Abstract

Curcumin has been widely used as a food additive for centuries and has been recently explored for its anti-inflammatory and antitumor properties. Although curcumin is pharmacologically safe and efficacious to certain cancers, its role against acute myeloid leukemia (AML) still remains unclear, and it lacks clinical application due to low water solubility and low in vivobioavailability. To address these issues, we developed a novel curcumin liposome modified with hyaluronan (HA-Cur-LPs) to specifically deliver curcumin to AML by targeting CD44 on AML cell surface. When compared with free curcumin and nontargeted liposome (Cur-LPs), the HA-Cur-LPs exhibited good stability, high affinity to CD44, increased cellular uptake, and more potent activity on inhibiting AML cell proliferation. The KG-1 cell implanted AML mice had significantly delayed, or even prevented, AML progression following treatment with 50 mg/kg of curcumin dose in the HA-Cur-LPs every 2 days for 2 weeks. Mechanistically, the anti-AML effects of HA-Cur-LPs were achieved by inhibiting Akt/ERK pathways and activating caspase-dependent apoptosis. Moreover, HA-Cur-LPs played a critical role in downregulation of DNMT1 expression in AML, leading to DNA hypomethylation and reactivation of tumor suppressor genes such as miR-223. The development and assessment of the HA-Cur-LPs in this study provide another potential choice for AML therapy, using HA-Cur-LPs as either a single treatment agent or in combination with other treatments.

42. Stray light and its reduction of focal plane arrays

Abstract

Existing studies have paid much attention to stray light of lenses. However, the ghost reflection inside FPA (Focal Plane Array) detector is almost ignored which is, in fact, also an unnegligible source of stray light in digital imaging systems. Ghosting between window surfaces and CCD photosensitive area is often a major source of stray light in FPA detectors, it may lead to image blur, color distortion, and contrast reduction. Besides, diffraction pattern can be observed from front-illuminated CCD as the incident beam can be diffracted by its polysilicon electrode gates at the surface. In this paper, we study the generation mechanism and the reduction approaches to such stray light. Both front-illuminated and back-illuminated CCDs (Charge Coupled Device) are investigated in our study. We build models to identify stray light paths and predict window ghost image characteristics. Furthermore, three methods, i.e., anti-reflective coating method, fluid-filled method and deconvolution method are presented to decrease the stray light. The first anti-reflective coating method can be effectively used in back-thinned CCDs to reduce the reflection and to maximize quantum efficiency. The second fluid-filled method attempts to reduce the effects of unwanted light contamination by simulating some characteristics of the human eyes. The use of liquid can reduce the fresnel reflectance of interface. In addition, the acquired images contaminated by FPA's stray light are post-processed with the deconvolution method. Effectiveness of our proposed methods is verified with experiments. It is shown that stray light of FPA can be efficiently reduced.

Abstract

Distant metastasis is a major contributor to cancer-related mortality. However, the role of circRNAs in this process remains unclear. Herein, we profiled the circRNA expression in a cohort of 68 colorectal carcinoma (CRC) primary tumors and their paired liver metastatic lesions. By overlapping with the TGFβ-responsive circRNAs, circNEIL3 (hsa_circ_0001460) was identified as a TGFβ-repressive and metastasis-related circRNA. Functionally, circNEIL3 effectively inhibited tumor metastasis in both and in vivo and in vivo models of various cancer types. Mechanistically, circNEIL3 exerts its metastasis-repressive function through its direct interaction with oncogenic protein, Y-box-binding protein 1 (YBX1), which consequently promotes the Nedd4L-mediated proteasomal degradation of YBX1. Importantly, circNEIL3 expression was negatively correlated to YBX1 protein level and metastatic tendency in CRC patient samples. Collectively, our findings indicate the YBX1-dependent antimetastatic function of circNEIL3 and highlight the potential of circNEIL3 as a biomarker and therapeutic option in cancer treatment.

Abstract

Objective: To analyze the mutation patters of HBV P gene and genotyping of heptitis B virus in patients with chronic hepatitis B (CHB) after the emergence of drug-resistance during lamivudine (LAM) therapy., Methods: LAM-resistant mutations and genotype of HBV were dectected in patients with LAM-resistant CHB in our hospital from Sep. 2008 to June. 2010., Result: 107 patients had 8 mutation patterns. YMDD mutations happened in 100%, only YMDD mutation were 43 patients, while others were YMDD combined with rtL180M mutations; the HBV genotype among 107chronic hepatitis was mainly B (25.2%) and C (73.8%) and only 1 patient was happend B and C mixed infection. Genetype C was mainly YMDD combined with rtL180M mutations pattern, the rate is 60.7% (48/79); while genetype B was mainly rtM204 mutation pattern, the rate is 66.7% (18/27); there were significant difference between the genetype B and C in mutation pattern (chi2 = 8.4, 7.2, respectively, P < 0. 01)., Conclusion: YMDD mutation is the major mutation pattern of HBV P gene after emergence of LAM-resistance. Genotypes of hepatitis B virus can determine the related mulation patterns of HBV P gene.

Abstract

Objective: To explore the alone and joint diagnostic value of serum golgi protein 73 (GP73), alpha-fetoprotein (AFP) and the percentage of lectin-reactive aipha-fetoprotein (AFP-L3) of primary hepatic carcinoma (PHC), and provide a novel method for diagnosis for PHC and screening for high-risk population., Methods: ELISA was used to detect the serum level of GP73, AFP and AFP-L3% in 81 cases of PHC,176 cases chronic hepatitis and liver cirrhosis, 30 cases other tumber cancer and 40 cases of health people., Results: The sensitivity of GP73, AFP and AFP-L3% in PHC is 77.78%, 62.69% and 51.85%, and the specificity is 84.55%, 86.99% and 96.34%, respectively. Joint detection could increase the sensitivity up to 88.89%., Conclusion: GP73 was a high sensitivity mark for dignosis of PHC, while AFP-L3% was a high specificity mark for dignosis of PHC. The joint detection could improve PHC diagnostic performance.
