Major histocompatibility complex Class II (MHCII) proteins initiate and regulate immune responses by presentation of antigenic peptides to CD4+ T-cells and self-restriction. The interactions between MHCII and peptides determine the specificity of the immune response and are crucial in immunotherapy and cancer vaccine design. With the ever-increasing amount of MHCII-peptide binding data available, many computational approaches have been developed for MHCII-peptide interaction prediction over the last decade. There is thus an urgent need to provide an up-to-date overview and assessment of these newly developed computational methods. To benchmark the prediction performance of these methods, we constructed an independent dataset containing binding and non-binding peptides to 20 human MHCII protein allotypes from the Immune Epitope Database, covering DP, DR and DQ alleles. After collecting 11 known predictors up to January 2022, we evaluated those available through a webserver or standalone packages on this independent dataset. The benchmarking results show that MixMHC2pred and NetMHCIIpan-4.1 achieve the best performance among all predictors. In general, newly developed methods perform better than older ones due to the rapid expansion of data on which they are trained and the development of deep learning algorithms. Our manuscript not only draws a full picture of the state-of-art of MHCII-peptide binding prediction, but also guides researchers in the choice among the different predictors. More importantly, it will inspire biomedical researchers in both academia and industry for the future developments in this field.

A bullet point cloud registration algorithm with a low overlap rate based on line feature detection was proposed to solve the problem of the difficulty and low efficiency of point cloud registration due to the low overlap rate among point clouds sampled by the bullet model. In this paper, voxel downsampling is used to remove some noise points and outliers from the bullet point cloud and applied to the specified resolution to reduce the calculation cost. The bullet point cloud is transformed to a better initial position by fitting the central axis with the geometrical features of the bullet. Then, the direction vector of the bullet linear features is obtained by using an icosahedral fitting discrete Hough transform to simplify the parameter space of the search transformation. Finally, the optimal rotation angle is searched for in the parameter space by using the improved Cuckoo algorithm to realize the registration of the bullet point cloud with a low overlap rate. Simulation and experimental results show that the proposed registration method can accurately register bullet point clouds of different densities with a low overlap rate. Compared with the commonly used ICP, GICP, and TRICP algorithms, the registration error of the proposed algorithm is reduced by 92.68% on average when the overlap rate is 52.85%. The registration error is reduced by 98.87% in the case of a 41.36% overlap rate, by 99.52% in the case of a 33.02% overlap rate, and by 98.89% in the case of a 22.75% overlap rate.

The amount of cultivated land per capita in China is relatively low, and the phenomenon of non-cultivated land (NCL) in recent years has negatively impacted the stability of grain production in China. In this study, long-time series images obtained via satellite remote sensing were used to monitor spatio-temporal changes in NCL at the county scale. Seven-phase images were acquired from 1990 to 2020 (every five years) using medium-resolution Landsat MSS, TM, ETM+, and Sentinel MSI. Vegetation indices and texture features were extracted for all images. Terrain features such as slope, aspect and elevation were extracted from the DEM data. Combining vegetation index features, texture features, terrain features and multispectral bands, the image classification was performed using the random forest (RF) algorithm. The indices of classification accuracy assessment indices included overall accuracy (OA) and multiclass F-scores (Fm). Zonal statistics were used to calculate the area of cultivated land in towns for 1990 and 2020, and to create grades for the reduction of cultivated land. Finally, indicators including land use dynamic degree (LUDD), land use type change (LUTC) and land use change rate (LUCR) were adopted to reflect the spatio-temporal of NCL in the study area. The results show that RF classification algorithm achieves accurate and efficient land use extraction. The OA were greater than 86%, and the Fm were over 0.88. The cultivated land area in the study area showed decreasing trend. From 1990 to 2020, the ratio of cultivated land decreased from 59.75% to 50.21%. Meanwhile, the dynamic degree of cultivated land increased annually. The conversion of cultivated land into construction land was dominant, accounting for 31.84% of the total change in cultivated land over the past 30 years. This study also reveals that NCL is highly related to local economic and land-use policies. Multi-source remote sensing data have been used to quantitatively analyse the spatio-temporal changes in cultivated land conversion, providing a reference for relevant land management departments to master cultivated land use changes and adjust land management policies.

Netted melons are welcomed for their soft and sweet pulp and strong aroma during the best-tasting period. The best-tasting period was highly correlated with its soluble solid content (SSC). However, the SSC of the intact melon was difficult to determine due to the low relationship between the hardness, color, or appearance of fruit peel and its SSC. Consequently, a rapid, accurate, and non-destructive method to determine the SSC of netted melons was the key to determining the best-tasting period. A hyperspectral model was constructed to estimate the SSC of intact netted melons. The combination of continuous wavelet transform and partial least squares or random forest algorithm was employed to improve the estimation accuracy of the hyperspectral model. Specifically, the hyperspectra of the diffuse reflection and SSC of 261 fruit samples were collected. The sensitivity band was screened based on the correlation analysis and continuous wavelet transform decomposition. The correlation coefficient and RMSE of the random forest regression model decomposed by the continuous wavelet transform were 0.72 and 0.98%, respectively. The decomposition of the continuous wavelet transform improved the correlation coefficient by 5 and 1.178 times at 754 and 880 nm, respectively. The random forest regression model enhanced the determination coefficient by at least 56.5% than the partial least squares regression model, and the continuous wavelet transform decomposition further enhanced the determination coefficient of the random forest regression model by 4.34%. Meanwhile, the RMSE of the random forest regression model was reduced. Therefore, the decomposition of the continuous wavelet transform improved the stability and prediction ability of the random forest regression model.

Invalid points, such as shadow and background, in the captured fringe patterns of fringe projection profilometry (FPP) are often inevitable due to the limited field of view measurements of three-dimensional (3D) imaging equipment. To ensure the quality of 3D reconstruction data, these invalid points must be identified and removed. FPP captures co-frequency-based fringe pattern sequences and approximately distributes this data along an ideal cosine curve. We propose an invalid points removal method based on an error energy function. By analyzing the relationship between the pixel values of a series of captured co-frequency patterns and an ideal cosine curve, we quantize the error energy by using a Gaussian weighted Euclidean distance. An improved Gaussian filtering method based on modulation intensity is used to significantly increase the error energy difference between the valid points of the target and the invalid points of the shadow/background area. Finally, the points whose error energy is greater than a certain error threshold are removed as invalid points. Experimental results indicate that this method can efficiently remove invalid points in fringe patterns and performs better than existing traditional methods.

The point cloud data from actual measurements are often sparse and incomplete, making it difficult to apply them directly to visual processing and 3D reconstruction. The point cloud completion task can predict missing parts based on a sparse and incomplete point cloud model. However, the disordered and unstructured characteristics of point clouds make it difficult for neural networks to obtain detailed spatial structures and topological relationships, resulting in a challenging point cloud completion task. Existing point cloud completion methods can only predict the rough geometry of the point cloud, but cannot accurately predict the local details. To address the shortcomings of existing point cloud complementation methods, this paper describes a novel network for adaptive point cloud growth, MAPGNet, which generates a sparse skeletal point cloud using the skeletal features in the composite encoder, and then adaptively grows the local point cloud in the spherical neighborhood of each point using the growth features to complement the details of the point cloud in two steps. In this paper, the Offset Transformer module is added in the process of complementation to enhance the contextual connection between point clouds. As a result, MAPGNet improves the quality of the generated point clouds and recovers more local detail information. Comparing our algorithm with other state-of-the-art algorithms in different datasets, experimental results show that our algorithm has advantages in dense point cloud completion.

With the rapid increase in the costs of rural labour and the adjustment of planting structures, the phenomenon of farmland abandonment has appeared in China. It is of great significance to promptly and accurately grasp the information on dynamic temporal and spatial changes in abandoned farmland to ensure national food security and the sustainable use of cultivated land. Luquan District in Hebei, China was selected as the research area based on multispectral images from Sentinel-2A, Landsat-7, and Landsat-8 combined with methods of random forest (RF) classification and vegetation index change detection. Rules for the identification of abandoned farmland were also developed, and remote sensing monitoring of the abandonment status of the cultivated land was also carried out in the study area. We also obtained the spatial distribution of abandoned and reclaimed farmland and analysed the frequency of farmland abandonment. The results show that the overall accuracy of the land-use time-series map ranged from 90.20% to 96.92% for the study period of 2010–2020. The average rate of farmland abandonment in the study area was 10.62%, with the lowest rate (5.83%) in 2020 and the highest (14.09%) in 2012. From 2011 to 2020, the maximum farmland abandonment area was 3906.02 hm2, and the minimum area was 1618.74 hm2. The farmland abandonment area showed a trend of first increasing and then decreasing. From 2012 to 2020, the maximum area of reclaimed farmland was 291.49 hm2, and the highest rate of reclamation was 14.26%. The overall reclamation rate was low. The abandonment frequency of most of the abandoned farmland was 1–3 years, covering an area of 8193.73 hm2, which comprised 79% of the total area of abandoned farmland. The frequency of abandonment was inversely proportional to the area of abandoned farmland. Farmland abandonment mainly occurred in hilly areas. We expect that our results can provide case studies for long time series in farmland abandonment research and can provide a reference for studying the driving factors, risk assessment, and policymaking with respect to abandoned farmland.

Lodging is one of the main problems in maize production. Assessing the self-recovery ability of maize plants after lodging at different growth stages is of great significance for yield loss assessment and agricultural insurance claims. The objective of this study was to quantitatively analyse the effects of different growth stages and lodging severity on the self-recovery ability of maize plants using UAV-LiDAR data. The multi-temporal point cloud data obtained by the RIEGL VUX-1 laser scanner were used to construct the canopy height model of the lodging maize. Then the estimated canopy heights of the maize at different growth stages and lodging severity were obtained. The measured values were used to verify the accuracy of the canopy height estimation and to invert the corresponding lodging angle. After verifying the accuracy of the canopy height, the accuracy parameter of the tasselling stage was R2 = 0.9824, root mean square error (RMSE) = 0.0613 m, and nRMSE = 3.745%. That of the filling stage was R2 = 0.9470, RMSE = 0.1294 m, and nRMSE = 9.889%, which showed that the UAV-LiDAR could accurately estimate the height of the maize canopy. By comparing the yield, canopy height, and lodging angle of maize, it was found that the self-recovery ability of maize at the tasselling stage was stronger than that at the filling stage, but the yield reduction rate was 14.16~26.37% higher than that at the filling stage. The more serious the damage of the lodging is to the roots and support structure of the maize plant, the weaker is the self-recovery ability. Therefore, the self-recovery ability of the stem tilt was the strongest, while that of root lodging and root stem folding was the weakest. The results showed that the UAV-LiDAR could effectively assess the self-recovery ability of maize after lodging.

Tracking objects over time, i.e., identity (ID) consistency, is important when dealing with multiple object tracking (MOT). Especially in complex scenes with occlusion and interaction of objects this is challenging. Significant improvements in single object tracking (SOT) methods have inspired the introduction of SOT to MOT to improve the robustness, that is, maintaining object identities as long as possible, as well as helping alleviate the limitations from imperfect detections. SOT methods are constantly generalized to capture appearance changes of the object, and designed to efficiently distinguish the object from the background. Hence, simply extending SOT to a MOT scenario, which consists of a complex scene with spatially mixed, occluded, and similar objects, will encounter problems in computational efficiency and drifted results. To address this issue, we propose a binary-channel verification model that deeply excavates the potential of SOT in refining the representation while maintaining the identities of the object. In particular, we construct an integrated model that jointly processes the previous information of existing objects and new incoming detections, by using a unified correlation filter through the whole process to maintain consistency. A delay processing strategy consisting of the three parts—attaching, re-initialization, and re-claiming—is proposed to tackle drifted results caused by occlusion. Avoiding the fuzzy appearance features of complex scenes in MOT, this strategy can improve the ability to distinguish specific objects from each other without contaminating the fragile training space of a single object tracker, which is the main cause of the drift results. We demonstrate the effectiveness of our proposed approach on the MOT17 challenge benchmarks. Our approach shows better overall ID consistency performance in comparison with previous works.

Under the dynamic working conditions of a star sensor, motion blur of the star will appear due to its energy dispersion during imaging, leading to the degradation of the star centroid accuracy and attitude accuracy of the star sensor. To address this, a restoration method of a blurred star image for a star sensor under dynamic conditions is presented in this paper. First, a kinematic model of the star centroid and the degradation function of blurred star image under different conditions are analyzed. Then, an improved curvature filtering method based on energy function is proposed to remove the noise and improve the signal-to-noise ratio of the star image. Finally, the Richardson Lucy algorithm is used and the termination condition of the iterative equation is established by using the star centroid coordinates in three consecutive frames of restored images to ensure the restoration effect of the blurred star image and the accuracy of the star centroid coordinates. Under the dynamic condition of 0~4°/s, the proposed algorithm can effectively improve the signal-to-noise ratio of a blurred star image and maintain an error of the star centroid coordinates that is less than 0.1 pixels, which meets the requirement for high centroid accuracy.

Road detection is a crucial research topic in computer vision, especially in the framework of autonomous driving and driver assistance. Moreover, it is an invaluable step for other tasks such as collision warning, vehicle detection, and pedestrian detection. Nevertheless, road detection remains challenging due to the presence of continuously changing backgrounds, varying illumination (shadows and highlights), variability of road appearance (size, shape, and color), and differently shaped objects (lane markings, vehicles, and pedestrians). In this paper, we propose an algorithm fusing appearance and prior cues for road detection. Firstly, input images are preprocessed by simple linear iterative clustering (SLIC), morphological processing, and illuminant invariant transformation to get superpixels and remove lane markings, shadows, and highlights. Then, we design a novel seed superpixels selection method and model appearance cues using the Gaussian mixture model with the selected seed superpixels. Next, we propose to construct a road geometric prior model offline, which can provide statistical descriptions and relevant information to infer the location of the road surface. Finally, a Bayesian framework is used to fuse appearance and prior cues. Experiments are carried out on the Karlsruhe Institute of Technology and Toyota Technological Institute (KITTI) road benchmark where the proposed algorithm shows compelling performance and achieves state-of-the-art results among the model-based methods.
