29. cyy-287, a novel pyrimidine-2,4-diamine derivative, inhibits tumor growth of EGFR-driven non-small cell lung cancer via the ERK pathway

Abstract

In recent decades, EGFR-targeted tyrosine kinase inhibitors (TKIs) have been proven to be an effective therapy for EGFR-mutant non-small cell lung cancer (NSCLC). However, resistance to EGFR-TKIs limits their clinical application. In the present study, we investigate the antitumor effect and underlying mechanism of a novel pyrimidine-2,4-diamine derivative, cyy-287, in NSCLC. We find that cyy-287 has a high affinity for lung tissue and inhibits the proliferation of NSCLC cells. Interestingly, the significant suppression of migration and induction of apoptosis by cyy-287 are only observed in EGFR-driven but not in EGFR-wild-type (wt) cells. According to the RNA sequencing and KEGG enrichment analysis results, cyy-287 markedly inhibits the MAPK pathway in EGFR-driven PC9 cells, and western blot analysis results further indicate that cyy-287 selectively blocks the ERK pathway in EGFR-driven cells. Meanwhile, apoptosis induced by cyy-287 could be partially reversed by ERK pathway inhibition. Further experiment indicates that cyy-287 inhibits the EGFR pathway in both EGFR-driven and EGFR-overexpressing cells. Interestingly, it only induces apoptosis in EGFR-driven cells, not in EGFR-overexpressing cells. The growth of EGFR-driven cells is suppressed by cyy-287 in vivo, with fewer side effects. Our results suggest that cyy-287 may be a potential therapeutic drug with promising antitumor effects against NSCLC.

42. Impact of Multiple Radar Wind Profilers Data Assimilation on Convective Scale Short-Term Rainfall Forecasts: OSSE Studies over the Beijing-Tianjin-Hebei region.

Abstract

The optimal spatial layout for a radar wind profiler (RWP) network in rainfall forecasting, especially over complex terrain, remains uncertain. This study explores the benefits of assimilating vertical wind measurements from various RWP network layouts into convective-scale numerical weather prediction (NWP) through observing system simulation experiments (OSSEs). Synthetic RWP data were assimilated into the Weather Research and Forecasting (WRF) model using the National Severe Storms Laboratory three-dimensional variational data assimilation (DA) system for three southwest (SW)-type heavy rainfall events in the Beijing-Tianjin-Hebei region. Four types of DA experiments were conducted and compared: a control experiment (CTL) that assimilates data solely from the operational RWP network, and three additional experiments incorporating foothill (FH), ridge (RD), and combined foothill-ridge (FH_RD) RWP network layouts. A detailed examination of the 21 July 2023 case reveals that the FH_RD experiment generally exhibits more skillful storm forecasts in terms of areal coverage, storm mode, and orientation, benifiting from refined mesoscale wind analysis. Particularly, in the RD experiment, RWP data assimilation notably reduces wind errors and enhances mesoscale dynamics near the Taihang Mountains upstream of Beijing, crucial for convective initiation (CI). Aggregated score metrics across all cases also indicate that both FH and RD experiments offer substantial added value over the operational network alone. Further sensitivity experiments on vertical resolution and maximum detection height indicate that the RWP system configuration with the highest detection height achieves the best performance, while lower detection height degrades forecast quality. These findings highlight the importance of strategic RWP network placement along the Taihang Mountains' ridge and foothill for short-term quantitative precipitation forecast in the Beijing-Tianjin-Hebei region. Abstract

Vascular aging is an important factor leading to cardiovascular diseases such as hypertension and atherosclerosis. Hyperlipidemia or fat accumulation may play an important role in vascular aging and cardiovascular disease. Isopropyl 3-(3,4-dihydroxyphenyl)-2-hydroxypropanoate (IDHP) has biological activity and can exert cardiovascular protection, which may be related to ferroptosis. However, the exact mechanism remains undefined. We hypothesized that IDHP may have a protective effect on blood vessels by regulating vascular aging caused by hyperlipidemia or vascular wall fat accumulation. The aim of this study is to investigate the protective effect and mechanism of IDHP on palmitic acid-induced human umbilical vein endothelial cells (HUVEC) based on senescence and ferroptosis. We found that IDHP could delay vascular aging, reduce the degree of ferrous ion accumulation and lipid peroxidation, and protect vascular cells from injury. These effects may be achieved by attenuating excessive reactive oxygen species (ROS) and ferroptosis signaling pathways generated in vascular endothelial cells. In short, our study identified IDHP as one of the antioxidant agents to slow down lipotoxicity-induced vascular senescence through the ROS/ferroptosis pathway. Abstract

Obesity is associated with enhanced colonic inflammation, which is a major risk factor for colorectal cancer. Considering the obesity epidemic in Western countries, it is important to identify novel therapeutic targets for obesity-induced colonic inflammation, to develop targeted strategies for prevention. Eicosanoids are endogenous lipid signaling molecules involved in regulating inflammation and immune responses. Using an LC-MS/MS-based lipidomics approach, we find that obesity-induced colonic inflammation is associated with increased expression of soluble epoxide hydrolase (sEH) and its eicosanoid metabolites, termed fatty acid diols, in colon tissue. Furthermore, we find that pharmacological inhibition or genetic ablation of sEH reduces colonic concentrations of fatty acid diols, attenuates obesity-induced colonic inflammation, and decreases obesity-induced activation of Wnt signaling in mice. Together, these results support that sEH could be a novel therapeutic target for obesity-induced colonic inflammation and associated diseases.
