17. Activation of LONP1 by 84-B10 alleviates aristolochic acid nephropathy via re-establishing mitochondrial and peroxisomal homeostasis.

Abstract

Pharmaceutical formulations derived from Aristolochiaceae herbs, which contain aristolochic acids (AAs), are widely used for medicinal purposes. However, exposure to these plants and isolated AAs is linked to renal toxicity, known as AA nephropathy (AAN). Currently, the mechanisms underlying AAN are not fully understood, leading to unsatisfactory treatment strategies. In this study, we explored the protective role of 84-B10 (5-[[2-(4-methoxyphenoxy)-5-(trifluoromethyl) phenyl] amino]-5-oxo-3-phenylpentanoic acid) against AAN. RNA-seq analysis revealed that the mitochondrion and peroxisome were the most affected cellular components following 84-B10 treatment in AAN mice. Consistently, 84-B10 treatment preserved mitochondrial ultrastructure, restored mitochondrial respiration, enhanced the expression of key transporters (carnitine palmitoyltransferase 2) and enzymes (acyl-Coenzyme A dehydrogenase, medium chain) involved in mitochondrial fatty acid β -oxidation, and reduced mitochondrial ROS generation in both aristolochic acid I (AAI)-challenged mice kidneys and cultured proximal tubular epithelial cells. Additionally, 84-B10 treatment increased the expression of key transporters (ATP binding cassette subfamily D) and rate-limiting enzymes (acyl-CoA oxidase 1) involved in peroxisomal fatty acid β -oxidation and restored peroxisomal redox balance. Knocking down LONP1 expression diminished the protective effects of 84-B10 against AAN, suggesting LONP1-dependent protection. In conclusion, our study provides evidence that AAN is associated with significant disturbances in both mitochondrial and peroxisomal functions. The LONP1 activator 84-B10 demonstrates therapeutic potential against AAN, likely by maintaining homeostasis in both mitochondria and peroxisomes. [ABSTRACT FROM AUTHOR]

Abstract

The channel sand bodies in the lower section of the Sifangtai Formation in the Songliao Basin are rich in uranium resources. The development of channel sand body distribution prediction is of great significance for guiding the exploration of sandstone-type uranium deposits. In this study, the wave-indicated inversion technology was used to predict the thickness of the sand body, the root mean square amplitude attribute was used to identify the plane shape of the river channel, and the seismic facies analysis and tracking technology of the section was used to identify the channel period and finely describe the channel distribution. The results show that there are two stages of channel sand bodies in the lower part of Sifangtai Formation, and the sand bodies are distributed in a wide range. The maximum thickness of single stage sand bodies is about 15m, with an average of 8 meters. The thickness of the sand body where the two phases of the river are superimposed is the largest, up to 28m, which is a favorable ore-bearing location revealed by current exploration.

31. Melilite-derived mineral inclusions in chromite from the Gaositai complex: Implications for an extensional tectonic setting in Early Permian at the north North China Craton

Abstract

The Early Permian mafic-ultramafic concentrically zoned Gaositai intrusion at Chengde, on the northern margin of the North China Craton (NCC), is a cumulative complex emplaced along a giant fracture that penetrates deeply into the continental lithosphere. Melt inclusions are present in chromite crystals from the inner dunite and chromitite zones of the Gaositai complex. The melt inclusions have experienced post-trap crystallization and resulted in multiple mineral phases, including melilite, garnet, phlogopite, magnesite and apatite, which can indicate the liquidus minerals of the primitive magma. The characteristics of the melilite+melanite+clinopyxene assemblage indicate that the primary parental magma was highly undersaturated and derived from an alkali-rich mantle source. The crystallization of phlogopite, magnesite and apatite suggests a primary magma rich in K, H2O and CO2. When compared with experimental data, the primary magma of the Gaositai intrusion is concordant with a kamafugite magma originating from partial melting of enriched mantle with H2O and CO2at pressures greater than 2.7 GPa. This magmatic process would have been related to extensional thinning of the continental lithosphere. The Gaositai primary magmas have high Nb/La ratios, which are similar to those of ocean island basalts, but different from arc-related magmas. This suggests that the northern margin of the NCC was not an active continental margin of the Paleo-Asian Ocean subduction zone during the Early Permian: an extensional tectonic setting during the emplacement of the Gaositai intrusion is more likely.
