8. Mechanism of human leukocyte antigen-G inhibiting T cell immune function in triple-negative breast cancer

Abstract

Objective To investigate the mechanism of human leukocyte antigen-G (HLA-G) expression and T cells activation in triple-negative breast cancer(TNBC) cells to exert immunosuppressive function. Methods From three groups of breast cancer cell lines（MDA-MB-231，HCC1937， MDA-MB-468）,real-time quantitative polymerase chain reaction (RT-qPCR) and Western blot were detected and screened that the breast cancer cell line was HCC1937,which with high expression of HLA-G, and low expression of fibrinogen-like protein 1 (FGL1), programmed death factor ligand 1 (PD-L1), as well as sialic acid-binding immunoglobulin-like lectin 15 (SIGLEC15) . HCC1937 cells were divided into NC group (control group) and KD group (knockdown group), and RNA interference (RNAi) lentiviral vectors were constructed and transfected into KD cells to knock down HLA-G gene, and the verification was carried out. HCC1937 cells were treated with HLA-G antibody blocking/nonblocking, and Jurkat cells were treated with CD3 and CD28 activation/non-activation, respectively, and the two types of cells were co-cultured into 8 groups: NC--, KD--, NC+-, KD+-, NC-+, KD-+, NC++, and KD++ groups. Cell scratch assay, MTT and ELISA method were used to detect cell migration rate, proliferation rate, and interferon (IFN)-γ and interleukin (IL)-2 levels in cell supernatants, respectively. Results RT-qPCR showed that the HLA-G gene was successfully knocked down in HCC1937 cells. Compared with the NC++ group, the migration rate of the KD++ group was significantly increased (P＜0.05), but there was no statistical difference in the cell proliferation rate between each pair of co-culture groups (P＞0.05). The concentration of IFN-γ in the KD++ group was lower than that in the NC++ group and the NC-+ group (P＜0.05), but there was no significant difference in the concentration of IL-2 between each pair of co-culture groups (P＞0.05). Conclusion HLA-G in TNBC cells may promote the secretion of IFN-γ by activing T cells, and IFN-γ can inhibit the immune function of T cells and promote tumor metastasis after HLA-G decreased."

Abstract

Using the density function theory in combination with the non-equilibrium Green's function method, the thermoelectric properties of molecular devices based on transition metal–terpyridine complexes are investigated. The results show that their thermoelectric properties can be significantly improved by changing the transition metal and the twist angle of the complex molecule, which is caused by shifting the molecular energy levels, resulting in increased coupling strength between the electrodes and the central molecule. The ZT value of the Ru-containing molecular device can reach up to 0.9 at room temperature, which is three orders of magnitude greater than that of the graphene nanoribbons of the same width. In addition, its thermoelectric performance can be further promoted by suppressing phonon thermal conductance through enhanced isotope scattering. The ZT value of doped devices can reach up to 1.0 in the range of 300–700 K. This work may help in the design and fabrication of transition metal-containing twistable molecular devices and provide effective methods to regulate their thermoelectric properties. Abstract

Abstract Purpose To evaluate the safety and effectiveness of endovascular treatment for massive haemoptysis caused by pulmonary pseudoaneurysm (PAP). Methods The clinical data, imaging data, and endovascular treatment protocol of 23 patients with massive haemoptysis caused by continuous PAP were retrospectively analysed. The success, complications, postoperative recurrence rate, and influence of the treatment on pulmonary artery pressure were also evaluated. Results Nineteen patients with a bronchial artery-pulmonary artery (BA-PA) and/or nonbronchial systemic artery-pulmonary artery (NBSA-PA) fistula underwent bronchial artery embolization (BAE) and/or nonbronchial systemic artery embolization (NBSAE) + pulmonary artery embolization (PAE). The pulmonary artery (PA) pressures before and after embolization were 52.11 ± 2.12 (35–69 cmH2O) and 33.58 ± 1.63 (22–44 cmH2O), respectively (P = 0.001). Four patients did not have a BA-PA and/or NBSA-PA fistula. Embolization was performed in two patients with a distal PAP of the pulmonalis lobar arteria. Bare stent-assisted microcoils embolization was performed in the other two patients with a PAP of the main pulmonary lobar arteries. The PA pressures of the four patients before and after treatment were 24.50 ± 1.32 (22–28 cmH2O) and 24.75 ± 1.70 (22–29 cmH2O), respectively (P = 0.850). The technique had a 100% success rate with no serious complications and a postoperative recurrence rate of 30%. Conclusion Endovascular treatment is safe and effective for massive haemoptysis caused by PAP. BAE and/or NBSAE can effectively reduce pulmonary hypertension in patients with a BA-PA and/or NBSA-PA fistula.
