1. Cultivation of novel Atribacterota from oil well provides new insight into their diversity, ecology, and evolution in anoxic, carbon-rich environments

Abstract: Abstract Background The Atribacterota are widely distributed in the subsurface biosphere. Recently, the first Atribacterota isolate was described and the number of Atribacterota genome sequences retrieved from environmental samples has increased significantly; however, their diversity, physiology, ecology, and evolution remain poorly understood. Results We report the isolation of the second member of Atribacterota, Thermatribacter velox gen. nov., sp. nov., within a new family Thermatribacteraceae fam. nov., and the short-term laboratory cultivation of a member of the JS1 lineage, Phoenicimicrobium oleiphilum HX-OS.bin.34TS, both from a terrestrial oil reservoir. Physiological and metatranscriptomics analyses showed that Thermatribacter velox B11T and Phoenicimicrobium oleiphilum HX-OS.bin.34TS ferment sugars and n-alkanes, respectively, producing H2, CO2, and acetate as common products. Comparative genomics showed that all members of the Atribacterota lack a complete Wood-Ljungdahl Pathway (WLP), but that the Reductive Glycine Pathway (RGP) is widespread, indicating that the RGP, rather than WLP, is a central hub in Atribacterota metabolism. Ancestral character state reconstructions and phylogenetic analyses showed that key genes encoding the RGP (fdhA, fhs, folD, glyA, gcvT, gcvPAB, pdhD) and other central functions were gained independently in the two classes, Atribacteria (OP9) and Phoenicimicrobiia (JS1), after which they were inherited vertically; these genes included fumarate-adding enzymes (faeA; Phoenicimicrobiia only), the CODH/ACS complex (acsABCDE), and diverse hydrogenases (NiFe group 3b, 4b and FeFe group A3, C). Finally, we present genome-resolved community metabolic models showing the central roles of Atribacteria (OP9) and Phoenicimicrobiia (JS1) in acetate- and hydrocarbon-rich environments. Conclusion Our findings expand the knowledge of the diversity, physiology, ecology, and evolution of the phylum Atribacterota. This study is a starting point for promoting more incisive studies of their syntrophic biology and may guide the rational design of strategies to cultivate them in the laboratory. Video Abstract

2. Study on Synthesis, Characterization and in Vitro Activity of Sweet Corncob Polysaccharide Nano-Silver

Abstract: Silver nanoparticles (AgNPs) are widely used in the field of biomedicine. However, the synthesis of AgNPs by traditional physical and chemical methods has problems such as pollution, product toxicity, and time-consuming. Therefore, it is extremely important to explore the preparation of AgNPs. Silver nanoparticles (AgNPs) of sweet corncob polysaccharide (SCP-AgNPs) were successfully synthesized by green method using sweet corncob polysaccharide (SCP) as raw material and the reaction conditions were optimized. The generated AgNPs were characterized by transmission electron microscopy (TEM), X-ray diffraction, X-ray photoelectron spectroscopy and Fourier transform infrared spectroscopy, and their antioxidant and anti-diabetic abilities in vitro were investigated. The results showed that the optimum reaction conditions of SCP-AgNPs were as follows: The concentration of silver nitrate in the solution was 0.75×10−3 mol/L, the pH was 10.0, and the reaction time was 80 min. The prepared SCP-AgNPs were spherical with an average particle size of 13.5 nm. The structure showed a face-centered cubic crystal structure, and Ag was determined to be Ag0 state. The synthesized SCP-AgNPs had strong free radical scavenging effect in vitro. At the same time, SCP-AgNPs had a good inhibitory effect on the activity of α-glucosidase and α-amylase. SCP-AgNPs has a significant inhibitory effect on key enzymes of diabetes and has potential anti-diabetic activity. It is a nano-drug suitable for nano-biomedical applications and will be widely used in food processing, medicine and other fields in the future.

3. Efficacy of acupuncture as an adjunctive treatment to patients with stable COPD: a multicenter, randomized, sham-controlled trial protocol

Abstract: Abstract Background Chronic obstructive pulmonary disease (COPD) is a common respiratory disease and the third leading cause of death worldwide. Previous evidence has shown that acupuncture may be an effective complementary alternative therapy for stable COPD. However, large-sample, rigorously designed long-term follow-up studies still need to be completed. Notably, the relationship between the frequency of acupuncture and clinical efficacy in studies on acupuncture for stable COPD still needs further validation. This study aims to evaluate the efficacy and safety of acupuncture for stable COPD and further investigate the dose–effect relationship of acupuncture. Methods/design This is a multicenter, randomized, controlled trial that uses central randomization to randomly allocate 550 participants in a 1:1:1:1:1 ratio to once a week acupuncture group, twice a week acupuncture group, three times a week acupuncture group, sham acupuncture group and waiting-list control group. The sham acupuncture group will receive placebo acupuncture treatments three times per week, and the waiting-list control group will not receive any form of acupuncture intervention. The study consists of a 2-week baseline, 12-week of treatment, and 52-week of follow-up. Patients with COPD between 40 to 80 years old who have received stable Western medication within the previous 3 months and have had at least 1 moderate or severe acute exacerbation within the past 1 year will be included in the study. Basic treatment will remain the same for all participants. The primary outcome is the proportion of responders at week 12. Secondary outcomes include the proportion of responders at week 64, change in the St. George's Respiratory Questionnaire (SGRQ) Scale, change in the Modified-Medical Research Council (mMRC) Scale, change in the COPD Assessment Test (CAT) Scale, change in the Lung Function Screening Indicators (LFSI), change in the 6-min walk distance (6-MWD), change in Short-Form 36 Health Survey (SF-36) Scale, the number of moderate and severe acute exacerbations and adverse event rate during the follow-up period. Discussion This study will provide robust evidence on whether acupuncture is safe and effective for treating stable COPD. Meanwhile, comparing the differences in efficacy between different acupuncture frequencies will further promote the optimization of acupuncture for stable COPD. Trial registration This study was registered in the Chinese Clinical Trial Registry (ChiCTR2200058757), on April 16, 2022.

4. Evaluating the fatigue life of notched components based on the stress gradient model with variable support effects

Abstract: As the notch location of engineered components suffers from size effect, thereby making it difficult to accurately predict the fatigue life of engineered components. This study briefly analyses the limitations of the traditional relative stress gradient model for analyzing the fatigue life of notched members. Subsequently, a relative stress gradient model with variable support effects is proposed. The fatigue data of various notched specimens with different materials were employed to verify the performance of the proposed model for size effect. The results show that the proposed model has satisfactory predictive performance. Compared with the conventional relative stress gradient model and critical distance method, the proposed model exhibited superior predictive capabilities.

5. Inhibitory Effect of Selenium Sweet Corncob Polysaccharide on Non-Enzymatic Glycosylation

Abstract: A selenized polysaccharide (Se-SCP) with selenium content of (7.19±0.067) mg/g was synthesized by imidazole-mediated method using sweet corncob polysaccharide (SCP) as raw material. In order to explore the inhibitory effect of Se-SCP on non-enzymatic glycosylation reaction, fructose and bovine serum albumin (BSA) were selected as substrates to establish albumin/fructose simulation system. Fructosamine→α-dicarbonyl compounds→advanced glycation end products (AGEs), the three characteristic products in the non-enzymatic glycosylation process, were determined and analyzed by transmission electron microscopy (TEM) and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The effects of protopolysaccharides and selenium polysaccharides on the non-enzymatic glycosylation process were compared and analyzed in terms of microstructure. The results showed that Se-SCP had different degrees of inhibition on the three stages of protein non-enzymatic glycosylation process, and the inhibition rates of characteristic products in non-enzymatic glycosylation process reached 89.5%, 74.9% and 71.7%, respectively. In terms of inhibiting protein cross-linking structure, compared with the SCP group, the texture of Se-SCP was relatively clear, the structural density was small, and the inhibition effect was obvious. The results of SDS-PAGE showed that Se-SCP reduced the formation of high molecular weight cross-linked protein induced by fructose, and the effect of Se-SCP was better, which was similar to that of positive drug. In summary, Se-SCP has good anti-nonenzymatic glycation activity and has great potential in inhibiting diabetes and its complications.

6. Historical development of karst evergreen broadleaved forests in East Asia has shaped the evolution of a hemiparasitic genus Brandisia (Orobanchaceae)

Abstract: Brandisia is a shrubby genus of about eight species distributed basically in East Asian evergreen broadleaved forests (EBLFs), with distribution centers in the karst regions of Yunnan, Guizhou, and Guangxi in southwestern China. Based on the hemiparasitic and more or less liana habits of this genus, we hypothesized that its evolution and distribution were shaped by the development of EBLFs there. To test our hypothesis, the most comprehensive phylogenies of Brandisia hitherto were constructed based on plastome and nuclear loci (nrDNA, PHYA and PHYB); then divergence time and ancestral areas were inferred using the combined nuclear loci dataset. Phylogenetic analyses reconfirmed that Brandisia is a member of Orobanchaceae, with unstable placements caused by nuclear-plastid incongruences. Within Brandisia, three major clades were well supported, corresponding to the three subgenera based on morphology. Brandisia was inferred to have originated in the early Oligocene (32.69 Mya) in the Eastern Himalayas–SW China, followed by diversification in the early Miocene (19.45 Mya) in karst EBLFs. The differentiation dates of Brandisia were consistent with the origin of keystone species of EBLFs in this region (e.g., Fagaceae, Lauraceae, Theaceae, and Magnoliaceae) and the colonization of other characteristic groups (e.g., Gesneriaceae and Mahonia). These findings indicate that the distribution and evolution of Brandisia were facilitated by the rise of the karst EBLFs in East Asia. In addition, the woody and parasitic habits, and pollination characteristics of Brandisia may also be the important factors affecting its speciation and dispersal.

7. Preparation, Structural Characterization and in Vitro Activity of Chromium Complexes of Sweet Corn Cob Polysaccharide

Abstract: To enhance the biological activity of sweet corn cob polysaccharide, a sweet corn cob polysaccharide chromium complex (SCP80-Cr) was prepared, characterized and its in vitro activity was also explored. The results showed that the optimal process conditions for the preparation were pH10.1, reaction temperature 74 ℃, polysaccharide concentration of 3 mg/mL, and the reaction time of 60 min. The weight percentage of Cr(III) in SCP80-Cr was 21.08% and the elemental percentage was 6.67%. The characterization revealed that SCP80-Cr contained almost no protein, nucleic acid or pigment, while Cr was conjugated with SCP80 in the form of Cr-O and Cr-OH and had strong molecular agglomeration properties. The molar percentage of the monosaccharide composition of SCP80-Cr was mannose: glucose: galactose: xylose: arabinose = 0.58: 89.50: 4.65: 1.33: 3.58. The results of in vitro hypoglycemia experiments showed that the IC50 of SCP80-Cr on α-amylase and α-glucosidase inhibitions were 4.70±0.38 mg/mL and 3.99±0.26 mg/mL, respectively. Moreover, the results of in vitro antioxidant experiments showed that SCP80-Cr scavenged DPPH radicals and hydroxyl radicals with IC50 of 4.80±0.34 mg/mL and 4.99±0.28 mg/mL, respectively, indicating that SCP80-Cr had better in vitro hypoglycemic and antioxidant activities compared with SCP80. The results of the study provided some references for the study of sweet corn cob polysaccharides in functional foods.

Abstract: BackgroundThe acute exacerbation of chronic obstructive pulmonary disease (AECOPD) is a common respiratory disease among older adults, which imposes a significant burden on individuals and society and poses a major challenge to the global public health system due to its high morbidity and mortality. Acupuncture is effective for AECOPD, but its efficacy has been questioned due to the limited methodological quality. Thus, we aim to investigate the efficacy of acupuncture as adjunctive therapy for AECOPD and determine whether the efficacy of acupuncture differs with the type of acupoint combinations.Methods and analysisThis study proposes a prospective, multicenter randomized controlled trial that will comprise four groups, including two acupuncture treatment groups, one sham acupuncture group, and one basic treatment group. The acupuncture treatment groups will be distinguished by their focus on different patterns of acupoint combination, namely the Xi-cleft and He-sea acupoint combination and the Eight Confluence points acupoint combination, which may vary in clinical efficacy based on traditional acupuncture theories. The study aims to randomize 556 patients in a 1:1:1:1 ratio across the four groups. Each patient in acupuncture group or sham acupuncture group will receive routine drug therapy and 7 sessions of acupuncture treatment over 1 week. Participants in the basic treatment group will only receive routine drug therapy. The trial will be conducted in seven hospitals located in China. The primary outcomes in this trial will include differences in the Breathlessness, Cough, and Sputum Scale (BCSS) before randomization, 7 days after randomization, 5 and 9 weeks after randomization.Ethics and disseminationEthical approval was obtained from the Sichuan Regional Ethics Review of Committee on Traditional Chinese Medicine (Approval ID: 2022KL-068). 9. High-Temperature-Resistant Epoxy Resin Gel Behavior and Profile Control in Heavy Oil Steam Drive

Abstract: In recent years, the prominence of conformance control technology in heavy oil steam flooding has significantly increased in oilfield development. However, the high-temperature demands of heavy oil steam flooding require more resilient plugging agents. Resin-based plugging agents, known for their exceptional temperature resistance and strength, have emerged as a viable solution within this domain. Yet, they face issues like rapid curing at high temperatures and limited sealing reach. Thus, we introduce a novel approach: epoxy resin gel (EHRB), consisting of epoxy resin (ER) as the curing agent, urotropine (HMTA) and pyrocatechol (RO) as cross-linking agents, and n-butyl glycidyl ether (BGE) as a diluent. EHRB gels at 130 °C in 5.4 h, with curing commencing at 160 °C, extending resin curing time and expanding the sealing radius. This study assessed EHRB’s performance and high-temperature stability through displacement experiments, TGA, and DSC analysis. The results indicate that the EHRB has low viscosity, high compressive strength, and minimal mass loss at high temperatures. At 260 °C, the mass loss is less than 15%. The plugging rate exceeds 90%, and it can withstand steam flushing for over 6 PV. Additionally, it demonstrates a 17% improvement in EOR. EHRB demonstrates outstanding capabilities in proficient channel control for heavy oil steam flooding, offering vital technical support for conformance control in this challenging environment.

10. Screening for inhibitors against SARS-CoV-2 and its variants

Abstract: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to evolve, generating new variants that pose a threat to global health; therefore, it is imperative to obtain safe and broad-spectrum antivirals against SARS-CoV-2 and its variants. To this end, we screened compounds for their ability to inhibit viral entry, which is a critical step in virus infection. Twenty compounds that have been previously reported to inhibit SARS-CoV-2 replication were tested by using pseudoviruses containing the spike protein from the original strain (SARS-CoV-2-WH01). The cytotoxicity of these compounds was determined. Furthermore, we identified six compounds with strong antagonistic activity against the WH01 pseudovirus, and low cytotoxicity was identified. These compounds were then evaluated for their efficacy against pseudoviruses expressing the spike protein from B.1.617.2 (Delta) and B.1.1.529 (Omicron), the two most prevalent circulating strains. These assays demonstrated that two phenothiazine compounds, trifluoperazine 2HCl and thioridazine HCl, inhibit the infection of Delta and Omicron pseudoviruses. Finally, we discovered that these two compounds were highly effective against authentic SARS-CoV-2 viruses, including the WH01, Delta, and Omicron strains. Our study identified potential broad-spectrum SARS-CoV-2 inhibitors and provided insights into the development of novel therapeutics.

11. Recent trends in acupuncture for chronic pain: A bibliometric analysis and review of the literature

Abstract: Background: Acupuncture has been increasingly used in patients with chronic pain, yet no bibliometric analysis of acupuncture studies for chronic pain exists. Objectives: To investigate the characteristics, hotspots and frontiers of global scientific output in acupuncture research for chronic pain over the past decade. Methods: We retrieved publications on acupuncture for chronic pain published from 2011 to 2022 from the Science Citation Index Expanded (SCI-expanded) of the Web of Science Core Collection (WoSCC). The co-occurrence relationships of journals/countries/institutions/authors/keywords were performed using VOSviewer V6.1.2, and CiteSpace V1.6.18 analyzed the clustering and burst analysis of keywords and co-cited references. Results: A total of 1616 articles were retrieved. The results showed that the number of annual publications on acupuncture for chronic pain has increased over time, with the main types of literature being original articles (1091 articles, 67.5 %) and review articles (351 articles, 21.7 %). China had the most publications (598 articles, 37 %), with Beijing University of Traditional Chinese Medicine (93 articles, 5.8 %) and Evidence-based Complementary and Alternative Medicine ranked first (169 articles, 10.45 %) as the most prolific affiliate and journal, respectively. Liang FR was the most productive author (43 articles), and the article published by Vickers Andrew J in 2012 had the highest number of citations (625 citations). Recently, ''acupuncture'' and ''pain'' appeared most frequently. The hot topics in acupuncture for chronic pain based on keywords clustering analysis were experimental design, hot diseases, interventions, and mechanism studies. According to burst analysis, the main research frontiers were functional connectivity (FC), depression, and risk. Conclusion: This study provides an in-depth perspective on acupuncture for chronic painstudies, revealing pivotal points, research hotspots, and research trends. Valuable ideas are provided for future research activities.

12. Seasonal variation of microbial community and methane metabolism in coalbed water in the Erlian Basin, China

Abstract: Coalbed water is a semi-open system connecting underground coalbeds with the external environment. Microorganisms in coalbed water play an important role in coal biogasification and the carbon cycle. The community assemblages of microorganisms in such a dynamic system are not well understood. Here, we used high-throughput sequencing and metagenomic analysis to investigate microbial community structure and identify the potential functional microorganisms involved in methane metabolism in coalbed water in the Erlian Basin, a preferred low-rank coal bed methane (CBM) exploration and research area in China. The results showed that there were differences in the responses of bacteria and archaea to seasonal variation. Bacterial community structure was affected by seasonal variation but archaea was not. Methane oxidation metabolism dominated by Methylomonas and methanogenesis metabolism dominated by Methanobacterium may exist simultaneously in coalbed water.

13. A wavelet based data coupling method for spatial damage detection in beam-type structures.

Abstract: Spatial damage identification is of great significance in mechanical, aerospace, and civil engineering. In this study, a data coupling method based on continuous wavelet transform (CWT) is proposed to identify the spatial damage location of beam-type structures. The singularity of the wavelet coefficient can be used to identify the signal singularity, and data coupling method calculates the spatial location of the damage. Numerical simulations and experimental analyses of different type of beams with transfixion damage are carried out to evaluate the accuracy of the method. The results show that the wavelet based data coupling method (W-DCM) can identify the minimum 4.9% damage severity of fixed beam and continuous beam, and can also identify the damage of non-free end of cantilever beam. However, the 9.7% damage severity of the free end of the cantilever beam cannot be identified. It is also found that the W-DCM can effectively circumvent the problem of wavelet coefficients edge effect. 14. Sensing of cytoplasmic chromatin by cGAS activates innate immune response in SARS-CoV-2 infection

Abstract: Abstract The global coronavirus disease 2019 (COVID-19) pandemic is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a positive-sense RNA virus. How the host immune system senses and responds to SARS-CoV-2 infection remain largely unresolved. Here, we report that SARS-CoV-2 infection activates the innate immune response through the cytosolic DNA sensing cGAS-STING pathway. SARS-CoV-2 infection induces the cellular level of 2′3′-cGAMP associated with STING activation. cGAS recognizes chromatin DNA shuttled from the nucleus as a result of cell-to-cell fusion upon SARS-CoV-2 infection. We further demonstrate that the expression of spike protein from SARS-CoV-2 and ACE2 from host cells is sufficient to trigger cytoplasmic chromatin upon cell fusion. Furthermore, cytoplasmic chromatin-cGAS-STING pathway, but not MAVS-mediated viral RNA sensing pathway, contributes to interferon and pro-inflammatory gene expression upon cell fusion. Finally, we show that cGAS is required for host antiviral responses against SARS-CoV-2, and a STING-activating compound potently inhibits viral replication. Together, our study reported a previously unappreciated mechanism by which the host innate immune system responds to SARS-CoV-2 infection, mediated by cytoplasmic chromatin from the infected cells. Targeting the cytoplasmic chromatin-cGAS-STING pathway may offer novel therapeutic opportunities in treating COVID-19. In addition, these findings extend our knowledge in host defense against viral infection by showing that host cells’ self-nucleic acids can be employed as a “danger signal” to alarm the immune system.

15. Experimental comparison on condensation and evaporation outside double-side enhanced horizontal tubes

Abstract: Experimental investigations of the condensation and evaporation of R410A on the outside of two double-side enhanced heat transfer tubes were conducted, and the tube performance was compared with that of a smooth copper tube. The research object consisted of a smooth tube with a length of 2000 mm and an outer diameter of 25.4 mm and two double-side enhanced tubes of the same size. The evaporation experiments were run under the following conditions: the evaporation saturation temperature was 7°C, water inlet temperature was 8°C–18°C, and water flow rate was 0.6–1.6 m 3 /h. The condensation experiments were run under the following conditions: the saturation temperature was 35°C, water inlet temperature was 21°C–30°C, and water flow rate was 0.4–1.6 m 3 /h. During the data processing, the heat transfer coefficient of the water side was obtained using the Gnielinski-Wilson method, and that of the refrigerant side was obtained using the thermal resistance separation method. The effect of the surface structure on the heat transfer performance was analyzed, and a strengthening measure was proposed. The experimental results indicate that the evaporation heat transfer coefficients outside the smooth tube increased with the raise of the inlet water temperature, while the double-side enhanced tubes decreased; both the condensation heat transfer coefficients outside the smooth tube and the enhanced tubes increased with the increase of the water flow rate. Due to the different type of enhanced tube ribs, tube #3 surface structure is more conducive to the formation of vaporization core, which is about 30% higher than the outer heat transfer coefficient of tube #2. It is more in line with the requirements of strengthening horizontal single tube external pool boiling heat transfer. In the condensation experiment, the condensation heat transfer coefficients outside the smooth tube increased with the raise of the inlet water temperature, while the double enhanced tubes decreased; both the condensation heat transfer coefficients outside the smooth tube and the enhanced tubes increased with the increase of the water flow rate. With the analysis of the rib structure, the outer surface structure of tube #2 has stronger drainage capacity, and the external average condensation heat transfer coefficient is about 7% higher than that of tube #3, which is more in line with the requirements of condensing heat transfer outside the single tube.

Abstract: How to theoretically describe the process of turbulence formation emains an unsolved problem. A new method is presented in this paper for quantitatively researching the turbulence phase transition based on a dimensionless method and catastrophe theory. Then, the formula of energy spectrum density Ek, with the wave number and time-scale index strictly derived from the folding catastrophe model, can quantitatively explain the energy in the turbulent system, inherited from large eddies and transferred to small eddies until viscous dissipation occurs, theoretically and physically. As an example, solid–liquid two-phase flow in a five-stage lifting pump for a 6000 m deep-sea mining system is analyzed based on a structured mesh by numerical simulation, in which this turbulence model is validated. This method provides equations that can give a quantitative analysis with a physical explanation for turbulence formation, which not only provides a new insight into the turbulence, but also can be applied to other complex phase transitions.

Here, we report a strategy, PArsing fragmented DNA Sequences from CRISPR Tiling MUtagenesis Screening (PASTMUS), which provides a streamlined workflow and a bioinformatics pipeline to identify critical amino acids of proteins in their native biological contexts. Using this approach, we map six proteins—three bacterial toxin receptors and three cancer drug targets, and acquire their corresponding functional maps at amino acid resolution.

Abstract: Although the material has developed micro-scale cracks, the micro-cracks stop propagating and transform into non-propagating cracks (NPCs) under fatigue limit loading. The movement of the crack tip position caused by the non-propagating crack will generate a small change in the notch geometry, which easily triggers the geometric size effect. Since the critical distance method focuses on evaluating the limit of fatigue in terms of the material’s cracking conditions, the present study attempted to develop a notched fatigue analysis model to consider the effect of non-propagating cracks on fatigue limit prediction in the critical distance method framework. The effectiveness and capability of the proposed model were demonstrated by the fatigue experimental data of Q345qD low carbon steel.

Abstract: Under the cloud manufacturing (CMfg) environment, a multilayer semantic matching model of design requirement and cloud service resource based on ontology was proposed to guarantee the rapid and accurate matching of resource services to meet the cloud user design requirement. First, the CMfg classification of demand and service was analyzed, and the ontology description was derived according to the service resource classification. From the aforementioned service resource classification, the multilayer matching model was established. The model included six matching stages, namely, base information matching, function information matching, state information matching, quality of service information matching, manufacturing capability information, and comprehensive matching. Then, the matching algorithm of each layer was quantitatively established and described to achieve intelligent matching of the service resource. Meanwhile, an improved analytic hierarchy process was presented to express the influence of each layer on its matching goal scientifically, ensuring that each affecting factor is reasonable. Finally, a case was used to prove the correctness and practicality of the proposed method.

Abstract: The complete chloroplast genome of Poikilospermum lanceolatum was sequenced using HiSeq4000 of Illumina. The length of this genome was 153, 454 bp, including a large single copy (LSC) region (84,202 bp), a small single copy (SSC) region (18,172 bp), and two inverted repeat (IR) regions (25,540 bp). The overall GC content of the genome of P. lanceolatum was 36.9%. The genome included 111 unique genes (78 protein-coding genes, 29 tRNA genes, and four rRNA genes). Phylogenetic analysis based on 67 protein-coding genes showed that Boehmerieae was sister to P. lanceolatum, with 100% bootstrap values.

Abstract: In high-energy collisions, the spatio-temporal size of the particle production region can be measured using the Bose–Einstein correlations of identical bosons at low relative momentum. The source radii are typically extracted using two-pion correlations, and characterize the system at the last stage of interaction, called kinetic freeze-out. In low-multiplicity collisions, unlike in high-multiplicity collisions, two-pion correlations are substantially altered by background correlations, e.g. mini-jets. Such correlations can be suppressed using three-pion cumulant correlations. We present the first measurements of the size of the system at freeze-out extracted from three-pion cumulant correlations in pp, p–Pb and Pb–Pb collisions at the LHC with ALICE. At similar multiplicity, the invariant radii extracted in p–Pb collisions are found to be 5–15% larger than those in pp, while those in Pb–Pb are 35–55% larger than those in p–Pb. Our measurements disfavor models which incorporate substantially stronger collective expansion in p–Pb as compared to pp collisions at similar multiplicity.

26. Highly mutable tandem DNA repeats generate a cell wall protein variant more frequent in disease-causing Candida albicans isolates than in commensal isolates.

Abstract: During adaptation to host environments, many microorganisms alter their cell surface. One mechanism for doing so is variation in the number of amino acid repeats in cell surface proteins encoded by hypermutable DNA tandem repeats. In the yeast Candida albicans, an opportunistic human pathogen, the gene SSR1 encodes a GPI-anchored cell wall protein with a structural role. It contains two regions consisting of tandem repeats, almost exclusively encoding the amino acid pair Ser-Ala. As expected, the repeat regions make SSR1 highly mutable. New SSR1 alleles arose with a frequency of 1.11×10-4 per cell division in serially propagated cells. We also observed a large number (25) of SSR1 alleles with different repeat lengths in a survey of 131 isolates from a global strain collection. C. albicans is diploid, and combinations of these allele generated 41 different SSR1 genotypes. In both repeat regions, nonsynonymous mutations were largely restricted to one particular repeat unit. Two very similar allele combinations were largely restricted to one clade, clade 1. Each combination was present in ~30% of 49 infection-causing clade 1 strains, but one was rare (2%), the other absent in 46 infection-causing strains representing the remainder of the species (P < 0.00018 and 0.00004; Fisher's exact test). These results indicate that both repeat regions are under selection and that amino acid repeat length polymorphisms generate Ssr1 protein variants most suitable for specific genetic backgrounds. One of these two allele combinations was 5.51 times more frequent, the other 1.75 times less frequent in 49 clade 1 strains that caused disease than in 36 commensal clade 1 strains (P = 0.0105; Chi2 test). This indicates that insertion and deletion of repeats not only generates clade-optimized Ssr1p variants, but may also assist in short-term adaptation when C. albicans makes the transition from commensal to pathogen.

Abstract: Quantum-behaved particle swarm optimization (QPSO) is an efficient and powerful population-based optimization technique, which is inspired by the conventional particle swarm optimization (PSO) and quantum mechanics theories. In this paper, an improved QPSO named SQPSO is proposed, which combines QPSO with a selective probability operator to solve the economic dispatch (ED) problems with valve-point effects and multiple fuel options. To show the performance of the proposed SQPSO, it is tested on five standard benchmark functions and two ED benchmark problems, including a 40-unit ED problem with valve-point effects and a 10-unit ED problem with multiple fuel options. The results are compared with differential evolution (DE), particle swarm optimization (PSO) and basic QPSO, as well as a number of other methods reported in the literature in terms of solution quality, convergence speed and robustness. The simulation results confirm that the proposed SQPSO is effective and reliable for both function optimization and ED problems.

31. Climatic Factors Drive Population Divergence and Demography: Insights Based on the Phylogeography of a Riparian Plant Species Endemic to the Hengduan Mountains and Adjacent Regions.

Abstract: Quaternary climatic factors have played a significant role in population divergence and demography. Here we investigated the phylogeography of Osteomeles schwerinae, a dominant riparian plant species of the hot/warm-dry river valleys of the Hengduan Mountains (HDM), Qinling Mountains (QLM) and Yunnan-Guizhou Plateau (YGP). Three chloroplast DNA (cpDNA) regions (trnD-trnT, psbD-trnT, petL-psbE), one single copy nuclear gene (glyceraldehyde 3-phosphate dehydrogenase; G3pdh), and climatic data during the Last Interglacial (LIG; c. 120-140 ka), Last Glacial Maximum (LGM; c. 21 ka), and Current (c. 1950-2000) periods were used in this study. Six cpDNA haplotypes and 15 nuclear DNA (nDNA) haplotypes were identified in the 40 populations of O. schwerinae. Spatial Analysis of Molecular Variance, median-joining networks, and Bayesian phylogenetic trees based on the cpDNA and nDNA datasets, all suggested population divergence between the QLM and HDM-YGP regions. Our climatic analysis identified significant heterogeneity of the climatic factors in the QLM and HDM-YGP regions during the aforementioned three periods. The divergence times based on cpDNA and nDNA haplotypes were estimated to be 466.4-159.4 ka and 315.8-160.3 ka, respectively, which coincide with the time of the weakening of the Asian monsoons in these regions. In addition, unimodal pairwise mismatch distribution curves, expansion times, and Ecological Niche Modeling suggested a history of population expansion (rather than contraction) during the last glaciation. Interestingly, the expansion times were found being well consistent with the intensification of the Asian monsoons during this period. We inferred that the divergence between the two main lineages is probably caused by disruption of more continuous distribution because of weakening of monsoons/less precipitation, whilst subsequent intensification of the Asian monsoons during the last glaciation facilitated the expansion of O. schwerinae populations.

32. Conserved B-cell epitopes among human bocavirus species indicate potential diagnostic targets.

Abstract: BACKGROUND: Human bocavirus species 1-4 (HBoV1-4) have been associated with respiratory and enteric infections in children. However, the immunological mechanisms in response to HBoV infections are not fully understood. Though previous studies have shown cross-reactivities between HBoV species, the epitopes responsible for this phenomenon remain unknown. In this study, we used genomic and immunologic approaches to identify the reactive epitopes conserved across multiple HBoV species and explored their potential as the basis of a novel diagnostic test for HBoVs. METHODOLOGY/PRINCIPAL FINDINGS: We generated HBoV1-3 VP2 gene fragment phage display libraries (GFPDLs) and used these libraries to analyze mouse antisera against VP2 protein of HBoV1, 2, and 3, and human sera positive for HBoVs. Using this approach, we mapped four epitope clusters of HBoVs and identified two immunodominant peptides--P1 (¹MSDTDIQDQQPDTVDAPQNT²⁰), and P2 (¹⁶²EHAYPNASHPWDEDVMPDL¹⁸⁰)--that are conserved among HBoV1-4. To confirm epitope immunogenicity, we immunized mice with the immunodominant P1 and P2 peptides identified in our screen and found that they elicited high titer antibodies in mice. These two antibodies could only recognize the VP2 of HBoV 1-4 in Western blot assays, rather than those of the two other parvoviruses human parvovirus B19 and human parvovirus 4 (PARV4). Based on our findings, we evaluated epitope-based peptide-IgM ELISAs as potential diagnostic tools for HBoVs IgM antibodies. We found that the P1+P2-IgM ELISA showed a higher sensitivity and specificity in HBoVs IgM detection than the assays using a single peptide. CONCLUSIONS/SIGNIFICANCE: The identification of the conserved B-cell epitopes among human bocavirus species contributes to our understanding of immunological cross-reactivities of HBoVs, and provides important insights for the development of HBoV diagnostic tools.

33. TRIM38 negatively regulates TLR3-mediated IFN-β signaling by targeting TRIF for degradation.

Abstract: Toll-like receptors (TLRs) mediated immune response is crucial for combating pathogens and must be tightly controlled. Tripartite motif (TRIM) proteins are a family of proteins that is involved in a variety of biological and physiological processes. Some members of the TRIM family are important in the regulation of innate immunity. Although it has been shown that TRIM38 negatively regulates innate immunity, the mechanisms by which it does so have not been fully addressed. In this study, we demonstrated that TRIM38 negatively regulates Toll-like receptor 3 (TLR3)-mediated type I interferon signaling by targeting TIR domain-containing adaptor inducing IFN-β (TRIF). We found that overexpression of TRIM38 inhibits TLR3-mediated type I interferon signaling, whereas knockdown of TRIM38 has the reverse effects. We further showed that TRIM38 targets TRIF, a critical adaptor protein downstream of TLR3. TRIF is co-immunoprecipitated with TRIM38, and domain mapping experiments show that PRYSPRY of TRIM38 interacts with the N-terminus of TRIF. Overexpression of TRIM38 decreased expression of overexpressed and endogenous TRIF. This effect could be inhibited by MG132 treatment. Furthermore, the RING/B-box domain of TRIM38 is critical for K48-linked polyubiquitination and proteasomal degradation of TRIF. Collectively, our results suggest that TRIM38 may act as a novel negative regulator for TLR3-mediated type I interferon signaling by targeting TRIF for degradation.

Abstract: Introduction: Depression is the most significant contributor to non-fatal health reductions worldwide. Acupuncture is the most commonly used complementary alternative therapy to relieve major depressive disorder (MDD) effectively. Nevertheless, the effects of acupuncture for MDD are uncertain. This review aimed to determine the efficacy and safety of acupuncture for MDD. Methods: Meta-analysis was performed for randomized controlled trials of acupuncture for MDD data from eight databases searched from inception until February 10, 2022. All RCTs with adult participants undergoing acupuncture treatment for MDD were included. The primary outcome measure was the Hamilton rating scale for depression (HAMD). We used random-effects meta-analysis to synthesize the results with a mean difference or odds ratio. Furthermore, the potential heterogeneity was tested through meta-regression/subgroup analyses/sensitive analysis. The quality of evidence for each outcome was assessed by the Grading of Recommendations Assessment, Development, and Evaluation approach. Results: Forty-three studies were included: 9 acupuncture versus sham acupuncture (n = 920), 26 acupuncture versus antidepressants (n = 2169), and 9 acupuncture plus antidepressants versus antidepressants (n = 667). Of the 43 high-quality articles, 24 and 8 were determined to have a low and moderate risk of bias, respectively. The pooled results for HAMD and SDS revealed the clinical benefits of acupuncture or acupuncture plus antidepressants compared to sham acupuncture or antidepressants, with high-quality evidence. Furthermore, high-quality evidence showed that acupuncture led to fewer adverse effects than antidepressants. Conclusions: Acupuncture or acupuncture plus antidepressants were significantly associated with reduced HAMD scores, with high-quality evidence. Also, more rigorous trials are needed to identify the optimal frequency of acupuncture for MDD and integrate such evidence into clinical care to reduce antidepressant use.

45. Effectiveness and Safety of Electroacupuncture for Depression: A Systematic Review and Meta-Analysis

Abstract: Purpose. The purpose of this systematic review and meta-analysis was to comprehensively evaluate the efficacy and safety of electroacupuncture as an effective adjunctive therapy for patients with depression. Methods. Randomized controlled trials (RCTs) about the treatment of depression by electroacupuncture therapy from inception to September 2021 were searched and collected in eight databases. HAMD, SDS, and Adverse Reactions were used as outcome indicators. The quality of relevant articles was evaluated using the Cochrane Collaboration’s risk of bias tool. The quality of evidence for each outcome was assessed through the Grading of Recommendations Assessment and the Development and Evaluation approach. Stata 15.0 software was used for data analysis. Results. A total of 16 depression-related RCTs were included in this meta-analysis. For the main outcome, electroacupuncture significantly reduced HAMD scores (I2:0.0%, SMD: −2.28% (95% CI−3.16 to −1.39)), and the quality of evidence was moderate. The improvement effect of electroacupuncture plus antidepressants was better than that of western drugs in patients with depression (I2:26.2%, SMD: −1.18% (95% CI−1.42 to −0.94)), and the quality of evidence was moderate. Electroacupuncture significantly reduced HAMD scores without significant heterogeneity (I2:0.0%, SMD: −3.76% (95%CI−5.78−1.73)). Studies with very low quality of evidence found that electroacupuncture was as effective as antidepressants in reducing SDS scores (I2:36.4%, WMD: −1.15% (95%CI−2.93–0.63)), and electroacupuncture was found to be more effective than sham electroacupuncture stimulation as well. Moderate quality evidence showed no statistical difference between electroacupuncture plus antidepressants/electroacupuncture and antidepressants (I2:0%, RR:1.05% (95%CI 0.73 to 1.53)). Conclusions. Our meta-analysis shows that electroacupuncture reduces HAMD scores. It is suggested to use electroacupuncture plus antidepressants to improve the curative effect and effectively reduce drug side effects.

46. Mutational landscape and clinical outcome of pediatric acute myeloid leukemia with 11q23/ <scp> KMT2A </scp> rearrangements

Abstract: Alterations of 11q23/KMT2A are the most prevalent cytogenetic abnormalities in acute myeloid leukemia (AML) and the prognostic significance of 11q23/KMT2A-rearranged AML based on various translocation partners varies among different studies. However, few studies evaluated the molecular characteristics of 11q23/KMT2A-rearranged pediatric AML. We aim to analyze the mutational landscape of 11q23/KMT2A-rearranged AML and assess their prognostic value in outcomes.The mutational landscape and clinical prognosis of 105 children with 11q23/KMT2A-rearranged AML in comparison with 277 children with non-11q23/KMT2A-rearranged AML were analyzed using publicly accessible next-generation sequencing data from Therapeutically Applicable Research to Generate Effective Treatments (TARGET) dataset.Pediatric AML patients with 11q23/KMT2A-rearrangements harbored a low number of mutations (Median, 1 mutation/patient, range, 1-22), 58% of which involved in RAS pathway mutations (KRAS, NRAS, and PTPN11) and 10.5% of which comprised of SETD2 mutations. Compared with non-11q23/KMT2A-rearranged AML, the incidence of KRAS (32.4% vs. 10.1%, P〈0.001) and SETD2 (10.5% vs. 1.4%, P=0.001) gene mutations in 11q23/KMT2A-rearranged AML was significantly higher. Both KRAS and SETD2 mutations occurred more often in t(10;11)(p12;q23). KRAS mutations were correlated with worse 5-year event-free survival [EFS] (Plog-rank = 0.001) and 5-year overall survival [OS] (Plog-rank = 0.009) and the presence of SETD2 mutations increases the 5-year relapse rate (PGray = 0.004). Multivariate analyses confirmed KRAS mutations in 11q23/KMT2A-rearranged AML as an independent predictor for poor EFS (hazard ratio [HR] = 2.10, P=0.05) and OS (HR = 2.39, P=0.054).Our findings show that pediatric patients with 11q23/KMT2A rearrangements have characteristic mutation patterns and varying clinical outcomes depending on different translocation partners, which could be utilized to develop more accurate risk stratification and tailored therapies.

Abstract: The purpose of this study is to analyze and visualize the research trends on acupuncture therapy for postoperative pain over the past 20 years to identify hotspots and frontiers, and provide new research ideas.A search of the Web of Science database, with a time frame of 2001-01-01 to 2022-02-28, was conducted to collect literatures related to acupuncture therapy for postoperative pain. A bibliometric analysis and visualization of results was performed using CiteSpace software for the volume of annual publications, journals, countries, institutions, authors, keywords, and references.A total of 840 literatures were eventually included in the analysis. The number of publications has fluctuated upwards each year over the past 20 years and reached a peak in the latest three years.The field of acupuncture therapy for postoperative pain is currently in a period of high growth. China and the USA have made the largest contribution to the volume of publications. The most influential institutions and authors are mainly from China and South Korea. The overall collaborative network needs to be strengthened. Electroacupuncture and auricular acupuncture (therapeutic techniques), low back surgery (types of surgery), and "postoperative pain, nausea and vomiting" are research hotspots in this field. Improvement of postoperative life quality, proof of clinical efficacy and evidence-based evaluation are the current research trends and frontiers.

50. Optical gain calculation and structure optimization of type-II GaAsSb/InAs/InGaAs quantum well heterostructures

Abstract: This paper reports an optimization scheme of type-II GaAsSb/InAs/InGaAs quantum wells using the six bands k · p method. By calculating the optical gain, the optimal structures of the quantum wells are obtained. We observed that without an electric field, the thickness values of symmetrical quantum wells designed by the AdaGrad algorithm are 2.97, 2.81, and 6.21 nm, respectively, and the gain intensity is 6489 cm−1 that is about 1600 cm−1 higher than the original empirical design scheme. The thickness of asymmetric quantum wells designed by the Adam algorithm is 5.56, 2.72, 1.88, 3.16, and 2.46 nm under the external electric field of 75 kV/cm, respectively, and the gain intensity is 5729 cm−1 that is about 2300 cm−1 higher than the original empirical design scheme. The optimized scheme effectively suppresses the gain attenuation caused by the electric field and is helpful to improve the performance of interband cascade lasers.
