Pemphigus vulgaris (PV) stands as a rare autoimmune bullous disease, while the precise underlying mechanism remains incompletely elucidated. High‐throughput proteomic methodologies, such as LC‐MS/MS, have facilitated the quantification and characterisation of proteomes from clinical skin samples, enhancing our comprehension of PV pathogenesis. The objective of this study is to elucidate the signalling mechanisms underlying PV through proteomic analysis. Proteins and cell suspension were extracted from skin biopsies obtained from both PV patients and healthy volunteers and subsequently analysed using LC‐MS/MS and scRNA‐seq. Cultured keratinocytes were treated with PV serum, followed by an assessment of protein expression levels using immunofluorescence and western blotting. A total of 880, 605, and 586 differentially expressed proteins (DEPs) were identified between the lesion vs. control, non‐lesion vs. control, and lesion vs. non‐lesion groups, respectively. The oxidative phosphorylation (OXPHOS) pathway showed activation in PV. Keratinocytes are the major cell population in the epidermis and highly expressed ATP5PF, ATP6V1G1, COX6B1, COX6A1, and NDUFA9. In the cellular model, there was a notable increase in the expression levels of OXPHOS‐related proteins (V‐ATP5A, III‐UQCRC2, II‐SDHB, I‐NDUFB8), along with STAT1, p‐STAT1, and p‐JAK1. Furthermore, both the OXPHOS inhibitor metformin and the JAK1 inhibitor tofacitinib demonstrated therapeutic effects on PV serum‐induced cell separation, attenuating cell detachment. Metformin notably reduced the expression of V‐ATP5A, III‐UQCRC2, II‐SDHB, I‐NDUFB8, p‐STAT1, p‐JAK1, whereas tofacitinib decreased the expression of p‐STAT1 and p‐JAK1, with minimal impact on the expression of V‐ATP5A, III‐UQCRC2, II‐SDHB, and I‐NDUFB8. As one of the key components of the circuit breaker operating mechanism, the relay can experience performance degradation due to harsh environmental factors, such as salt spray, during operation, which can ultimately affect the normal operation of the circuit breaker and threaten the safe and stable operation of the power system. To effectively evaluate the condition of the relay operating in salt spray environments over the long term, this study conducted accelerated aging experiments on a certain model of auxiliary switch under salt spray conditions using an existing test platform. The relay's pull-in voltage, release voltage, pull-in time, and release time were analyzed as characteristic parameters affected by salt spray. The results showed that the intrusion of salt spray caused a decline in the coil performance of the relay, which required higher voltage to provide the electromagnetic force needed for operation, leading to an increase in operating voltage. On the other hand, coil degradation also caused a decrease in the electromagnetic force generated by the step voltage, resulting in a slower operating time. Subsequently, a Genetic Algorithm_Back Propagation Neural Network GA_BP) algorithm model suitable for identifying the relay status in this study was established. After optimization by a Genetic Algorithm Neural Network (GA), the recognition accuracy increased from 78.6% to 91.8%, which showed significant improvement. By clarifying the changes in the characteristic parameters of the relay in a salt spray environment and relying on experimental data, this study established a relevant state recognition model. The research results have important engineering application value for understanding the relay operating status of the circuit breaker operating mechanism in salt spray environments, conducting circuit breaker operating mechanism relay life prediction, and preventing circuit breaker operating mechanism failures. Background: Previous studies have shown that individuals with schizophrenia have a greater risk for psoriasis than a typical person. This suggests that there might be a shared genetic etiology between the 2 conditions. We aimed to characterize the potential shared genetic susceptibility between schizophrenia and psoriasis using genome-wide marker genotype data. Methods: We obtained genetic data on individuals with psoriasis, schizophrenia and control individuals. We applied a marker-based coheritability estimation procedure, polygenic score analysis, a gene set enrichment test and a least absolute shrinkage and selection operator regression model to estimate the potential shared genetic etiology between the 2 diseases. We validated the results in independent schizophrenia and psoriasis cohorts from Singapore. Results: We included 1139 individuals with psoriasis, 744 with schizophrenia and 1678 controls in our analysis, and we validated the results in independent cohorts, including 441 individuals with psoriasis (and 2420 controls) and 1630 with schizophrenia (and 1860 controls). We estimated that a large fraction of schizophrenia and psoriasis risk could be attributed to common variants ([h.sup.2.sub.SNP] = 29% ± 5.0%, p = 2.00 x [10.sup.-8]), with a coheritability estimate between the traits of 21%. We identified 5 variants within the human leukocyte antigen (HLA) gene region, which were most likely to be associated with both diseases and collectively conferred a significant risk effect (odds ratio of highest risk quartile = 6.03, p < 2.00 x [10.sup.-16]). We discovered that variants contributing most to the shared heritable component between psoriasis and schizophrenia were enriched in antigen processing and cell endoplasmic reticulum. Limitations: Our sample size was relatively small. The findings of 5 HLA gene variants were complicated by the complex structure in the HLA region. Conclusion: We found evidence for a shared genetic etiology between schizophrenia and psoriasis.
