Abstract The combination of micron zero-valent iron (mZVI) and microorganisms is an effective method for trichloroethylene (TCE) degradation, but electron transfer efficiency needs improvement. A new chem-bio hybrid process using a composite material (S-ZVI@biochar) was developed, consisting of sulfurized mZVI and biochar as a chemical remover, and Shewanella oneidensis MR-1 and dechlorinating bacteria (DB) as a biological agent for TCE degradation. S-ZVI@biochar showed improved stability, biocompatibility, and TCE removal compared to ZVI and S-ZVI. The hybrid system DB + MR-1 + S-ZVI@biochar exhibited the highest TCE removal efficiency at 96.5% after 30 days, which was 3.7 times higher than that of bare ZVI. The study revealed that the enhanced dechlorination performance was due to improved electron transfer efficiency, adjustment of microbial community structure, and iron recycling. S-ZVI@biochar constructed electron transport channels in the composite system, improving the overall dechlorination capacity. This system shows promise for long-term TCE removal in anaerobic environments.

Abstract Double-layer island working face main roadway coal pillars are affected by complex mining stress superposition, when different coal pillar width combinations, the surrounding rock stress field will produce different degrees of regional loading increase effect; the study of the surrounding rock stress field regional superposition loading increase law is meaningful to explaining the failure mode of the roadway and determining the critical control area. This study combines numerical simulation with on-site monitoring and other methods and draws the following conclusions: The superimposed loading increase law (“decreasing” → “increasing”) of the abutment pressure and deviatoric stress in the lower coal seam of the double-layer island working face during the mining; the type of the principal stress deflection in the advance working face region; and by obtaining the three types of development morphology of the deviatoric stress peak zone of the roadway and its corresponding nine evolution modes (one type of circular tube → four types of inverse hyperbolic body → four types of hyperbolic body) in the double-layered island working face mining. Indicated the critical reinforcement area corresponding to the main roadway when at different combinations of coal pillar widths; determined the main track roadway protective coal pillars width for 40 m and the shape of the roadway peak deviatoric stress zone is the inverse class hyperbolic body mode; according to the evolution mode of the peak deviatoric stress zone, determined the synergistic failure control program for the asymmetric critical zone of the roadway surrounding rock which is a targeted scientific support method; after the feedback of on-site monitoring and, the support program is reasonable and effective.

Abstract Sweet corn rapidly loses its sweetness, along with other nutrients, once it is harvested, which reduces its economic value. The present study was conducted to elucidate the regulatory mechanisms by which a rapid cooling slurry ice (SI) treatment followed by storage at 0 ± 0.5°C extends the postharvest quality of sweet corn relative to slower direct cooling at 0 ± 0.5°C. In this study, we found that SI treatment can maintain the appearance quality of sweet corn and inhibit grain shrinkage, respiration, and ethylene release. SI treatment delayed the aging of corn by regulating the expression levels of genes and the levels of metabolites. SI treatment also maintains its carbohydrate content, which is mainly related to starch and sucrose metabolism, glycolysis, citric acid cycle, pentose phosphate pathway, and cutin biosynthesis. The results provide a possible target for improving the postharvest quality of sweet corn.

Abstract Background The number of spikelets per spike is a key trait that affects the yield of bread wheat (Triticum aestivum L.). Identification of the QTL for spikelets per spike and its genetic effects that could be used in molecular assistant breeding in the future. Results In this study, four recombinant inbred line (RIL) populations were generated and used, having YuPi branching wheat (YP), with Supernumerary Spikelets (SS) phenotype, as a common parent. QTL (QSS.sicau-2 A and QSS.sicau-2D) related to SS trait were mapped on chromosomes 2 A and 2D through bulked segregant exome sequencing (BSE-Seq). Fourteen molecular markers were further developed within the localization interval, and QSS.sicau-2 A was narrowed to 3.0 cM covering 7.6 Mb physical region of the reference genome, explaining 13.7 − 15.9% the phenotypic variance. Similarly, the QSS.sicau-2D was narrowed to 1.8 cM covering 2.4 Mb physical region of the reference genome, and it explained 27.4 − 32.9% the phenotypic variance. These two QTL were validated in three different genetic backgrounds using the linked markers. QSS.sicau-2 A was identified as WFZP-A, and QSS.sicau-2D was identified a novel locus, different to the previously identified WFZP-D. Based on the gene expression patterns, gene annotation and sequence analysis, TraesCS2D03G0260700 was predicted to be a potential candidate gene for QSS.sicau-2D. Conclusion Two significant QTL for SS, namely QSS.sicau-2 A and QSS.sicau-2D were identified in multiple environments were identified and their effect in diverse genetic populations was assessed. QSS.sicau-2D is a novel QTL associated with the SS trait, with TraesCS2D03G0260700 predicted as its candidate gene.

Abstract Objective This study aimed to establish the dose-response relationship between volume base dose and tumor local control for vaginal cancer, including primary vaginal cancer and recurrent gynecologic malignancies in the vagina. Materials and methods We identified studies that reported volume base dose and local control by searching the PubMed, the Web of Science, and the Cochrane Library Database through August 12, 2023. The regression analyses were performed using probit model between volume based dose versus clinical outcomes. Subgroup analyses were performed according to stratification: publication year, country, inclusion time of patients, patients with prior radiotherapy, age, primaries or recurrent, tumor size, concurrent chemoradiotherapy proportion, dose rate, image modality for planning, and interstitial proportion. Results A total of 879 patients with vaginal cancer were identified from 18 studies. Among them, 293 cases were primary vaginal cancer, 573 cases were recurrent cancer in the vagina, and 13 cases were unknown. The probit model showed a significant relationship between the HR-CTV (or CTV) D90 versus the 2-year and 3-year local control, P values were 0.013 and 0.014, respectively. The D90 corresponding to probabilities of 90% 2-year local control were 79.0 GyEQD2,10 (95% CI: 75.3–96.6 GyEQD2,10). Conclusions A significant dependence of 2-year or 3-year local control on HR-CTV (or CTV) D90 was found. Our research findings encourage further validation of the dose-response relationship of radical radiotherapy for vaginal cancer through protocol based multicenter clinical trials.

Abstract Background Carbapenem-resistant Escherichia coli (CREC) has been considered as WHO priority pathogens, causing a great public health concern globally. While CREC from patients has been thoroughly investigated, the prevalence and underlying risks of CREC in healthy populations have been overlooked. Systematic research on the prevalence of CREC in healthy individuals was conducted here. We aimed to characterize CREC collected from healthy populations in China between 2020 and 2022 and to compare the genomes of CREC isolates isolated from healthy individuals and clinical patients. Methods We present a nationwide investigation of CREC isolates among healthy populations in China, employing robust molecular and genomic analyses. Antimicrobial susceptibility testing, whole-genome sequencing, and bioinformatics were utilized to analyze a cohort of CREC isolates (n = 113) obtained from fecal samples of 5 064 healthy individuals. Representative plasmids were extracted for third-generation nanopore sequencing. We previously collected 113 non-duplicate CREC isolates (59 in 2018, 54 in 2020) collected from ICU patients in 15 provinces and municipalities in China, and these clinical isolates were used to compare with the isolates in this study. Furthermore, we employ comparative genomics approaches to elucidate molecular variations and potential correlations between clinical and non-clinical CREC isolates. Results A total of 147 CREC isolates were identified from 5 064 samples collected across 11 provinces in China. These isolates were classified into 64 known sequence types (STs), but no dominant STs were observed. In total, seven carbapenemase genes were detected with bla NDM-5 (n = 116) being the most prevalent one. Genetic environments and plasmid backbones of bla NDM were conserved in CREC isolated from healthy individuals. Abstract Background The interplay between exosomes and the tumor microenvironment (TME) remains unclear. We investigated the influence of exosomes on the TME in hepatocellular carcinoma (HCC), focusing on their mRNA expression profile. Methods mRNA expression profiles of exosomes were obtained from exoRBase. RNA sequencing data from HCC patients’ tumors were acquired from The Cancer Genome Atlas (TCGA) and the International Cancer Genome Consortium (ICGC). An exosome mRNA-related risk score model of prognostic value was established. The patients in the two databases were divided into high- and low-risk groups based on the median risk score value, and used to validate one another. Functional enrichment analysis was performed based on a differential gene prognosis model (DGPM). CIBERSORT was used to assess the abundance of immune cells in the TME. The correlation between the expression levels of immune checkpoint-related genes and DGPM was analyzed alongside the prediction value to drug sensitivity. Results A prognostic exosome mRNA-related 4-gene signature (DYNC1H1, PRKDC, CCDC88A, and ADAMTS5) was constructed and validated. A prognostic nomogram had prognostic ability for HCC. The genes for this model are involved in extracellular matrix, extracellular matrix (ECM)-receptor interaction, and the PI3K-Akt signaling pathway. Expression of genes here had a positive correlation with immune cell infiltration in the TME. Conclusions Our study results demonstrate that an exosome mRNA-related risk model can be established in HCC, highlighting the functional significance of the molecules in prognosis and risk stratification.

In the current study, the CN-CD30V10A battery tester, K-type temperature sensor, and RS485 are utilized to perform charging and discharging tests on the Panasonic 21,700 lithium-ion battery at various rates. The voltage and temperature changes are measured in real-time, with the data transmitted to the computer via RS485. Then a micro-thermal resistance method is proposed to calculate the equivalent thermal resistance of the cell core along the axial positive and negative poles, as well as along the radial outer circumference surface under anisotropic conditions. Based on the experimental results and the equivalent thermal resistance, an electrothermal coupling model is established for battery internal temperature estimation, taking into account anisotropy and temperature effects on heat transfer. Through the integration of the model and externally measured temperature, the internal temperature is accurately estimated. Moreover, the proposed model (Root-Mean-Square Error, RMSE=0.329) exhibits a significant improvement in accuracy compared to equivalent circuit model (RMSE=1.08), extended Kalman filter (RMSE=0.95), network extended Kalman filter (RMSE=0.58), and neural network unscented Kalman filter (RMSE=0.61). Specifically, the model achieves an approximately 76 % to 228 % reduction in RMSE compared to these methods.

Soybean yield is affected by soil, terrain, biology, and other factors. However, few studies have considered the main and secondary influencing factors of soybean yield and the interaction mechanisms among these factors within a catchment area in the black soil region. This study aims to explore the main influencing factors and the coupling mechanisms affecting soybean yield in China’s northeast black soil region. Using 46 samples from Heshan Farm, Nenjiang City, a driving factor model for soybean yield was developed using PLS-SEM, incorporating latent variables such as soybean yield, topographic characteristics, soil physical properties, soil chemical properties, and biological factors. The main factors, secondary factors, and their interactions affecting soybean yield were discussed. Topographic factors, soil physical properties, and soil chemical properties had positive total effects on soybean yield, while biological factors had a direct negative impact on soybean yield, with the total effects ranging from soil physical properties > biological factors > topographic factors > soil chemical properties. In terms of soil physical properties, the load value of soil bulk density was the largest (−0.917). This was followed by saturated water content (0.886), total porosity (0.855), capillary water holding capacity (0.837), field water holding capacity (0.763), black soil thickness (0.695), sand particle size (−0.664), clay particle size (0.450), and gravel ratio (−0.439). These findings demonstrate soil physical properties are the key factors affecting soybean yield. Thus, soil improvement in small catchment, by reducing bulk density, increasing porosity, and improving soil water retention capacity are key to improving soybean yield.

Background: The clinical applications of endoscope-assisted transoral release for irreducible atlantoaxial dislocations are limited. This study aimed to investigate the clinical effect and prognostic factors of traditional and endoscope-assisted transoral release, as well as posterior reduction and fixation, in treating irreducible atlantoaxial dislocations. Materials and methods: We conducted a retrospective study on 59 patients with irreducible atlantoaxial dislocation who underwent either traditional or endoscope-assisted transoral release, posterior fixation, and fusion between January 2018 and January 2023. Various data, including surgical time, blood loss, drainage volume, oral intake, hospital stay, complications, and neurological status (assessed by the Japanese Orthopedic Association [JOA] score and Oswestry Disability Index [ODI]), were recorded. Imaging parameters such as the atlantodontoid interval (ADI), space available for the cord (SAC), and cervicomedullary angle (CMA) were analyzed and compared. In addition, the correlation between ODI, JOA and patient age, course of disease, preoperative ADI, SAC and CMA were analyzed. Results: No significant differences were observed in age, sex, BMI, preoperative ADI, preoperative SAC, or preoperative CMA. All patients achieved excellent reduction with no significant differences between the two groups. Abstract The continued miniaturization of chips demands highly thermally conductive materials and effective thermal management strategies. Particularly, the high‐field transport of the devices built with 2D materials is limited by self‐heating. Here a systematic control of heat flow in single‐side fluorinated graphene (FG) with varying degrees of fluorination is reported, revealing a superior room‐temperature thermal conductivity as high as 128 W m−1 K−1. Monolayer graphene/FG lateral heterostructures with seamless junctions are approached for device fabrication. Efficient in‐plane heat removal paths from graphene channel to side FG are created, contributing significant reduction of the channel peak temperature and improvement in the current‐carrying capability and power density. Molecular dynamics simulations indicate that the interfacial thermal conductance of the heterostructure is facilitated by the high degree of overlap in the phonon vibrational spectra. The findings offer novel design insights for efficient heat dissipation in micro‐ and nanoelectronic devices.

In order to investigate the effect of different water filling times on the compression deformation of filled gangue, a large-scale crushed gangue deformation testing system was utilized, and two sets of gradient loading experiments were designed. Through rigorous experimental analysis, distinct patterns of deformation during loading and constant load stages are elucidated, with a particular focus on the influence of water filling timing. Quantitative assessment reveals that early water filling induces rapid gangue deformation, primarily during loading stages, while later water filling stages exhibit a heightened impact on constant load stages. Moreover, the study identifies a stepped deformation phenomenon attributed to the irregularities in crushed gangue particles, shedding light on the underlying mechanisms governing gangue behavior under varying water filling conditions. Notably, an in-depth analysis of energy consumption patterns underscores the dominant role of strain, especially in later water filling stages, significantly influencing deformation characteristics. The study further explores the implications of water filling timing on mining subsidence effects, offering quantitative insights into the stability of gangue backfilling. Under the action of the cumulative deformation and concentrated filling effect, the equivalent filling height is slightly higher than that of the early water filling state, but this trend weakens with the increase of mining height. The conclusions of this study can provide experimental data for stability analysis of crushed gangue filling the abandoned mines with different water filling timing.

Abstract To assess the skiing economy (SE) and kinematics during double poling (DP) roller skiing between two groups of skiers in a field setting. Five experienced and five novice male skiers performed a SEDP test at 16 km∙h−1 on an outdoor athletics track. Gas exchange parameters were measured to determine SEDP. A two-dimensional video was filmed to measure the kinematics variables. Abstract In this paper we address the question of whether high-temperature superconductors have anything in common with BCS-BEC crossover theory. Towards this goal, we present a proposal and related predictions which provide a concrete test for the applicability of this theoretical framework. These predictions characterize the behavior of the Ginzburg-Landau coherence length, $${\xi }_{0}^{{{{\rm{coh}}}}}$$ ξ 0 coh , near the transition temperature T c, and across the entire superconducting T c dome in the phase diagram. That we are lacking a systematic characterization of $${\xi }_{0}^{{{{\rm{coh}}}}}$$ ξ 0 coh in the entire class of cuprate superconductors is perhaps surprising, as it is one of the most fundamental properties of any superconductor. This paper is written to motivate further experiments and, thus, address this shortcoming. Here we show how measurements of $${\xi }_{0}^{{{{\rm{coh}}}}}$$ ξ 0 coh contain direct indications for whether or not the cuprates are associated with BCS-BEC crossover and, if so, where within the crossover spectrum a particular superconductor lies.

Abstract Background Cytokine storm is known to impact the prognosis of coronavirus disease 2019 (COVID-19), since pro-inflammatory cytokine variants are associated with cytokine storm. It is tempting to speculate that pro-inflammatory cytokines variants may impact COVID-19 outcomes by modulating cytokine storm. Here, we verified this hypothesis via a comprehensive analysis. Methods PubMed, Cochrane Library, Central, CINAHL, and ClinicalTrials.gov were searched until December 15, 2023. Case–control or cohort studies that investigated the impacts of rs1800795 or rs1800629 on COVID-19 susceptibility, severity, mortality, IL-6, TNF-α, or CRP levels were included after an anonymous review by two independent reviewers and consultations of disagreement by a third independent reviewer. Results 47 studies (8305 COVID-19 individuals and 17,846 non-COVID-19 individuals) were analyzed. The rs1800629 A allele (adenine at the −308 position of the promoter was encoded by the A allele) was associated with higher levels of tumor necrosis factor-α (TNF-α) and C-reactive protein (CRP). In contrast, the rs1800795 C allele (cytosine at the −174 position of the promoter was encoded by the C allele) was linked to higher levels of interleukin-6 (IL-6) and CRP. In addition, the A allele of rs1800629 increased the severity and mortality of COVID-19. However, the C allele of rs1800795 only increased COVID-19 susceptibility. Conclusions rs1800629 and rs1800795 variants of pro-inflammatory cytokines have significant impacts on systemic inflammatory profile and COVID-19 clinical outcomes. rs1800629 may serve as a genetic marker for severe COVID-19.

PURPOSE: Through three neurocritical care unit (NCCU) surveys in China, we tried to understand the development status of neurocritical care and clarify its future development. METHODS: Using a cross-sectional survey method and self-report questionnaires, the number and quality of NCCUs were investigated through three steps: administering the questionnaire, sorting the survey data, and analyzing the survey data. RESULTS: At the second and third surveys, the number of NCCUs (76/112/206) increased by 47% and 84%, respectively. The NCCUs were located in tertiary grade A hospitals or teaching hospitals (65/100/181) in most provinces (24/28/29). The numbers of full-time doctors (359/668/1337) and full-time nurses (904/1623/207) in the NCCUs increased, but the doctor–bed ratio and nurse–bed ratio were still insufficient (0.4:1 and 1.3:1). CONCLUSION: In the past 20 years, the growth rate of NCCUs in China has accelerated, while the allocation of medical staff has been insufficient. Although most NCCU hospital bed facilities and instruments and equipment tend to be adequate, there are obvious defects in some aspects of NCCUs.

Abstract In northern Australia, a region with limited access to healthcare and a substantial population living remotely, antibiotic resistance adds to the complexity of treating infections. Focussing on Escherichia coli urinary tract infections (UTIs) and Staphylococcus aureus skin & soft tissue infections (SSTIs) captured by a northern Australian antibiotic resistance surveillance system, we used logistic regression to investigate predictors of a subsequent resistant isolate during the same infection episode. We also investigated predictors of recurrent infection. Our analysis included 98,651 E. coli isolates and 121,755 S. aureus isolates from 70,851 patients between January 2007 and June 2020. Following an initially susceptible E. coli UTI, subsequent recovery of a cefazolin (8%) or ampicillin (13%) -resistant isolate during the same infection episode was more common than a ceftriaxone-resistant isolate (2%). For an initially susceptible S. aureus SSTI, subsequent recovery of a methicillin-resistant isolate (8%) was more common than a trimethoprim-sulfamethoxazole-resistant isolate (2%). For UTIs and SSTIs, prior infection with a resistant pathogen was a strong predictor of both recurrent infection and resistance in future infection episodes. This multi-centre study demonstrates an association between antibiotic resistance and an increased likelihood of recurrent infection. Particularly in remote areas, a patient’s past antibiograms should guide current treatment choices since recurrent infection will most likely be at least as resistant as previous infection episodes. Using population-level surveillance data in this way can also help clinicians decide if they should switch antibiotics for patients with ongoing symptoms, while waiting for diagnostic results.

In order to improve the control accuracy and synchronous performance of the multi-motor control system, this paper proposes an optimal position collaborative control strategy for dual permanent magnet synchronous motor system. Firstly, based on the mathematical model of permanent magnet synchronous motor, an incremental predictive model is established to suppress constant and slow time-varying disturbances caused by changes in rotational inertia and load torque. Afterwards, the predicted compensation amount after multi-step prediction is obtained by minimizing the cost function which contains the synchronous error and tracking error simultaneously, and compensated to the given current loop in advance without changing the original dual motor system structure. Then the controllability and observability of the system are analyzed, and it can be proved that the system is asymptotically stable based on the optimal control theory. Secondly, considering the constraints such as maximum voltage and current limits, the optimal solution under the constraints is output to obtain the reference currents iq1 and iq2. Finally, it is demonstrated through the experiments that the proposed control strategy can achieve advance prediction compensation control of synchronous error, and the dynamic response speed is improved, while ensuring its steady-state performance remains unchanged.

Apple quality is closely related to its cultivar and origin. However, the apple quality characteristics of different cultivars and origins are unclear. The hypothesis that some quality indicators can effectively distinguish the cultivar and origin of apples. The result indicated that the discriminant accuracy of the models was above 90%, and the multilayer perceptron neural network (MLP–NN) model was superior to the linear discriminant analysis (LDA) model. The identification accuracy of cultivars was higher than origins. The main reason was that the single fruit weight, vitamin C, total soluble solid, soluble sugar, sweetness value, sorbitol, glucose, fructose, sucrose, malic acid, quinic acid and citric acid of ‘Fuji’ apples were significantly higher than ‘Gala’ apples. This study provides a foundation for the quality evaluation and further geographical traceability studies of apples. Further studies related to the regulatory mechanism of environmental conditions on apple quality characteristics should be explored for theoretical confirmation.

Duchenne muscular dystrophy (DMD) is the most prevalent herediatry disease in men, characterized by dystrophin deficiency, progressive muscle wasting, cardiac insufficiency, and premature mortality, with no effective therapeutic options. Here, we investigated whether adenine base editing can correct pathological nonsense point mutations leading to premature stop codons in the dystrophin gene. We identified 27 causative nonsense mutations in our DMD patient cohort. Treatment with adenine base editor (ABE) could restore dystrophin expression by direct A-to-G editing of pathological nonsense mutations in cardiomyocytes generated from DMD patient-derived induced pluripotent stem cells. We also generated two humanized mouse models of DMD expressing mutation-bearing exons 23 or 30 of human dystrophin gene. Intramuscular administration of ABE, driven by ubiquitous or muscle-specific promoters could correct these nonsense mutations in vivo, albeit with higher efficiency in exon 30, restoring dystrophin expression in skeletal fibers of humanized DMD mice. Moreover, a single systemic delivery of ABE with human single guide RNA (sgRNA) could induce body-wide dystrophin expression and improve muscle function in rotarod tests of humanized DMD mice. These findings demonstrate that ABE with human sgRNAs can confer therapeutic alleviation of DMD in mice, providing a basis for development of adenine base editing therapies in monogenic diseases.

BackgroundHemorrhagic Fever with Renal Syndrome (HFRS) continues to pose a significant public health threat to the well-being of the population. Given that the spread of HFRS is susceptible to meteorological factors, we aim to probe into the meteorological drivers of HFRS. Thus, novel techniques that can discern time-delayed non-linear relationships from nonlinear dynamical systems are compulsory.MethodsWe analyze the epidemiological features of HFRS in Weifang City, 2011-2020, via the employment of the Empirical Dynamic Modeling (EDM) method. Our analysis delves into the intricate web of time-delayed non-linear associations between meteorological factors and HFRS. Additionally, we investigate the repercussions of minor perturbations in meteorological variables on future HFRS incidence.ResultsA total of 2515 HFRS cases were reported in Weifang from 2011 to 2020. The number of cases per week was 4.81, and the average weekly incidence was 0.52 per 1,000,000. The propagation of HFRS is significantly impacted by the mean weekly temperature, relative humidity, cumulative rainfall, and wind speed, and the ρCCM converges to 0.55,0.48,0.38 and 0.39, respectively. The graphical representation of the relationship between temperature (lagged by 2 weeks) and the incidence of HFRS exhibits an inverted U-shaped curve, whereby the incidence of HFRS culminates as the temperature reaches 10 °C. Moreover, temperature, relative humidity, cumulative rainfall, and wind speed exhibit a positive correlation with HFRS incidence, with a time lag of 4-6 months.ConclusionsOur discoveries suggest that meteorological factors can drive the transmission of HFRS both at a macroscopic and microscopic scale. Prospective alterations in meteorological conditions, for instance, elevations in temperature, relative humidity, and precipitation will instigate an upsurge in the incidence of HFRS after 4-6 months, and thus, timely public health measures should be taken to mitigate these changes.

BackgroundRenshen Yangrong decoction (RSYRD) has been shown therapeutic effects on secondary malaise and fatigue (SMF). However, to date, its bioactive ingredients and potential targets remain unclear.PurposeThe purpose of this study is to assess the potential ingredients and targets of RSYRD on SMF through a comprehensive strategy integrating network pharmacology, Mendelian randomization as well as molecular docking verification.MethodsSearch for potential active ingredients and corresponding protein targets of RSYRD on TCMSP and BATMAN-TCM for network pharmacology analysis. Mendelian randomization (MR) was performed to find therapeutic targets for SMF. The eQTLGen Consortium (sample sizes: 31,684) provided data on cis-expression quantitative trait loci (cis-eQTL, exposure). The summary data on SMF (outcome) from genome-wide association studies (GWAS) were gathered from the MRC-IEU Consortium (sample sizes: 463,010). We built a target interaction network between the probable active ingredient targets of RSYRD and the therapeutic targets of SMF. We next used drug prediction and molecular docking to confirm the therapeutic value of the therapeutic targets.ResultsIn RSYRD, network pharmacology investigations revealed 193 possible active compounds and 234 associated protein targets. The genetically predicted amounts of 176 proteins were related to SMF risk in the MR analysis. Thirty-seven overlapping targets for RSYRD in treating SMF, among which six (NOS3, GAA, IMPA1, P4HTM, RB1, and SLC16A1) were prioritized with the most convincing evidence. Finally, the 14 active ingredients of RSYRD were identified as potential drug molecules. The strong affinity between active components and putative protein targets was established by molecular docking.ConclusionThis study revealed several active components and possible RSYRD protein targets for the therapy of SMF and provided novel insights into the feasibility of using Mendelian randomization for causal inference between Chinese medical formula and disease.

Abstract Background According to the studies, more than 80% of pediatric patients with cancer can achieve a survival rate greater than 5 years; however, long‐term chemotherapy and/or radiation therapy may seriously affect their reproductive ability. Fertility preservation in adolescents with cancer in China was initiated late, and related research is lacking. Analyze data to understand the current situation and implement measures to improve current practices. Methods From 2011 to 2020, data on 275 male adolescents with cancer whose age ranged from 0 to 19 years old were collected from 16 human sperm banks for this retrospective study. Methods include comparing the basic situation of male adolescents with cancer, the distribution of cancer types, and semen quality to analyze the status of fertility preservation. Results The mean age was 17.39 ± 1.46 years, with 13 cases (4.7%) aged 13–14 years and 262 cases (95.3%) aged 15–19 years. Background: Gukang Capsule has been used as a complementary and alternative medicine (CAM) for the treatment of primary osteoporosis (POP) in China. The primary aim of this study was to assess the clinical effectiveness and safety of Gukang Capsule in POP patients.Methods: A systematic search was conducted across multiple academic databases including PubMed, Web of science, Cochrane Library, China National Knowledge Infrastructure, Chongqing VIP Information, and Wanfang database to identify randomized controlled trials investigating the Gukang Capsule in the treatment of POP. The screening process, data extraction, and assessment of methodological quality were conducted independently by two reviewers. Statistical analysis was performed using the Rev Man 5.3 software. Subgroup analysis was carried out through the combination of OPF. Subgroup analysis was performed according to whether OPF were combined. Stata 12.0 was used for sensitivity and bias analysis.Results: Nineteen studies were assessed that included 1804 participants. It was found that compared with the control group, the total effective rate (RR = 1.26, 95% CI, 1.20, 1.33), the Medical Outcomes Study Short-form 36 [RR = 1.26, 95% CI(1.20, 1.33)], the bone mineral density (BMD) of lumbar vertebra (SMD = 0.77, 95% CI, 0.48, 1.07), the BMD of femoral neck [SMD = 0.84, 95% CI(0.53, 1.14)], and the BMD of Ward’s triangle (SMD = 0.64, 95% CI, 0.44, 0.85) of the Gukang Capsule experimental group were higher. Compared with the control group, the fracture healing time (SMD = −2.14, 95% CI, −2.45, −1.84), the bone specific alkaline phosphatase (BALP) levels in serum (SMD = −2.00, 95% CI, −2.83, −1.17), the tartrate resistant acid phosphatase 5b (TRACP-5b) levels in serum (SMD = −2.58, 95% CI, −3.87, −1.29) of the Gukang Capsule experimental group were lower. Abstract Background This study aims to explore the effect of nutritional impact symptoms (NIS) on oral nutritional supplements (ONS) energy intake and use days among head and neck cancer (HNC) patients. Methods A cross‐sectional study was conducted among HNC patients in a hospital in western China between January 2019 and June 2020. The NIS was from the Patient‐Generated Subjective Global Assessment (PG‐SGA) scale. Mann–Whitney test was used to examine the differences between different kinds of NIS and ONS use days. Binary logistic regression was used to determine the effect of NIS on ONS energy intake. Results The most prevalent four NIS were no appetite (35.3%), dysphagia (29.4%), vomiting (13.2%) and oral pain (12.5%), respectively. All patients in the study were malnutrition. Patients with xerostomia or oral pain had less ONS use days than those without these symptoms. Patients with vomiting (OR 0.09, 95% CI 0.02–0.50) or pain (OR 0.15, 95% CI 0.02–0.89) were less likely to have ONS energy intake ≥400 kcal/day than those without these symptoms after adjusting the confounding factors. In addition, one‐point increase in total NIS score was associated with a lower proportion of ONS energy intake ≥400 kcal/day (OR 0.77, 95% CI 0.59–0.99). Conclusion Xerostomia, oral pain, vomiting and pain should be strengthened and intervened to improve ONS use and nutritional status among HNC patients with malnutrition.

As the inflammatory subtype of nonalcoholic fatty liver disease (NAFLD), the progression of nonalcoholic steatohepatitis (NASH) is associated with disorders of glycerophospholipid metabolism. Scoparone is the major bioactive component in Artemisia capillaris which has been widely used to treat NASH in traditional Chinese medicine. However, the underlying mechanisms of scoparone against NASH are not yet fully understood, which hinders the development of effective therapeutic agents for NASH. Given the crucial role of glycerophospholipid metabolism in NASH progression, this study aimed to characterize the differential expression of glycerophospholipids that is responsible for scoparone’s pharmacological effects and assess its efficacy against NASH. Liquid chromatography-multiple reaction monitoring-mass spectrometry (LC-MRM-MS) was performed to get the concentrations of glycerophospholipids, clarify mechanisms of disease, and highlight insights into drug discovery. Additionally, pathologic findings also presented consistent changes in high-fat diet-induced NASH model, and after scoparone treatment, both the levels of glycerophospholipids and histopathology were similar to normal levels, indicating a beneficial effect during the observation time. Altogether, these results refined the insights on the mechanisms of scoparone against NASH and suggested a route to relieve NASH with glycerophospholipid metabolism. In addition, the current work demonstrated that a pseudotargeted lipidomic platform provided a novel insight into the potential mechanism of scoparone action.

Global warming may exacerbate cropland soil drought, affect microbial systems, and decrease grain yield. However, the compensatory effects of supplementary irrigation on winter wheat yield and soil microbial communities under increased temperatures have not been fully studied. Here, a three-year field experiment was conducted, to verify the effects of warming treatments (WS: winter and spring warming; FF: flowering and filling period warming) and irrigation (W0: conventional irrigation; W1: supplemental irrigation) on crop-soil systems during the winter wheat growth stages. The results demonstrated that warming measures have significant negative effects on the net assimilation rate of flag leaves and yield formation, which led to a reduction of wheat yield by 14–40%. However, increasing irrigation can compensate for a yield of 9.7–15%. In addition, warming increases the soil temperature and decreases the soil water content, resulting in decreased soil microbial diversity. The effect size (β) = −0.23 to −0.53 for index of phylogenetic diversity (PD) under warming, while β= 0.16 under increased irrigation. The SEM model further showed that supplementary irrigation during anthesis to filling can improve the soil environment and extend the validity of photosynthetic to increase production, which was an important mitigation measure to adapt to the warming climate in wheat production.

Current research has increasingly focused on the preventive role of individual legal socialization in crime. The socialization of legal emotions is an important part of legal socialization. Building upon existing literature, this study, conducted through two sub-studies, investigated the influencing factors of legal emotions in N mainland Chinese university students and the mechanisms through which legal emotions impact aggressive behavior. In study 1, the results indicated that mother-child attachment, innovation spirit, and positive emotional expression positively predicted positive legal emotion, while mother-child attachment, dependency dimension in adult attachment, and positive emotional expression negatively predicted negative legal emotions. The anxiety dimension in adult attachment and negative emotional expression positively predicted negative legal emotions. In study 2, Positive legal emotion among university students could directly negatively predict aggressive behavior or exert influence through social alienation. Negative legal emotions could not only directly positively predict aggressive behavior but also partly affect it through social alienation. In summary, our study not only identified factors that influence legal emotions, but also found that legal emotions have an impact on aggressive behavior directly or indirectly through social alienation. Our research findings have significant implications for cultivating positive legal emotion in university students and curbing aggressive behavior. This can be achieved by promoting the legal socialization of university students and ultimately contributing to crime prevention.

A bedside rehabilitation robot is developed to address the challenge of motor rehabilitation for patients with lower limb paralysis. Firstly, based on the principles of physical rehabilitation, a two-link planar robot model is used to simulate both the robot and human lower limbs, and the coupling characteristics between the human and robot are thoroughly analyzed. Then, the lower limb rehabilitation robot, fitted with an end-effector and ankle wearable feature, is designed according to the structural parameters. To enhance patient safety during rehabilitation, the device incorporates a freely rotating leg support mechanism that reduces the load on the ankle due to gravitational forces, and a two-stage series elastic mechanism is integrated below the foot support to provide a passive compliant output of robot power, allowing for more natural movement and reducing the risk of injury. Secondly, dynamic modeling is used to determine the dynamic parameters of the robot by conducting simulation calculations based on the inertia parameters of the human body and the robot model design parameters. Finally, an experimental platform is established using the structural and dynamic parameters, and the robot’s reliability is validated through experimentation. Results indicate that the robot can accurately complete passive rehabilitation training tasks, and the dynamic parameters meet the expected requirements.

ABSTRACTTigecycline and colistin were referred to as the “last resort” antibiotics in defending against carbapenem-resistant, Gram-negative bacterial infections, and are currently widely used in clinical treatment. However, the emergence and prevalence of plasmid-mediated tet(X4) and mcr-1 genes pose a serious threat to the therapeutic application of tigecycline and colistin, respectively. In this research, a tigecycline- and colistin-resistant bacteria resensitization system was developed based on efficient and specific DNA damage caused by Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-Associated Protein 9 (Cas9) nucleases. A conjugation method was used to deliver the resensitization system, which harbors two single-guide RNAs targeting tet(X4) and mcr-1 genes and constitutively expressed Cas9. The conjugation efficiency was nearly 100% after conjugation condition optimization in vitro, and the resensitivity efficiency for clinical isolates was over 90%. In addition, when performing resensitization in vivo, the resistance marker was replaced with a glutamate-based, chromosomal, plasmid-balanced lethal system to prevent the introduction of additional resistance genes in clinical settings, making this strategy a therapeutic approach to combat the in vivo spread of antibiotic resistance genes (ARGs) among bacterial pathogens. As a proof of concept, this resensitive system can significantly decrease the counts of tigecycline- and colistin-resistant bacteria to 1% in vivo. Our study demonstrates the efficacy and adaptability of CRISPR-Cas systems as powerful and programmable antimicrobials in resensitizing tet(X4)- and mcr-1-mediated, tigecycline- and colistin-resistant strains, and opens up new pathways for the development of CRISPR-based tools for selective bacterial pathogen elimination and precise microbiome composition change.IMPORTANCEThe emergence of plasmid-encoded tet(X4) and mcr-1 isolated from human and animal sources has affected the treatment of tigecycline and colistin, and has posed a significant threat to public health. Tigecycline and colistin are considered as the “last line of defense” for the treatment of multidrug-resistant (MDR) Gram-negative bacterial infections, so there is an urgent need to find a method that can resensitize tet(X4)-mediated tigecycline-resistant and mcr-1-mediated colistin-resistant bacteria. In this study, we developed a glutamate-based, chromosomal, plasmid-balanced lethal conjugative CRISPR/Cas9 system, which can simultaneously resensitize tet(X4)-mediated tigecycline-resistant and mcr-1-mediated colistin-resistant Escherichia coli. The counts of tigecycline- and colistin-resistant bacteria decreased to 1% in vivo after the resensitization system was administered. This study opens up new pathways for the development of CRISPR-based tools for selective bacterial pathogen elimination and precise microbiome composition change.

Abstract WRKY transcription factors are pivotal regulators in various aspects of plant biology, including growth, development, secondary metabolic biosynthesis, and responses to both biotic and abiotic stresses. The legume plant Grona styracifolia is widely utilized for its medicinal properties in treating urinary calculi and combating SARS-CoV-2, owing to its bioactive component schaftoside. However, the regulatory function of GsWRKYs in schaftoside biosynthesis within G. styracifolia remains elusive. In the G. styracifolia genome, we identified a total of 102 GsWRKYs, classified phylogenetically into Group I (18), II (68), and III (16). Genomic analysis revealed an uneven distribution of GsWRKYs on chromosomes (Chr), with prevalence on Chr 1, followed by Chr 2, 3, 5, and 6. Among the 82 duplicated GsWRKYs, comprising 12, 54, and 16 members in Group I, II, and III respectively, 11 GsWRKYs were tandemly duplicated genes located across Chr 2 (2), Chr 5 (7), and Chr 9 (2). Weighted gene co-expression network analysis unveiled that 2 Group I (GsWRKY44 and GsWRKY95) and 14 Group II GsWRKYs, including two pairs of segmentally duplicated Group II GsWRKYs associated with thermomorphogenesis, exhibited coexpression with Grona styracifolia C-glycosyltransferases (GsCGT), a gene encoding a C-glucosyltransferase involved in schaftoside biosynthesis. Furthermore, GsWRKY95 demonstrated coexpression with other schaftoside biosynthetic genes. Dual-luciferase and yeast one-hybrid assays provided additional evidence that GsWRKY95 binds to the W-box of GsCGT, activating its expression. In addition, GsWRKY95- and GsCGT-coexpressing G r o na styracifolia chalcone synthase (GsCHSs), along with 11 pairs of segmentally duplicated Group II GsWRKYs, responded to both abiotic and biotic stresses. Notably, certain GsWRKYs were identified as regulators specific to schaftoside biosynthesis in stems, roots, and leaves. These findings suggest that duplication events, particularly in segmentally duplicated Group II GsWRKYs, play a pivotal role in orchestrating the hierarchical regulation of schaftoside biosynthesis. Overall, our results establish a foundation for genetically enhancing G. styracifolia to abundantly produce schaftoside, thereby contributing to its medicinal efficacy.

Hyperspectral imaging technology has emerged as a prominent research area for quantitatively estimating soil nutrient content owing to its non-destructive, rapid, and convenient features. Our work collected the data from soil samples using the hyperspectrometer. Then, the data were processed. The competitive adaptive reweighted sampling (CARS) algorithm reduced the original 148 bands to 13, which accounted for 8.8% of the total bands. These selected bands possess a certain level of interpretability. Based on the modelling results, it can be concluded that the prediction model constructed by the least squares support vector machine (LSSVM) exhibited the highest accuracy. The coefficient determination, root mean square error, and ratio performance deviation were 0.8295, 2.95, and 2.42, respectively. These findings can provide theoretical support for the application of hyperspectral technology in detecting the content of the AHN in soil. Moreover, they can also serve as a reference for the rapid detection of other soil components.

Abstract Manatees are aquatic mammals with voracious appetites. They rely on sea grass as the main food source, and often spend up to eight hours a day grazing. They move slow and frequently stay in groups (i.e. aggregations) in shallow water to search for food, making them vulnerable to environment change and other risks. Accurate counting manatee aggregations within a region is not only biologically meaningful in observing their habit, but also crucial for designing safety rules for boaters, divers, etc., as well as scheduling nursing, intervention, and other plans. In this paper, we propose a deep learning based crowd counting approach to automatically count number of manatees within a region, by using low quality images as input. Because manatees have unique shape and they often stay in shallow water in groups, water surface reflection, occlusion, camouflage etc. making it difficult to accurately count manatee numbers. To address the challenges, we propose to use Anisotropic Gaussian Kernel (AGK), with tunable rotation and variances, to ensure that density functions can maximally capture shapes of individual manatees in different aggregations. After that, we apply AGK kernel to different types of deep neural networks primarily designed for crowd counting, including VGG, SANet, Congested Scene Recognition network (CSRNet), MARUNet etc. to learn manatee densities and calculate number of manatees in the scene. By using generic low quality images extracted from surveillance videos, our experiment results and comparison show that AGK kernel based manatee counting achieves minimum Mean Absolute Error (MAE) and Root Mean Square Error (RMSE). The proposed method works particularly well for counting manatee aggregations in environments with complex background.

Abstract Under double-seam mining, the main roadway surrounding rock is affected by the superposition of the advanced stress of the two-seam coal working faces. The stress superposition mode and degree are of great significance to the width calculation of the protective coal pillar and the determination of the critical control direction of the surrounding rock. This paper uses theoretical analysis, numerical simulation, and site engineering practice to carry out targeted research. The conclusions are as follows: Under different lateral pressure coefficients, the superposition evolution law of maximum principal stress direction of two coal seams with different offsets; Two developmental trends and three types of evolution models of J 2 peak zone (the critical area of the stress increase and deflection changes) under different superimposed loading modes are summarized. Based on the typical asymmetric evolution model of the J 2 peak zone, an asymmetric truss-cable co-anchoring method is proposed aimed at the J 2 critical zone. The field monitoring results show that the main roadway surrounding rock is stable after support when the upper coal seam protective coal pillar is left 80 m, and the lower one is 60 m wide. It is of great reference importance for similar engineering practices.

Abstract The significant impact of vegetation restoration on the turnover of organic carbon (OC) is mainly attributed to biological processes. However, the contribution of abiotic processes associated with reactive oxygen species (ROS) during vegetation restoration has remained largely overlooked. Therefore, we systematically explored ROS distribution and production mechanisms in sandy soil and their corresponding effects on OC oxidation through field monitoring and incubation experiments. The cumulative concentrations of hydroxyl radicals (•OH) produced within 24 h varied in different soils undergoing vegetation restoration, ranging from 2.36 to 22.05 μmol kg‒1, which were derived from the Fenton-like reaction of hydrogen peroxide (H2O2) and biotic-induced Fe(II). In addition, during the oxidation of reductive SOC, H2O2 can be formed and react with Fe(II) to generate •OH. Further, •OH significantly induced the abiotic transformation of OC, accounting for 15.93%‒25.80% of carbon dioxide (CO2) efflux. Specifically, 1 mol •OH can induce the production of 0.01–0.02 mol CO2 for particulate organic carbon (POC) and 0.03‒0.23 mol CO2 for mineral-associated organic carbon (MOC), as demonstrated by chemical quenching and sterilisation experiments. This indicated that MOC was more sensitive to •OH attacks. Collectively, our findings provide insights into the accumulation of ROS in sandy soils during vegetation restoration, with •OH playing a key role in SOC oxidation, as well as the patterns of SOC turnover and its response to changes in the soil environment. As new advanced vehicles, the safety and stability of electromagnetic suspension maglev trains have always been a subject of concern. This study introduces the improved R index and τ-distance index into the performance evaluation of the suspension control system, respectively assessing the stability of the suspension gap and the smoothness of train operation, combining them with grey relational analysis to achieve data-driven comprehensive evaluation. Furthermore, feasibility tests on the Fenghuang Maglev Express validate the effectiveness and superiority of the comprehensive evaluation method based on measured data. Experimental results demonstrate that the data-driven comprehensive evaluation method, through designing specialized evaluation metrics and increasing assessment dimensions, effectively evaluates the performance of the suspension system control loop. Compared to a traditional error integral comprehensive performance index, it offers greater comprehensiveness and accuracy, along with real-time state-monitoring capabilities.

The rise of antibiotic tolerance in bacteria harboring genetic elements conferring resistance to antibiotics poses an increasing threat to public health. However, the primary factors responsible for the emergence of antibiotic tolerance and the fundamental molecular mechanisms involved remain poorly comprehended. Here, we demonstrate that the commonly utilized food additive Benzaldehyde (BZH) possesses the capacity to induce a significant level of fluoroquinolone tolerance in vitro among resistant Escherichia coli. Our findings from animal models reveal that the pre-administration of BZH results in an ineffective eradication of bacteria through ciprofloxacin treatment, leading to similar survival rates and bacterial loads as observed in the control group. These results strongly indicate that BZH elicits in vivo tolerance. Mechanistic investigations reveal several key factors: BZH inhibits the formation of bacterial flagella and releases proton motive force (PMF), which aids in expelling antibiotics from within cells to reducing their accumulation inside. In addition, BZH suppresses bacterial respiration and inhibits the production of reactive oxygen species (ROS). Moreover, exogenous pyruvate successfully reverses BZH-induced tolerance and restores the effectiveness of antibiotics, highlighting how crucial the pyruvate cycle is in combating antibiotic tolerance. The present findings elucidate the underlying mechanisms of BZH-induced tolerance and highlight potential hazards associated with the utilization of BZH.

In the past decade, Spodoptera frugiperda has emerged as a significant invasive pest globally, posing a serious threat to agriculture due to its broad diet, migratory behavior, and ability to cause extensive plant damage. While extensive research has focused on the olfactory capabilities of adult S. frugiperda, understanding of the olfactory process in larvae remains limited, despite larvae playing a crucial role in crop damage. To address this gap, we identified an odorant receptor (OR), SfruOR40, expressed in the first-instar larvae through phylogenetic analysis. Using quantitative real-time PCR, we compared SfruOR40 expression levels in larvae and adults. We then characterized the function of SfruOR40 against 67 compounds using the Xenopus oocyte expression system and found that SfruOR40 responded to three plant volatiles. Further, behavioral experiments revealed a larval attraction to (−)-trans-Caryophyllene oxide. This study elucidates SfruOR40’s role in the olfactory recognition of newly hatched S. frugiperda larvae, expanding our knowledge of such mechanisms in Noctuid moths. Furthermore, it highlights the potential of plant-derived natural products for biological pest control from a behavioral ecology perspective.

Currently, there are limitations in the research on the use of carbon fiber geogrids to prevent low-temperature cracking in asphalt pavements. This study aims to comparatively investigate the effects of carbon fiber-based geogrid type and dense-graded asphalt concrete mixture (AC) surface combined body (SCB) type on the low-temperature cracking resistance of reinforced asphalt pavement through low-temperature bending damage tests. Two geogrid types were prepared: a carbon fiber geogrid (CCF) and a glass/carbon fiber composite qualified geogrid (GCF). Two SCB types were studied: AC-13/AC-20 and AC-20/AC-25. The results show that the improvement in the flexural tensile strength of CCF is similar to that for GCF. Moreover, under reinforced conditions, the improvement in the low-temperature cracking resistance of AC-20/AC-25 is better than that for AC-13/AC-20 by 16.26–24.57%. Based on the analysis, the reasonable ratio range of the aperture sizes to the major particle sizes in the dense gradation can achieve a more effective interlocking effect. This can improve the low-temperature cracking resistance of carbon fiber-based geogrid-reinforced samples. Then, increasing the bending absorption energy is a key way of improving the low-temperature cracking resistance of carbon fiber-based geogrid reinforcements. Eventually, the fracture type of carbon fiber-based geogrid-reinforced samples is a mixed plastic–brittle fracture. These results can provide a reference for the road failure analysis of geogrid-reinforced asphalt pavement.

Background COVID-19 carriers experience psychological stresses and mental health issues such as varying degrees of stigma. The Social Impact Scale (SIS) can be used to measure the stigmatisation of COVID-19 carriers who experience such problems. Aims To evaluate the reliability and validity of the Chinese version of the SIS, and the association between stigma and depression among asymptomatic COVID-19 carriers in Shanghai, China. Method A total of 1283 asymptomatic COVID-19 carriers from Shanghai Ruijin Jiahe Fangcang Shelter Hospital were recruited, with a mean age of 39.64 ± 11.14 years (59.6% male). Participants completed questionnaires, including baseline information and psychological measurements, the SIS and Self-Rating Depression Scale. The psychometrics of the SIS and its association with depression were examined through exploratory factor analysis, confirmatory factor analysis and receiver operating characteristic analysis. Results The average participant SIS score was 42.66 ± 14.61 (range: 24–96) years. Analyses suggested the model had four factors: social rejection, financial insecurity, internalised shame and social isolation. The model fit statistics of the four-factor SIS were 0.913 for the comparative fit index, 0.902 for the Tucker–Lewis index and 0.088 for root-mean-square error of approximation. Standard estimated factor loadings ranged from 0.509 to 0.836. After controlling for demographic characteristics, the total score of the 23-item SIS predicted depression (odds ratio: 1.087, 95% CI 1.061–1.115; area under the curve: 0.84, 95% CI 0.788–0.892). Conclusions The Chinese version of the SIS showed good psychometric properties and can be used to assess the level of perceived stigma experienced by asymptomatic COVID-19 carriers.

Summary: Background: Neoadjuvant chemoimmunotherapy has offered novel therapeutic options for patients with locally advanced oesophageal squamous cell carcinoma (ESCC). Depicting the landscape of genomic and immune profiles is critical in predicting therapeutic responses. Methods: We integrated whole-exome sequencing, single-cell RNA sequencing, and immunofluorescence data of ESCC samples from 24 patients who received neoadjuvant treatment with PD-1 inhibitors plus paclitaxel and platinum-based chemotherapy to identify correlations with therapeutic responses. Findings: An elevation of small insertions and deletions was observed in responders. DNA mismatch repair (MMR) pathway alternations were highly frequent in patients with optimal responses and correlated with tumour infiltrating lymphocytes (TILs). Among the TILs in ESCC, dichotomous developing trajectories of B cells were identified, with one lineage differentiating towards LMO2+ germinal centre B cells and another lineage differentiating towards CD55+ memory B cells. While LMO2+ germinal centre B cells were enriched in responding tumours, CD55+ memory B cells were found to correlate with inferior responses to combination therapy, exhibiting immune-regulating features and impeding the cytotoxicity of CD8+ T cells. The comprehensive evaluation of transcriptomic B cell lineage features was validated to predict responses to immunotherapy in patients with cancer. Interpretation: This comprehensive evaluation of tumour MMR pathway alternations and intra-tumoural B cell features will help to improve the selection and management of patients with ESCC to receive neoadjuvant chemoimmunotherapy. In China, slope engineering occasionally faces landslides in rocky slopes containing muddy interlayers, primarily triggered by excavation activities. These incidents lead to considerable human casualties and substantial economic losses. However, existing studies predominantly concentrate on the excavation-induced impacts on the stability of rocky slopes characterized by single-layered soft and weak interlayers. Conversely, reports on how excavation influences the stability of bedding cataclastic rock mass high slope containing multimuddy interlayers remain notably absent in the literature. Moreover, unloading due to excavation can swiftly compromise the mechanical integrity and overall quality of the rock mass, consequently impacting the stability of slopes postexcavation. Therefore, this paper modeled the unstable slope excavated at a waste incineration power plant in Yuxi, Yunnan, using the finite element strength reduction method. This approach was employed to comprehensively simulate the entire process of artificial multilevel excavation in a bedding cataclastic rock mass high slope containing multimuddy interlayers. This study investigated the impact of multilevel artificial excavation on slope stability by thoroughly considering factors including alterations in slope morphology, unloading effects, and the degradation of geotechnical parameters. The research yielded the subsequent conclusions. Multimuddy interlayers were the key to the slope’s instability. For slopes subjected to such multilevel excavation, efforts were made to minimize the exposure of muddy interlayers. Slopes above exposed muddy interlayers did not require additional support, while those below needed prioritized reinforcement. The likely instability mode of the actual slope was local destabilization leading to landslides. Furthermore, when using numerical simulation methods to study the impact of excavation disturbances on the stability of such slopes, it was necessary to consider the deterioration of geotechnical parameters to obtain results more reflective of actual conditions. These research findings provided valuable theoretical and empirical support for studies on similar excavated slopes.

Summary: Background: Duchenne muscular dystrophy (DMD) is a disabling and life-threatening, X-linked recessive disorder caused by mutations in dystrophin. Natural history studies can inform the disease characteristics of DMD, and data from these studies can be used to plan and design clinical trials and as external controls for long-term studies. Abstract Red leaves in autumn are characteristic of a very early-maturing ripening variety of peach (Prunus persica). Analysis of the genetic factors and molecular mechanisms associated with the red-leaf phenotype can help breed peach very early maturing peach varieties. This study investigated the mechanisms underlying the red- and green-leaf phenotypes in autumn. Red compounds accumulated in the older but not younger leaves in the extremely early-maturing peach variety, ‘99-30-33’, and the leaves of the medium-maturing variety, ‘Zhongtao5 (CP5)’. Metabolic analysis showed that cyanidin-3-O-glucoside was the most abundant anthocyanin in the red leaves. The segregation of the progenies obtained from crossing revealed that the red leaves are a unique hereditary phenomenon not in line with Mendel’s law. BSA-seq and RNA-seq analyses suggest that PpNAC1 was essential for enhancing anthocyanin biosynthesis and was highly upregulated in red than in green leaves. Similar to PpNAC1, the anthocyanin activator, PpMYB10.1, was the only gene highly expressed in red leaves. Moreover, the functional genes involved in anthocyanin biosynthesis, such as Prunus persica Flavonoid 3'-hydroxylase (PpF3'H), Prunus persicaDihydroflavonol reductase (PpDFR), Prunus persica Leucoantho-cyanidin dioxygenase (Pp LDOX), Prunus persica Glutathione S-transferase (PpGST), and Prunus persica UDP-glucose, flavonoid-3-O-glucosyltransferase (PpUFGT), were upregulated in the older red leaves of 99-30-33 but downregulated in the younger 99-30-33 and green CP5 leaves. Yeast one-hybrid and dual-luciferase assays further confirmed that PpNAC1, which refers to 'Prunus persica NAC (NAM、A TAF1/2、CUC1/2)' bound to the promoter of PpMYB10.1, PpMYB10.1 stands for 'Prunus persica MYB (v-myb avian myeloblastosis viral) 10.1' and activated its expression along with those of PpGST and PpUFGT. These results provide insights into the mechanisms responsible for the development of red color in peach leaves in autumn.

Abstract Objectives This study aimed to investigate the long-term effect of vitamin D supplementation compared to placebo over 5 years in participants with knee osteoarthritis (OA). We also aimed to describe the effect of maintaining sufficient serum vitamin D levels over five years in knee OA. Methods Participants (n = 173) from the Hobart centre of the Vitamin D Effects on Osteoarthritis (VIDEO) trial were extensively followed up 3 years after the cessation of 2-year investigational treatment. Participants were classified as maintaining sufficient vitamin D (n = 79) and not maintaining sufficient vitamin D (n = 61) groups. Results There was no significant difference in change in the knee symptoms, depression, and serum levels of IL6 and hs-CRP between both comparisons after 3 years of cessation of the clinical trial. However, among participants who reported no knee surgery (KS), there was a significant improvement in WOMAC function (β: − 83.7, 95% CI: − 167.3, 0) and depression scores (β: − 1.3, 95% CI: − 2.3, − 0.2) in vitamin D group compared to the placebo group. Similarly, those who maintained adequate vitamin D levels over 5 years had significantly less WOMAC knee pain (β: − 33.9, 95% CI: − 65.7, − 2) and physical dysfunction (β: − 105.5, 95% CI: − 198.2, − 12.8) than participants with vitamin D deficiency over 5 years. Conclusion Vitamin D supplementation over 2 years or maintaining vitamin D sufficiency for 5 years was not associated with statistically significant differences in change in knee symptom scores over 5 years. However, among participants who did not report KS, 2-year vitamin D supplementation and maintaining sufficient vitamin D was linked to modest improvements in knee symptoms and depression scores in knee OA.
