Crops have diverse structures and complex growth environments. RGB image data can reflect the texture and color features of plants accurately, while 3D data contains information about crop volume. The combination of RGB image and 3D point cloud data can achieve the extraction of two-dimensional and three-dimensional phenotypic parameters of crops, which is of great significance for the research of phenomics methods. In this study, potatoe plants were chosen as the research subject, and RGB cameras and laser scanners were used to collect 50 potato RGB images and 3D laser point cloud data. The segmentation accuracy of four deep learning semantic segmentation methods, OCRNet, UpNet, PaNet, and DeepLab v3+, were compared and analyzed for the RGB images. OCRNet, which demonstrated higher accuracy, was used to perform semantic segmentation on top-view RGB images of potatoes. Mean shift clustering algorithm was optimized for laser point cloud data processing, and single-plant segmentation of laser point cloud data was completed. Stem and leaf segmentation of single-plant potato point cloud data were accurately performed using Euclidean clustering and K-Means clustering algorithms. In addition, a strategy was proposed to establish a one-to-one correspondence between RGB images and point clouds of single-plant potatoes using pot numbering. 8 2D phenotypic parameters and 10 3D phenotypic parameters, including maximum width, perimeter, area, plant height, volume, leaf length, and leaf width, etc., were extracted from RGB images and laser point clouds, respectively. Finally, the accuracy of three representative and easily measurable phenotypic parameters, leaf number, plant height, and maximum width were evaluated. The mean absolute percentage errors (MAPE) were 8.6%, 8.3% and 6.0%, respectively, while the root mean square errors (RMSE) were 1.371 pieces, 3.2 cm and 1.86 cm, respectively, and the determination coefficients (R2) were 0.93, 0.95 and 0.91, respectively. The research results indicated that the extracted phenotype parameters can accurately and efficiently reflect the growth status of potatoes. Combining the RGB image data of potatoes with three-dimensional laser point cloud data can fully exploit the advantages of the rich texture and color characteristics of RGB images and the volumetric information provided by three-dimensional point clouds, achieving non-destructive, efficient, and high-precision extraction of two-dimensional and three-dimensional phenotype parameters of potato plants. The achievements of this study could not only provide important technical support for the cultivation and breeding of potatoes but also provide strong support for phenotype-based research.

This paper proposes a generalized active disturbance rejection controller (GADRC) based hierarchical control structure for the boilerturbine unit. In the lower layer, a multivariable extended state observer (MESO) is developed to estimate the values of the lumped disturbances caused by modelling mismatches, fuel quality variation and wide range load variation. The influence of the disturbances is then compensated at the input side as a feedforward control. In the upper layer, the multi-objective optimization is devised to obtain the set-points by removing the plant behaviour variation from the optimized model in a feasible way. The lowpass filter acting on the lumped disturbances is designed to bridge the gap between the lower and upper layer. The impact of the feedthrough item is approximated by a first-order system and a two degree-of-freedom (2-DOF) control strategy is established to illustrate the set-point tracking and disturbance rejection properties of the controller. Simulation studies on a 1000MWe coal-fired ultra-supercritical boiler-turbine unit demonstrate that the proposed control strategy can achieve a satisfactory performance in cases of fuel quality variations, model-plant mismatches and wide range load variation.

Abstract Background Kaposi's sarcoma-associated herpesvirus (KSHV) is causally linked to several acquired immunodeficiency syndrome-related malignancies, including Kaposi's sarcoma (KS), primary effusion lymphoma (PEL) and a subset of multicentric Castleman's disease. Regulation of viral lytic replication is critical to the initiation and progression of KS. Recently, we reported that herpes simplex virus type 1 (HSV-1) was an important cofactor that activated lytic cycle replication of KSHV. Here, we further investigated the possible signal pathways involved in HSV-1-induced reactivation of KSHV. Results By transfecting a series of dominant negative mutants and protein expressing constructs and using pharmacologic inhibitors, we found that either Janus kinase 1 (JAK1)/signal transducer and activator of transcription 3 (STAT3) or JAK1/STAT6 signaling failed to regulate HSV-1-induced KSHV replication. However, HSV-1 infection of BCBL-1 cells activated phosphatidylinositol 3-kinase (PI3K)/protein kinase B (PKB, also called AKT) pathway and inactivated phosphatase and tensin homologue deleted on chromosome ten (PTEN) and glycogen synthase kinase-3β (GSK-3β). PTEN/PI3K/AKT/GSK-3β pathway was found to be involved in HSV-1-induced KSHV reactivation. Additionally, extracellular signal-regulated protein kinase (ERK) mitogen-activated protein kinase (MAPK) pathway also partially contributed to HSV-1-induced KSHV replication. Conclusions HSV-1 infection stimulated PI3K/AKT and ERK MAPK signaling pathways that in turn contributed to KSHV reactivation, which provided further insights into the molecular mechanism controlling KSHV lytic replication, particularly in the context of HSV-1 and KSHV co-infection.

Simple Summary: In addition to porcine parvovirus 1 (PPV1), seven new PPVs (PPV2-PPV8) have been identified in the last two decades. However, the prevalence and evolution of PPVs in slaughtered pigs in China are still unclear. The infection status in slaughtered pigs could reflect the overall health situation during pig production in swine herds. Therefore, we detected the infection status of PPVs in 353 samples collected from slaughtered pigs in six regions of China in 2023. Overall, 79.32% of the samples were PPV positive, with 67.50% PPV-positive samples co-infecting with two to six PPVs. Six species of PPVs were detected, except for PPV4 and PPV8. Representative PPV genomes were determined for evaluating evolutionary relationships and detecting recombination events. A genome-based phylogenetic tree confirmed the PCR results, and cross-over events were detected in the PPV2 and PPV3 strains identified in this study. This study provided the first clue on the prevalence and evolution of PPVs in slaughtered pigs in China. Porcine parvoviruses one through eight (PPV1-PPV8) are prevalent in Chinese swine herds. However, the infection status of all these PPVs in slaughtered pigs is still unclarified. In this study, we detected PPV1-PPV8 in 353 tissue samples collected from slaughtered pigs from six regions of China in 2023. At least one species of PPV was detected in 79.32% of the samples (280 out of 353). Six PPV species were detected, except for PPV4 and PPV8, in slaughtered pigs, within which PPV3 (49.86%), PPV2 (42.49%), and PPV7 (42.21%) were predominant, followed by PPV1 (13.31%), PPV6 (13.31%), and PPV5 (8.22%). Noticeably, co-infection was frequently detected, with 67.50% of PPV-positive samples (189 out of 280) co-infecting with two to six PPVs. In addition, one representative genome for each detected PPV was determined. Multiple sequence alignment determined a large number of substitutions in capsid proteins of PPVs. Genome-based phylogenetic analysis confirmed the PCR detection results. Recombination detection identified two potential recombinants (PPV2 GDCZ2023-2088 strain and PPV3 HLJSYS2023-1654 strain) in slaughtered pigs. Mixed-flow pumps, optimized for marine engineering, provide a balance of high efficiency and adaptability, accommodating varied flow and head demands across challenging oceanic settings and are essential for reliable operations in tidal energy and subsea applications. The primary purpose of this paper is to perform a numerical analysis of the cavitation flow characteristics of the mixed-flow pump under differing operational circumstances. The cavitation simulation was implemented to explore the cavitation bubbles evolution and the pressure pulsation characteristics in the impeller region under diverse flow rates, utilizing the Shear Stress Transport (SST) turbulence model and the Zwart-Gerber-Belamri cavitation model as a foundation. The findings indicate that cavitation bubbles initially distribute at the leading edge of blade suction surfaces at the cavitation growth stage. The bubbles spread gradually with the decline of the available net positive suction head (NPSHa). At the same time, many bubbles appear in the area below the blade and extend to the rim of the suction side of blades. As the flow rate decreases, the critical net positive suction head (NPSHc) gradually declines. The dominant pressure pulsation frequency at the impeller inlet is the blade passing frequency, and the vibration at the impeller shroud inlet is more intense than that at the hub. The dominant frequency at the impeller outlet is mainly the blade passing frequency. With the development of cavitation, it changes to impeller rotation frequency at low flow rates, while the dominant frequency remains unchanged at high flow rates. [ABSTRACT FROM AUTHOR]

To investigate the clinical value of multi-slice spiral computed tomography (CT) angiography (MSCTA) combined with MRI in the diagnosis of cerebral aneurysm. A total of 90 patients with cerebral aneurysms diagnosed by DSA were selected as the subjects of this study. Another 30 patients with cerebral infarction were selected as negative controls (NC). Before diagnosis, all patients underwent comprehensive examination using MSCTA and MRI. The results of the comparison and the clinical data of all patients were retrospectively analyzed. MSCTA and MRI examinations can clearly show the specific location, shape, size and anatomical relationship with surrounding tissues of cerebral aneurysms. MSCTA diagnosed 82 patients and missed or misdiagnosed 8 patients in the 90 patients with cerebral aneurysm. The diagnostic sensitivity and accuracy of MSCTA were 91.1 (82/90) and 89.2 (107/120), respectively. MRI examination diagnosed 87 patients and missed or misdiagnosed 3 patients in the 90 patients with cerebral aneurysm. The diagnostic sensitivity and accuracy of MRI were 96.7 (87/90) and 96.7 (116/120), respectively. The sensitivity and accuracy of MSCTA combined with MRI were 100.0 (90/90) and 99.2 (119/120), respectively. MSCTA combined with MRI can not only display the whole picture of brain tissue, but also display the size, shape and relationship with the parent vessel of the aneurysm. The combination of MSCTA and MRI has high sensitivity and accuracy in diagnosing intracranial aneurysms, which provides a promising diagnostic protocol for patients with aneurysms. African swine fever (ASF), a highly infectious and devastating disease affecting both domestic pigs and wild boars, is caused by the African swine fever virus (ASFV). ASF has resulted in rapid global spread of the disease, leading to significant economic losses within the swine industry. A significant obstacle to the creation of safe and effective ASF vaccines is the existing knowledge gap regarding the pathogenesis of ASFV and its mechanisms of immune evasion. The cyclic GMP–AMP synthase (cGAS)–stimulator of interferon genes (STING) pathway is a major pathway mediating type I interferon (IFN) antiviral immune response against infections by diverse classes of pathogens that contain DNA or generate DNA in their life cycles. To evade the host's innate immune response, ASFV encodes many proteins that inhibit the production of type I IFN by antagonizing the cGAS-STING signaling pathway. Multiple proteins of ASFV are involved in promoting viral replication by protein–protein interaction during ASFV infection. The protein QP383R could impair the function of cGAS. The proteins EP364R, C129R and B175L could disturb the function of cyclic guanosine monophosphate-adenosine monophosphate (cGAMP). The proteins E248R, L83L, MGF505-11L, MGF505-7R, H240R, CD2v, E184L, B175L and p17 could interfere with the function of STING. The proteins MGF360-11L, MGF505-7R, I215L, DP96R, A151R and S273R could affect the function of TANK Binding Kinase 1 (TBK1) and IκB kinase ε (IKKε). The proteins MGF360-14L, M1249L, E120R, S273R, D129L, E301R, DP96R, MGF505-7R and I226R could inhibit the function of Interferon Regulatory Factor 3 (IRF3). Different gravity fields have important effects on the structural morphology of bone. The fluid flow caused by loadings in the bone lacunar-canalicular system (LCS), converts mechanical signals into biological signals and regulates bone reconstruction by affecting effector cells, which ensures the efficient transport of signaling molecules, nutrients, and waste products. In this study, the fluid flow and mass transfer effects of bone lacunar-canalicular system at multi-scale were firstly investigated, and a three-dimensional axisymmetric fluid-solid coupled finite element model of the LCS within three continuous osteocytes was established. The changes in fluid pressure field, flow velocity field, and fluid shear force variation on the surface of osteocytes within the LCS were studied comparatively under different gravitational fields (0 G, 1 G, 5 G), frequencies (1 Hz, 1.5 Hz, 2 Hz) and forms of cyclic compressive loading. The results showed that different frequencies represented different exercise intensities, suggesting that high-intensity exercise may accelerate the fluid flow rate within the LCS and enhance osteocytes activity. Hypergravity enhanced the transport of solute molecules, nutrients, and signaling molecules within the LCS. Conversely, the mass transfer in the LCS may be inhibited under microgravity, which may cause bone loss and eventually lead to the onset of osteoporosis.
