2. Genome-Wide Identification of NAC Gene Family Members of Tree Peony (Paeonia suffruticosa Andrews) and Their Expression under Heat and Waterlogging Stress.

Abstract

An important family of transcription factors (TFs) in plants known as NAC (NAM, ATAF1/2, and CUC2) is crucial for the responses of plants to environmental stressors. In this study, we mined the NAC TF family members of tree peony (Paeonia suffruticosa Andrews) from genome-wide data and analyzed their response to heat and waterlogging stresses in conjunction with transcriptome data. Based on tree peony's genomic information, a total of 48 PsNAC genes were discovered. Based on how similar their protein sequences were, these PsNAC genes were divided into 14 branches. While the gene structures and conserved protein motifs of the PsNAC genes within each branch were largely the same, the cis-acting elements in the promoter region varied significantly. Transcriptome data revealed the presence of five PsNAC genes (PsNAC06, PsNAC23, PsNAC38, PsNAC41, PsNAC47) and one PsNAC gene (PsNAC37) in response to heat and waterlogging stresses, respectively. qRT-PCR analysis reconfirmed the response of these five PsNAC genes to heat stress and one PsNAC gene to waterlogging stress. This study lays a foundation for the study of the functions and regulatory mechanisms of NAC TFs in tree peony. Meanwhile, the NAC TFs of tree peony in response to heat and waterlogging stress were excavated, which is of great significance for the selection and breeding of new tree peony varieties with strong heat and waterlogging tolerance. Abstract

In this study, the effects of salicylic acid to antioxidative activity and photosynthetic characteristics in waterlogging stress of two peony cultivars (‘Fengdanbai’ and ‘Mingxing’) were investigated. 4-year-old peony grown in different levels of waterlogging stress and then different concentration prepared SA (0.0, 0.1, 0.5 and 1.0 mmol L–1) sprayed on fresh leaves of peony. The antioxidative enzymes activities include superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), chlorophyll content, relative conductivity and MDA content were measured in leaves about different waterlogging treatment, the photosynthetic characteristics were also measured using photosynthetic measurement system. The results showed that waterlogging stress decreased the chlorophyll content in all peony cultivars leaves, but with SA treatment can inhibit the decrease of chlorophyll content. Relative conductivity increased as the extension of waterlogging time in two cultivars. SA treatment could effectively inhibit the increase of relative conductivity, and 0.5 mmol L–1 of SA was the most suitable concentration. SOD, POD, CAT activity increased first and then decreased in different waterlogging condition, SA significantly increased the activity of various enzymes. MDA content was increase as the expansion of waterlogging time in two cultivars. SA inhibits the increase of MDA content. Of all concentration of SA, 0.5 mmol L–1 was the best concentration to inhibit the waterlogging stress. For the photosynthetic characteristics, the net assimilation rate (Pn), stomatal conductance (Gs), transpiration rate (Tr) and intercellular CO2 (Ci) were decreased under different waterlogging condition. SA treatment can increase Pn, Gs, Tr and Ci of peony.

Abstract

Tree peony (Paeonia suffruticosa Andr.) is a perennial deciduous shrub with ornamental and medicinal value. Waterlogging stress is an agricultural problem for peony in Jiangnan of China. This study investigated the growth, biomass, cell membrane permeability and antioxidant enzymes of two P. suffruticosa cultivars ‘Feng Dan Bai (FDB)’ and ‘MingXing (MX)’. The response of roots and upper ground to different levels of waterlogging stress in two peony cultivars was also evaluated. Results showed that mild waterlogging stress (MWS) and severe waterlogging stress (SWS) significantly decreased the increment of seedling height, diameter and biomass of leaves. The root biomass increased in FDB but no significant changes in MX. Moreover, cell membrane permeability of leaves and roots also increased, while the chlorophyll content of leaves decreased. Antioxidant enzyme activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) of leaves and roots all increased, along with a gradual increase in malondialdehyde (MDA). Of this two cultivars, the root system of FDB is more susceptible to waterlogging than the upper ground, and the root system can improve the resistance to waterlogging by increasing the root system biomass, Peony adapted to the waterlogging environment by changing its external form. 26. NSs Filament Formation Is Important but Not Sufficient for RVFV Virulence In Vivo

Abstract

Rift Valley fever virus (RVFV) is a mosquito-borne phlebovirus that represents as a serious health threat to both domestic animals and humans. The viral protein NSs is the key virulence factor of RVFV, and has been proposed that NSs nuclear filament formation is critical for its virulence. However, the detailed mechanisms are currently unclear. Here, we generated a T7 RNA polymerase-driven RVFV reverse genetics system based on a strain imported into China (BJ01). Several NSs mutations (T1, T3 and T4) were introduced into the system for investigating the correlation between NSs filament formation and virulence in vivo. The NSs T1 mutant showed distinct NSs filament in the nuclei of infected cells, the T3 mutant diffusively localized in the cytoplasm and the T4 mutant showed fragmented nuclear filament formation. Infection of BALB/c mice with these NSs mutant viruses revealed that the in vivo virulence was severely compromised for all three NSs mutants, including the T1 mutant. This suggests that NSs filament formation is not directly correlated with RVFV virulence in vivo. Results from this study not only shed new light on the virulence mechanism of RVFV NSs but also provided tools for future in-depth investigations of RVFV pathogenesis and anti-RVFV drug screening.
