Abstract Objective This study aims to assess the consistency of various CT-FFR software, to determine the reliability of current CT-FFR software, and to measure relevant influence factors. The goal is to build a solid foundation of enhanced workflow and technical principles that will ultimately improve the accuracy of measurements of coronary blood flow reserve fractions. This improvement is critical for assessing the level of ischemia in patients with coronary heart disease. Methods 103 participants were chosen for a prospective research using coronary computed tomography angiography (CCTA) assessment. Heart rate, heart rate variability, subjective picture quality, objective image quality, vascular shifting length, and other factors were assessed. CT-FFR software including K software and S software are used for CT-FFR calculations. The consistency of the two software is assessed using paired-sample t-tests and Bland-Altman plots. IntroductionWe verified that Akabane virus (AKAV) is transmitted through biting midges and infects local domestic animals.MethodsIn 2013, viruses were isolate from biting midges in Yunnan, China, using BHK-21 and C6/36 cells. Two AKAV strains (No. 52 and 55) that induced cytopathogenic effects (CPE) in BHK-21, MDBK, and Vero cells were characterized.ResultsThe complete genomic sequence of both viruses consisted three RNA segments (S, M, and L). The S segment (856 nucleotides) encoded a 233-amino-acid nucleocapsid protein and a 91-amino-acid nonstructural protein, while the M segment (4309 nucleotides) encoded a 1401-amino-acid polyprotein. The L segment (6869 nucleotides) encoded a 2511-amino-acid RNA-dependent RNA polymerase. Phylogenetic analysis revealed that specimen Nos. 52 and 55 clustered with AKAV genotype Ia viruses isolated from Asia. The AKAV strain (55) neutralizing antibody exhibited a total positive rate of 43.55% (202/466) against serum samples from cattle and goats collected in Yunnan Province. Specifically, the positive rates were 48.77% (139/285) for cattle and 34.81% (63/181) for goats. Neutralizing antibody titers in cattle (1:32–1:128) were higher than those in goats (1:4–1:16).DiscussionThis study represents the first isolation of AKAV from biting midges in China, along with the detection of high neutralizing antibody titers against AKAV in the serum samples of local cattle and goats. These findings suggested that biting midges are involved in AKAV transmission among domestic animals in Yunnan Province, China.

Abstract The continuous production of mature blood cell lineages is maintained by hematopoietic stem cells but they are highly susceptible to damage by ionizing radiation (IR) that induces death. Thus, devising therapeutic strategies that can mitigate hematopoietic toxicity caused by IR would benefit acute radiation syndrome (ARS) victims and patients receiving radiotherapy. Herein, we describe the preparation of an injectable hydrogel formulation based on Arg‐Gly‐Asp‐alginate (RGD‐Alg) and Laponite using a simple mixing method that ensured a slow and sustained release of interleukin‐12 (IL‐12) (RGD‐Alg/Laponite@IL‐12). The local administration of RGD‐Alg/Laponite@IL‐12 increased survival rates and promoted the hematopoietic recovery of mice who had received sublethal‐dose irradiation. Local intra‐bone marrow (intra‐BM) injection of RGD‐Alg/Laponite@IL‐12 hydrogel effectively stimulated IL12 receptor‐phosphoinositide 3‐kinase/protein kinase B (IL‐12R‐PI3K/AKT) signaling axis, which promoted proliferation and hematopoietic growth factors secretion of BM mesenchymal stem/stromal cells. This signaling axis facilitates the repair of the hematopoietic microenvironment and plays a pivotal role in hematopoietic reconstitution. In conclusion, we describe a biomaterial‐sustained release of IL‐12 for the treatment of irradiated hematopoietic injury and provide a new therapeutic strategy for hematopoietic ARS.

Straw return is an effective agricultural measure that influences the ecological environment. In the context of global climate change and the need for sustainable development, it is essential to explore how straw return methods affect the environment. This study conducted a two-year field experiment comparing different straw return treatments: no straw return (CK), straw return directly (SR), and straw returned in deep ditches (ISR). The results showed that SR treatment increased soil dissolved organic carbon (DOC) (21.7~25.8%) and dissolved organic nitrogen (DON) (8.5~43.8%) compared to CK. The ISR treatment significantly enhanced DOC (13.1~33.0%) and DON (14.2~50.8%). Both SR and ISR treatments also improved maize growth indicators, such as leaf area index (LAI), stem biomass, leaf biomass, and grain biomass. In terms of environmental effects, the ISR measure reduced N2O emissions and significantly lowered the carbon footprint (CF) and water footprint (WF). Specifically, the yield-scale carbon footprint (CFy) and yield-scale water footprint (WFy) of ISR were reduced by 12.0% and 9.1%, respectively, in 2023. Correlation analysis revealed that soil DON and DOC were key factors in these environmental benefits. Furthermore, the reduction in WF and CF was closely linked to the increased SWC and higher biomass in all maize organs. Therefore, we demonstrated that the ISR measure not only promotes maize biomass accumulation but also effectively mitigates carbon emissions and water consumption on farmland. It may be related to the boosting soil DON and DOC levels. Overall, the ISR method holds promise for supporting sustainable farming practices.

Stroke is a major disorder of the central nervous system that poses a serious threat to human life and quality of life. Many stroke victims are left with long-term neurological dysfunction, which adversely affects the well-being of the individual and the broader socioeconomic impact. Currently, post-stroke brain dysfunction is a major and difficult area of treatment. Vagus nerve stimulation is a Food and Drug Administration-approved exploratory treatment option for autism, refractory depression, epilepsy, and Alzheimer's disease. It is expected to be a novel therapeutic technique for the treatment of stroke owing to its association with multiple mechanisms such as altering neurotransmitters and the plasticity of central neurons. In animal models of acute ischemic stroke, vagus nerve stimulation has been shown to reduce infarct size, reduce post-stroke neurological damage, and improve learning and memory capacity in rats with stroke by reducing the inflammatory response, regulating blood-brain barrier permeability, and promoting angiogenesis and neurogenesis. At present, vagus nerve stimulation includes both invasive and non-invasive vagus nerve stimulation. Clinical studies have found that invasive vagus nerve stimulation combined with rehabilitation therapy is effective in improving upper limb motor and cognitive abilities in stroke patients. Further clinical studies have shown that non-invasive vagus nerve stimulation, including ear/cervical vagus nerve stimulation, can stimulate vagal projections to the central nervous system similarly to invasive vagus nerve stimulation and can have the same effect. In this paper, we first describe the multiple effects of vagus nerve stimulation in stroke, and then discuss in depth its neuroprotective mechanisms in ischemic stroke. We go on to outline the results of the current major clinical applications of invasive and non-invasive vagus nerve stimulation. Finally, we provide a more comprehensive evaluation of the advantages and disadvantages of different types of vagus nerve stimulation in the treatment of cerebral ischemia and provide an outlook on the developmental trends. We believe that vagus nerve stimulation, as an effective treatment for stroke, will be widely used in clinical practice to promote the recovery of stroke patients and reduce the incidence of disability.

Returning straw to the field is a crucial practice for enhancing soil quality and increasing efficient use of secondary crop products. However, maize straw has a higher carbon-to-nitrogen ratio compared to other crops. This can result in crop nitrogen loss when the straw is returned to the field. Therefore, it is crucial to explore how different methods of straw return affect maize (Zea mays L.) farmland. In this study, a field experiment was performed with three treatments (I, no straw returned, CK; II, direct straw return, SR; and III, straw returned in deep furrows, ISR) to explore the effects of the different straw return modes on soil carbon and nitrogen content and greenhouse gas emissions. The results indicated that the SR and ISR treatments increased the dissolved organic carbon (DOC) content in the topsoil (0–15 cm). Additionally, the ISR treatment boosted the contents of total nitrogen (TN), nitrate nitrogen (NO3−-N), ammonium nitrogen (NH4+-N), dissolved organic nitrogen (DON), and DOC in the subsurface soil (15–30 cm) compared with CK. When it comes to greenhouse gas emissions, the ISR treatment led to an increase in CO2 emissions. However, SR and ISR reduced N2O emissions, with ISR showing a more pronounced reduction. The ISR treatment significantly increased leaf and grain biomass compared to CK and SR. The correlation analyses showed that the yield was positively correlated with soil DOC, and soil greenhouse gas emission was correlated with soil NO3−-N. The ISR technology has great potential in sequestering soil organic matter, improving soil fertility, and realizing sustainable agricultural development.

Abstract Background Long non encoding RNA (lncRNA) plays a crucial role in breast cancer. However, the prognostic role of AFAP1‐AS1 in breast cancer remains unclear. Aims To investigate the relationship between the expression of long non‐coding RNA actin filament‐associated protein1 antisense RNA1 (AFAP1‐AS1) and prognosis of breast cancer. Methods and Results Meta‐analysis was performed to explore the correlation between AFAP1‐AS1 and breast cancer. The AFAP1‐AS1expression in patients with breast cancer tissue and adjacent normal tissue from 153 patients was determined by qRT‐PCR. Bioinformatics and Cox proportional‐hazards risk model were used to explore the relationship between expression of AFAP1‐AS1 and prognosis. Developing new photocatalytic materials is one of the most promising strategies to address bacterial infection during wound healing without bacterial drug resistance, but their poor photocatalytic activity ultimately limits their therapeutic efficacy. In this work, a metal–organic framework‐based S‐scheme heterostructure (Ag2S@PB) is prepared to rapidly treat open wounds caused by bacterial infection, which effectively establishes and retains spatially isolated redox centers through an S‐scheme charge‐transfer pathway. Additionally, the formation of an interface electric field and covalent FeS bridge bonding in the heterogeneous interface endows the heterojunction with a much stronger charge separation and transfer capability than bare Ag2S, which significantly enhances the photocatalytic performance and stability, consequently generating more radical oxygen species. Meanwhile, the introduction of Prussian blue greatly improves the photothermal effect of the Ag2S@PB under 808 nm near‐infrared light. Therefore, under illumination for 20 min, the Ag2S@PB shows a desirable antibacterial efficiency of 99.92% against Staphylococcus aureus and 99.86% against Escherichia coli. The in vivo wound repair experiment shows that the Ag2S@PB has good tissue repair effects and can expedite wound healing. This work will provide insights for designing highly efficient photoresponsive materials to treat bacterial wound infections.

IntroductionBanna virus (BAV), a potential pathogen that may cause human encephalitis, is the prototype species of genus Seadornaviru within the family Reoviridae, and has been isolated from a variety of blood-sucking insects and mammals in Asia.MethodsCulicoides, Mosquitoes, and Ticks were collected overnight in Yunnan, China, during 2016-2023 using light traps. Virus was isolated from these collected blood-sucking insects and grown using Aedes albopictus (C6/36) cells. Preliminary identification of the virus was performed by agarose gel electrophoresis (AGE). The full genome sequences of the BAVs were determined by full-length amplification of cDNAs (FLAC) and sequenced using next-generation sequencing.ResultsIn this study, 13 strains BAV were isolated from Culicoides, Mosquitoes and Ticks. Their viral genome consisted of 12 segments of double-stranded RNA (dsRNA), and with three distinct distribution patterns. Sequence analysis showed that Seg-5 of four strains (SJ_M46, SJ_M49, JC_M19-13 and JC_C24-13) has 435 bases nucleotide sequence insertions in their ORF compared to other BAVs, resulting in the length of Seg-5 up to 2128 nt. There are 34 bases sequence deletion in Seg-9 of 3 strains (WS_T06, MS_M166 and MS_M140). Comparison of the coding sequences of VP1, VP2, VP5, VP9 and VP12 of the 13 BAV strains, the results show that VP1, VP2 and VP12 are characterised by high levels of sequence conservation, while VP9 is highly variable, under great pressure to adapt and may be correlated with serotype. While also variable, VP5 appears to be under less adaptive pressure than VP9. Additionally, phylogenetic analysis indicates that the 13 BAV strains locate in the same evolutionary cluster as BAVs isolated from various blood-sucking insects, and are clustered according to geographical distribution.ConclusionThe data obtained herein would be beneficial for the surveillance of evolutionary characteristics of BAV in China and neighboring countries as well as extend the knowledge about its genomic diversity and geographic distribution.

Abstract Background Iatrogenic injury to the radial nerve is a risk in surgical treatment for extraarticular fractures of the middle and distal third of the humerus. We aimed to investigate the safety, feasibility and advantages of minimally invasive percutaneous plate osteosynthesis (MIPPO) via an anteromedial approach in the treatment of middle and middle-distal humeral fractures and to evaluate proximity to neurovascular structures. Materials and methods In 2016, 13 adult cadaver arms were used to simulate a minimally invasive surgical approach to the anteromedial humerus followed by fixation with a locking compression plate (LCP), and several sets of anatomical data were measured to clarify the possible risk of iatrogenic vascular and nerve injury in this surgical approach. Then, a case series study of 12 patients with humeral fractures who were treated with this surgical approach was conducted between 2017 and 2020. Results The average humeral length was 29.22 ± 1.62 cm, the average width of the medial epicondyle of the humerus was 1.31 ± 0.17 cm, and the average distance from the vertex of the medial epicondyle to the median nerve was 2.96 ± 1.62 cm. Furthermore, the safe area for distal humeral screw placement was 6.28 ± 0.39 cm, and the average distance from the tip of the distal end of the screw in the medial epicondyle to the ulnar nerve was 1.7 ± 1.25 mm. None of the 12 patients had nerve damage or an incisional infection after the operation. Conclusions The new approach was performed as described, and no cases of iatrogenic nerve palsy occurred. This approach can be used as an alternative for the treatment of extraarticular fractures of the middle and distal thirds of the humerus. Level of Evidence: Level IV, therapeutic study.

Objective: To investigate the value of dual-source CT dual-energy virtual plain scan in the preoperative diagnosis of extramural vascular invasion (EMVI) in rectal cancer. Methods: A total of 150 patients with rectal cancer (64 females (42.7%) and 86 males (57.3%), with an average age of (62.3±11.8) years) who were scheduled for surgical treatment in our hospital from November 2019 to December 2021 were selected for the preoperative dual-source CT dual-energy virtual plain scan;. Two senior radiologists independently analyzed preoperative imaging data (including ctEMVI status, local lymph node infiltration, and distant metastasis) to determine the existence of preoperative EMVI. With pathological results as the gold standard, the sensitivity, specificity, accuracy, positive predictive value, and negative predictive value of VNC in the diagnosis of EMVI were evaluated, and the area under the receiver operating characteristic curve (ROC) was used to evaluate the diagnostic efficiency. Results: Among 150 patients with rectal cancer, 56 (37.3%) were positive for EMVI and 94 (62.7%) were negative for EMVI. The accuracy, sensitivity, specificity, positive predictive value, and negative predictive value of EMVI evaluation by physician 1 were 86%, 80.36%, 89.36%, 81.82%, and 88.42%, respectively, and the AUC was 0.831 (95%CI, 0.756~0.905). The accuracy, sensitivity, specificity, positive predictive value, and negative predictive value of physician 2 were 88.67%, 80.36%, 93.62%, 88.24%, and 88.89%, respectively, and the AUC was 0.870 (95%CI, 0.802~0.938). The consistency of the evaluation results between physician 1 and physician 2 was high (k=0.943). Conclusion: Dual-source CT dual-energy virtual plain scan has a certain diagnostic value in the preoperative evaluation of EMVI of rectal cancer.

Biting midges are one of the most common hematophagous insects. They are capable of transmitting a wide range of arboviruses and have a significant impact on public health and veterinary medicine. Herein, from midge samples collected in 2013 in Yunnan, China, one sample induced a cell cytopathic effect (CPE) in BHK-21, MA104, and PK15 cell lines. Next-generation sequencing data, RACE and PCR determined the genome sequence of the sample and designated as an Oya virus (OYAV) isolate SZC50. Phylogenetic analysis of the sample revealed that it was cluster into viruses from species Orthobunyavirus catqueense. The open reading frames of S, M, and L segment of OYAV SZC50 were closest to those of OYAV SC0806. Moreover, 831 serum samples (736 pigs, 45 cattle, and 50 sheep) were gathered from 13 cities in Yunnan Province to detect neutralizing antibody of OYAV SZC50. A significant proportion of OYAV SZC50 antibody (more than 30%) was found in Yunnan pig populations, with the positive rate of OYAV SZC50 antibody in pigs from Malipo reaching 95%. To determine the pathogenicity of OYAV SZC50, we chose three animal models: specific pathogen-free Kunming mice, C57BL/6 mice lacking the interferon α/β receptor, and chicken embryos. At 5, 6, and 7 days post-infection, all adult and suckling C57BL/6 mice, and specific pathogen-free suckling Kunming mice were dead. Our finding was expanding the knowledge about the infection and pathogenic risk of the neglected virus in the Orthobunyavirus.

Two strains of viruses, JC13C644 and JC13C673, were isolated from Culicoides tainanus collected in Jiangcheng County, Yunnan Province, situated along the border area shared by China, Laos, and Vietnam. JC13C644 and JC13C673 viruses can cause cytopathic effect (CPE) in mammalian cells BHK21 and Vero cells, and cause morbidity and mortality in suckling mice 48 h after intracerebral inoculation. Whole-genome sequencing was performed, yielding complete sequences for all 10 segments from Seg-1 (3942nt) to Seg-10 (810nt). Phylogenetic analysis of the sub-core-shell (T2) showed that the JC13C644 and JC13C673 viruses clustered with the Epizootic Hemorrhagic Disease Virus (EHDV) isolated from Japan and Australia, with nucleotide and amino acid homology of 93.1% to 98.3% and 99.2% to 99.6%, respectively, suggesting that they were Eastern group EHDV. The phylogenetic analysis of outer capsid protein (OC1) and outer capsid protein (OC2) showed that the JC13C644 and JC13C673 viruses were clustered with the EHDV-10 isolated from Japan in 1998, with the nucleotide homology of 98.3% and 98.5%, and the amino acid homology of 99.6% and 99.6–99.8%, respectively, indicating that they belong to the EHDV-10. Seroepidemiological survey results demonstrated that JC13C644 virus-neutralizing antibodies were present in 29.02% (177/610) of locally collected cattle serum and 11.32% (89/786) of goat serum, implying the virus’s presence in Jiangcheng, Yunnan Province. This finding suggests that EHDV-10 circulates not only among blood-sucking insects in nature but also infects local domestic animals in China. Notably, this marks the first-ever isolation of the virus in China and its discovery outside of Japan since its initial isolation from Japanese cattle. In light of these results, it is evident that EHDV Serotype 10 exists beyond Japan, notably in the natural vectors of southern Eurasia, with the capacity to infect local cattle and goats. Therefore, it is imperative to intensify the surveillance of EHDV infection in domestic animals, particularly focusing on the detection and monitoring of new virus serotypes that may emerge in the region and pose risks to animal health.

During tunnel excavation using a tunnel boring machine (TBM), the interaction between the cutter and the rock causes the cutterhead to generate abrupt vibrations, which could lead to engineering problems, such as bearing seal failure and even cutterhead cracking. In this study, large-scale physical model tests with various cutter spacings were performed to study the vibration and load characteristics of the cutterhead during the rock-breaking process. Through a TBM modal comprehensive test bench, the real-time acceleration of the cutterhead was obtained using a monitoring system. The test results showed that the complete time-domain curves were stepped and periodic. When the cutterhead was stuck or rock peeled off during the excavation, the monitoring curves showed abrupt vibrations, which were mainly concentrated at 0–3 Hz and were typical low-frequency vibrations. Taking measured data as the driving parameter, the real-time mechanical characteristics of the cutterhead were determined through transient dynamic analysis. Numerical simulation results showed that the vibration trend during excavation was the superposition of random vibrations generated by rock breaking and tunneling without rock cutting. During the tunneling process, the wear of the edge cutters should be highlighted in actual construction. The related methods and results can provide an essential reference for the selection of cutterheads.

IntroductionThis article aimed to investigate the effects of the endoscopic-assisted microsurgery technique on the resection of large (Koos grade IV) vestibular schwannoma (VS) and provide a prognosis analysis of the patients.MethodsA retrospective analysis of the use of the endoscopic-assisted microsurgery technique in 16 cases of large vestibular schwannoma surgery was carried out. Intraoperative nerve electrophysiological monitoring was conducted to explore the effect of neuroendoscopy on the resection of internal auditory canal tumors, protection of the facial nerve, and minimizing postoperative complications.ResultsTumors were completely removed in all 16 cases, and the facial nerve was anatomically preserved in 14 cases (87.5%). There was no postoperative cerebrospinal fluid leakage and no intracranial infection complications occurred.Following the House-Brackmann (H-B) grading system, post-operative facial nerve function was grade I in 5 cases, grade II in 6 cases, grade III in 3 cases, and grade V in 2 cases. As a result, the preservation rate of facial nerve function (H-B grade I-II) was 68.8%. All 16 patients were followed up for 3 to 24 months, and no tumor recurrence was found on enhanced MRI.DiscussionUsing the endoscopic-assisted microsurgery technique in the retrosigmoid approach has many advantages over the microscopic-only approach. When compared to the microscopy-only approach, the endoscope can provide a wide-angle surgical field superior to that of a microscope in areas such as the internal auditory canal in the resection of large VS, minimize iatrogenic injuries, ensure complete removal of internal auditory canal tumors, and well as reducing postoperative complications such as cerebrospinal fluid leakage and the loss of facial and auditory nerve functions.

Introductionbluetongue virus (BTV) infection triggers dramatic and complex changes in the host's transcriptional profile to favor its own survival and reproduction. However, there is no whole-transcriptome study of susceptible animal cells with BTV infection, which impedes the in-depth and systematical understanding of the comprehensive characterization of BTV-host interactome, as well as BTV infection and pathogenic mechanisms.Methodsto systematically understand these changes, we performed whole-transcriptome sequencing in BTV serotype 1 (BTV-1)-infected and mock-infected sheep embryonic testicular cells, and subsequently conducted bioinformatics differential analyses.Resultsthere were 1504 differentially expressed mRNAs, 78 differentially expressed microRNAs, 872 differentially expressed long non-coding RNAs, and 59 differentially expressed circular RNAs identified in total. Annotation from the Gene Ontology, enrichment from the Kyoto Encyclopedia of Genes and Genomes, and construction of competing endogenous RNA networks revealed differentially expressed RNAs primarily related to virus-sensing and signaling transduction pathways, antiviral and immune responses, inflammation, and development and metabolism related pathways. Furthermore, a protein-protein interaction network analysis found that BTV may contribute to abnormal spermatogenesis by reducing steroid biosynthesis. Finally, real-time quantitative PCR and western blotting results showed that the expression trends of differentially expressed RNAs were consistent with the whole-transcriptome sequencing data.Discussionthis study provides more insights of comprehensive characterization of BTV-host interactome, and BTV infection and pathogenic mechanisms.

Endovascular mechanical thrombectomy, combined with a tissue plasminogen activator (t-PA), is efficacious as a standard care for qualifying ischemic stroke patients. However, > 50% of thrombectomy patients still have poor outcomes. Manganese porphyrins, commonly known as mimics of superoxide dismutases, are potent redox-active catalytic compounds that decrease oxidative/nitrosative stress and in turn decrease inflammatory responses, mitigating therefore the secondary injury of the ischemic brain. This study investigates the effect of intracarotid MnTnBuOE-2-PyP5+ (BMX-001) administration on long-term, 28-day post-stroke recovery in a clinically relevant setting. The 90 min of transient middle cerebral artery occlusion was performed in young, aged, male, female, and spontaneous hypertension rats. All physiological parameters, including blood pressure, blood gas, glucose, and temperature, were well controlled during ischemia. Either BMX-001 or a vehicle solution was infused through the carotid artery immediately after the removal of filament, mimicking endovascular thrombectomy, and was followed by 7 days of subcutaneous injection. Neurologic deficits and infarct volume were assessed at 28 days in a blinded manner. The effects of BMX-001 on the carotid arterial wall and blood–brain barrier permeability and its interaction with t-PA were assessed in normal rats. There were no intra-group differences in physiological variables. BMX-001-treated stroke rats regained body weight earlier, performed better in behavioral tests, and had smaller brain infarct size compared to the vehicle-treated group. No vascular wall damage and blood–brain barrier permeability changes were detected after the BMX-001 infusion. There was no drug interaction between BMX-001 and t-PA. Intracarotid BMX-001 infusion was safe, and it significantly improved stroke outcomes in rats. These findings indicate that BMX-001 is a candidate drug as an adjunct treatment for thrombectomy procedure to further improve the neurologic outcomes of thrombectomy patients. This study warrants further clinical investigation of BMX-001 as a new stroke therapy.

Sustaining proliferative signaling and enabling replicative immortality are two important hallmarks of cancer. The complex of cyclin-dependent kinase (CDK) and its cyclin plays a decisive role in the transformation of the cell cycle and is also critical in the initiation and progression of cancer. CRIF1, a multifunctional factor, plays a pivotal role in a series of cell biological progresses such as cell cycle, cell proliferation, and energy metabolism. CRIF1 is best known as a negative regulator of the cell cycle, on account of directly binding to Gadd45 family proteins or CDK2. In addition, CRIF1 acts as a regulator of several transcription factors such as Nur77 and STAT3 and partly determines the proliferation of cancer cells. Many studies showed that the expression of CRIF1 is significantly altered in cancers and potentially regarded as a tumor suppressor. This suggests that targeting CRIF1 would enhance the selectivity and sensitivity of cancer treatment. Moreover, CRIF1 might be an indispensable part of mitoribosome and is involved in the regulation of OXPHOS capacity. Further, CRIF1 is thought to be a novel target for the underlying mechanism of diseases with mitochondrial dysfunctions. In summary, this review would conclude the latest aspects of studies about CRIF1 in cancers and mitochondria-related diseases, shed new light on targeted therapy, and provide a more comprehensive holistic view.

ObjectiveTo assess the predictive value of magnetic resonance imaging (MRI) radiomics for progression-free survival (PFS) in patients with prostate cancer (PCa).Methods191 patients with prostate cancer confirmed by puncture biopsy or surgical pathology were included in this retrospective study, including 133 in the training group and 58 in the validation group. All patients underwent T2WI and DWI serial scans. Three radiomics models were constructed using univariate logistic regression and Gradient Boosting Decision Tree(GBDT) for feature screening, followed by Cox risk regression to construct a mixed model combining radiomics features and clinicopathological risk factors and to draw a nomogram. The performance of the models was evaluated by receiver operating characteristic curve (ROC), calibration curve and decision curve analysis. The Kaplan-Meier method was applied for survival analysis.ResultsCompared with the radiomics model, the hybrid model consisting of a combination of radiomics features and clinical data performed the best in predicting PFS in PCa patients, with AUCs of 0.926 and 0.917 in the training and validation groups, respectively. Decision curve analysis showed that the radiomics nomogram had good clinical application and the calibration curve proved to have good stability. Survival curves showed that PFS was shorter in the high-risk group than in the low-risk group.ConclusionThe hybrid model constructed from radiomics and clinical data showed excellent performance in predicting PFS in prostate cancer patients. The nomogram provides a non-invasive diagnostic tool for risk stratification of clinical patients.

Tunnel Boring Machine (TBM), as the most advanced construction equipment for rock tunnels, has been widely used in railway tunnel engineering, water conservancy tunnel engineering, urban rail engineering, coal mine roadway engineering, and other fields. However, the disadvantage of TBM lies in its poor adaptability to complex geological conditions, resulting in a significant reduction in tunneling efficiency and even major accidents due to improper TBM construction measures, especially in weak and fractured strata, which pose a great challenge to TBM construction. Therefore, it is necessary to study the key technologies of TBM construction in weak and fractured strata in order to improve the efficiency of TBM construction. Based on the background of the TBM construction of the Gaoligongshan Tunnel on the Southwest Darui Railway, this paper provides a reference for safe and efficient TBM excavation in weak and fractured areas through research on equipment, excavation parameter control, and construction measures.

The purpose of this study was to identify the biomarkers implicated in the development of intracranial hemorrhage (ICH) and potential regulatory pathways. In the transcriptomic data for patients with ICH, we identified DEmiRNAs and DEmRNAs related to hypoxia, inflammation, and their transcription factors (TFs). An ICH-based miRNA-TF-mRNA regulatory network was thus constructed, and four biomarkers (TIMP1, PLAUR, DDIT3, and CD40) were screened for their association with inflammation or hypoxia by machine learning. Following this, SP3 was found to be a transcription factor involved in hypoxia and inflammation, which regulates TIMP1 and PLAUR. From the constructed miRNA-TF-mRNA regulatory network, we identified three axes, hsa-miR-940/RUNX1/TIMP1, hsa-miR-571/SP3/TIMP1, and hsa-miR-571/SP3/PLAUR, which may be involved in the development of ICH. Upregulated TIMP1 and PLAUR were validated in an independent clinical cohort 3 days after ICH onset. According to Gene Set Enrichment Analysis (GSEA), SP3 was discovered to be important in interleukin signaling and platelet activation for hemostasis. Transcription factor SP3 associated with hypoxia or inflammation plays an important role in development of ICH. This study provides potential targets for monitoring the severity of inflammation and hypoxia in patients with ICH.

In order to study the effect of fracture zone on tunnel stability under high ground stress and to reveal the disaster mechanism of fracture zone surrounding rock compressor, the TBM tunnel of Fragrant Furnace Mountain Tunnel in the F12 fracture zone in Dali I section of central Yunnan was experimentally studied. The results show that the TBM cutter head has a significant effect on the surrounding rock pressure in the range of 0.43 D~0.5 D (hole diameter), with a maximum increase of 1.19-fold. As the measurement point approaches the tunnel contour, the displacement of surrounding rock increases. As TBM traverses the fracture zone interface, the displacement of surrounding rock increases at the maximum rate. The effect of TBM shear rock formation failure on the internal displacement of rock formation is 1.21 D in the front and not more than 1.5 D in the top. Sliding and loosening of the fault zone looser above the interface can easily lead to blockage of the fault zone, and the pressure of the surrounding rocks at front edge does not affect more than 1.73 H (the length of horizontal projection of the fault zone interface). Therefore, in the course of construction, the interface of fracture zone should be promoted to reduce the construction risks.

Soil eukaryotic microorganisms are important to biodiversity, and returning different kinds of organic materials to the field could improve the biodiversity of soil eukaryotic microorganisms. However, no detailed research has been conducted in the northern China semi-arid area in returning the different organic materials to the field and determining the status of eukaryotic microorganisms. Therefore, we explored the effects of various organic materials returning to the soil in combination with inorganic fertilizer on the diversity and community structure of eukaryotic microorganisms in Shanxi province, China. Soil samples were collected from five different fertilization treatments: chemical fertilizer (F), chemical fertilizer + cattle manure (FM), chemical fertilizer + straw (FS), chemical fertilizer + pig manure (FC) and control without fertilizers (CK). High-throughput sequencing was applied to analyze the eukaryotic diversity and community structure. Results showed that the dominant eukaryotic microorganisms among soil samples were Fungi, Viridiplantae, Metazoa and Protist. Although α-diversity was not significantly different among the five treatments, principal coordinate analysis and permutational multivariate analysis of variance illustrated significant differences (p < 0.001) in β-diversity of eukaryotic microorganism under treatments with different organic materials. Redundancy analysis showed that the soil properties, including total potassium, available nitrogen, available potassium and organic matter were the main factors attributed to eukaryotic microorganisms’ community structure in this region.

In 2018, a strain of epizootic hemorrhagic disease virus (EHDV), named YNDH/V079/2018, was isolated from a sentinel calf in Mangshi County, Yunnan Province, China. Nucleotide sequencing and neutralization tests indicated that the virus belongs to a novel serotype of EHDV that had not been reported previously.

Abstract Background The expression level of ribonucleotide reductase subunit M1 (RRM1) is closely related to the effect of gemcitabine-based therapy in advanced bladder cancer. However, the value of RRM1 expression in predicting progression-free survival in non-muscle-invasive bladder cancer (NMIBC) patients treated with intravesical gemcitabine chemotherapy has not been elucidated. Methods This study randomly assigned 162 patients to either the RRM1-known group or the unknown group. We collected cancer tissues from 81 patients to evaluate the mRNA expression of RRM1 by using liquid chip technology. All patients were diagnosed and then treated with intravesical gemcitabine monotherapy immediately after transurethral resection of the bladder tumour (TURBT). Results RRM1 expression was high in 21% (17/81) of patients. The RRM1 mRNA level was not correlated with sex, age, weight, performance status, or CUA/EAU risk (p > 0.05). Progression-free survival (PFS) was significantly longer for patients with low RRM1 expression than for patients with high and unknown RRM1 expression (p = 0.009). Additionally, the 1- and 2-year relapse rates also differed according to RRM1 expression level. The 1-year relapse rates for RRM1-low, RRM1-high and RRM1-unknown patients were 0, 17.7 and 6.2% (p = 0.009), while the 2-year relapse rates for these groups were 3.1, 29.4, and 11.1% (p = 0.005), respectively. Conclusions This preliminary study showed that low RRM1 expression was associated with longer progression-free survival and lower 1-year/2-year relapse rates in NMIBC patients treated with intravesical gemcitabine monotherapy, despite the need for further verification with large sample sizes and considering more mixed factors and biases.

The congenital intellectual disability (ID)-causing gene mutations remain largely unclear, although many genetic variations might relate to ID. We screened gene mutations in Chinese Han children suffering from severe ID and found a single-nucleotide polymorphism (SNP) in the 5′-untranslated region (5′-UTR) of fibroblast growth factor 13 (FGF13) mRNA (NM_001139500.1:c.-32c>G) shared by three male children. In both HEK293 cells and patient-derived induced pluripotent stem cells, this SNP reduced the translation of FGF13, which stabilizes microtubules in developing neurons. Mice carrying the homologous point mutation in 5′-UTR of Fgf13 showed delayed neuronal migration during cortical development, and weakened learning and memory. Furthermore, this SNP reduced the interaction between FGF13 5′-UTR and polypyrimidine-tract-binding protein 2 (PTBP2), which was required for FGF13 translation in cortical neurons. Thus, this 5′-UTR SNP of FGF13 interferes with the translational process of FGF13 and causes deficits in brain development and cognitive functions.

Non-invasive biomarkers to identify patients with bladder outlet obstruction (BOO)-related dysfunction are still needed to guide clinical practice. The current study aims to investigate molecular alterations and biomarkers associated with partial BOO (PBOO) in rats. Sprague–Dawley rats were used to establish the BOO model. Serum samples from 60 patients with benign prostatic hyperplasia (BPH) were used for enzyme-linked immunosorbent assay analysis. RNA sequencing and TMT-labeling proteomic analyses were conducted to identify molecular alterations. Masson’s trichrome, H&E, and immunohistochemical staining and western blotting were conducted by using conventional methods following the manufacturer’s instructions. Rats with PBOO experienced hypertrophy of smooth muscle cells and hyperplasia of interstitial cells during the first 4 weeks after the initiation of obstruction. Four weeks later, rats with PBOO showed activation of the adaptive immune response, cell death and apoptosis. The levels of brain-derived neurotrophic factor (BDNF) and fibroblast growth factor 2 (FGF2) in the serum gradually increased in the first 4 weeks and gradually decreased after week 4. FGF2 levels slightly correlated with prostate volume (R = 0.156, P = 0.0028) but not with age or BMI in BPH patients. No correlations were found between BDNF levels and prostate volume, age or BMI. BOO induces a change from bladder compensation to decompensation at week 4. FGF2 is involved in the development of hypertrophy in the PBOO bladder and shows a positive correlation with prostate volume in BPH patients.

Sedoreoviridae is a family of viruses belonging to the order Reovirales and comprises six genera, two of which, Orbivirus and Seadornavirus, contain arboviruses that cause disease in humans and livestock. Areas such as Yunnan Province in southwestern China, have high arboviral activity due in part to warm and wet summers, which support high populations of biting flies such as mosquitoes and Culicoides. Three viral isolates previously obtained from Culicoides collected at cattle farms in Shizong County of Yunnan Province, China, between 2019 and 2020 were completely sequenced and identified as Banna virus (BAV) genotype A of Seadornavirus and serotypes 1 and 7 of epizootic hemorrhagic disease virus (EHDV) of Orbivirus. These results suggest that Culicoidestainanus and C. orientalis are potential vectors of BAV and EHDV, respectively, and represent the first association of a BAV with C. tainanus and of an arbovirus with C. orientalis. Analysis using VP9 generally agreed with the current groupings within this genus based on VP12, although the classification for some strains should be corrected. Furthermore, the placement of Kadipiro virus (KDV) and Liao ning virus (LNV) in Seadornavirus may need confirmation as phylogenetic analysis placed these viruses as sister to other species in the genus.

Excessive use of chemical fertilizers in agricultural practices have demonstrated a significant impact on microbial diversity and community in soil by altering soil physical and chemical properties, thereby leading to a certain degree of soil salinization and nutritional imbalances. As an organic amendment, maize straw has been widely used to improve soil quality; however, its effect on the soil bacterial community remains limited in Calcarie-Fluvie Cambisols soil in semi-humid arid plateau of North China. In the present experiment, we investigated the effects of continuous straw utilization and fertilization on bacterial communities in Shouyang, Shanxi province, China. Soil samples were collected from 5 different straw utilization and fertilization modes in the following ways: straw mulching (SM), straw crushing (SC), cattle manure (CM), in which way straw is firstly used as silage and then organic fertilizer, control with no straw return (NSR), and control without fertilizers (CK), same amount of N+P fertilizer was applied to the regimes except CK. High-throughput sequencing approaches were applied to the V3-V4 regions of the 16S ribosomal RNA for analysis of the bacterial abundance and community structures. Different long-term straw returning regimes significantly altered the physicochemical properties and bacterial communities of soil, among which CM had the most significant effects on soil fertility and bacterial diversity. Proteobacteria, Actinobacteria, Chloroflexi, Acidobacteria, and Gemmatimonadetes were consistently dominant in all soil samples, and Redundancy analysis (RDA) showed significant association of total nitrogen (TN), total phosphorus (TP) and available potassium (AK) with alternation of the bacterial community. Cattle manure had the most beneficial effects on soil fertility and bacterial diversity among different straw utilization and fertilization modes.

Background/Aims: Chemoresistance is largely responsible for relapses of bladder cancer during clinical therapy. However, the molecular mechanisms involved in the chemoresistance of bladder cancer are unclear. Growing evidence supports the theory that microRNAs (miRNAs) play an important role in chemotherapeutic drug resistance because they are downregulated in many malignancies that have been implicated in the regulation of diverse processes in cancer cells. More specifically, the extent and precise mechanism of the involvement of miR-34as in chemoresistance to epirubicin (EPI) in the treatment of bladder cancer remains unclear. Methods: In this study, real-time quantitative polymerase chain reaction (PCR) was used to analyze the expression of miR-34a in bladder cancer cell line BIU87 and its EPI chemoresistant cell line BIU87/ADR. The miR-34a profiles in bladder cancer tissues were obtained from The Cancer Genome Atlas database. The effect of miR-34a on chemosensitivity was evaluated by cell viability assays, colony formation assays, and in vivo experimentation. Apoptosis and the cell cycle were examined by flow cytometry. A luciferase reporter assay was used to assess the target genes of miR-34a. Western blot and qPCR were used to analyze the expression of target proteins and downstream molecules. Results: The downregulation of miR-34a in bladder cancer serves as an independent predictor of reduced patient survival. The CCK-8 assay showed that miR-34a overexpression resulted in increased sensitivity to EPI, while miR-34a downregulation resulted in chemoresistance to EPI in vitro. Moreover, it was found that miR-34a increased the sensitivity of BIU87/ADR cells to chemotherapy in vivo. The luciferase reporter assay ascertained that TCF1 and LEF1 are direct target genes of miR-34a. It was found that miR-34a increased chemosensitivity in BIU87/ADR cells by inhibiting the TCF1/LEF1 axis. Conclusions: The results of this study indicate that miR-34a contributes to the chemosensitivity of BIU87/ADR by inhibiting the TCF1/LEF1 axis. Consequently, miR-34a is a determinant of BIU87 chemosensitivity and may therefore serve as a potential therapeutic target in bladder cancer treatment.

Chronic cystitis: proteins may open a channel to treatment A protein that controls the passage of ions through cell membranes is implicated in interstitial cystitis/painful bladder syndrome (IC/PBS). This condition causes chronic pelvic pain and increased urinary frequency and urgency. Current treatment options are unsatisfactory. Researchers led by Longkun Li at the Third Military Medical University in Chongqing, China, and Mingjia Tan at the University of Michigan, Ann Arbor, USA, studied the role of this membrane channel protein, called Piezo1. Increased activity of Piezo1 was linked to bladder hyperactivity in rats with drug-induced cystitis. The research also identified a synergistic interaction between Piezo1 and a second membrane channel protein. A drug that inhibits Piezo1 activity reduced bladder hyperactivity in the rats. Drugs targeting these two proteins might help to treat the chronic cystitis of patients with IC/PBS.

Background/Aims: Cyanidin is an anthocyanin found in many foods. Although its variable antioxidant levels are well-documented, little is known about its effects on renal cell carcinoma (RCC) tumorigenesis. This study, therefore, investigated the effects of cyanidin on the proliferation, migration, and invasion of renal cell carcinoma lines and demonstrated, for the first time, significant inhibitory effects of cyanidin on RCC tumorigenesis. Methods: RCC cells were treated with different doses of cyanidin and the effects were tested by Cell Counting Kit-8 reagent, clone formation assay, transwell assay, and flow cytometry. Moreover, the cyanidin-mediated mechanism that curtailed tumorigenesis was analyzed by RNA sequencing (RNA-seq). Sequencing data from The Cancer Genome Atlas (TCGA) were used to compare the expression of both early growth response protein 1 (EGR1) and selenoprotein W (SEPW1) in RCC and tumor-free adjacent normal tissue samples. Real-time PCR (RT-PCR) and/or western blot were used to assess the expression of E-cadherin, cleaved-caspase3, Bcl2, p62, and ATG4. Results: We found significantly greater induction of cell-cycle arrest, apoptosis, and suppression of RCC cell invasion and migration at concentrations of 25 µM and 100 µM than at a concentration of 50 µM. It was also discovered, first through RNA-seq then confirmed by RT-PCR, that cyanidin (100 µM) inhibited RCC carcinogenesis through EGR1 and SEPW1. TCGA data indicated that the expression level of EGR1 was lower and that of SEPW1 was higher in RCC tumor tissue than in normal tissues. Moreover, western blot and/or RT-PCR indicated that cleaved-caspase3 was enhanced and E-cadherin was inhibited by cyanidin treatment. Furthermore, western blot and RT-PCR also showed regulation of p62 and ATG4, which are associated with autophagy. Cyanidin in vivo significantly inhibited the growth of xenografts in nude mice. Conclusions: The results of this study showed the therapeutic potential of cyanidin for the treatment of RCC and the prevention of recurrence and metastasis.

In this paper, a design approach for a high-efficiency concurrent dual-band power amplifier (PA) with precise harmonic control up to third order is proposed. With the precise harmonic control approach, the high-efficiency performance of the PA can be improved at two arbitrary wide interval frequencies. Based on the inherent impedance matching flexibility of the continuous Class-F (CCF) operating mode, the design spaces of fundamental and harmonic impedances of the PA are largely expanded. The optimal impedances of a CCF PA mode at the internal current-generator (I-gen) plane and package plane are investigated, respectively, and a novel dual-band matching network topology is designed to simultaneously present required load impedances at the fundamental and harmonic impedances of both operation frequencies. First, a harmonic control network is designed to control the harmonic impedances precisely for CCF operation. Then, the fundamental impedances are synthesized using the real frequency technique. To verify the validity of proposed methodology, a dual-band CCF PA operating at 2.6 and 3.5 GHz is designed, fabricated, and measured. The measurement results show that the peak drain efficiency is 76.7% with an output power 42.4 dBm at the lower band and 72.8% with an output power 41.1 dBm at the upper band.

In this paper, a new methodology using a distributed ladder structure with finite transmission zero at finite frequencies is proposed for designing a broadband high-efficiency continuous Class-F (CCF) power amplifier (PA). The approach of realizing finite transmission zeros in Richard domain using Real Frequency Technique is first presented. By applying the proposed approach, the characteristics of the driving point impedance (DPI) of the proposed structure can be deduced and obtained in theory. Furthermore, the synthesis methods in Richard domain are extended to synthesize the DPI of the proposed structure. Four types of distributed elements are investigated to synthesize the DPI of the proposed structure. This proposed structure can improve the performance of the CCF PA when the bandwidth approaches an octave as the finite transmission zero can be used to control the second harmonic impedance of the lower frequency which is near the upper frequency of the operation band. To verify the validity of the proposed method, a broadband high-efficiency CCF PA working from 1.15 to 2.2 GHz is designed. Experimental results show that the fabricated PA achieves 11.3-13.7 dB power gain and 40.5-43.2 dBm output power in the operation band. And the PA has also achieved a high-efficiency characteristic of 70%-83% drain efficiency (DE) over the whole operation bandwidth.

Abstract Background Previous studies have confirmed a family risk of nephrolithiasis (NL), but only 15% of all cases are associated with an identified monogenic factor. In clinical practice, our group encountered a patient with NL combined with cystic kidney disease that had 3 affected family members. No known mutations association with NL was detected in this family, and thus further investigation of the molecular cause of NL was deemed to be necessary. Results Quality analysis from the sequencing stage showed a more than 80-fold average depth and 95% coverage for each sample, and six mutations within six genes were chosen as candidate variants for further validation. Genotyping of rs182089527in the phosphodiesterase 1A (PDE1A) gene in the validation cohort indicated that the alternative allele was present in 15 patients with heterozygosity and in 1 patient with homozygosity, and exhibited significant enrichment in NL patients (Fisher’s exact test, adjusted p = 0.0042) and kidney cystic patients (Fisher’s exact test, adjusted p = 0.067) compared to controls. In addition, function analysis displayed a significant decrease in the protein and mRNA expression levels resulting from the rs182089527 mutant sequence compared with the wild-type sequence. Moreover, patients with this mutation displayed a high level of creatinine and urea in urinalysis. Conclusions Our study provides genetic evidence that the rs182089527 mutation in PDE1A is involved in the development of NL and kidney cysts, which should help to improve personalized medicine for diagnosis and treatment.

The application of organic fertilizer affects soil microbes and enzyme activities. In this study, we explored the effects of various long-term different fertilization treatments (manure, M; chemical fertilizer, NP; manure + chemical fertilizer, MNP; and no fertilizer, CK) on bacterial community structure and soil sucrase, urease, and alkaline phosphatase activities in Shaping, Hequ, China. High-throughput sequencing was used to amplify the third to the fourth hypervariable region of the 16S ribosomal RNA for analysis of the bacterial community structure. Enzyme activities were determined by colorimetry. Soil treated with MNP had the highest bacterial Abundance-based Coverage Estimator index and enzyme activities. The principal coordinates analysis results showed significant differences among the various fertilization treatments (p < 0.001). Proteobacteria, Actinobacteria, Acidobacteria, Gemmatimonadetes, and Chloroflexi were consistently dominant in all soil samples. The redundancy analysis and Monte Carlo permutation tests showed that the soil bacterial communities were significantly correlated with alkali-hydrolyzable nitrogen, organic matter, urease, and alkaline phosphatase. Our results reveal the fundamentally different effects that organic and inorganic fertilizers have on soil bacterial communities and their functions.

Terahertz reflection behaviors of metallic-grating-dielectric-metal (MGDM) microcavity with a monolayer graphene embedded into the dielectric layer are theoretically investigated. A tunable wideband reflection dip at about the Fabry–Pérot resonant frequency of the structure is found. The reflectance at the dip frequency can be electrically tuned in the range of 96.5% and 8.8%. Because of the subwavelength distance between the metallic grating and the monolayer graphene, both of the evanescent grating slit waveguide modes and the evanescent Rayleigh modes play key roles in the strong absorption by the graphene layer. The dependence of reflection behaviors on the carrier scattering rate of graphene is analyzed. A prototype MGDM-graphene structure is fabricated to verify the theoretical analysis. Our investigations are helpful for the developments of electrically controlled terahertz modulators, switches, and reconfigurable antennas based on the MGDM-graphene structures.

Acute kidney injury (AKI) is an extremely dangerous clinical syndrome with high morbidity and mortality. Stem cell-based therapies have shown great promise for AKI treatment. Urine-derived stem cells (USCs) are a novel cell source in tissue engineering and cell therapy which provide advantages of simple, noninvasive, and low-cost harvest methods, efficient proliferation, and multi-differentiation potential. Here, we described the therapeutic effects of USCs in a rat model of cisplatin-induced AKI as a novel therapy. In vivo, the intravenous administration of USCs alleviated the renal functional damage in AKI rats, for the levels of blood urea nitrogen (BUN) and serum creatinine (SCr) were significantly decreased. The USCs-treated group also exhibited improved histological and ultrastructural changes, promoted proliferation, and inhibited apoptosis in renal tissues. After the USC therapy, the expression levels of proinflammatory cytokines (TNF-α and IL-6) and apoptosis-related proteins (BAX and cleaved caspase-3) were downregulated. In addition, the presence of a few GFP-labeled USCs was confirmed in rat renal tissues. In vitro, rat tubular epithelial (NRK-52E) cells were incubated with cisplatin to induce cell damage and then cocultured with USCs. After coculture with USCs, the cisplatin-induced NRK-52E cells showed higher cell viability and a lower apoptosis ratio than those of the control group, and cell cycle arrest was improved. In conclusion, our results demonstrated that USC therapy significantly improved the renal function and histological damage, inhibited the inflammation and apoptosis processes in the kidney, and promoted tubular epithelial proliferation. Our study exhibited the potential of USCs in the treatment of AKI, representing a new clinical therapeutic strategy.

BackgroundNumerous studies have demonstrated the presence of microRNA-124 abnormalities involving gene expression, methylation, and single nucleotide polymorphism (SNP) in multiple and diverse cancers, but the prognostic value of these abnormalities in cancer remains inconclusive.ObjectiveThe aim of this study is to determine the prognostic value of miR-124 in cancer.MethodsWe scrutinized the electronic databases and estimate the association between miR-124 expression, methylation and single nucleotide polymorphisms (SNPs), and prognosis in cancers. The pooled hazard ratios with 95% confidence intervals (CIs) for overall survival (OS), and disease-free survival/recurrence-free survival (RFS)/progression-free survival (PFS) were calculated to estimate the effects of miR-124 expression, methylation, and SNPs on cancer prognosis. The Quality in Prognosis Studies and Newcastle-Ottawa Scale were utilized to assess the quality of included studies.ResultsA total of 20 studies involving 3,574 participants were analyzed in evidence synthesis. Our findings showed that the low expression of miR-124 was significantly associated with poor OS (HR = 2.37, 95% CI: 1.91–2.94, P = 0.00; HR = 3.10, 95% CI: 2.04–4.70, P = 0.00) and PFS/RFS (HR = 2.21, 95% CI: 1.50–3.26, P = 0.00; HR = 2.12, 95% CI: 1.20–3.74, P = 0.00). The hyper-methylation of miR-124 was associated with poor OS (HR = 2.09, 95% CI: 1.48–2.95, P = 0.00) and PFS (HR = 3.70, 95% CI: 1.72–7.97, P = 0.00) (Table 3). The patients carrying with Allele C of miR-124 rs5315649 had a worse OS (HR = 1.50, 95% CI: 1.09–2.07, P = 0.00) and PFS (HR = 1.67, 95% CI: 1.20–2.33, P = 0.00) than the carriers with Allele G.ConclusionThe low expression and hyper-methylation of miR-124 was strongly associated with poor prognosis, and genetic variations of miR-124 rs531564 affected prognosis in cancer patients.

OBJECTIVE: To identify and investigate the susceptibility genes of Kashin-Beck disease (KBD) in Chinese population. METHODS: Whole-exome capturing and sequencing technology was used for the detection of genetic variations in 19 individuals from six families with high incidence of KBD. A total of 44 polymorphisms from 41 genes were genotyped from a total of 144 cases and 144 controls by using MassARRAY under the standard protocol from Sequenom. Association was applied on the data by using PLINK1.07. RESULTS: In the sequencing stage, each sample showed approximately 70-fold coverage, thus covering more than 99% of the target regions. Among the single nucleotide polymorphisms (SNPs) used in the transmission disequilibrium test, 108 had a p-value of
