Abstract Background Atropine, specifically 0.05% eyedrops, has proven effective in slowing myopia progression. This study aims to investigate peripheral refraction (PR) characteristics in myopic children treated with 0.05% atropine eyedrops at different frequencies. Methods One hundred thirty-eight myopic children completed this one-year prospective study, randomly assigned to once daily (7/7), twice per week (2/7), or once per week (1/7) groups. Spherical equivalent (SE) and axial length (AL) were measured. PR was assessed using a custom-made Hartmann-Shack wavefront peripheral sensor, covering a visual field of horizontal 60° and vertical 36°. Relative peripheral refraction (RPR) was calculated by subtracting central from peripheral measurements. Results After one year, SE increased more significantly in the 1/7 group compared to the 7/7 group (P

The work in this paper incorporated printed circuit board (PCB) technology into micro-direct methanol fuel cells (µDMFCs) and conjectured and verified the performance degradation factors of PCB current collectors in µDMFCs by testing different designed configuration µDMFCs. The experiment results showed that all kinds of PCB coating can benefit from the porous stainless-steel plates covering to a great extent. At the end of 48 h discharging, µDMFCs with porous stainless-steel plates between MEA and PCB coating achieved higher performance than those of the direct contacting series. It can be inferred from various types of experimental data that because of stainless-steel porous plate isolating, the impact of corrosion on the surface of the PCB electrode plate was reduced to a certain extent. The corrosion of the electrode plate was slowed down in the µDMFC discharging as a result of the passivation behavior on the iron surface and a decrease in corrosion current. Consequently, the attenuation of the PCB performance was delayed. The conclusion of this work explores a practical direction to enhance the cost-effectiveness of fuel cells, promoting the large-scale application of DMFCs in the future.

Abstract In contrast to rodents, the mechanisms underlying human trophectoderm and early placenta specification are understudied due to ethical barriers and the scarcity of embryos. Recent reports have shown that human pluripotent stem cells (PSCs) can differentiate into trophectoderm (TE)-like cells (TELCs) and trophoblast stem cells (TSCs), offering a valuable in vitro model to study early placenta specification. Here, we demonstrate that the VGLL1 (vestigial-like family member 1), which is highly expressed during human and non-human primate TE specification in vivo but is negligibly expressed in mouse, is a critical regulator of cell fate determination and self-renewal in human TELCs and TSCs derived from naïve PSCs. Mechanistically, VGLL1 partners with the transcription factor TEAD4 (TEA domain transcription factor 4) to regulate chromatin accessibility at target gene loci through histone acetylation and acts in cooperation with GATA3 and TFAP2C. Our work is relevant to understand primate early embryogenesis and how it differs from other mammalian species.

With the rapid development of smart cities and the swift transition toward digital governance, optimizing urban spatial governance through digital technology remains underexplored in the Global South, particularly from the perspective of resident perception and interaction. Digitization of government services is a key area of interest in digital governance research; this study investigates the impact of government self-service systems on the spatial perception and behavior of residents in Guangzhou and Foshan, China. Through a mixed-method approach, combining questionnaire surveys and semi-structured interviews and analyzing them using a structural equation model, the findings reveal that government self-service systems significantly influence residents’ spatial behavior and perception. These systems enhance the efficiency of administrative processes, increase convenience, and lead to temporal-spatial compression, thereby reshaping residents’ physical interactions with urban spaces. The findings provide practical insights for policymakers to enhance urban governance by integrating digital technologies to improve residents’ interaction with government services. These insights can guide the development of more efficient, resident-centered digital governance frameworks, particularly in rapidly urbanizing regions. This research contributes to a deeper understanding of how digital technology transforms urban spatial governance, highlighting the critical interplay between individuals, technology, and the urban environment. The study likewise provides examples of the ongoing digital transformation of public services in countries of the Global South that are lagging behind in the area of digital governance.

Abstract Background Shoot branching of flowering plants exhibits phenotypic plasticity and variability. This plasticity is determined by the activity of axillary meristems, which in turn is influenced by endogenous and exogenous cues such as nutrients and light. In many species, not all buds on the main shoot develop into branches despite favorable growing conditions. In petunia, basal axillary buds (buds 1–3) typically do not grow out to form branches, while more apical axillary buds (buds 6 and 7) are competent to grow. Results The genetic regulation of buds was explored using transcriptome analyses of petunia axillary buds at different positions on the main stem. To suppress or promote bud outgrowth, we grew the plants in media with differing phosphate (P) levels. Using RNA-seq, we found many (> 5000) differentially expressed genes between bud 6 or 7, and bud 2. In addition, more genes were differentially expressed when we transferred the plants from low P to high P medium, compared with shifting from high P to low P medium. Buds 6 and 7 had increased transcript abundance of cytokinin and auxin-related genes, whereas the basal non-growing buds (bud 2 and to a lesser extent bud 3) had higher expression of strigolactone, abscisic acid, and dormancy-related genes, suggesting the outgrowth of these basal buds was actively suppressed. Consistent with this, the expression of ABA associated genes decreased significantly in apical buds after stimulating growth by switching the medium from low P to high P. Furthermore, comparisons between our data and transcriptome data from other species suggest that the suppression of outgrowth of bud 2 was correlated with a limited supply of carbon to these axillary buds. Candidate genes that might repress bud outgrowth were identified by co-expression analysis. Conclusions Plants need to balance growth of axillary buds into branches to fit with available resources while allowing some buds to remain dormant to grow after the loss of plant parts or in response to a change in environmental conditions. Here we demonstrate that different buds on the same plant with different developmental potentials have quite different transcriptome profiles.

Abstract Introduction The purpose of this study was to investigate the characteristics of objective ocular surface parameters using non-invasive objective instruments in children with myopia who are candidates for orthokeratology lens wear. Methods Children with myopia who are candidates for orthokeratology lens wear were retrospectively investigated. The subjects were divided into three age groups. The Keratograph 5M and LipiView interferometry were used to assess non-invasive tear meniscus height (TMH), non-invasive tear film break-up time (NIBUT), conjunctival hyperemia redness score (RS), meibomian gland loss (MGL) score, lipid layer thickness (LLT), and blink pattern analysis, including the number of partial blinks (PB) and total blinks (TB), as well as the partial blink rate (PBR). Results A total of 1119 children with myopia (2070 eyes) aged 7–18 years were selected. The mean TMH, NIBUT, and LLT of the subjects was 0.21 mm, 12.45 s, and 65.28 nm, respectively. The mean RS and upper and lower MGL scores were 0.64, 1.00, and 1.06, respectively. The mean PB, TB, and PBR was 5.13, 6.46, and 0.81, respectively. Age was significantly correlated to all ocular surface parameters (p = 0.00), except for PB. NIBUT and LLT did not differ between male participants and female participants (all p > 0.05). TMH, RS, and upper and lower MGL were significantly higher in male participants than in female participants (all p 0.05). In addition, PBR was positively correlated with NIBUT and LLT (all p = 0.00) but not with TMH, RS, or MGL (all p > 0.05). Overall, 57.00% had a TMH ≤ 0.2 mm, 43.20% had a NIBUT ≤ 10 s, 48.10% had an LLT ≤ 60 nm, and 88.10% had a PBR > 0.4. Conclusions Child orthokeratology candidates have enhanced tear secretion and increased meibomian gland deficiency with aging. In addition, the adult dry eye diagnostic criteria may apply to orthokeratology candidates aged 12–18 years but should be lower for younger candidates. Given the proportion of abnormal risk, it is necessary to assess tear film status and blink pattern by reliable and feasible objective examination before fitting orthokeratology.

Abstract Given the challenge of pear leaf disease recognition caused by uneven illumination, overlapping leaves, and other green plants in the background, a two‐stage strategy‐based framework for pear leaf segmentation and disease classification is proposed. Initially, a double branch polymerization net fusing features of the low‐level feature branch and semantic branch is constructed to extract the target diseased pear leaf and eliminate background interference. Then an improved lightweight neural network (Inverted‐Inception Efficient‐Excitation‐and‐Filtering‐Bottleneck MobileNet‐v2, I2EMv2) is used to capture multi‐scale lesion information and ainhibit invalid feature channels and filter redundant features for the final classification. The experimental results show that the proposed framework can accurately extract the pear leaf region with complete boundary from the complex background, maximize the retention of lesion information, and achieve high‐precision pear leaf disease identification. The mean absolute error, F‐measure, and intersection over union of leaf segmentation are 0.027, 0.947, and 0.880, respectively, and the average recognition accuracy of leaf disease is 92.05%. Compared with others, the method proposed in this paper has superior performance on segmentation and classification, which provides a reference for pear leaf disease classification under complex background.

The action of the petunia strigolactone (SL) hormone receptor DAD2 is dependent not only on its interaction with the PhMAX2A and PhD53A proteins, but also on its expression patterns within the plant. Previously, in a yeast-2-hybrid system, we showed that a series of a single and double amino acid mutants of DAD2 had altered interactions with these binding partners. In this study, we tested the mutants in two plant systems, Arabidopsis and petunia. Testing in Arabidopsis was enabled by creating a CRISPR-Cas9 knockout mutant of the Arabidopsis strigolactone receptor (AtD14). We produced SL receptor activity in both systems using wild type and mutant genes; however, the mutants had functions largely indistinguishable from those of the wild type. The expression of the wild type DAD2 from the CaMV 35S promoter in dad2 petunia produced plants neither quite like the dad2 mutant nor the V26 wild type. These plants had greater height and leaf size although branch number and the plant shape remained more like those of the mutant. These traits may be valuable in the context of a restricted area growing system such as controlled environment agriculture.

PurposeThis aim of this study was to evaluate the effect of 3% Diquafosol Ophthalmic Solution (DQS) on children with dry eye from wearing overnight orthokeratology (OrthoK) lenses.MethodsMyopic children aged 8–18 years with dry eye syndrome were enrolled in this prospective observational study, and they were grouped according to their OrthoK treatment history for at least 1 year. All participants received DQS 4 times per day for 1 month. The following indicators were measured at baseline 1 month after treatment: the Dry Eye Questionnaire-5 (DEQ-5), non-invasive tear meniscus height (TMH), non-invasive tear film break-up time (first and average, NIBUT-F and NIBUT-A), meibomian gland score (MG score), conjunctival hyperemia redness score (R-scan), and blink pattern analysis.ResultsA total of 104 participants (189 eyes) including 40 OrthoK wearers (72 eyes) and 64 Orthok candidates (117 eyes) completed the study. Of all, after DQS treatment for 1 month, DEQ-5 scores reduced from 5.54 ± 3.25 to 3.85 ± 2.98 (t = −3.36, p = 0.00). TMH increased from 0.20 ± 0.05 mm to 0.21 ± 0.05 mm (t = 2.59, p = 0.01), NIBUT-F and NIBUT-A were prolonged from 6.67 ± 4.71 s to 10.32 ± 6.19 s and from 8.86 ± 5.25 s to 13.30 ± 6.03 s (all p = 0.00), respectively. R-scan decreased from 0.69 ± 0.28 to 0.50 ± 0.25 (t = −9.01, p = 0.00). Upper MG scores decreased from 1.04 ± 0.32 to 0.97 ± 0.36 (t = −2.14, p = 0.03). Lower MG scores, partial blink rate, partial blinks, and total blinks did not change significantly. Both break-up time (BUT) and R-scan improved significantly after DQS treatment for 1 month (all p = 0.00) in OrthoK candidates and OrthoK wearers. Among the OrthoK wearers, TMH and dry eye symptoms increased significantly (all p = 0.00) but did not increase in OrthoK candidates (p > 0.05). There were no adverse events related to DQS.ConclusionDiquafosol Ophthalmic Solution was effective for children wearing overnight orthokeratology in relieving dry eye symptoms and improving ocular surface parameters, which may help improve children's OrthoK wearing tolerance and compliance.

Inverse optimal control is a method for recovering the cost function used in an optimal control problem in expert demonstrations. Most studies on inverse optimal control have focused on building the unknown cost function through the linear combination of given features with unknown cost weights, which are generally considered to be constant. However, in many real-world applications, the cost weights may vary over time. In this study, we propose an adaptive online inverse optimal control approach based on a neural-network approximation to address the challenge of recovering time-varying cost weights. We conduct a well-posedness analysis of the problem and suggest a condition for the adaptive goal, under which the weights of the neural network generated to achieve this adaptive goal are unique to the corresponding inverse optimal control problem. Furthermore, we propose an updating law for the weights of the neural network to ensure the stability of the convergence of the solutions. Finally, simulation results for an example linear system are presented to demonstrate the effectiveness of the proposed strategy. The proposed method is applicable to a wide range of problems requiring real-time inverse optimal control calculations.

To examine the three-dimensional texture structure of SMA-13 asphalt pavement and assess its anti-skid performance, a light gradient-boosting machine evaluation model was developed using non-contact three-dimensional laser-scanning technology. The study focused on collecting three-dimensional texture data from newly laid SMA-13 asphalt pavement. Subsequently, wavelet transform was employed to reconstruct the pavement’s three-dimensional texture, and discrete Fourier transform was utilized to separate macro- and microtextures, enabling the calculation of their characteristics. The macro- and micro-characteristics of the three-dimensional texture and friction coefficient were input into the model. A comparative analysis with linear regression and a random forest model revealed superior accuracy and efficiency in the model. The training set R2 is 0.948, and the testing set R2 is 0.842, effectively enabling the evaluation of pavement anti-skid performance. An analysis of parameter importance indicated that Rku and MPD are still effective indicators for evaluating skid resistance. Furthermore, diverse texture indexes exhibited varying effects on the anti-skid performance. The established asphalt pavement anti-skid evaluation model serves as a theoretical foundation for understanding the actual influence on pavement anti-skid performance.

The optical generation of pulsed ultrasound is attractive to nondestructive testing and biological imaging, especially for those involving narrow operation space or strong electro-magnetic interference. However, conventional techniques based on the photoacoustic effect inevitably required an expensive high-energy short pulsed laser and dedicated preparation of the optically absorptive composite film. Here, a fiber-optic ultrasound pulse transmitter based on continuous-wave (CW) laser triggered thermo-cavitation was demonstrated. The fiber-delivered CW laser light heated the highly-absorptive copper nitrate solution and generated explosive bubbles , which emitted strong ultrasound waves. Omnidirectional ultrasound pulses with an amplitude up to 0.3 MPa and a repetition rate of 5 kHz in the frequency range of 5–12 MHz were obtained by using a 50 mW optical heating power at a wavelength of 980 nm. The fiber-tip ultrasound transmitter was integrated with a polymer-cavity-based fiber ultrasound detector to construct an all-fiber ultrasound endoscopic imaging probe. Without the need for a wavelength-tunable laser, the ultrasound detector was interrogated by CW laser light with a fixed wavelength, coupled with feedback-controlled heating of the cavity to stabilize its spectral fringe. The CW laser-driven fiber ultrasound transmitter, in combination with the photothermally stabilized fiber ultrasound detector, opens new routes for a number of ultrasound-related industrial and biomedical applications.

ObjectiveTo study the brain network mechanism of cognitive control in the elderly with brain aging.Materials and methods21 normal young people and 20 elderly people were included in this study. Mini-mental State Examination and functional near-infrared spectroscopy (fNIRS) synchronous judgment test (including forward tests and reverse judgment tests) were performed on all subjects. To observe and compare differences in brain region activation and brain functional connectivity between subjects and forward and reverse trials by recording functional connectivity (FC) in different task paradigms and calculating bilateral prefrontal and primary motor cortical (PMC) areas.ResultsIn the forward and reverse judgment tests, the reaction time of the elderly group was significantly longer than the young group (P < 0.05), and there was no significant difference in the correct rate. In the homologous regions of interest (ROI) data, the FC of PMC and prefrontal cortex (PFC) in the elderly group was significantly decreased (P < 0.05). In the heterologous ROI data, except for left primary motor cortex (LPMC)-left prefrontal cortex (LPFC), the other PMC and PFC of the elderly group were significantly lower than the young group (P < 0.05) while processing the forward judgment test. However, the heterologous ROI data of LPMC-right prefrontal cortex (RPFC), LPMC-LPFC and RPFC-LPFC in the elderly group were significantly lower than the young group (P < 0.05) while processing the reverse judgment test.ConclusionThe results suggest that brain aging affected degeneration of whole brain function, which reduce the speed of information processing and form a brain network functional connection mode different from that of young people.

This paper proposes a new classification method, which can adaptively select effective features and give different classification weights according to the classification ability of features, this method effectively eliminates the influence of invalid features in classification process. For the classification problem of fault diagnosis, this method judges the fault type from the azimuth relative to the normal state in multi-dimensional space, and judges the fault degree from the ratio of distance, the method can not only give the fault category, but also give the fault probability, fault degree and whether there is a new fault type. In the process of updating the classification center, the classification method proposed in this paper solves the problem that the location of the classification center changes with the sample processing order. The effectiveness of this method is tested by the transformer vibration data (TVIB) and five data sets published in MATLAB and UCI, and compared with K-Mean, self-organizing map (SOM) methods, it is proved that considering the classification weight can effectively improve the classification accuracy, and the proposed method has strong applicability for different classification problems and good processing ability for large capacity and multi feature data.

Anthracnose caused by Colletotrichum spp. was widespread in recent years and resulted in great damage to strawberry production. Soil microbial communities were key contributors to host nutrition, development, and immunity; however, the difference between the microbial communities of healthy and anthracnose-infected strawberry rhizosphere soils remains unclear. In this study, the Illumina sequencing technique was used to comparatively study the prokaryotic and fungal community compositions and structures between healthy and anthracnose-infected strawberry rhizosphere soils in Yuxi, Yunnan Province. Both microbial community diversities and richness of anthracnose-infected strawberry rhizosphere soils were higher than those of healthy strawberry rhizosphere soils. A total of 2,518 prokaryotic and 556 fungal operational taxonomic units (OTUs) were obtained at the 97% similarity threshold. Proteobacteria, Thaumarchaeota, and Acidobacteria were the dominant prokaryotic phyla; Ascomycota, unclassified_k__Fungi, and Mortierellomycota were the dominant fungal phyla. The relative abundances of beneficial bacterial phyla Actinobacteria and Firmicutes, genera Streptomyces, Azospirillum, and Bacillus were significantly reduced in anthracnose-infected strawberry rhizosphere soils; the relative abundance of beneficial fungal species Trichoderma asperellum shows a similar tendency with bacterial abundance. Besides Colletotrichum, 15 other potential fungal pathogen genera and seven fungal pathogen species were identified; among the potential pathogen genera and species, eight pathogen genera and Fusarium oxysporum showed significant differences between healthy and anthracnose-infected strawberry rhizosphere soils. The results suggested that strawberry planted in this area may be infected by other fungal pathogens except for Colletotrichum spp. Our present research will provide theoretical basis and data reference for the isolation and identification of strawberry pathogens and potential probiotics in future works.

A building curtain wall is an outer protective structure of a building composed of a panel and a supporting structure system. As the main materials of a building curtain wall, the optimization of the keel and panel is a problem that has attracted much attention from decoration enterprises. In order to reduce the cost of constructing a single building curtain wall and improve the economic benefits of decoration enterprises, in this study, the keel is taken as the key optimization objective, and a comprehensive optimization scheme is proposed for the deepening design stage of a single building curtain wall. This scheme firstly reduces the cost of the keel by optimizing the radius of the keel and then considers the overall degree of fit between the panel and the keel while optimizing the keel. Finally, in order to improve the processing efficiency of the keel, a reverse-cutting method is proposed based on the traditional forward-cutting method and then combined with the forward-cutting method to form a comprehensive cutting method. The feasibility of the optimization of both design and driving cost is verified with practical cases. The results show the following: Firstly, compared with the theoretical scheme, the average rate of cost saving for the keel when using the comprehensive optimization scheme is 9.56%. Secondly, the overall degree of fit between the optimized keel and the panel is evaluated, which effectively reduces the discontinuity of panel installation. Finally, the comprehensive cutting method proposed here is shown to improve the material utilization rate compared with the single forward-cutting method.

Detection of voice changes in Parkinson's Disease (PD) patients would make it possible for early detection and intervention before the onset of disabling physical symptoms. This study explores static and dynamic speech features relating to PD detection. A comparative analysis of the articulation transition characteristics shows that the number of articulation transitions and the trend of the fundamental frequency curve are significantly different between HC speakers and PD patients. Motivated by this observation, we propose to apply Bidirectional long-short term memory (LSTM) model to capture time-series dynamic features of a speech signal for detecting PD. The dynamic speech features are measured based on computing the energy content in the transition from unvoiced to voiced segments (onset), and in the transition from voiced to unvoiced segments (offset). Under the two evaluation methods of 10-fold cross validation (CV) and splitting the dataset without samples overlap of one individual, the experimental results show that the proposed method remarkably improves the accuracy of PD detection over traditional machine learning models using static features.

In this study, we investigated dynamic control strategies for over-constrained cable-driven robots. In order to control a cable-driven robot, it is essential to address issues that arise from the restriction of cable tension, as well as to the redundancy issues that arise from an over-constrained cable-driven system. In contrast to previous research that required consideration of the relationship between tension constraints and computed control wrench in tension distribution problems, we developed a tension function that incorporates the hyperbolic tangent function, which allows tension to always satisfy tension constraints and eliminates the consideration of constraints at each step. The gradient descent method was applied to this tension function to determine an appropriate distribution of tension for the computed wrench. In order to manage tension distribution optimization for achieving objectives such as energy conservation, we provide a practical method to simultaneously realize the necessary wrench and the appropriate tension distribution. Compared with studies that focus on the complex analysis of the structure matrix to solve the tension distribution problem, the tension distribution issue is handled in a straightforward manner in our method, providing the solutions to other problems, such as discontinuity in the calculated wrench, and requirements of changing the cable’s force level during movement. The simulation results and results of comparison with other methods show the effectiveness of the method.

Class IIa histone deacetylases (HDACs) are a subfamily of HDACs with important functions in development and adult tissue homeostasis. As opposed to other HDACs, they lack catalytic function and bind transcription factors to recruit transcriptional co-regulators, mostly co-repressors such as nuclear receptor co-repressor (NCoR)/silencing mediator of retinoid and thyroid hormone receptor (SMRT). Class IIa HDACs enhance mouse somatic cell reprogramming to induced pluripotent stem cells (iPSCs) by repressing the function of the pro-mesenchymal transcription factor myocyte enhancer factor 2 (MEF2), which is upregulated during this process. Here, we describe, using HDAC4 and 7 as examples, that class IIa HDACs exhibit nuclear-cytoplasmic trafficking in reprogramming, being mostly cytoplasmic in donor fibroblasts and intermediate cells but translocating to the nucleus in iPSCs. Importantly, over-expressing a mutant form of HDAC4 or 7 that becomes trapped in the nucleus enhances the early phase of reprogramming but is deleterious afterwards. The latter effect is mediated through binding to the exogenous reprogramming factors at pluripotency loci, and the subsequent recruitment of NCoR/SMRT co-repressors. Thus, our findings uncover a context-dependent function of class IIa HDACs in reprogramming and further reinforce the idea that recruitment of co-repressors by the exogenous factors is a major obstacle for reactivating the pluripotency network in this process. A key challenge is to find/re-purpose approved drugs that could be used in humans to induce autophagy-associated clearance of neurodegenerative proteins. Here, authors demonstrate that felodipine, an anti-hypertensive drug, can induce autophagy and clear a variety of aggregated neurodegenerative disease-associated proteins in mouse brains at plasma concentrations similar to those that would be seen in humans taking the drug.

The water–energy nexus has become a key factor in the implementation of low-carbon green development, which has led to the need for exploring effective management within the coupled integrated system with multi-energy flow supplies. In this study, the coupled relationship between water resources and energy in the integrated energy system was systematically analyzed, and a system operation optimization model was proposed through comprehensively considering cold, heat and electricity load, and nine kinds of energy conversion and supply equipment/technology from the perspective of a water resources and energy nexus in a typical industry park. The system operation scheme, energy supply mode, net benefit and water resource consumption under different water resource control scenarios were obtained. The results show that water resource control would directly bring about a directly positive influence on renewable energy utilization and energy storage reduction, and that a system’s external dependence and benefits, renewable energy utilization potential and other factors in an integrated energy system should be comprehensively considered. The development of more effective control indicators could be better to promote the effectiveness of bidirectional regulation in a water–energy nexus.

Piezoelectric ceramics have recently become one of the most common materials in electromechanical industries, especially in the microelectronic packaging industry. In a jetting dispenser, the presence of imbalanced stress-induced defects in such materials prevents them from providing high stability during the jetting dispensing process. In this work, lead zirconate titanate piezoelectric ceramics (PZT ceramics) were used to actuate the amplifying mechanism of the jetting dispenser. However, during the cyclic dispensing, several ceramics were found to be damaged. In order to figure out what actually caused the damage, analytical and experimental studies were carried out to acquire characteristics of PZT ceramics with damages, and simulation studies of overstress were also performed. Details of the damage characteristics in PZT ceramics are discussed in this paper. This work will improve the stability and life of the piezoelectric dispenser and further promote its wide application in various fields.

Mutations occurring in the gene body of PARK7 (encoding DJ-1/PARK7) cause autosomal recessive early-onset parkinsonism (AREP). These mutations produce a loss of function and have been reported to lead to dopaminergic neuron degeneration in the substantia nigra. However, the underlying mechanisms are largely unknown. Here, we report the generation of a patient-derived induced pluripotent stem cell (iPSC) line carrying mutant DJ-1 (p.L10P). This cell line is a valuable tool for in vitro Parkinson’s disease (PD) modeling and mechanistic studies.

However, the mechanistic links between PARK2 mutations and dopaminergic neuron degeneration are unclear. Here, we have generated three patient-derived induced pluripotent stem cell (iPSC) lines from the same donor with mutant PARKIN (p. C253Y). These well characterized cell lines will facilitate the study of PARKIN function in disease relevant cell types in vitro.

Mutations in the Leucine rich repeat kinase 2 (LRRK2) gene are found in both familial and sporadic Parkinson's disease (PD), and are also associated with immune-related disorders including Crohn's disease (CD) and leprosy. We have generated two homozygous LRRK2 knockout human induced pluripotent stem cell (iPSC) lines using CRISPR-Cas9 in a well-characterized human iPSC clone. The LRRK2 knockout cell lines retained normal morphology, gene expression, and the capacity to differentiate into cell types of the three germ layers. These cell lines are valuable for elucidating the role of LRRK2 in innate immunity and PD.

Loss of function mutations in PARK2 (encoding PARKIN) cause autosomal recessive Parkinson's disease (PD), which often manifests at a juvenile age. Molecular and biochemical studies show that PARKIN functions as an E3 ubiquitin ligase controlling mitochondrial homeostasis. Yet, the exact mechanisms are unclear due to the use of sub-optimal models including cancer cells and fibroblasts. We have generated a PARK2 knockout (KO) isogenic cell line using a well-characterized induced pluripotent stem cell (iPSC) clone with good differentiation potential. This cell line lacks the expression of all PARKIN isoforms and is valuable for elucidating the role of PARK2 mutations in PD.

Familial Parkinson's disease (PD) can be caused by deleterious mutations in PINK1 (encoding PINK1) in an autosomal recessive manner. Functional studies suggest that PINK1 works as a regulator of mitochondrial homeostasis. However, how loss of PINK1 induces dopaminergic neuron degeneration is still unclear. Here, we have generated a patient-derived induced pluripotent stem cell (iPSC) line with mutant PINK1 (p. I368N). This cell line will facilitate PD disease modeling in vitro and can be used for generating isogenic cell lines through gene correction.

Abstract Background Most published genome sequences are drafts, and most are dominated by computational gene prediction. Draft genomes typically incorporate considerable sequence data that are not assigned to chromosomes, and predicted genes without quality confidence measures. The current Actinidia chinensis (kiwifruit) ‘Hongyang’ draft genome has 164 Mb of sequences unassigned to pseudo-chromosomes, and omissions have been identified in the gene models. Results A second genome of an A. chinensis (genotype Red5) was fully sequenced. This new sequence resulted in a 554.0 Mb assembly with all but 6 Mb assigned to pseudo-chromosomes. Pseudo-chromosomal comparisons showed a considerable number of translocation events have occurred following a whole genome duplication (WGD) event some consistent with centromeric Robertsonian-like translocations. RNA sequencing data from 12 tissues and ab initio analysis informed a genome-wide manual annotation, using the WebApollo tool. In total, 33,044 gene loci represented by 33,123 isoforms were identified, named and tagged for quality of evidential support. Of these 3114 (9.4%) were identical to a protein within ‘Hongyang’ The Kiwifruit Information Resource (KIR v2). Some proportion of the differences will be varietal polymorphisms. However, as most computationally predicted Red5 models required manual re-annotation this proportion is expected to be small. The quality of the new gene models was tested by fully sequencing 550 cloned ‘Hort16A’ cDNAs and comparing with the predicted protein models for Red5 and both the original ‘Hongyang’ assembly and the revised annotation from KIR v2. Only 48.9% and 63.5% of the cDNAs had a match with 90% identity or better to the original and revised ‘Hongyang’ annotation, respectively, compared with 90.9% to the Red5 models. Conclusions Our study highlights the need to take a cautious approach to draft genomes and computationally predicted genes. Our use of the manual annotation tool WebApollo facilitated manual checking and correction of gene models enabling improvement of computational prediction. This utility was especially relevant for certain types of gene families such as the EXPANSIN like genes. Finally, this high quality gene set will supply the kiwifruit and general plant community with a new tool for genomics and other comparative analysis.

Abstract Hand segmentation is one of the most fundamental and crucial steps for egocentric human-computer interaction. The special egocentric view brings new challenges to hand segmentation tasks, such as the unpredictable environmental conditions. The performance of traditional hand segmentation methods depend on abundant manually labeled training data. However, these approaches do not appropriately capture the whole properties of egocentric human-computer interaction for neglecting the user-specific context. It is only necessary to build a personalized hand model of the active user. Based on this observation, we propose an online-learning hand segmentation approach without using manually labeled data for training. Our approach consists of top-down classifications and bottom-up optimizations. More specifically, we divide the segmentation task into three parts, a frame-level hand detection which detects the presence of the interactive hand using motion saliency and initializes hand masks for online learning, a superpixel-level hand classification which coarsely segments hand regions from which stable samples are selected for next level, and a pixel-level hand classification which produces a fine-grained hand segmentation. Based on the pixel-level classification result, we update the hand appearance model and optimize the upper layer classifier and detector. This online-learning strategy makes our approach robust to varying illumination conditions and hand appearances. Experimental results demonstrate the robustness of our approach.

This paper studies a novel recurrent neural network (RNN) with hyperbolic secant (sech) in the gate for a specific medical application task of Parkinson’s disease (PD) detection. In detail, it focuses on the fact that patients with PD have motor speech disorders, by converting the voice data into black-and-white images of a recurrence plot (RP) at specific time intervals and constructing the detection model that combines RNN and convolutional neural network (CNN); the study evaluates the performance of the RNN with sech gate compared with long short-term memory (LSTM) and gated recurrent unit (GRU) with conventional gates. As a result, the proposed model obtained similar results to LSTM and GRU (an average accuracy of about 70%) with less hyperparameters, resulting in faster learning. In addition, in the framework of the RNN with sech in gate, the accuracy obtained by using tanh as the output activation function is higher than using the relu function. The proposed method will see more improvement by increasing the data in the future. More analysis on the input sound type, the RP image size, and the deep learning structures will be included in our future work for further improving the performance of PD detection from voice.

Familial hypercholesterolemia (FH) causes elevation of low-density lipoprotein cholesterol (LDL-C) in blood and carries an increased risk of early-onset cardiovascular disease. A caveat for exploration of new therapies for FH is the lack of adequate experimental models. We have created a comprehensive FH stem cell model with differentiated hepatocytes (iHeps) from human induced pluripotent stem cells (iPSCs), including genetically engineered iPSCs, for testing therapies for FH. We used FH iHeps to assess the effect of simvastatin and proprotein convertase subtilisin/kexin type 9 (PCSK9) antibodies on LDL-C uptake and cholesterol lowering in vitro. In addition, we engrafted FH iHeps into the liver of Ldlr−/−/Rag2−/−/Il2rg−/− mice, and assessed the effect of these same medications on LDL-C clearance and endothelium-dependent vasodilation in vivo. Our iHep models recapitulate clinical observations of higher potency of PCSK9 antibodies compared with statins for reversing the consequences of FH, demonstrating the utility for preclinical testing of new therapies for FH patients.

Based on 243 current valid distribution records for six wild strawberry species in China and data on 20 environmental variables, the geographical distributions of and potentially suitable areas for the wild strawberry species in Yunnan Province (China) under the current climate scenario were explored using the MaxEnt model and ArcGIS software, and major environmental variables affecting their geographical distributions were evaluated. In addition, the spatio-temporal dynamic patterns of the suitable areas for the six wild strawberry species in Yunnan Province in the 2050s and 2070s under the two climate models of RCP2.6 and RCP8.5 were predicted. Under the current climate scenario, the six wild strawberry species have suitable areas in Yunnan Province, which were mainly distributed in the high-altitude and low-temperature regions in the northwest and northeast, such as Diqing and Zhaotong. In addition, the average size of the highly suitable area for diploid wild strawberry species was greater than that for tetraploid species. Under the future climate scenarios, the average size of the highly suitable area for diploid species showed a tendency to expand, while that of tetraploid species showed a tendency to shrink. Altitude was a critical variable affecting the distribution of tetraploid species. Under the two future climate models of RCP2.6 and RCP8.5, the suitable areas for wild strawberry species shifted to the regions of high latitude, high altitude, and low temperature. In addition, the average distance in the shift of the suitable area for tetraploid strawberry species was greater than that for the suitable area for diploid strawberry species. The above results provide valuable information for the management and protection of the germplasm resources of Fragaria.

The exponentially increasing size of biomedical literature and the limited ability of manual curators to discover protein–protein interactions (PPIs) in text has led to delays in keeping PPI databases updated with the current findings. The state-of-the-art text mining methods for PPI extraction are primarily based on deep learning (DL) models, and the performance of a DL-based method is mainly affected by the architecture of DL models and the feature embedding methods. In this study, we compared different architectures of DL models, including convolutional neural networks (CNN), long short-term memory (LSTM), and hybrid models, and proposed a hybrid architecture of a bidirectional LSTM+CNN model for PPI extraction. Pretrained word embedding and shortest dependency path (SDP) embedding are fed into a two-embedding channel model, such that the model is able to model long-distance contextual information and can capture the local features and structure information effectively. The experimental results showed that the proposed model is superior to the non-hybrid DL models, and the hybrid CNN+Bidirectional LSTM model works well for PPI extraction. The visualization and comparison of the hidden features learned by different DL models further confirmed the effectiveness of the proposed model.

Abstract Fruit shape represents a key trait that consumers use to identify and select preferred cultivars, and although the manipulation of this trait is an opportunity to create novel, differentiated products, the molecular mechanisms regulating fruit shape are poorly understood in tree fruits. In this study, we have shown that ectopic expression of Malus domestica PISTILLATA (MdPI), the apple ortholog of the floral organ identity gene PISTILLATA (PI), regulates apple fruit tissue growth and shape. MdPI is a single‐copy gene, and its expression is high during flower development but barely detectable soon after pollination. Transgenic apple plants with ectopic expression of MdPI produced flowers with white sepals and a conversion of sepals to petals. Interestingly, these plants produced distinctly flattened fruit as a consequence of reduced cell growth at the basipetal position of the fruit. These altered sepal and fruit phenotypes have not been observed in studies using Arabidopsis. This study using apple has advanced our understanding of PI functions outside the control of petal and stamen identity and provided molecular genetic information useful for manipulating fruit tissue growth and fruit shape.

Both commercial and experimental genotypes of kiwifruit (Actinidia spp.) exhibit large differences in response to insect pests. An understanding of the vine's physiological response to insect feeding and its genetic basis will be important in assisting the development of varieties with acceptable levels of pest resistance. This experiment describes transcriptome changes observed in the bark of kiwifruit 2 and 7 days after the commencement of feeding by the armored scale insect pest, Hemiberlesia lataniae. Using a cDNA microarray consisting of 17,512 unigenes, we measured transcriptome changes and analyzed these into functional ontology categories using MapMan. Results are available in the GEO database GSE73922 and are described fully in Ref. Hill et al. (2015) [1]. After 7 days, transcripts associated with photosynthesis were down-regulated and secondary metabolism was up-regulated. Differential expression of transcripts associated with stress response was consistent with a defense response involving both effector and herbivore-triggered immunities, with predominant involvement of the salicylic acid phytohormonal pathway. This hypothesis was supported by the results of two laboratory experiments. The methods described here could be further adapted and applied to the study of plant responses to a wide range of sessile sucking pests.

Somatic cells can be reprogrammed to induced pluripotent stem cells (iPSCs) using oncogenic transcription factors. However, this method leads to genetic aberrations in iPSCs via unknown mechanisms, which may limit their clinical use. Here, we demonstrate that the supplementation of growth media with antioxidants reduces the genome instability of cells transduced with the reprogramming factors. Antioxidant supplementation did not affect transgene expression level or silencing kinetics. Importantly, iPSCs made with antioxidants had significantly fewer de novo copy number variations, but not fewer coding point mutations, than iPSCs made without antioxidants. Our results suggest that the quality and safety of human iPSCs might be enhanced by using antioxidants in the growth media during the generation and maintenance of iPSCs.

The real-time observation of electrohydrodynamic printing (EHDP) process is of importance in practical applications. The electric current is a reflection of charge transfer and might provide a potential method to detect the liquid behavior. In this paper, current during EHDP process is measured and studied to investigate the relationship with liquid behaviors. Experimental results show that the liquid ejection can be accurately reflected by the current signal. Current of various ejection modes in EHDP are then examined, and various characteristic numbers are summarized to identify the present liquid ejection mode. This work proposes a simple and efficient method by combining current detection and visual observation to achieve better real-time monitoring and controlling of EHDP process.

Featured Application: The proposed Uncertainty Flow framework may benefit the facial analysis with its promised elevation in discriminability in multi-label affective classification tasks. Moreover, this framework also allows the efficient model training and between tasks knowledge transfer. The applications that rely heavily on continuous prediction on emotional valance, e.g., to monitor prisoners’ emotional stability in jail, can be directly benefited from our framework. Abstract: To lower the single-label dependency on affective facial analysis, it urges the fruition of multi-label affective learning. The impediment to practical implementation of existing multi-label algorithms pertains to scarcity of scalable multi-label training datasets. To resolve this, an inductive transfer learning based framework, i.e.,Uncertainty Flow, is put forward in this research to allow knowledge transfer from a single labelled emotion recognition task to a multi-label affective recognition task. I.e., the model uncertainty—which can be quantified in Uncertainty Flow—is distilled from a single-label learning task. The distilled model uncertainty ensures the later efficient zero-shot multi-label affective learning. On the theoretical perspective, within our proposed Uncertainty Flow framework, the feasibility of applying weakly informative priors, e.g., uniform and Cauchy prior, is fully explored in this research. More importantly, based on the derived weight uncertainty, three sets of prediction related uncertainty indexes, i.e., soft-max uncertainty, pure uncertainty and uncertainty plus are proposed to produce reliable and accurate multi-label predictions. Validated on our manual annotated evaluation dataset, i.e., the multi-label annotated FER2013, our proposed Uncertainty Flow in multi-label facial expression analysis exhibited superiority to conventional multi-label learning algorithms and multi-label compatible neural networks. The success of our proposed Uncertainty Flow provides a glimpse of future in continuous, uncertain, and multi-label affective computing.

The kiwifruit cultivar Actinidia chinensis 'Hort16A' is resistant to the polyphagous armoured scale insect pest Hemiberlesia lataniae (Hemiptera: Diaspididae). A cDNA microarray consisting of 17,512 unigenes selected from over 132,000 expressed sequence tags (ESTs) was used to measure the transcriptomic profile of the A. chinensis 'Hort16A' canes in response to a controlled infestation of H. lataniae. After 2 days, 272 transcripts were differentially expressed. After 7 days, 5,284 (30%) transcripts were differentially expressed. The transcripts were grouped into 22 major functional categories using MapMan software. After 7 days, transcripts associated with photosynthesis (photosystem II) were significantly down-regulated, while those associated with secondary metabolism were significantly up-regulated. A total of 643 transcripts associated with response to stress were differentially expressed. This included biotic stress-related transcripts orthologous with pathogenesis related proteins, the phenylpropanoid pathway, NBS-LRR (R) genes, and receptor-like kinase-leucine rich repeat signalling proteins. While transcriptional studies are not conclusive in their own right, results were suggestive of a defence response involving both ETI and PTI, with predominance of the SA signalling pathway. Exogenous application of an SA-mimic decreased H. lataniae growth on A. chinensis 'Hort16A' plants in two laboratory experiments.

To improve the jetting performance of liquid metals, an electromagnetic micro-jetting (EMJ) valve that realizes drop-on-demand (DOD) jetting while not involving any valve core or moving parts was designed. The influence of the lead angle of the nozzle on the jetting of liquid metal gallium (Ga) was investigated. It was found that the Lorentz force component parallel to the nozzle that jets the electrified liquid Ga is always larger than its internal friction; thus, jet can be generated with any lead angle but with different kinetic energies. Experimental results show that the mass of the jetting liquid, the jetting distance, the initial velocity of the jet, and the resulting kinetic energy of the jet increase first and then decrease. When the lead angle is 90°, the mass of the jetting liquid and the kinetic energy are at their maximum. When the angle is 80°, the initial velocity achieves its maximum, with a calculated value of 0.042 m/s. Moreover, very close and comparatively high kinetic energies are obtained at 80° and 90°, indicating that angles in between this range can produce a preferable performance. This work provides an important theoretical basis for the design of the EMJ valve, and may promote the development and application of micro electromagnetic jetting technology.
