Drought stress adversely impacts crop development and yield. Maize frequently encounters drought stress during its life cycle. Improvement of drought tolerance is a priority of maize breeding programs. Here, we identified a novel transcription factor encoding gene, APETALA2 (AP2)/Ethylene response factor (ERF), which is tightly associated with drought tolerance in maize seedlings. ZmERF21 is mainly expressed in the root and leaf and it can be highly induced by polyethylene glycol treatment. Genetic analysis showed that the zmerf21 mutant plants displayed a reduced drought tolerance phenotype, accompanied by phenotypical and physiological changes that are commonly observed in drought conditions. Overexpression of ZmERF21 in maize significantly increased the chlorophyll content and activities of antioxidant enzymes under drought conditions. RNA-Seq and DNA affinity purification sequencing analysis further revealed that ZmERF21 may directly regulate the expression of genes related to hormone (ethylene, abscisic acid) and Ca signaling as well as other stress-response genes through binding to the promoters of potential target genes. Our results thereby provided molecular evidence of ZmERF21 is involved in the drought stress response of maize.

In order to improve the level of solid waste comprehensive utilization and reduce the cost of filling raw materials for mining, a study is carried out on the preparation of cementing agents (CA) for underground mine filling using secondary smelting water-granulated nickel slag (SWNS) as the main raw material. The mechanical properties test, X-ray diffraction (XRD), flourier transform-infrared spectroscopy (FT-IR), thermogravimetric/differential scanning calorimeter (DSC-TG), scanning electron microscope (SEM) and other test methods were used to study the working performance and mechanical properties of the cemented paste backfill (CPB) material, and the types of hydration products, micro-morphology and hydration characteristics of CA of SWNS for CPB material were analyzed. The results show that when the ratio of CA used for CPB material is SWNS:cement clink (CC):Na2SO4:flue gas desulphurization gypsum (FGDG):carbide slag (CS) = 85:2:3:5:5, the binder content on dry basis of tailings (CA/iron ore tailings (IOT)) 1:4, the slurry concentration 82%, and the addition of 0.156% PC water-reducer, the static compressive strength of CPB material 28 days is 7.94 MPa, which is 57% higher under dynamic loading conditions than that under static loading, the tensile strength is increased by 72%, and the shear strength is increased by 127%, which meets the strength requirements of CPB materials used in underground mines. CA of SWNS is a CPB material containing glass phase and crystalline materials. Under the action of various activators, it can participate in the hydration reaction, and the main hydration products are ettringite (AFt) and hydrated aluminosilicate gel material (C–M–S–H) containing Ca2+ and Mg2+; (C–M–S–H) is the main source of strength in the later stage of CPB material.

Flowering time is an important agronomic trait that determines the distribution and adaptation of plants. The accurate prediction of flowering time in elite germplasm is critical for maize breeding. However, the molecular mechanisms underlying the photoperiod response remain elusive in maize. Here we cloned the flowering time-controlling gene, ZmNF-YC2, by map-based cloning and confirmed that ZmNF-YC2 is the nuclear transcription factor Y subunit C-2 protein and a positive regulator of flowering time in maize under long-day conditions. Our results show that ZmNF-YC2 promotes the expression of ZmNF-YA3. ZmNF-YA3 negatively regulates the transcription of ZmAP2. ZmAP2 suppresses the expression of ZMM4 to delay flowering time. We then developed a gene regulatory model of flowering time in maize using ZmNF-YC2, ZmNF-YA3, ZmAP2, ZMM4, and other key genes. The cascading regulation by ZmNF-YC2 of maize flowering time has not been reported in other species.

This paper establishes shipping enterprises' safety culture system model based on the dynamic calculation matrix model. This paper calculates the deep learning theory's shipping safety early warning system. At the same time, we use the fuzzy evaluation method to establish the relevant weights of the safety culture index system. The method model mentioned in this paper effectively avoids the subjectivity of the pure expert evaluation method. We use experimental simulations to verify the effectiveness of our model. Experiments show that the system in this paper has the advantages of reliable prediction results and high precision.

Maize is a model plant species often used for genetics and genomics research because of its genetic diversity. There are prominent morphological, genetic, and epigenetic variations between tropical and temperate maize lines. However, the genome-wide chromatin conformations of these two maize types remain unexplored. We applied a Hi-C approach to compare the genome-wide chromatin interactions between temperate inbred line D132 and tropical line CML288. A reconstructed maize three-dimensional genome model revealed the spatial segregation of the global A and B compartments. The A compartments contain enriched genes and active epigenome marks, whereas the B compartments are gene-poor, transcriptionally silent chromatin regions. Whole-genome analyses indicated that the global A compartment content of CML288 was 3.12% lower than that of D132. Additionally, global and A/B sub-compartments were associated with differential gene expression and epigenetic changes between two inbred lines. About 25.3% of topologically associating domains (TADs) were determined to be associated with complex domain-level modifications that induced transcriptional changes, indicative of a large-scale reorganization of chromatin structures between the inbred maize lines. Furthermore, differences in chromatin interactions between the two lines correlated with epigenetic changes. These findings provide a solid foundation for the wider plant community to further investigate the genome-wide chromatin structures in other plant species.

Esophageal squamous cell carcinoma (ESCC), the main subtype of esophageal cancer (EC), is a common lethal type of cancer with a high mortality rate. The aim of the present study was to select key relevant genes and identify potential mechanisms involved in the development of ESCC based on bioinformatics analysis. Minichromosome maintenance 6 complex component (MCM6) has been identified to be upregulated in multiple malignancies; however, its contributions to ESCC remain unclear. For the purposes of the present study, four datasets were downloaded from the Gene Expression Omnibus (GSE63941, GSE26886, GSE17351 and GSE77861), and the intersection of the differentially expressed genes was obtained using a Venn diagram. The protein‑protein interaction was then constructed, and the modules were verified by Cytoscape, in which the key genes have a high connectivity degree with other genes. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway were subsequently filtered out to analyze the development of ESCC. MCM6, an upregulated gene, was selected and connected with most of the other genes, for further research validation. The expression levels of MCM6 were then assessed using the Oncomine, GEPIA and UALCAN databases and validated in both ESCC tissues samples and cell lines by immunohistochemistry and RT‑qPCR. Cell counting kit‑8 (CCK‑8), flow cytometry, wound healing and Transwell assays were used to determine the proliferation, apoptosis, cell cycle, migration and invasion of ESCC cells. A total of 24 genes were identified by a series of bioinformatics analyses and the results revealed that the genes were associated with DNA replication and cell cycle. Experimental validation revealed that MCM6 expression was significantly elevated in both ESCC tissues and cell lines. The results were consistent with those of bioinformatics analysis. Furthermore, the knockdown of MCM6 inhibited cell proliferation, migration and invasion and promoted cell apoptosis, and made cells arrested in S stage. In summary, the findings of bioinformatics analysis provided a novel hypothesis for ESCC progression. In particular, the aberrantly elevated expression of MCM6 is a potential biomarker for ESCC diagnosis and treatment.

The present study aimed to investigate the role of Wiskott‑Aldrich syndrome verprolin‑homologous protein 3 (WAVE3) in the progression of esophageal squamous cell carcinoma (ESCC), and to explore its effect on the migration of esophageal cancer cell lines in vitro. The expression level of WAVE3 in ESCC tissues was determined via immunohistochemistry, and the relative levels of WAVE3 mRNA and micro (mi)RNA200b were assessed in the serum of patients with ESCC using reverse transcription‑quantitative PCR (RT‑qPCR). Following cell transfection, the levels of miRNA200b and WAVE3 protein were determined via RT‑qPCR and western blot analysis, and cell migration was examined using a Transwell assay. Subsequently, the clinical parameters were used to analyze whether the expression of WAVE3 in tissues and serum was associated with the occurrence and development of ESCC. The results demonstrated that the expression of WAVE3 was increased in ESCC tissues compared with normal tissues. The results also revealed increased expression levels of WAVE3 and decreased expression levels of miRNA200b in the serum of patients with ESCC, compared with healthy volunteers. High expression of WAVE3 was significantly associated with tumor TNM stage, invasion depth and lymphatic invasion of ESCC. In cells transfected with miRNA200b mimic, the miRNA200b was overexpressed, WAVE3 protein was downregulated and cell migration ability was decreased. The results of the present study suggest that WAVE3 may serve as an oncogene in ESCC, and its inhibition via miRNA200b decreased tumor cell migration. Therefore, WAVE3 may serve as a novel biological marker and therapeutic target for ESCC.

Leaf angle (LA) is a critical agronomic trait in maize, with more upright leaves allowing higher planting density, leading to more efficient light capture and higher yields. A few genes responsible for variation in LA have been identified by map-based cloning. In this study, we cloned maize ZmIBH1-1, which encodes a bHLH transcription factor with both a basic binding region and a helix-loop-helix domain, and the results of qRT-PCR showed that it is a negative regulator of LA. Histological analysis indicated that changes in LA were mainly caused by differential cell wall lignification and cell elongation in the ligular region. To determine the regulatory framework of ZmIBH1-1, we conducted RNA-seq and DNA affinity purification (DAP)-seq analyses. The combined results revealed 59 ZmIBH1-1-modulated target genes with annotations, and they were mainly related to the cell wall, cell development, and hormones. Based on the data, we propose a regulatory model for the control of plant architecture by ZmIBH1-1 in maize., ZmIBH1-1 encodes a bHLH transcription factor that affects the leaf angle in maize primarily through differential cell wall lignification and cell elongation in the ligular region.

Leaf angle (LA) is a critical agronomic trait enhancing grain yield under high-density planting in maize. A number of researches have been conducted in recent years to investigate the quantitative trait loci/genes responsible for LA variation, while only a few genes were identified through map-based cloning. Here we cloned the ZmDWF1 gene, which was previously reported to encode Δ24-sterol reductase in the brassinosteroids (BRs) biosynthesis pathway. Overexpression of ZmDWF1 resulted in enlarged LA, indicating that ZmDWF1 is a positive regulator of LA in maize. To reveal the regulatory framework of ZmDWF1, we conducted RNA-Sequencing and yeast-two hybrid (Y2H) screening analysis. RNA-Sequencing analyzing results indicate ZmDWF1 mainly affected expression level of genes involved in cell wall associated metabolism and hormone metabolism including BR, gibberellin, and auxin. Y2H screening with Bi-FC assay confirmed three proteins (ZmPP2C-1, ZmROF1, and ZmTWD1) interacting with ZmDWF1. We revealed a new regulatory network of ZmDWF1 gene in controlling plant architecture in maize.

The present study aimed to investigate the expression of LRP16 in the development of ESCC and the relationship between Hippo signaling pathway and LRP16. Immunohistochemistry was used to detect the expression of LRP16 in ESCC tissues. After transfection, the expression of LRP16 was detected by reverse transcription quantitative PCR (RT-qPCR) and western blot techniques. Cell counting kit (CCK-8), clone formation experiment, flow cytometry and wound healing were used to determine the proliferation, apoptosis, cell cycle and migration of ESCC cells. The changes of factors related to Hippo signaling pathway were determined via RT-qPCR and western blot experiments. The results showed that the LRP16 expression in ESCC tissues was higher than that in normal tissues. High expression of LRP16 was related to the depth of invasion, TNM stage and lymph node metastasis of ESCC. Furthermore, the knockdown of LRP16 inhibited proliferation, migration and promoted cell apoptosis and made cells arrested in G2/M phase. It also resulted in decreased expression of Yes-associated protein (YAP), and increased expression of mammalian STE20-like protein kinase (MST1/2), suggesting that LRP16 promoted the development of ESCC through Hippo signaling pathway. The results of this study suggest that LRP16 may be a carcinogenic gene of ESCC and promotes the progression of ESCC through the regulation of Hippo signaling pathway. Our study provides a new idea for the diagnosis and treatment of ESCC in the future.

Background Appropriate flowering time is very important to the success of modern agriculture. Maize (Zea mays L.) is a major cereal crop, originated in tropical areas, with photoperiod sensitivity. Which is an important obstacle to the utilization of tropical/subtropical germplasm resources in temperate regions. However, the study on the regulation mechanism of photoperiod sensitivity of maize is still in the early stage. Although it has been previously reported that ZmCCT is involved in the photoperiod response and delays maize flowering time under long-day conditions, the underlying mechanism remains unclear. Results Here, we showed that ZmCCT overexpression delays flowering time and confers maize drought tolerance under LD conditions. Implementing the Gal4-LexA/UAS system identified that ZmCCT has a transcriptional inhibitory activity, while the yeast system showed that ZmCCT has a transcriptional activation activity. DAP-Seq analysis and EMSA indicated that ZmCCT mainly binds to promoters containing the novel motifs CAAAAATC and AAATGGTC. DAP-Seq and RNA-Seq analysis showed that ZmCCT could directly repress the expression of ZmPRR5 and ZmCOL9, and promote the expression of ZmRVE6 to delay flowering under long-day conditions. Moreover, we also demonstrated that ZmCCT directly binds to the promoters of ZmHY5, ZmMPK3, ZmVOZ1 and ZmARR16 and promotes the expression of ZmHY5 and ZmMPK3, but represses ZmVOZ1 and ZmARR16 to enhance stress resistance. Additionally, ZmCCT regulates a set of genes associated with plant development. Conclusions ZmCCT has dual functions in regulating maize flowering time and stress response under LD conditions. ZmCCT negatively regulates flowering time and enhances maize drought tolerance under LD conditions. ZmCCT represses most flowering time genes to delay flowering while promotes most stress response genes to enhance stress tolerance. Our data contribute to a comprehensive understanding of the regulatory mechanism of ZmCCT in controlling maize flowering time and stress response.

LiDAR has the advantages of small size, high imaging accuracy, high resolution, all-day survey, and strong adaptability to complex environments. With the continuous improvement of technical performance, LiDAR is widely used in aerospace, mapping, military, and civil fields. In this study, the principle of LiDAR survey technology was introduced, and the key technology and application method of using LiDAR to measure the initial section of rocket launch were discussed in detail; a LiDAR survey system was designed on this basis. Afterward, the feasibility and rationality of the survey system and survey method were verified by rocket launch tests. The results show that compared with the traditional survey methods, LiDAR survey technology has obvious advantages in terms of survey accuracy, automation, and real-time visualization of survey data, and it can greatly reduce labor intensity and improves work efficiency, which provides an important reference for the design and application mode of LiDAR survey system in space launch sites.

The purpose of the current study was to construct a β-cyclodextrin drug carrier for rubropunctatin to improve its water solubility and light stability for future cytotoxicity studies. The inclusion complexation behavior of rubropunctatin with β-cyclodextrin was investigated using FESEM, FT-IR and XRD. A molecular docking study was performed to elucidate the most probable inclusion structure. The inclusion complex could be completely dispersed in water and had a small size of 121.87 ± 2.13 nm (n = 3), a good PDI (0.320 ± 0.017), and an acceptable potential value of −27.7 ± 0.32 mV (n = 3). Furthermore, the stability of the rubropunctatin in water under light irradiation was found to be greatly enhanced after being encapsulated in cyclodextrin, and it exhibited a retention rate of over 70% vs. 10.17%. In addition, the cytotoxicity of the inclusion complex was evaluated by MTT assay and Annexin V-FITC/PI detection using cervical adenocarcinoma HeLa cells. The results showed that the inclusion complex had comparable toxicity compared to rubropunctatin solubilized with 0.4% DMSO. More importantly, the formation of the inclusion complex contributed greatly to the intensification of the bioavailability of rubropunctatin because the use of organic solvent was avoided.

The present work investigated the antioxidative, anti-inflammatory and pulmonary protective effects of enzymatic- and acid- hydrolysed mycelia polysaccharides (En-MPS and Ac-MPS) from Oudemansiella radicata on LPS-induced acute lung injury (ALI) mice. The results demonstrated that both En-MPS and Ac-MPS showed potential pulmonary protective effects by decreasing serum levels of hs-CRP and C3, increasing pulmonary enzyme values of SOD, GSH-Px, CAT and the level of T-AOC; reducing the activity of MPO; and down-regulating the contents of MDA and LPO. In addition, the levels of TNF-ɑ, IL-1β, and IL-6 in BALF of mice treated with En-MPS at a dosage of 400 mg/kg/d were significantly lower than those in the ALI mice. The in vitro antioxidant effects also showed that the En-MPS was more effective than Ac-MPS. Furthermore, the physical properties of polysaccharides were also investigated by GC, HPGPC, FT-IR and NMR. These results indicated that both En-MPS and Ac-MPS possessed potent antioxidant and anti-inflammatory activities, which could be used as an ingestible drug in preventing lung injury.

The present work investigated the antioxidative, anti-inflammatory and organ protective effects of residue polysaccharide (RPS) and its enzymatic-RPS (ERPS) of Lentinula edodes against the LPS-induced sepsis in mice. The results demonstrated that ERPS showed superior effects on inhibiting serum activities of CKMB, LDH, AST, ALP and ALT, lowing the serum levels of BUN, CRE and UA, as well as improving the antioxidant status (SOD, CAT, GSH-PX and T-AOC) than RPS. Moreover, the levels of TNF-α (667.17 ± 36.72 pg/mL), IL-6 (288.47 ± 16.48 pg/mL), and IL-1β (61.62 ± 3.54 pg/mL) on ERPS-treated mice (600 mg/kg bw) significantly lower than that in the LPS-induced mice. Furthermore, the histological observations also indicated the significant recovery effect on organs. The RPS yield was 5.03%, and the ERPS yield was 73.65%. Moreover, the physical properties of polysaccharides were investigated by HPGPC, FT-IR, NMR, GC–MS and AFM analysis, and the results indicated that ERPS was a homogeneous heteropolysaccharide by (1 → 6)- and (1 → 3)-linkages. These conclusions suggested that ERPS might be suitable for functional foods and potentially effective candidate medicine for the treatment of sepsis and its complications.

The present work investigated the antioxidant and hepatoprotective effects of acidic- and enzymatic-hydrolysis mycelium polysaccharide (AcMPS and EnMPS) from Pleurotus geesteranus on alcoholic liver disease (ALD) mice. The animal studies demonstrated that the polysaccharides had potential effects reflected by remitting alcoholic hepatitis, reducing lipid accumulation, preventing oxidative stress, improving inflammatory symptoms, and alleviating the liver functions by histopathologic observation. Results showed that AcMPS (yield of 84%) was composed of L-Rha, D-Rib, L-Ara, D-Glc, D-Man and D-Gal with the Mw of 3.49 × 104 Da, while EnMPS (yield of 90%) was contained L-Rha, L-Ara, D-Gal and D-Glc with the Mw of 3.67 × 104 Da. Furthermore, the GC–MS analysis indicated that both AcMPS and EnMPS were β-pyranoside polysaccharides with the (1→3)- and (1→6)-linkages. The conclusions indicated that AcMPS and EnMPS could be used as natural drugs for preventing the ALD, and providing underlying hepatoprotective mechanisms, pharmaceutically.

The present work was designed to investigate the antioxidant and hepatoprotective effects of acidic-hydrolysis Lentinus edodes residue polysaccharides (Ac-LRP) on lipopolysaccharide-induced liver injury in mice. Hepatoprotective effects were observed in liver treated with Ac-LRP at doses of 200, 400, and 600 mg/kg body weight, respectively. Activities of superoxide dismutase, glutathione peroxide, catalase, total antioxidant capacity, alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase and levels of total cholesterol, triglyceride, malondialdehyde, and lipid peroxidation in liver or serum samples were analyzed. Ac-LRP reduced the incidence of liver necrosis detected via histological observations. In addition, Ac-LRP chemical bonds and ultrastructure were measured. These results provided valuable evidence supporting the utilization of Ac-LRP as a functional food and natural medicine for the treatment of liver injury.

Foamed concrete (FC) was prepared from raw materials of vanadium tailings (VTs), reservoir sediment (RS), and phosphogypsum (PG). The physicochemical properties of the raw materials were studied by using X-ray diffraction (XRD), X-ray fluorescence (XRF) analysis, and inductively coupled plasma emission spectrometer (ICP-OES). The preparation and properties of FC were investigated by particle size analysis and strength test. The hydration products and microstructure of FC were analyzed by XRD and field emission scanning electron microscopy (FE-SEM). The results show that when the specific surface area (SSA) of VTs is 768 m2·kg−1 and the content is 40%, the products with a compressive strength of 3.56 MPa and density of 619.1 kg·m−3 meet the requirements of JG/T 266-2011 standard on FC of grades A06 and C3.0; the main mineral phases in the products are calcium silicate hydrate (C-S-H) gel, ettringite (AFt), and calcite, as well as the residual mineral phases after the system reaction include quartz, orthoclase, mullite, pyrite, and PG.

B lymphocyte-induced maturation protein-1 (Blimp1) is a key regulator that promotes the terminal differentiation of mature B lymphocytes into plasma cells, and is essential for the survival of Multiple myeloma (MM)cells. However, the expression of Blimp1 in MM and its effect on the signaling pathway remain unknown. Studies have found that during long-term endoplasmic reticulum (ER) stress, activated ATF4 may also stimulate the CCAAT-enhancer-binding protein homologous protein (CHOP) gene, triggering the unfolded protein response (UPR) terminal apoptotic pathway in plasma cells. Moreover Aspirin can induce MM cell apoptosis through mitochondria and death receptor pathway. Therefore, we aim to explore whether Aspirin could induce AFT4/CHOP apoptosis pathway in MM by inhibiting Blimp1 expression, thereby promoting MM cell apoptosis and exerting anti-tumor effects.

The growth and development of maize are negatively affected by various abiotic stresses including drought, high salinity, extreme temperature, and strong wind. Therefore, it is important to understand the molecular mechanisms underlying abiotic stress resistance in maize. In the present work, we identified that a novel NAC transcriptional factor, ZmNST3, enhances maize lodging resistance and drought stress tolerance. ChIP-Seq and expression of target genes analysis showed that ZmNST3 could directly regulate the expression of genes related to cell wall biosynthesis which could subsequently enhance lodging resistance. Furthermore, we also demonstrated that ZmNST3 affected the expression of genes related to the synthesis of antioxidant enzyme secondary metabolites that could enhance drought resistance. More importantly, we are the first to report that ZmNST3 directly binds to the promoters of CESA5 and Dynamin-Related Proteins2A (DRP2A) and activates the expression of genes related to secondary cell wall cellulose biosynthesis. Additionally, we revealed that ZmNST3 directly binds to the promoters of GST/GlnRS and activates genes which could enhance the production of antioxidant enzymes in vivo. Overall, our work contributes to a comprehensive understanding of the regulatory network of ZmNST3 in regulating maize lodging and drought stress resistance.

Leaf angle in cereals is an important agronomic trait contributing to plant architecture and grain yield by determining the plant compactness. Although ZmCLA4 was identified to shape plant architecture by affecting leaf angle, the detailed regulatory mechanism of ZmCLA4 in maize remains unclear. ZmCLA4 was identified as a transcriptional repressor using the Gal4-LexA/UAS system and transactivation analysis in yeast. The DNA affinity purification (DAP)-seq assay showed that ZmCLA4 not only acts as a repressor containing the EAR motif (CACCGGAC), but was also found to have two new motifs, CCGARGS and CDTCNTC. On analyzing the ZmCLA4-bound targeted genes, we found that ZmCLA4, as a cross node of multiple plant hormone-mediated pathways, directly bound to ARF22 and IAA26 to regulate auxin transport and mediated brassinosteroid signaling by directly binding to BZR3 and 14-3-3. ZmCLA4 bound two WRKY genes involved with abscisic acid, two genes (CYP75B1, CYP93D1) involved with jasmonic acid, B3 involved in the response to ethylene, and thereby negatively regulated leaf angle formation. We built a new regulatory network for the ZmCLA4 gene controlling leaf angle in maize, which contributed to the understanding of ZmCLA4’s regulatory mechanism and will improve grain yields by facilitating optimization of plant architecture.

DNA affinity purification sequencing (DAP-Seq) analysis revealed that ZmCLA4 mainly binds to promoters containing the EAR motif (CACCGGAC) as well as to two other motifs (CCGARGS and CDTCNTC) to inhibit the expression of its target genes. Further analysis of ZmCLA4 target genes indicated that ZmCLA4 functions as a hub of multiple plant hormone signaling pathways: ZmCLA4 was found to directly bind to the promoters of multiple genes including ZmARF22 and ZmIAA26 in the auxin transport pathway, ZmBZR3 in the brassinosteroid signaling pathway, two ZmWRKY genes involved in abscisic acid metabolism, ZmCYP genes (ZmCYP75B1, ZmCYP93D1) related to jasmonic acid metabolism, and ZmABI3 involved in the ethylene response pathway. Overall, our work provides deep insights into the ZmCLA4 regulatory network in controlling leaf angle in maize.

Flowering time is an important agronomic trait that determines the distribution and adaptation of plants. The accurate prediction of flowering time in elite germplasm is very critical for maize breeding. However, the molecular mechanisms underlying photoperiod response remain elusive in maize. Here we cloned the flowering time controlling gene, ZmDPS10-2, by map-based cloning and confirmed that ZmDPS10-2 is a positive regulator of flowering time in maize under long-day conditions using ChIP-Seq and CRISPER-Cas9 mutagenesis system. More specifically ZmDPS10-2 promotes ZmNF-YA3 transcription through binding to the promoter region of ZmNF-YA3. Then ZmNF-YA3 negatively regulates the transcription of ZmAP2 by binding ZmAP2 promoter. Finally, ZmAP2 suppresses the expression of ZMM4 to delay flowering time. The cascading regulation of flowering time by ZmDPS10-2 in maize that has not been reported in other species such as rice or Arabidopsis thaliana. Then we created an integrated gene regulatory network of flowering time in maize using ZmDPS10-2, ZmNF-YA3, ZmAP2, ZMM4, and CONZ1 as the backbone plus 12 previous reported key maize flowering genes, ten enhancers via ChIA-PET, 41 genes identified by ChIP-Seq, and 49 through Hi-C technology. The PIF3 being identified by both ChIP-Seq and Hi-C experiments is a hub linking DPS10-2 and other known flowering genes. This network provides in-depth understanding of genetic mechanism of maize flowering time and serves as invaluable resource for the plant community to tackle this very important agronomic trait.

The selenium polysaccharides (SPS) by Oudemansiella radicata were isolated, and two hydrolysates (enzymatic-SPS (ESPS) and acidic-SPS (ASPS)) were obtained by enzymolysis and acidolysis. The endo-toxemic-induced lung and kidney damages were successfully established by the lipopolysaccharides (LPS) injection (5 mg/kg/d). The in vivo mice experiments indicated that the ESPS showed superior antioxidant and protective effects against the LPS-toxicities by increasing the antioxidant activities, reducing lipid peroxidation, improving the inflammatory response and elevating the kidney and lung functions than SPS and ASPS. Furthermore, the selenium contents and structure of SPS, ESPS and ASPS were investigated through FAAS, GC, UV, HPGPC, FT-IR, NMR and the results indicated that SPS, ESPS and ASPS were homogeneous heteropolysaccharide with an average molecular weight of 3.12 × 104, 1.77 × 104, 1.41 × 104 Da, respectively. The conclusions demonstrated that the polysaccharides by O. radicata might be suitable for functional foods and natural drugs in preventing the endo-toxemia and its complications, and providing references for exploitation and utilization of natural substances.

Rubropunctatin is a naturally occurring constituent of polyketide compounds that has great potential in the development of cancer-assisted chemotherapy. However, it has certain shortcomings such as water insolubility and photo instability that limit its clinical application. In this study, we constructed a Rubropunctatin-loaded liposome (R-Liposome) anticancer drug carrier for the first time. The results indicate that R-Liposome is water soluble, has spherical morphology, great homogeneity and dispersibility with high encapsulation efficiency (EE%, 90 ± 3.5%) and loading rate (LR%, 5.60 ± 2.5%) values. Moreover, the carrier improves the photostability, storage and pH stabilities of Rubropunctatin. The R-Liposome also prolongs the release of Rubropunctatin, enhances the anticancer activity of Rubropunctatin and encourages the mechanism of Rubropunctatin to promote apoptosis. Therefore, liposomal nanoparticles have great potential as drug delivery vehicles of Rubropunctatin for cancer treatment.

Nuclear factor-Y (NF-Y) transcription factors are important regulators of several essential biological processes, including embryogenesis, drought resistance, meristem maintenance, and photoperiod-dependent flowering in Arabidopsis. However, the regulatory mechanisms of NF-Ys in maize (Zea mays) are not well understood yet. In this study, we identified an NF-Y transcription factor, ZmNF-YA3. Genome-wide analysis showed that ZmNF-YA3 bound to >6000 sites in the maize genome, 2259 of which are associated with genic sequences. ZmNF-YA3 was found to interact with CONSTANS-like (CO-like) and flowering promoting factor1 (FPF1) through yeast two-hybrid and bimolecular fluorescence complementation (BiFC) assays. Quantitative real-time reverse transcription-PCR (qRT-PCR) combined with yeast one-hybrid assay and EMSA suggested that NF-YA3 could promote early flowering by binding to the FLOWERING LOCUS T-like12 (FT-like12) promoter in maize. Morerover, we also showed that ZmNF-YA3 could improve drought and high-temperature tolerance through binding to the promoter regions of bHLH92, FAMA, and the jasmonic acid activator MYC4, respectively. These results contribute to a comprehensive understanding of the molecular mechanisms and regulatory networks of NF-Y transcription factors in regulating maize flowering time and stress response in maize.

In the present study, we investigated the antioxidant and hepatoprotective effects of intracellular mycelium polysaccharides from Pleurotus geesteranus (IMPP) to address acute alcoholic liver diseases (ALD). Animal studies have demonstrated that the hepatic parameters of CYP2E1, MPO, TC, ALT, AST and ALP, HDLC, LDL-C and VLDL-C were improved; the inflammatory factor levels, including TNF-α, IL-1β and IL-6, were decreased; the hepatic ADH and ALDH activities were inhibited; the activities of the antioxidant enzymes SOD, GSH-Px, CAT and T-AOC were enhanced; and the MDA and LPO contents were reduced by IMPP treatment. Furthermore, the liver histopathology confirmed the beneficial effects of IMPP against ALD in mice, demonstrating that IMPP had the potential to remediate alcoholic hepatitis. Besides the properties of typical pyran-type polysaccharides obtained by β-glycosidic bonding rich in fucose, the results demonstrated that IMPP might be used as a functional food and natural medicine for preventing ALD and its complications.
