Abstract Background The symbiosis between arbuscular mycorrhizal fungi (AMF) and plants often stimulates plant growth, increases agricultural yield, reduces costs, thereby providing significant economic benefits. AMF can also benefit plants through affecting the rhizosphere microbial community, but the underlying mechanisms remain unclear. Using Rhizophagus intraradices as a model AMF species, we assessed how AMF influences the bacterial composition and functional diversity through 16 S rRNA gene sequencing and non-targeted metabolomics analysis in the rhizosphere of aluminum-sensitive soybean that were inoculated with pathogenic fungus Nigrospora oryzae and phosphorus-solubilizing fungus Talaromyces verruculosus in an acidic soil. Results The inoculation of R. intraradices, N. oryzae and T. verruculosus didn’t have a significant influence on the levels of soil C, N, and P, or various plant characteristics such as seed weight, crude fat and protein content. However, their inoculation affected the structure, function and nutrient dynamics of the resident bacterial community. The co-inoculation of T. verruculosus and R. intraradices increased the relative abundance of Pseudomonas psychrotolerans, which was capable of N-fixing and was related to cry-for-help theory (plants signal for beneficial microbes when under stress), within the rhizosphere. R. intraradices increased the expression of metabolic pathways associated with the synthesis of unsaturated fatty acids, which was known to enhance plant resistance under adverse environmental conditions. The inoculation of N. oryzae stimulated the stress response inside the soil environment by enriching the polyene macrolide antifungal antibiotic-producing bacterial genus Streptomyces in the root endosphere and upregulating two antibacterial activity metabolic pathways associated with steroid biosynthesis pathways in the rhizosphere. Although inoculation of pathogenic fungus N. oryzae enriched Bradyrhizobium and increased soil urease activity, it had no significant effects on biomass and N content of soybean. Lastly, the host niches exhibited differences in the composition of the bacterial community, with most N-fixing bacteria accumulating in the endosphere and Rhizobium vallis only detected in the endosphere. Conclusions Our findings demonstrate that intricate interactions between AMF, associated core fungi, and the soybean root-associated ecological niches co-mediate the regulation of soybean growth, the dynamics of rhizosphere soil nutrients, and the composition, function, and metabolisms of the root-associated microbiome in an acidic soil.

Selenium (Se) pollution is mainly caused by anthropogenic activities, and the resulting biosecurity concerns have garnered significant attention in recent years. Using one-compartmental toxicokinetic (TK) modelling, this study explored the kinetic absorption, sub-tissue distribution, and elimination processes of the main Se species (selenate, Se(VI)) in the cultivated aerobic soil of the earthworm Eisenia fetida. The bio-accessibility of earthworm-derived Se was assessed using an in vitro simulated gastrointestinal digestion test to evaluate its potential trophic risk. The results demonstrated that Se accumulated in the pre-clitellum (PC) and total tissues (TT) of earthworms in a time- and dose-dependent manner. The highest Se levels in the PC, post-clitellum (PoC), and TT were 70.54, 57.93, and 64.26 mg/kg during the uptake phase, respectively. The kinetic Se contents in the earthworms PC and TT were consistent with the TK model but not with PoC. The earthworm TT exhibited a faster uptake (Kus = 0.83–1.02 mg/kg/day) and elimination rate of Se (Kee = 0.044–0.049 mg/kg/day), as well as a shorter half-life time (LT1/2 = 15.88–14.22 days) than PC at low soil Se levels (≤5 mg/kg). Conversely, the opposite trend was observed with higher Se concentrations (10 and 20 mg/kg). These results are likely attributable to the tissue specificity and concentration of the toxicant. Earthworms PC and TT exhibited a higher kinetic Se accumulation factor (BAFk) than steady-state BAF (BAFss), with values ranging from 8 to 24 and 3–13, respectively. Furthermore, the bio-accessibility of earthworm-derived Se to poultry ranged from 66.25 % to 84.35 %. As earthworms are at the bottom of the terrestrial food chain, the high bio-accessibility of earthworm-derived Se poses a potential risk to predators. This study offers data support and a theoretical foundation for understanding the biological footprint of soil Se and its toxicological impacts and ecological hazards.

Abstract Background Plant diseases caused by pathogenic fungi are devastating. However, commonly used fungicides are harmful to the environment, and some are becoming ineffective due to fungal resistance. Therefore, eco-friendly biological methods to control pathogenic fungi are urgently needed. Results In this study, a strain, Paenibacillus sp. lzh-N1, that could inhibit the growth of the pathogenic fungus Mycosphaerella sentina (Fr) Schrorter was isolated from the rhizosphere soil of pear trees, and the complete genome sequence of the strain was obtained, annotated, and analyzed to reveal the genetic foundation of its antagonistic ability. The entire genome of this strain contained a circular chromosome of 5,641,488 bp with a GC content of 45.50%. The results of species identification show that the strain belongs to the same species as P. polymyxa Sb3-1 and P. polymyxa CJX518. Sixteen secondary metabolic biosynthetic gene clusters were predicted by antiSMASH, including those of the antifungal peptides fusaricidin B and paenilarvins. In addition, biofilm formation-related genes containing two potential gene clusters for cyclic lactone autoinducer, a gene encoding S-ribosylhomocysteine lyase (LuxS), and three genes encoding exopolysaccharide biosynthesis protein were identified. Conclusions Antifungal peptides and glucanase biosynthesized by Paenibacillus sp. lzh-N1 may be responsible for its antagonistic effect. Moreover, quorum sensing systems may influence the biocontrol activity of this strain directly or indirectly.

Phenazine-1-carboxylic acid (PCA) is a biologically active substance with the ability to prevent and control crop diseases. It was certified as a pesticide by the Ministry of Agriculture of China in 2011 and was named “Shenzimycin.” Lzh-T5 is a Pseudomonas chlororaphis strain found in the rhizosphere of tomatoes. This strain can produce only 230 mg/L of PCA. We used LDA-4, which produces the phenazine synthetic intermediate trans-2,3-dihydro-3-hydroxyanthranilic acid in high amounts, as the starting strain. By restoring phzF and knocking out phzO, we achieved PCA accumulation. Moreover, PCA production was enhanced after knocking out negative regulators, enhancing the shikimate pathway, and performing fed-batch fermentation, thus resulting in the production of 10,653 mg/L of PCA. It suggested that P. chlororaphis Lzh-T5 has the potential to become an efficiency cell factory of biologically active substances.

The clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR‐associated (Cas) genome editing system provides a powerful toolbox for genetic engineering. However, safe and efficient in vivo delivery of all CRISPR components into the nuclei of target organs remains challenging toward CRISPR/Cas clinical translation. With the rapid development of nanotechnology, the nonviral CRISPR/Cas mRNA delivery systems are emerging as one of the most clinically advanced platforms. CRISPR/Cas mRNA formulated in lipid nanoparticles is currently under clinical trials for treating hereditary transthyretin amyloidosis. Nonviral delivery of CRISPR/Cas systems in mRNA format not only minimizes immunogenicity and carcinogenicity, but also enables transient genome editing, thereby reducing potential off‐target effects. The recent advances in the nonviral CRISPR/Cas mRNA delivery systems should facilitate CRISPR technologies toward clinical applications.

Continuous-variable quantum key distribution (CV–QKD) is a system that provides secret keys for symmetric key systems. In the application of CV–QKD, the practical security of the system is crucial. In this article, we investigate the practical security issues caused by non–ideal Gaussian modulation, which is caused by fitting defects of the amplitude modulator’s (AM) modulation curve. We provide the effect of fitting error on parameter estimation. We also give the relationship between the fitting order and the secret key rate. The simulation results indicate that the system is completely unable to communicate during first–order fitting. During second–order fitting, the system’s performance decreases by more than half. During third–order fitting, the system’s performance will be consistent with the ideal. Therefore, to ensure the performance of the CV–QKD system, the fitting order must be at least three or higher.

Cyanobacterial blooming is an increasing environmental issue all over the world. Algicidal bacteria are potential tools for the control of algal blooms. The algicidal activity in many bacteria exhibits quorum-sensing (QS) dynamics and the regulatory mechanism of this activity in these bacteria is unclear. In this study, combining genomic sequencing and genome editing, we have identified that the primary quorum-sensing system in the isolated algicidal strain Shewanella xiamenensis Lzh-2 is the LuxS/AI-2 signaling pathway. Disruption of the QS system through recombination deletion of the LuxS gene led to a loss of algicides production and algicidal activity. Restoration of the LuxS gene in the deletion mutant compensated the QS system and recovered the algicidal activity. Consequently, we proved that Lzh-2 regulates the algicidal activity through LuxS/AI-2 quorum-sensing system.

Heating network constraint is one of the important factors that affect the scale of electro-thermal coupling scheduling. This paper first establishes an electrothermal coupling scheduling model considering the multi-source ring heating network pipe structure and then proposes a method of simplifying a multi-source cyclic heating network topology approximation. Second, the electrothermal coupling scheduling system is coordinated and solved. Finally, Through the simulation example results, the annular heating network topological approximate equivalent can simplify the model complexity of the original heating network while also retaining the thermal dynamic characteristics of the initial multi-source ring heating network. This study will greatly improve the efficiency of solving the electrothermal coupling system.

The clinical translation of messenger mRNA (mRNA)-based therapeutics requires safe and effective delivery systems. Although considerable progress has been made on the development of mRNA delivery systems, many challenges, such as the dose-limiting toxicity and specific delivery to extrahepatic tissues, still remain. Cell-derived vesicles, a type of endogenous membranous particle secreted from living cells, can be leveraged to load mRNA during or after their biogenesis. Currently, they have received increasing interest for mRNA delivery due to their natural origin, good biocompatibility, cell-specific tropism, and unique ability to cross physiological barriers. In this review, we provide an overview of recent advances in the naturally occurring mRNA delivery platforms and their biomedical applications. Furthermore, the future perspectives on clinical translation of cell-derived vesicles have been discussed.

Abstract Background Mechanical loading is an essential factor for bone formation. A previous study indicated that mechanical tensile strain of 2500 microstrain (με) at 0.5 Hz for 8 h promoted osteogenesis and corresponding mechanoresponsive microRNAs (miRs) were identified in osteoblasts. However, in osteocytes, it has not been identified which miRs respond to the mechanical strain, and it is not fully understood how the mechanoresponsive miRs regulate osteoblast differentiation. Methods Mouse MLO-Y4 osteocytes were applied to the same mechanical tensile strain in vitro. Using molecular and biochemical methods, IGF-1 (insulin-like growth factor-1), PGE2 (prostaglandin E2), OPG (osteoprotegerin) and NOS (nitric oxide synthase) activities of the cells were assayed. MiR microarray and reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assays were applied to select and validate differentially expressed miRs, and the target genes of these miRs were then predicted. MC3T3-E1 osteoblasts were stimulated by the mechanical tensile strain, and the miR-29b-3p expression was detected with miR microarray and RT-qPCR. Additionally, the effect of miR-29b-3p on IFG-1 secretion of osteocytes and the influence of conditioned medium of osteocytes transfected with miR-29b-3p on osteoblast differentiation were investigated. Results The mechanical strain increased secretions of IGF-1 and PGE2, elevated OPG expression and NOS activities, and resulted in altered expression of the ten miRs, and possible target genes for these differentially expressed miRs were revealed through bioinformatics. Among the ten miRs, miR-29b-3p were down-regulated, and miR-29b-3p overexpression decreased the IGF-1 secretion of osteocytes. The mechanical strain did not change expression of osteoblasts’ miR-29b-3p. In addition, the conditioned medium of mechanically strained osteocytes promoted osteoblast differentiation, and the conditioned medium of osteocytes transfected with miR-29b-3p mimic inhibited osteoblast differentiation. Conclusions In osteocytes (but not osteoblasts), miR-29b-3p was responsive to the mechanical tensile strain and regulated osteoblast differentiation via regulating IGF-1 secretion of mechanically strained osteocytes.

Abstract The construction and control of high‐order coupled vortices are a significant challenge for promoting the application of magnetic vortices. Thus far, only double‐coupled vortices have been produced and modulated in some ferromagnetic nanostructures. Here, an effective approach is provided to obtain a high‐order coupled vortex structure by using a chiral nanostructure. Double‐vortex, triple‐vortex, and n‐vortex chains can be successfully constructed using structured Fe4N nanostrips and bias nanomagnets. The designed chiral nanostructure cannot only control the transport and hybridization of vortices but also modulate the domain walls of the vortex chain for spin wave (SW) propagation. At the exciting frequency of 1.2 GHz, the SW propagates along the domain walls formed in the vortex chain. Upon increasing the frequency to 5.0 GHz, the SW gradually spreads from the domain walls into domains. This technique will present a new perspective for the design and application of magnetic vortex‐based devices.

The clinical manifestations of variant angina is unevenly distributed during the 24 h, thus the in vivo performance of drugs should be tailored according to the angina circadian rhythm. Cryptotanshinone (CTN) is one of the representative bioactive lipid-soluble components of Danshen which has been commonly used for cardiovascular diseases such as angina pectoris. The aim of this study was to develop a novel CTN sustained-released pellets (CTN-SRPs) to precisely synchronize the CTN plasma concentrations with predicted occurrence of angina pectoris for angina chronotherapy. A deconvolution-based method was applied to develop and optimize the CTN-SRPs. The plasma concentration-time curve of CTN immediate-released formulation after oral administration in rats was used as the weight function. The predicted plasma concentration-time curve of CTN-SRPs simulated according to the incidence of variant angina during 24 h was used as the response function. Then the desired drug release profile of CTN-SRPs was calculated based on deconvolution using weight function and response function, and subsequently used for guiding the formulation optimization. CTN-SRPs were prepared with the combinations of PVP, poloxamer 127 and EC as matrix using fluidized bed technology. An orthogonal design was employed to obtain the optimal formulation with its release profile similar with the desired one. Pharmacokinetic studies validated that the actual plasma concentration-time curve of these optimized CTN-SRPs was similar with the predicted one. In addition, the percent errors (%PE) of CTN plasma concentrations in 8–12 h were less than 10%. In conclusion, this deconvolution-based method could be applied to adjust the in vivo performance of drugs for angina chronotherapy. We numerically simulated dynamic propagation of in-phase and out-of-phase finite-energy Airy pulses in anomalous regions of optical fibers and analyzed the effects of cubic-quintic (CQ) competing nonlinearity and separation parameter on evolution properties. Numerical results showed entirely different effects of quintic nonlinearity on in-phase and out-of-phase Airy pulses that vary with different separation parameters. For in-phase Airy pulses, a central quasi-soliton pulse is shed from all finite-energy Airy pulses when separation parameter is certain. CQ competing nonlinearity exhibits profound effects on propagation dynamics of soliton pulses. However, for out-of-phase Airy pulses, two quasi-soliton pulses and more dispersion wave pulses are symmetrically shed from the main lobe of out-of-phase Airy pulses. CQ competing nonlinearity slightly impacts evolution of out-of-phase Airy pulses depending on peak intensity, which varies with propagation distance and unlike in cases of in-phase Airy pulses.

In this work, SiCp/7085 composites with a volume fraction of 10% were fabricated by ultrasonic stirring casting. The evolution of the microstructure and wear properties of SiCp/7085 composites, as well as the mechanism of ultrasonic action in the process of preparation, when the ultrasonic treatments last for 0 min, 5 min, 10 min, and 15 min, respectively, were studied using the optical effects (OM), scanning electron microscope (SEM), and X-ray diffraction (XRD). The experimental results show that the high temperature and high pressure formed by the cavitation of ultrasound can effectively eliminate the cluster of particles, improve the distribution of SiC particles, increase the interfacial wettability of the composites, and form MgAl2O4 and MgO on the particle surface, thereby ultimately improving the wear resistance of the composites. Because the particles in the melt are affected by the segregation effect of ultrasonic standing wave field, there exists an optimal value for the ultrasonic treatment time. When the melt is treated with the ultrasonic wave with an amplitude of 12 μm and a treatment duration of 10 min in the test conditions given in this paper, the composites have the most uniform distribution of particles, the best wear resistance, a large and stable friction coefficient, and the minimum weight loss of sample wear: 12 mg. The change of wear resistance is consistent with the variation of solidification structure and interface properties.

In situ TiB2/2A14 composites with a 3% volume fraction were prepared by mixing salt reaction and high energy ultrasound. The effects of high-intensity ultrasonic on the microstructure and mechanical properties of TiB2/2A14 composites were systematically investigated. The microstructures of the composites were analyzed using scanning electron microscopy (SEM) and electron backscattered diffraction (EBSD). The phase composition was examined by X-ray diffraction (XRD) and energy dispersive spectroscopy (EDS). The results showed that after introducing ultrasonic vibration into the melt, due to the cavitation and acoustic streaming effect, the particle agglomerations were significantly reduced and particles of different sizes were evenly dispersed in the matrix. With ultrasonic vibration treatment of 120 s, the agglomerations were basically eliminated, and the particles were uniformly distributed to the most. The yield strength, tensile strength and elongation of the composites were increased by 53%, 21% and 30%, respectively, compared with that without ultrasonic vibration treatment (UVT).
