4. Effect of in Vitro Gastrointestinal Environment on Astxanthin-Loaded Pickering High Internal Phase Emulsions Stabilized by Silkworm Pupa Protein

Abstract

In this study, the stability of Pickering high internal phase emulsions (HIPEs) stabilized by silkworm pupa protein in an in vitro gastrointestinal environment and their astaxanthin loading capacity as well as the bioaccessibility of astaxanthin-loaded HIPEs were characterized, and the mechanism of the targeted and controlled release of astaxanthin-loaded HIPEs in the simulated gastrointestinal environment was elucidated by measuring the particle size distribution and zeta potential as well as using confocal laser scanning microscopy (CLSM), reversed-phase high performance liquid chromatography (RP-HPLC) and electrophoresis. The results showed that silkworm pupa protein stabilize effectively stabilized Pickering HIPEs with an internal phase as 78%, and the encapsulation efficiency of AST in the HIPEs was nearly 88%. The Pickering HIPEs remained stable in the simulated gastric environment for 90 min and became progressively unstable with prolonged digestion in the simulated intestinal environment, significantly increasing the bioaceessibility of astaxanthin. The interfacial protein composition of the emulsion indicated that silkworm pupa protein, with a molecular mass of 70 kDa, was the major protein stabilizing the Pickering HIPEs, and its hydrolysis by the intestinal digestive enzymes was the major cause of the destabilization of the emulsion and the release of astaxanthin. The present study provides a theoretical basis for the application of Pickering HIPEs stabilized by silkworm pupa protein for targeted intestinal delivery of hydrophobic bioactive substances, and opens up new possibilities for the application of astaxanthin in the food and nutraceutical fields.

9. Ag Nanoparticles on MXene Nanosheets for Combined Ionic and Photothermal Therapy of Bacterial Infections.

Abstract

In view of the increasing bacterial resistance, 2D MXenes are promising alternatives to antibiotics. However, MXene-based photothermal therapy (PTT) suffers from unsatisfactory antibacterial efficiency and heat-resistant strains. Here, we prepared a Ti3C2 MXene and Ag hybridized antibacterial nanocomposite [MXene/metal-polyphenol networks (MPNs)/Ag] through the in situ reduction of Ag nanoparticles on MPN wrapped MXene matrix. The use of MPNs as the reducing agents of Ag+ and anchoring agents of Ag nanoparticles endowed MXene/MPN/Ag with a tight immobilization capacity and improved colloidal dispersion stability of Ag nanoparticles. The pH-triggered decomposition of MPNs led to the pH-responsive release of Ag to achieve combined MXene-based PTT and Ag-mediated therapy for enhanced antibacterial efficiency. In vitro antibacterial experiments revealed its satisfactory bactericidal activities against both planktonic bacteria and bacteria in stubborn biofilms. In vivo antibacterial assays solidly confirmed its high antibacterial therapeutic efficiency, strong anti-inflammatory ability, and good biosafety. Therefore, the in situ combination of Ag nanoparticles with MXenes offers a promising microenvironment-responsive 2D bactericidal candidate for infection that could be applied in future antibacterial treatments. Abstract

Human pregnane X receptor (PXR) is critical for regulating the expression of key drug‐metabolizing enzymes such as CYP3A and CYP2C. Our recent study revealed that treatment with rodent‐specific PXR agonist pregnenolone‐16α‐carbonitrile (PCN) significantly induced hepatomegaly and promoted liver regeneration after two‐thirds partial hepatectomy (PHx) in mice. However, it remains unclear whether PXR activation induces hepatomegaly and liver regeneration and simultaneously promotes metabolic function of the liver. Here, we investigated the metabolism activity of CYP1A2, CYP3A1/2 and CYP2C6/11 during PXR activation‐induced liver enlargement and regeneration in rats after cocktail dosing of CYP probe drugs. For PCN‐induced hepatomegaly, a notable increase in the metabolic activity of CYP3A1/2 and CYP2C6/11, as evidenced by the plasma exposure of probe substrates and the AUC ratios of the characteristic metabolites to its corresponding probe substrates. The metabolic activity of CYP1A2, CYP3A1/2 and CYP2C6/11 decreased significantly after PHx. However, PCN treatment obviously enhanced the metabolic activity of CYP2C6/11 and CYP3A1/2 in PHx rats. Furthermore, the protein expression levels of CYP3A1/2 and CYP2C6/11 in liver were up‐regulated. Taken together, this study demonstrates that PXR activation not only induces hepatomegaly and liver regeneration in rats, but also promotes the protein expression and metabolic activity of the PXR downstream metabolizing enzymes such as CYP3A1/2 and CYP2C6/11 in the body. 17. Gut Microbiota Affects Mouse Pregnane X Receptor Agonist Pregnenolone 16α-Carbonitrile-Induced Hepatomegaly by Regulating Pregnane X Receptor and Yes-Associated Protein Activation

Abstract

Pregnane X receptor (PXR) is essential in the regulation of liver homeostasis, and the gut microbiota is closely linked to liver physiologic and pathologic status. We previously found that activation of PXR significantly promotes liver enlargement through interaction with yes-associated protein (YAP). However, whether gut microbiota contributes to PXR-induced hepatomegaly and the involved mechanisms remain unclear. In this study, C57BL/6 mice were administered the mouse-specific agonist pregnenolone 16α-carbonitrile (PCN) for 5 days. Depletion of gut microbiota was achieved using broad-spectrum antibiotics (ABX) and fecal microbiota transplantation (FMT) was performed to restore the gut microbia. The composition of gut microbiota was analyzed by 16S rRNA sequencing, while the expression of PXR, YAP, and their downstream target genes and proteins were assessed. The results indicated that PCN treatment altered the composition and abundance of specific bacterial taxa. Furthermore, depletion of gut microbiota using ABX significantly attenuated PCN-induced hepatomegaly. FMT experiments further demonstrated that the fecal microbiota from PCN-treated mice could induce liver enlargement. Mechanistic studies revealed that ABX treatment impeded the PXR and YAP activation induced by PCN, as evidenced by decreased expression of PXR, YAP, and their downstream targets. Moreover, alterations in PXR and YAP activation were likely contributing to hepatomegaly in recipient mice following FMT from PCN-treated mice. Collectively, the current study demonstrated that gut microbiota is involved in PCN-induced hepatomegaly via regulating PXR and YAP activation, providing potential novel insights into the involvement of gut microbiota in PXR-mediated hepatomegaly.SIGNIFICANCE STATEMENTThis work describes that the composition of gut microbiota is altered in mouse pregnane X receptor (PXR) agonist pregnenolone 16α-carbonitrile (PCN)-induced hepatomegaly. Treatment with an antibiotic cocktail depletes the intestinal microbiota, leading to the impairment of liver enlargement caused by PCN. Additionally, fecal microbiota transplantation from PCN-treated mice induces liver enlargement. Further study revealed that gut microbiota is involved in hepatomegaly via regulating PXR and yes-associated protein activation.

20. Traditional village digital archival conservation: a case study from Gaoqian, China.

Abstract

Archiving in traditional villages has been carried out for nearly 20 years in China. The conventional practice is to establish registration records for the basic information and cultural heritage of traditional villages combined with collecting historical records. This research develops a digital archival model to strengthen traditional village conservation. The theoretical framework of this model arises from a case study of Gaoqian, a typical traditional Chinese village with 1000 years of history. Through field investigation and document analysis, Gaoqian's experience is summarized into a paradigm of traditional village digital archival conservation. There are three modules in this paradigm: a digital archival resource system, a digital archival management system, and a digital archival service system adhering to the culture-centred principle. This model transforms the initial cultural resources to digital archival resources and then to digital cultural products through strict life-cycle management. The whole working process needs the cooperation and participation of multiple subjects. The case of Gaoqian demonstrates that traditional village digital archival conservation is simultaneously universal and scene-oriented, which can provide replicable solutions for traditional villages or rural landscape conservation in other regions during ongoing world urbanization, with the appropriate archival perspective and methods. Abstract

The paper analyzes the open data action framework of local governments in China from the policy level, aiming to clarify its practical progress and optimization direction. Based on the elements of government open data derived from the literature, the paper clusters and analyzes the policies of regions above sub-provincial level, and summarizes the primary contents of China's government open data action framework from the aspects of open management system, data opening process and data opening guarantee. The study shows that systematization of the action framework revealed by the policies is limited, based on the limitation, the directions of optimization are proposed: to strengthen the connotation of openness for social utilization, to establish a multi-body synergistic mechanism with internal and external governmental linkage, and to guarantee the systematic expansion of elements by institutional optimization as the leader. Abstract

The type-II InAs/GaSb superlattices (T2SLs)-based 640 × 512 long wavelength infrared (LWIR) Focal Plane Array (FPA) detector with15 μm pitch and 50% cut-off wavelength of 10.5 μm demonstrates a peak quantum efficiency of 38.6% and peak detectivity of 1.65 × 1011 cm Hz1/2 W−1 at 8.1 μm, high pixel operability of 99.5% and low responsivity non-uniformity of 2.69% at 80 K. The FPA exhibits clear infrared imaging at 110 K and diffusion-limited dark current densities below Tennant’s ‘Rule07’ at temperature above 100 K, which is attributed to the efficient suppression of diffusion dark current and surface leak current by introducing M-structure barrier and double hetero-structure passivation layers.

Abstract

For type-II InAs/GaSb superlattice (T2SL) photodetectors with deep and vertical mesa sidewalls, passivation for suppression of surface leakage currents is facing two key challenges that in the conformal coating of sharp sidewalls and increasing dominance of surface leakage currents as device dimensions shrink. In this work, atomic layer deposited (ALD)-Al2O3 and SiO2 method was employed as conformal passivation for deep and vertical mesa sidewalls. And a stable and high-quality interface between the ALD-Al2O3 and T2SL mesa sidewalls was obtained by the combined pretreatment of trimethylaluminum (TMA) and H2 plasma exposure (Al2O3TH), and thus a significant improvement in electrical performances has been achieved. The electrical performances before and after baking were compared among the Al2O3TH, conventional sulfur plus SiO2 passivated photodiodes. The dark current density of Al2O3TH passivated photodiodes were improved by 81% at 77 K under 0.1 V reverse bias and exhibited no deterioration after 90 °C-4 days baking treatment. Investigation on the effects of TMA and/or H2 plasma pretreatments demonstrates that combined pretreatment of TMA and H2 plasma, rather than TMA or H2 plasma alone, prior to the deposition of ALD-Al2O3 leads to a more thorough elimination of interface native oxides and surface states introduced in mesa process. The innovative application of this effective surface pretreatment technique is very encouraging for the passivation of deep and vertical sidewalls in III-V compound-based devices, such as large-format T2SL infrared detectors.

Abstract

This paper deals with the collaborative construction of private-natured yet social-oriented family archives by families, state archives and community management agencies. The construction of these archives has fostered an important practice model for national memory construction in China and can be used to build a base for education on patriotism and cultivation of fine family traditions. We conducted a case study on the Chen Yao Family Archives, the first social-oriented family archive in Sichuan Province. In this article we explore the methods of multi-actor collaborative construction and the specific contributions of different actors in terms of motives, contents and procedures. We also propose strategies for achieving successful construction of social-oriented family archives in terms of archival resources, multi-actor collaboration and utilization mechanisms. Abstract

Supplemental Digital Content is available in the text, Patients with cirrhosis are vulnerable to acute hepatic insults and are more likely to develop rapid hepatic deterioration. The aim of this study is to describe the clinical course of patients with cirrhosis and hospitalized for acute hepatic deterioration (AHD). This is a prospective study involving 163 patients with cirrhosis and AHD. The occurrence of organ failures, systemic inflammatory response syndrome (SIRS), and infections during hospital stay were recorded and the relationship between organ failure and death or SIRS/infection was subsequently analyzed. Of 163 patients, 35 did not develop any organ failure during in-hospital follow-ups (90-day mortality: 0%); 84 had intrahepatic organ failures (IH-OFs, defined by liver and/or coagulation failure) (90-day mortality: 22.0%); and 44 patients developed extra-hepatic organ failures (EH-OFs, defined by kidney, cerebral, circulation, and respiratory failure) on the basis of IH-OF with a 90-day mortality of 90.9%. On multivariable analysis by a Cox proportion hazard model, age, WBC, presence of IH-OF, and EH-OF all predicted 90-day death. A logistic regression analysis identified SIRS being associated with the development of EH-OF. Furthermore, IH-OF at admission and infections occurred during the hospital stay were shown to be another 2 potential risk factors. The clinical course of cirrhosis patients with acute hepatic injury was characterized by 3 consecutive stages (AHD, IH-OF, and EH-OF), which provided a clear risk stratification. The PIRO criteria provided an accurate frame for prognostication of those patients. The systemic inflammatory response syndrome may be a target for blocking the progression to the EH-OF stage.

Abstract

Objective: This study is to establish the rhesus monkey model of lymphedema in the upper limbs, and assess the suitability of this model. Methods: An animal model of lymphedema was established by the combined irradiation and surgical techniques in the upper limbs of these rhesus monkeys. Physical examination, high-resolution MR lymphangiography, bioelectrical impedance analysis (BIA), and immunohistochemical staining were performed to determine the severity of the edema in the upper limbs of the animal model. Results: Our results from physical examination indicated that the rhesus monkey model present with typical appearance and features of lymphedema. MR lymphangiography further demonstrated pathologically modified lymphatic vessels in our rhesus monkey model. BIA revealed increased water content in the upper limb in these rhesus monkeys, which was in line with the pathology of lymphedema. Immunohistochemical staining showed the curvature of the lymphatic vessels in the rhesus monkey model, typical pathological changes in lymphedema. Conclusion: Rhesus monkey lymphedema model provides a more consistent background to elucidate the pathophysiology of the disease. This new model would help to increase our understanding of acquired upper limb lymphedema, and promote the development of new treatments for this intractable disorder.
