11. Research progress of m6A methylation modification in regulating tumor immunity

Abstract

N6-methyladenosine (m6A) is the most prevalent modification that regulates gene expression in eukaryotes. It regulates splicing, degradation, stability, and translation of RNA. Numerous studies have demonstrated the close association between m6A methylation and tumor development, highlighting its crucial role in regulating tumor immune response. The m6A modification actively participates in governing immune cell differentiation and maturation as well as modulating anti-tumor immune responses. Within the tumor microenvironment, m6A modification can also impact the recruitment, activation, and polarization of immune cells, thereby either promoting or inhibiting tumor cell proliferation and metastasis. Consequently, it plays a pivotal role in reshaping the tumor immune microenvironment. In recent years, immunotherapy for tumors has been increasingly applied to clinical practice with notable success achieved through approaches such as immune checkpoint inhibitor therapy and adoptive cell immunotherapy. Targeting m6A modifications to interfere with the immune system, such as targeting dysregulated m6A regulators through small molecule inhibitors and inducing immune cell reprogramming, can improve anti-tumor immune response and strengthen immune cells′ ability to recognize and kill tumor cells. The m6A modification represents a novel avenue for potential clinical application within tumor immunotherapy. This review provides a comprehensive summary of the regulatory impact of m6A methylation modification on immune cells in the context of cancer, while also delving into novel targets for tumor immunotherapy.

42. Genetic variations in NER pathway gene polymorphisms and Wilms tumor risk: A six‐center case–control study in East China.

Abstract

The nucleotide excision repair (NER) system is one of the main ways to protect organisms from DNA damage caused by endogenous and exogenous carcinogens. NER deficiency increases genome mutations, chromosomal aberrations, and cancer viability. However, the genetic association between Wilms tumor and NER pathway gene polymorphisms needs to be further validated. We assessed the associations between 19 NER gene polymorphisms and Wilms tumor susceptibility in 416 cases and 936 controls from East China via the TaqMan method. We found that xeroderma pigmentosum group D (XPD) rs238406 and rs13181 significantly decreased the risk of Wilms tumor [adjusted odds ratio (OR) = 0.59, 95% confidence interval (CI) = 0.46–0.75, p <.0001; adjusted OR = 0.63, 95% CI = 0.44–0.89, p = .009, respectively]. Furthermore, the rs751402 and rs2296147 polymorphisms in the xeroderma pigmentosum group G (XPG) gene were significantly correlated with an increased risk for Wilms tumor (adjusted OR = 1.47, 95% CI = 1.03–2.09, p = .034; adjusted OR = 2.14, 95% CI = 1.29–3.56, p = .003, respectively). Expression quantitative trait loci (eQTL) analysis revealed that these four polymorphisms may affect the expression of genes that are adjacent to XPD and XPG. Our study provides evidence that XPD and XPG gene polymorphisms are associated with Wilms tumor risk. Nonetheless, these findings should be confirmed in a larger sample size. [ABSTRACT FROM AUTHOR]

43. Study on the effect of residential interior decoration on children's respiratory diseases: A field trial conducted in Chongqing, China.

Abstract

Pollutants such as formaldehyde, benzene series compounds and total volatile organic compounds (TVOC) released by residential interior decoration activities (including household renovation and new furniture) pose a severe threat to children's respiratory health. To study the effect of indoor decoration on children's respiratory diseases and provide suggestions to prevent it, we conducted a cross-sectional questionnaire survey amongst 6547 preschoolers from 34 kindergartens in Chongqing, China, and selected 39 houses to measure their indoor air pollutants concentrations. The results showed that household decoration, during pregnancy and early childhood, would increase the risk of children's rhinitis, asthma and pneumonia. Vacancy time after the completion of decoration, furniture pieces and decoration materials significantly affected the concentration of formaldehyde, xylene and TVOC, which could pose a potential risk to children's respiratory health. The concentration of xylene and TVOC and the incidence of children's asthma and pneumonia were both negatively correlated with the vacancy time. Decoration materials such as composite and ceramic tile/stone/cement flooring and water-based/latex/oil-based paints emit more gaseous pollutants than solid wood flooring and wallpaper, respectively, which were more likely to increase the risk of children's respiratory diseases. Theoretically, it bridges national identity theory and political ideology using spatial network analysis; expanding understanding of how cultural heritage tourism contributes to national identity consolidation and the political narrative within a spatial framework. This contributes to the literature by demonstrating the role of SCNs in promoting cultural tourism's sustainable growth, addressing the call for methodological advances in cultural tourism research. In terms of the managerial side, it guides policymakers in leveraging SCNs for balanced cultural tourism growth through strategic planning and digital innovation.
