BackgroundThe 24-Hour Movement Guidelines (24-HMG) recommend a balanced combination of physical activity (PA), sedentary behavior (SB) and sleep (SLP) for optimal health. However, there is limited understanding of how well U.S. adolescents adhere to these guidelines. This study aims to analyze the prevalence trends of meeting the 24-HMG among a nationally representative sample of U.S. general adolescents.MethodsThe study included 2,273 adolescents (55.3% boys) aged 16–19 who participated in the National Health and Nutrition Examination Surveys (NHANES) from 2007 to 2016. The researchers categorized the adolescents based on whether they met various PA, SB, and SLP recommendations, as well as different combinations of these recommendations, separately for boys and girls. The prevalence rate, weighted by survey data, was calculated along with a 95% confidence interval (CI) to assess the changes in meeting the 24-HMG among U.S. adolescents across different survey years and sociodemographic subgroups.ResultsIn the 2015–2016 cycle, approximately 6.3% of adolescents did not meet any of the three recommendations, while only 19.2% of adolescents achieved all three guidelines. Compliance with PA and SB recommendations among adolescents has decreased over time, from 72.5% (65.9% to 79.2%) to 64.2% (57.4% to 70.9%) for PA, and from 59.0% (49.6% to 68.4%) to 46.6% (37.8% to 55.5%) for SB, respectively, from 2007–2008 cycle to 2015–2016 cycle. Revealing the relationship between the mating system (i.e., the outcrossing/inbreeding degree) and the fitness of seeds in tree species under wild conditions is essential for understanding the ecological adaptability and evolutionary stability of the species. This study collected open-pollinated seeds from seven wild populations of Pinus yunnanensis var. tenuifolia that exhibited fragmentation in the Nanpan–Hongshui River basin, an ecologically fragile area in China. The seeds and sprouts (germinated seeds) from 20 families were genotyped (24 seeds and 24 sprouts per family) using twelve microsatellite loci to reveal the genetic diversity, mating status, and effect of inbreeding on the three seed quality indicators (thousand-seed weight, germination rate, and germination potential). The three seed quality indicators differed significantly between families (p < 0.001). Higher values of genetic diversity (except the observed heterozygosity) were observed in the sprout group than those in the seed group. Families from different populations showed a notable genetic differentiation (Φst = 0.12), and a large part of families from the common populations had a high degree of coancestry, which signified that the current habitat fragmentation is limiting gene flow between populations. High levels of outcrossing rates (tm) were observed in both the seed group (tm = 0.974) and the sprout group (tm = 0.978), indicating that a low proportion of seeds were self-fertilized. Although there was a slightly higher single outcrossing rate (ts = 0.888) and a lower proportion of biparental inbreeding (tm − ts = 0.077) in sprouts compared to the seeds (ts = 0.871, tm − ts = 0.091), indicating that a part of inbred seeds were purged during the germination stage, curve fitting between the outcrossing rate and seed quality indicators showed that a certain degree of biparental inbreeding (ts between 0.89 and 0.91 and tm − ts between 0.09 and 0.11) did have a positive effect on seed germination ability. This highlights that excessive inbreeding or outbreeding seems to be unfavorable to seed viability. The peculiar relationship between seed viability and the mating system in P. yunnanensis var. tenuifolia was likely an evolutionary consequence of a trade-off between the nature of mixed mating and its specific ecological niche.

Increasing evidence indicates that mitochondrial fission imbalance plays an important role in delayed neuronal cell death. Our previous study found that photobiomodulation improved the motor function of rats with spinal cord injury. However, the precise mechanism remains unclear. To investigate the effect of photobiomodulation on mitochondrial fission imbalance after spinal cord injury, in this study, we treated rat models of spinal cord injury with 60-minute photobiomodulation (810 nm, 150 mW) every day for 14 consecutive days. Transmission electron microscopy results confirmed the swollen and fragmented alterations of mitochondrial morphology in neurons in acute (1 day) and subacute (7 and 14 days) phases. Photobiomodulation alleviated mitochondrial fission imbalance in spinal cord tissue in the subacute phase, reduced neuronal cell death, and improved rat posterior limb motor function in a time-dependent manner. These findings suggest that photobiomodulation targets neuronal mitochondria, alleviates mitochondrial fission imbalance-induced neuronal apoptosis, and thereby promotes the motor function recovery of rats with spinal cord injury.

Alcohol use accounts for a large variety of diseases, among which alcoholic liver injury (ALI) poses a serious threat to human health. In order to overcome the limitations of chemotherapeutic agents, some natural constituents, especially polysaccharides from edible medicinal plants (PEMPs), have been applied for the prevention and treatment of ALI. In this review, the protective effects of PEMPs on acute, subacute, subchronic, and chronic ALI are summarized. The pathogenesis of alcoholic liver injury is analyzed. The structure–activity relationship (SAR) and safety of PEMPs are discussed. In addition, the mechanism underlying the hepatoprotective activity of polysaccharides from edible medicinal plants is explored. PEMPs with hepatoprotective activities mainly belong to the families Orchidaceae, Solanaceae, and Liliaceae. The possible mechanisms of PEMPs include activating enzymes related to alcohol metabolism, attenuating damage from oxidative stress, regulating cytokines, inhibiting the apoptosis of hepatocytes, improving mitochondrial function, and regulating the gut microbiota. Strategies for further research into the practical application of PEMPs for ALI are proposed. Future studies on the mechanism of action of PEMPs will need to focus more on the utilization of multi-omics approaches, such as proteomics, epigenomics, and lipidomics.

The difference (DIF) between land surface temperature (Ts) and near surface air temperature (Ta) is the key indicator of the energy budget of the land surface, which has a more complex process than the individual Ts or Ta. However, the spatiotemporal variations and influencing factors of DIF remain incomplete. The contribution of vegetation and soil moisture (SM) as key driving factors to DIF is not yet clear. Here, we analyzed the spatiotemporal variation patterns of DIF in China from 2011 to 2023 using in situ Ts and Ta data. A convergent cross-mapping method was employed to explore the causal relationship between SM, NDVI and DIF, and subsequently calculated the contribution of NDVI and SM variations to DIF under different climatic backgrounds. The results indicate that during the study period, DIF values were all above 0 °C and showed a significant increasing trend with a national mean slope of 0.02 °C/a. In general, vegetation and SM have a driving effect on DIF, with vegetation contributing more to DIF (0.11) than SM (0.08) under different surface properties. The background values of SM and temperature have a significant effect on the spatial and temporal distribution of DIF, as well as the correlation of vegetation and soil moisture to DIF. The study outcomes contribute to a better understanding of the coupling relationship between the land surface and atmosphere, which are also crucial for addressing climate change and ecological environmental management. Climate change impacts on species and ecosystem functioning may depend on climatic context and study systems. Climate warming and intensified herbivory are two stressors to plants that often appear in combination and are predicted to increase in cold environments. Effects of multiple drivers on plant performance are difficult to predict and warrant studies that use experimental manipulations along climatic gradients to produce more realistic knowledge. Our three study sites by the Sognefjord in Norway, that differed mainly in climatic conditions (ca. 5°C growing season difference), ranged from hemi-boreal lowland (100 masl, Low), via boreal mid-montane (500 masl) to alpine timberline (900 masl, High) bioclimates. At each site, in a randomized block design, we simulated growing-season warming using open-top chambers (OTCs) and experimentally induced herbivory resistance using the plant hormone methyl jasmonate (MeJA). We recorded growth, mortality, flower and fruit numbers, and insect herbivory on tagged ramets in permanent plots across three years (2016-2018) in three open woodland populations of two functionally important plant species with contrasting traits, Vaccinium myrtillus (bilberry) and V. vitis-idaea (lingonberry). Growth of both dwarf shrubs decreased with warming in the warm lowland populations (Low) but increased in the alpine populations (High). Shoot mortality increased most with warming at Low but was reduced at High. Reproduction, both flowering and fruiting, decreased with induced resistance treatment, but the effect was larger when warmed for bilberry and increased with elevation for both species. Leaf herbivory in bilberry increased with warming at Low but decreased at High. The combined warming and resistance treatment had only synergistic negative interaction effects on fruit numbers in bilberry. The clear context-and species-dependent effects of climate warming and increased resistance in this study may predict a potential decline in performance, as well as abundance and distribution, of these functionally important Vaccinium species at our lowest site. Bilberry reproduction appeared to be particularly susceptible to both climate warming and induced resistance in the manipulated populations. Spinal fusion, a common surgery performed for degenerative lumbar conditions, often uses recombinant human bone morphogenetic protein 2 (rhBMP-2) that is associated with adverse effects. Mesenchymal stromal/stem cells (MSCs) and their extracellular vesicles (EVs), particularly exosomes, have demonstrated efficacy in bone and cartilage repair. However, the efficacy of MSC exosomes in spinal fusion remains to be ascertained. This study investigates the fusion efficacy of MSC exosomes delivered via an absorbable collagen sponge packed in a poly Ɛ-caprolactone tricalcium phosphate (PCL–TCP) scaffold in a rat posterolateral spinal fusion model. Herein, it is shown that a single implantation of exosome-supplemented collagen sponge packed in PCL–TCP scaffold enhanced spinal fusion and improved mechanical stability by inducing bone formation and bridging between the transverse processes, as evidenced by significant improvements in fusion score and rate, bone structural parameters, histology, stiffness, and range of motion. This study demonstrates for the first time that MSC exosomes promote bone formation to enhance spinal fusion and mechanical stability in a rat model, supporting its translational potential for application in spinal fusion.
