Abstract

Objective To study the effects and mechanisms of molecular targeted drug combination on multi-driven proliferation hepatocellular carcinoma SK-Hep-1 cells. Methods Four molecular targeted drugs (HG6-64-1, Dasatinib, Crizotinib, and Sunitinib) were used to treat SK-Hep-1 cells, and the monophasic kinetic analysis curve and two-phase analysis curve were drawn. Western blot analysis was used to detect the effects of the above drugs on key signaling pathways in SK-Hep-1 cells. MTT assay was used to detect the effects of the above drugs and their combination on the proliferation of SK-Hep-1 cells. Results Compared with the monophasic kinetic analysis curve, the biphase analysis curve could better fit the effects of molecular targeted drugs on SK-Hep-1 cells, which predicted that the combination of HG6-64-1, Dasatinib, and MK-2206 could effectively inhibit the proliferation of SK-Hep-1 cells. Conclusion Two-phase kinetic analysis can quantitatively describe the response of multi-driven proliferation hepatocellular carcinoma SK-Hep-1 cells to molecular targeted therapy. The combination of HG6-64-1, Dasatinib, and MK-2206 is a potential drug combination for the treatment of hepatocellular carcinoma.

Abstract

China is one of the major sources of student migrants to many Western countries, growing rapidly during the last couple of decades. In China, several national and regional level policy schemes have been set up to incentivise the return of the high-skilled overseas population. Data to monitor these migration patterns are typically lacking. In this article, we explore the spatial patterns of overseas alumni populations from 106 leading Chinese universities using data gathered from the LinkedIn advertising platform. We first assess the suitability of the LinkedIn data for measuring overseas migrant distributions and then adapt an extended gravity model to aid the interpretation of the relationships between countries, universities and intermediate characteristics and the size of the overseas alumni populations. We find that the LinkedIn data provide plausible measures of Chinese university alumni networks. Alumni populations are in general larger from highly ranked universities, in greater numbers from universities in Beijing and Shanghai and universities with higher numbers of foreign students. These findings help better understand human capital flight, where conventional studies use migration data that do not typically have breakdowns to sub-national units or specify where emigrants received their education. Abstract

A hierarchical consensus control algorithm based on value function decomposition is proposed for hierarchical multi-agent systems. To implement the consensus control algorithm, the reward function of the multi-agent systems can be decomposed, and two value functions can be obtained by analyzing the communication content and the corresponding control objective of each layer in the hierarchical multi-agent systems. Therefore, for each agent in the systems, a dual-critic network and a single-actor network structure are applied to realize the objective of each layer. In addition, the target network is introduced to prevent overfitting in the critic network and improve the stability of the online learning process. During the updating of network parameters, a soft updating mechanism and experience replay buffer are introduced to slow down the update rate of the network and improve the utilization rate of training data. The convergence and stability of the consensus control algorithm with the soft updating mechanism are analyzed theoretically. Finally, the correctness of the theoretical analysis and the effectiveness of the algorithm were verified by two experiments. [ABSTRACT FROM AUTHOR]

48. Higher Dietary Inflammatory Index and Increased Mortality Rate of Adults With Hyperuricemia: Findings From the National Health and Nutritional Examination Survey (2001–2018).

Abstract

Objective: This study aimed to assess the association between Dietary Inflammatory Index (DII) score and death among adults with hyperuricemia. Methods: We collected data from the 2001 to 2018 cohorts of the National Health and Nutritional Examination Survey. Death information was obtained based on death certificate records from the National Death Index through December 31, 2019. The associations between DII score and all‐cause, cardiovascular disease (CVD), and cancer death were investigated by using Cox proportional hazards regression models. Results: We enrolled 7,786 participants with hyperuricemia in this study. The DII score ranged from −4.42 to 4.61. Higher DII score was significantly associated with higher levels of body mass index, glycohemoglobin, glucose, low‐density lipoprotein–cholesterol, and C‐reactive protein (all P < 0.05). During 67,851 person‐years of follow‐up, deaths of 1,456 participants were identified, including 532 CVD deaths and 246 cancer deaths. After adjusting for potential variables, significant higher risk of all‐cause (hazard ratio [HR] 1.18, 95% confidence interval [95% CI] 1.03–1.36, P = 0.01) and CVD (HR 1.30, 95% CI 1.03–1.63, P = 0.02) death was observed for individuals with higher DII scores. Considering the DII score as a continuous variable, the risk of all‐cause and CVD death increases 5% (HR 1.05, 95% CI 1.01–1.08) and 8% (HR 1.08, 95% CI 1.02–1.15) with each one‐unit increment in DII score, respectively. Subgroup analysis indicated that the association between DII score and all‐cause death among participants with hyperuricemia was more significant in males. Conclusion: DII score is found to be positively associated with all‐cause and CVD death of adults with hyperuricemia. Controlling the intake of proinflammatory food might be a potential strategy to reduce the mortality rate. [ABSTRACT FROM AUTHOR]
