Abstract

Messenger RNA (mRNA) is the molecule which carries genetic information from DNA and guides the synthesis of protein. It participates in many cellular cascades and exhibits substantial associations with certain diseases. The development of advanced approaches for mRNA sensing and imaging is of utmost importance for biological studies and early clinical diagnosis. Herein, a novel fluorescent method is demonstrated for highly sensitive analysis of mRNA based on entropy‐driven strand displacements around three‐dimensional DNA nanostructures. Tetrahedral DNA with four pendant linear sequences provides excellent molecular scaffold loading at multiple sites for target mRNA‐initiated cascade toehold strand displacement reactions. The variations of fluorescence resonance energy transfer states can be used to indicate the presence and level of target mRNA. Besides, MnO2 nanosheets are introduced for intracellular transportation of the reaction system for bioimaging. The operation is relatively simple and the developed approach provides a promising tool for mRNA‐related biological researches and clinical applications. [ABSTRACT FROM AUTHOR]

Abstract

PurposeTo assess the feasibility of a new shape configuration involving a partially hollow and porous lower part for dental implants.Materials and methodsCylindrical zirconia dental implants coated with bioactive glass were fabricated in the laboratory. Each implant has a solid upper part and a partially hollow lower part. It is open at the bottom with holes through the lower cylindrical walls. This hollow and porous configuration permits bone growth into the lower part of the implant that, over time, forms an interlinked network to lock the implant into the alveolar bone. Biomechanical properties of the new design were evaluated through material testing and experiments with dogs.ResultsMechanical testing of bending strength, hardness, fracture toughness, and fatigue life indicated that zirconia implants with the proposed partially hollow configuration can be fabricated to have structural properties comparable to or exceeding the usual requirements for implants. Animal testing suggests that there is appreciable improvement in lock-in strength and osteointegration due to the hollow and porous configuration.ConclusionThe new shape configuration is biomechanically feasible and further research is warranted to improve the design for human use.

Abstract

Objectives: Hemifacial microsomia (HFM) is a common birth defect involving the first and second branchial arch derivatives. Although several chromosomal abnormalities and causal gene variants have been identified, genetic etiologies in a majority of cases with HFM remain unknown. This study aimed to identify genetic mutations in affected individuals with HFM. Methods: Whole‐exome sequencing and bioinformatics analysis were performed for 16 affected individuals and their family members. Sanger sequencing was applied for confirmation of selected mutations. Zebrafish embryos were used for in situ hybridization of candidate gene, microinjection with antisense morpholino, and cartilage staining. Results: A homozygous missense mutation (c.484G > A; p.V162I) in the FRK gene was identified in an 18‐year‐old girl with HFM and dental abnormalities. Heterozygous mutation of this mutation was identified in her parents, who are first cousins in a consanguineous family. FRK is highly expressed in the Meckel's cartilage during embryonic development in mouse and zebrafish. Knockdown of frk in zebrafish showed a lower length and width ratio of Meckel's cartilage, abnormal mandibular jaw joint, and disorganized ceratobranchial cartilage and bone. Conclusions: We identified a recessive variant in the FRK gene as a novel candidate gene for a patient with HFM and mandibular hypoplasia and revealed its effects on craniofacial and embryonic development in zebrafish. Abstract

Background and importance: Scheuermann’s disease (SD), intradural-extramedullary epidermoid cysts and idiopathic scoliosis (IS) are all rare conditions. The co-occurrence of the three condition on one case has not been reported in literature before. We describe an extremely rare case of SD co-occurrence with IS and intradural-extramedullary epidermoid cyst, which is the first in the literature. Clinical presentation: A 13-year old boy and his parents presented at Honghui Hospital complaining of progressive hyperkyphosis for 5 years and back pain for 2 months. Thorough medical examination and radiology confirmed the diagnosis of SD kyphosis, IS, and intradural-extramedullary epidermoid cyst. Medical panel with spine surgeons, oncologists, neurosurgeons, histopathologists and radiologists were consulted. Tumor excise, kyphosis recorrection and spine reconstruction surgery was conducted. Postoperative histological examination detected desquamation of keratin from the epithelial lining and stratified squamous epithelium without skin adnexa. The boy was ambulant 2 days postoperative with walking aids. Conclusion: This is the first case report of Scheuermann’s disease (SD) co-occurrence with idiopathic scoliosis (IS) and intradural-extramedullary epidermoid cyst. The co-occurrence of intradural epidermoid cyst, SD and IS might not be a coincidence, indicating a kind of genetic etiology need to be illuminated.

Abstract

BACKGROUND The purpose of this study was to investigate whether Orai1 plays a role in the metastasis of osteosarcoma. MATERIAL AND METHODS The expression of Orai1 was silenced by small interfering RNAs against Orai1 (Orai1 siRNA) in osteosarcoma MG-63 cells. Various experiments were carried out to detect the changes in migration, invasion, and adhesion ability of these osteosarcoma cells. Furthermore, the activity of Rac1, Wave2, and Ras was detected using Western blot analysis. Moreover, the Rac1 and Ras inhibitors were used to confirm whether the Ras-Rac1-WAVE2 signaling pathway was involved in osteosarcoma metastasis promoted by Orai1.
