure prophylaxis (PrEP), 29.9% said they stopped taking their PrEP entirely, and 14.2% started selectively skipping doses. For those who had been taking PrEP, discontinuing PrEP was associated with having no new sex partners (β = 0.90, 95% CI 0.40-1.40). Among the 152 HIV-positive participants, 30.9% said they were unable to maintain an HIV-related medical appointment because of the pandemic and 13.8% said they had been unable to retrieve HIV medications. Additionally, 35.3% of participants were experiencing moderate to severe anxiety because of the pandemic and 36.7% reported symptoms of depression. In a multivariable logistic regression, reporting a new sex partner in the prior 30 days was significantly associated with being aged 30 or older (vs. not, AOR = 1.21), being Black (AOR = 1.79) or Latinx (AOR = 1.40, vs. white), and being unsure if they had been in close contact with someone diagnosed with COVID-19 (AOR = 1.32, vs. no contact). It was unassociated with COVID-19-induced anxiety, depression, or knowing someone hospitalized with COVID-19. The pandemic has caused disruptions in sexual behavior (partner reduction) as well as difficulties navigating PrEP and HIV care continua. The particularly interdisciplinary nature of human microbiome research makes the organization and reporting of results spanning epidemiology, biology, bioinformatics, translational medicine and statistics a challenge. Commonly used reporting guidelines for observational or genetic epidemiology studies lack key features specific to microbiome studies. Therefore, a multidisciplinary group of microbiome epidemiology researchers adapted guidelines for observational and genetic studies to culture-independent human microbiome studies, and also developed new reporting elements for laboratory, bioinformatics and statistical analyses tailored to microbiome studies. The resulting tool, called 'Strengthening The Organization and Reporting of Microbiome Studies' (STORMS), is composed of a 17-item checklist organized into six sections that correspond to the typical sections of a scientific publication, presented as an editable table for inclusion in supplementary materials. The STORMS checklist provides guidance for concise and complete reporting of microbiome studies that will facilitate manuscript preparation, peer review, and reader comprehension of publications and comparative analysis of published results., (© 2021. Background: Primary healthcare (PHC) is essential for equitable access and cost-effective healthcare. This makes PHC a key factor in the global strategy for universal health coverage (UHC). Implementing PHC requires an understanding of the health system under prevailing circumstances, but for most countries, no data are available. Objectives: This paper describes and analyses the health systems of Algeria, Kuwait, Morocco, Saudi Arabia, Jordan and Iraq to PHC. Methods: Data were collected during a workshop at the Wonca East Mediterranean Regional Conference in 2018. Academic family physicians (FP) presented their country; using the Wonca framework of 11 PowerPoint slides, with queries of the country demographics, main health challenges, and the position of PHC in the health system. Results: The six countries had achieved a significant improvement in populations' health but currently face challenges of health financing, a small number of certified FPs, difficulties in accessing services and bureaucratic procedures. Primary concerns were the absence of a family practice model, brain drain and immigration of FPs. Countries differed in building a coherent policy. Conclusion: Priorities should be focused on: developing PHC model in Eastern Mediterranean region with advocacy for community-based PHC to policymakers; capacity building for strengthening PHC-oriented health systems with FP specialty training and restrict practising to fully trained FPs; engage communities to improve understanding of PHC; adopt quality and accreditation policies for better services; validation of the referral and follow-up process; and, develop public-private partnership mechanisms to enhance PHC for UHC.

Background: Current COVID-19 researches suggest that both general population and health-care providers (HCPs) are at risk of elevated psychological sufferings including suicidality. However, suicidality has not been addressed properly, although mental health problems are studied globally. Besides, the extreme fear of COVID-19 infection is being existed among the Bangladeshi HCPs, that is reported by a recent patients' suicide because of HCPs treatment negligence., Methods: A web-based cross-sectional study was administered through the social media platforms. A total 3,388 respondents took part in the survey (mean age 30.1 ± 6.4 years) among them 834 were frontline HCPs (30.7 ± 5.6 years). The measures included socio-demographics, PPE-related and patient-care related information and a question concerned with the COVID-19 suicidal behavior., Results: About 6.1% of the total participants had suicidal behavior, with no detectable differences within the groups (i.e., general population and HCPs). Regression analysis showed that being female, being divorced, and having no child were emerged as independent predictors for suicidality. There was no significant association between the PPE-related or patient-care related variables and suicidal behavior of the HCPs. Majority of the participants sometimes had fear of death although no significant relation of the factor was found with suicidality., Limitations: The study can be limited because of its nature (i.e., cross-sectional self-reporting online survey) and not considering non-COVID-19 related suicide risk factors etc., Conclusions: The present findings identified the substantial proportion of the HCPs and general population had COVID 19 related suicidal behavior. In this work, an advance approach is reported for the water treatment technology using nanohybrid composite Fe 2 O 3 -SnO 2 /BC prepared by incorporation of iron-tin binary oxide into the cellulosic framework of medicinally active Nigella sativa (Black cumin) seed powder. The co-precipitation method was followed to prepare the nanohybrid composite which was subjected to investigate its physiochemical properties using spectroscopic and microscopic techniques. Fourier-transform infrared spectroscopy analysis confirmed the formation of highly functionalized nanocomposite through the hydrogen and electrostatic interactions between the functional groups of seeds and Fe 2 O 3 -SnO 2 . X-ray and selected area electron diffraction pattern revealed the presence of cubic phase of γ-Fe 2 O 3 and tetragonal phase of SnO 2 in the composite. The scanning electron microscopic images suggested the porous and relatively smooth surface of the composite, and transmittance electron microscopic images showed the trapping of nano-cubes of Fe 2 O 3 -SnO 2 , having particles size in the range 95-185 nm, into the organic framework of Black cumin seeds, whose zero point charge was found at pH 7.2. The composite was investigated for adsorption of Methylene blue dye from water for which the results revealed that 2.0 gL -1 amount of Fe 2 O 3 -SnO 2 /BC was sufficient to remove more than 95% dye, within 15 min, at 6-9 pH, from its 10 mgL -1 concentration. The thermodynamic studies established spontaneity, feasibility, and endothermic nature of the adsorption process. The adsorption data was satisfactorily described by the Freundlich isotherm which indicated inhomogeneous surface of the composite. Application of Temkin isotherm revealed the same extent of bonding probability and heat of adsorption at 27, 35, and 45 °C. The free energy change calculated from Dubinin-Radushkevich isotherm suggested weak interaction between Methylene blue and Fe 2 O 3 -SnO 2 /BC. The process satisfactorily followed the pseudo-second order kinetics that was controlled by the film diffusion step which indicated interaction of Methylene blue with functional sites of the Fe 2 O 3 -SnO 2 /BC. The Fourier-transform infrared spectroscopy analysis gave the confirmatory evidence for interaction of Methylene blue to Fe 2 O 3 -SnO 2 /BC. The maximum Langmuir adsorption capacity of the Fe 2 O 3 -SnO 2 /BC was found to be 58.82 mgg -1 at 27 °C which is higher than the previously reported adsorbents, MnFe 2 O 4 /BC [J. Clean. Prod. 2018. 200, 996-1008], and Fe 2 O 3 -ZrO 2 /BC [J. Clean. Prod. 2019. 223, 849-868]. Different desert truffles, collected from Algerian Saharan soils, were identified and their capacity to produce bioactive substances with antimicrobial activity was analyzed. Based on morphological characterization using Melzer's reagent staining, the collected strains were identified as Terfezia arenaria . The bioactive substances from T. arenaria were extracted using the following techniques: maceration with methanol and Soxhlet with dichloromethane. The former led to a yield much higher than that of the latter (i.e., 15% and 0.48%, respectively). Both extracts presented antifungal activities against all the tested strains (i.e., A. niger , Penicillium sp., and C. albicans ). However, the dichloromethane extracts showed much higher antibacterial activities against all the tested bacteria (i.e., S. aureus, E. faecalis, E. coli, and P. aeruginosa ) than the methanol extracts. Extreme ecosystems can be a source of untapped microorganisms to produce novel bioactive compounds of industrial interest. Consequently, in this work, 32 actinomycetes were isolated from 6 soil samples collected from Algerian Sahara in searching for untapped producers of novel antimicrobial compounds. All the isolates were further subjected to antimicrobial screening against pathogenic bacteria, yeast and fungi. The obtained results indicated that three of the isolates (named C, MS1 and 10) showed antimicrobial activities against most of the tested pathogenic microorganisms. Therefore, these three promising isolates, previously identified as Streptomyces by morphological, biochemical and physiological methods, were selected for their subsequent identification by the whole cell matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) analysis. Thus, the isolates C, MS1 and 10 were identified as Streptomyces violaceoruber B263 UFL, Streptomyces albus B262 UFL and Streptomyces badius B192 UFL, respectively. These results pointed out actinomycetes from Sahara soils as potential sources of novel antimicrobial compounds. Cytotoxic T-lymphocyte activation and extension of the cell life span is necessary in order to enable immunotherapy to perform effectively against cancer. In the present study, mucin 1 (MUC1)-stimulated human mononuclear cells (M1SHMCs) were costimulated with bead-attached monoclonal antibodies specific for cluster of differentiation (CD)3 and CD28 receptors. The study was undertaken to determine whether costimulation was capable of enhancing the killing of cancer cells in vitro and of protecting non-obese diabetic severe combined immunodeficient mice from tumor development. Lysis of MCF-7 tumor cells by M1SHMCs was reduced following costimulation with anti-CD3 and anti-CD28. Furthermore, costimulation with anti-CD3 and anti-CD28 eliminated the protective effects of M1SHMCs on MCF-7 breast cancer cell growth in the non-obese diabetic severe combined immunodeficient mice. The present study suggested that costimulation with anti-CD3 and anti-CD28 is not advisable following antigen activation of lymphocytes under the conditions used here. Using a lower anti-CD3/CD28 bead to T-cell ratio may prevent immune suppression, however, further studies are required to support this hypothesis.

mRNA instead of DNA provides a new and attractive approach for gene therapy and genetic vaccination. Current technologies for mRNA delivery are based on cationic lipids with DOTAP being the most efficient one. We previously reported on the synthesis of an inorganic-organic hybrid carrier by embedding inorganic nano-particles of carbonate apatite onto liposomal carrier DOTAP and demonstrated its high transfection potency of luciferase mRNA both in mitotic and non-mitotic cells. Here we show that in addition to the carrier design for effective endocytosis and release of mRNA to the cytoplasm, enhancement of mRNA translation efficiency is a prerequisite for maximum protein expression. We used the modified cap analog (ARCA) during in vitro transcription of luciferase DNA for proper cap orientation and demonstrated that transfection with ARCA-mRNA resulted in higher protein expression than the mRNA with usual cap structure for both DOTAP and DOTAP-apatite complex. Secondly, exogenous poly(A) was co-delivered with mRNA by the DOTAP-apatite, resulting in very significant expression compared to mRNA delivery only. Finally, when combined both of the effects of smart carrier and the modifications at mRNA translational level, a notable enhancement (100 times) was achieved as compared to the existing DOTAP-based liposome technology. Our findings, therefore, unveiled a novel approach that an effective delivery system can be developed by the improvement of the gene expression level in combination with the enhancement of the carrier potency.

Chronic granulomatosis disease (GCD) is a hereditary abnormality of phagocytic cells, frequently revealed by invasive pulmonary Aspergillosis. The authors report the case of 8 year old girl investigates for hypotrophia, reccurent lung disease and hemoptysia. She was treated for pulmonary tuberculosis because of biological inflammatory syndrome, right alveolar opacity and positive tuberculin skin test. The course was marked by the developement of a thoracic wall mass. Anti- aspergillus serology was positive; culture of the thoracic mass puncture and bronchoalveolar lavage isolated fumigatus Aspergillus. GCD was suggested and confirmed by the NBT test.
