15. Molecular dissection of an intronic enhancer governing cold-induced expression of the vacuolar invertase gene in potato.

Abstract

Potato (Solanum tuberosum) is the third most important food crop in the world. Potato tubers must be stored at cold temperatures to minimize sprouting and losses due to disease. However, cold temperatures strongly induce the expression of the potato vacuolar invertase gene (VInv) and cause reducing sugar accumulation. This process, referred to as "cold-induced sweetening," is a major postharvest problem for the potato industry. We discovered that the cold-induced expression of VInv is controlled by a 200 bp enhancer, VInvIn2En, located in its second intron. We identified several DNA motifs in VInvIn2En that bind transcription factors involved in the plant cold stress response. Mutation of these DNA motifs abolished VInvIn2En function as a transcriptional enhancer. We developed VInvIn2En deletion lines in both diploid and tetraploid potato using clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated nuclease 9 (Cas9)-mediated gene editing. VInv transcription in cold-stored tubers was significantly reduced in the deletion lines. Interestingly, the VInvIn2En sequence is highly conserved among distantly related Solanum species, including tomato (Solanum lycopersicum) and other non-tuber-bearing species. We conclude that the VInv gene and the VInvIn2En enhancer have adopted distinct roles in the cold stress response in tubers of tuber-bearing Solanum species. 40. Modulating the Electronic Coordination Configuration and d-Band Center in Homo-Diatomic Fe2N6Catalysts for Enhanced Peroxymonosulfate Activation

Abstract

The electronic coordination configuration of metal active sites and the reaction mechanism were investigated by constructing homo-diatomic Fe sites for visible-light-assisted heterogeneous peroxymonosulfate (PMS) activation. A novel Fe2N6catalyst was synthesized by selecting uniform pyridinic-N of graphitic carbon nitride (g-C3N4) as anchoring sites. The results demonstrated that homo-diatomic Fe sites modulated the d-band center and electron delocalization and thus enhanced the PMS activation kinetics (3.58 times vs single-atom Fe catalyst) with kobsof 0.111 min–1owing to the synergistic effect between adjacent Fe atoms. New Fe–Fe coordination significantly decreased the contribution of the antibonding state in the Fe–O bond due to the coupling of the Fe-3d orbitals, which facilitated the O–O bond cleavage of the Fe2–HOO–SO3complex with a reduced thermodynamic energy barrier of only −0.29 eV. This work provided comprehensive mechanistic insights into developing homo-diatomic catalysts governed by the coordination configuration and radical pathway for efficient heterogeneous PMS catalysis.

Abstract

This study investigated the changes of toxic compounds in coking wastewater with biological treatment (anaerobic reactor, anoxic reactor and aerobic-membrane bioreactor, A1/A2/O-MBR) and advanced physicochemical treatment (Fenton oxidation and activated carbon adsorption) stages. As the biological treatment stages preceding, the inhibition effect of coking wastewater on the luminescence of Vibrio qinghaiensis sp. Nov. Q67 decreased. Toxic units (TU) of coking wastewater were removed by A1/A2/O-MBR treatment process, however approximately 30 % TU remained in the biologically treated effluent. There is a tendency that fewer and fewer residual organic compounds could exert equal acute toxicity during the biological treatment stages. Activated carbon adsorption further removed toxic pollutants of biologically treated effluent but the Fenton effluent increased acute toxicity. The composition of coking wastewater during the treatment was evaluated using the three-dimensional fluorescence spectra, gas chromatography-mass spectrometry (GC-MS). The organic compounds with high polarity were the main cause of acute toxicity in the coking wastewater. Aromatic protein-like matters in the coking wastewater with low biodegradability and high toxicity contributed mostly to the remaining acute toxicity of the biologically treated effluents. Chlorine generated from the oxidation process was responsible for the acute toxicity increase after Fenton oxidation. Therefore, the incorporation of appropriate advanced physicochemical treatment process, e.g., activated carbon adsorption, should be implemented following biological treatment processes to meet the stricter discharge standards and be safer to the environment.

Abstract

Background: Oil pollution of water causes extensive damage to the environment and human health. Adsorption techniques are environmentally friendly and relatively feasible to treat oil contamination and reclaim oil resources, and it is essential to develop adsorbents that have high adsorption capacity for oil which can be recycled and regenerated conveniently to save money and resources. Results: Hydrophobic nonwoven fabrics coated with polydivinylbenzene nanofibres (HNF‐PDVB) were constructed by a one‐step cation polymerization reaction. The results showed that the optimal concentration of OTS (octadecylsilane) for the modification of nonwoven fibers was 1.22 mL L–1of n‐hexane. The HNF‐PDVB showed relatively good performance in adsorbing oil from water, with adsorption capacity as high as 7.5 g g–1. Freundlich and Dubinin–Radushkevic isotherms fitted well with the experimental data. The kinetic data fitted better to the pseudo‐second‐order and intraparticle diffusion models, and the parameters indicated that the adsorption by HNF‐PDVB involved macropores that contributed to the rapid and high‐capacity adsorption, as well as micropores that resulted in the diffusion of oil in the nanofibres. Thermodynamics studies revealed that the adsorption was spontaneous and endothermic, with an increasing performance when the temperature rose from 10 to 50 °C. The material possessed good stability and the adsorption capacity reduced from 7.5 g g–1 to 6.6 g g–1 after ten cycles of repeated use, recovered by squeezing and ethanol extraction.
