Abstract Background Temperature is a crucial environmental determinant for the vitality and development of teleost fish, yet the underlying mechanisms by which they sense temperature fluctuations remain largely unexplored. Transient receptor potential (TRP) proteins, renowned for their involvement in temperature sensing, have not been characterized in teleost fish, especially regarding their temperature-sensing capabilities. Results In this study, a genome-wide analysis was conducted, identifying a total of 28 TRP genes in the mandarin fish Siniperca chuatsi. These genes were categorized into the families of TRPA, TRPC, TRPP, TRPM, TRPML, and TRPV. Despite notable variations in conserved motifs across different subfamilies, TRP family members shared common structural features, including ankyrin repeats and the TRP domain. Tissue expression analysis showed that each of these TRP genes exhibited a unique expression pattern. Furthermore, examination of the tissue expression patterns of ten selected TRP genes following exposure to both high and low temperature stress indicated the expression of TRP genes were responsive to temperatures changes. Moreover, the expression profiles of TRP genes in response to mandarin fish virus infections showed significant upregulation for most genes after Siniperca chuatsi rhabdovirus, mandarin fish iridovirus and infectious spleen and kidney necrosis virus infection. Conclusions This study characterized the TRP family genes in mandarin fish genome-wide, and explored their expression patterns in response to temperature stress and virus infections. Our work will enhance the overall understanding of fish TRP channels and their possible functions.

Abstract Iridoviruses are nucleocytoplasmic large dsDNA viruses that infect invertebrates and ectothermic vertebrates. The hypermethylated genome of vertebrate iridoviruses is unique among animal viruses. However, the map and function of iridovirus genomic methylation remain unknown. Herein, the methylated genome of Infectious spleen and kidney necrosis virus (ISKNV, a fish iridovirus), and its role in viral infection, are investigated. The methylation level of ISKNV is 23.44%. The hypermethylated genome is essential for ISKNV amplification, but there is no correlation between hypermethylation and viral gene expression. The hypomethylated ISKNV (obtained via 5-Azacytidine) activates a strong immunoreaction in vitro and reduces its pathogenicity in vivo. The unmethylated viral DNA can induce a stronger immunoreaction in vitro, whereas inactivated hypomethylated ISKNV can induce a stronger immunoreaction in vivo, suggesting ISKNV may evade from immune system by increasing its genome methylation level. Our work provides new insights into the role of genome methylation in viral infection.

Background: Infectious spleen and kidney necrosis virus (ISKNV) poses a significant threat to aquaculture sustainability, particularly affecting mandarin fish (Siniperca chuatsi) and causing significant economic losses. Methods: To address this challenge, this study developed an ISKNV Δorf037l vaccine strain, where the orf037l gene was knocked out. Infection assays conducted at 28 °C showed that the knocking out the orf037l gene decreased the virulence of ISKNV and reduced lethality against mandarin fish by 26.7% compared to wild-type ISKNV. To further diminish residual virulence, the effect of low-temperature (22 °C) immersion immunization was evaluated. Results: The results indicate that low temperature significantly diminished the virulence of the Δorf037l vaccine strain, elevating the survival rate of mandarin fish to 90%. Furthermore, the vaccine strain effectively triggered the expression of crucial immune-related genes, such as IFN-h, IL-1, IκB, Mx, TNF-α, and Viperin, while inducing the production of specific neutralizing antibodies. Low-temperature immersion with Δorf037l achieved a high relative percentage of survival of 92.6% (n = 30) in mandarin fish, suggesting the potential of Δorf037l as a promising immersion vaccine candidate. Conclusions: These findings contribute to advancing fish immersion vaccine development and demonstrate the importance and broad applicability of temperature optimization strategies in vaccine development. Our work carries profound implications for both the theoretical understanding and practical application in aquaculture disease control.

In recent years, the levels of cadmium (Cd) in agricultural soils have been increasing. Cd is highly toxic and can enter the human body through the food chain, threatening human health, therefore, reducing the Cd content in vegetables and producing green and non-polluting food has become a common concern in society. However, the physiological properties of exogenous selenium in alleviating Cd stress in cabbage seedlings have not been thoroughly investigated. In this study, exogenous Se (10 μMol/L) was applied under Cd (25 μMol/L) stress and the physiological characteristics such as biomass, photosynthetic fluorescence parameters, Se and Cd contents, chloroplast ultrastructure, leaf membrane esterification, and antioxidant enzyme activities were determined. The results showed that the exogenous application of Se could effectively alleviate the decrease in growth, photosynthetic pigment, and the gas exchange characteristics of the cabbage seedlings under Cd stress, improve cabbage root vitality, reduce root leaf Cd content, and alleviate the Cd stress-induced damage. Ultrastructural observation showed that the Cd stress caused the disruption to the chloroplasts’ internal structure in the cabbage leaves, while an exogenous Se treatment alleviated the chloroplast damage to some extent, improved the stability of the inner capsule membrane, and alleviated the Cd stress-induced damage to the photosynthetic organs. Cd stress also caused oxidative damage and the excessive accumulation of reactive oxygen species (ROS) in the leaves of cabbage seedlings, as evidenced by the significant accumulation of superoxide anion (O2−), hydrogen peroxide (H2O2), malondialdehyde (MDA), and electrolyte leakage. On the other hand, after the exogenous Se treatment, the Cd stress-induced oxidative damage could be reduced by up-regulating the activities of the antioxidant enzymes, such as superoxide dismutase (SOD), peroxidase (POD), and ascorbate peroxidase (APX). At the same time, Cd stress significantly increased glutathione (GSH) levels, and the exogenous Se treatment further increased the GSH levels, thereby increasing the tolerance of the cabbage to Cd stress. In conclusion, exogenous Se can further improve the Cd tolerance of the cabbage seedlings by protecting the photosynthetic system, eliminating excessive accumulation of reactive oxygen species under Cd stress, alleviating oxidative stress, and reducing Cd levels in plants, among other physiological properties.

Abstract Background Chronic Kidney Disease - Mineral and Bone Disorder (CKD-MBD) is a significant cause of morbidity among haemodialysis patients and is associated with pathological changes in phosphate, calcium and parathyroid hormone (PTH). In the ACTIVE Dialysis study, extended hours dialysis reduced serum phosphate but did not cause important changes in PTH or serum calcium. This secondary analysis aimed to determine if changes in associated therapies may have influenced these findings and to identify differences between patient subgroups. Methods The ACTIVE Dialysis study randomised 200 participants to extended hours haemodialysis (≥24 h/week) or conventional haemodialysis (≤18 h/week) for 12 months. Mean differences between treatment arms in serum phosphate, calcium and PTH; and among key subgroups (high vs. low baseline phosphate/PTH, region, time on dialysis, dialysis setting and frequency) were examined using mixed linear regression. Results Phosphate binder use was reduced with extended hours (− 0.83 tablets per day [95% CI -1.61, − 0.04; p = 0.04]), but no differences in type of phosphate binder, use of vitamin D, dose of cinacalcet or dialysate calcium were observed. The impairment of amyloid-β (Aβ) clearance in the brain plays a causative role in Alzheimer’s disease (AD). Polarity distribution of aquaporin-4 (AQP4) is important to remove Aβ from brain. AQP4 polarity can be influenced by the ratio of two AQP4 isoforms M1 and M23 (AQP4-M1/M23), however, it is unknown whether the ratio of AQP4-M1/M23 changes in AD. Histone deacetylase 3 has been reported to be significantly increased in AD brain. Moreover, evidence indicated that microRNA-130a (miR-130a) possibly mediates the regulation of histone deacetylase 3 on AQP4-M1/M23 ratio by repressing the transcriptional activity of AQP4-M1 in AD. This study aimed to investigate whether intermittent fasting (IF), increasing the level of an endogenous histone deacetylases inhibitor β-hydroxybutyrate, restores AQP4 polarity via miR-130a mediated reduction of AQP4-M1/M23 ratio in protection against AD. The results showed that IF ameliorated cognitive dysfunction, prevented brain from Aβ deposition, and restored the AQP4 polarity in a mouse model of AD (APP/PS1 double-transgenic mice). Additionally, IF down-regulated the expression of AQP4-M1 and histone deacetylase 3, reduced AQP4-M1/M23 ratio, and increased miR-130a expression in the cerebral cortex of APP/PS1 mice. In vitro, β-hydroxybutyrate was found to down-regulate the expression of AQP4-M1 and histone deacetylase 3, reduce AQP4-M1/M23 ratio, and increase AQP4-M23 and miR-130a expression in 2 μM Aβ-treated U251 cells. Interestingly, on the contrary to the result observed in 2 μM Aβ-treated cells, AQP4 expression was obviously decreased in cells exposed to 10 μM Aβ. miR-130a mimic decreased the expression of AQP4-M1 and the ratio of AQP4-M1/M23, as well as silencing histone deacetylase 3 caused the up-regulation of AQP4 and miR-130a, and the reduction of AQP4-M1/M23 ratio in U251 cells. In conclusion, IF exhibits beneficial effects against AD. The mechanism may be associated with recovery of AQP4 polarity, resulting from the reduction of AQP4-M1/M23 ratio. Furthermore, β-hydroxybutyrate may partly mediate the effect of IF on the reduction of AQP4-M1/M23 ratio in AD, in which miR-130a and histone deacetylase 3 may be implicated.

Alzheimer's disease is an irreversible, progressive neurodegenerative disorder. The accumulation of Aβ in the brain is thought to play a causative role in the development of cognitive dysfunction in Alzheimer's disease. The p75 neurotrophin receptor is of great importance to protect against the Aβ burden and its expression is regulated by histone acetylation. This study investigated whether the phytochemical sulforaphane, a pan-histone deacetylase inhibitor, up-regulates the p75 neurotrophin receptor expression via affecting histone acetylation in protection against Alzheimer's disease. We found that sulforaphane ameliorated behavioral cognitive impairments and attenuated brain Aβ burden in Alzheimer's disease model mice. Additionally, sulforaphane reduced the expression of histone deacetylase1, 2, and 3, up-regulated p75 neurotrophin receptor, and increased levels of acetylated histone 3 lysine 9 and acetylated histone 4 lysine 12 in the cerebral cortex of Alzheimer's disease model mice as well as in Aβ-exposed SH-SY5Y cells. Furthermore, silencing of histone deacetylase1 and 3, but not histone deacetylase2, gene expression with small interfering RNA caused up-regulation of p75 neurotrophin receptor in SH-SY5Y cells. In conclusion, this study demonstrates that sulforaphane can ameliorate neurobehavioral deficits and reduce the Aβ burden in Alzheimer's disease model mice, and the mechanism underlying these effects may be associated with up-regulation of p75 neurotrophin receptor mediated, apparently at least in part, via reducing the expression of histone deacetylase1 and 3.

Abstract Oral ulcers are a common oral mucosal disease that seriously affect the quality of life. Traditional drug treatments have shown unsatisfactory efficacy and potential adverse reactions. In this study, curcumin-loaded multifunctional magnesium metal–organic framework-embedded hyaluronic acid-soluble microneedles patches were developed to optimize treatment strategies for oral ulcers. This microneedles patch achieves efficient release of curcumin and Mg2+ in the ulcer through precisely targeted delivery and controllable release mechanism, significantly regulates inflammation, promotes cell migration and angiogenesis, and accelerates the ulcer healing process. At the same time, the synergistic effect of curcumin and gallic acid effectively alleviated oxidative stress, while the backplate ε-poly-l-lysine and needle tip Mg2+ jointly constructed an antibacterial barrier to effectively inhibit pathogens. Verification using an oral ulcer rat model showed that the microneedles patch exhibited excellent therapeutic effects. This not only opens up a new avenue for clinical oral treatment but also marks a breakthrough in nanobiomaterials science and drug delivery technology and heralds a broad prospect in the field of oral ulcer treatment in the future. Abstract Background Chlorophenoxy compounds represent a group of selective herbicides widely used around the world. Chlorophenoxy herbicides are toxic, chemically stable, and can migrate into groundwater through soil leaching, posing a significant threat to drinking water safety and human health. Chlorophenoxy herbicides in groundwater aquifers are subject to anaerobic processes; however, the pathway and microbiology involved in the attenuation of chlorophenoxy herbicides under anaerobic condition are largely unknown. Here, the anaerobic degradation process of 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), a typical chlorophenoxy herbicide, was investigated. Results The initial 52.5 ± 2.3 μM 2,4,5-T was completely degraded by a sediment-derived microbial consortium, with 3,4-dichlorophenol, 2,5-dichlorophenol, 3-chlorophenol (3-CP) and phenol being identified as the intermediate products. Reductive dechlorination of 3-CP to phenol and the subsequent elimination of phenol were the key transformation steps in the overall degradation process of 2,4,5-T. Amplicon sequencing suggested that Dehalobacter, Sulfuricurvum, Bacteroides, Acetobacterium, and Clostridium sensu stricto 7 might contribute to the transformation of 2,4,5-T to phenol, and Smithella, Syntrophorhabdus, Methanofollis and Methanosaeta likely cooperated to accomplish the complete mineralization of phenol. Conclusions This study reported the anaerobic degradation of 2,4,5-T via reductive dechlorination and the subsequent syntrophic metabolization of phenol, an intermediate product transformed from 2,4,5-T. Dehalobacter was identified as the organohalide-respiring population catalyzing the reductive dechlorination reaction. Syntrophorhabdus and methanogenic populations were likely involved in anaerobic phenol oxidation and facilitated the complete mineralization of 2,4,5-T.

Abstract Background The updated status of hypertension and its risk factors are poorly evaluated in Tibetan highland areas. We initiated a large-scale cross-sectional survey to provide updated status of hypertension and its risk factors (especially salt intake) in the Ganzi Tibetan Plateau, China. Methods Stratified multistage random sampling was performed to obtain a representative sample of 4,036 adult residents from 4 counties in the Ganzi Tibetan area. The whole survey population was used to present the epidemiology and risk factors of hypertension. The participants with blood and urine biochemistry data were used to analyze the relationship between salt intake parameters and hypertension. Results Stratified multistage random sampling was performed to obtain a representative sample of 4,036 adult residents. The overall prevalence rate of hypertension was 33.5% (the age-adjusted prevalence rate was 28.9%). A total of 50.9% of the hypertensive patients knew their conditions; 30.1% of them received antihypertensive treatment; and 11.2% of them had their blood pressure controlled. Age, male sex, living altitude ≥ 3500 m, overweight and abdominal obesity were positively correlated with hypertension. In addition, the adjusted odds ratio (OR) for hypertension was 1.33 (95% CI: 1.01–1.74) for drinking tea with salt, and 1.51 (95% CI: 1.32–1.72) for per SD increase in the estimation of 24-hour urinary sodium excretion (e24hUSE). Furthermore, per 100mmol/day increase in e24hUSE was associated with elevation of blood pressure (+ 10.16, 95% CI: 8.45–11.87 mmHg for SBP; +3.83, 95% CI: 2.74–4.93 mmHg for DBP) in this population. Conclusions Our survey suggests a heavy disease burden of hypertension in the Ganzi Tibetan Plateau. Age, male sex, altitude of residence ≥ 3500 m, overweight, abdominal obesity, and excessive salt intake (shown as drinking tea with adding salt and a higher level of e24hUSE) all increased the risk of hypertension in this highland area.

Abstract Tandem repeats (TRs) are genomic regions that tandemly change in repeat number, which are often multiallelic. Their characteristics and contributions to gene expression and quantitative traits in rice are largely unknown. Here, we survey rice TR variations based on 231 genome assemblies and the rice pan-genome graph. We identify 227,391 multiallelic TR loci, including 54,416 TR variations that are absent from the Nipponbare reference genome. Only 1/3 TR variations show strong linkage with nearby bi-allelic variants (SNPs, Indels and PAVs). Using 193 panicle and 202 leaf transcriptomic data, we reveal 485 and 511 TRs act as QTLs independently of other bi-allelic variations to nearby gene expression, respectively. Using plant height and grain width as examples, we identify and validate TRs contributions to rice agronomic trait variations. These findings would enhance our understanding of the functions of multiallelic variants and facilitate rice molecular breeding.

Abstract Background Understanding the release characteristics of soil colloids is a prerequisite for studying the co-transport of colloids and pollutants in subsurface environment. As a crucial agricultural management measure, fertilization not only alters the material composition of farmland soil, but also significantly regulates the properties and release patterns of soil colloids. This study systematically investigated the regulatory mechanism of short-term cattle manure fertilization on the macroscopic release and microscopic properties of soil colloids with different particle sizes, providing a theoretical foundation for subsequent research on the fate and transport of agricultural non-point source pollutants. Results The colloids in natural agricultural soil primarily consist of inorganic components. Graded extraction of the colloids has revealed that the combined proportion of colloids with particle sizes of 1–2 μm and 0.45–1 μm accounts for approximately 80.5%. Applying cattle manure inhibits the release of soil colloids, and the content of large particle size (1–2 μm) components increases. The content of organic colloids is increased due to the high total organic carbon (TOC) in cattle manure, particularly those with a particle size less than 1 μm. The characterization of organic colloid components revealed a significant increase in aromatic carbon and oxygen-containing functional groups, while the aliphatic content decreased. The response sequence regarding changes in functional groups within organic colloids induced by fertilization was as follows: –CH3, –CH2 > C–O > –OH > C=C. Fertilization promotes the release of 1:1-type inorganic mineral colloids, increasing the content of poorly crystalline minerals. The retention of aromatic carbon and oxygen-containing functional groups by poorly crystalline mineral colloids served as the primary mechanism leading to their increased content levels. Changes in environmental factors significantly impacted the release and properties of soil colloids. Conditions such as low cationic valence, high ionic strength, and high pH promoted the release of soil colloids. Conclusions The short-term fertilization resulted in a reduction in the release of soil colloids and brought about significant alterations in their particle size composition and properties. The findings of this study provide valuable insights into understanding the impact of fertilization-induced colloid release on the environmental behavior of agricultural non-point source pollutants.

Background Low-intensity pulsed ultrasound (LIPUS) is a special type of low-intensity ultrasound. In periodontal disease, LIPUS is applied as an adjuvant and non-invasive treatment. It has been reported that LIPUS significantly shifts the macrophage phenotype from M1 to M2, but the specific mechanism behind this shift is still unknown. Methods RAW264.7 cells were induced to M1/M2 polarization with lipopolysaccharide (LPS)/interleukin-4 (IL4). LIPUS was performed for 25 min two times, 24 h apart, at an intensity of 45 mW/cm2 to stimulate RAW264.7 cells. PolyA mRNA sequencing was conducted of both the LPS-induced RAW264.7 cells and the LPS-induced RAW264.7 cells with LIPUS treatment. The expression of Wnt2b in RAW264.7 cells was downregulated by siRNA. The macrophage surface markers and downstream inflammatory cytokines were detected using flow cytometry. The relative expression of proteins in the Wnt2b/AXIN/β-catenin pathway was assessed using reverse transcription real-time polymerase chain reaction (RT-qPCR) and Western blot. Results LIPUS reversed the M1 polarization of RAW264.7 cells, with decreased expression of CD80 and CD86. In addition, LIPUS enhanced the M2 polarization of RAW264.7 cells, with upregulated expression of CD163 and CD206. The polyA mRNA sequencing results indicated that the Wnt signaling pathway participated in the M1 polarization of LIPUS-treated RAW264.7. The results of the RT-qPCR showed a higher expression of Wnt2b in LIPUS-treated and M1- or M2-polarized RAW264.7 cells. Knocking down Wnt2b was shown to reverse the inhibitory effect of LIPUS on M1 polarization and increase the expression of CD80 and CD86. Wnt2b knockdown also regulated downstream AXIN, β-catenin, and inflammatory factors such as tumor necrosis factor alpha (TNFα) and interleukin-6 (IL6). Conclusions LIPUS plays an anti-inflammatory role by inhibiting LPS-induced M1 polarization of RAW264.7 cells in a Wnt2b/AXIN/β-catenin-dependent way. LIPUS may play a therapeutic role in periodontal diseases by inhibiting inflammation through the regulation of macrophage differentiation.

Urticaria is characterized by transient itchy symptoms on the skin, usually accompanied by swelling, which is caused by mast cell activation leading to increased vascular permeability and dilation of the dermis. Urticaria involves recurrent activation of mast cells, T cells, eosinophils, and other immune cells around lesioned venules, with complex regulatory systems affecting mast cell functions, potentially contributing to urticaria pathogenesis. The direct causal relationship between immune cells and urticaria is currently unclear. To address this, our study utilized a bidirectional Mendelian randomization analysis, employing instrumental variables (IVs) associated with immune cells and urticaria, to investigate this causal relationship. First, by utilizing Genome-wide Association Study (GWAS) data, we identified 31 immunophenotypes associated with urticaria risk, with 18 increasing and 13 decreasing the risk. Through rigorous criteria, we identified 4 immunophenotypes that have a strong causal relationship with urticaria. Notably, HLA DR+ CD4+AC, CD45 on CD8br, and HLA DR on plasmacytoid dendritic cells were associated with an increased risk, while CD8dim NKT %lymphocyte was identified as a protective factor. Sensitivity analyses, including the MR-Egger intercept test, scatter plots, funnel plots, and leave-one-out analysis, supported the robustness of the findings. Reverse MR analysis suggested an inverse causal effect of urticaria on CD8dim NKT %lymphocyte, reinforcing the potential bidirectional nature of the relationship between urticaria and immune cell phenotypes. Our research substantiates the bidirectional causal relationship between immune cells and urticaria, thus benefiting for urticaria-targeted therapy development.

Extracellular protein coronas (exPCs), which have been identified in various biofluids, are recognized for their pivotal role in mediating the interaction between nanoparticles and the cytomembrane. However, it is still unclear whether various exPCs can induce different levels of intracellular proteostasis, which is of utmost importance in preserving cellular function, and eliciting distinct intracellular biological behaviors. To investigate this, two types of exPC-coated iron oxide nanoparticles (IONPs) are prepared and used to investigate the influence of exPCs on extracellular and intracellular biological outcomes. The results demonstrate that the formation of exPCs promotes the colloidal stability of IONPs, and the discrepancies in the components of the two exPCs, including opsonin, dysopsonin, and lipoprotein, are responsible for the disparities in cellular uptake and endocytic pathways. Moreover, the differential evolution of the two exPCs during cellular internalization leads to distinct autophagy and glycolysis activities, which can be attributed to the altered depletion of angiopoietin 1 during the formation of intracellular protein coronas, which ultimately impacts the PI3K/AKT-mTOR signaling. These findings offer valuable insights into the dynamic characteristics of exPCs during cellular internalization, and their consequential implications for cellular internalization and intracellular metabolism activity, which may facilitate the comprehension of PCs on biological effects of NPs and expedite the design and application of biomedical nanoparticles.

ABSTRACT Backgroud According to the 2020 statistics from the World Health Organization’s International Agency for Research on Cancer (IARC), it is projected that there will be over 1 million new cases of gastric cancer (GC) patients worldwide in 2020, resulting in approximately 770 000 deaths. Gastric cancer ranks fifth in terms of incidence rate and forth in death rate among malignant tumors. Despite advancements in early diagnostic techniques, the incidence of GC has exhibited a marginal decline; nevertheless, the mortality rate remains elevated for advanced inoperable patients with no currently available efficacious treatment options. Recent Finding Chinese medicine (CM) has emerged as an efficacious treatment for GC, gradually gaining acceptance and widespread usage in China. It exhibits distinctive advantages in the prevention and treatment of metastasis. CM and natural medicine possess the ability to elicit antitumor effects by augmenting immune cell population, enhancing immune cell activity, and improving the tumor immune microenvironment. CMs and natural remedies encompass a diverse range of types, characterized by multiple targets, pathways, and extensive pharmacological effects. Consequently, they have become a prominent research area among oncologists worldwide. Numerous studies have demonstrated that CM and natural medicine can directly or indirectly enhance innate immune system components (including macrophages, natural killer cells, and myeloid suppressor cells), adaptive immune system elements (such as T lymphocytes and regulatory T cells), relevant cytokines (e.g., IL‐2, IL‐4, IL‐10, TNF‐α), and PD‐1/PD‐L1 axis regulation, thereby bolstering the cytotoxicity of immune cells against tumor cells. Conclusions This ultimately leads to an improved tumor immune microenvironment facilitating superior antitumor efficacy. This paper critically examines the role of CM and natural medicine in regulating immunotherapy for GC, aiming to establish a new theoretical framework for the clinical treatment and prevention of gastric cancer within the realm of CM.

Objective Few studies have examined the relationship between systemic oxidative stress and prostate cancer (PCa) risk. This study aimed to explore potential correlations between PCa and oxidative balance score (OBS), which measures systemic oxidative stress.Design A cross-sectional study.Setting The National Health and Nutrition Examination Survey.Participants A total of 8156 individuals were included in this study.Primary and secondary outcome measures Weighted logistic regression with multivariable adjustment and restricted cubic splines (RCS) were used to assess the correlation between PCa and OBS. A sensitivity analysis was conducted specifically on patients with PCa to verify the results.Results The prevalence of PCa was 2.55%. The multivariable logistic regression model revealed no correlation between OBS, dietary OBS, lifestyle OBS and PCa. Compared with the lowest quartile of OBS, the adjusted ORs for the highest quartile of OBS, dietary OBS and lifestyle OBS were 1.852 (95% CI 1.028–3.339), 1.565 (95% CI 0.841–2.913) and 1.575 (95% CI 0.915–2.710), respectively. Additionally, all p values for trend were greater than 0.05. Subgroup analysis revealed a consistent lack of association between OBS and PCa across various population settings. Furthermore, analysis using RCS confirmed this absence of association, indicating no significant relationship in either a linear or non-linear context. A sensitivity analysis focusing exclusively on patients with PCa showed a strong association (OR=2.737, p=0.008).Conclusion This cross-sectional study reveals no significant association between systemic oxidative stress, measured by OBS, and PCa risk. Notably, a sensitivity analysis focusing solely on PCa patients suggested a potential link, warranting further investigation.

In recent years, research on enjoyment in foreign language (FL) learning has flourished. To help illuminate the existing scope of inquiry and guide future research, this paper presents a systematic review of 118 empirical studies on FL learning enjoyment published between 2014 and 2023. Each study was coded according to its research context, methodological features, and research focus. The results indicate (1) a heavy focus on adult English as a foreign language (EFL) learners whose first languages are Chinese or Persian within traditional classroom learning settings; (2) a strong preference for quantitative methods; and (3) a prominent focus on enjoyment's antecedents and effects. Drawing upon these findings, we recommend that future research (1) addresses the experiences of language learners from diverse demographic backgrounds in a wider variety of learning settings; (2) applies multimodal methods to thoroughly assess the experience of enjoyment from both objective and subjective perspectives; and (3) explores the nature of enjoyable teacher-student or student-student socio-emotional interaction in greater depth.

China is currently the world’s largest carbon emitter and has pledged to achieve carbon neutrality by 2060, which requires significant reductions in emissions and the removal of carbon dioxide removal. Precise and efficient forestation has become a key strategy to increase carbon sequestration and mitigate the effects of climate change. In order to obtain a high-accuracy distribution area of potential forestation land, a variety of influencing factors on potential forestation land were selected in this study, including vegetation factors and environmental factors. Combining the two models “law of the minimum factors (LMF)” and “equal weight classification (EWC)” predicts the spatial distribution pattern of potential forestation land in China. And, we calculated the available forestation area in each province and predicted future forest coverage. The results showed that the potential forestation area using the LMF method reached approximately 5.65 × 105 km2, and the potential forestation area using the EMC method reached approximately 4.95 × 105 km2. Combining the two models, the final potential forestation area in China was approximately 6.29 × 105 km2, of which approximately 1.02 × 105 km2 was evaluated as a high-confidence area. If all potential forestation land obtained from the two models was successfully planted, China’s forest coverage rate would reach nearly 26 %. Two newly introduced vegetation factors, namely vegetation resilience and top vegetation succession, were considered in this study to make the prediction results of potential forestation distribution more accurate and reasonable. In addition, the results obtained in this study can provide certain assistance for the specific implementation of national forestation policies, and can provide a reference for promoting forestation actions and achieving the goal of carbon neutrality.

Abstract Background Zoonotic viruses cause substantial public health and socioeconomic problems worldwide. Understanding how viruses evolve and spread within and among wildlife species is a critical step when aiming for proactive identification of viral threats to prevent future pandemics. Despite the many proposed factors influencing viral diversity, the genomic diversity and structure of viral communities in East Africa are largely unknown. Results Using 38.3 Tb of metatranscriptomic data obtained via ultradeep sequencing, we screened vertebrate-associated viromes from 844 bats and 250 rodents from Kenya and Uganda collected from the wild. The 251 vertebrate-associated viral genomes of bats (212) and rodents (39) revealed the vast diversity, host-related variability, and high geographic specificity of viruses in East Africa. Among the surveyed viral families, Coronaviridae and Circoviridae showed low host specificity, high conservation of replication-associated proteins, high divergence among viral entry proteins, and frequent recombination. Despite major dispersal limitations, recurrent mutations, cocirculation, and occasional gene flow contribute to the high local diversity of viral genomes. Conclusions The present study not only shows the landscape of bat and rodent viromes in this zoonotic hotspot but also reveals genomic signatures driven by the evolution and dispersal of the viral community, laying solid groundwork for future proactive surveillance of emerging zoonotic pathogens in wildlife. Abstract Background The hemoglobin glycation index (HGI) is the difference between the observed and predicted values of glycosylated hemoglobin (HbA1c), which is closely associated with a variety of poor prognoses. However, there are still no studies on the correlation between HGI and poor prognosis in patients with critical coronary artery disease. The purpose of this study was to analyze the correlation between HGI and all-cause mortality in patients with critical coronary artery disease using the MIMIC-IV database. Methods The HGI was calculated by constructing a linear regression equation between HbA1c and fasting plasma glucose (FPG). A Kaplan‒Meier survival analysis model was constructed based on the HGI quartiles to clarify the differences in all-cause mortality rates between groups, and the log-rank test was used to assess the differences between groups. The hazard ratio (HR) of HGI as a risk factor for outcome events was assessed using the Cox proportional risk model and restricted cubic spline (RCS), with the Q2 group serving as the reference group. Abstract Background Chronic kidney disease (decreased kidney function) is common in hypertensive patients. The SIRI is a novel immune biomarker. We investigated the correlation between the SIRI and kidney function in hypertensive patients. Methods The present study analyzed data from participants who suffered from hypertension in the NHANES from 2009 to 2018. Multivariate regression analysis and subgroup analysis were used to clarify whether the SIRI was an independent risk factor for decreased kidney function. RCSs were utilized to evaluate the correlation between the SIRI and the eGFR and between the SIRI and the ACR. In addition, we modeled the mediating effect of the SIRI on the eGFR and the ACR using blood pressure as a mediating variable. Results The highest SIRI was an independent risk factor for a decreased eGFR [odds ratio (OR) = 1.46, 95% CI (1.15, 1.86)] and an increased ACR [OR = 2.26, 95% CI (1.82, 2.82)] when the lowest quartile was used as the reference. The RCS results indicated an inverted U-shaped relationship between the SIRI and the eGFR and between the SIRI and the ACR (the inflection points were 1.86 and 3.09, respectively). The mediation effect analysis revealed that the SIRI was the main factor influencing kidney function, and diastolic blood pressure was a mediating variable. In particular, there was a fully mediating effect between the SIRI and UCr, with a mediating effect value of -0.61 (-0.90, -0.36). Conclusions The association between the SIRI and renal function in hypertensive patients was significant and was particularly dominated by the association between the SIRI and the ACR. This difference may be due to the mediating effect of diastolic blood pressure.

Abstract Background Comprehending the correlation between body conformation traits of cows at the early stages of lactation and prevalent lactation diseases might facilitate the execution of selection and feeding strategies that prioritize cow health. This study aimed to evaluate the impact of body conformation traits on the incidence of clinical mastitis and lameness in Chinese Holstein cows. From a pasture herd of 1472 early lactating Chinese Holstein cows, we evaluated 20 body conformation traits. During lactation, this pasture herd was visited weekly to gather clinical mastitis and lameness data. A nine-point scale was used to determine the conformation traits of cows to clarify their linear characters, including frame capacity, rump (RU), feet and leg (FL), mammary system (MS), and dairy character. A longitudinal binary disease (0 = healthy; 1 = diseased) data structure was created by allocating disease records to adjacent official test dates. The impact of body conformation traits on the risk of developing diseases (clinical mastitis and lameness) was analyzed using the logistic regression models. The spring system functions as a pivotal element of the check valve, with its compression performance significantly influencing the valve's time, flow stability, and other characteristics. Taking cues from conventional springs, a refined rectangular helical spring with integrated support features was thoroughly evaluated for its compression performance. The design approach for this spring was elucidated, encompassing the utilization of the Finite Element Method (FEM) to model its compression behavior. Additionally, a laboratory configuration was implemented to authenticate the findings derived from the FEM simulation. Subsequently, a comparative investigation was carried out between an engineered spring and a conventional spring subjected to analogous processing. The comparative analysis unveiled that the support-featured spring exhibited a diminished lateral offset of 2.84 mm (equivalent to a reduction of 10.1 %) and a force-displacement curve with narrower vibration intervals and smaller amplitudes. Moreover, it exhibited an enhanced force feedback of 17.5 % under identical compression displacement conditions.

Summary: The Streptomyces antibiotic regulatory proteins (SARPs) are ubiquitously distributed transcription activators in Streptomyces and control antibiotics biosynthesis and morphological differentiation. However, the molecular mechanism behind SARP-dependent transcription initiation remains elusive. We here solve the cryo-EM structure of an AfsR-loading RNA polymerase (RNAP)-promoter intermediate complex (AfsR-RPi) including the Streptomyces coelicolor RNAP, a large SARP member AfsR, and its target promoter DNA that retains the upstream portion straight. The structure reveals that one dimeric N-terminal AfsR-SARP domain (AfsR-SARP) specifically engages with the same face of the AfsR-binding sites by the conserved DNA-binding domains (DBDs), replacing σHrdBR4 to bind the suboptimal −35 element, and shortens the spacer between the −10 and −35 elements. Notably, the AfsR-SARPs also recruit RNAP through extensively interacting with its conserved domains (β flap, σHrdBR4, and αCTD). Thus, these macromolecular snapshots support a general model and provide valuable clues for SARP-dependent transcription activation in Streptomyces.

Abstract Background Studies have indicated a close association between dysbiosis of the gut microbiota and chronic sinusitis. However, the causal relationship between the gut microbiota and the risk of chronic sinusitis remains unclear. Methods Using genome‐wide association study (GWAS) data for the gut microbiota and chronic sinusitis, we conducted a two‐sample Mendelian randomization (MR) study to determine the potential causal relationship between the microbiota and chronic sinusitis. We employed the inverse variance‐weighted (IVW) method as the primary analytical approach to estimate the effect. Additionally, sensitivity, heterogeneity, and pleiotropy analyses were conducted to evaluate the robustness of the results. Reverse MR analysis was also applied to investigate potential reverse causality. Results Through MR analysis, we identified 17 gut microbiota classifications that are closely associated with chronic sinusitis. However, after Bonferroni multiple correction, only class Bacilli (odds ratio: 0.785, 95% confidence interval: 0.677–0.911, p = .001, false discovery rate = 0.023) maintained a significant causal negative relationship with chronic sinusitis. Sensitivity analysis did not reveal any evidence of heterogeneity or horizontal pleiotropy. Reverse MR analysis found five gut microbiota classifications that are significantly associated with chronic sinusitis, but they were no longer significant after Bonferroni multiple correction. There was no evidence to suggest a reverse causal relationship between chronic sinusitis and class Bacilli. Conclusion Specific gut microbiota predicted by genetics exhibit a potential causal relationship with chronic sinusitis, and class Bacilli may have a protective effect on chronic sinusitis.

In recent years, the ever-rising penetration of distributed photovoltaics (PV) power has presented substantial challenges in power system dispatch due to its inherent randomness and unpredictability. To bridge this gap, this paper proposes a two-stage robust optimization method for power system security dispatch considering traditional generators as well as flexible resources, such as load demand response and energy storage systems. Specifically, a price-based demand response model is established to optimize the system’s load curve during a day. On this basis, a two-stage optimization problem for day-ahead and intra-day power system dispatch model is proposed. The dispatch objective is to minimize the overall cost in worst-case scenarios through properly scheduling unit commitment (UC) as well as flexible resources in each dispatch interval. Column and constraint generation (C&CG) algorithm is adopted for problem solving. The effectiveness of the proposed method is validated by case studies based on a modified 6-node system and a 24-node system. Simulation results indicate that through appropriately scheduling the energy storage system and load demand response, the proposed dispatch method can significantly reduce the total operation cost of a PV rich power system, which in turn facilitates the integration of PV power.

Abstract Metabolic abnormalities contribute to the pathogenesis of obesity and its complications. Yet, the understanding of the interactions between critical metabolic pathways that underlie obesity remains to be improved, in part owing to the lack of comprehensive metabolomics studies that reconcile data from both hydrophilic and lipophilic metabolome analyses that can lead to the identification and characterization of key signaling networks. Here, the study conducts a comprehensive metabolomics analysis, surveying lipids and hydrophilic metabolites of the plasma and omental adipose tissue of obese individuals and the plasma and epididymal adipose tissue of mice. Through these approaches, it is found that a significant accumulation of ceramide due to inhibited sphingolipid catabolism, while a significant reduction in the levels of uridine monophosphate (UMP), is critical to pyrimidine biosynthesis. Further, it is found that UMP administration restores sphingolipid homeostasis and can reduce obesity in mice by reversing obesity‐induced inhibition of adipocyte hypoxia inducible factor 2a (Hif2α) and its target gene alkaline ceramidase 2 (Acer2), so as to promote ceramide catabolism and alleviate its accumulation within cells. Using adipose tissue Hif2α‐specific knockout mice, the study further demonstrates that the presence of UMP can alleviate obesity through a HIF2α‐ACER2‐ceramide pathway, which can be a new signaling axis for obesity improvement.

To study the variation rule of workpiece temperature concerning machining parameters in the cylindrical transverse grinding process of gear steel, 18CrNiMo7-6 gear steel was selected as the workpiece material, and a CBN grinding wheel was used as the grinding tool. An infrared thermal imaging instrument was used to detect the temperature during the grinding process. A calibration scheme of thermal emissivity value, incorporating an additional air curtain device, was proposed. The high-speed airflow generated by the air curtain was used to slow down the entry of external air into the heating furnace, preventing the oxidation of samples during calibration, thus ensuring the accuracy of the emissivity value calibrated under high-temperature conditions. Based on the test results, the influence of workpiece speed \begin{document}${v_{\text{w}}}$\end{document}, workpiece grinding depth per revolution \begin{document}${f_{\text{a}}}$\end{document}, grinding wheel grain size \begin{document}${d_{\text{g}}}$\end{document}, grinding wheel diameter \begin{document}${d_{\text{s}}}$\end{document} and grinding width \begin{document}$b$\end{document} on the temperature was analyzed. The workpiece grinding depth per revolution \begin{document}${f_{\text{a}}}$\end{document} was identified as the most significant factor affecting the temperature change. The empirical formula between grinding temperature and aforementioned test parameters was obtained by fitting, with an error of 9.27%. The test measurement results of grinding temperature were compared with those derived from the theory of a moving heat source, Rayleigh distribution model and dry grinding heat distribution ratio model, resulting in a deviation of 8.51%.

Abstract Background Solar radiation is primarily composed of ultraviolet radiation (UVR, 200 − 400 nm) and photosynthetically active radiation (PAR, 400 − 700 nm). Ultraviolet-B (UVB) radiation accounts for only a small proportion of sunlight, and it is the primary cause of plant photodamage. The use of chlorofluorocarbons (CFCs) as refrigerants caused serious ozone depletion in the 1980s, and this had led to an increase in UVB. Although CFC emissions have significantly decreased in recent years, UVB radiation still remains at a high intensity. UVB radiation increase is an important factor that influences plant physiological processes. Ulva prolifera, a type of macroalga found in the intertidal zone, is intermittently exposed to UVB. Alternative oxidase (AOX) plays an important role in plants under stresses. This research examines the changes in AOX activity and the relationships among AOX, photosynthesis, and reactive oxygen species (ROS) homeostasis in U. prolifera under changes in UVB and photosynthetically active radiation (PAR). Results UVB was the main component of solar radiation impacting the typical intertidal green macroalgae U. prolifera. AOX was found to be important during the process of photosynthesis optimization of U. prolifera due to a synergistic effect with non-photochemical quenching (NPQ) under UVB radiation. AOX and glycolate oxidase (GO) worked together to achieve NADPH homeostasis to achieve photosynthesis optimization under changes in PAR + UVB. The synergism of AOX with superoxide dismutase (SOD) and catalase (CAT) was important during the process of ROS homeostasis under PAR + UVB. Conclusions AOX plays an important role in the process of photosynthesis optimization and ROS homeostasis in U. prolifera under UVB radiation. This study provides further insights into the response of intertidal macroalgae to solar light changes.

Shade tolerance is essential for soybeans in inter/relay cropping systems. A genome-wide association study (GWAS) integrated with transcriptome sequencing was performed to identify genes and construct a genetic network governing the trait in a set of recombinant inbred lines derived from two soybean parents with contrasting shade tolerance. An improved GWAS procedure, restricted two-stage multi-locus genome-wide association study based on gene/allele sequence markers (GASM-RTM-GWAS), identified 140 genes and their alleles associated with shade-tolerance index (STI), 146 with relative pith cell length (RCL), and nine with both. Annotation of these genes by biological categories allowed the construction of a protein–protein interaction network by 187 genes, of which half were differentially expressed under shading and non-shading conditions as well as at different growth stages. From the identified genes, three ones jointly identified for both traits by both GWAS and transcriptome and two genes with maximum links were chosen as beginners for entrance into the network. Altogether, both STI and RCL gene systems worked for shade-tolerance with genes interacted each other, this confirmed that shade-tolerance is regulated by more than single group of interacted genes, involving multiple biological functions as a gene network.

Point cloud segmentation is a crucial technique for object recognition and localization, widely employed in various applications such as point cloud registration, 3D reconstruction, object recognition, and robotic grasping. However, in practical scenarios, challenges such as sparse object features, significant pose variations, or instances of object adhesion, stacking, and occlusion can lead to difficulties and poor stability in point cloud segmentation. In response to these challenges, this paper proposes a novel point cloud segmentation algorithm based on enhanced Euclidean clustering. To address the issue of over-segmentation and under-segmentation in cases of object adhesion and stacking in unordered scenes, a method is introduced for discriminating collision points based on normal angle constraints. Subsequently, an adaptive search radius is employed to determine the clustering distance threshold, enhancing the algorithm’s ability to handle different object arrangements. Finally, the collision points are reintegrated into the clustering results to ensure the completeness of the segmented target point cloud. Experimental results using the ROBI public dataset and real-world point cloud segmentation scenarios demonstrate that the proposed algorithm effectively resolves challenges associated with the segmentation of adhering and stacking objects. The improved algorithm exhibits higher accuracy and robustness in multi-object segmentation across diverse scenes.

Effective extraction of building edge information based on high-resolution remote sensing images is the basis for efficient urban 3D modeling. Existing building extraction methods still have some problems, such as an uncertain segmentation scale, effective feature selection, and sample selection. In this paper, we propose a practical building extraction method based on convolutional network edge-enhanced attention U2-Net (EA U2-Net) to accurately achieve multi-scale extraction of buildings from remote sensing imagery. First, the U2-Net is used as the backbone network for building extraction because each stage of the network is filled by residual U-block (RSU), and the network can better aggregate multi-scale features. Second, the building edge feature map is introduced into the generation network to compensate for the problems of insufficient extracted building edge features and loss of detail. Finally, the convolutional block attention module is used to achieve effective feature extraction of buildings. We performed the experiment on the WHU building dataset, and the experimental results showed that the EA U2-Net model has significantly improved the ability to extract buildings, with an accuracy of 96.30%, a recall rate of 94.91%, f1 of 95.26%, and iou of 91.57%. This proves that EA U 2 - Net can achieve better remote sensing image-building segmentation results. Finally, in view of the problem that the deep learning network relies on training samples, this study examined the influence of the number of building samples, sample purity, and sample resolution on the effect of building extraction. The results confirmed that reasonable sample parameter settings can improve the target extraction accuracy and the optimal sample parameter combination was verified in this experiment.

A torque control strategy based on acceleration intention recognition is proposed to address the issue of insufficient power performance in linear torque control strategies for electric racing cars, aiming to better reflect the acceleration intention of racing drivers. First, the support vector machine optimized by the sparrow search algorithm is used to recognize the acceleration intention, and the running mode of the racing car is divided into two types: Starting mode and driving mode. In driving mode, based on the recognition results of acceleration intention, fuzzy control is used for torque compensation. Based on the results of simulation and hardware in the loop testing, we can conclude that the support vector machine model optimized using the sparrow search algorithm can efficiently identify the acceleration intention of racing drivers. Furthermore, the torque control strategy can compensate for positive and negative torque based on the results of intention recognition, significantly improving the power performance of the racing car.

Abstract Previous studies examining the association between hemoglobin concentration and hypertension have yielded inconsistent results. There is still a lack of evidence regarding the association between hemoglobin concentration and hypertension risk in native Tibetans at high altitude. We performed this cross‐sectional study in Luhuo County of Ganzi Tibetan Autonomous Prefecture (average altitude of 3500 m). In this study, we enrolled 1547 native Tibetans. The association between hemoglobin concentration and hypertension risk was examined by multivariate binary logistic regression and smooth curve fitting. Abstract Background Malignant pleural effusion is mostly a complication of advanced malignant tumors. However, the cancer markers such as carbohydrate antigen 125 (CA 125), carbohydrate antigen 15-3 (CA 15-3), carbohydrate antigen 19-9 (CA 19-9), and cytokeratin fragment 21-1 (CYFRA 21-1) have low sensitivity and organ specificity for detecting malignant pleural effusion. Research question Is IR808@MnO nano-near infrared fluorescent dye worthy for the diagnosis in differentiating benign and malignant pleural effusions. Study design and methods This experiment was carried out to design and characterize the materials for in vitro validation of the new dye in malignant tumor cells in the A549 cell line and in patients with adenocarcinoma pleural effusion. The dye was verified to possess tumor- specific targeting capabilities. Subsequently, a prospective hospital-based observational study was conducted, enrolling 106 patients and excluding 28 patients with unknown diagnoses. All patients underwent histopathological analysis of thoracoscopic biopsies, exfoliative cytological analysis of pleural fluid, and analysis involving the new dye. The objective of this study was to explore the relationship between calf birth traits and their susceptibility to diseases before reaching adulthood. A total of 5253 birth traits of Chinese Holstein calves were examined, including gestation length (GL), calf weight at birth (CW), and calving ease score (CES), which ranges from 1 (easy) to 5 (very difficult). Furthermore, monthly medical records were scrutinized for pneumonia and diarrhea in these calves. The study assessed five aspects of disease manifestation in calves: age at first onset, frequency of illness, longest duration of treatment, and total duration of treatment. The link between age at onset and disease manifestation prior to adulthood was analyzed using general linear models and regression models. The GL of calves significantly correlated with the risk of pneumonia, with the risk decreasing as the GL increases. A higher CES was associated with a later onset of diarrhea in calves. Furthermore, the CES was significantly negatively correlated with the duration of diarrhea treatment in calves. These results suggest that implementing different preventive measures for calves with different birth traits and modifying treatment protocols for affected calves could enhance the productivity of dairy cows and reduce losses on farms.

Using rapeseed straw as a raw material and potassium bicarbonate (KHCO3) and urea (CO(NH2)2) as modification reagents, the pyrolysis raw materials were mixed in a certain proportion, and the unmodified biochar GBC800, KHCO3-modified biochar KGBC800, and (KHCO3)/(CO(NH2)2) co-modified biochar N-KGBC800 were, respectively, prepared using the one-pot method at 800 °C. The physicochemical properties, such as surface morphology, pore characteristics, functional group distribution, and elemental composition of the three biochars, were characterized, and the adsorption performance and mechanism of the typical antibiotic tetracycline (TC) in water were studied. The results showed that the surface of GBC800 was smooth and dense, with no obvious pore structure, and both the specific surface area and total pore volume were small; the surface of KGBC800 showed an obvious coral-like three-dimensional carbon skeleton, the number of micropores and the specific surface area were significantly improved, and the degree of carbonization and aromatization was enhanced; N-KGBC800 had a coral-like three-dimensional carbon skeleton similar to KGBC800, and there were also many clustered carbon groups. The carbon layer changed significantly with interlayer gaps, presenting a multi-level porous structure. After N doping, the content of N increased, and new nitrogen-containing functional groups were formed. When the initial TC concentration was 100 mg/L, pH ≈ 3.4, the temperature was 25 °C, and the dosage of the three biochars was 0.15 g/L, the adsorption equilibrium was reached before 720 min. The adsorption capacities of GBC800, KGBC800, and N-KGBC800 for TC were 16.97 mg/g, 294.86 mg/g, and 604.71 mg/g, respectively. Fitting the kinetic model to the experimental data, the adsorption of TC by the three biochars was more in line with the pseudo-second-order adsorption kinetic model, and the adsorption isotherm was more in line with the Langmuir model. This adsorption process was a spontaneous endothermic reaction, mainly chemical adsorption, specifically involving multiple adsorption mechanisms such as pore filling, electrostatic attraction, hydrogen bonds, n−π interaction, Lewis acid–base interaction, π−π stacking, or cation −π interaction between the aromatic ring structure of the carbon itself and TC. A biochar-adsorption column was built to investigate the dynamic adsorption process of tetracycline using the three biochars against the background of laboratory pure water and salt water. The adsorption results show that the Thomas model and the Yoon–Nelson model both provide better predictions for dynamic adsorption processes. The modified biochars KGBC800 and N-KGBC800 can be used as preferred materials for the efficient adsorption of TC in water.

Abstract Background Helicobacter pylori (H. pylori) accounts for the majority of gastric cancer (GC) cases globally. The present study found that H. pylori promoted GC stem cell (CSC)‐like properties, therefore, the regulatory mechanism of how H. pylori promotes GC stemness was explored. Methods Spheroid‐formation experiments were performed to explore the self‐renewal capacity of GC cells. The expression of R‐spondin 3 (RSPO3), Nanog homeobox, organic cation/carnitine transporter‐4 (OCT‐4), SRY‐box transcription factor 2 (SOX‐2), CD44, Akt, glycogen synthase kinase‐3β (GSK‐3β), p‐Akt, p‐GSK‐3β, β‐catenin, and G protein subunit gamma 7 (GNG7) were detected by RT‐qPCR, western blotting, immunohistochemistry (IHC), and immunofluorescence. Co‐immunoprecipitation (CoIP) and liquid chromatography coupled with tandem mass spectrometry (LC–MS/MS) were performed to identify proteins interacting with RSPO3. Lentivirus‐based RNA interference constructed short hairpin (sh)‐RSPO3 GC cells. Small interfering RNA transfection was performed to inhibit GNG7. The in vivo mechanism was verified using a tumor peritoneal seeding model in nude mice. Results H. pylori extracts promoted a CSC‐like phenotype in GC cells and elevated the expression of RSPO3. RSPO3 knockdown significantly reduced the CSC‐like properties induced by H. pylori. Previous studies have demonstrated that RSPO3 potentiates the Wnt/β‐catenin signaling pathway, but the inhibitor of Wnt cannot diminish the RSPO3‐induced activation of β‐catenin. CoIP and LC–MS/MS revealed that GNG7 is one of the transmembrane proteins interacting with RSPO3, and it was confirmed that RSPO3 directly interacted with GNG7. Recombinant RSPO3 protein increased the phosphorylation level of Akt and GSK‐3β, and the expression of β‐catenin in GC cells, but this regulatory effect of RSPO3 could be blocked by GNG7 knockdown. Of note, GNG7 suppression could diminish the promoting effect of RSPO3 to CSC‐like properties. In addition, RSPO3 suppression inhibited MKN45 tumor peritoneal seeding in vivo. IHC staining also showed that RSPO3, CD44, OCT‐4, and SOX‐2 were elevated in H. pylori GC tissues. Conclusion RSPO3 enhanced the stemness of H. pylori extracts‐infected GC cells through the GNG7/β‐catenin signaling pathway.

Isorhamnetin (ISO) is a phenolic compound belonging to flavonoid family, showcasing important in vitro pharmacological activities such as antitumor, anti-inflammation, and organ protection. ISO is predominantly extracted from Hippophae rhamnoides L. This plant is well-known in China and abroad because of its “medicinal and food homologous” characteristics. As a noteworthy natural drug candidate, ISO has received considerable attention in recent years owing to its low cost, wide availability, high efficacy, low toxicity, and minimal side effects. To comprehensively elucidate the multiple biological functions of ISO, particularly its antitumor activities and other pharmacological potentials, a literature search was conducted using electronic databases including Web of Science, PubMed, Google Scholar, and Scopus. This review primarily focuses on ISO’s ethnopharmacology. By synthesizing the advancements made in existing research, it is found that the general effects of ISO involve a series of in vitro potentials, such as antitumor, protection of cardiovascular and cerebrovascular, anti-inflammation, antioxidant, and more. This review illustrates ISO’s antitumor and other pharmacological potentials, providing a theoretical basis for further research and new drug development of ISO.

Abstract Background The predictive utility of QTc values, calculated through various correction formulas for the incidence of postoperative major adverse cardiovascular and cerebrovascular events (MACCE) in patients experiencing acute myocardial infarction (AMI), warrants further exploration. This study endeavors to ascertain the predictive accuracy of disparate QTc values for MACCE occurrences in patients with perioperative AMI. Methods A retrospective cohort of three hundred fourteen AMI patients, comprising 81 instances of in-hospital MACCE and 233 controls, was assembled, with comprehensive collection of baseline demographic and clinical data. QTc values were derived employing the correction formulas of Bazett, Fridericia, Hodges, Ashman, Framingham, Schlamowitz, Dmitrienko, Rautaharju, and Sarma. Abstract Introduction Hypertension increases the risk of cardiovascular disease. Uncontrolled nocturnal blood pressure is prevalent in patients taking antihypertensive medication, with an incidence rate of 30–60%. Although chronotherapy with antihypertensive agents may provide a new direction for effective control of nocturnal blood pressure, the clinical evidence base remains controversial. This research is presently underway to compare the effects of morning and bedtime administration of antihypertensive medication on nocturnal reduction and circadian rhythm of blood pressure in patients with hypertension. Methods and analysis This study is being performed as a randomized, multicenter, open-label, parallel-group, clinical trial in which 720 participants are to undergo 24-h ambulatory blood pressure measurement (ABPM) and office blood pressure measurement (OBPM) at baseline before being randomly assigned to a morning (6–10 am) or a bedtime (6–10 pm) administration group. Each participant receives one 20/5-mg tablet of olmesartan/amlodipine (OA) daily for 4 weeks and is then followed up at 4-week intervals for a total of 12 weeks. During follow-up, the OA dosage is adjusted according to the ABPM and OBPM results. Patients with uncontrolled hypertension at the first follow-up visit will receive an increase in OA dosage to 1.5 tablets/day. For patients with blood pressure that is still uncontrolled after a further 4 weeks, the dosage of OA can be increased to 2 tablets/day. The primary objective is the reduction in mean nocturnal systolic blood pressure between baseline and week 12. The secondary objectives are the reduction in ambulatory blood pressure at weeks 4 and 12 and the blood pressure control rate at weeks 4, 8, and 12. Discussion Antihypertensive chronotherapy remains controversial. A superiority test hypothesis design has been adopted for this trial, in which all participants will be taking the same antihypertensive medication. We anticipate that our findings will determine if nocturnal blood pressure control in Chinese patients with essential hypertension varies according to whether antihypertensive medication is taken in the morning or at bedtime. This study may provide scientific evidence for the application of chronotherapy in clinical practice. Trial registration ChiCTR2200059719. Acquiring labels in anomaly detection tasks is expensive and challenging. Therefore, as an effective way to improve efficiency, pretraining is widely used in anomaly detection models， which enriches the model's representation capabilities, thereby enhancing both performance and efficiency in anomaly detection. In most pretraining methods, the decoder is typically randomly initialized. Drawing inspiration from the diffusion model, this paper proposed to use denoising as a task to pretrain the decoder in anomaly detection, which is trained to reconstruct the original noise-free input. Denoising requires the model to learn the structure, patterns, and related features of the data, particularly when training samples are limited. This paper explored two approaches on anomaly detection: simultaneous denoising pretraining for encoder and decoder, denoising pretraining for only decoder. Experimental results demonstrate the effectiveness of this method on improving model’s performance. Particularly, when the number of samples is limited, the improvement is more pronounced.

Abstract Urban railways in coastal areas are exposed to the risk of extreme weather conditions. A cost-effective and robust wind monitoring system, as a vital part of the railway infrastructure, is essential for ensuring safety and efficiency. However, insufficient sensors along urban rail lines may result in failure to detect local strong winds, thus impacting urban rail safety and operational efficiency. This paper proposes a hybrid method based on historical wind speed data analysis to optimize wind monitoring system deployment. The proposed methodology integrates warning similarity and trend similarity with a linear combination and develops a constrained quadratic programming model to determine the combined weights. The methodology is demonstrated and verified based on a real-world case of an urban rail line. The results show that the proposed method outperforms the single similarity-based method and spatial interpolation approach in terms of both evaluation accuracy and robustness. This study provides a practical data-driven tool for urban rail operators to optimize their wind sensor networks with limited data and resources. It can contribute significantly to enhancing railway system operational efficiency and reducing the hazards on rail infrastructures and facilities under strong wind conditions. Additionally, the novel methodology and evaluation framework can be efficiently applied to the monitoring of other extreme weather conditions, further enhancing urban rail safety.
