Abstract N6-methyladenosine (m6A) is important in regulating mRNA stability, splicing, and translation, and it also contributes to tumor development. However, there is still limited understanding of the comprehensive effects of m6A modification patterns on the tumor immune microenvironment, metabolism, and drug resistance in hepatocellular carcinoma (HCC). In this study, we utilized unsupervised clustering based on the expression of 23 m6A regulators to identify m6A clusters. We identified differential m6A modification patterns and characterized m6A-gene-cluster A, which exhibited poorer survival rates, a higher abundance of Treg cells, and increased expression of TGFβ in the tumor microenvironment (TME). Additionally, m6A-gene-cluster A demonstrated higher levels of glycolysis activity, cholesterol metabolism, and fatty acid biosynthesis. We also found that the m6A score was associated with prognosis and drug resistance. Patients with a low m6A score experienced worse prognoses, which were linked to an abundance of Treg cells, upregulation of TGFβ, and increased metabolic activity. HCC patients with a higher m6A score showed improved prognosis following sorafenib treatment and immunotherapy. In conclusion, we reveals the association between m6A modification patterns and the tumor immune microenvironment, metabolism, and drug resistance in HCC. Furthermore, the m6A score holds potential as a predictive factor for the efficacy of targeted therapy and immunotherapy in HCC.

Objective: α-Klotho is a potential biological marker of aging with satisfactory clinical applicability. However, its prognostic significance in age-related diseases has largely been undermined. Therefore, we aimed to report the prognostic value of serum α-klotho levels in age-related diseases. Methods: Participants with available serum α-klotho data from the National Health and Nutrition Examination Survey (2007–2016) were included. Their survival status was collected at 7.62 ± 2.99 years after serum α-klotho data was collected, and the endpoint was all-cause and cardiovascular mortality. A Cox regression model was established to examine the association between serum α-klotho levels and all-cause and cardiovascular mortality. Results: The present study included 13,746 U.S. adults with a survey-weighted mean age of 56.19 ± 10.42 years old. Of these, 52.2 % were female and 72.9 % were non-Hispanic whites. Most studies on building height estimation using remote sensing imagery mainly focus on urban buildings with relatively regular shape and relatively flat terrain, and pay little attention to large buildings with complex terrain like dams. A new dam height measurement method was proposed in this paper, which used shadow measurement data and metadata from multiple image sources. The method not only considers the geometric relationship between the sun, the satellite and the dam, but also considers the influence of the satellite zenith and introduces the correction factor, which brings the higher precision. Two dams with a maximum height of more than 200 m were studied, and the dam height is estimated by considering the topography around the dam and the shape of the dam. The experimental results show that the Mean Relative Error (MRE) of the estimated dam height using our proposed method are 3.1% and 4.7% for our two study areas, while the MRE of the traditional models is more than 13%. By doing so, we are able to calculate dam height and obtain information on temporal changes in dam height during the construction process, even in situations without a Digital Surface Model (DSM). Therefore, the proposed method will be propitious to the dynamic supervision of the construction process of the dams effectively.

In recent years, the global outbreak of COVID-19 has posed an extremely serious life-safety risk to humans, and in order to maximize the diagnostic efficiency of physicians, it is extremely valuable to investigate the methods of lesion segmentation in images of COVID-19. Aiming at the problems of existing deep learning models, such as low segmentation accuracy, poor model generalization performance, large model parameters and difficult deployment, we propose an UNet segmentation network integrating multi-scale attention for COVID-19 CT images. Specifically, the UNet network model is utilized as the base network, and the structure of multi-scale convolutional attention is proposed in the encoder stage to enhance the network's ability to capture multi-scale information. Second, a local channel attention module is proposed to extract spatial information by modeling local relationships to generate channel domain weights, to supplement detailed information about the target region to reduce information redundancy and to enhance important information. Moreover, the network model encoder segment uses the Meta-ACON activation function to avoid the overfitting phenomenon of the model and to improve the model's representational ability. A large number of experimental results on publicly available mixed data sets show that compared with the current mainstream image segmentation algorithms, the pro-posed method can more effectively improve the accuracy and generalization performance of COVID-19 lesions segmentation and provide help for medical diagnosis and analysis.

AbstractAmphotericin B (AmB) is regarded as a first-line therapy against life-threatening invasive fungal infections. Due to its poor oral bioavailability, AmB is restricted to intravenous administration in clinical practice. As science continues to move forward, two lipid-based formulations are successfully developed for oral AmB administration, currently undergoing phase I clinical trials. Encouragingly, lipid-AmB conjugates with emulsions also exhibit a better bioavailability, which may be another strategy to design oral AmB formulation in clinical practice. Thus, this review mainly focused on the two lipid-based formulations in clinical trials, and discussed the potential perspectives of AmB-lipid conjugation-loaded nanocochleates and emulsions.

Abstract The power system dynamics have consistently challenged the rapid and persistent proliferation of photovoltaic (PV) systems in power systems. In this paper, the authors propose the generalized discrete‐time equivalent model (GDEM) of PV power generation system using a fourth‐order dynamic equivalent model for representing the physical characteristics of PV power stations in power system dynamic studies. In addition, the authors propose a grid‐connected system model which includes modelling interfaces (MIs) for the GDEM of power grid and the GDEM of PV power generation system in dynamic studies. The proposed equivalent models of the power grid and PV power generation system will improve the simulation accuracy and speed in dynamic power system studies. The authors have used the IEEE 14‐bus system and simulated various types of short‐circuit faults and PV penetration levels in their study to demonstrate the merits of the proposed GDEM of grid‐connected system in power systems. The pertinent simulation results are analyzed and conclusions are presented.

The motion responses, mooring tensions, and submergence depth are the dominant factors for the arrangement of the Submerged Floating Tunnel (SFT) subjected to waves. Generally, the maximum values of motion responses, mooring tensions, and absolute submergence depth are mainly focused on. In the present study, experiments are implemented to measure the motion responses and mooring tensions of the SFT with different mooring patterns and submergence depths under waves with different characteristic wave heights and periods. In order to evaluate the arrangement of the SFT more effectively and comprehensively, besides the maximum values, several new characteristic parameters are introduced. Such parameters account for the motion responses in the frequency domain, the uniformity of the tension distribution, the length of time during which the cable reaches a relaxed condition during wave action, the KC number, the dimensionless period, the wave height, and the submergence depth. The results from the optimization analysis show the following: according to the characteristic values of motion responses and mooring tensions, the pattern of diagonal cables is better than that of diagonal cables + vertical cables; and within the range of the present experiments, there are optimal dimensionless parameters—the dimensionless submergence depth d0/LP ≥ 0.15, the KC number ≤ 0.8, or the dimensionless wave height Hs/d0 ≤ 0.10—for the condition of which the dynamic responses and mooring tensions vary slightly.

Abstract Objective Gastric cancer (GC) remains a prevailing threat to life. Baicalein exhibits anti-cancer properties. This study estimated the mechanism of baicalein in GC cell apoptosis by mediating endoplasmic reticulum stress (ERS) through the PI3K/AKT pathway. Methods After treatment with different concentrations of baicalein, GC cell (HGC-27 and AGS) viability was detected by MTT assay. AGS cells more sensitive to baicalein treatment were selected as study subjects. The IC50 of baicalein on AGS cells was determined. Colony formation, cell cycle, and apoptosis were detected using crystal violet staining and flow cytometry. Levels of ERS-related and BTG3/PI3K/AKT pathway-related proteins were determined by Western blot. Intracellular Ca2+ level was measured using Fluo-3 AM fluorescence working solution. GC mouse models were established by subcutaneously injecting AGS cells into the right rib and were intragastrically administrated with baicalein. Tumor volume and weight were recorded. Expression of Ki67 in tumor tissues and positive expression of apoptotic cells were detected by immunohistochemistry and TUNEL staining. Results Baicalein inhibited cell proliferation and induced G0/G1 arrest and apoptosis by regulating the cell cycle, and triggered ERS in GC cells. Baicalein impeded the PI3K/AKT pathway by activating BTG3, thereby triggering ERS and inducing apoptosis. BTG3 inhibition reversed baicalein-induced apoptosis and ERS. Baicalein regulated GC cells in a concentration-dependent manner. Moreover, in xenograft mice, baicalein prevented tumor growth, decreased Ki67-positive cells, activated BTG3, and inhibited the PI3K/AKT pathway, thus activating ERS and increasing apoptotic cells. Conclusion Baicalein facilitates GC cell apoptosis by triggering ERS via repression of the PI3K/AKT pathway.

Summary: How neuronal signaling affects brain myelination remains poorly understood. We show dysregulated neuronal RHEB-mTORC1-DLK1 axis impairs brain myelination. Neuronal Rheb cKO impairs oligodendrocyte differentiation/myelination, with activated neuronal expression of the imprinted gene Dlk1. Neuronal Dlk1 cKO ameliorates myelination deficit in neuronal Rheb cKO mice, indicating that activated neuronal Dlk1 expression contributes to impaired myelination caused by Rheb cKO. The effect of Rheb cKO on Dlk1 expression is mediated by mTORC1; neuronal mTor cKO and Raptor cKO and pharmacological inhibition of mTORC1 recapitulate elevated neuronal Dlk1 expression. We demonstrate that both a secreted form of DLK1 and a membrane-bound DLK1 inhibit the differentiation of cultured oligodendrocyte precursor cells into oligodendrocytes expressing myelin proteins. Finally, neuronal expression of Dlk1 in transgenic mice reduces the formation of mature oligodendrocytes and myelination. This study identifies Dlk1 as an inhibitor of oligodendrocyte myelination and a mechanism linking altered neuronal signaling with oligodendrocyte dysfunction.

Herein, activated red mud particles are used as adsorbents for phosphorus adsorption. HCl solutions with different concentrations and deionized water are employed for desorption tests, and the desorption mechanism under the following optimal conditions is investigated: HCl concentration = 0.2 mol/L, desorbent dosage = 0.15 L/g, desorption temperature = 35 °C, and desorption time = 12 h. Under these conditions, the phosphate desorption rate and amount reach 99.11% and 11.29 mg/g, respectively. Notably, the Langmuir isothermal and pseudo-second-order kinetic linear models exhibit consistent results: monomolecular-layer surface desorption is dominant, and chemical desorption limits the rate of surface desorption. Thermodynamic analysis indicates that phosphorus desorption by the desorbents is spontaneous and that high temperatures promote such desorption. Moreover, an intraparticle diffusion model demonstrates that the removal of phosphorus in the form of precipitation from the surface of an activated hematite particle adsorbent primarily occurs via a chemical reaction, and surface micromorphological analysis indicates that desorption is primarily accompanied by Ca dissolution, followed by Al and Fe dissolutions. The desorbents react with the active elements in red mud, and the vibrations of the [SiO4]4− functional groups of calcium–iron garnet and calcite or aragonite disappear. Further, in Fourier-transform infrared spectra, the intensities of the peaks corresponding to the PO43− group considerably decrease. Thus, desorption primarily involves monomolecular-layer chemical desorption.

The J-TEXT capability is enhanced compared to two years ago with several upgrades of its diagnostics and the increase of electron cyclotron resonance heating (ECRH) power to 1 MW. With the application of electron cyclotron wave (ECW), the ECW assisted plasma startup is achieved; the tearing mode is suppressed; the toroidal injection of 300 kW ECW drives around 24 kA current; fast electrons are generated with toroidal injected ECW and the runaway current conversion efficiency increases with ECRH power. The mode coupling between 2/1 and 3/1 modes are extensively studied. The coupled 2/1 and 3/1 modes usually lead to major disruption. Their coupling can be either suppressed or avoided by external resonant magnetic perturbation fields and hence avoids the major disruption. It is also found that the 2/1 threshold of external field is significantly reduced by a pre-excited 3/1 mode, which can be either a locked island or an external kink mode. The disruption control is studied by developing prediction methods capable of cross tokamak application and by new mitigation methods, such as the biased electrode or electromagnetic pellet injector. The high-density operation and related disruptions are studied from various aspects. Approaching the density limit, the collapse of the edge shear layer is observed and such collapse can be prevented by applying edge biasing, leading to an increased density limit. The density limit is also observed to increase, if the plasma is operated in the poloidal divertor configuration or the plasma purity is increased by increasing the pre-filled gas pressure or ECRH power during the start-up phase.

Shield tunneling underneath existing rectangular pipe jacking can often lead to uneven settling. To solve the problem of excessive deformation of the existing underground pedestrian passages, excavated via the rectangular pipe-jacking method due to the adjacent shield tunnels construction beneath, a safety control plan has been proposed based on the shield tunnels constructed beneath a well-type underground pedestrian passage in Hohhot Subway. This plan involves the use of both numerical simulations and on-site monitoring to investigate the deformation patterns of the rectangular pipe jacking. The results suggest that the combined approach of interlayer soil grouting and steel support reinforcement is not only appropriate but also feasible for on-site implementation. It can be concluded that interlayer soil grouting for reinforcement was applied successfully to improve the strength of surrounding soils. Steel support reinforcement is advantageous for controlling the relative deformation between pipe segments. The maximum settlement induced by the shield tunneling for Passage A and Passage B was measured at 17.67 mm and 10.08 mm, respectively, with the final settlement distribution of the rectangular pipe jacking taking on a “W” shape. This study could provide a reference for the design and construction of shield tunnels that undercross existing rectangular pipe-jacking tunnels.

FOXP1, known as a Forkhead-box (FOX) family protein, plays an important role in human tumorigenesis. However, the function and molecular mechanism of FOXP1 in pancreatic cancer (PC) remain unclear. Here, we report that PC patients with FOXP1 overexpression had a higher survival rate compared to patients with low- FOXP1 expression. Additionally, high expression of FOXP1 can markedly inhibit the growth of pancreatic cancer in vivo and in vitro, whereas low expression of FOXP1 effectively promoted the tumorigenesis. Mechanistically, FOXP1 could directly bind the IRF1 promoter, which triggered the transcriptional activity of IRF1. Taken together, FOXP1 suppressed PC growth via IRF1-dependent manner, serving as a potential prognostic biomarker for patients with PC.

As the rapid and continually proliferation of photovoltaic (PV) systems are connected to the power system, the load structure are changeable to lack an accurate dynamic discrete equivalent model to describe its characteristics of power grid. In this study, the generalized discrete-time equivalent model (GDEM) of PV system using a fourth-order dynamic equivalent model for representing the physical characteristics of PV power stations are proposed in power system dynamic studies. The paper then investigates the inherent relations among GDEM parameters in the discrete-time models of PV system to facilitate the GDEM parameters estimation in PV system. Finally, the least square method (LSM) was to identify the GDEM of PV system parameters, and various types of ground faults and PV penetration rate levels is adopted to verify the dynamic characteristics of the proposed GDEM of PV system in power system simulations.

Caspase recruitment domain-containing protein 9 (CARD9) is highly expressed in myeloid cells and has been identified as a central regulator of innate immunity. Increasingly, studies demonstrate that CARD9 also plays a critical role in the development of lung cancer. This review focuses on the clinical significance and potential molecular mechanisms that CARD9 plays in lung cancer.

CARD9 is a cytosolic adaptor in myeloid cells, has a critical role in inflammatory disorders, and provides a protective function against microbial pathogen, especially fungal infection. Recently, CARD9 polymorphisms are of interest, showing a positive correlation with the elevated risk of fungal infection, inflammatory bowel disease, and other autoimmune diseases. Mechanistically, CARD9 polymorphisms impair the activation of RelB, a subunit of non-canonical NF-κB, which lead to the reduced cytokine and chemokine production by innate immune cells. In addition, CARD9 polymorphisms show a defective neutrophil accumulation in infectious sites. Furthermore, CARD9 polymorphisms could alter the composition of the gut microbiome. In this review, we summarize the latest findings of CARD9 polymorphisms with respect to inflammatory diseases.

Abstract Transarterial chemoembolization (TACE) is the major treatment for advanced hepatocellular carcinoma (HCC), but it may cause hypoxic environment, leading to rapid progression after treatment. Here, using high‐throughput sequencing on different models, S100 calcium binding protein A9 (S100A9) is identified as a key oncogene involved in post‐TACE progression. Depletion or pharmacologic inhibition of S100A9 significantly dampens the growth and metastatic ability of HCC. Mechanistically, TACE induces S100A9 via hypoxia‐inducible factor 1α (HIF1A)‐mediated pathway. S100A9 acts as a scaffold recruiting ubiquitin specific peptidase 10 and phosphoglycerate mutase family member 5 (PGAM5) to form a tripolymer, causing the deubiquitination and stabilization of PGAM5, leading to mitochondrial fission and reactive oxygen species production, thereby promoting the growth and metastasis of HCC. Higher S100A9 level in HCC tissue or in serum predicts a worse outcome for HCC patients. Collectively, this study identifies S100A9 as a key driver for post‐TACE HCC progression. Targeting S100A9 may be a promising therapeutic strategy for HCC patients.

Brake emissions from vehicles are increasing as the number of vehicles increases. However, current research on brake emissions, particularly the intensity and characteristics of emissions under real road conditions, is significantly inadequate compared to exhaust emissions. To this end, a dataset of 600 (200 unique real-world braking events simulated using three types of brake pads) real-world braking events (called brake pad segments) was constructed and a mapping function between the average brake emission intensity of PM2.5 from the segments and the segment features was established by five algorithms (multiple linear regression (MLR) and four machine learning algorithms). Based on the five algorithms, the importance of the different features of the fragments was discussed and brake energy intensity (BEI) and metal content (MC) of the brake pad emissions were identified as the most significant factors affecting brake emissions and used as the final modeling features. Among the five algorithms, categorical boosting (CatBoost) had the best prediction performance, with a mean R2 and RMSE of 0.83 and 0.039 respectively for the tenfold cross-validation. In addition, the CatBoost-based model was further compared with the MOVES model to demonstrate its applicability. The CatBoost-based model has better prediction performance than the MOVES model. The MOVES model overpredicts brake fragment emissions for urban roads and underpredicts brake fragment emissions for motorways. Furthermore, the CatBoost-based model was interpreted and visualized by an individual conditional expectation (ICE) plot to break the machine learning “black box”, with BEI and MC showing nonlinear monotonic increasing relationships with braking emissions. ICE plot also provides viable technical solutions for controlling brake emissions in the future. Both avoiding aggressive braking driving behavior (e.g., the application of smart transportation technologies) and using brake pads with less metal content (e.g., using ceramic brake pads) can effectively reduce brake emissions. The construction of a machine learning-based brake emission model and the white-boxing of its model provide excellent insights for the future detailed assessment and control of brake emissions.

In implosion experiments, bremsstrahlung radiation ratios of broad-energy-band x-ray emission intensities (sampled by Ross pair) and narrow-energy-band x-ray emission intensities (sampled by multilayer) are typically used to extract the hotspot electron temperature. The latter method could potentially be more accurate because it does not require any additional theoretical arithmetic. However, the boundary conditions of the energy band, drastic influence on the measured electron temperature resulting from response differences of recording devices in the energy band, evident impact from uncertainties of the detector aiming, and coordinate interrelations for the two narrow-energy-band x-ray images have not been explored. These problems should be overcome to obtain the accurate hotspot electron temperature using the narrow-energy-band x-ray emission intensities method. This study solves the problems indicated above by exploring a diagnosis technique to extract the accurate hotspot electron temperature. In particular, we determine that the effect of the response differences and uncertainties could be ignored when the width of the sampled narrow energy band is approximately ±0.5 keV in the linear spectrum response regions of the imaging plate, and the reflectivity of the multilayer is uniform and constant in that energy band and the viewing field of the detector (≥±110 µm). This study is the first to consider the linear spectrum response of the imaging plate in different energy regions, eliminating the effect of the response differences. Finally, the maximal emission intensities in the two recorded-energy-band x-ray images can be used for coordinate interrelation.

BackgroundThyroid-associated ophthalmopathy (TAO) is one of the most common orbital diseases that seriously threatens visual function and significantly affects patients’ appearances, rendering them unable to work. This study established an intelligent diagnostic system for TAO based on facial images.MethodsPatient images and data were obtained from medical records of patients with TAO who visited Shanghai Changzheng Hospital from 2013 to 2018. Eyelid retraction, ocular dyskinesia, conjunctival congestion, and other signs were noted on the images. Patients were classified according to the types, stages, and grades of TAO based on the diagnostic criteria. The diagnostic system consisted of multiple task-specific models.ResultsThe intelligent diagnostic system accurately diagnosed TAO in three stages. The built-in models pre-processed the facial images and diagnosed multiple TAO signs, with average areas under the receiver operating characteristic curves exceeding 0.85 (F1 score >0.80).ConclusionThe intelligent diagnostic system introduced in this study accurately identified several common signs of TAO.

Abstract Background Elafin is a serine protease inhibitor critical for host defence. We previously reported that Elafin was associated with the recurrence of early-stage hepatocellular carcinoma (HCC) after surgery. However, the exact role of Elafin in HCC remains obscure. Methods HCC tissue microarrays were used to investigate the correlation between Elafin expression and the prognosis of HCC patients. In vitro migration, invasion and wound healing assays and in vivo lung metastasis models were used to determine the role of Elafin in HCC metastasis. Mass spectrometry, co-immunoprecipitation, western blotting, and immunofluorescence staining assays were performed to uncover the mechanism of Elafin in HCC. Dual-luciferase reporter and chromatin immunoprecipitation assays were employed to observe the transcriptional regulation of Elafin. Results Elafin expression was frequently increased in HCC tissues compared to normal tissues, and high Elafin expression in HCC tissues was correlated with aggressive tumour phenotypes and a poor prognosis in HCC patients. Elafin dramatically enhanced the metastasis of HCC cells both in vitro and in vivo by interacting with EGFR and activating EGFR/AKT signalling. Moreover, Elafin attenuated the suppressive effects of erlotinib on HCC metastasis. Besides, Elafin was transcriptionally regulated by Sp1 in HCC cells. Clinically, Elafin expression was positively correlated with Sp1, Vimentin, and EGFR signalling in both our HCC tissue microarrays and TCGA database analysis. Conclusions Upregulation of Elafin by Sp1 enhanced HCC metastasis via EGFR/AKT pathway, and overexpression of Elafin attenuated the anti-metastatic effects of erlotinib, suggesting a valuable prognostic biomarker and therapeutic target for HCC.

The aim of this study is to investigate the effects of seawater retention inside subsea oil and gas pipelines on the corrosion of several corrosion resistant alloys commonly used in seawater. Pitting corrosion, crevice corrosion, and electrochemical test samples were immersed in coastal waters at the depth of about 170 m in South China Sea (western Pacific Ocean). Assessments of the pitting and crevice corrosion for different alloys were carried out through electrochemical measurements and surface characterizations. The results showed that, the order of the pitting corrosion resistance of five alloys in marine environment was: 825 Ni-based alloy ∼ 2507 super duplex stainless steel > 2205 duplex stainless steel ∼ 317L ∼ 316L, while the crevice corrosion resistance was: 825 Ni-based alloy ∼ 2507 super duplex stainless steel > 2205 duplex stainless steel > 317L > 316L. The pitting initiations of 2205 duplex stainless steel, 2507 super duplex stainless steel, and 825 Ni-based alloy were later than those of 316L and 317L stainless steels. After the exposure for 592 d, the passivation for 316L, 317L and 2205 stainless steels were completely broken down while 2507 super duplex stainless steel and 825 Ni-based alloy still had a wide and stable passive zone. In crevice corrosion samples, the most severe corrosion was observed in the crevice edge area. A boat shaped corrosion pit was found in the inner coverage area far from the edge of crevice, which was probably related to the distribution of microorganisms in this area.

Objective: Cancer stem cell is one of the important causes of tumorigenesis as well as a drug target in the treatment of malignant tumor. However, at present, there is no immune vaccine targeting these cells. Octamer-binding transcription factor 4 (OCT4), a marker of embryonic stem cells and germ cells, often highly expresses in the early stages of tumorigenesis and is therefore a good candidate for cancer vaccine development. Methods: To identify the optimal carrier and adjuvant combination, we chemically synthesized and linked three different OCT4 epitope antigens to a carrier protein, keyhole limpet hemocyanin (KLH), combined with Toll-like receptor 9 agonist (TLR9). Results: Immunization with OCT4-3 + TLR9 produced the strongest immune response in mice. In prevention assays, significant tumor growth inhibition was achieved in BABL/c mice treated with OCT4-3 + TLR9 (P < 0.01). Importantly, the results showed that cytotoxic T lymphocyte activity and the inhibition of tumor growth were enhanced in mice immunized with OCT4-3 combined with TLR9. Meanwhile, multiple cytokines [such as interferon (IFN)-γ (P < 0.05), interleukin (IL)-12 (P < 0.05), IL-2 (P < 0.01), and IL-6 (P < 0.05)] promoting cellular immune responses were shown to be greatly enhanced in mice immunized with OCT4-3 + TLR9. Moreover, we considered safety considerations in terms of the composition of the vaccines to help facilitate the development of effective next-generation vaccines. Conclusions: Collectively, these experiments demonstrated that combination therapy with TLR9 agonist induced a tumor-specific adaptive immune response, leading to the suppression of primary tumor growth in testis embryonic carcinoma.

The transient simulation of CO2 and NOX from motor vehicles has essential applications in evaluating vehicular greenhouse gas emissions and pollutant emissions. However, accurately estimating vehicular transient emissions is challenging due to the heterogeneity between different vehicles and the continuous upgrading of vehicle exhaust purification technology. To accurately characterize the transient emissions of motor vehicles, a Super-learner model is used to build CO2 and NOx transient emission models. The actual onboard test data of 9 China VI N2 vehicles were used to train the model, and the test data of another China VI N2 vehicle were selected for further robustness verification. There were significant differences in the emissions between the vehicles, but the constructed transient model could capture the common law of transient emissions from China VI N2 vehicles. The R2 values of CO2 and NOx emission in the test data of the validation vehicle were 0.71 and 0.82, respectively. In addition, to further prove the model's robustness, the training data were synchronously modelled based on the Moves-method. The Super-learner model has a smaller RMSE on the validation set than the model based on the Moves-method, indicating that the Super-learner model has more transient simulation advantages. The marginal contributions of the model characteristics to the model results were analysed by SHapley Additive exPlanation (SHAP) value interpretation, and the marginal contributions of different pollutant characteristic parameters varied. Therefore, when establishing transient models of different pollutants, the selection of the model parameters demands considering the generation and purification process of different pollutants. The present work provides novel insights into the parameter selection, construction, and interpretation of the transient vehicle emission model.

The adaptor protein CARD9 plays an important role in anti-fungal immunity responses, linking detection of fungi by surface receptors to activation of the transcription factor nuclear factor κB (NF-κB). Recent studies indicate that CARD9 also plays different but vital roles during the development of colitis-associated colorectal cancer (CAC). This review summarizes the current understanding of CARD9 functions in CAC, and we discuss its potentially carcinogenic mechanisms. The adaptor protein Caspase Recruitment Domain Family Member 9 (CARD9) plays an indispensable role in innate immunity. Recent studies indicate that dysregulated CARD9 is a critical risk factor in the progression of colorectal cancer (CRC). This review provides novel insights into the functions of CARD9 in CRC, particularly in delineating its role in disrupting the host microbe balance, fueling gut microbiota metabolism and inducing systemic immunoglobulin G (IgG) antifungal antibodies. These pathways provide important information that can potentially be used for therapeutic innovation in developing potential vaccines for CRC.

Background: Thrombolysis in Myocardial Infarction (TIMI) Risk Index (TRI) is a simple risk assessment tool for patients with ST-segment elevation myocardial infarction (STEMI). However, its applicability to elderly patients with STEMI undergoing percutaneous coronary intervention (PCI) is uncertain.Methods: This was a retrospective analysis of elderly (≥60 years) patients who underwent PCI for STEMI from January 2010 to April 2016. TRI was calculated on admission using the following formula: heart rate × (age/10)2/systolic blood pressure. Discrimination and calibration of TRI for in-hospital events and 1 year mortality were analyzed.Results: Totally 1,054 patients were divided into three groups according to the tertiles of the TRI: 36 (n = 346). The incidence of acute kidney injury (AKI; 7.8 vs. 8.6 vs. 24.0%, p < 0.001), AHF (3.5 vs. 6.6 vs. 16.2%, p < 0.001), in-hospital death (0.6 vs. 3.3 vs. 11.6%, p < 0.001) and MACEs (5.2 vs. 5.8 vs. 15.9%, p < 0.001) was significantly higher in the third tertile. TRI showed good discrimination for in-hospital death [area under the curve (AUC) = 0.804, p < 0.001; Hosmer-Lemeshow p = 0.302], which was superior to its prediction for AKI (AUC = 0.678, p < 0.001; Hosmer-Lemeshow p = 0.121), and in-hospital MACEs (AUC = 0.669, p < 0.001; Hosmer-Lemeshow p = 0.077). Receiver-operation characteristics curve showed that TRI > 42.0 had a sensitivity of 64.8% and specificity of 82.2% for predicting in-hospital death. Kaplan-Meier analysis showed that patients with TRI > 42.0 had higher 1 year mortality (Log-rank = 79.2, p < 0.001).Conclusion: TRI is suitable for risk stratification in elderly patients with STEMI undergoing PCI, and is thus of continuing value for an aging population.

Gesture recognition technology is widely used in the flexible and precise control of manipulators in the assisted medical field. Our MResLSTM algorithm can effectively perform dynamic gesture recognition. The result of surface EMG signal decoding is applied to the controller, which can improve the fluency of artificial hand control. Much current gesture recognition research using sEMG has focused on static gestures. In addition, the accuracy of recognition depends on the extraction and selection of features. However, Static gesture research cannot meet the requirements of natural human-computer interaction and dexterous control of manipulators. Therefore, a multi-stream residual network (MResLSTM) is proposed for dynamic hand movement recognition. This study aims to improve the accuracy and stability of dynamic gesture recognition. Simultaneously, it can also advance the research on the smooth control of the Manipulator. We combine the residual model and the convolutional short-term memory model into a unified framework. The architecture extracts spatiotemporal features from two aspects: global and deep, and combines feature fusion to retain essential information. The strategy of pointwise group convolution and channel shuffle is used to reduce the number of network calculations. A dataset is constructed containing six dynamic gestures for model training. The experimental results show that on the same recognition model, the gesture recognition effect of fusion of sEMG signal and acceleration signal is better than that of only using sEMG signal. The proposed approach obtains competitive performance on our dataset with the recognition accuracies of 93.52%, achieving state-of-the-art performance with 89.65% precision on the Ninapro DB1 dataset. Our bionic calculation method is applied to the controller, which can realize the continuity of human-computer interaction and the flexibility of manipulator control.

Urea regulators (UR) have generally been employed against diesel trucks to save urea usage and thus contribute to the reduction in excessive emissions, while their usage is generally difficult to supervise and enforce. By conducting real driving emission measurements on a China IV heavy-duty diesel truck, a “trade-off” effect caused by UR was found between nitrogen oxides (NOx) and particle number (PN) emissions. The usage of UR contributes to 1.04 times higher NOx but 0.28 times lower PN emissions for the whole trip. In particular, the increasing effects on NOx are most efficient on the highway and least effectual on the urban road, while the decreasing effects on PN exhibit an opposite trend under different road types. From low- and medium- to the high-speed bin, the peak average vehicle-specific power NOx emission rates exhibit markedly increasing but slightly decreasing trends for the truck with and without UR, respectively. Furthermore, the NOx emissions in units of CO2 and the linear correlational relationship between CO2 and NOx instantaneous mass emission rates, especially those on the highway, are significantly enhanced. This study directly clarifies the effects of UR on real-world emissions, providing a scientific basis for the real-time identification of the malfunction of the selective catalytic reduction system.

Reflected appraisals refer to the perceptions of individuals of how they are perceived by others. Numerous studies in cultural psychology have revealed that individuals in the Eastern collectivist culture show an interdependent self-construal, which depends much on the social culture. Hence, the research on reflected appraisals in the Eastern culture can improve the understanding of how the social environment shapes the self-perception of an individual. In this study, we aimed to explore the relationships among self-appraisals, reflected appraisals, and actual appraisals of peers of the Big Five personality for Chinese late adolescents. Participants were divided into 16 groups, with two to four people of each group who were familiar with each other. Each participant was told to fill out the questionnaires of reflected appraisals, actual appraisals of peers, and self-appraisals. Through analyzing 164 sets of data, the results showed the following: (a) The scores of reflected appraisals are significantly lower than that of the actual appraisals of peers. (b) The relationships among the reflected appraisals, actual appraisals of peers, and self-appraisals are distinct on different personalities. For extroversion, there are significant medium- to high-degree relationships among the three types of appraisals; while for the agreeableness, conscientiousness, emotional stability, and openness, self-appraisals are highly correlated with reflected appraisals, and reflected appraisals show a low-degree correlation with the actual appraisals of peers. (c) Reflected appraisals play a mediating role between actual appraisals of peers and self-appraisals. Our study suggests that individuals in Chinese culture generally underestimate how their peers perceive them. Furthermore, actual appraisals of peers affect the self-concepts of individuals through reflected appraisals. This study revealed the unique personality feature of self-modesty under the background of Chinese culture and the importance of peers on the development of self-concepts for Chinese late adolescents. This study can shed new light on the understanding of the development of self-concepts for late adolescents under different cultural backgrounds.

Abstract Background Gender information frequently exists in the eligibility criteria of clinical trial text as essential information for participant population recruitment. Particularly, current eligibility criteria text contains the incompleteness and ambiguity issues in expressing transgender population, leading to difficulties or even failure of transgender population recruitment in clinical trial studies. Methods A new gender model is proposed for providing comprehensive transgender requirement specification. In addition, an automated approach is developed to extract and summarize gender requirements from unstructured text in accordance with the gender model. This approach consists of: 1) the feature extraction module, and 2) the feature summarization module. The first module identifies and extracts gender features using heuristic rules and automatically-generated patterns. The second module summarizes gender requirements by relation inference. Results Based on 100,134 clinical trials from ClinicalTrials.gov, our approach was compared with 20 commonly applied machine learning methods. It achieved a macro-averaged precision of 0.885, a macro-averaged recall of 0.871 and a macro-averaged F1-measure of 0.878. The results illustrated that our approach outperformed all baseline methods in terms of both commonly used metrics and macro-averaged metrics. Conclusions This study presented a new gender model aiming for specifying the transgender requirement more precisely. We also proposed an approach for gender information extraction and summarization from unstructured clinical text to enhance transgender-related clinical trial population recruitment. The experiment results demonstrated that the approach was effective in transgender criteria extraction and summarization.

Increased applications of quantum dots (QDs) in the biomedical field have aroused attention for their potential toxicological effects. Although numerous studies have been carried out on the toxicity of QDs, their effects on reproductive and development are still unclear. In this study, we systematically evaluated the male reproductive toxicity and developmental toxicity of CdSe/ZnS QDs in BALB/c mice. The male mice were injected intravenously with CdSe/ZnS QDs at the dosage of 2.5 mg/kg BW or 25 mg/kg BW, respectively, and the survival status, biodistribution of QDs in testes, serum sex hormone levels, histopathology, sperm motility and acrosome integrity was measured on Day 1, 7, 14, 28 and 42 after injection. On Day 35 after treatment, male mice were housed with non-exposed female mice, and then offspring number, body weight, organ index and histopathology of major organs, blood routine and biochemical tests of offspring were measured to evaluate the fertility and offspring health. The results showed that CdSe/ZnS QDs could rapidly distribute in the testis, and the fluorescence of QDs could still be detected on Day 42 post-injection. QDs had no adverse effect on the structure of testis and epididymis, but high-dose QDs could induce apoptosis of Leydig cells in testis at an early stage. No significant differences in survival of state, body weight organ index of testis and epididymis, sex hormones levels, sperm quality, sperm acrosome integrity and fertility of male mice were observed in QDs exposed groups. However, the development of offspring was obviously influenced, which was mainly manifested in the slow growth of offspring, changes in organ index of main organs, and the abnormality of liver and kidney function parameters. Our findings revealed that CdSe/ZnS QDs were able to cross the blood-testis barrier (BTB), produce no discernible toxic effects on the male reproductive system, but could affect the healthy growth of future generations to some extent. In view of the broad application prospect of QDs in biomedical fields, our findings might provide insight into the biological safety evaluation of the reproductive health of QDs.

The widespread application of cadmium-free CuInS2/ZnS QDs has raised great concern regarding their potential toxicity to humans. To date, toxicological data related to CuInS2/ZnS QDs are scarce. Neurons play extraordinary roles in regulating the activities of organs and systems, and serious consequences occur when neurons are damaged. Currently, the potential toxicity of CuInS2/ZnS QDs on neurons has not been fully elucidated. Here, we investigate the neurotoxicity of PEGylated CuInS2/ZnS (CuInS2/ZnS-PEG) QDs on neuron-like PC12 cells. We found that CuInS2/ZnS-PEG QDs were taken up by PC12 cells, but at a concentration range from 0 to 100 μg/mL, they did not affect the survival rate of the PC12 cells. In addition, we found that CuInS2/ZnS-PEG QDs significantly inhibited neurite outgrowth from and the differentiation of PC12 cells in the presence of NGF, while COOH-modified CuInS2/ZnS QDs or free PEG did not have a similar effect. Further studies showed that CuInS2/ZnS-PEG QDs obviously downregulated the expression of low-affinity NGF receptor (p75NTR) and subsequently negatively regulated the downstream MAPK cascade by dephosphorylating ERK1/2 and AKT. Taken together, these results suggest that CuInS2/ZnS-PEG QDs disturb NGF signal transduction from external stimuli to relevant internal signals, thus affecting normal biological processes such as neurite outgrowth and cell differentiation.

Neutrophils play a critical role in innate immune defense and directly contribute to infectious and autoimmune ailments. Great efforts are underway to better understand the nature of neutrophilic inflammation. Of note, CARD9, a myeloid cell-specific signaling protein that mainly expresses in macrophages and dendritic cells, is also present in neutrophils, emerging as a critical mediator for intercellular communication. CARD9–deficiency neutrophils display an increased susceptibility to fungal infection that primarily localize to the central nervous system, subcutaneous, and skin tissue. Additionally, CARD9–deficiency neutrophils are associated with some autoimmune diseases and even provide protection against a few bacteria. Here, the review summarizes recent preclinical and clinical advances that have provided a novel insight into the pathogenesis of CARD9 deficiency in neutrophils.

Chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) is a genitourinary disease commonly seen in males, with symptoms involving pelvic pain, urinary system disease, and sexual dysfunction, which seriously affects physical and mental health, and it also influences the quality of life of patients. At present, the disease’s aetiology and pathogenesis are unclear, and there is also no effective treatment for it. Acupuncture and moxibustion have been a way to CP/CPPS, showing good curative effect with advantages of safety and affordability. However, the relevant research in this field is less discussed. By adopting databases, such as CNKI, VIP, Wanfang, PubMed, and Medline, this review article used keywords including chronic prostatitis, chronic pelvic pain syndrome, and electric acupuncture, manual acupuncture, moxibustion, and animal experiments, rats, mice, and mechanism research and reviewing research papers published from 1998 to 2021. Then, it further summarized and evaluated the mechanism research and gave a brief comment about modeling methods, acupoints selection, and stimulus parameters that have been used in the selected research papers. Equally important, this review article proposes a reference for the in-depth study of the mechanism of acupuncture and moxibustion on CP/CPPS and provides a theoretical basis to better treat the disease in the clinic.

Based on the demand of vehicle emission research and control, this paper presents the development of a portable vehicle measurement system (PEMS) based on SEMTECH-DS and ELPI+, the vehicle emission tests carried out on actual roads, and the data obtained for the establishment and validation of a vehicle emission model. Based on the results of the vehicle emission test, it was found that vehicle driving conditions (speed, acceleration, vehicle specific power (VSP), etc.) had a significant impact on the pollutant emission rate. In addition, local driving cycles were generated and the frequency distribution of VSP-bin under different cycles was analyzed. Then, through the establishment of an emission rate database, calculation of emission factors and validation of the emission model, a vehicle emission model based on actual road driving conditions was developed by taking VSP as the “surrogate variables”. It showed that the emission factor model established in this study could better reflect the vehicle transient emissions on the actual road with high accuracy and local adaptability. Through this study, it could be found that due to the great differences in traffic development modes and vehicle driving conditions in different cities in China, the emission model based on driving conditions was a better choice to carry out the research on vehicle emission in Chinese cities. Compared with directly applying international models or quoting the recommended values of relevant macroscopic guidelines, the emission factor model established in this study, using actual driving conditions, could better reflect the vehicle transient emissions on the actual road with high accuracy and local adaptability. In addition, due to the rapid development of China’s urban traffic and the rapid change of driving conditions, it was of great significance to regularly update China’s urban conditions to improve the accuracy of the model, no matter which model was chosen.

Jack-up offshore platform is a type of important marine structure, which is mainly used for satellite launch, oil exploitation, and other engineering tasks in the offshore area. The offshore platform is bound to be subjected to wave loading in the course of use. Whether it can withstand the wave impact is an important engineering problem. To solve this engineering problem, the self-developed fluid–structure–foundation interaction coupling model OlaFlow-ABAQUS is used to explore the dynamic response characteristics of a jack-up offshore platform and its seabed foundation under three conventional wave conditions (wave height is 3, 5, and 7 m, respectively) in a coupled way. The numerical results show that only a small amplitude of periodic sloshing occurs for the jack-up offshore platform under the three conventional wave conditions. The maximum sloshing amplitude is up to 8 cm, and there is no visible residual displacement. It is indicated that there is no plastic deformation zone in the seabed foundation near the pile legs of the jack-up platform. It can thus be concluded that the jack-up platform has excellent stability under conventional wave conditions. Under conventional wave loading, momentary liquefaction occurs in the seabed foundation around the pile legs of the platform, and the maximum liquefaction depth is about 1 m. This study indicates that the coupling model OlaFlow-ABAQUS for the fluid–structure–foundation interaction is feasible, and has some advantages to study the dynamic response and to evaluate the stability of large-scale marine structures and their seabed foundations under ocean waves.

Graphene, known as “black gold”, has important applications in various fields. In previous studies, it has been proved that graphene oxide (GO) which is a derivative of graphene has low toxicity. However, the immunotoxicity of GO has not been fully elucidated. In this work, we used DC2.4 cell line to investigate the in vitro immunotoxicity of two types of GO, mono-layer GO (mono-GO) and multi-layer GO (multi- GO). We found that mono-GO had less effect on cell viability than multi-GO, but both mono-GO and multi-GO significantly induced the generation of ROS in DC2.4 cells. Interestingly, mono-GO caused DC2.4 cells to aggregate, thus changed the cell morphology significantly. However, no similar influence occurred for multi-GO. In addition, the results showed that these two GOs obviously enhance the release of TNF-α by DC2.4 cells with and without LPS stimulation. GO did not affect the level of IL-6 released from DC2.4 cells, but multi-GO promoted the release of IL-6 while mono-GO inhibited the production of IL-6 when cells were in response to LPS stimulation. Whole-transcriptome sequencing analysis found some immune-related differentially expressed genes including H2-DMb1, Ncbp3, Oas2, Men1, Fas, Cd320, Cd244, and Tinagl1 which are engaged in the immune system process. These results suggested that both mono-GO and multi-GO are immunotoxic to DC2.4 cells, which provides important basis for subsequent biological and clinical medical applications.

Deep rock mass theory has not yet been completely established, which leads to a lack of theoretical guidance for deep resource development and poor continuity among engineering activities. The foundation of deep rock mechanics theory is to achieve the deep in situ rock fidelity coring (including the retaining of the pore pressure and temperature). To realize this, pressure-retaining coring technology is required. A self-triggered pressure-retaining controller based on magnetic control is proposed in this paper. The pressure-retaining controller realizes pressure-retaining coring in any direction by triggering the closure of the valve cover by a magnetic force, forming a magnetic seal. Fifteen combined magnetic circuit design schemes are proposed. The magnetic flux density norm distribution and magnetic force evolution law of different schemes are then quantitatively analyzed by the finite element method. The results show that a complex magnetization combination can weaken the nonlinear negative correlation between the magnetic force and distance. The optimal design of the valve seat magnetic circuit is Scheme 9, with the valve seat consisting of four shape identical tile magnets. Among the schemes, for Scheme 9, the magnetic flux density norm of the valve cover is the most concentrated. The maximum magnetic flux density norm is in the middle, and the magnetic force at 35 mm from the valve cover to the valve seat is 2.915 N. Scheme 9 satisfies the minimum condition of the mechanical model and verifies the feasibility of magnetic field triggering. This research can be used to gain a better understanding of deep Earth properties and provides technology for the improved design of deep in situ pressure-retaining coring devices.

Abstract Background The optimal follow-up strategy after curative thermal ablation of hepatocellular carcinoma (HCC) remains unclear. Methods We retrospectively analyzed a prospective series of 616 patients who underwent curative thermal ablation for HCC within the Milan criteria. Multivariate Cox model was used to identify independent predictive factors for recurrence; accordingly, patients were stratified into 2 groups with different relapse risks: a low-risk group (solitary tumor ≤3 cm) and a high-risk group (multiple tumors ≤3 cm or solitary tumor between 3 and 5 cm). Abstract Background Average postoperative follow-up intervals vary in patients undergoing hepatocellular carcinoma (HCC) resection because of limited evidence regarding the optimal interval. We aimed to compare the associations of long-versus short-interval follow-up with survival and recurrence in risk-stratified HCC patients. Methods We performed a retrospective cohort study between 2007 and 2014. In total, 1227 patients treated by curative resection of Barcelona Clinic Liver Cancer stage A or B HCC were stratified as having a low (n = 865) or high (n = 362) risk of early recurrence (within the first 2 years after resection) based on prognostic factors identified by the least absolute shrinkage and selection operation algorithm. Patients were further classified into long-interval (every 4–6 months) and short-interval (every 2–4 months) follow-up subgroups based on follow-up within 2 years after resection (low risk, long vs. short: n = 390 vs. n = 475; high-risk, long vs. short: n = 149 vs. n = 213). Results The short-interval follow-up did not prolong overall survival in either the low-risk (hazard ratio [HR] = 1.152; 95% confidence interval [CI] 0.720–1.843) or high-risk (HR = 1.213; 95% CI 0.702–2.094) patients. Early recurrence occurred in 401 patients. For high-risk patients, the short-interval follow-up subgroup exhibited smaller intrahepatic recurrence than did the long-interval group (2.6 vs. 3.5 cm, respectively, P = 0.045). However, no significant difference in the rate of Barcelona Clinic Liver Cancer stage 0/A recurrence was found between the long- and short-interval follow-up groups in either low- or high-risk patients (63.1% vs. 68.2%, respectively, P = 0.580; 31.3% vs. 41.5%, respectively, P = 0.280). The rate of curative intent treatment for recurrence (34.5% vs. 39.7%, respectively, P = 0.430; 14.6% vs. 20.3%, respectively, P = 0.388) was also similar between the follow-up groups for low- and high-risk patients. Conclusions Shortening the postoperative follow-up interval from every 4–6 months to every 2–4 months within the first 2 years after resection did not increase the rate of curative intent treatment or prolong the overall survival of patients with Barcelona Clinic Liver Cancer stage A or B HCC.

Tumor-associated macrophages (TAMs) are one of the most abundant immune cells in the tumor microenvironment, and they play a pivotal role in prompting the various tumor growth. However, the role of TAMs in colorectal carcinoma (CRC) is controversial, because a few papers report that TAMs is beneficial to CRC patients. In this review, we discuss the good or bad roles of TAMs in CRC progression. Interestingly, recent studies provide strong evidence that TAMs facilitate CRC growth, but do not exert tumor suppressive activities. TAMs can stimulate CRC growth by altering extracellular matrix remodeling, tumor metabolism, angiogenesis, as well as the tumor microenvironment. Therefore, TAMs could serve as a target for CRC therapeutic treatment.

We present our recent laser-plasmas instability (LPI) comparison experiment at the SGIII laser facility between the spherical and cylindrical hohlraums. Three kinds of filling are considered: vacuum, gas-filling with or without a capsule inside. A spherical hohlraum of 3.6 mm in diameter, and a cylindrical hohlraum of 2.4 mm × 4.3 mm are used. The capsule diameter is 0.96 mm. A flat-top laser pulse with 3 ns duration and up to 92.73 kJ energy is used. The experiment has shown that the LPI level in the spherical hohlraum is close to that of the outer beam in the cylindrical hohlraum, while much lower than that of the inner beam. The experiment is further simulated by using our 2-dimensional radiation hydrodynamic code LARED-Integration, and the laser back-scattering fraction and the stimulated Raman scatter (SRS) spectrum are post-processed by the high efficiency code of laser interaction with plasmas HLIP. According to the simulation, the plasma waves are strongly damped and the SRS is mainly developed at the plasma conditions of electron density from 0.08 nc to 0.1 nc and electron temperature from 1.5 keV to 2.0 keV inside the hohlraums. However, obvious differences between the simulation and experiment are found, such as that the SRS back-scattering is underestimated, and the numerical SRS spectrum peaks at a larger wavelength and at a later time than the data. These differences indicate that the development of a 3D radiation hydrodynamic code, with more accurate physics models, is mandatory for spherical hohlraum study.
