ABSTRACT: Ruminants may be affected by a wide variety of central nervous system (CNS) diseases. Cerebrospinal fluid (CSF) analysis forms the basis for ante mortem diagnostic evaluation of ruminants with clinical signs involving the CNS. Despite its importance as a tool to aid diagnosis, data regarding CSF examinations in spontaneous cases of CNS diseases in ruminants from Brazil are limited, and most reports involve experimental studies. Therefore, this study aimed to report the results of CSF analysis in 58 ruminants showing signs of neurological disorders. CSF samples for analysis were obtained from 32 cattle, 20 sheep, and 6 goats by cerebello-medullary cistern (n=54) or lumbosacral space (n=4) puncture. These ruminants showed neurological signs related to viral (n=13), mycotic (n=3), or bacterial (n=15) infections, and toxic (n=21), traumatic (n=4), or congenital disorders (n=2). CSF analysis from ruminants with viral infections presented lymphocytic pleocytosis, even though CSF showed no changes in several cases of rabies. Neutrophilic pleocytosis, cloudiness, presence of fibrin clots, and abnormal coloration were evident in the CSF of most cases of CNS bacterial infection, such as meningoencephalitis, meningitis, abscesses, myelitis, and a case of conidiobolomycosis. On the other hand, CSF was unchanged in most cases of toxic disorders, as botulism and hepatic encephalopathy. Elevated CSF density was observed in 60% of ruminants diagnosed with polioencephalomalacia. Our findings show that evaluation of CSF is a valuable diagnostic tool when used in association with epidemiological, clinical and pathological findings for diagnosis of CNS diseases in ruminants.

Abstract Recent work has highlighted the tumor microenvironment as a central player in cancer. In particular, interactions between tumor and immune cells may help drive the development of brain tumors such as glioblastoma multiforme (GBM). Despite significant research into the molecular classification of glioblastoma, few studies have characterized in a comprehensive manner the immune infiltrate in situ and within different GBM subtypes. In this study, we use an unbiased, automated immunohistochemistry-based approach to determine the immune phenotype of the four GBM subtypes (classical, mesenchymal, neural and proneural) in a cohort of 98 patients. Tissue Micro Arrays (TMA) were stained for CD20 (B lymphocytes), CD5, CD3, CD4, CD8 (T lymphocytes), CD68 (microglia), and CD163 (bone marrow derived macrophages) antibodies. Using automated image analysis, the percentage of each immune population was calculated with respect to the total tumor cells. Mesenchymal GBMs displayed the highest percentage of microglia, macrophage, and lymphocyte infiltration. CD68+ and CD163+ cells were the most abundant cell populations in all four GBM subtypes, and a higher percentage of CD163+ cells was associated with a worse prognosis. We also compared our results to the relative composition of immune cell type infiltration (using RNA-seq data) across TCGA GBM tumors and validated our results obtained with immunohistochemistry with an external cohort and a different method. The results of this study offer a comprehensive analysis of the distribution and the infiltration of the immune components across the four commonly described GBM subgroups, setting the basis for a more detailed patient classification and new insights that may be used to better apply or design immunotherapies for GBM.

During the seventeenth century, in both France and the Grand Duchy of Lithuania, the rulers formed their own parties to help them rule the state. The main purpose of the article is to present how rulers of the Grand Duchy of Lithuania and of France chose their favourites, what was their background, why were they chosen and how they could retain or lose the king’s favour. Common elements appeared in the seventeenth-century system of appointing favourites in both France and the Polish-Lithuanian Commonwealth. In Poland and Lithuania, however, kings valued the maintaining of a balance of power, in order to prevent the dominant influence of any person over the ruler, while in France, certain favourites – Richelieu, Mazarin – took on a dominant role. When Louis XIV came to power, he introduced personal and independent government yet also had several favourites whose influence he tried to balance. In Lithuania, the situation began to change over time, when, in the second half of the seventeenth century, the hegemony of two families was established, that of the Pacs and the Sapiehas, who could follow their own political agenda, independently of the court of Warsaw, and which the king was obliged to take into account.

Uznaje się, że jedną z przyczyn polowań na czarownice były przemiany społeczno-ekonomiczne, do których doszło we wczesnonowożytnej Europie. Jak słusznie zauważył Janusz Tazbir, duży związek z oskarżeniami o czary w Rzeczypospolitej, podobnie jak w całej Europie, miały konflikty społeczne na wsi, które nabrały szczególnie ostrego charakteru w XVII w. Niekiedy wynikały one z zarazy, która pojawiła się na danym terenie i doprowadziła do śmierci zwierząt hodowlanych (w procesach czarownic występuje głównie bydło) lub ludzi. Mieszkańcy XVII-wiecznej Rzeczypospolitej, jak i całej Europy, nie potrafili wytłumaczyć, dlaczego doszło do masowego zgonu zwierzyny, w związku z tym najczęściej posądzano o to samotne kobiety, często te, które parały się ziołolecznictwem, lub te żyjące na uboczu osady lub wykluczone ze społeczności lokalnej, czymś wyróżniające się (cechy charakteru, wygląd), kłótliwe. Można stwierdzić z dużą dozą prawdopodobieństwa, że zmiany i kryzysy społeczno-ekonomiczne dominowały jako przyczyny wszczynania pozwów przeciwko czarownicom na Litwie. Należy także zwrócić uwagę na to, że w wielu procesach najprawdopodobniej powodem odprawiania czarów przez mieszkańców wsi i miast była zawiść ekonomiczna nie tylko wobec sąsiadów, ale również właścicieli, dzierżawców i administratorów określonych dóbr.

Während des Interregnums, also zwischen dem Tod des alten Königs und dem Zeitpunkt der Neuwahl, wurde der zukünftige König der Rzeczpospolita in einem hochspannenden politischen Prozess bestimmt. Die Thronprätendenten haben viele Möglichkeiten versucht, um die Unterstützung des Adels zu gewinnen. Zweifellos war eines der wichtigsten Themen bei den Verhandlungen mit den Kandidaten für den Thron deren Verhältnis zu anderen Religionen und die erklärte Toleranz (oder Intoleranz) gegenüber den Dissidenten. Ihre Einstellung zur Religion sowie die diesbezüglichen Versprechen waren manchmal ein entscheidendes Element in der politischen Auseinandersetzung.

Este artigo, inserido no âmbito da sintaxe comparativa, estabelece uma comparação entre variedades do português, a saber: o português europeu (PE), o português brasileiro (PB), o português de Moçambique (PM), de Angola (PA), de Guiné-Bissau (PGB), de São Tomé e Príncipe (PST) e de Cabo Verde (PCV), centrando a atenção no licenciamento de duplos sujeitos (DSs). Para tanto, o corpus da pesquisa constitui-se de contextos declarativos finitos extraídos de entrevistas informais. Estudos têm revelado que em PB predominam DSs com o traço de 3ª pessoa, ao contrário de outras línguas, como é o caso do crioulo caboverdiano e do Santome cuja predominância é da 1ª pessoa do singular. Visando estabelecer o estudo interlinguístico proposto, abordamos os traços de pessoa e de definitude e propomos uma reinterpretação de uma geometria de traços que culmina em uma hierarquia interna aos pronomes relevante para sua realização morfológica e para a sintaxe. Com base nesse modelo, propomos a inclusão do traço pessoa (a saber, [π]) considerado empiricamente necessário, o que implica considerarmos de que esse traço é inerente ao pronome e não ao verbo. Retomando também a proposta de composição de traços e observando os traços relevantes para a valoração de pessoa, levantamos como hipótese que entre o PB e as demais variedades do português, diferentes subespecificações de traços de pessoa estão envolvidas nas estruturas de DSs entendidas como estruturas tópicas que possuem um pronome resumptivo na posição do sujeito movido.

No texto são discutidos os planos e as ações elaboradas para a salvaguarda da diversidade florística e faunística existente nas terras que foram alagadas para a formação do lago da Hidrelétrica Itaipu, entre o Brasil e o Paraguai. Para isso são analisadas as diretrizes elaboradas pela empresa responsável pela obra, que orientaram a produção dos inventários da fauna e da flora local, num contexto desenvolvimentista. A perda da biodiversidade, a transformação da dinâmica do rio, a destruição e a fragmentação de habitats e o desaparecimento da paisagem cênica das Sete Quedas são marcas deixadas pelo empreendimento.

Introduction. Macrophages are phenotypically heterogeneous cells that play a vital role in hepatic fibrogenesis. We aimed to compare the macrophage profiles between normal livers and those with various chronic liver diseases in the precirrhotic fibrosis stage. Methods. Immunohistochemistry was performed for three macrophage markers (CD163, CD68, and IBA1) on 48 liver biopsies. Digital image analysis and automated cell count were used to calculate the densities of immunostained cells in two selected regions of interest: the periportal region and the perivenous region. Results. The absolute and relative densities of the macrophage phenotypes in relationship with zones and etiologies showed four distinct patterns by hierarchical cluster analysis: (1) no significant increase in the macrophage densities in either periportal or perivenous regions - nonalcoholic steatohepatitis; (2) significant increase in the selected macrophage densities in both periportal and perivenous regions - Hepatitis C; (3) significant increase in the macrophage densities only in periportal region - alcoholic liver disease, primary sclerosing cholangitis, and primary biliary cholangitis; and (4) significant increase in the densities of all types of macrophages in both periportal and perivenous regions - autoimmune hepatitis. Conclusions. There are distinct macrophage phenotypic and zonal geographic signatures correlating to etiologies of chronic liver disease in the precirrhotic stage. [ABSTRACT FROM AUTHOR]

An analysis was made of the spatial and temporal distribution of the abundance and composition of fish larvae assemblies, aiming to ascertain correlations with biotic and abiotic factors in marginal lagoons of the Cuiabá River floodplain in Brazil's Pantanal wetlands. Samples were collected bimonthly in the pelagic region of the marginal lagoons (fourteen lagoons) close to the littoral zone covered by aquatic macrophytes from December 2006 to April 2007. The captured individuals consisted of 2,739 larvae, represented predominantly by Triportheus spp. (Characidae), Parodontidae, Anostomidae, Characidae and Serrasalmidae. Larval density varied among the sample lagoons in December, but the biotic (zooplankton density) and abiotic (water temperature, electrical conductivity, dissolved oxygen, pH, water transparency and lagoon depth) factors did not significantly affect larval composition and abundance. Larvae were captured throughout the sample period, with the highest densities occurring in December and early January. Temporally, lagoon depth, water transparency and fluviometric level showed a negative correlation with larval density. Both the zooplankton and the larval density vary positively, responding likewise to the environmental conditions imposed by flooding. The highest larval density was also found to coincide with the first rains (early December), when the rainfall has not yet reached its peak.Foi realizada uma análise da distribuição espaço-temporal da abundância e composição da assembleia de larvas de peixes, com o objetivo de verificar as correlações com os fatores bióticos e abióticos em lagoas marginais da planície de inundação do rio Cuiabá, Pantanal, Brasil. As amostras foram coletadas quinzenalmente na região pelágica das lagoas, perto da zona litorânea coberta por macrófitas aquáticas, de dezembro de 2006 à março de 2007, na planície de inundação. Foram capturadas 2.739 larvas, representadas predominantemente por Triportheus spp. (Characidae), Parodontidae, Anostomidae, Characidae e Serrasalmidae. A densidade de larvas variou entre as lagoas amostradas em dezembro, mas as variáveis bióticas (densidade de zooplâncton) e abióticas (temperatura da água, condutividade elétrica, oxigênio dissolvido, pH, transparência e profundidade da lagoa) não afetaram significativamente a composição e a abundância das larvas. As larvas foram capturadas ao longo de todo o período de amostragem, com maiores densidades no mês de dezembro e início de janeiro. Temporalmente, a profundidade da lagoa, a transparência e o nível fluviométrico mostraram uma correlação negativa com a densidade de larvas. Tanto a abundância de zooplâncton, quanto à abundância de larvas variaram positivamente, respondendo da mesma forma às condições ambientais impostas pela cheia. A maior densidade de larvas coincidiu com as primeiras chuvas (início de dezembro), quando a pluviosidade ainda não tinha atingido seu pico.

This work aimed to evaluate female lineage broilers for halothane sensitivity and for their susceptibility to the subsequent development of PSE meat. The halothane test was carried out in an anesthetic chamber with 3.0% halothane. The unconscious birds were examined for leg muscle rigidity. If one or both legs became extended and rigid, the birds were classified as halothane sensitive (HAL+), while unresponsive birds were classified as halothane negative (HAL-). The results showed that of 298 birds aged 42 days old, 95.6% were HAL- and 4.4% were HAL+. A sample of pectoralis major muscle was collected from HAL- (n=105) and HAL+ (n=13) birds. The pH and breast fillet color were determined at 4ºC, 24 hours post-mortem. Interestingly, only 2.5% of HAL+ birds displayed PSE meat characteristics compared to 12.7% of HAL- individuals. The halothane test demonstrated that female lineage broilers displayed very little sensitivity towards halothane, indicating that the development of PSE meat is related to other environmental factors.O objetivo deste trabalho foi avaliar a sensibilidade de aves de uma linhagem fêmea ao halotano e sua relação com o desenvolvimento de carnes PSE. O teste do halotano foi conduzido com o auxílio de uma câmara anestésica com 3,0% de halotano volatilizado. As aves inconscientes foram examinadas pelo enrijecimento dos seus membros inferiores. Quando ambos ou um dos membros permaneceram rígidos, os frangos foram classificados como sensíveis ao halotano (HAL+) e os frangos sem enrijecimento dos membros foram classificados como não-sensíveis (HAL-). Os resultados mostraram que de 298 frangos com 42 dias de idade, 95,6% foram HAL-, e apenas 4,4% HAL+. O peito foi coletado das aves HAL- (n=105) e HAL+ (n=13) em que o pH e Cor (L*,a*,b*) foram determinados a 4ºC, 24h postmortem. Interessantemente, apenas 2,5% das aves HAL+ demonstraram carnes PSE, enquanto que as aves HAL- apresentaram 12,7% de carnes PSE, em relação ao total de aves abatidas. O teste do halotano demonstrou que frangos da linhagem fêmea mostraram pouquíssima sensibilidade ao halotano, indicando que a ocorrência de carnes PSE está mais associada a fatores ambientais.

Samples of Pectoralis major m. were collected, and an RT-PCR analysis of the a-Ryanodine receptor (a RYR) from chicken mRNA hotspot region spanning aminoacid residues 386 to 540, numbered according to the turkey sequence, revealed two classes of transcripts. The sequences of the first class were similar to turkey and human with 97% and 74% of identity, respectively, and included all transcripts with substitutions in the nucleotide sequence. The second class was characterized by the deletion of nucleotides, leading to a premature stop codon and coding for a truncated and nonfunctional protein. These results are to date the first report related to the sequencing of the chicken αRYR hotspot region 1, which will possibility serve as a guide for further studies regarding a solution in the poultry production chain related to the problem of pale, soft and exudative (PSE) meat.Amostras do músculo Pectoralis major foram coletadas e uma RT-PCR foi conduzida para avaliar a sequência do mRNA do αRYR, região compreendida entre os resíduos de aminoácido 386-540, numerado de acordo com a sequência de perus. Os resultados revelaram duas classes de transcritos. O primeiro teve 97% e 74% de identidade com as sequências de αRyR e RyR1 de perus e humanos, respectivamente, e incluiu todos os transcritos com substituições de nucleotídeos. A segunda classe de transcritos foi caracterizada pela deleção de bases que levaram a um stop códon prematuro e a uma proteína truncada não-funcional. Esses resultados são até o momento, o primeiro relato de sequenciamento do αRYR, região hotspot1 de frangos e podem servir como guia para estudos futuros na tentativa de se encontrar uma solução para os problemas na cadeia de produção de frangos relacionados com as carnes PSE (pálida, flácida e exsudativa).

The objective of this work was to investigate the use of a halothane to screen broiler chickens prone to develop PSE (Pale, Soft, Exudative) meat through a special gas chamber connected to a veterinarian anaesthetic apparatus developed in our laboratory. Anaesthesia was induced with approx. 3.0% halothane at a flow rate of 6.0 Lm-1 in pure oxygen for 5 min. Commercial male broilers (n=342) aged 46 days old were evaluated and classified as either halothane sensitive (HAL+) or insensitive (HAL-), depending on the leg rigidity response. Approximately 27.5% (n=94) of broilers were HAL+ and 72.5% (n=248) were HAL-. This is a simple and rapid technique to evaluate broiler sensitivity to halothane and identify broilers prone to develop PSE meat. The occurrence of PSE meat in HAL insensitive broiler chicken samples suggests that other factors are related to PSE occurrence.O objetivo deste trabalho foi investigar o uso do halotano para avaliar e identificar frangos propensos ao desenvolvimento de carnes PSE (Pálida, Flácida, Exsudativa) por meio de uma câmara desenvolvida em nossos laboratórios. Esta foi conectada a um equipamento de anestesia veterinária e a anestesia foi então induzida com aproximadamente 3,0% de halotano a um fluxo de 6,0 L/min em puro oxigênio. A sensibilidade de três frangos foi avaliada simultaneamente. Os frangos de linhagem comercial (n=352) com 46 dias de idade foram expostos à 3,0% de halotano por 5 min e classificados como halotano sensível (HAL +) e não-sensível (HAL -), baseado no enrijecimento dos seus membros inferiores. Aproximadamente 27,5% (n=94) frangos foram HAL+ e 72,5% (n=248) HAL-. Amostras do músculo Pectoralis major (n=75) foram coletadas após o abate dos animais para as medidas de pH e cor, determinadas nos filés mantidos a 4 C por 24 h postmortem. Frangos HAL+ apresentaram maior porcentagem de carnes PSE quando comparadas ao grupo HAL-. Este é um teste simples e rápido para a avaliação da sensibilidade das aves ao halotano e identificar frangos propensos ao desenvolvimento de carnes PSE embora amostras de HAL - também apresentaram carnes PSE sugerindo que outros fatores estão relacionados com a formação da anormalidade.
