Abstract Background Drought stress is a significant abiotic stressor that hinders growth, development, and crop yield in soybeans. Strigolactones (SLs) positively regulate plant resistance to drought stress. However, the impact of foliar application of SLs having different concentrations on soybean growth and metabolic pathways related to osmoregulation remains unknown. Therefore, to clarify the impact of SLs on soybean root growth and cellular osmoregulation under drought stress, we initially identified optimal concentrations and assessed key leaf and root indices. Furthermore, we conducted transcriptomic and metabolic analyses to identify differential metabolites and up-regulated genes. Results The results demonstrated that drought stress had a significant impact on soybean biomass, root length, root surface area, water content and photosynthetic parameters. However, when SLs were applied through foliar application at appropriate concentrations, the accumulation of ABA and soluble protein increased, which enhanced drought tolerance of soybean seedlings by regulating osmotic balance, protecting membrane integrity, photosynthesis and activating ROS scavenging system. This also led to an increase in soybean root length, lateral root number and root surface area. Furthermore, the effects of different concentrations of SLs on soybean leaves and roots were found to be time-sensitive. However, the application of 0.5 µM SLs had the greatest beneficial impact on soybean growth and root morphogenesis under drought stress. A total of 368 differential metabolites were screened in drought and drought plus SLs treatments. The up-regulated genes were mainly involved in nitrogen compound utilization, and the down-regulated metabolic pathways were mainly involved in maintaining cellular osmoregulation and antioxidant defenses. Conclusions SLs enhance osmoregulation in soybean plants under drought stress by regulating key metabolic pathways including Arachidonic acid metabolism, Glycerophospholipid metabolism, Linoleic acid metabolism, and Flavone and flavonol biosynthesis. This study contributes to the theoretical understanding of improving soybean adaptability and survival in response to drought stress.

Potatoes (Solanum tuberosum L.) are the third largest food crop globally and are pivotal for global food security. Widespread N fertilizer waste in potato cultivation has caused diverse environmental issues. This study employed microbial metagenomic sequencing to analyze the causes behind the declining N use efficiency (NUE) and escalating greenhouse gas emissions resulting from excessive N fertilizer application. Addressing N fertilizer inefficiency through breeding has emerged as a viable solution for mitigating overuse in potato cultivation. In this study, transcriptome and metabolome analyses were applied to identify N fertilizer-responsive genes. Metagenomic sequencing revealed that excessive N fertilizer application triggered alterations in the population dynamics of 11 major bacterial phyla, consequently affecting soil microbial functions, particularly N metabolism pathways and bacterial secretion systems. Notably, the enzyme levels associated with NO3- increased, and those associated with NO and N2O increased. Furthermore, excessive N fertilizer application enhanced soil virulence factors and increased potato susceptibility to diseases. Transcriptome and metabolome sequencing revealed significant impacts of excessive N fertilizer use on lipid and amino acid metabolism pathways. Weighted gene co‑expression network analysis (WGCNA) was adopted to identify two genes associated with N fertilizer response: PGSC0003DMG400021157 and PGSC0003DMG400009544.

The mechanism by which nitrate inhibits nitrogen fixation in soybean (Glycine max L.) is not fully understood. Accumulation of ureide in soybean plant tissues may regulate the nitrogen fixation capacity through a feedback pathway. In this study, unilaterally nodulated dual-root soybeans prepared by grafting were grown in sand culture. They were subjected to the removal of the nodulated side roots, and were given either nitrate supply or no supply to the non-nodulated side roots for 3 days (experiment I). Additionally, they received nitrate supply to the non-nodulated side roots for 1–14 days (experiment II). The results showed that nitrate supply increased the levels of asparagine and ureide in soybean shoots (Experiment I). In Experiment II, nodule dry weight, nodule number, nodule nitrogenase activity, and nodule urate oxidase activity decreased significantly after 3, 7, and 14 days of nitrate supply. Ureide content in the shoots and nodules increased after 1, 3, and 7 days of nitrate supply, but decreased after 14 days of nitrate supply. There was a significant positive correlation between urate oxidase activity and nitrogenase activity. Hence, we deduced that nitrate supply increased the asparagine content in soybean shoots, likely inhibiting ureide degradation, which induced the accumulation of ureide in soybean shoots and nodules, and, in turn, feedback inhibited the nodule nitrogen fixation. In addition, urate oxidase activity can be used to assess the nitrogen fixation capacity of nodules.

To improve nitrogen use efficiency (NUE) during rice cultivation, it is essential to comprehend the morphological and physiological traits of rice roots. However, in high-fertility black soil regions of Northeast China, the effects of combining biochar application with water-saving irrigation (WSI) conditions on rice root development and nitrogen utilization are still unknown. To address this knowledge gap, a combination of field experiments and 15N tracer micro-area investigations was conducted in this study. Four treatments were implemented: (i) controlled irrigation without biochar application (CB0); (ii) controlled irrigation with 2.5 t ha−1 biochar application (CB1); (iii) controlled irrigation with 12.5 t ha−1 biochar application (CB2); and (iv) controlled irrigation with 25 t ha–1 biochar application (CB3). Flooded irrigation conditions without biochar treatment (FB0) were used as the control. The primary objective of this research was to identify the mechanisms by which combined WSI conditions and biochar application affect rice root development and nitrogen utilization. Biochar application enhanced rice root morphological and physiological characteristics. Optimal biochar application increased the longest root length (RL), root volume (RV), root fresh weight (RFW), root active absorption area, root bleeding intensity, and root activity (RA) of rice while also optimizing the root–shoot ratio and facilitating nitrogen absorption by roots. These changes in root morphological and physiological characteristics facilitated the absorption of fertilizer-15N and soil nitrogen by rice roots, ultimately leading to improvements in rice yields and NUEs. Notably, the rice yields, NUE, nitrogen agronomic efficiency (NAE), and nitrogen partial factor productivity (NPFP) of CB2 plants were 16.45%, 39.42%, 24.48%, and 16.45% higher than those of FB0 plants, respectively. These results highlight the effectiveness of biochar application as a strategy to ensure food security and enhance NUE under WSI conditions. Furthermore, this study suggests that the recommended optimal application amount of biochar for the black soil area of Northeast China is 12.5 t ha−1.

Pumpkin is one of the region’s main cashcrops in northeast China cultivated for its edible seed and sarcocarp. Gummy stem blight (GSB), caused by the main pathogenic fungus of Stagonosporopsis cucurbitacearum (Sc.) which was identified in our previous study, has hampered pumpkin industry development with the affection of its reduced biological yield and edible quality. To clear the plant immune response mechanisms against Sc. infection, RNA-seq technology was employed to sequence pumpkin leaf transcriptome, the total number of distinct differentially expressed genes (DEGs) between the treatments and control was 2351, 4892, 5285, 4698, and 4213 for 2, 6, 12, 24, and 96 h treatments, respectively. KEGG analysis results revealed 16 distinct significantly up-regulated DEGs associated with MAPK signaling and plant-pathogen interaction pathways that may actively participate in Sc. infection-associated processes, while other five DEGs exhibited significantly altered expression during GSB infection. An apyrase-like gene, Gene3360 (LOC111476496), with significantly different expression levels, was possessing an early-stage primary infection-based function, which was hypothesized an important role in the resistance of pumpkin to Sc. infection. This study further deepens our understanding of the mechanism and mechanism of pumpkin leaf resistance to fungal infection.

Introduction: Spinocerebellar ataxias 36 (SCA36) is the neurodegenerative disease caused by the GGCCTG Hexanucleotide repeat expansions in NOP56, which is too long to sequence using short-read sequencing. Single molecule real time (SMRT) sequencing can sequence across disease-causing repeat expansion. We report the first long-read sequencing data across the expansion region in SCA36.Methods: We collected and described the clinical manifestations and imaging features of Han Chinese pedigree with three generations of SCA36. Also, we focused on structural variation analysis for intron 1 of the NOP56 gene by SMRT sequencing in the assembled genome.Results: The main clinical features of this pedigree are late-onset ataxia symptoms, with a presymptomatic presence of affective and sleep disorders. In addition, the results of SMRT sequencing showed the specific repeat expansion region and demonstrated that the region was not composed of single GGCCTG hexanucleotides and there were random interruptions.Discussion: We extended the phenotypic spectrum of SCA36. We applied SMRT sequencing to reveal the correlation between genotype and phenotype of SCA36. Our findings indicated that long-read sequencing is well suited to characterize known repeat expansion.

In this study, the systematic analysis of nitrate assimilation and transport in soybean roots was further improved by analyzing the concentrations of nitrate assimilates, asparagine and glutamine, in soybean roots and the related enzyme activities. This provided a theoretical basis for the efficient utilization of nitrogen fertilizer in soybean farming. A dual-root soybean system with both sides being nodulated was used to provide nitrate withdrawal and resupply in three phases on one side, while the other side received nutrients without nitrogen under sand culture conditions. Measurements were taken of the root’s nitrate reductase and glutamine synthetase activities, as well as the concentrations of asparagine. Measurements were also taken of the asparagine concentration in the basal root peeled skin (where the primary transport pathway is the phloem) and the woody parts (where the primary transport pathway is the xylem). Furthermore, the concentration of glutamine in the roots was also assessed. The findings indicated a positive correlation between the nitrate concentration in the roots and the activity of glutamine synthetase in the roots on both sides. The levels of asparagine in the roots, specifically those in the basal root peeled skin and woody part on both sides, rose when nitrate was added and declined when nitrate was removed. There was no significant change in glutamine concentration within the roots of soybeans. Hence, we deduced that the local provision of nitrate to the dual-root soybeans might enhance the absorption of nitrate into the roots on both sides. Additionally, the assimilated substances were predominantly carried as asparagine through the peeled skin and woody sections of the basal root, facilitating transportation in both directions within the soybean plants (from shoot to root and from root to shoot).

To investigate the effect of different organic matter returns on soil organic carbon (SOC) fractions, phosphorus (P) fractions and microbial communities, a pot experiment was conducted in a cold region of China for three years. There were six treatments in this study, including no rice straw return (S0), rice straw return (SR), decomposed rice straw return (DS), rice-straw-burned return (BS), rice root return (RR) and decomposed cattle manure return (DM). The results indicated that the organic matter returns had no significant effect on the rice yield after three years. The SR, DS and DM treatments significantly increased the content of the soil’s total organic carbon (TOC), light fraction organic carbon (LFOC), particulate organic carbon (POC), dissolved organic carbon (DOC) and microbial biomass carbon (MBC). The BS treatment decreased the soil MBC content. The SR, DS, BS and DM treatments significantly increased the content of the soil’s total P, NaHCO3-P, NaOH-P and residual-P. The proportion of nonlabile P (HCl-P and residual-P) was reduced by the organic matter returns. The SOC fractions were positively correlated to the soil P fractions (except HCl-P). The organic matter returns did not affect the microbial diversity but did change the microbial community composition. The dominant phyla included Proteobacteria, Firmicutes, Chloroflexi, and Bacteroidetes. Compared with the S0 treatment, the organic matter returns increased the relative abundance of Actinobacteria, Anaerolineae and Alphaproteobacteria and decreased the relative abundance of Bacteroidetes, Clostridia and Bacteroidia. The contents of MBC, DOC and NaOH-P were the main factors affecting the microbial community composition, and the soil’s P fractions had a larger influence on the microbial community than the SOC fractions. These results indicated that the incorporation of rice straw, decomposed rice straw and decomposed cattle manure might be an effective practice for maintaining soil fertility in the cold region of China.

BackgroundThe nitrate regulates soybean nodulation and nitrogen fixation systemically, mainly in inhibiting nodule growth and reducing nodule nitrogenase activity, but the reason for its inhibition is still inconclusive.MethodsThe systemic effect of nitrate on nodule structure, function, and carbon distribution in soybean (Glycine max (L.) Merr.) was studied in a dual-root growth system, with both sides inoculated with rhizobia and only one side subjected to nitrate treatment for four days. The non-nodulating side was genetically devoid of the ability to form nodules. Nutrient solutions with nitrogen concentrations of 0, 100, and 200 mg L-1 were applied as KNO3 to the non-nodulating side, while the nodulating side received a nitrogen-free nutrient solution. Carbon partitioning in roots and nodules was monitored using 13C-labelled CO2. Other nodule responses were measured via the estimation of the nitrogenase activity and the microscopic observation of nodule ultrastructure.ResultsElevated concentrations of nitrate applied on the non-nodulating side caused a decrease in the number of bacteroids, fusion of symbiosomes, enlargement of the peribacteroid spaces, and onset of degradation of poly-β-hydroxybutyrate granules, which is a form of carbon storage in bacteroids. These microscopic observations were associated with a strong decrease in the nitrogenase activity of nodules. Furthermore, our data demonstrate that the assimilated carbon is more likely to be allocated to the non-nodulating roots, as follows from the competition for carbon between the symbiotic and non-symbiotic sides of the dual-root system.ConclusionWe propose that there is no carbon competition between roots and nodules when they are indirectly supplied with nitrate, and that the reduction of carbon fluxes to nodules and roots on the nodulating side is the mechanism by which the plant systemically suppresses nodulation under nitrogen-replete conditions.

Straw return is crucial for the sustainable development of rice planting, but no consistent results were observed for the effect of straw return on rice growth. To investigate the response of rice leaves to rice straw return in Northeast China, two treatments were set, no straw return (S0) and rice straw return (SR). We analyzed the physiological index of rice leaves and measured differentially expressed proteins (DEPs) and differentially expressed metabolites (DEMs) levels in rice leaves by the use of proteomics and metabolomics approaches. The results showed that, compared with the S0 treatment, the SR treatment significantly decreased the dry weight of rice plants and non-structural carbohydrate contents and destroyed the chloroplast ultrastructure. In rice leaves of SR treatment, 329 DEPs were upregulated, 303 DEPs were downregulated, 44 DEMs were upregulated, and 71 DEMs were downregulated. These DEPs were mainly involved in photosynthesis and oxidative phosphorylation, and DEMs were mainly involved in alpha-linolenic acid metabolism, galactose metabolism, glycerophospholipid metabolism, pentose and gluconic acid metabolism, and other metabolic pathways. Rice straw return promoted the accumulation of scavenging substances of active oxygen and osmotic adjustment substances, such as glutathione, organic acids, amino acids, and other substances. The SR treatment reduced the photosynthetic capacity and energy production of carbon metabolism, inhibiting the growth of rice plants, while the increase of metabolites involved in defense against abiotic stress enhanced the adaptability of rice plants to straw return stress.

Most phosphorus (P) in soil exists in nonlabile forms, leading to poor soil P supply capacity and limiting crop growth. This study evaluated the effect of 10 years of rice straw return on rice yield, soil P budget, P fractions, and phosphatase activity to establish the relationship between soil P fractions and related microbial communities. Four treatments, i.e., no rice straw return (S0), low amount of rice straw return (S1), high amount of rice straw return (S2), and abandoned farmland (AL), were used in the evaluation. The results showed that rice straw return had no effect on the rice yield and P uptake, and the P budget was positive in the S2 treatment. Rice straw return increased the phosphatase activity and content of soil Olsen-P, total P, NaHCO3-, and NaOH-extractable P, and the phosphatase activity and P fractions were both increased with the amount of straw returned. There was a positive correlation between most soil P fractions and active organic carbon fractions. Rice straw return changed the composition and abundance of soil phosphate-solubilizing bacteria (PSB). The findings showed that straw return decreased the proportion of soil nonlabile P, enhancing the soil P supply capacity, and they further showed that the abundance of PSB was not consistent with soil P content.

The potato is the third largest food crop, and nitrogen fertilizer is important for increasing potato yields; however, the shallow root system of potatoes causes the nitrogen fertilizer utilization rate to be low, which results in waste and environmental pollution, meaning that high nitrogen efficiency breeding is highly significant for potatoes. In the high nitrogen efficiency breeding of potatoes, genes with a nitrogen-deficient response should first be identified, and RNA-seq is an efficient method for identifying nitrogen-deficiency-response genes. In this study, two potato cultivars, Dongnong 322 (DN322) and Dongnong 314 (DN314), were utilized, and two nitrogen fertilizer application rates (N0 and N1) were set for both cultivars. Through the determination of physiological indicators, we identified that DN314 is more sensitive to nitrogen fertilizer, while DN322 is relatively insensitive to nitrogen fertilizer. Samples were taken at the seedling and tuber formation stage. At the seedling stage, DN322 and DN314 had 573 and 150 differentially expressed genes (DEGs), while at the tuber formation stage, they had 59 and 1905 DEGs, respectively. A total of three genes related to a low-nitrogen response were obtained via the combined analysis of differentially expressed genes (DEGs) and weighted correlation network analysis (WGCNA), of which two genes were obtained at the tuber formation stage and one gene in the seedling stage, providing theoretical guidance for the high nitrogen efficiency breeding of potatoes.

The present study carried out with a field plot experiment and light environment simulation experiment in 2017 to assess the effect of spectral distribution on internode elongation in soybean canopy. Henong 60 and Heinong 48 were used as the experimental materials. The spectral distribution in the soybean canopy was studied under four planting densities 200,000(D20), 300,000(D30), 400,000(D40) and 500,000(D50) plant ha−1. Meanwhile, a pot experiment of light environment simulation with light-emitting diode (LED) was carried out to further analyze the effects of light quantity and quality on the elongation of soybean seedlings. The results showed that the intensity of PAR, blue light, red light and far-red light in soybean canopy decreased by varying degrees with density and canopy depth increasing, and the decrease of PAR and R/FR led to significant elongation of internodes and plant height in high density. The spectral intensity in the middle and base of the canopy was strongly reduced, resulting in the strongest internode elongation in the middle of soybean stem. The increase in soybean plant height was also mainly due to the internode elongation in the middle at high density. In addition, the mono-light of far-red light could promote the internode elongation, while the red light and blue light acted as inhibitors and the inhibitory effect of blue light was even greater.

Nitrogen (N) inhibits soybean (Glycine max L.) nodulation and N2 fixation. Isoflavones secreted by soybean roots can stimulate signal transduction for symbiotic nodules, thus playing a key role in root nodule development and N2 fixation. The relationship between the inhibition of soybean nodulation, N2 fixation and isoflavones by N is still unclear. In this study, dual-root soybean plants were prepared by grafting, and N or isoflavones were supplied to unilateral roots. The number and dry weight of the soybean nodules, nitrogenase activity, isoflavone concentrations and relative changes in the level of expression of nodulation-related genes were measured to study the response relationship between the N systemic regulation the soybean nodule N2 fixation and changes in the concentrations of isoflavones in its roots. The results showed that N supply to one side of the dual-root soybeans systematically affected the N2 fixation of root nodules on both sides, and this effect began in the early stage of nodulation. Moreover, a unilateral supply of N systematically affected the concentrations of daidzein and genistein on both sides of the roots. The concentrations of isoflavones were consistent with the change trend of soybean root nodule and nodulation-related gene expression level. Treatment with unilateral N or isoflavones affected the soybean nodule N2 fixation and its nodulation-related genes, which had the same response to the changes in concentrations of root isoflavones. N regulates soybean nodulation and N2 fixation by systematically affecting the concentrations of isoflavones in the roots.

In agroecosystems, a plant-usable form of nitrogen is mainly generated by legume-based biological nitrogen fixation, a process that requires phosphorus (P) as an essential nutrient. To investigate the physiological mechanism whereby phosphorus influences soybean nodule nitrogen fixation, soybean root nodules were exposed to four phosphate levels: 1 mg/L (P stress), 11 mg/L (P stress), 31 mg/L (Normal P), and 61 mg/L (High P) then proteome analysis of nodules was conducted to identify phosphorus-associated proteome changes. We found that phosphorus stress-induced ribosomal protein structural changes were associated with altered key root nodule protein synthesis profiles. Importantly, up-regulated expression of peroxidase was observed as an important phosphorus stress-induced nitrogen fixation-associated adaptation that supported two nodule-associated activities: scavenging of reactive oxygen species (ROS) and cell wall growth. In addition, phosphorus transporter (PT) and purple acid phosphatase (PAPs) were up-regulated that regulated phosphorus transport and utilization to maintain phosphorus balance and nitrogen fixation function in phosphorus-stressed root nodules.

Appropriate irrigation schedules could minimize the existing imbalance between agricultural water supply and crop water requirements (ETc), which is severely impacted by climate change. In this study, different hydrological years (a wet year, normal year, dry year, and an extremely dry year) in Heilongjiang Province were calculated by hydrological frequency methods. Then, the single crop coefficient method was used to calculate the maize ETc, based on the daily meteorological data of 26 meteorological stations in Heilongjiang Province from 1960 to 2020. Afterward, the CROPWAT model was used to calculate the effective precipitation (Pe) and irrigation water requirement (Ir), and formulate the irrigation schedules of maize in Heilongjiang Province under different hydrological years. The results showed that ETc and Ir decreased first and then increased from west to east. The Pe and crop water surplus deficit index increased first and then decreased from west to east in Heilongjiang Province. Meanwhile, the average values of the Ir in were 171.14 mm, 232.79 mm, 279.08 mm, and 334.47 mm in the wet year, normal year, dry year, and extremely dry year, respectively. Heilongjiang Province was divided into four irrigation zones according to the Ir of different hydrological years. Last, the irrigation quotas for the wet year, normal year, dry year, and extremely dry year were 0~180 mm, 20~240 mm, 60~300 mm, and 80~430 mm, respectively. This study provides reliable support for maize irrigation practices in Heilongjiang Province, China.

Abstract Outbreaks of gummy stem blight (GSB), an emerging seed pumpkin disease, have increased in number and have become more widespread in recent years. Previously we reported that Stagonosporopsis cucurbitacearum (Sc.) is the dominant fungal cause of pumpkin seedling GSB in Northeast China, where it has greatly reduced crop yields in that region. Here, high-throughput whole-genome sequencing and assembly of the Sc. genome were conducted toward revealing pathogenic molecular regulatory mechanisms involved in fungal growth and development. Zq-1 as representative Sc. strain, DNA of Zq-1was prepared for genomic sequencing, we obtained 5.24 Gb of high-quality genomic sequence data via PacBio RS II sequencing. After sequence data was processed to filter out low quality reads, a hierarchical genome-assembly process was employed that generated a genome sequence of 35.28 Mb in size. A total of 9844 genes were predicted, including 237 non-coding RNAs, 1024 genes encoding proteins with signal peptides, 2066 transmembrane proteins and 756 secretory proteins.Transcriptional identification revealed 54 differentially expressed secretory proteins. Concurrently, 605, 130 and 2869 proteins were matched in the proprietary databases Carbohydrate-Active EnZymes database (CAZyme), Transporter Classification Database (TCDB) and Pathogen–Host Interactions database (PHI), respectively. And 96 and 36 DEGs were identified form PHI database and CAZyme database, respectively. In addition, contig00011.93 was an up-regulated DEG involving ATP-binding cassette metabolism in the procession of infection. In order to test relevance of gene predictions to GSB, DEGs with potential pathogenic relevance were revealed through transcriptome data analysis of Sc. strains pre- and post-infection of pumpkin. Interestingly, Sc. and Leptosphaeria maculans (Lm.) exhibited relatively similar with genome lengths, numbers of protein-coding genes and other characteristics. This work provides a foundation for future exploration of additional Sc. gene functions toward the development of more effective GSB control strategies.

Climate change refers to the statistically significant changes in the mean and dispersion values of meteorological factors. Characterizing potential evapotranspiration (ET0) and its climatic causes will contribute to the estimation of the atmospheric water cycle under climate change. In this study, based on daily meteorological data from 26 meteorological stations in Heilongjiang Province from 1960 to 2019, ET0 was calculated by the Penman–Monteith formula, linear regression method and the Mann–Kendall trend test were used to reveal the seasonal and inter-annual changing trend of ET0. The sensitivity-contribution rate method was used to clarify the climatic factors affecting ET0. The results showed that: (1) From 1960 to 2019, the maximum temperature (Tmax), minimum temperature (Tmin) and average temperature (Tmean) showed an increasing trend, with climate tendency rate of 0.22 °C per decade (10a), 0.49 °C/(10a), 0.36 °C/(10a), respectively. The relative humidity (RH), wind speed (U) and net radiation (Rn) showed a decreasing trend, with a climate tendency rate of −0.42%/(10a), −0.18 m/s/(10a), −0.08 MJ/m2/(10a), respectively. (2) ET0 showed a decreasing trend on seasonal and inter-annual scales. Inter-annually, the average climate tendency rate of ET0 was −8.69 mm/(10a). seasonally, the lowest climate tendency rate was −6.33 mm/(10a) in spring. (3) ET0 was negatively sensitive to Tmin, and RH, while positively sensitive to Tmax, TmeanU and Rn, its sensitivity coefficient of U was the highest, which was 1.22. (4) The contribution rate of U to ET0 was the highest on an inter-annual scale as well as in spring and autumn, which were −8.96%, −9.79% and −13.14%, respectively, and the highest contribution rate to ET0 were Rn and Tmin in summer and winter, whose contribution rates were −4.37% and −11.46%, respectively. This study provides an understanding on the response of evapotranspiration to climatic change and further provides support on the optimal allocation of regional water resource and agricultural water management under climate change.

Crop water production function models (WPFMs) are required methods to study the relationships between yield and water consumption under regulated deficit irrigation (RDI). In this study, a pot experiment was established to study the effect of water deficit during both individual growth stages and across two consecutive growth stages of rice on yield, water consumption, and water use efficiency (WUE) in 2017 and 2018. Light, medium, and severe water deficits were set as 80~90%, 70~80%, and 60~70% of saturated soil moisture content, respectively. The accuracies of five WPFMs were tested based on the experimental results. The results showed that yields and WUE of a light water deficit were higher than those of medium and severe water deficits at each growth stage. The yields and WUE of light drought stress treatments in the flowering and milky stages were higher than the saturated soil moisture control by 4~7.4% and 5.3~20.6%, respectively. Water consumption decreased with increasing water deficit across two consecutive growth stages. The Minhas model had the highest simulation accuracy of the five WPFMs, with relatively lower AE, RMSE, Cv, CRM, and higher R2, which were 0.0002, 0.0634, 6.9965, 0.0002, and 0.9951 in 2017 and 0.0110, 0.0760, 8.9882, 0.0131, and 0.9923 in 2018, respectively. The sensitivity indices for the Minhas model more accurately reflected the sensitivity of rice yield to water deficit at different growth stages in 2017 and 2018, compared with the Jensen model, Stewart model, Blank model, and Singh model. Rice yield was most sensitive to water deficit at the jointing and booting stage. The results indicate that the Minhas model is the most suitable WPFM for guiding rice irrigation practices in cold and black soil regions of China.

The production of potato (Solanum tuberosum L.) faces a severe challenge due to the salinization of arable land worldwide. The cultivation of salt-tolerant potatoes is of great significance to ensure food security. In this study, two cultivars of ‘Longshu 5’ and ‘Qingshu 9’ were compared for physiological responses to salt stress, and then the salt tolerance of the two cultivars were assessed via principal component analysis. Furthermore, the Na+, K+, and Ca2+ flux of the cultivars under salt stress was recorded. Finally, the expression levels of ion transport-related genes and transcription factors in salt-tolerant cultivars were explored under NaCl stress. The results showed that the seven physiological indicators of salt tolerance were differed between the cultivars. Interestingly, soluble protein and sugar were early responsive to salt stress than proline in the salt-tolerance cultivar. Peroxidase and superoxide dismutase activity were significantly different in ‘Longshu 5’ under NaCl stress and without being significantly different in ‘Qingshu9’. In addition, the salt tolerance of ‘Longshu 5’ was more tolerant than ‘Qingshu 9’ based on principal component evaluation. Meanwhile, the strong efflux of Na+, the stability of K+, and the high absorption of Ca2+ in ‘Longshu 5’ indicated salt adaption mechanisms in the salt-tolerant potato. In addition, we found that ion transport-related genes and transcription factors, such as StSOS1, StNHX4, StAKT1, StNAC24, and StCYP707A, played a role in the salt tolerance of ‘Longshu 5’. In conclusion, the salt-tolerant potato can regulate physiological substances to adapt to salt stress, and ion transport related genes and transcription factors play a role in improving salt tolerance.

Nitrate absorbed by soybean (Glycine max L. Merr.) roots from the soil can promote plant growth, while nitrate transported to nodules inhibits nodulation and nodule nitrogen fixation activity. The aim of this study was to provide new insights into the inhibition of nodule nitrogen (N) fixation by characterizing the transport and distribution of nitrate in soybean plants. In this research, pot culture experiments were conducted using a dual root system of soybeans. In the first experiment, the distribution of 15N derived from nitrate was observed. In the second experiment, nitrate was supplied–withdrawal–resupplied to one side of dual-root system for nine consecutive days, and the other side was supplied with N-free solution. Nitrate contents in leaves, stems, petioles, the basal root of pealed skin and woody part at the grafting site were measured. Nitrate transport and distribution in soybean were analyzed combining the results of two experiments. The results showed that nitrate supplied to the N-supply side of the dual-root system was transported to the shoots immediately through the basal root pealed skin (the main transport route was via the phloem) and woody part (transport was chiefly related to the xylem). There was a transient storage of nitrate in the stems. After the distribution of nitrate, a proportion of the nitrate absorbed by the roots on the N-supply side was translocated to the roots and nodules on the N-free side with a combination of the basal root pealed skin and woody part. In conclusion, the basal root pealed skin and woody part are the main transport routes for nitrate up and down in soybean plants. Nitrate absorbed by roots can be transported to the shoots and then retranslocated to the roots again. The transport flux of nitrate to the N-free side was regulated by transient storage of nitrate in the stems.

The return of straw to soil can help closing the nutrient cycle in agricultural ecosystems and is an effective means of improving the physical and chemical properties of soil. The objective of this research was to evaluate the effect of rice straw return on soil organic carbon (SOC) fractions and microbial community with a 10-year plot experiment in Northeast China. The study involved four treatments: no rice straw return (S0), a low amount of rice straw return (S1), a high amount of rice straw return (S2), and abandoned farmland (AL). The results indicated that the labile organic carbon (LOC), particulate organic carbon (POC), dissolved organic carbon (DOC), and microbial biomass carbon (MBC) in the S1, S2, and AL treatments were significantly higher than those in the S0 treatment. Rice straw return increased soil microbial richness but did not affect the soil microbial diversity. Proteobacteria, Bacteroidetes, Firmicutes, Acidobacteria, and Verrucomicrobia were the dominant phyla in all treatments based on their relative abundances. Redundancy analysis indicated that MBC and DOC had relatively large influences on the microbial community.

The specific mechanisms by which nitrogen affects nodulation and nitrogen fixation in leguminous crops are still unclear. To study the relationship between nitrogen, nodulation and nitrogen fixation in soybeans, dual-root soybean plants with unilateral nodulation were prepared by grafting. At the third trifoliate leaf (V3) to fourth trifoliate leaf (V4) growth stages (for 5 days), nitrogen nutrient solution was added to the non-nodulated side, while nitrogen-free nutrient solution was added to the nodulated side. The experiment was designed to study the effects of exogenous nitrogen on proteins and metabolites in root nodules and provide a theoretical reference for analyzing the physiological mechanisms of the interaction between nitrogen application and nitrogen fixation in soybean root nodules. Compared with no nitrogen treatment, exogenous nitrogen regulated the metabolic pathways of starch and sucrose metabolism, organic acid metabolism, nitrogen metabolism, and amino acid metabolism, among others. Additionally, exogenous nitrogen promoted the synthesis of signaling molecules, including putrescine, nitric oxide, and asparagine in root nodules, and inhibited the transformation of sucrose to malic acid; consequently, the rhizobia lacked energy for nitrogen fixation. In addition, exogenous nitrogen reduced cell wall synthesis in the root nodules, thus inhibiting root nodule growth and nitrogen fixation.

Phosphorus (P) is an important nutrient affecting nodulation and nitrogen fixation in soybeans. To further investigate the relationship of phosphorus with soybean nodulation and nitrogen fixation, the seedling grafting technique was applied in this study to prepare dual-root soybean systems for a sand culture experiment. From the unfolded cotyledon stage to the initial flowering stage, one side of each dual-root soybean system was irrigated with nutrient solution containing 1 mg/L, 31 mg/L, or 61 mg/L of phosphorus (phosphorus-application side), and the other side was irrigated with a phosphorus-free nutrient solution (phosphorus-free side), to study the effect of local phosphorus supply on nodulation and nitrogen fixation in soybean. The results are described as follows: (1) Increasing the phosphorus supply increased the nodules weight, nitrogenase activity, ureide content, number of bacteroids, number of infected cells, and relative expression levels of nodule nitrogen fixation key genes (GmEXPB2, GmSPX5, nifH, nifD, nifK, GmALN1, GmACP1, GmUR5, GmPUR5, and GmHIUH5) in root nodules on the phosphorus-application side. Although the phosphorus-application and phosphorus-free sides demonstrated similar changing trends, the phosphorus-induced increases were more prominent on the phosphorus-application side, which indicated that phosphorus supply systematically regulates nodulation and nitrogen fixation in soybean. (2) When the level of phosphorus supply was increased from 1 mg/L to 31 mg/L, the increase on the P– side root was significant, and nodule phosphorus content increased by 57.14–85.71% and 68.75–75.00%, respectively; ARA and SNA were 218.64–383.33% and 11.41–16.11%, respectively, while ureide content was 118.18–156.44%. When the level of phosphorus supply was increased from 31mg/L to 61mg/L, the increase in the regulation ability of root and nodule phosphorus content, ARA, SNA, and ureide content were low for roots, and the value for nodules was lower than when the phosphorus level increased from 1 mg/L to 31 mg/L. (3) A high-concentration phosphorus supply on one side of a dual-root soybean plant significantly increased the phosphorus content in the aboveground tissues, as well as the roots and nodules on both sides. In the roots on the phosphorus-free side, the nodules were prioritized for receiving the phosphorus transported from the aboveground tissues to maintain their phosphorus content and functionality.

Excessive plant height is an important factor that can lead to lodging, which is closely related to soybean yield. Gibberellins are widely used as plant growth regulators in agricultural production. Gibberellic acid (GA3), one of the most effective active gibberellins, has been used to regulate plant height and increase yields. The mechanism through which GA3 regulates internode elongation has been extensively investigated. In 2019 and 2020, we applied GA3 to the stems, leaves, and roots of two soybean cultivars, Heinong 48 (a high-stalk cultivar) and Henong 60 (a dwarf cultivar), and GA3 was also applied to plants whose apical meristem was removed or to girded plants to compare the internode length and stem GA3 content of soybean plants under different treatments. These results suggested that the application of GA3 to the stems, leaves, and roots of soybean increased the internode length and GA3 content in the stems. Application of GA3 decreased the proportion of the pith in the soybean stems and primary xylem while increasing the proportion of secondary xylem. The apical meristem is an important site of GA3 synthesis in soybean stems and is involved in the regulation of stem elongation. GA3 was shown to be transported acropetally through the xylem and laterally between the xylem and phloem in soybean stems. We conclude that the GA3 level in stems is an important factor affecting internode elongation.

The specific mechanisms by which nitrogen affects the nodulation and nitrogen fixation of soybean plants are unclear. Investigating the relationship between nitrogen, nodulation and nitrogen fixation can provide new insights for the rational and proper use of nitrogen fertilizer in soybean plants. In this study, we grafted soybean roots to construct a dual-root system with a single nodulated side. Experiment I was performed at the third trifoliate leaf to initial seed filling (V3-R3) growth stages (for 30 days) for long-term nitrogen supply, and Experiment II was performed at the third trifoliate leaf to fourth trifoliate leaf (V3-V4) growth stages (for 5 days) for short-term nitrogen supply. For the two experiments, a nutrient solution providing 15NH415NO3 or NH4NO3 as the nitrogen source was added to the non-nodulated side, while a nitrogen-free nutrient solution was added to the nodulated side. The concentrations of nitrogen supplied were 0 mg/L, 25 mg/L, 50 mg/L, 75 mg/L, and 100 mg/L. The results showed the following: (1) Short-term nitrogen supply systematically regulated the specific nitrogenase activity (SNA), thereby inhibiting the acetylene reduction activity (ARA). Under long-term nitrogen supply, the recovery of SNA was generally consistent across treatments, and the concentration of nitrogen supplied systematically regulated the growth of root nodules, thereby inhibiting the ARA. (2) Using the 15N tracer method, the concentration of fertilizer nitrogen was positively correlated with the amount of nitrogen redistributed to other organs. Although the percentage of nitrogen derived from the atmosphere (Ndfa%) decreased significantly with increasing concentrations of nitrogen supplied, the effect on the accumulation of nitrogen fixed by nodules (Naccumulation of nodules) was not significant. By establishing the relationships between the ARA (measured by the acetylene reduction method), Ndfa% (based on 15N calculations), and Naccumulation nodules (based on 15N calculations), it was found that the ARA reliably reflected the Ndfa% but not the Naccumulation of nodules.

The soybean (Glycine max L. Merr.) is a crop with a high demand for nitrogen (N). The root nodules that form in soybeans can fix atmospheric N effectively, yet the goal of achieving high yields cannot be met by relying solely on nodule-fixed N. Nonetheless, the application of N fertilizer may inhibit nodule formation and biological N fixation (BNF), but the underpinning mechanisms are still unclear. In this study, we grafted the roots of non-nodulated soybeans onto nodulated soybeans to generate plants with dual root system. The experiment included three treatments conducted under sand culture conditions with NO 3 − and NH 4 + as N sources. Treatment I: The non-nodulated roots on one side received 50 mg·L−1 15 NO 3 − or 15NH4+, and the nodulated roots on the other side were not treated. Treatment II: The non-nodulated roots received 50 mg·L−1 15 NO 3 − or 15 NH 4 + , and the nodulated roots received 50 mg·L−1 14 NO 3 − or 14 NH 4 + . Treatment III: Both non-nodulated and nodulated roots received 50 mg·L−1 15 NO 3 − or 15 NH 4 + . The results showed the following: (1) Up to 81.5%−87.1% of the N absorbed by the soybean roots and fixed by the root nodules was allocated to shoot growth, leaving 12.9%−18.5% for root and nodule growth. Soybeans preferentially used fertilizer N in the presence of a NO 3 − or NH 4 + supply. After the absorbed fertilizer N and nodule-fixed N was transported to the shoots, a portion of it was redistributed to the roots and nodules. The N required for root growth was primarily derived from the NO 3 − or NH 4 + assimilated by the roots and the N fixed by the nodules, with a small portion translocated from the shoots. The N required for nodule growth was primarily contributed by nodule-fixed N with a small portion translocated from the shoots, whereas the NO 3 − or NH 4 + that was assimilated by the roots was not directly supplied to the nodules. (2) Based on observations of the shoots and one side of the roots and nodules in the dual root system as an N translocation system, we proposed a method for calculating the N translocation from soybean shoots to roots and nodules during the R1−R5 stages based on the difference in the 15N abundance. Our calculations showed that when adding N at a concentration of 50 mg·L−1, the N translocated from the shoots during the R1−R5 stages accounts for 29.6%−52.3% of the N accumulation in nodulated roots (Rootn) and 9.4%−16.6% of the N accumulation in Nodulen of soybeans. Through the study of this experiment, the absorption, distribution and redistribution characteristics of fertilizer N and root nodule N fixation in soybean can be clarified, providing a theoretical reference for analyzing the mechanisms of the interaction between fertilizer N and nodule-fixed N.
