Abstract Background The mature mouse oocyte contains the full complement of maternal proteins required for fertilization, reprogramming, zygotic gene activation (ZGA), and the early stages of embryogenesis. However, due to limitations of traditional proteomics strategies, only a few abundantly expressed proteins have yet been identified. Our laboratory applied a more effective strategy: one-dimensional sodium dodecyl sulfate polyacrylamide gel electrophoresis (1D SDS-PAGE) and reverse-phase liquid chromatography tandem mass spectrometry (RP-LC-MS/MS) were employed to analyze the mature oocyte proteome in depth. Results Using this high-performance proteomic approach, we successfully identified 625 different proteins from 2700 mature mouse oocytes lacking zona pellucidae. This is the largest catalog of mature mouse oocyte proteins compiled to date. According to their pattern of expression, we screened 76 maternal proteins with high levels of mRNA expression both in oocytes and fertilized eggs. Many well-known maternal effect proteins were included in this subset, including MATER and NPM2. In addition, our mouse oocyte proteome was compared with a recently published mouse embryonic stem cell (ESC) proteome and 371 overlapping proteins were identified. Conclusion This proteomics analysis will be a valuable resource to aid in the characterization of important maternal proteins involved in oogenesis, fertilization, early embryonic development and in revealing their mechanisms of action.

Abstract Background MicroRNAs (miRNAs), which are small, non-coding RNAs approximately 21-nucleotides in length, have become a major focus of research in molecular biology. Mammalian miRNAs are proposed to regulate approximately 30% of all protein-coding genes. Previous studies have focused on highly conserved miRNAs, but nonconserved miRNAs represent a potentially important source of novel functionalities during evolution. Results An analysis of the chromosome distribution of miRNAs showed higher densities of miRNAs on the X chromosome compared to the average densities on autosomes in all eight mammalian species analyzed. The distribution pattern did not, however, apply well to species beyond mammals. In addition, by comparing orthologous human and mouse miRNAs, we found that X-linked miRNAs had higher substitution rates than autosomal miRNAs. Since the highest proportion of X-linked miRNAs were found in mouse testis, we tested the hypothesis that testis miRNAs are evolving faster on the X chromosome than on autosomes. Mature X-linked testis miRNAs had an average substitution rate between mouse and human that was almost 25-fold higher than mature testis miRNAs on autosomes. In contrast, for mature miRNAs with precursors not expressed in testis, no significant difference in the substitution rate between the X chromosome and autosomes was found. Among mammals, the rapid evolution of X-linked testis miRNAs was also observed in rodents and primates. Conclusion The rapid evolution of X-linked testis miRNAs implies possible important male reproductive functions and may contribute to speciation in mammals.

Whole‐rock major‐trace elemental, whole‐rock Sr–Nd, and zircon U–Pb–Hf isotopic analyses have been carried out on two suites of Early Cretaceous granitic dykes in the western Dabie Orogen (Central China) with the aim of addressing their petrogenesis and geodynamic significance. LA‐ICPMS zircon U–Pb dating reveals that the E‐W‐trending granitic dykes have emplacement ages of 133.9 ± 1.6 and 132.8 ± 1.0 Ma, while the NW‐trending granitic dykes have emplacement ages of 121.4 ± 0.9 and 120.5 ± 0.7 Ma. These rocks have high concentrations of SiO2 and K2O, belonging to the high‐K calc‐alkaline and shoshonitic series. They are metaluminous to peraluminous and are mainly fractionated I‐type granite. The E‐W‐trending granites have moderate initial 87Sr/86Sr ratios (0.70614 to 0.70878) and negative εNd(t) values varying from −16.2 to −17.2 with two‐stage Nd model ages of 2.20 to 2.43 Ga. They exhibit a narrow range of zircon εHf(t) values (−22.34 to −23.70) with two‐stage Hf model ages of 2.31 to 2.36 Ga. These isotopic signatures suggest a homogeneous crustal source without significant magma contamination or mixing. The NW‐trending granites have initial 87Sr/86Sr ratios ranging from 0.70774 to 0.71034 and negative εNd(t) values varying from −10.9 to −9.5 with two‐stage Nd model ages of 1.69 to 1.80 Ga. They yield variable zircon εHf(t) values of −17.06 to −0.39 with two‐stage Hf model ages of 1.07 to 1.99 Ga, implying heterogeneous crustal signatures with minor mantle materials involved. Combining the geological, geochemical, and isotopic evidences, the E‐W‐trending granitic dykes are considered to be derived from partial melting of crustal materials under normal thickness during a transitional period following the tectonic collapse, while the NW‐trending granitic dykes resulted from crust melting in an extensional setting, most likely induced by the asthenospheric upwelling. Our results support that the western Dabie Orogen could have experienced S‐N‐directed compression (pre. 133 Ma), S‐N‐directed extension (ca. 133 Ma), and general E‐W‐directed extension (ca. 120 Ma). We propose that the Dabie Orogen has experienced tectonic collapse of the thickened lower crust and also has been affected by the subduction of the Izanagi (or Palaeo‐Pacific) Plate during the Early Cretaceous.
