Abstract

Aging is characterized by a progressive loss of physiological integrity, while cancer represents one of the primary pathological factors that severely threaten human lifespan and healthspan. In clinical oncology, drug resistance limits the efficacy of most anticancer treatments, and identification of major mechanisms remains a key to solve this challenging issue. Here, we highlight the multifaceted senescence-associated secretory phenotype (SASP), which comprises numerous soluble factors including amphiregulin (AREG). Production of AREG is triggered by DNA damage to stromal cells, which passively enter senescence in the tumor microenvironment (TME), a process that remarkably enhances cancer malignancy including acquired resistance mediated by EGFR. Furthermore, paracrine AREG induces programmed cell death 1 ligand (PD-L1) expression in recipient cancer cells and creates an immunosuppressive TME via immune checkpoint activation against cytotoxic lymphocytes. Targeting AREG not only minimized chemoresistance of cancer cells, but also restored immunocompetency when combined with classical chemotherapy in humanized animals. Our study underscores the potential of in vivo SASP in driving the TME-mediated drug resistance and shaping an immunosuppressive niche, and provides the proof of principle of targeting major SASP factors to improve therapeutic outcome in cancer medicine, the success of which can substantially reduce aging-related morbidity and mortality.

Abstract

Chemotherapy and radiation not only trigger cancer cell apoptosis but also damage stromal cells in the tumour microenvironment (TME), inducing a senescence-associated secretory phenotype (SASP) characterized by chronic secretion of diverse soluble factors. Here we report serine protease inhibitor Kazal type I (SPINK1), a SASP factor produced in human stromal cells after genotoxic treatment. DNA damage causes SPINK1 expression by engaging NF-κB and C/EBP, while paracrine SPINK1 promotes cancer cell aggressiveness particularly chemoresistance. Strikingly, SPINK1 reprograms the expression profile of cancer cells, causing prominent epithelial-endothelial transition (EET), a phenotypic switch mediated by EGFR signaling but hitherto rarely reported for a SASP factor. In vivo, SPINK1 is expressed in the stroma of solid tumours and is routinely detectable in peripheral blood of cancer patients after chemotherapy. Our study substantiates SPINK1 as both a targetable SASP factor and a novel noninvasive biomarker of therapeutically damaged TME for disease control and clinical surveillance.

Abstract

Objective To investigate the voice handicap index of teachers and its correlation with their work and lifestyle habits, and provide a basis for guiding the prevention and treatment of their voice disorders. Methods In 2020,a total of 494 teachers in 9 educational institutions in Shenzhen City were surveyed with a structured questionnaire to collect information on their work and personal lives and the Voice Handicap Index-10 (VHI) Scale to assess the voice disorder situation. A multifactorial unconditional logistic regression model was used to analyze the influencing factors of voice disorders. Results With a response rate of 91.30%, 451 complete and valid surveys were returned. The median voice handicap index of the surveyed teachers was 9, with a quartile range of (4, 13), and 167 teachers had voice disorders, accounting for 37.03%. Multivariate regression analysis showed that poor sleep quality, the occurrence of emotional distress within the past month, and a large class size were risk factors for voice disorders. For each decrease in sleep quality level (very good, good, poor), each increase in emotional distress severity level (none, mild, moderate, severe), and each increase in the scale of class size (≤ 30, 31-40, 41-50, and ≥ 51 students), the risk of voice disorders among teachers increased by 1.78, 1.90, and 1.58 times, respectively (P < 0.05). Conclusions Some teachers in educational institutions in Shenzhen had voice disorders. To improve their vocal health, it is recommended to organize teaching tasks reasonably, pay attention to mental health conditions and sleep quality, and promote healthy vocal practice. Abstract

Purpose Laser melting deposition (LMD) is an advanced additive manufacturing (AM) technology without powder waste, and nickel-based alloys with different Nb contents were created one-time by adjusting the ratio of mixed powders via a dual-feed system. Here, the authors provide a systematic report on the effects of the Nb content on the microstructure, Laves phase segregation and mechanical properties of as-received LMD nickel-based alloys. The effects of the Nb content on the microstructure, precipitation evolution and mechanical properties of the subsequent heat-treated LMD samples are also discussed in this paper. Design/methodology/approach Thus, the present research aims to obtain a better understanding of the effect of Nb content on the microstructural and mechanical properties of the as-received LMD Inconel 718 alloys through high-throughput sample fabrication. The microstructures were characterized by scanning electron microscopy and energy-dispersive spectroscopy, electron back-scattered diffraction and transmission electron microscopy methods. The mechanical properties were obtained from compressive tests and nano-indentation tests. Electrochemical tests, including electrochemical impedance spectroscopy and potentiodynamic polarizations, were carried out to evaluate the durability of the Inconel 718 alloys. Results can provide a factual basis for future applications of the functionally graded by AM technology. Findings The grain size of the as-received LMD Inconel 718 alloys decreased with the Nb content. The Laves phase distribution at the macro level was relatively uniform and the Laves phase exhibited a 1.5-fold nano-hardness compared with the matrix. The strength improvement for the as-received LMD Inconel 718 alloys with Nb content was attributed to grain refinement and enhancement of the Laves phase in terms of both hardness and content. Meanwhile, the corrosion resistance increased with the increase of the Nb content, especially for the pitting potential, which was attributed to the optimization of carbide precipitates due to the strong affinity between niobium and carbon. Originality/value The results provide a factual basis for the Nb content effect in LMD nickel-based alloys, and this method can greatly promote the development of new materials. The authors believe that this study makes a significant contribution to the literature and is suitable for publication.

Abstract

The corrosion behavior and mechanical properties of 316L stainless steel (SS) fabricated via selective laser melting (SLM) were clarified by potentiodynamic polarization measurements, immersion tests, and tensile experiments. The microstructural anisotropy of SLMed 316L SS was also investigated by electron back-scattered diffraction and transmission electron microscopy. The grain sizes of the SLMed 316L SS in the XOZ plane were smaller than those of the SLMed 316L SS in the XOY plane, and a greater number of low-angle boundaries were present in the XOY plane, resulting in lower elongation for the XOY plane than for the XOZ plane. The SLMed 316L was expected to exhibit higher strength but lower ductility than the wrought 316L, which was attributed to the high density of dislocations. The pitting potentials of the SLMed 316L samples were universally higher than those of the wrought sample in chloride solutions because of the annihilation of MnS or (Ca,Al)-oxides during the rapid solidification. However, the molten pool boundaries preferentially dissolved in aggressive solutions and the damage of the SLMed 316L in FeCl3 solution was more serious after long-term service, indicating poor durability.

Abstract

This work discussed the effects of different scanning speeds (800, 1083, 1200 and 1400 mm/s) on the microstructure of 316L stainless steel manufactured by selective laser melting (SLM) and the related corrosion behavior. Results showed that there were more voids with faster scanning speeds, and there were oxide powder and non-melt silicon inside the defects. The SLM 316L exhibited a full gamma austenite phase filled with sub-grains, and the average grain size of SLM-1083 mm/s 316L was approximately 42 μm, three times larger than that of the quenched 316L. The pitting potentials for the SLM 316L were all approximately 300 mV higher than that of the quenched due to the modification of inclusions in SLM, but the corrosion rate for the SLM 316L was faster, which was attributed to the voids and unstable passive film. The number of pitting sites increased with the scanning speed, and the pits occurred preferentially at the voids in SLM 316L.
