Abstract

Heparin is known to have anti-inflammatory effects, yet the mechanisms are not completely understood. In this study, we tested the hypothesis that heparin has a direct effect on activated polymorphonuclear leukocytes (PMNLs), changing their activation state, and can explain its anti-inflammatory effect. To test our hypothesis, we designed both in vitro and ex vivo studies to elucidate the mechanism by which heparin modulates PMNL functions and therefore the inflammatory response. We specifically tested the hypothesis that priming of PMNLs renders them more susceptible to heparin. Amplified levels of CD11b and increased rate of superoxide release manifested PMNL priming. Increase in cell priming resulted in a dose-dependent increase in heparin binding to PMNLs followed by augmented apoptosis. Blocking antibodies to CD11b inhibited heparin binding and abolished the apoptotic response. Moreover, heparin caused a significant dose-dependent decrease in the rate of superoxide release from PMNLs, which was blunted by blocking antibodies to CD11b. Altogether, this study shows that the interaction of heparin with the PMNL CD11b results in cell apoptosis and explains heparin's anti-inflammatory effects.

9. Genome dilution by cell growth drives starvation-like proteome remodeling in mammalian and yeast cells

Abstract

Cell size is tightly controlled in healthy tissues and single-celled organisms, but it remains unclear how cell size influences physiology. Increasing cell size was recently shown to remodel the proteomes of cultured human cells, demonstrating that large and small cells of the same type can be compositionally different. In the present study, we utilize the natural heterogeneity of hepatocyte ploidy and yeast genetics to establish that the ploidy-to-cell size ratio is a highly conserved determinant of proteome composition. In both mammalian and yeast cells, genome dilution by cell growth elicits a starvation-like phenotype, suggesting that growth in large cells is restricted by genome concentration in a manner that mimics a limiting nutrient. Moreover, genome dilution explains some proteomic changes ascribed to yeast aging. Overall, our data indicate that genome concentration drives changes in cell composition independently of external environmental cues.

26. Microsaccades Tracking by Secondary Speckle Pattern Analysis.

Abstract

Here we propose a not pupil-dependent microsaccades tracking technique and a novel detection method. We present a proof of concept for detecting microsaccades using a non-contact laser-based photonic system recording and processing the temporal changes of speckle patterns scattered from an eye sclera. The data, simultaneously recorded by the speckle-based tracker (SBT) and the video-based eye tracker (Eyelink), was analyzed by the frequently used detection method of Engbert and Kliegl (E&K) and by advanced machine learning detection (MLD) techniques. We detected 93% of microsaccades in the SBT data out of microsaccades detected in the Eyelink data with the E&K method. By utilizing MLD, a precision of 86% was achieved. The findings of our study demonstrate a potential improvement in measuring tiny eye movements, such as microsaccades, using speckle-based eye tracking and, thus, an alternative to video-based eye tracking for detecting microsaccades., (© 2024 The Author(s). Abstract

Increasing cell size was recently shown to remodel the proteomes of cultured human cells, demonstrating that large and small cells of the same type can be biochemically different. Here, we corroborate these results in mouse hepatocytes and extend our analysis using yeast. We find that size-dependent proteome changes are highly conserved and mostly independent of metabolic state. As eukaryotic cells grow larger, the dilution of the genome elicits a starvation-like proteome phenotype, suggesting that growth in large cells is limited by the genome in a manner analogous to a limiting nutrient. We also demonstrate that the proteomes of replicatively-aged yeast are primarily determined by their large size. Overall, our data suggest that genome concentration is a universal determinant of proteome content in growing cells.
