Background: Spinal cord injuries (SCIs) trigger a cascade of detrimental processes, encompassing neuroinflammation and oxidative stress (OS), ultimately leading to neuronal damage. Phillygenin (PHI), isolated from forsythia, is used in a number of biomedical applications, and is known to exhibit anti-neuroinflammation activity. In this study, we investigated the role and mechanistic ability of PHI in the activation of microglia-mediated neuroinflammation and subsequent neuronal apoptosis following SCI. Methods: A rat model of SCI was used to investigate the impact of PHI on inflammation, axonal regeneration, neuronal apoptosis, and the restoration of motor function. In vitro, neuroinflammation models were induced by stimulating microglia with lipopolysaccharide (LPS); then, we investigated the influence of PHI on pro-inflammatory mediator release in LPS-treated microglia along with the underlying mechanisms. Finally, we established a co-culture system, featuring microglia and VSC 4.1 cells, to investigate the role of PHI in the activation of microglia-mediated neuronal apoptosis. Results: In vivo, PHI significantly inhibited the inflammatory response and neuronal apoptosis while enhancing axonal regeneration and improving motor function recovery. In vitro, PHI inhibited the release of inflammation-related factors from polarized BV2 cells in a dose-dependent manner. The online Swiss Target Prediction database predicted that toll-like receptor 4 (TLR4) was the target protein for PHI. In addition, Molecular Operating Environment software was used to perform molecular docking for PHI with the TLR4 protein; this resulted in a binding energy interaction of −6.7 kcal/mol. PHI inhibited microglia-mediated neuroinflammation, the production of reactive oxygen species (ROS), and activity of the NF-κb signaling pathway. PHI also increased mitochondrial membrane potential (MMP) in VSC 4.1 neuronal cells. In BV2 cells, PHI attenuated the overexpression of TLR4-induced microglial polarization and significantly suppressed the release of inflammatory cytokines. Conclusion: PHI ameliorated SCI-induced neuroinflammation by modulating the TLR4/MYD88/NF-κB signaling pathway. PHI has the potential to be administered as a treatment for SCI and represents a novel candidate drug for addressing neuroinflammation mediated by microglial cells. The translational potential of this article: We demonstrated that PHI is a potential drug candidate for the therapeutic management of SCI with promising developmental and translational applications.

This study aims to explore how the environment affects the variation and trade-offs of anatomical traits in shrub plants, contributing to a deeper understanding of plant survival strategies and mechanisms of adaptation to the environment. The dominant shrub species Corylus mandshurica in the broad-leaved Korean pine (Pinus koraiensis) forest was selected as the research subject. Current-year twigs and leaves of C. mandshurica were collected in spring (May), summer (July), and autumn (September). Anatomical traits, including non-vascular column proportion (NVP), vascular column proportion (VP), phloem proportion (PP), and xylem proportion (XP), were measured for twigs, lower part of petioles, upper part of petioles, and leaf veins. We found that (1) Compared with seasons, organ type explained most of the variation in the four traits (NVP = 96 %, VP = 87 %, PP = 81 %, XP = 76 %). Lower part of petioles served as the turning point in the variation trend of tissue proportions in cross-sectional structure along the sequence of twigs, lower part of petioles, upper part of petioles, and leaf veins. (2) In spring, C. mandshurica exhibits higher NVP but lower VP and PP, while the results in summer and autumn are opposite to those in spring. Among different organs, twigs show lower NVP but higher VP and XP; whereas petioles and leaf veins show the opposite pattern. (3) Both hydrothermal conditions (monthly precipitation and mean monthly temperature) and light (canopy openness and total transmitted radiation) were the dominant environmental factors influencing anatomical trait variation. In different seasons, the response of NVP and VP in C. mandshurica twigs to dominant environmental factors was opposite to that of lower part of petioles, upper part of petioles, and leaf veins. The response of cross-sectional anatomical traits of current-year twigs, petioles, and leaf veins in C. mandshurica to seasonal changes exhibits distinct characteristics. Furthermore, twigs, petioles, and leaf veins demonstrate distinct survival strategies. Future research should focus on the variation of traits in different plant organs and their connecting parts.

Abstract Ubiquitin-specific protease 3 (USP3) plays an important role in the progression of various tumors. However, the role of USP3 in osteosarcoma (OS) remains poorly understood. The aim of this study was to explore the biological function of USP3 in OS and the underlying molecular mechanism. We found that OS had higher USP3 expression compared with that of normal bone tissue, and high expression of USP3 was associated with poor prognosis in patients with OS. Overexpression of USP3 significantly increased OS cell proliferation, migration, and invasion. Mechanistically, USP3 led to the activation of the PI3K/AKT signaling pathway in OS by binding to EPHA2 and then reducing its protein degradation. Notably, the truncation mutant USP3-F2 (159–520) interacted with EPHA2, and amino acid 203 was found to play an important role in this process. And knockdown of EPHA2 expression reversed the pro-tumour effects of USP3-upregulating. Thus, our study indicates the USP3/EPHA2 axis may be a novel potential target for OS treatment.

Helicobacter pylori causes globally prevalent infections that are highly related to chronic gastritis and even development of gastric carcinomas. With the increase of antibiotic resistance, scientists have begun to search for better vaccine design strategies to eradicate H. pylori colonization. However, while current strategies prefer to formulate vaccines with a single H. pylori antigen, their potential has not yet been fully realized. Outer membrane vesicles (OMVs) are a potential platform since they could deliver multiple antigens. In this study, we engineered three crucial H. pylori antigen proteins (UreB, CagA, and VacA) onto the surface of OMVs derived from Salmonella enterica serovar Typhimurium (S. Typhimurium) mutant strains using the hemoglobin protease (Hbp) autotransporter system. In various knockout strategies, we found that OMVs isolated from the ΔrfbP ΔfliC ΔfljB ΔompA mutants could cause distinct increases in immunoglobulin G (IgG) and A (IgA) levels and effectively trigger T helper 1- and 17-biased cellular immune responses, which perform a vital role in protecting against H. pylori. Next, OMVs derived from ΔrfbP ΔfliC ΔfljB ΔompA mutants were used as a vector to deliver different combinations of H. pylori antigens. The antibody and cytokine levels and challenge experiments in mice model indicated that co-delivering UreB and CagA could protect against H. pylori and antigen-specific T cell responses. In summary, OMVs derived from the S. Typhimurium ΔrfbP ΔfliC ΔfljB ΔompA mutant strain as the vector while importing H. pylori UreB and CagA as antigenic proteins using the Hbp autotransporter system would greatly benefit controlling H. pylori infection.

Abstract Chronic risk factors for pseudoaneurysm (PSA) or penetrating aortic ulcer (PAU) have not been fully clarified. This study aims to evaluate the association of aortic calcification with PSA or PAU of different etiologies. Totally 77 pseudoaneurysms, 80 PAU, and 160 healthy controls (HCs) were retrospectively included, of which 30 were infected, 34 were immunological, and 93 were atherosclerotic etiologies. The aortic calcification status, position of aortic tears/ulcers, and risk factors for disease or acute aortic syndrome (AAS) were identified. Atherosclerotic patients aged more than 65 and infective patients aged more than 60 had significantly higher calcification scores. The immunological group had a lower level of calcification in the infrarenal aorta. For patients of infective or atherosclerotic etiology, 60% (18/30) and 60.22% (56/93) of the tears/ulcers occurred at the aortic parts with the highest level of calcification. Patients with longitudinal calcification exceeding 1/3 of the aortic arch had an increased risk of acquiring diseases (OR = 13.231). The presence of longitudinal calcification of the descending aorta or cross-sectional calcification of the infrarenal aorta increased the risks of acquiring diseases (OR = 8.484 and 8.804). After adjusting for age, longitudinal calcification of the descending aorta exceeding 1/3 length was found to be associated with AAS (OR = 4.662). Tears/ulcers of pseudoaneurysm and PAU were both generally found at the part of the aorta with most calcification. Distinct aorta calcification characteristics were observed for lesions of different etiologies. Longitudinal thoracic and cross-sectional infrarenal abdominal aortic calcification increased the risk of acquiring diseases, and descending aortic calcification was associated with symptomatic patients.

Abstract Introduction Renal artery stenosis (RAS) is a significant reason for secondary hypertension. Impaired renal function and subsequent cardiopulmonary dysfunction could also occur. Patients of non-atherosclerotic RAS has a relatively young age and long life expectancy. Revascularization with percutaneous transluminal angioplasty (PTA) is a viable treatment option. However, restenosis is unavoidable which limits its use. Drug-coated balloon (DCB) has been proven to be effective in restenosis prevention in femoropopliteal arterial diseases and in patients with renal artery stenosis. And PTA for Renal artery fibromuscular dysplasia is safe and clinically successful. Therefore, we could speculate that DCB might have potential efficacy in non-atherosclerotic RAS treatment. Methods and analysis This will be a randomized multi-center-controlled trial. Eighty-four eligible participants will be assigned randomly in a 1:1 ratio to the control group (plain old balloon, POB) and the experimental group (DCB). Subjects in the former group will receive balloon dilatation alone, and in the latter group will undergo the DCB angioplasty. The DCB used in this study will be a paclitaxel-coated balloon (Orchid, Acotec Scientific Holdings Limited, Beijing, China). Follow-up visits will be scheduled 1, 3, 6, 9, and 12 months after the intervention. Primary outcomes will include controlled blood pressure and primary patency in the 9-month follow-up. Secondary outcomes will include technical success rate, complication rate, and bail-out stenting rate. Trial registration ClinicalTrials.gov (number NCT 05858190). Protocol version V.4 (3 May 2023).

In this paper, based on the regional ensemble CMA-REPS V3.1 system, the algorithm for the neighborhood ensemble probability method of precipitation is optimized. The daily 24-hour accumulated precipitation data from May to July 2021 are selected to calculate the neighborhood probability of precipitation. The grid precipitation product combined from three sources developed by the National Meteorological Information Center is selected as the observational data. The optimized method is evaluated using the area scoring method with the relative operating characteristic curve, and is compared with the scoring results of original neighborhood ensemble probability method and ensemble mean neighborhood probability method. At the same time, a typical precipitation case is selected to evaluate these three methods. It is found that the optimized method has the highest score, and its predicted information of precipitation is more consistent with the observations. In this paper, the three precipitation neighborhood probability prediction results are also used to calculate the FSS (Fractions Skill Score) of ensemble precipitation. It is found that the FSS score based on the optimized method is higher than that of ensemble mean neighborhood probability method. Both the optimized and original methods have some advantages in terms of the FSS score. The former one has better scoring for small precipitation, especially for light rain and moderate rain, and the latter one for large precipitation, especially, the scores of rainstorm is better. FSS score based on the optimized method is relatively more objective.

Plant water use efficiency (WUE) is a comprehensive physiological indicator of plant growth and ability to adapt to drought. However, research on the mechanisms controlling WUE during plant growth and development remains weak. Here, we studied Pinus koraiensis as a typical evergreen conifer species in Northeast China. After collecting 80 tree samples with varying diameters at breast height (DBH), we measured δ13C and δ18O as an indicator of WUE, leaf morphology (volume, dry weight, and total epidermal area), ecological stoichiometry (carbon, nitrogen, and phosphorus content), and abiotic factors (light environment, soil pH, soil water content, and soil nutrient content). Correlational analysis of these variables revealed distinct differences between smaller/younger and larger/older plants: (1) In plants with DBH less than 52 cm, δ13C was positively related to DBH, and δ18O was negatively related to DBH. Plants with DBH greater than 52 cm showed no relationship between δ13C and DBH, and δ18O was positively related to DBH. (2) In plants with DBH less than 52 cm, there was a negative correlation between δ13C and δ18O and between δ13C and leaf phosphorus content (LP), but a positive correlation between δ13C and DBH, leaf mass per area (LMA), and leaf density (LD). The slopes of DBH-δ13C, δ18O-δ13C, leaf nitrogen content (LN)-δ13C, and LMA-δ13C correlations were greater in smaller plants than large plants. (3) Structural equation modelling showed that in smaller plants, DBH had a direct positive effect on δ13C content and a direct negative effect on δ18O, and there was a direct positive effect of light environment on δ18O. In larger plants, there was a direct negative effect of light environment on δ13C and a direct positive effect of DBH on light environment, as well as a negative effect of soil nitrogen content on leaf nitrogen. In smaller plants, DBH was the most important factor influencing δ13C, followed by δ18O and soil moisture, with light and soil pH showing minimal influence. In larger plants, light environment influenced δ13C the most, followed by soil nitrogen content and soil moisture content, with leaf nitrogen and DBH contributing little. The results suggest that water use efficiency strategies of P. koraiensis vary according to growth stage, and the effects of abiotic factors and functional traits vary at different growth stages.

Abstract The canonical correspondence analysis (CCA) is a multivariate direct gradient analysis method performing well in many fields, however, when it comes to approximating the unimodal response of species to an environmental gradient, which still assumes that the relationship between the environment and the weighted species score is linear. In this work, we propose a nonlinear canonical correspondence analysis method (NCCA), which first determines the most appropriate nonlinear explanatory factor through two screenings by correlation and LASSO regression, and successively uses the linear regression method and the improved heuristic optimal quadratic approximation method to fit the chi-square transformation values of the response variables. Thus, our method effectively reflects the nonlinear relationship between the species and the environment factors, and a biplot is employed to visualize the effects of the later on the distribution of species. The results from applying this method over a real dataset show that the NCCA method not only maintains the advantages of the polynomial canonical correspondence analysis (PCCA) proposed by Makarenkov (2002), but also outperforms Makarenkov’s method in explaining the variance of response variables.

Litter decomposition is an important source of soil organic carbon, and it plays a key role in maintaining the stability of forest ecosystems. The microbial mechanism of soil organic carbon (SOC) formation in different urban forest planting patterns during litter lignocellulose degradation is still unclear. The key genes, microbes, and metabolites in the process of lignocellulose degradation and SOC formation were determined by metagenomics and metabolomics in different litter decomposition layers and soil layers in different urban forest planting patterns, including three types of broadleaf forests (BP forests), three types of coniferous forests (CP forests), and two types of mixed coniferous and broadleaf forests (MCBP forests). The results indicated that the cellulose, hemicellulose, and lignin concentrations from the undecomposed layer to the totally decomposed layer decreased by 70.07, 86.83, and 73.04% for CP litter; 74.30, 93.80, and 77.55% for BP litter; and 62.51, 48.58, and 90.61% for MCBP litter, respectively. The soil organic carbon of the BP forests and MCBP forests was higher than that of the CP forests by 38.06 and 94.43% for the 0–10 cm soil layer and by 38.55 and 20.87% for the 10–20 cm soil layer, respectively. Additionally, the gene abundances of glycoside hydrolases (GHs) and polysaccharide lyases (PLs) in the BP forests were higher than those in the MCBP forests and CP forests. Amino acid metabolism, sugar metabolism, TCA metabolism, and cAMP signaling metabolism were mainly between the CP forests and BP forests, while the TCA cycle, pyruvate metabolism, phenylalanine metabolism, and tyrosine metabolism were mainly between the BP forests and MCBP forests during litter decomposition. Additionally, ammonia nitrogen and hemicellulose were key factors driving SOC formation in the CP forests, while ammonia nitrogen, hemicellulose, and lignocellulose-degrading genes were key factors driving SOC formation in the BP forests. For the MCBP forests, cellulose, pH, ammonia nitrogen, and lignin were key factors driving SOC formation. Our findings revealed that the BP forests and MCBP forests had stronger lignocellulose degradation performance in the formation of SOC. This study provided a theoretical basis for the flow and transformation of nutrients in different urban forest management patterns.

Abstract Lung cancer is the most lethal malignancies with high aggressive and poor prognosis. Until now, the five-year survival rate has not been improved which brings serious challenge to human health. Lung cancer stem cells (LCSCs) serve as the root of cancer occurrence, progression, recurrence, and drug resistance. Therefore, effective anti-cancer agents and molecular mechanisms which could specifically eliminate LCSCs are urgently needed for drug design. In this article, we discovered Olig2 was overexpressed in clinical lung cancer tissues and acted as a transcription factor to regulate cancer stemness by regulating CD133 gene transcription. The results suggested Olig2 could be a promising target in anti-LCSCs therapy and new drugs targeted Olig2 may exhibit excellent clinical results. Furthermore, we verified ACT001, a guaianolide sesquiterpene lactone in phase II clinical trial with excellent glioma remission, inhibited cancer stemness by directly binding to Olig2 protein, inducing Olig2 ubiquitination degradation and inhibiting CD133 gene transcription. All these results suggested that Olig2 could be an excellent druggable target in anti-LCSCs therapy and lay a foundation for the further application of ACT001 in the treatment of lung cancer in clinical.

Abstract Aims This study was performed to identify and summarize systematic reviews focusing on the prevention of unplanned endotracheal extubation in the intensive care unit. Design Overview of systematic reviews. Methods This overview was conducted according to the Preferred Reporting Items for Overviews of Systematic Reviews, including the harms checklist. A literature search of PubMed, the Cochrane Library, CINAH, Embase, Web of Science, SINOMED and PROSPERO was performed from January 1, 2005–June 1, 2021. A systematic review focusing on unplanned extubation was included, resulting in an evidence summary. Results Thirteen systematic reviews were included. A summary of evidence on unplanned endotracheal extubation was developed, and the main contents were risk factors, preventive measures and prognosis. The most important nursing measures were restraint, fixation of the tracheal tube, continuous quality improvement, psychological care and use of a root cause analysis for the occurrence of unplanned endotracheal extubation. Conclusions This overview re‐evaluated risk factors and preventive measures for unplanned endotracheal extubation in the intensive care unit, resulting in a summary of evidence for preventing unplanned endotracheal extubation and providing direction for future research. Background Lung cancer (LC) is the most prevalent cancer with a poor prognosis. Semaphorin4A (Sema4A) is important in many physiological and pathological processes. This study aimed to explore the role and mechanism of Sema4A in LC. Methods Firstly, Sema4A expression was analyzed by the available dataset and detected in human normal bronchial epithelial cell line (HBE) and LC cell line (NCI-H460). Then, LC cells were transfected with Sema4A siRNA, and the cells were stimulated by PlexinB1, PlexinB2, PlexinD1 blocking antibodies, IgG antibody, BAY 11-7082 (an inhibitor for NF-κB pathway) and Sema4A-Fc protein, alone or in combination. After transfection, PlexinB1 mRNA expression was analyzed. Next, the biological functions, including proliferative, migratory, invasive abilities and viability of the cells were detected by colony formation, scratch, Transwell and MTT assays, respectively. NF-κB, Stat3 and MAPK protein expressions were determined by western blot. Furthermore, the secretion of IL-6 in LC cells was tested by ELISA. Results Sema4A was highly expressed in LC tissues and cells, could activate the NF-κB pathway and upregulate PlexinB1 mRNA expression. Furthermore, we observed that Sema4A knockdown suppressed the biological functions of NCI-H460 cells, while Sema4A-Fc protein reversed the situation. However, Sema4A-induced biological functions and activation in the NF-κB pathway were inhibited by PlexinB1 blocking antibody. Consistently, Sema4A promoted IL-6 production, which was down-regulated by PlexinB1 blocking antibody and BAY 11-7082. Conclusions Sema4A may facilitate LC development via the activation of the NF-κB pathway mediated by PlexinB1, suggesting that Sema4A would be a novel therapeutic target for LC treatment.

Abstract Purpose Current research has shown a link between ABO blood group and many diseases. The purpose of this study aimed to investigate the influence of the ABO blood group on the risk of developing different pathological types of lung cancer. Materials and methods This retrospective study was composed of 7681 patients with lung cancer and 12, 671 non-lung cancer patients who were admitted to the First Affiliated Hospital of Nanchang University from January 2016 to January 2021. The subjects with lung cancer were grouped into small cell lung cancer group (n = 725), lung adenocarcinoma group (n = 4520), and lung squamous cell carcinoma group (n = 2286) according to pathological types. The ABO blood group distribution of each lung cancer type group was compared with that of the control group. Statistical analysis was determined with chi-square and logistic regression. Results Univariate analysis showed that the ABO blood group distribution of lung adenocarcinoma, lung squamous cell carcinoma, and small cell lung cancer was different from that of the control group (P < 0.01). After adjusting for age, sex, smoking history, and drinking history, logistic regression analysis showed that the risk of lung adenocarcinoma in blood type O was higher than that in blood type A (P < 0.01). There was no significant difference in ABO blood group composition between small cell lung cancer group, lung squamous cell carcinoma group, and control group (P > 0.05). In addition, gender and age have an influence on all three types of lung cancer (P < 0.01). Smoking was a risk factor in lung squamous cell carcinoma and small cell carcinoma (P < 0.01). Alcohol consumption was a risk factor in lung adenocarcinoma (P < 0.01). Conclusion ABO blood group may be correlated with the occurrence of lung adenocarcinoma in Jiangxi province, but not with lung squamous cell carcinoma and small cell carcinoma.

The multidimensionality of leaf traits allows plants to have diverse survival strategies to adapt to complex living environments. Whether the anatomical traits of leaves are associated with leaf economic traits and which group of traits are more strongly correlated with soil fertility factors remains unclear. We measured four leaf economic traits, four anatomical traits, and five soil fertility factors of eight coexisting broadleaf species distributed in mixed broadleaved-Korean pine (Pinus koraiensis) forests located in Northeast China. Results show a strong interdependence between economic and anatomical traits (p < 0.05). The range of variation between economic and anatomical traits were almost equal, but the causes of variation were different. Specific leaf area was positively correlated with the abaxial epidermis, negatively correlated with the ratio of spongy tissue to leaf thickness (ST/LT), and not correlated with adaxial epidermis. Leaf dry matter content was negatively correlated with the abaxial epidermis and adaxial epidermis, positively correlated with ST/LT. Specific leaf area, palisade tissue, and ST/LT showed stronger correlation with soil fertility factors than other traits. Soil fertility factors dominating trait variation were dependent upon the trait. Our results suggest anatomical traits can be considered in economic trait dimension. The coupled relationship between anatomical and economic traits is potentially a cost-effective adaptation strategy for species to improve efficiency in resource utilization. Our results provide evidence for the complex soil-trait relationship and suggest that future studies should emphasize the role of anatomic traits in predicting soil fertility changes.

IntroductionVaricose veins are a common chronic disease that creates a significant economic burden on the healthcare system. Current treatment options, including pharmacological treatments, are not always effective, and there is a need for more targeted therapies. A Mendelian randomization (MR) method uses genetic variants as instrumental variables to estimate the causal effect of an exposure on an outcome, and it has been successful in identifying therapeutic targets in other diseases. However, few studies have used MR to explore potential protein drug targets for varicose veins.MethodsTo identify potential drug targets for varicose veins of lower extremities, we undertook a comprehensive screen of plasma protein with a two-sample MR method. We used recently reported cis-variants as genetic instruments of 2,004 plasma proteins, then applied MR to a recent meta-analysis of genome-wide association study on varicose veins (22,037 cases and 437,665 controls). Furthermore, pleiotropy detection, reverse causality testing, colocalization analysis, and external replication were utilized to strengthen the causal effects of prioritized proteins. ObjectiveTo explore the results of hypertension improvement and renal function preservation after renal artery aneurysm (RAA) repair.MethodsThis study retrospectively analyzed the change in blood pressure (BP) and renal outcomes of 59 RAA patients throughout either open or endovascular operations and follow-up at a large center. Patients were grouped according to the difference in their BP at the last follow-up vs. their baseline value. Logistic regression was conducted to explore risk factors for perioperative BP relief and long-term hypertension reonset. Previous studies of RAA with records of BP, blood creatinine level, or GFR/eGFR results are reviewed.ResultsHypertension was observed in 62.7% (37/59) of the patients included. Postoperative BP declined from 132.20 ± 16.46/79.92 ± 9.64 mmHg to 122.41 ± 11.17/71.10 ± 9.82 mmHg, while eGFR changed from 108.17 ± 24.73 to 98.92 ± 23.87 ml/min/1.73 m2. The median follow-up was 854 [IQR: 1,405] days. Both open and endovascular techniques significantly relieved hypertension and did not impair renal function much. Lower preoperative systolic BP (SBP) was significantly associated with hypertension relief (OR = 0.83, 95% CI: 0.70–0.99). Among patients with normal BP after the operation, higher postoperative SBP was significantly associated with new-onset hypertension (OR = 1.14, 95% CI: 1.01–1.29). Literature review indicated that renal function usually remained normal at follow-up, whereas relief of hypertension varied.ConclusionPatients with lower preoperative SBP were likely to benefit more from the operation, while higher postoperative SBP indicated a higher chance of hypertension reonset. Creatinine level and eGFR generally remained stable regardless of operation type.

The composition of non-structural carbohydrates (NSC), one of the photosynthetic products of plants, reflects the survival strategy of a plant. Although several studies have investigated variation of NSC content in woody plants over a set time scale, few studies have considered the dynamic variation of NSC over a continuous life history. In this study, the leaves, new twigs, and old branches of seven temperate broad-leaved tree species (diffuse-porous species: Betula platyphylla, Betula costata, Tilia amurensis, Acer pictum subsp. mono; ring-porous species: Ulmus davidiana var. japonica, Ulmus laciniata, Fraxinus mandshurica) were observed at three life history stages (seedling, sapling, and mature tree) to measure the dynamic changes of NSC and its influencing factors throughout the entire life cycles of these species. The results showed that life history, wood type, and environmental factors (soil nitrogen and phosphorus content, soil pH) significantly affected the NSC content in leaves and branches (including both new twigs and old branches). As plants grew, the NSC content in the leaves and branches generally showed an upward trend, meaning the total non-structural carbohydrate (TNC) content and soluble sugar (SS) content increased significantly, and the starch (ST) content was relatively stable. Lastly, there was no significant difference in NSC content between the canopy layers of mature trees. This indicates that the influence of life stage on NSC content in leaves and branches of plants may be dominated by genetics instead of being regulated by light factors.

Investigating the spatial distributions and associations of tree populations provides better insights into the dynamics and processes that shape the forest community. Korean pine (Pinus koraiensis) is one of the most important tree species in broad-leaved Korean pine mixed forests (BKMFs), and little is known about the spatial point patterns of and associations between Korean pine and community-level woody species groups such as coniferous and deciduous trees in different developmental stages. This study investigated the spatial patterns of Korean pine (KP) trees and then analyzed how the spatial associations between KP trees and other tree species at the community level vary in different BKMFs. Extensive data collected from five relatively large sample plots, covering a substantial area within the natural distribution range of KP in northeastern China, were utilized. Uni- and bivariate pair correlation functions and mark correlation functions were applied to analyze spatial distribution patterns and spatial associations. The DBH (diameter at breast height) histogram of KP trees in northeastern China revealed that the regeneration process was very poor in the Changbai Mountain (CBS) plot, while the other four plots exhibited moderate or expanding population structures. KP trees were significantly aggregated at scales up to 10 m under the HPP null model, and the aggregation scales decreased with the increase in size classes. Positive or negative spatial associations were observed among different life stages of KP trees in different plots. The life history stages of the coniferous tree group showed positive spatial associations with KP saplings and juvenile trees at small scales, and spatial independence or negative correlations with larger KP trees at greater scales. All broad-leaved tree groups (canopy, middle, and understory layers) exhibited only slightly positive associations with KP trees at small scales, and dominant negative associations were observed at most scales. Our results demonstrate that mature KP trees have strong importance in the spatial patterns of KP populations, and site heterogeneity, limited seed dispersal, and interspecific competition characterize the spatial patterns of KP trees and community-level spatial associations with respect to KP trees, which can serve as a theoretical basis for the management and restoration of BKMFs in northeastern China.

The resorption of nutrients by plants before litter fall and the mineralization of nutrients from plant litter by soil processes are both important pathways supporting primary productivity. While the positive relationship between plant biodiversity and primary productivity is widely accepted for natural ecosystems, the roles of nutrient resorption and mineralization in mediating that relationship remains largely unknown. Here, we quantified the relative importance of nitrogen (N) resorption and N mineralization in driving plant community N investment and the correlation between species diversity and community productivity along an N-limited successional chronosequence of the mixed broadleaved–Korean pine (Pinus koraiensis) forest in northeastern China. Leaf N resorption efficiency (NRE) at the community level increased significantly along the successional chronosequence, whereas litter N mineralization rate decreased significantly. Leaf NRE was more important than litter N mineralization rate in driving the diversity–productivity relationship. However, higher leaf NRE led to less N mineralization as succession progressed along the chronosequence. Our results highlight the importance of the N resorption pathway rather than the N mineralization pathway for forest N acquisition with community succession, and they provide mechanistic insights into the positive effects of biodiversity on ecosystem functioning. In future forest management practices, we recommend appropriate application of N fertilizer to mitigate the adverse effects of N-poor soil on seedling regeneration during late succession and thus maintain the sustainable development of temperate forest ecosystems.

Introduction: Chronic heart disease (CHD) is a common comorbidity of patients receiving endovascular aneurysm repair (EVAR) for abdominal aortic aneurysms (AAA). The explicit relationship between ventricular systolic function and EVAR complication of thrombotic events is unknown.Methods: In this study, we proposed a three-dimensional numerical model coupled with the lumped-elements heart model, which is capable of simulating thrombus formation in diverse systolic functions. The relation of cardiac functions and the predicted risk of thrombus formation in the aorta and/or endograft of 4 patients who underwent EVAR was investigated. Relative risks for thrombus formation were identified using machine-learning algorithms.Results: The computational results demonstrate that thrombus tended to form on the interior side of the aorta arch and iliac branches, and cardiac function can affect blood flow field and affect thrombus formation, which is consistent with the four patients' post-operative imaging follow-up. We also found that RRT, OSI, TAWSS in thrombosis area are lower than whole average. In addition, we found that the thrombus formation has negative correlations with the maximum ventricular contractile force (r = −.281 ± .101) and positive correlations with the minimum ventricular contractile force (r = .238 ± .074), whereas the effect of heart rate (r = −.015 ± .121) on thrombus formation is not significant.Conclusion: In conclusion, changes in ventricular systolic function may alter the risk of thrombotic events after EVAR repair, which could provide insight into the selection of adjuvant therapy strategies for AAA patients with CHD.

Abstract Background Hepatocellular carcinoma (HCC) is the fourth leading cause of cancer-related death. Traditional Chinese medicine (TCM) has special advantages in relieving HCC, while Astragalus membranaceus is commonly used in TCM treatment. However, its underlying mechanisms for treatment of HCC are unclear. Methods Differentially expressed genes (DEGs) of Astragalus membranaceus treatment in HepG2 cells were identified, and Astragalus membranaceus-gene network was constructed. The hub genes were then obtained via protein-protein interaction (PPI) analysis. Kyoto Encyclopedia of Genes and Genomes (KEGG), Gene Ontology (GO), and Gene Set Enrichment Analysis (GSEA) were subsequently performed. Furthermore, prognosis genes related to HCC from The Cancer Genome Atlas Program (TCGA) was identified to explore the correlation between Astragalus membranaceus treatment and prognosis of HCC. Finally, Astragalus membranaceus-component-target network was established through SymMap. Results Twenty five DEGs (15 up-regulated and 10 down-regulated) of Astragalus membranaceus treatment in HepG2 cells were identified. Abstract Background Mycotic aortic aneurysm is a rare and potentially life-threatening lesion, and endovascular repair has become increasingly accepted for intervention. Fenestrated endografts are available options to treat aneurysms involving visceral arteries. Here, we first report two patients with mycotic aortic aneurysm involving paraviscereal aorta who were successfully treated with custom-made fenestrated endograft. Case presentation Two patients were presented with mycotic aortic aneurysm. Due to their comorbidities and the involvement of the renal arteries, company-manufactured fenestrated stents were designed. Meanwhile, antibiotic therapy was administrated for 2 months before endovascular repair. Patients improved well without complications. Conclusions Custom-made fenestrated endovascular stent is an effective and feasible alternative solution to mycotic paravisceral aorta aneurysm.

ObjectiveThis article explores the effect of preoperative health education, in the form of animation videos, on postoperative self-reported pain levels and anxiety in femoral fractures.MethodsNinety cases of femoral fracture were divided at random into the oral instruction group, the recorded video group, and the animation video group, with 30 cases in each group. Sociodemographic data were collected the day before surgery. Health education was then offered in one of three ways: orally, using a recorded video, or using an animation video. On days 2, 4, and 7 after surgery, the state-trait anxiety inventory (STAI) and the visual analog scale (VAS) were used to assess postoperative anxiety and pain levels, respectively, in the participants.ResultsAt different time points during the evaluation, total anxiety scores in the animation and recorded video groups were significantly lower than in the oral instruction group (P < 0.01), and the pairwise comparisons indicated statistically significant differences (F = 11.04, 10.06, 10.37, P < 0.01). However, the levels of postoperative pain in the animation and recorded video groups were not significantly different (P > 0.05). STAI scores in the three groups were found to have significant interactions with the measurement time (F = 6.74, P < 0.01). However, there were no apparent interactions between the VAS score and the measurement time (F = 1.31, P > 0.05) in the three groups.ConclusionPreoperative health education with the aid of multimedia is more effective than oral instruction in lowering patients’ postoperative anxiety and pain levels. In addition, animation videos are superior to recorded videos in mitigating postoperative anxiety. Whether the two approaches differ in reducing postoperative pain in bone fractures remains to be further tested.

The timely discovery of low-frequency oscillations in power systems and accurate identification of their modal parameters is critical in numerous applications. Therefore, we investigated the feasibility of using multi-channel signals and established a relative theory. An algorithm based on the underdetermined blind source separation (UBSS) algorithm was proposed using this theory. First, the energy ratio function was used to determine the fault occurrence time. Then, the Bayesian information criterion was used to estimate the number of fault sources, and the boundary conditions were set to determine the number of fault sources. Next, the UBSS algorithm was used to analyze raw data, extract individual components that characterize faults, and subsequently measure low-frequency oscillation modal parameters through the Hilbert transform. Finally, the fast independent component analysis (FastICA) algorithm was used to separate noise signal from raw data. This separation considerably reduced noise disturbance and ensured the stability of the proposed method. Model simulation was conducted in MATLAB and experimental measurement revealed that the proposed method effectively reduced noise disturbance and could be applied to conditions with considerable disturbance.

Background: Among cancer-related deaths, hepatocellular carcinoma (HCC) ranks fourth, and traditional Chinese medicine (TCM) treatment is an important complementary alternative therapy for HCC. Curcumin is a natural ingredient extracted from Curcuma longa with anti-HCC activity, while the therapeutic mechanisms of curcumin remain unclear, especially on ferroptosis and cuproptosis. Methods: Differentially expressed genes (DEGs) of curcumin treatment in PLC, KMCH, and Huh7 cells were identified, respectively. The common genes among them were then obtained to perform functional enrichment analysis and prognostic analysis. Moreover, weighted gene co-expression network analysis (WGCNA) was carried out for the construction of the co-expression network. The ferroptosis potential index (FPI) and the cuproptosis potential index (CPI) were subsequently used to quantitatively analyze the levels of ferroptosis and cuproptosis. Finally, single-cell transcriptome analysis of liver cancer was conducted. Results: We first identified 702, 515, and 721 DEGs from curcumin-treated PLC, KMCH, and Huh7 cells, respectively. Among them, HMOX1, CYP1A1, HMGCS2, LCN2, and MTTP may play an essential role in metal ion homeostasis. By WGCNA, grey60 co-expression module was associated with curcumin treatment and involved in the regulation of ion homeostasis. Furthermore, FPI and CPI assessment showed that curcumin had cell-specific effects on ferroptosis and cuproptosis in different HCC cells. In addition, there are also significant differences in ferroptosis and cuproptosis levels among 16 HCC cell subtypes according to single-cell transcriptome data analysis. Conclusions: We developed CPI and combined it with FPI to quantitatively analyze curcumin-treated HCC cells. It was found that ferroptosis and cuproptosis, two known metal ion-mediated forms of programmed cell death, may have a vital effect in treating HCC with curcumin, and there are significant differences in various liver cancer cell types and curcumin treatment which should be considered in the clinical application of curcumin.

ObjectiveIliac arteriovenous fistulas (IAVFs) are rare abnormalities with limited reported cases. This study aimed to summarize the clinical features and experiences on the diagnosis and endovascular treatment of IAVFs.MethodsA single-center retrospective study of IAVFs from 2010 to 2019 was performed. Data including demographics, clinical characteristics, radiological images, surgical details, and follow-up were collected.ResultsA total of 16 patients diagnosed with IAVFs were identified. The female to male ratio was 3:1. The mean age was 47.7 ± 10.4 years (range: 35–73 years). Leg swelling and cardiac insufficiency, especially heart failure, were the most common primary symptoms in this series, which were revealed in 68.8 and 37.5% of patients, respectively. Iatrogenic, traumatic, and congenital IAVFs were diagnosed in 62.5, 12.5, and 25.0% of patients, respectively, among which hysterectomy was thought to be the main cause for female iatrogenic IAVFs (31.3%). Anatomic analysis found that internal iliac vessels were the predilected sites of IAVFs. All the patients were treated by endovascular procedures composed of transarterial embolization (50.0%), transarterial and stage II transvenous embolization (31.3%), stage I transarterial and transvenous embolization (12.5%), and transarterial embolization plus stent implantation (6.3%). The clinical success rate was 93.8%. Minor complications including fever (18.8%) and exudation at the puncture point (6.25%) were noted and well-treated. During a follow-up period of 51.3 ± 41.9 months after operations, only one patient experienced re-emergence of heart failure because of recurrence of leiomyosarcoma; other patients recovered uneventfully with symptoms relief and no severe embolization-related complications was encountered.ConclusionIAVFs are rare disorders and correct diagnosis requires careful history taking and physical examination, combined with proper imaging investigation. The primary goal for treatment of IAVFs was to relieve associated symptoms. Based on the results of this study, endovascular approach is safe and effective for treatment of IAVFs.

Non-structural carbohydrates (NSCs), including soluble sugar (SS) and starch (ST), are essential for the growth and survival of plants. However, few studies have investigated the effects of phenology, needle age, and life history together on NSCs. We measured NSCs of leaves for 120 Pinus koraiensis individuals of various sizes (0.3–100 cm diameter at breast height, DBH) across needle age and during three leaf development periods to evaluate the effects of phenology, needle age, and life history on NSCs. Concentration ranges were 6.60–12.07, 2.38–8.90 and 10.97–16.18 g·100 g−1 dry mass for SS, ST, and NSC, respectively. Needle age accounted for most of the variations (42.2%-84.4%) of NSCs, followed by phenology (9.2%-46.8%), while life history accounted for only a small proportion. In general, NSC reached its maximum value during the leaf unfolding period and the shift between SS and ST occurred during the leaf fall period. The slope of NSC with phenology generally increased with increasing needle age (absolute value). With increasing needle age, NSC and its components showed gradual upward trends. The slope of NSC with needle age decreased with leaf development (except during the leaf fall period). Life history had significant effects on NSCs in the leaf fall period for both current year and perennial needles, and in the leaf unfolding period only for the current year needles. Needle age significantly affected the correlations of NSC and SS (or ST) only at the leaf unfolding period. Phenology had significant effects on the correlations of NSC and SS (or ST) only for current year needle. Our results clearly showed that the variations of NSC and its components were highly dependent on needle age, followed by phenology. Unexpectedly, the effects of life history on NSCs were much less than those of needle age and phenology. Therefore, needle age and phenology dominate over life history in influencing NSCs, and the ecological significance of NSCs variations need to be reconsidered by effects of incorporating needle age and phenology.

ObjectiveThe objective of this study was to establish and validate novel individualized nomograms for predicting the overall survival (OS) and cancer-specific survival (CSS) in cervical cancer patients with lymph node metastasis.MethodsA total of 2,956 cervical cancer patients diagnosed with lymph node metastasis (American Joint Committee on Cancer, AJCC N stage=N1) between 2000 and 2018 were included in this study. Univariate and multivariate Cox regression models were applied to identify independent prognostic predictors, and the nomograms were established to predict the OS and CSS. The concordance index (C-index), calibration curves, and receiver operating characteristic (ROC) curves were applied to estimate the precision and discriminability of the nomograms. Decision-curve analysis (DCA) was used to assess the clinical utility of the nomograms.ResultsTumor size, log odds of positive lymph nodes (LODDS), radiotherapy, surgery, T stage, histology, and grade resulted as significant independent predictors both for OS and CSS. The C-index value of the prognostic nomogram for predicting OS was 0.788 (95% CI, 0.762–0.814) and 0.777 (95% CI, 0.758–0.796) in the training and validation cohorts, respectively. Meanwhile, the C-index value of the prognostic nomogram for predicting CSS was 0.792 (95% CI, 0.767–0.817) and 0.781 (95% CI, 0.764–0.798) in the training and validation cohorts, respectively. The calibration curves for the nomograms revealed gratifying consistency between predictions and actual observations for both 3- and 5-year OS and CSS. The 3- and 5-year area under the curves (AUCs) for the nomogram of OS and CSS ranged from 0.781 to 0.828. Finally, the DCA curves emerged as robust positive net benefits across a wide scale of threshold probabilities.ConclusionWe have successfully constructed nomograms that could predict 3- and 5-year OS and CSS of cervical cancer patients with lymph node metastasis and may assist clinicians in decision-making and personalized treatment planning.

Localization and mapping in a dynamic scene is a crucial problem for the indoor visual simultaneous localization and mapping (SLAM) system. Most existed visual odometry (VO) or SLAM systems are based on the assumption that the environment is static. The performance of a SLAM system may degenerate when it is operated in a severely dynamic environment. The assumption limits the applications of RGB-D SLAM in the dynamic environment. In this paper, we propose a workflow to segment the objects accurately, which will be marked as the potentially dynamic-object area based on the semantic information. A novel approach for motion detection and removal from the moving camera is introduced. We integrate the semantics-based motion detection and the segmentation approach with an RGB-D SLAM system. To evaluate the effectiveness of the proposed approach, we conduct the experiments on the challenging dynamic sequences of TUM-RGBD datasets. The experimental results suggest that our approach improves the accuracy of localization and outperforms the state-of-the-art dynamic-removal-based SLAM system in both severely dynamic and slightly dynamic scenes.

Soy yogurt is plant-based dairy of great nutritional interest that is widely accepted in developing countries as a milk alternative. Poor stability has been an urgent problem to solve of soy yogurt products over past several years. The present study aimed to construct multiple network composite gel by adding low acyl gellan gum (LAG) to improve the stability. The effect of addition of LAG on property of soy yogurt was investigated by determining water holding capacity, texture, rheology, particle size, and zeta potential. The results showed that water holding capacity was significantly higher than control. The soy yogurt with 0.1% LAG had a stable gel network with much gel strength and viscosity, and strengthened interaction between complex gel. The addition of LAG increased the particle size and decreased zeta potential. Furthermore, sensory properties were acceptable. Therefore, during industrial production, LAG could act as an appropriate stabilizer to inhibit poor body and bring more desirable sensory characteristics of soy yogurt.

DEDEQIPSHPPR, the calcium-binding peptide (CBP) identified in soy yogurt, was proven to be a potential cofactor in osteoporosis prevention in our previous study, but the mechanism was unknown. In this study, the activity of alkaline phosphatase (ALP) and osteocalcin (OCN), the regulation of RUNX2, and the expression of TβRI were investigated to elucidate the underlying mechanism. The results show that CBP upregulated ALP activity and OCN concentration and increased the expression of RUNX2 and the activation of the MAPK signaling pathway. Similarly, the expression of osteogenesis-related genes in osteoblasts also increased upon CBP treatment. Moreover, the CBP-induced enhancement of ALP activity and phosphorylation levels in the p38 pathway was inhibited by treatment with a p38 inhibitor (SB203538) and TβRI inhibitor (SB431542), respectively, suggesting that p38 and TβRI were involved in the osteogenic action. Based on the signaling pathways, the intracellular calcium concentration was significantly elevated by CBP, which was correlated with the increased behavioral functions and the relative fluorescence intensity of the bone mass. These findings suggest that CBP stimulates osteoblast differentiation and bone mineralization through the activation of RUNX2 via mechanisms related to the TβRI-p38-MAPK signaling pathways, further highlighting CBP’s important potential for treating osteoporosis.

Background: NSCLC (non-small-cell lung cancer) is the leading cause of cancer-related mortality worldwide. Both epigenetic and genetic changes contribute to the initiation, development and metastasis of NSCLC. Recently, accumulating data have begun to support the notion that long noncoding RNAs (lncRNAs) function as new crucial regulators of diverse biological processes, including proliferation, apoptosis and metastasis, and play crucial roles in tumorigenesis. Nevertheless, further study is warranted to comprehensively determine lncRNAs' functions and potential mechanism. Methods: In this study, we performed a comprehensive analysis of the lncRNA expression profile of NSCLC using data from TCGA and Gene Expression Omnibus (GEO). PCAT6 expression level in a cohort of 60 pairs of NSCLC tissues using quantitative real-time PCR (qRT-PCR). Additionally, Loss-of-function assays and gain-of-function assays were used to assess the role of PCAT6 in promoting NSCLC progression. Tumor formation assay in a nude mouse model was performed to verity the role of PCAT6 in NSCLC in vivo. Meanwhile, RIP, ChIP, resue experiment and western blot assays were used to highlights the potential molecular mechanism of PCAT6 in NSCLC. Findings: We identified that an oncogene, PCAT6, was upregulated in NSCLC, and this upregulation was verified in a cohort of 60 pairs of NSCLC tissues. Additionally, the expression level of PCAT6 was correlated with tumor size (P = .036), lymph node metastasis (P = .029) and TNM stage (P = .038). Loss-of-function and gain-of-function assays were used to assess the role of PCAT6 in promoting NSCLC progression. The results revealed that PCAT6 knockdown mitigated NSCLC cell growth by inducing G1-phase cell cycle arrest and apoptosis in vitro and in vivo. Whereas, PCAT6 overexpression could promoted tumor cell growth. Meanwhile, PCAT6 additionally promoted NSCLC cell migration and invasion. Furthermore, mechanistic investigation demonstrated that the oncogenic activity of PCAT6 is partially attributable to its repression of LATS2 via association with the epigenetic repressor EZH2 (Enhancer of zeste homolog 2). Overall, our study highlights the essential role of PCAT6 in NSCLC, suggesting that PCAT6 might be a potent therapeutic target for patients with NSCLC. Plant functional traits play an important role in plant growth and community composition in forest ecosystem. However, how the intraspecific trait variations for both leaf and branch were driven by organ age is less clear. We chose three evergreen coniferous species in the mixed broadleaved-Korean pine (Pinus koraiensis) forest in Northeast China and measured the functional traits for leaf and branch across organ ages. We mainly evaluated the influences of needle or branch age on the intraspecific trait (both leaf and branch) variations and bivariate correlation of traits for three species. We found that organ age had the largest contributions to the intraspecific variations in most traits for the three species. Current organ had high specific leaf area, high leaf nitrogen content, high specific branch length and low leaf and branch dry matter content, while perennial organ showed the opposite trend, indicating the differentiation of ecological strategies at different organ ages. The organ age significantly modified most of the trait-trait relationships, indicating that the transformation of the plant ecological strategy was driven by organ age. Unexpectedly, the variation in plant hydraulic traits was not affected by branch age and decoupled with most other traits for the three species. Our results suggest that the organ age effect should be incorporated into future studies of trait-based ecology for evergreen coniferous species.

Objective Popliteal artery entrapment syndrome is a rare cause of popliteal artery aneurysms. We present a rare case of a false aneurysm associated with popliteal artery entrapment syndrome that was treated with endovascular repair that initially failed. Case report A 60-year-old man with a false popliteal artery aneurysm and limb ischemia was treated with endovascular repair that initially failed. The popliteal artery was suspected to be compressed by an abnormal bundle of muscle according to the findings of a subsequent magnetic resonance imaging examination. The popliteal artery was entrapped by an abnormal slip of the medial gastrocnemius muscle head. Parts of the popliteus muscle were also involved in compression of the popliteal artery, which was not distinguished on preoperative magnetic resonance imaging. Thus, the patient was diagnosed with a mixed type of popliteal artery entrapment syndrome (types III and IV). Bypass with the small saphenous vein was then performed. The patient was finally discharged with satisfactory relief of his ischemic symptoms. Conclusion Popliteal artery entrapment syndrome should be considered before treating popliteal artery aneurysms, especially atypical pseudoaneurysms without significant atherosclerosis. Definitive surgical management rather than endoluminal treatment is required unless combined with open decompressive surgery to correct the musculotendinous anatomy.

Neurodegenerative diseases encompass a wide variety of pathological conditions caused by a loss of neurons in the central nervous system (CNS) and are severely debilitating. Exosome contains bio-signatures of great diagnostic and therapeutic value. There is proof that exosomal proteins can be biomarkers for Alzheimer’s disease (AD) and Parkinson’s disease (PD). MicroRNAs in exosome has potential to be an important source of biomarkers for neurodegenerative diseases. Here, we report exosomal microRNA performance of human plasma in neurodegenerative diseases by small RNA sequencing. A wide range of altered exo-miRNA expression levels were detected in both AD and PD patients. Down-regulated miRNAs in AD samples were enriched in ECM-receptor interaction pathway and both up-/down-regulated miRNAs in PD samples were enriched in fatty acid biosynthesis pathway. Compared to the control, 8 miRNAs were found to be significantly elevated/declined in AD and PD samples, of which 4 miRNAs were newly identified. Additionally, two exosome isolating methods were compared and the reproducibility of plasma exo-miRNA expression was confirmed, suggesting the feasibility of large-scale clinical application of this method. This study revealed exo-miRNA expression levels in neurodegenerative diseases, proposed new biomarkers and their potential functional pathway for AD and PD, confirmed the reproductivity of exo-miRNA profiles by using a different exosome isolating method, and compared the results with plasma miRNA expression. Therefore, this study also provides a precedent for identifying exosomal biomarkers of neurodegenerative diseases in plasma by high-throughput sequencing and it could extend the therapeutic repertoire of exosomal biomarkers.

Abstract Background Long noncoding RNAs (lncRNAs) have emerged as critical regulators in a variety of human cancers, including gastric cancer (GC). However, the function and mechanisms responsible for these molecules in GC are not fully understood. In our previous study, we found that GC associated lncRNA HOXA11-AS is significantly upregulated in GC tissues. Over-expressed HOXA11-AS promotes GC cells proliferation and invasion through scaffolding the chromatin modification factors PRC2, LSD1 and DNMT1. Methods HOXA11-AS expression levels in GC cells was detected by quantitative real-time PCR (qPCR). HOXA11-AS siRNAs and overexpression vector were transfected into GC cells to down-regulate or up-regulate HOXA11-AS expression. In vitro and in vivo assays were performed to investigate the functional role of HOXA11-AS in GC cells cell cycle progression, invasion and metastasis. RIP and ChIP assays were used to determine the mechanism of HOXA11-AS’s regulation of underlying targets. Results We found that knockdown of HOXA11-AS induced GC cells G0/G1 phase arrest and suppressed GC cells migration, invasion and metastasis in vivo. Moreover, mechanistic investigation showed that HOXA11-AS could interact with WDR5 and promote β-catenin transcription, bind with EZH2 and repress P21 transcription, and induce KLF2 mRNA degradation via interacting with STAU1. Conclusions Taken together, these findings show that HOXA11-AS not only could promote GC cells migration and invasion in vitro, but also promotes GC cells metastasis in vivo, at least in part, by regulating β-catenin and KLF2.

Urban forests affect the filtration and absorption of airborne particulate matter, which can minimize the harmful effects to human health caused by airborne particulate pollution. Evergreen plants in urban forests play a major role in absorbing dust and purifying the air, especially in winter. Studying the spatial distribution of leaf dust and exploring the dust retention effect of evergreen shrubs are important for scientifically guiding urban forest construction and improving the living environment of cities in winter. The purpose of this study was to establish a dust inversion model by correlation analysis of spectral reflectance and the amount of dust absorption (ADA) of vegetation, using Sentinel-2 satellite remote-sensing images to obtain the dust distribution of the evergreen vegetation (mainly Euonymus japonicus Thunb.) in the Beijing urban area, and to determine the effect of the spatial pattern of E. japonicus woodland on ADA intensity. The result showed that the red band and near-infrared band are most sensitive to dust. The normalized difference phenology index (NDPI) is more suitable for building an inversion model, where the determination coefficient (R2) of the inversion model constructed by the ratio of the NDPI (RNDPI) was 0.879. The inversion results show that the mean ADA in the enclosed area is smaller than that in semi-enclosed and open areas, and the regional distribution of high ADA in the urban area of Beijing was higher in the south with a tendency of the ADA to decrease from city center to the surrounding area. The size, shape, and percentage of landscape (PLAND) of E. japonicus woodland have a significant effect on ADA intensity. We found that, in the study area, when the PLAND of E. japonicus woodland is higher than 40%, its ADA intensity remains basically unchanged. When the vegetation coverage is fixed, the landscape shape index is negatively correlated with ADA intensity, and reduction of the overall shape complexity of forestland can enhance its dust retention effect and improve the air environment of the surrounding areas. The results of this study can be used as a reference for urban planners and landscape architects when building urban forests, providing a scientific basis for controlling and reducing air particulate pollution in Beijing’s winter and improving the living environment.

Spinal tuberculosis is still prevalent in some developing countries. The purpose of this study is to investigate the efficacy and safety of one-stage posterior debridement, autogenous bone grafting, and transpedicular screw fixation in treating monosegmental thoracic and lumbar tuberculosis in adults. 37 patients were retrospectively reviewed in this study. The data of images, operative time and blood loss volume, perioperative complications, time to achieve bony fusion, VAS score, and neurologic function preoperatively and postoperatively were collected. The mean follow-up period was 21.5±3.5 months. The tuberculosis was cured after surgery in all patients, and no recurrence was observed. Bony fusion was achieved in all patients with a mean time of 5.6±2.5 months. Neurological outcome did not change in one case with grade A, and increased by 1–3 grades in the other patients with nerve deficit. HOTAIR, a long intervening non-coding RNA (lincRNA), associates with the Polycomb Repressive Complex 2 (PRC2) and is reported to reprogram chromatin organization and promote tumor progression. However, little is known about the roles of this gene in the development of chemoresistance phenotype of lung adenocarcinoma (LAD). Thus, we investigated the involvement of HOTAIR in the resistance of LAD cells to cisplatin. In this study, we show that HOTAIR expression was significantly upregulated in cisplatin-resistant A549/DDP cells compared with in parental A549 cells. Knockdown of HOTAIR by RNA interference could resensitize the responses of A549/DDP cells to cisplatin both in vitro and in vivo. In contrast, overexpression of HOTAIR could decrease the sensitivity of A549 and SPC-A1 cells to cisplatin. We also found that the siRNA/HOTAIR1-mediated chemosensivity enhancement was associated with inhibition of cell proliferation, induction of G0/G1 cell-cycle arrest and apoptosis enhancement through regulation of p21(WAF1/CIP1) (p21) expression. Also, pcDNA/p21or siRNA/p21 could mimic the effects of siRNA/HOTAIR1 or pcDNA/HOTAIR on the sensitivity of LAD cells to cisplatin. Importantly, siRNA/p21 or pcDNA/p21 could partially rescue the effects of siRNA/HOTAIR1 or pcDNA/HOTAIR on both p21 expression and cisplatin sensitivity in LAD cells. Further, HOTAIR was observed to be significantly downregulated in cisplatin-responding LAD tissues, and its expression was inversely correlated with p21 mRNA expression. Taken together, our findings suggest that upregulation of HOTAIR contributes to the cisplatin resistance of LAD cells, at least in part, through the regulation of p21 expression.

The energy cost of a rail transport system is very big especially for the tractions of trains in its daily operation. Continual effort on decreasing the traction energy cost intensities of various rail transport modes has been made by many researchers and practitioners on different aspects for a long time. From the rail transport operation perspective, this study reviews such energy-saving research mainly focusing on the optimizations of train coasting schemes, the rational designs and utilizations of track alignments, the ameliorations of train attributes, and the improvements of system operations. According to the review work, it is confirmed that in sound responses to distinct track alignments, dynamically optimizing control programs of trains with their reasonably improved attributes ought to be further studied in view of the systematic transport operation of a rail line or network in an integrated manner as much as possible in the future research on traction energy saving of rail transport.

Leaf area index (LAI) is an important variable in the study of forest ecosystem processes, but very few studies are designed to monitor LAI and the seasonal variability in a mixed forest using non-destructive sampling. In this study, first, true LAI from May 1(st) and November 15(th) was estimated by making several calibrations to LAI as measured from the WinSCANOPY 2006 Plant Canopy Analyzer. These calibrations include a foliage element (shoot, that is considered to be a collection of needles) clumping index measured directly from the optical instrument, TRAC (Tracing Radiation and Architecture of Canopies); a needle-to-shoot area ratio obtained from shoot samples; and a woody-to-total area ratio. Second, by periodically combining true LAI (May 1(st)) with the seasonality of LAI for deciduous and coniferous species throughout the leaf-expansion season (from May to August), we estimated LAI of each investigation period in the leaf-expansion season. Third, by combining true LAI (November 15(th)) with litter trap data (both deciduous and coniferous species), we estimated LAI of each investigation period during the leaf-fall season (from September to mid-November). Finally, LAI for the entire canopy then was derived from the initial leaf expansion to the leaf fall. The results showed that LAI reached its peak with a value of 6.53 m(2) m(-2) (a corresponding value of 3.83 m(2) m(-2) from optical instrument) in early August, and the mean LAI was 4.97 m(2) m(-2) from May to November using the proposed method. The optical instrument method underestimated LAI by an average of 41.64% (SD = 6.54) throughout the whole study period compared to that estimated by the proposed method. The result of the present work implied that our method would be suitable for measuring LAI, for detecting the seasonality of LAI in a mixed forest, and for measuring LAI seasonality for each species.
