PurposeThe primary cause of pulp and periapical diseases is the invasion of bacteria into the root canal, which results from the continuous destruction of dental hard tissues. Effective management of infections during root canal therapy necessitates effectively irrigation. This study aims to investigate the effects of two antimicrobial peptides (AMPs), buCaTHL4B and Im-4, on root canal biofilms in vitro.MethodsTwo-species biofilms (Enterococcus faecalis and Fusobacterium nucleatum) were selected and anaerobically cultivated. The following treatments were applied: 10 μg/mL buCaTHL4B, 10 μg/mL Im-4, 5 μg/mL buCaTHL4B, 5 μg/mL Im-4, 1 μg/mL buCaTHL4B, 1 μg/mL Im-4, 1% NaOCl, and sterile water. Each group was treated for 3 min. Subsequently, the two strains were co-cultured with 10 μg/mL buCaTHL4B, 10 μg/mL Im-4, 1% NaOCl, and sterile water for 24, 48, and 72 h. The biofilms were examined using confocal laser scanning microscopy (CLSM) with fluorescent staining, and the percentages of dead bacteria were calculated. Quantitative real-time PCR (qRT-PCR) was employed to assess the variations in bacterial proportions during biofilm formation.ResultsCompared to 1% NaOCl, 10 μg/mL buCaTHL4B or Im-4 exhibited significantly greater bactericidal effects on the two-species biofilms (p < 0.05), leading to their selection for subsequent experiments. Over a 48-hour period, 10 μg/mL Im-4 demonstrated a stronger antibiofilm effect than buCaTHL4B (p < 0.05). Following a 24-hour biofilm formation period, the proportion of F. nucleatum decreased while the proportion of E. faecalis increased in the sterile water group. In the buCaTHL4B and 1% NaOCl groups, the proportion of F. nucleatum was lower than that of E. faecalis (p < 0.05), whereas in the Im-4 group, the proportion of F. nucleatum was higher than that of E. faecalis (p < 0.05). The proportions of bacteria in the two AMPs groups gradually stabilized after 24 h of treatment.ConclusionbuCaTHL4B and Im-4 exhibited remarkable antibacterial and anti-biofilm capabilities against pathogenic root canal biofilms in vitro, indicating their potential as promising additives to optimize the effectiveness of root canal treatment as alternative irrigants.

Abstract Purpose To characterize the bacterial community in the primarily infected root canals. Methods A total of 13 samples were collected from the primarily infected root canals. 16 S rDNA sequencing was performed to define bacterial community. Taxonomic annotation, bacterial hierarchical structures, community richness and diversity, and inter-subject variability of the bacterial community in the root canal samples were analyzed. Gender, age, and duration of the toothache-specific bacterial community associated with the patient groups were analyzed. Results A total of 359 Species were annotated and identified in the whole study cohort. The Alpha diversity analysis showed that the species diversity and detection rate of the 13 samples were high, which reflected the authenticity of sequencing results. The Beta diversity analysis was used to compare the degree of difference between different root canal samples. The 13 samples were divided into two groups according to the results, group A was samples I1-I12, and group B was samples I13. The bacterial species of group A samples were analyzed with the clinical characteristics of patients, and it was found that gender, and duration specific differences in bacterial species, and there was no significant difference in species types among different ages of patients. Conclusion There were a wide diversity and inter-subject variability in the bacterial community in the primary infected root canals. While Porphyromonas gingivalis was the most abundant species, Fusobacterium nucleatum was the most variable species in the bacterial community of the root canal. The bacterial community at different taxonomic levels varied from sample to sample, despite consistent disease diagnoses. There was gender, duration-specific differences in the bacterial species in the primary infected root canals.

Summary: Accumulating evidence has shown that fibromodulin (FMOD) plays a pivotal role in tumorigenesis and metastasis. However, the biological function of FMOD in oral squamous cell carcinoma (OSCC) remains largely unclear to date. In this study, we confirmed that FMOD was overexpressed and showed a significant association with malignant progression and lymph node metastasis in OSCC. Depletion of FMOD inhibited OSCC proliferation and metastasis in vitro and in vivo. RNA sequencing, western blotting, and rescue assays verified that FMOD exerted oncogenic roles in OSCC via activation of EGFR signaling. In addition, FMOD was proved to be a putative target gene of miR-338-3p. Taken together, FMOD overexpression due to the reduced level of miR-338-3p promotes OSCC by activating EGFR signaling. Our findings provide direct evidence that targeting FMOD could be a promising therapeutic strategy for OSCC patients.

The goal of this paper is to explore a method for virus inactivation based on ultrasonic treatment, and on this basis, to explore the synergistic effect of methylene blue as a sonosensitizer in virus inactivation. The titer of human parainfluenza virus type 3 (HPIV3), Autographacaliforinica nuclear polyhedrosis virus carried a green fluorescent reporter gene (AcNPV-GFP) and Enterovirus group D 68 (EV-D68) were determined by plaque assays or TCID50 methods after treatment by MB combined with illumination (MB photochemical, MBP) or ultrasonic excitation. Different ultrasonic power and time, MB concentration gradient were set to determine the best anti-viral combination. For the enveloped virus HPIV3 and AcNPV-GFP, pure ultrasonic or MBP treatment could reduce viral titer more than 104，and ultrasonic combined with MB could completely inactivate HPIV3 and AcNPV-GFP in 5min while traditional MBP methods could only reduce viral titer about 10X. For the non-enveloped virus EV-D68, pure ultrasonic or MBP treatment only reduced the viral titer about 102, but ultrasonic combined with MB treatment could reduce the viral titer about 103 in 5min and completely inactive EV-D68 in 10min (reduced 105). Compared with MBP method and pure ultrasonic inactivation, ultrasonic combined with MB has better inactivation effect on either enveloped or non-enveloped viruses, and the appropriate combination of parameters is expected to be a new blood transfusion transmitted virus inactivation method.

Gas leakage is the largest risk to the safe operation of natural gas transmission stations and valve chambers of long-distance gas pipelines, and it is of high probability for the leakage of instrument pipes. In order to effectively control the leakage risk of the instrument pipes, it is necessary to develop a gauge valve that can automatically shut off when the instrument pipe leaks. Hence, an automatic shut-off valve was developed for the instrument pipes with the closing pressure difference set by the spring structure, so that the valve could be shut down automatically by the pressure difference generated during leakage. The valve is simple in structure and easy to install, consisting of a metal seat, a throttle element, a regulation spring and a needle valve. It can be cut off immediately once a large amount of leakage occurs in the instrument pipe so that the gas supply can be disconnected for the purpose of effectively eliminating the risks caused by leakage. As shown in a series of field application test, the valve is capable of realizing good shutoff effect and stable operation with various performance satisfying the operating requirements, so it can be further promoted and applied.

ABSTRACT: The threat of antimicrobial resistance (AMR) is on the rise globally, especially with the development of animal husbandry and the increased demand for antibiotics. Livestock and poultry farms, as key sites for prevalence of antibiotic-resistant bacteria (ARB), can spread antimicrobial resistance genes (ARGs) through microbial aerosols and affect public health. In this study, total suspended particulate matter (TSP) and airborne culturable microorganisms were collected from duck houses in Tai'an, Shandong Province, and the bacterial communities and airborne ARGs were analyzed using metagenomics and PCR methods. The results showed that the bacterial communities in the air of duck houses were mainly Actinobacteria, Firmicutes, Proteobactria, Chlamydia, and Bcateroidetes at the phylum level. At the genus level, the air was dominated by Corynebacterium, Jeotgalicoccus, Staphylococcus, Brevibacterium, and Megacoccus, and contained some pathogenic bacteria such as Staphylococcus aureus, Corynebacterium diphtheriae, Klebsiella oxytoca, Acinetobacter baumannii, and Pseudomonas aeruginosa, which were also potential hosts for ARGs. The airborne ARGs were mainly macrolides (10.97%), penicillins (10.73%), cephalosporins (8.91%), streptozotocin (8.91%), and aminoglycosides (8.02%). PCR detected 27 ARGs in airborne culturable microorganisms, and comparative analysis between PCR and the metagenomic data revealed that a total of 9 ARGs were found to the same, including macrolides ErmA, ErmF, tetracyclines tetG, tetX, methicarbamazepines dfrA12, dfrA15, aminoglycosides APH3-VI, ANT2-Ⅰ, and sulfonamides sul2. Moreover, inhalation exposure modeling showed that the workers in duck houses inhaled higher concentrations of ARB, human pathogenic bacteria (HPB) and human pathogenic antibiotic-resistant bacteria (HPARB) than hospital workers. These results provide new insights into airborne microorganisms and ARGs in animal farms and lay the foundation for further study.

To effectively control the stress state and spatial alignment of arch ribs in the cable hoisting construction of a long-span, concrete-filled, steel tube arch bridge and ensure the safety of the structure, it is necessary to calculate and determine the appropriate cable force. Based on the actual project of a double-span, concrete-filled, steel tubular arch bridge, the construction stage of the left span of the bridge from the beginning of construction to the closure is taken as an example. The linear control method of “quiet do not move” is adopted. Based on the principle that the vertical displacement of the front end of the installed segment caused by the self-weight of the new hoisting segment is equal to the vertical displacement of the front end of the previous segment caused by the tension of the new hoisting segment, the tension cable force is calculated by forward iteration. Finally, based on the theory of the stress-free state method, the ideal linear design of the structure was achieved. The results show that after the closure of the bridge, the error range of the cable tension force is −13.33–15.40% on the left bank and −8.37–11.00% on the right bank. The elevation error of the arch rib is −0.003–0.043 m on the left bank and −0.007–0.032 m on the right bank. The overall stress error of the bridge arch is ±7.0 MPa. The error between the theoretical value and the actual value is within the scope of the specification requirements, which meets the specification requirements. After the closure, the arch shape of the bridge meets the smooth requirements.

Considering the characteristics of lower power resources in multi-point wireless body area networks (WBANs), the wireless energy and information transmission in full duplex (FD) mode is considered. We propose a transmission protocol based on time division multiple access, which includes two phases. In the inactive phase, the signal access point (AP) broadcasts Radio Frequency (RF) energy to the sensors. When the energy collected by all sensors reaches their respective energy thresholds, the active phase will start. In the active phase, the sensors send back the collected physiological information parameters to the AP, while sensors still collecting the energy transmitted from the AP to achieve the purpose of energy accumulating. Our goal is to obtain the optimal time allocation ratio through the joint objective linear programming method to obtain the maximum average sum-throughput. The simulation results compared with the AP with FD and the sensors with half duplex (HD) show that our proposed optimization method can improve system throughput obviously.
