Due to the use of lumbar folding steering for heavy-duty articulated vehicles in coal mines, the lateral stiffness is weak and the disturbance factors are complex, making it more difficult to operate and easily causing safety problems. A new pump controlled electro-hydraulic steering system with lateral motion feedback compensation was proposed to improve the safe driving and handling performance of coal mine heavy-duty articulated vehicles in wet and low adhesion road environments, and further enhance the intelligent level of coal mine auxiliary transportation. A four-degree-of-freedom dynamics model of articulated vehicle and a mathematical model of pump-controlled electro-hydraulic steering system were established, including front body longitudinal, lateral, yaw and rear body yaw motion. Based on the yaw velocity and lateral acceleration of the decoupling dynamics model, the two-channel articulated steering control strategy of “steering control follow - yaw stability compensation” was designed. The steering control follow channel can realize the real-time tracking of the steering input of the driver by the articulated angle. The yaw stability compensation channel ensures that the actual yaw velocity is close to the ideal yaw velocity by actively adjusting the articulated angle. By setting up a pre-filters and attenuation integrator, an active steering control system was designed, which provides corrective action by adjusting the steering angle to ensure that the vehicle can follow the driver's intention and maintain path following. Based on the dSPACE/DS1007 semi physical simulation platform, some simulation tests were conducted on the low adhesion steering conditions of articulated mining vehicles. The effectiveness of the active controller for tracking the desired path of articulated vehicles was verified by simulating the J-shaped turning condition and selecting the low friction coefficient road conditions for the medium to low speed operation. Meanwhile, based on the experimental conditions of equivalent low adhesion coefficient road surface covered with ice and snow, a 25 t coal mine heavy-duty articulated support truck was used for actual vehicle turning experiments. The results show that the dynamics model of articulated vehicle and a mathematical model of pump-controlled electro-hydraulic steering system established can truly reflect the lumbar folding motion state of this type of vehicle by decoupling the yaw rate and lateral acceleration. The two-channel articulated steering control strategy of "steering control follow - yaw stability compensation" based on the analysis of articulated vehicle yaw stability control, as well as the designed active control system, can effectively improve the lateral stability of articulated vehicles under low adhesion coefficient road conditions, and the system has not changed the driver's driving experience, and after active control intervention, it can timely, safely, and smoothly hand over the vehicle's driving rights to the driver.

Peripheral nervous system (PNS) injuries often lead to significant sensory and motor impairments. Traditional artificial nerve conduits, lacking anisotropic structures, have been associated with prolonged repair time and failures in nerve regeneration. This study aimed to address these challenges by developing a novel approach for rapid repair of peripheral nerve injuries (PNI). A 3D oriented fibers scaffold featuring distinct radial (RFs) and longitudinal (LFs) fibers orientations was engineered using coaxial electrospinning and gas directional foaming techniques. This scaffold was then integrated with a shape memory conduit to form a directional multi-channel nerve conduit with micro/nanostructures. The results revealed that the grooved surface of the fibers significantly improved cellular directional guidance, effectively facilitating the migration of SCs from the periphery towards the center and from the base to the apex of the scaffold. In a rat model with a 10 mm nerve defect, the ND-PLATMC/LF ND-PCL scaffold significantly enhanced nerve regeneration and motor function recovery within 4 weeks. These results suggest the potential of this innovative scaffold for efficient repair of the nerve injuries.

ObjectivePrevious studies have confirmed a positive correlation between the Triglyceride-Glucose (TyG) index and future risk of diabetes. BackgroundThe red blood cell distribution width (RDW) is closely linked to the prognosis of multiple diseases. However, the connection between RDW and gastrointestinal bleeding (GIB) in stroke patients is not well understood. This study aimed to clarify this association.MethodsThis retrospective study involved 11,107 hospitalized patients from 208 hospitals in the United States, admitted between January 1, 2014, and December 31, 2015. We examined clinical data from 7,512 stroke patients in the intensive care unit (ICU). Multivariate logistic regression assessed the link between RDW and in-hospital GIB in stroke patients. Generalized additive model (GAM) and smooth curve fitting (penalty spline method) were utilized to explore the non-linear relationship between RDW and GIB in stroke patients. The inflection point was calculated using a recursive algorithm, and interactions between different variables were assessed through subgroup analyses.ResultsAmong the 11,107 screened stroke patients, 7,512 were included in the primary analysis, with 190 identified as having GIB. Abstract Previous research has established a strong link between pulse pressure (PP) and diabetes, but there is limited investigation into the connection between PP and prediabetes. This study aims to explore the potential association between PP and prediabetes. A retrospective cohort study encompassed 202,320 Chinese adults who underwent health check-ups between 2010 and 2016. Prediabetes was defined in accordance with the World Health Organization criteria, indicating impaired fasting glucose, with fasting blood glucose levels ranging from 6.1 to 6.9 mmol/L. To assess the PP-prediabetes relationship, we employed Cox regression analysis, sensitivity analysis, and subgroup analysis. Abstract This primary objective of our study was to investigate the relationship between serum calcium levels and the occurrence of sudden cardiac arrest (SCA) in stroke patients. We analyzed the clinical data of 10,423 acute stroke patients admitted to the intensive care unit. The association between serum calcium and SCA following an acute stroke was assessed through multivariate logistic regression. We explored the non-linear connection between serum calcium levels and SCA in stroke patients using a generalized additive model and smooth curve fitting. Our study uncovered that serum calcium serves as an independent risk factor for sudden cardiac arrest in stroke patients. Notably, we observed that the relationship between serum calcium levels upon admission and the occurrence of SCA in stroke patients within the hospital was non-linear. Furthermore, we identified inflection points in serum calcium levels at 8.2 and 10.4 mg/dL. These findings emphasize a non-linear relationship between serum calcium levels and the risk of SCA in stroke patients. Maintaining serum calcium within the range of 8.2–10.4 mg/dL could lead to a significant reduction in the incidence of cardiac arrest among stroke patients.

Wave energy conversion holds promise for renewable energy, but challenges like high initial costs hinder commercialization. Integrating wave-energy converters (WECs) into shore-protection structures creates dual-function structures for both electricity generation and coastal protection. Oscillating water columns (OWCs) have been well studied in the past with their simple generation mechanism and their out-of-water power take-off (PTO) system, which can minimize bio-fouling effects and maintenance costs compared to other submerged WECs. In addition, a slotted barrier allows for better circulation behind the breakwater while dissipating incoming wave energy through viscous damping. This study examines the performance of a new design which combines an OWC with a slotted breakwater. Small-scale (1:49) laboratory tests were performed with a piston-type wave generator. The performance is evaluated in terms of wave transmission, wave energy extraction, and wave loading under various wave conditions while focusing on the effects of the porosity of the slotted barrier and tide level changes. Results show that under larger waves, a decreasing wave transmission, increasing power extraction from the OWC, and energy dissipation from the slotted barrier are observed. On the other hand, under increasing wavelengths, wave transmission is observed to be constant; this is important for harbor design, which means that the breakwater is effective under a wider range of wavelengths. Porosity allows for more transmission while inducing less horizontal force on the structure.

Under the background of global carbon neutrality, soil carbon accumulation and transformation become a hot topic in academic circles. In the process of land reclamation and restoration, the change and its mechanism of soil carbon pool in coal mining subsidence area under the influence of groundwater are complicated. The study of carbon dynamics in reclaimed soil is an important basis for a scientific restoration of reclaimed soil ecosystem. In this study, the age chronosequense approach and the location-monitoring method were used to study the change of soil carbon pool after the reclamation of coal mining subsidence area in the area with higher level of underground water. Soil samples from the study areas were collected twice, respectively in 2012 and 2020. The content of soil total carbon (CTC, total organic carbon (CTOC), total inorganic carbon (CTIC) were measured, and soil carbon storage of TC pool (STC), TOC pool (STOC), TIC pool (STIC) and rates of carbon sequestration were calculated. The soil properties (i.e., soil bulk density, compaction, porosity and soil structure, etc.) associated with the change of soil carbon were also analyzed simultaneously. The results show that: ① the STC, STOC and STIC of soil in non-subsidence area (CK) were 268.34, 81.63, 186.70 t/hm2 respectively. Land subsidence caused the decrease of STC and STIC, but with no significant change of STOC. ② A quadratic equation curve was shown in the change of soil carbon density and carbon storage in the restoration process of reclaimed soil, the CTC and CTIC of 0−20 cm layer reaching the maximum in 14−18 years after reclamation, and exceeding CK. The STOC could exceed CK in 13−14 years of reclamation. ③ There was no significant difference in carbon density and carbon pool between layers in 0−40 cm of newly reclaimed soil. In the process of restoration after reclamation, the largest increase of carbon density and carbon storage was found in the surface layer (0−10 cm), and there was no regular variation of soil CTIC in the profile for study. ④ The soil CTOC, STOC and CTC were positively correlated with soil porosity, soil clay, fractal dimension (D) and content of aggregate larger than 3.2 mm, but negatively correlated with bulk density, compactness, pH and aggregate content of 2−0.50 mm. There was great potential for carbon sequestration in the reclaimed soil. Soil moisture affected the carbon fixation and transformation of reclaimed soil by influencing soil aeration, aggregate formation and particle change in the area with higher level of underground water. The cultivation could improve soil structure and ventilation condition, promote the growth of plants and soil microorganisms, increase the source of soil organic carbon, and promote the accumulation of reclaimed soil organic carbon by ploughing, crushing, applying organic fertilizer and water level adjustment.

Abstract Background Circulating tumor DNA (ctDNA) has been becoming a novel convenient and noninvasive method for dynamically monitoring landscape of genomic information to guild personalized cancer treatment. In this study we comprehensively evaluated the additional value of plasma ctDNA to routine tissue next generation sequencing (NGS) of therapeutically targetable mutations in lung cancers. Methods The tumor tissues and peripheral blood samples from 423 cases of patients with lung cancer were subjected to NGS of mutations in oncodrivers (EGFR, ERBB2, ALK, ROS1, C-MET, KRAS, BRAF, RET, BRCA1 and BRCA2). Results One hundred and ninety-seven cases showed both plasma and tissue positive and 96 showed both negative. The concordance for tissue and blood detection was 69.27% (293/423). 83 (19.62%) cases showed positive by tissue NGS alone and 47 (11.11%) positive by plasma ctDNA alone. The sensitivity of tissue and plasma detection was 85.63%, and 74.62%, respectively. Plasma had lower detection and sensitivity than tissue, but plasma additionally detected some important mutations which were omitted by tissue NGS. Plasma plus tissue increased the detection rate of 66.19% by tissue alone to 77.30% as well as the sensitivity of 85.63–100%. Similar results were also observed when the cases were classified into subpopulations according to different stages (IV vs. III vs. I-II), grades (low vs. middle grade) and metastatic status (metastasis vs. no metastasis). Conclusion Plasma ctDNA shares a high concordance with tissue NGS, and plasma plus tissue enhances the detection rate and sensitivity by tissue alone, implying that the tissue and plasma detection should be mutually complementary in the clinical application.

Osteosarcoma (OS) is a malignant tumor with an extremely poor prognosis, especially in progressive patients. Immunotherapy based on immune checkpoint inhibitors (ICIs) is considered to be a promising treatment option for OS. Due to tumor heterogeneity, only a minority of patients benefit from immunotherapy. Therefore, it is urgent to explore a model that can accurately assess the response of OS to immunotherapy. In this study, we obtained the single-cell RNA sequencing datasets of OS patients from public databases and defined 34 cell clusters by dimensional reduction and clustering analysis. PTPRC was applied to identify immune cell clusters and nonimmune cell clusters. Next, we performed clustering analysis on the immune cell clusters and obtained 25 immune cell subclusters. Immune cells were labeled with CD8A and CD8B to obtain CD8+ T cell clusters. Meanwhile, we extracted the differentially expressed genes (DEGs) of CD8+ T cell clusters and other immune cell clusters. Furthermore, we constructed a prognostic model (CD8-DEG model) based on the obtained DEGs of CD8+ T cells, and verified the excellent predictive ability of this model for the prognosis of OS. Moreover, we further investigated the value of the CD8-DEG model. The results indicated that the risk score of the CD8-DEG model was an independent risk factor for OS patients. Finally, we revealed that the risk score of the CD8-DEG model correlates with the immune profile of OS and can be used to evaluate the response of OS to immunotherapy. In conclusion, our study revealed the critical role of CD8 cells in OS. The risk score model based on CD8-DEGs can provide guidance for prognosis and immunotherapy of OS.

The study of oscillating water column (OWC)-type wave energy converters (WEC) has primarily focused on applications in the nearshore environment with an end use in residential power grids. This study examines the power performance of a new OWC geometry relative to blue economy energy objectives that focus on providing power in the intermediate-water-depth environment. The method examines power performance through the performance indicators of extraction efficiency, coefficient of amplification, and coefficient of pressure. The study discusses the implications of these coefficients relative to a new end use and examines if an analytical relationship between each indicator exists. The power performance is evaluated through experimental testing on a fixed-geometry OWC that consists of a cylindrical OWC chamber affixed above a heave plate with V-shaped channels. In evaluating power performance, the impact of different representative power take-off (PTO) damping values and directional dependence is investigated. It was found that incident wave angle has a minimal impact on extraction efficiency for this geometry and that a relationship exists between extraction efficiency, wave frequency, and PTO damping. It can be concluded that extraction efficiency can be expressed as a function of each coefficient, that an emphasis on extraction efficiency may be misguided for at-sea power applications, and that performance indicators have functionality in OWC design outside of power performance.

Abstract Background Depression is a common, easily ignored, accompanied disease of gastric cancer (GC) patients and is often observed with elevated plasma catecholamine levels. Depression frequently promotes GC progression and leads to poor clinical outcomes; however, the molecular mechanisms underlying depression‐induced GC progression remain poorly understood. We aimed to study the effects of depression on GC progression and explore possible mechanisms mediating the action of depression‐associated catecholamines on GC. Methods Depression states of GC patients were graded using the Patient Health Questionnaire‐9, and plasma catecholamine levels were examined by high performance liquid chromatography coupled with tandem mass spectrometry. Migrative and invasive GC cells were examined using transwell assays, and metastatic GC niches were imaged using bioluminescence technology in a depression mouse model established with chronic unpredictable mild stress. Mouse depression‐like behaviors were assessed through sucrose preference, forced swimming, and tail suspension tests. Characteristics of the neuroendocrine phenotype were observed via RT‐PCR, Western blotting, flow cytometry, and transmission electron microscopy. Results Fifty‐one GC patients (age: 53.61 ± 1.79 years; cancer duration: 3.71 ± 0.33 months; depression duration: 2.37 ± 0.38 months; male‐to‐female ratio: 1.55:1) were enrolled in the study. Depression grade was significantly higher in GC patients showing higher plasma levels of catecholamines (epinephrine: P = 0.018; noradrenaline: P = 0.009), higher oncogene metastasis‐associated in colon cancer‐1 (MACC1) level (P = 0.018), and metastasis (P < 0.001). Further, depression‐associated catecholamine specifically bound to the beta‐2 adrenergic receptor (β2‐AR) and upregulated MACC1 expression, and thus promoting neuroendocrine phenotypic transformation through direct binding between MACC1 and synaptophysin. Eventually, the neuroendocrine phenotypic transformation accelerated GC invasion in vitro and metastasis in vivo. However, β2‐AR antagonist ICI‐118,551 or MACC1 silencing effectively blocked the catecholamine‐induced neuroendocrine phenotypic transformation and eliminated depression‐enhanced GC migration and invasion. Moreover, β2‐AR blocking or MACC1 silencing prevented GC metastasis attributed to a neuroendocrine phenotype in a depression mouse model. Conclusions Catecholamine‐induced neuroendocrine phenotypes of GC cells led to depression‐accelerated GC invasion and metastasis via the β2‐AR/MACC1 axis, while β2‐AR antagonist or MACC1 silencing could reverse it, showing promising potential therapeutic strategies for improving the outcome of GC patients with comorbid depression.

Antimicrobial-resistant (AMR) foodborne bacteria causing bacterial infections pose a serious threat to human health. In addition, the ability of some of these bacteria to form biofilms increases the threat level as treatment options may become compromised. The extent of antibiotic resistance and biofilm formation among foodborne pathogens remain uncertain globally due to the lack of systematic reviews. We performed a meta-analysis on the global prevalence of foodborne pathogens exhibiting antibiotic resistance and biofilm formation using the methodology of a Cochrane review by accessing data from the China National Knowledge Infrastructure (CNKI), PubMed, and Web of Science databases between 2010 and 2020. A random effects model of dichotomous variables consisting of antibiotic class, sample source, and foodborne pathogens was completed using data from 332 studies in 36 countries. The results indicated AMR foodborne pathogens has become a worrisome global issue. The prevalence of AMR foodborne pathogens in food samples was greater than 10% and these foodborne pathogens were most resistant to β-lactamase antibiotics with Bacillus cereus being most resistant (94%). The prevalence of AMR foodborne pathogens in human clinical specimens was greater than 19%, and the resistance of these pathogens to the antibiotic class used in this research was high. Independently, the overall biofilm formation rate of foodborne pathogenic bacteria was 90% (95% CI, 68%–96%) and a direct linear relationship between biofilm formation ability and antibiotic resistance was not established. Future investigations should document both AMR and biofilm formation of the foodborne pathogen isolated in samples. The additional information could lead to alternative strategies to reduce the burden cause by AMR foodborne pathogens.

Inappropriate use of antibiotics eventually leads to the emergence of antibiotic-resistant strains and invalidates the treatment of infectious diseases. Aminoglycoside antibiotics (AGAs) are a class of broad-spectrum cationic antibiotics widely used for the treatment of Gram-negative bacterial infections. Understanding the AGA resistance mechanism of bacteria would increase the efficacy of treating these infections. This study demonstrates a significant correlation between AGA resistance and the adaptation of biofilms by Vibrio parahaemolyticus (VP). These adaptations were the result of challenges against the aminoglycosides (amikacin and gentamicin). Confocal laser scanning microscope (CLSM) analysis revealed an enclosure type mechanism where the biological volume (BV) and average thickness (AT) of V. parahaemolyticus biofilm were significantly positively correlated with amikacin resistance (BIC) (p < 0.01). A neutralization type mechanism was mediated by anionic extracellular polymeric substances (EPSs). The biofilm minimum inhibitory concentrations of amikacin and gentamicin were reduced from 32 µg/mL to 16 µg/mL and from 16 µg/mL to 4 µg/mL, respectively, after anionic EPS treatment with DNase I and proteinase K. Here, anionic EPSs bind cationic AGAs to develop antibiotic resistance. Transcriptomic sequencing revealed a regulatory type mechanism, where antibiotic resistance associated genes were significantly upregulated in biofilm producing V. parahaemolyticus when compared with planktonic cells. The three mechanistic strategies of developing resistance demonstrate that selective and judicious use of new antibiotics are needed to win the battle against infectious disease.

Dexmedetomidine is a very useful anesthetic, and it currently has less improvement in early childhood surgery. In order to study the effect of dexmedetomidine in improving the complex congenital heart surgery in infants and young children, this paper used machine learning algorithm to improve the image, and combined MRI (Magnetic Resonance Imaging) images for diagnostic analysis. At the same time, this paper collected experimental data to carry out algorithm analysis, collected cases to design experiments, used the method proposed in this paper to conduct research, and recorded the research results. Through comparative analysis, the algorithm proposed in this study has certain effects. Moreover, it has been proved that the dexmedetomidine infants have complicated effects in complex anesthesia surgery and can provide theoretical reference for subsequent related research.

Advanced oxidation processes (AOPs) based on peroxymonosulfate (PMS) activation have been developed as an ideal pathway for completely eradication of recalcitrant organic pollutants from water environment. Herein, the V-doped graphitic carbon nitride (g-C3N4) is rationally fabricated by one-step thermal polymerization method to activate PMS for contamination decontamination. The results demonstrate the V atoms are successfully integrated into the framework of g-C3N4, which can effectively improve light absorption intensity and enhance charge separation. The V-doped g-C3N4 displays superior catalytic performance for PMS activation. Moreover, the doping content has a great influence on the activation performances. The radical quenching experiments confirm •O2−, SO4•−, and h+ are the significant species in the catalytic reaction. This work would provide a feasible strategy to exploit efficient g-C3N4-based material for PMS activation.

In recent years, with the development of green environmental protection, starch film has become of interest due to the wide availability of sources, low price, and biodegradability. Amylose/polyvinyl alcohol (PVA) blend films crosslinked with different amounts of glutaraldehyde (GLU) were prepared by a solution casting method. The cross-linking degree, water sorption, tensile property, crystallization and section morphology of the films were examined. With the increase in glutaraldehyde concentration, the cross-linking degree of the blend film was improved. The wide-angle X-ray scattering (WAXS) result indicated that cross-linking hindered the crystallization of film. The section morphology of films was examined by scanning electron microscope (SEM). The results showed that the cross-linking degree of amylose film improved while the crystallinity decreased with the increase in glutaraldehyde content. Cross-linking had no obvious effect on the water sorption property of the blend films. The cross-linking modification significantly enhanced the tensile strength and Young’s modulus, while it reduced the elongation at break of the blend films. It was found that the film with 0.5 wt % glutaraldehyde possessed the best performance: the tensile strength increased by 115%, while the elongation at break decreased by 18% even at high relative humidity (RH) of 90% compared to non-crosslinked films. The developed amylose/PVA blend films have promising application prospects as agricultural mulch films and packaging materials.

Mine tires are an essential and expensive component of heavy mining machinery. This study explored the grounding characteristics and temperature field distribution of mining tires during driving as well as the relationships between the maximum temperature and tire inflation pressure, load, and speed. Two-dimensional and three-dimensional finite element models of mine tires were established. Steady-state rolling simulation analysis was conducted based on inflation and static load simulations. Temperature field simulation analysis was conducted with the tire section as a research object. The accuracy of the finite element model was verified. Analysis results demonstrated that the grounding contact area decreased with an increase in charging pressure and increased with an increase in load. With an increase in inflation pressure, the maximum normal grounding stress increased in the middle part of the tread and decreased near the shoulder. The maximum normal grounding stress continuously deviated in the shoulder direction with an increase in load. Temperature field analysis indicated that the tire had the maximum temperature at the binder position, where the first belt layer was connected to the second belt layer, which corresponds to the maximum stress position in the steady-state rolling simulations. Tire temperature increased with driving speed. The maximum temperature increased with an increase in tire deflection, whereas the deflection decreased with an increase in inflation pressure and increased with an increase in load. Speed had the greatest influence on the maximum temperature, followed by load, with inflation pressure having the smallest influence. The results of this research can be used to improve the service life of mine tires to improve productivity and reduce costs.

Abstract Background Atherosclerosis, the underlying cause of the majority of cardiovascular diseases, is a lipid-driven, inflammatory disease of the large arteries. Atherosclerotic cardiovascular disease (ASCVD) threatens human lives due to high morbidity and mortality. Many studies have demonstrated that atherosclerosis is accelerated via activation of the NLRP3 inflammasome. The NLRP3 inflammasome plays a critical role in the development of vascular inflammation and atherosclerosis. In atherosclerotic plaques, excessive generation of reactive oxygen species (ROS) activates the NLRP3 inflammasome. 13-Methylberberine (13-MB) is a newly synthesized compound used in traditional Chinese medicine that has outstanding antibacterial, antitumor, and antiobesity activities, especially anti-inflammatory activity. However, the role of 13-MB in atherosclerosis needs to be explored. Methods CCK-8 assays and flow cytometry were conducted to determine the cell viability and apoptotic profiles of human umbilical vein endothelial cells (HUVECs) treated with 13-MB. Carboxy-DCFH-DA and JC-10 assays were used to measure ROS and determine mitochondrial membrane potential. Western blot analysis was performed to investigate proteins that are associated with the NLRP3 inflammasome and autophagy. ELISA was used to detect and quantify inflammatory cytokines related to the NLRP3 inflammasome. Transfection and confocal microscopy were conducted to observe autophagy. Results Pretreatment with 13-MB markedly reduced cytotoxicity and apoptosis, as well as intracellular ROS production, in H2O2-induced HUVECs. Moreover, 13-MB showed a protective effect in maintaining mitochondrial membrane potential. 13-MB also suppressed NLRP3 inflammasome activation and promoted autophagy induction in HUVECs. Conclusion 13-MB exerts cytoprotective effects in an H2O2-induced cell injury model by inhibiting NLRP3 inflammasome activation via autophagy induction in HUVECs. These anti-inflammatory and autophagy induction activities may provide valuable evidence for further investigating the potential role of 13-MB in atherosclerosis.

Driven by the vision of Internet of Things, some research efforts have already focused on designing a network of efficient speech recognition for the development of edge computing. Other researches (such as tpool2) do not make full use of spatial and temporal information in the acoustic features of speech. In this paper, we propose a compact speech recognition network with spatio-temporal features for edge computing, named EdgeRNN. Alternatively, EdgeRNN uses 1-Dimensional Convolutional Neural Network (1-D CNN) to process the overall spatial information of each frequency domain of the acoustic features. A Recurrent Neural Network (RNN) is used to process the temporal information of each frequency domain of the acoustic features. In addition, we propose a simplified attention mechanism to enhance the portion of the network that contributes to the final identification. The overall performance of EdgeRNN has been verified on speech emotion and keywords recognition. The IEMOCAP dataset is used in speech emotion recognition, and the unweighted average recall (UAR) reaches 63.98%. Speech keywords recognition uses Google's Speech Commands Datasets V1 with a weighted average recall (WAR) of 96.82%. Compared with the experimental results of the related efficient networks on Raspberry Pi 3B+, the accuracies of EdgeRNN have been improved on both of speech emotion and keywords recognition.

The domain of Aspect Level Sentiment Classification, in which the sentiment toward a given aspect is analyzed, attracts much attention in NLP. Recently, the state-of-the-art Aspect Level Sentiment Classification methods are devised by using the Graph Convolutional Networks to deal with both the semantics and the syntax of the sentence. Generally, the parsing of syntactic structure inevitably incorporates irrelevant information toward the aspect. Besides, the syntactic and semantic alignment and uniformity that contribute to the sentiment delivery is currently neglected during processing. In this work, a Triplet Contrastive Learning Network is developed to coordinate the syntactic information and the semantic information. To start with, the aspect-oriented sub-tree is constructed to replace the syntactic adjacency matrix. Further, a sentence-level contrastive learning scheme is proposed to highlight the features of sentiment words. Based on The Triple Contrastive Learning, the syntactic information and the semantic information are thoroughly interacted and coordinated whilst the global semantics and syntax can be exploited. Extensive experiments are performed on three benchmark datasets and achieve accuracies (BERT-based) of 87.40, 82.80, 77.55 on Rest14, Lap14, and Twitter datasets, which demonstrate that our approach achieves state-of-the-art results in Aspect Level Sentiment Classification task.

This study investigates the alteration in function and number of circulating endothelial progenitor cells (EPCs) in patients with aortic dissection (AD), compared with hypertensive patients, and its possible mechanism. Thirty-four patients with acute aortic dissection (AAD) and 20 patients with primary hypertension were involved. Flow cytometry analysis was performed to detect the number of CD34+/KDR+ cells, and acetylated low density lipoprotein (ac-LDL) and lectin fluorescent staining method was applied to test the number of cultured EPCs. In addition, EPC migration and proliferation were measured, and plasma interleukin 6 (IL-6) and interleukin 17 (IL-17) levels were investigated. The number of circulating EPCs in the AAD group was lower than that in the non-AD group, and the proliferation and migration of circulating EPCs in the AAD group were lower than that in the non-AD group. In addition, the number, proliferation, and migration of circulating EPCs were significantly inversely correlated with the aortic dissection detection risk score (ADD-RS). More importantly, increased plasma IL-6 and IL-17 level was found in the AAD group, and the two inflammatory factors were inversely associated with the function and number of circulating EPCs in the AAD group. We first demonstrated that the number and function of circulating EPCs are reduced in the AAD group, which may be partly related to upregulated plasma IL-6 and IL-17. Our study provides novel insight on the underlying mechanism and potential therapeutic target of AAD.

Recently, deep learning has gained great popularity in the area of recommender systems. Various combinations of deep learning, collaborative recommendation and content-based recommendation have occurred. However, as one of the three most significant recommendation techniques, hybrid recommendation has little cooperation with deep learning. Besides, most current deep hybrid models only incorporate two simple recommendation methods together in post-fusion, leaving massive space for further exploration of better combinations. In this paper, we apply deep learning to hybrid recommendation, proposing a deep hybrid recommendation model DMFL (Deep Metric Factorization Learning). In DMFL, we combine deep learning with improved machine learning models to learn the interaction between users and items from multiple perspectives. Such deep hybrid learning helps to reflect the user preference more comprehensively and strengthen model's ability of generalization. We also propose a more accurate method of user feature representation, taking both long-term static characteristics and short-term dynamic interest changes of users into consideration. Furthermore, thorough experiments have been conducted on real-world datasets, which strongly proves the effectiveness and efficiency of the proposed model.

An information dissemination network (i.e., a cascade) with a dynamic graph structure is formed when a novel idea or message spreads from person to person. Predicting the growth of cascades is one of the fundamental problems in social network analysis. Existing deep learning models for cascade prediction are primarily based on recurrent neural networks and representation on random walks or propagation paths. However, these models are not sufficient for learning the deep spatial and temporal features of an entire cascade. Therefore, a new model, called Cascade2vec, is proposed to learn the dynamic graph representation of cascades based on graph recurrent neural networks. To learn more effective graph-level representation of cascades, the current graph neural networks are improved by designing a graph residual block, which shares attention weights between nodes, and by transforming features through perception layers. Furthermore, the proposed graph neural network is integrated into a recurrent neural network to learn the temporal features between graphs. With this method, both the spatial and temporal characteristics of cascades are learned in Cascade2vec. The experimental results show that our method significantly reduces the mean squared logarithmic error and median squared logarithmic error by 16.1% and 12%, respectively, in the cascade prediction at one hour in the Microblog network dataset compared with strong baselines.

Emotional intelligence plays an important role in human intelligence and is a recent research hotspot. With the rapid development of deep learning techniques in recent years, several neural network-based emotional text generation methods have been investigated. However, the existing emotional text generation approaches often suffer from the problem of requiring large-scale annotated data. Generative adversarial network (GAN) has shown promising results in natural language generation and data enhancement. In order to solve the above problem, this paper proposes a GAN-based cross-domain text sentiment transfer model, which uses annotated data from other domains to assist in the training of emotional text generation network. By combining adversarial reinforcement learning with supervised learning, our model is able to extract patterns of sentiment transformation and apply them in emotional text generation. The experimental results have shown that our approach outperforms the state-of-the-art methods and is able to generate high-quality emotional text while maintaining the consistency of domain information and content semantics.

Meningioma (MEN) is a common central nervous system disease. Accumulating evidence indicated that long non-coding RNA maternally expressed gene 3 (MEG3) participated in the progression of MEN. However, the potential mechanisms of MEG3 in altering the aggressive phenotypes of MEN need further exploration. Levels of MEG3, microRNA (miR)-29c, and A-kinase anchor protein 12 (AKAP12) were determined using quantitative real-time Polymerase Chain Reaction (qRT-PCR) assay. Dual-luciferase reporter and RNA immunoprecipitation (RIP) assays were performed to verify the relationship between miR-29c and MEG3 or AKAP12. The protein level of AKAP12 was detected by western blot. Moreover, cell-cycle arrest, migration, invasion, and proliferation were assessed by flow cytometry, wound healing, transwell assays, and CCK-8 assay, respectively. Levels of MEG3 and AKAP12 were downregulated, while miR-29c was effectively increased in MEN tissues and cell line. Mechanically, MEG3 was a sponge of miR-29c to regulate the expression of AKAP12. Functionally, increase of MEG3 diminished cell-cycle, migration, invasion, and proliferation in MEN cells, and reintroduction of miR-29c could eliminate these effects. In addition, AKAP12 depletion overturned the inhibitory effects of miR-29c absence on cell-cycle, migration, invasion, and proliferation in vitro. Also, AKAP12 was co-regulated by MEG3/miR-29c axis. MEG3 mediated the aggressive behaviors of MEN cells via miR-29c/AKAP12 axis, supporting that MEG3 served as a promising biomarker for the diagnosis and treatment of human MEN.

Hyperhomocysteinemia (HHcy) induced endothelial dysfunction is associated with disturbance in circulating endothelial progenitor cells (EPCs). Nevertheless, whether this unfavorable effect of HHcy on circulating EPCs also exists in premenopausal women is still unknown. Therefore, this leaves an area for the investigation of the difference on the number and activity of circulating EPCs in premenopausal women with hyperhomocysteinemia and its underlying mechanism. The number of circulating EPCs was measured by fluorescence-activated cell sorter analysis, as well as DiI-acLDL and lectin fluorescent staining. The migration and proliferation of circulating were evaluated by the Transwell chamber assay and MTT. Additionally, the endothelial function and levels of nitric oxide (NO), VEGF, and GM-CSF in plasma and culture medium were determined. The number or activity of circulating EPCs and flow-mediated dilatation (FMD) in premenopausal women with or without HHcy were higher than those in postmenopausal women. However, no significant effect of HHcy on the number or activity of circulating EPCs in premenopausal women was observed. A similar alteration in NO level between the four groups was observed. There was a correlation between FMD and the number or activity of EPCs, as well as NO level in plasma or secretion by EPCs. For the first time, our findings illuminated the quantitive or qualitative alterations of circulating EPCs and endothelial function in premenopausal patients with HHcy are preserved, which was associated with retained NO production. The recuperated endothelial repair capacity is possibly the potential mechanism interpreting cardiovascular protection in premenopausal women with HHcy.

Out-of-hospital cardiac arrest (OHCA) due to drowning carries high morbidity and mortality. There are a few studies on drowning-related out-of-hospital cardiac arrest (OHCA), in which patients are followed from the scene to hospital discharge. This study aims to compare patient characteristics between the survival group and mortality group of OHCA due to drowning. OHCA due to drowning cases were selected from the North America Termination of Resuscitation Association database between 2011 and 2015. The retrospective analysis of epidemiological characteristics and clinical features of all OHCA patients were performed. Of the 17,094 OHCA cases in the registry, 54 cases of OHCA due to drowning were included in this study. Among the 54 OHCAs due to drowning, 7 (13.0%) survived, while 47 (87.0%) died. It has been proven that vitamin D was decreased and function of circulating endothelial progenitor cells (EPCs) was injured in systemic lupus erythematosus (SLE) patients. However, the effect of vitamin D on the function of EPCs in vitro and its mechanism need further study. Therefore, we investigated whether vitamin D improved the function of EPCs in vitro. The peripheral blood mononuclear cells of the participants were isolated from SLE patients and control subjects and cultured to EPCs. After the EPCs were treated with vitamin D (1,25-(OH)2D3), we evaluated the number, migratory and proliferative activities, and nitric oxide (NO) production of EPCs in vitro and detected vascular endothelial function by flow-mediated dilatation (FMD). We found that vitamin D in a dose-dependent manner improved number and migratory and proliferative activities of EPCs from SLE patients. Additionally, vitamin D upregulated NO production from EPCs in vitro. A significant correlation between the FMD and plasma NO level was found. There was also a correlation between number, migration, and proliferation of EPCs and NO production. Thus, the present findings indicated that vitamin D improved the function of EPCs from SLE patients via NO secretion.

Abstract Background Long non-coding RNAs (lncRNAs) have emerged as important regulators of human cancers. However, the functional roles of lncRNAs and the mechanisms responsible for their aberrant expression in gastric cancer (GC) have not been well characterized. Methods In this study, we examined the expression of lncRNA UFC1 in GC by qRT-PCR and explored its correlation with clinicopathological parameters. In vitro cell functional assays and in vivo animal studies were performed to determine the roles of UFC1 in GC progression. Results UFC1 was elevated and predicted poorer prognosis in GC. UFC1 knockdown inhibited while UFC1 overexpression promoted GC cell proliferation, migration, and invasion. UFC1 bound to miR-498 to antagonize its tumor suppressive effect on Lin28b. Suppression of Lin28b by miR-498 could be rescued by UFC1 overexpression, whereas Lin28b overexpression partially rescued UFC1 knockdown-mediated inhibition of GC cell function. Lin28b expression was increased in GC and suggested a co-expression pattern with UFC1. Conclusions UFC1 has a promoting role in GC progression, at least in part, by acting as a miR-498 sponge and derepressing Lin28b expression, which would provide a novel biomarker for GC diagnosis and prognosis and offer a potential target for GC therapy.

Abstract Background Gastric cancer is common in developing regions, and we hope to find out an economical but practical prognostic indicator. It was reported that pre-treatment peripheral neutrophil-to-lymphocyte ratio (NLR) and platelet-to-lymphocyte ratio (PLR), as well as differentiation status, were associated with cancer progression. Hence, we introduced a novel combined Neutrophil/platelet/lymphocyte/differentiation Score (cNPLDS) to improve the prediction value of palliative chemotherapeutic response in advanced gastric cancer. Methods According to statistical sample size estimation, 136 primary diagnosed unresectable advanced ptaients were included for a retrospective study. The follow-up end-point was progression free survival (PFS) during the first-line palliative chemotherapy. Differentiation stratified patients into well, medium and poor groups by score 1 to 3, while patients with neither elevated NLR and PLR, only one elevated, or both elevated were of the combined NLR-PLR score (cNPS) 1 to 3, respectively. The cNPLDS was calculated by multiplying the tumor differentiation score and cNPS. Results Determined by the receiver operating characteristic (ROC) curve, the optimal cut-off points for NLR and PLR were 3.04 and 223. Through univariate analysis and survival analysis, poor differentiation, high NLR, high PLR, high cNPS, and high cNPLDS respectively indicated inferior PFS during the first-line palliative chemotherapy. Patients were furhter classified into low to high risk groups by cNPLDS. Groups of elevated NLR, PLR, cNPS, and cNPLDS showed lower disease control rate. Compared to other parameters, cNPLDS significantly improved the accuracy in predicing the first-progression. Conclusions This study indicates that the novel parameter cNPLDS is superior to NLR or PLR alone, or even cNPS, in predicting the first-line chemosensitivity in advanced gastric cancer.

Abstract Background Osteosarcoma is one of the most malignant primary bone cancers, while is rarely reported in China. Of note, very few data of prognosis has been documented in this region. Thus, we carried a retrospective study to identify prognostic factors and to analyze outcomes in patients of both classic and non-classic high-grade osteosarcomas. Classic osteosarcoma is defined as of high-grade histology, age below 40 years, with extremity localized primary tumor, and without detectable metastasis at primary diagnosis. Methods A total of 98 patients (68 classic and 30 non-classic) aged from 4 to 64 years old were diagnosed as high-grade osteosarcoma from 2008 to 2015 in Nanfang Hospital, Guangzhou, China. Univariate and multivariate analyses were performed to identify the independent predictors for overall survival and event-free survival. Kaplan-Meier method was used for survival analysis. Results The median overall survival was 117 vs. 21 months, and the median event-free survival was 31 vs. 6 months in classic and non-classic osteosarcoma, respectively. The most frequently found tumor site was around the knee. The classic osteosarcoma had better overall survival and event-free survival than the non-classics. Tumor site and primary metastasis were found to be associated with overall survival and event-free survival in the univariate analysis. In the multivariate Cox regression analysis, tumor site and primary metastasis were each verified as independent prognostic factors. However, no similar result was found in elevated serum alkaline phosphatase or lactate dehydrogenase. Amputation or limb salvage surgery had no significant effect on overall survival and event-free survival in the extremity osteosarcomas. Classic osteosarcomas with extremity tumor site and free of primary metastasis exhibited better overall survival and event-free survival, while the axial and metastatic non-classics exhibited the worse. Conclusions The extremity classic osteosarcomas have better survivals than the axial non-classic cases. Amputation and limb salvage surgery make no significant change in overall survival and event-free survival in the extremity osteosarcomas. The connectivity of a large-scale heterogeneous Internet of Vehicles (IoV) is an important challenge for the environment of urban road scenes, which feature crossroads, buildings, and communication devices. The uneven density and wide distribution of vehicles in the city, the building barriers, and the interference of other communication technology will make the connectivity weaker. To the best of our knowledge, there is no formal method to model and analyze connectivity by considering the environment of urban road scenes. This paper presents such a theoretical method to investigate four connectivity properties- i.e., possibility, data forwarding time, link forwarding capability, and packet error rate-and deduce a connectivity model. Using experiments, we prove that the proposed model can ensure that the connectivity is effective and reliable in an urban road scene, and it can be used to accurately evaluate the network connectivity of a highly dynamic IoV.

In general, the stock prices of the same industry have a similar trend, but those of different industries do not. When investing in stocks of different industries, one should select the optimal model from lots of trading models for each industry because any model may not be suitable for capturing the stock trends of all industries. However, the study has not been carried out at present. In this paper, firstly we select 424 S&P 500 index component stocks (SPICS) and 185 CSI 300 index component stocks (CSICS) as the research objects from 2010 to 2017, divide them into 9 industries such as finance and energy respectively. Secondly, we apply 12 widely used machine learning algorithms to generate stock trading signals in different industries and execute the back-testing based on the trading signals. Thirdly, we use a non-parametric statistical test to evaluate whether there are significant differences among the trading performance evaluation indicators (PEI) of different models in the same industry. Finally, we propose a series of rules to select the optimal models for stock investment of every industry. The analytical results on SPICS and CSICS show that we can find the optimal trading models for each industry based on the statistical tests and the rules. Most importantly, the PEI of the best algorithms can be significantly better than that of the benchmark index and "Buy and Hold" strategy. Therefore, the algorithms can be used for making profits from industry stock trading.

Crystallinity is an essential indicator for evaluating the quality of fiber materials. Terahertz spectroscopy technology has excellent penetrability, no harmful substances, and commendable detection capability of absorption characteristics. The terahertz spectroscopy technology has great application potential in the field of fiber material research, especially for the characterization of the crystallinity of cellulose. In this work, the absorption peak of wood cellulose, microcrystalline cellulose, wood nano cellulose, and cotton nano cellulose were probed in the terahertz band to calculate the crystallinity, and the result compared with XRD and FT-IR analysis. The vibration model of cellulose molecular motion was obtained by density functional theory. The results showed that the average length of wood cellulose (WC) single fiber was 300 μm. The microcrystalline cellulose (MCC) was bar-like, and the average length was 20 μm. The cotton cellulose nanofiber (C-CNF) was a single fibrous substance with a length of 50 μm, while the wood cellulose nanofiber (W-CNF) was with a length of 250 μm. The crystallinity of cellulose samples in THz was calculated as follows: 73% for WC, 78% for MCC, 85% for W-CNF, and 90% for C-CNF. The crystallinity values were obtained by the three methods which were different to some extent. The absorption peak of the terahertz spectra was most obvious when the samples thickness was 1 mm and mixed mass ratio of the polyethylene and cellulose was 1:1. The degree of crystallinity was proportional to the terahertz absorption coefficients of cellulose, the five-movement models of cellulose molecules corresponded to the five absorption peak positions of cellulose.

Super-hydrophobic phenomena generally exist in nature, and wood can also obtain hydrophobicity by specific processing on the surface, being like the construction of microscale rough surface or decoration with low surface energy materials. In this research, the formation of hydrophobic layers on wood surface was investigated without breaking the wood’s original structure. The core-shell structure particles were prepared by penetrating orthosilicate and polystyrene into the hollow mesoporous microsphere structure with tetrahydrofuran. A wood sample was coated with polydimethylsiloxane (PDMS) resin layer to enhance the adhesion of nano and micron hollow mesoporous microsphere on its surface. According to the surface structure of super-hydrophobic subjects in nature, the nano and micron hollow mesoporous microsphere were sprayed with different ratios several times to form a hydrophobic surface. The water contact angle could reach 150°, revealing that the hydrophobic behavior of the nano and micron hollow mesoporous microsphere coating was achieved. The microstructures of wood samples were examined by the scanning electron microscopy, and the chemical functional groups were investigated by the Fourier transform infrared; both verified that the hydrophobic surface was successfully coated. The thermogravimetric examination revealed the improved thermal stability of the hydrophobic wood. The scratch test was used to measure the abrasion resistance of the nano and micron hollow mesoporous microsphere coatings on wood surface. It was suggested that the nano and micron hollow mesoporous microsphere coating was an effective method to fabricate extremely hydrophobic wood products.

An eco-friendly and novel water treatment material was synthesized using sodium lignosulfonate modified polystyrene (SLPS), which can be used to eliminate phenols in aqueous solution. SLPS was characterized by BET, FTIR, SEM, and EDS. The effect of the initial pH value, phenol content, adsorption time, and temperature on the absorbability of phenol in SLPS was investigated through adsorption experiments. It was found that SLPS could efficiently adsorb phenol in aqueous solution at a pH value of about 7. The test results revealed that the kinetic adsorption and isotherm adsorption could be successfully described using the pseudo second-order and Langmuir models, respectively. It was illustrated that the phenol adsorption on SLPS was dominated by chemisorption and belonged to monolayer adsorption. The max. phenol adsorption value of SLPS was 31.08 mg/g at 30 °C. Therefore, SLPS displayed a great potential for eliminating phenol from polluted water as a kind of novel and effective adsorbent.

The passive surface wave survey is a practical, non-invasive seismic exploration method that has increasingly been used in geotechnical engineering. However, in situ deployment of traditional wired geophones is labor intensive for a dense sensor array. Alternatively, stand-alone seismometers can be used, but they are bulky, heavy, and expensive because they are usually designed for long-term monitoring. To better facilitate field applications of the passive surface wave survey, a low-cost energy-efficient geophone system was developed in this study. The hardware design is presented in this paper. To validate the system’s functionality, both laboratory and field experiments were conducted. The unique feature of this newly-developed cableless geophone system allows for rapid field applications of the passive surface wave survey with dense array measurements.

This study developed a new cable-less seismograph system, which can transmit seismic data in real-time and automatically perform high-precision differential self-positioning. Combining the ZigBee technology with the high-precision differential positioning module, this new seismograph system utilized the wireless personal area network (WPAN) and real-time kinematic (RTK) technologies to improve its on-site performances and to make the field quality control (QC) and self-positioning possible. With the advantages of low-cost, good scalability, and good compatibility, the proposed new cable-less seismograph system can improve the field working efficiency and data processing capability. It has potential applications in noise seismology and mobile seismic monitoring.

Endothelial progenitor cells (EPCs) have endogenous endothelium-reparative potential, but obesity impairs EPCs. Overweight premenopausal women have a normal number of circulating EPCs with functional activity, but whether EPCs in overweight postmenopausal women can repair obesity-related endothelial damage requires further investigation. For this purpose, we examined the function and number of circulating EPCs, evaluated vascular endothelial function, and explored the underlying mechanism. Compared with normal weight or overweight age-matched men, postmenopausal women (overweight or normal weight) had a diminished number of circulating EPCs and impaired vascular endothelial function, as detected by flow-mediated dilatation. Moreover, GTCPH I expression and the nitric oxide level in overweight postmenopausal women and men were significantly decreased. Together, our findings demonstrate that the number or function of circulating EPCs and endothelial function, which is partially regulated by the GTCPH I/BH4 signaling pathway, is not preserved in overweight postmenopausal women. The GTCPH I/BH4 pathway in circulating EPCs may be a potential therapeutic target for endothelial injury in overweight postmenopausal women.

Autonomous vehicles are considered to have great potentials in improving transportation safety and efficiency. Autonomous follow driving is one of the highly probable application forms of autonomous vehicles in the near future. In this article, we aim at the basic autonomous following form with one follower and one leader. Proper longitudinal regulation of the follower vehicle is essential for the driving quality of the two-vehicle platoon. Focusing on this problem, a novel longitudinal control method composing of a learning-based acceleration decision phase and an internal model–based acceleration tracking phase is proposed for the follower vehicle. In the acceleration decision phase, proper acceleration commands of the follower that adjusts the following distance converging to the target value are determined by a near-optimal acceleration policy which is obtained through an online reinforcement learning algorithm named neural dynamic programming. In the acceleration tracking phase, throttle and brake control commands that drive the vehicle as the decided acceleration are derived by an internal model control structure. The performance of our proposed method is verified by simulation experiments conducted with CarSim, an industry recognized vehicle dynamic simulator.

Technological advances have led to the emergence of wireless sensor nodes in wireless networks. Sensor nodes are usually battery powered and hence have strict energy constraints. As a result, energy conservation is very important in the wireless sensor network protocol design and the limited power resources are the biggest challenge in wireless network channels. Link adaptation techniques improve the link quality by adjusting medium access control (MAC) parameters such as frame size, data rate, and sleep time, thereby improving energy efficiency. In this paper we present an adaptive packet size strategy for energy efficient wireless sensor networks. The main goal is to reduce power consumption and extend the whole network life. In order to achieve this goal, the paper introduces the concept of a bounded MAB to find the optimal packet size to transfer by formulating different packet sizes for different arms under the channel condition. At the same time, in achieve fast convergence, we consider the bandwidth evaluation according to ACK. The experiment shows that the packet size is adaptive when the channel quality changes and our algorithm can obtain the optimal packet size. We observe that the MAB packet size adaptation scheme achieves the best energy efficiency across the whole simulation duration in comparison with the fixed frame size scheme, the random packet size and the extended Kalman filter (EKF).

The water-soluble protein from Phascolosoma esculenta was hydrolyzed by pepsin to obtain the hydrolysate with angiotensin I-converting enzyme (ACE) inhibitory activity. The hydrolysate (PEPH) was then further separated by membrane bioreactor system, ion-exchange chromatography, gel filtration, and reversed phase high-performance liquid chromatography (RP-HPLC) and a novel ACE inhibitory peptide named as PeP with the IC50 value of 135 M was isolated. The amino acid sequence, Ala-Trp-Leu-His-Pro-Gly-Ala-Pro-Lys-Val-Phe, was identified by matrix-assisted laser desorption ionization-time of flight/time of flight (MALDI-TOF/TOF). Inhibitory kinetics study suggested that PeP acted as competitive inhibitor against ACE. Single oral administration of synthesized PeP at 10 mg/kg dose in spontaneously hypertensive rats could reduce the systolic blood pressure around 30 mmHg and the effect could last for more than 8 h. The results suggest that peptide from P. esculenta could be a potent natural ingredient for functional foods or pharmaceuticals against hypertension.

Three sulfated polysaccharides from Bullacta exarata were isolated, purified, and named BEP1, BEP2 and BEP3, respectively. The antitumor and antioxidant activities, in vitro, of three polysaccharides were investigated, including inhibition of cells proliferation, hydroxyl radical scavenging effect, superoxide radical scavenging capacity and reducing power assay. The results suggested that three polysaccharides possessed antioxidant activities in a dose-dependent manner, and BEP3 exhibited stronger antioxidant activities than BEP1 and BEP2. Furthermore, BEP3 showed significant inhibitory effects on growth of Bcap37 breast cancer cells, SW1990 pancreatic cancer cells and HeLa cervical cancer cells, and the IC50 were 135.3, 147.5 and 172.6 μg/ml, respectively. The highest inhibition rates of BEP1 and BEP2 were approximately 10% against three cancer cells. The data obtained in vitro models indicates that polysaccharides from B. exarata could be explored as novel and potential natural antioxidants and cancer prevention agents for use in functional foods.
