Abstract Objective Construction of a neuroblastoma (NB) prognostic predictive model based on pyroptosis-related genes (PRGs) to improve individualized management of NB patients. Methods The NB cohort GSE49711 was obtained from the Gene Expression Omnibus (GEO) database, and a total of 498 patients were enrolled into the study, which were randomized into a training set and a test set at a ratio of 1:1, with 250 patients in the training set and 248 patients in the test set. A risk prediction model was constructed using the training set, and the GSE49711 cohort and test set were used as internal validation to verify the reliability of the model. Independent predictors associated with prognosis were screened using univariate and multivariate COX regression analyses, and risk score models were constructed. Single-cell gene set enrichment analysis (ssGSEA) was used to assess the relationship between PRGs and the tumor immune microenvironment. Nomograms were constructed to extend the clinical usability of the model and the reliability of the model was verified using ROC curves and calibration curves. Protein interaction networks of risk genes were mapped using the String database, and the expression of PRGs in NB cell lines was staged using the CCLE database. Results A prognostic model was first developed with the training set: the risk score formula was (− 0.30 × GSDMB) + (− 0.46 × IL-18) + (− 0.21 × NLRP3) + (0.56 × AIM2). Patients were categorized into high- and low-risk groups based on the median risk score value. BackgroundOverweight/obesity is a serious public health problem that affects a large part of the world population across all age and racial/ethnic groups. However, there has not been a meta-analysis of the prevalence of childhood and adolescent overweight/obesity in China during the past 30 years.MethodsThe China National Knowledge Infrastructure and Wanfang DATA, MEDLINE, EMBASE and Cumulative Index to Nursing and Allied Health Literature were searched for relevant studies published between January 1970 and June 2012. The prevalence of overweight/obesity over time was pooled using Stata/SE, version 9. Summary statistics (odds ratios, ORs) were used to compare sex-specific and urban-rural preponderance of overweight/obesity using Review Manager.ResultsAfter screening 1326 papers, we included 35 papers (41 studies), most of medium quality. The prevalence of overweight/obesity increased from 1.8% (95% confidence interval [CI], 0.4%-3.1%) and 0.4% (95% CI, -0.1% to -0.8%) respectively in 1981-1985 to 13.1% (95% CI, 11.2%-15.0%) and 7.5% (95% CI, 6.6%-8.4%) respectively in 2006-2010. The average annual increase was 8.3% and 12.4% respectively. Boys were more likely to be overweight/obese than girls (OR, 1.36; 95% CI, 1.24-1.49 and OR, 1.68; 95% CI, 1.52-1.86 respectively). The prevalence of overweight/obesity was higher in urban areas than in rural areas (OR, 1.66; 95% CI, 1.54-1.79 and OR, 1.97; 95% CI, 1.68-2.30 respectively). For age-specific subgroup analyses, both overweight and obesity increased more rapidly in the toddler stage than in other developmental stages. Sensitivity analyses showed that sample-size differences, study quality, overweight/obesity criteria and geographical distribution affected overweight/obesity prevalence.ConclusionsToddlers and urban boys were at particularly high risk; the prevalence in these groups increased more rapidly than in their counterparts. Public health prevention strategies are urgently needed to modify health behaviors of children and adolescents and control overweight/obesity in China.

Background: Glutamine is crucial for the activation and efficacy of T cells, and may play a role in regulating the immune environment. This study aimed to investigate the potential role of glutamine in the activation and proliferation of induced regulatory T cells (iTregs). Methods: CD4+CD45RA1T cells were sorted from peripheral blood mononuclear cells and cultured to analyze iTreg differentiation. Glutamine was then added to the culture system to evaluate the effects of glutamine on iTregs by determining oxidative phosphorylation (OXPHOS), apoptosis, and cytokine secretion. Additionally, a humanized murine graft-versus-host disease (GVHD) model was constructed to confirm the efficacy of glutaminetreated iTregs in vivo. Results: After being cultured in vitro, glutamine significantly enhanced the levels of Foxp3, CTLA-4, CD39, CD69, IL-10, TGF-ß, and Ki67 (CTLA-4, IL-10, TGF-ß are immunosuppressive markers of iTregs) compared with that of the control iTregs (P < 0.05). Furthermore, the growth curve showed that the proliferative ability of glutaminetreated iTregs was better than that of the control iTregs (P < 0.01). Compared with the control iTregs, glutamine supplementation significantly increased oxygen consumption rates and ATP production (P < 0.05), significantly downregulated Annexin V and Caspase 3, and upregulated BCL2 (P < 0.05). However, GPNA significantly reversed the effects of glutamine (P < 0.05). Finally, a xeno-GVHD mouse model was successfully established to confirm that glutamine-treated iTregs increased the mice survival rate, delayed weight loss, and alleviated colon injury. Conclusion: Glutamine supplementation can improve the activity and immunosuppressive action of iTregs, and the possible mechanisms by which this occurs are related to cell proliferation, apoptosis, and OXPHOS. Abstract Understanding the diversity of life history, life stage connectivity and population is essential to determine the spatial scale over which fish populations operate. Otolith microchemistry analysis is a powerful tool to elucidate the life history and population connectivity of fish, providing important insights to the natal origin and population structure. In this study, we used laser ablation inductively coupled plasma mass spectrometry to analyze the chemical composition of otoliths throughout the entire lifetime of endangered fourfinger threadfin species, Eleutheronema tetradactylum. We reconstructed the life history of E. tetradactylum from Southern China collected from different locations over a spatial scale of 1200 km. Sr:Ca and Ba:Ca ratios profiles from otolith core-to-edge analysis suggested two contrasting life history patterns. Based on the differences in early life stages, we identified some fish spending their first year in an estuarine environment with subsequent movement to marine coastal systems, while some fish remaining in the coastal systems throughout their entire early life history stages. The non-metric multi-dimensional scaling showed a strong overlap in otolith core elemental composition, indicating a large-scale connectivity in the life history of E. tetradactylum. The immature fish from different natal origins mixed to a large extent when they fed and overwintered in the extensive offshore waters. Clustering of near core chemistry pointed to three possible sources of nursery for the threadfin fish. This study demonstrated the diversity of life history patterns of E. tetradactylum in Southern Chinese waters. Restoration in egg and larvae densities in coastal waters and estuaries may enhance their population abundances.

This study was performed by using D-loop region sequences to investigate genetic diversity and population structure across eight representative groups of M. nipponense from south to north China. Eight populations comprising a total of 144 individuals were from Songhua River (SHJ), Yellow River (HH), Yangtze River (YZ), Pearl River (ZJ), Weishan Lake (WSH), Taihu Lake (TH), Bosten Lake (BH) and Wanquan River (WQH). The 1130-bp D-loop region exhibited a higher content of A + T (79.04%) than C + G (20.95%). In addition, 239 variable sites and 73 haplotypes were detected. Loss of valued diadromous fishes and their habitats is one of the most critical problems in aquatic habitat connection and resource management worldwide. In China, the Poyang, Dongting, Gaobao, Gucheng, Dongping, and Taihu lakes were known to be historical migratory spawning sites of the anadromous estuarine tapertail anchovy Coilia nasus. However, except for Poyang Lake, it is believed that these lakes are no longer used by anadromous fish owing to overfishing, water pollution, and loss of connectivity. To confirm this assumption, we used an electron probe microanalyzer to analyze elemental strontium (Sr) and calcium (Ca) microchemical patterns in the otoliths of C. nasus individuals sampled from these lakes, in accordance with our previous analysis of the otolithic patterns of the same species sampled from habitat areas characterized by different salinity gradients. The results of line transect analysis of Sr/Ca ratios and Sr X-ray intensity maps of the otoliths indicated that all individuals from Dongting, Gaobao, Gucheng, Taihu, and Dongping lakes were characterized by a freshwater-resident life history. In contrast, individuals from Poyang Lake exhibited both freshwater-resident and anadromous life histories. The findings of this pilot study suggest that anadromous C. nasus can be found in Poyang Lake but are unlikely to be found in Dongting, Gaobao, Gucheng, Dongping, or Taihu lakes, despite these lakes being historical distribution areas or even spawning sites. This anchovy can possibly be used as a good model species for understanding the aforementioned global problem. Given that C. nasus is a commercially important species, restoration of its natural habitats and maintenance of their connections are recommended for its management and conservation.
