14. Optimizing Kernel Density Estimation Bandwidth for Road Traffic Accident Hazard Identification: A Case Study of the City of London.

Abstract

Road traffic accidents pose significant challenges to sustainable urban safety and intelligent transportation management. The effective hazard identification of crash hotspots is crucial in implementing targeted safety measures. A severity-weighted system was adopted to quantify crash hazard levels. Using 1059 valid crash records of the City of London, the spatial correlations of crash points were first examined via average nearest neighbor analysis. Then, the optimal KDE bandwidth was determined via ArcGIS's automatic extraction method, multi-distance spatial cluster analysis, and incremental spatial autocorrelation (ISA) analysis. The predictive accuracy index (PAI) was used to evaluate the accuracy of KDE results at various bandwidths. The results revealed a clustered spatial distribution of crash points. The optimized KDE bandwidth obtained via ISA analysis was 134 m, and the yielded PAI was 4.381, indicating better predictive accuracies and balanced hotspot distributions and reflecting both local concentrations and the overall continuity of crash hazard hotspots. Applying this bandwidth to the validation data allowed the successful identification of most high-risk areas and potential crash hazard hotspots attributed to traffic environmental factors; this method exhibits reliability, accuracy, and robustness over medium to long time scales. This workflow can serve as an analytical template for assisting planners in improving the identification accuracy of hazard hotspots, thereby reducing crash occurrences, actively promoting sustainable traffic safety development, and providing valuable insights for targeted crash prevention and intelligent traffic safety management in urban areas. 26. Real‐world application of a fast stool DNA test for colorectal cancer screening in primary screening positive population.

Abstract

Background: Stool DNA test has been emerged as an effective noninvasive method for colorectal cancer (CRC) screening, but the real‐world performance of stool DNA test in Chinese population has rarely been reported. Methods: A total of 36,527 subjects were recruited in Haining City from January 2021 to December 2021. Participants underwent primary screening by taking both two‐samples fecal immunochemical tests (FITs) and high‐risk factor questionnaire (HRFQ), and those who tested either positive by FITs or evaluated to be high risk by HRFQ were recommended to undertake subsequent stool DNA test and colonoscopy. Results: Of 36,527 participants, 34,778 (95%) completed both HRFQ and FITs, 9947 (29%) showed positive results during primary screening, and the colonoscopy compliance rate was 49%. Of primary screening positives, 8733 (88%) completed stool sample collections, and colonoscopy results from 4293 eligible participants were used for analyzing the performance of stool DNA test. The sensitivities for detecting CRC and advanced adenomas (AA) were 100% (95% CI: 60–100%) and 40% (95% CI: 34–46%), and the area under curve (AUC) was 0.961 (95% CI:0.954–0.967) and 0.625 (95% CI: 0.609–0.641), respectively. The specificity of stool DNA test was 84% (95% CI: 82–85%). The false‐positive rate for stool DNA test is about 10% less than that of primary screening. Conclusion: Stool DNA test is a cost‐effective and promising alternative strategy for CRC screening in China. [ABSTRACT FROM AUTHOR]

Abstract

This study proposed a method for identifying traffic accident (TA) hotspots. The method combines kernel density estimation (KDE) with network kernel density estimation (NKDE). The hotspots identified through NKDE can be overlaid on the high-risk areas (hot zones) identified using planar KDE, resulting in accurate hotspot identification. The research site was Zhenjiang City in Jiangsu Province, China; data on 410 fatal traffic accidents from 2017 to 2020 were collected. An average nearest neighbour (ANN) ratio (0.563) was obtained using the global auto nearest neighbour distance method; thus, the fatal TAs had a cluster-type distribution in the city. Subsequently, the maximum clustering distance (7812.842591 m) in the research site was analysed using Ripley's K-function, which yielded a reasonable KDE bandwidth and enabled identifying the distribution of hot zones for fatal TAs. Precise hotspot identification was achieved by overlaying the results of NKDE-based analysis in these hot zones. The proposed method was verified using data on 131 fatal TAs during January to July 2021. The results revealed that, in Zhenjiang City, the identification rate for hot zones and hotspots in hot zones was 71.75% and 38.29%, respectively, and the overall hotspot identification rate was 27.48%, demonstrating the method's ability to relatively accurately identify hotspot locations, information that can help traffic authorities implement preventive measures. 50. A cautionary tale of cross-contamination among plasmids from commercial suppliers

Abstract

Many researchers have switched to purchasing their desired plasmids from commercial suppliers to save time and resources, as we did for 17 high-risk human papillomavirus plasmids. To our surprise, they were shown to be cross-contaminated with one another. Comparison between the production schedule and the pattern of contaminations proved that this contamination occurred during the production process, which was also shown for another two sets of commercial plasmids. Our experience indicates that the absolute purity of plasmids obtained from external sources cannot be guaranteed. Extreme caution should be exercised, especially when such plasmids are used for human gene therapies and DNA vaccines, where even a minute amount of contamination may pose significant risks to patients.
