Carbon fibre reinforced silicon carbide (C/C-SiC) composite materials have attracted increasing attention in brake system of high-speed trains, due to their low density, high specific strength, thermal stability, and frictional wear performance. This study aims to explore the mechanical properties of the 2.5D C/C-SiC composites, in order to characterise its potential application to the friction braking system of high-speed trains. To comprehensively evaluate the mechanical performance of the 2.5D C/C-SiC composites, the chemical vapor infiltration (CVI) method was adopted to prepare novel samples with high densification and low content of residual Si. The mechanical properties and failure mechanisms of the composites were investigated under various loading conditions, including tension, compression, bending, and shear tests. The experimental results indicated that the 2.5D C/C-SiC composites exhibited superior mechanical properties, with average in-plane tensile strength, compressive strength, bending strength, and interlaminar shear strength reaching 127.67 MPa, 326.92 MPa, 355.74 MPa, and 9.77 MPa, respectively, which are 2–8 times higher than the mechanical properties of C/C-SiC composites in existing publications. Under different loading conditions, the composites demonstrated characteristics of ductile fracture, pseudo-plastic compression fracture, pseudo-plastic bending fracture, and brittle shear fracture. Both high fibre content and the formation of SiC structure were advantageous for enhancing the load-bearing performance of C/C-SiC composites. The outstanding mechanical properties of the 2.5D C/C-SiC composite render the brake disc highly resistant to deformation and cracking under high stress, thereby enhancing its durability and establishing it as the ideal material for the next generation of high-speed train brake discs.

Objective To explore the mechanism of melatonin (Mel) alleviating cholestatic liver injury in a mouse cholestasis model induced by cholic acid (CA) feeding. Methods A total of 15 8-week-old male C57BL/6J mice were randomly divided into control group, 1%CA group, and 1%CA+Mel group, with 5 animals in each group. The control group was fed with normal chow diet, and the other 2 groups were fed with a diet containing 1%CA for 14 d to construct a model of cholestasis, and intraperitoneal injection with 100 mg/kg Mel was given to the mice from the 1%CA+Mel group. Immunohistochemical assay of α-SMA was applied for the liver tissues in the 1%CA group and the 1%CA+Mel group. The mRNA expression levels of fibrosis-related indicators in mouse liver tissue were examined by RT-qPCR. Liquid chromatography tandem mass spectrometry (LC-MS/MS) and automated biochemical analyzer were used to detect the contents of bile acids in the liver tissues and the serum of mouse, respectively. Then real-time qPCR and Western blotting were applied to detect the expression of bile acid synthesis and liver detoxification enzymes related indicators at mRNA and protein levels, respectively, to further investigate the mechanism of bile acid metabolism. In the context of global climate change, the research on eco-physiological responses to drought and rewatering among provenances of trees species will help to understand the effect of climate conditions of provenances on drought resistance of trees, further providing theoretical references for cultivation and management of forests in the future. In this study, Schima superba from two provenances in southern China were selected. Potted seedlings were subjected to simulated drought and rewatering conditions. Eco-physiological traits (photosynthesis traits, hydraulic traits, non-structural carbohydrates-NSC, proline concentrations-Pro and superoxide dismutase-SOD activity) of seedlings were examined. The results were as follows: (1) Under well-watered condition, stem xylem water potential (Ψxylem), leaf relative water content (RWC), photosynthetic rate (Asat) and stomatal conductance (Gs) were lower in seedlings from Guangdong provenance compared with Fujian provenance. (2) Responses of hydraulic traits, Pro and SOD to drought and rewatering were consistent between the two provenances. Ψxylem, RWC and Pro could recover to control values rapidly, while percent of stem xylem embolism and SOD could not recover to control values in the end. (3) Asat was more sensitive to drought from Fujian provenance than Guangdong provenance, and it took more time to recover to control in Fujian provenance. (4) The recovery rate of NSC from Fujian provenance was faster than Guangdong provenance. Collectively, seedlings from both provenances could not repair their embolized xylem in the short-term rewatering period (30 days). Although photosynthesis rate in Guangdong provenance could recover to control faster than from Fujian provenance, it was lower than that from Fujian provenance. Furthermore, the recovery rate of NSC was slower in Guangdong provenance than Fujian provenance, indicating that growth and survivals of seedlings from Guangdong provenance may experience greater risk in future characterized with increasing drought stress.

This study aims to experimentally and numerically explore the bearing capacity of windshield glass for rail vehicles under projectile impact by gravel along the railway. The gravel particle sizes, ranging from 0.5 to 10 mm, were divided into 7 levels, and two conditions were chosen based on loading angles: 90° vertical impact and 45° oblique impact. Multiple gravel impacts were conducted based on the conversion of wind-blown sand flow intensity, analysing the difference in fracture velocity of windshield glass compared with single gravel impact. Finally, a stable 4 kPa wind pressure was applied to the windshield glass, and simultaneous gravel impact was performed. By comparing the impact test results without wind pressure, the weakening effect of wind pressure load on the impact resistance of windshield glass was analysed. Lastly, the dynamic process of projectile penetration and fracture of windshield glass was numerically simulated. This involved studying the crack propagation process and analysing the fracture velocity of windshield glass under projectiles made of different materials. By comparing the simulation and experimental results, the accuracy and reliability of this study were verified. The research results show that the larger the gravel particle size, the lower the chip and fracture velocities of the windshield glass. The chip velocity and fracture velocity of windshield glass under single gravel oblique impact at 45° are higher than those under vertical impact conditions, approximately 1.2 times and 1.13 times higher, respectively. Moreover, the fracture velocities under 3 and 6 gravel impacts are approximately 12 % and 20 % lower, respectively, compared to single gravel impact condition. Additionally, stress oscillations were observed to occur due to the cyclic process of stress accumulation, energy release through crack propagation, stress decline, and further stress accumulation. This was observed by tracking the stress state near the crack tip of the windshield glass.

Objective To explore the clinical practice effect and optimization method of optimizing the Traditional Chinese Medicine (TCM)nursing plan for chest obstruction and heart pain (coronary heart disease) based on Feng Yunhua's academic viewpoint of blood stasis syndrome and nursing for promoting blood circulation and resolving stasis. Methods The clinical data of 793 patients with chest obstruction and heart pain admitted to the hospital from June 2021 to May 2022 were retrospectively analyzed. Totally 390 patients with hospital admission between June 2021 and November 2021 were recruited as the control group, receiving routine nursing plan of promoting blood circulation and resolving blood stasis. Another 403 patients with hospital admission between December 2021 and May 2022 were included as the observation group, received modified nursing plan. Effect of nursing interventions on main symptoms and syndrome integral of blood stasis syndrome were measured, and patient satisfaction and evaluation of nursing program by nurses were investigated. Aiming at the problem of spectrum scarcity faced by Vehicle to Everything (V2X) communication, a deep reinforcement learning method is proposed to manage V2X spectrum resources. Firstly, the V2X communication model of a single vehicle to infrastructure link is established. Combined with the constraints such as frequency spectrum subband and transmission power, the optimization problem is constructed to maximize the comprehensive efficiency of V2X communication network. Secondly, considering the non-convexity of the optimization problem, the communication model can be regarded as a Markov decision process. Then, the Dueling-Deep Q Network (Dueling-DQN) algorithm is introduced to obtain the optimal spectrum subband selection and transmission power allocation strategy to maximize the comprehensive efficiency of V2X communication network. Finally, the simulation is carried out on tensorflow software platform to verify the performance of the proposed method. The simulation results show that Dueling-DQN algorithm can obtain higher link performance and V2X communication network efficiency compared with other algorithm.

Objective To systematically evaluate the efficacy and current application of emotional nursing of Traditional Chinese Medicine(TCM) in psycho-somatic rehabilitation for early-stage colorectal cancer(CRC) patients after radical surgery, so as to provide reference for clinical practice and future research. Methods CNKI, VIP, Wanfang, Embase, Pubmed, Conchrane database were retrieved, and the randomized controlled clinical trials of TCM emotional nursing on psychosomatic rehabilitation of early-stage colorectal cancer patients after radical surgery from 2010 to 2022 was collected. The literatures met the inclusion and exclusion criteria were selected for data extraction and quality evaluation. Meta-analysis was conducted through RevMan5. 3 statistical software. Results Totally 13 studies were included, and 1034 patients were enrolled. Meta-analysis results showed that TCM emotional nursing could significantly reduce the scores of Self-rating Anxiety Scale (SAS) ［MD=-3. 57, 95% CI: (-4. 02, -3. 12), P＜0. 01］and Self-rating Depression Scale (SDS)［MD=-6. 70, 95% CI: (-7. 28, -6. 11), P＜0. 01］ of patients after intervention compared with conventional nursing, and could improve the quality of life, sleep, pain of patients, and improve the compliance and satisfaction of nursing and cancer treatment. Conclusion TCM emotional nursing could promote the psychosomatic rehabilitation of early-stage colorectal cancer patients after radical surgery, but it still needs to further improve the quality of research, optimize and integrate the nursing practice process and details, and strive to obtain higher level of evidence, so as to better help patients benefit. (目的 系统评价中医情志护理在早中期结直肠癌根治术后患者心身康复中应用效果及现状, 为临床实践及后续研究开展提供依据。方法 检索CNKI、维普、万方、Embase、Pubmed、Conchrane, 收集2010年—2022年中医情志护理干预早中期结直肠癌术后患者心身康复的随机对照临床试验研究, 筛选符合纳入排除标准的文献进行资料提取和质量评价, 通过RevMan5. 3 统计软件进行Meta分析。结果 共纳入13项研究, 共计1034例患者。Meta分析结果显示, 中医情志护理相比常规护理能够显著降低干预后患者焦虑(SAS)［MD=-3. 57, 95% CI: (-4. 02, -3. 12), P＜0. 01］、抑郁(SDS)［MD=-6. 70, 95% CI: (-7. 28, -6. 11), P＜0. 01］评分, 并有可能改善患者生活质量、睡眠、疼痛, 并且提高护理及治疗依从性和满意度。结论 中医情志护理能够促进结直肠癌术后患者心身康复, 但仍需进一步提高研究质量、优化整合护理实践流程和细节, 争取获得更高级别循证医学证据, 从而更好地帮助患者获益。)

Objective To explore the effect and molecular mechanism of artesunate (ART) on cholestatic liver fibrosis induced by 3, 5-diethoxycarbonyl-1, 4-dihydro-2, 4, 6-collidine (DDC). Methods 8-week-old male C57BL/6J mice(n=24) were randomly divided into 3 groups: the control group(n=6), the 0.1% DDC group(n=9) and the 0.1% DDC+ART group(n=9). Mice have been fed with diet containing 0.1% DDC for 2 weeks to establish cholestatic mouse model, and ART was given intramuscular injection (30.0 mg/kg) once a day in the 0.1% DDC+ART group. The indicators of liver injury in serum of mice were detected. HE staining and Sirius red staining were performed, and histopathological scores were obtained according to Scheuer scoring system. RT-qPCR was used to detect the levels of α-SMA, Col1a1, Col1a2 and Tgfβ1 in mouse liver. Hydroxyproline (HYP) content was detected with HYP detection kit. LX2 cells were treated with TGF-β combined with ART, and the proteins related to PI3K/Akt/mTOR pathway were detected by Western blot assay. The expression of α-SMA, hepatic stellate cell (HSC) activation marker, was detected after ART and pathway inhibitors were used to treated with LX2 cells. Results ART significantly ameliorated DDC-induced cholestatic liver injury and liver fibrosis (P < 0.05). ART markedly decreased the levels of ALT, AST, ALP and TBA in serum of mice (P < 0.05). ART inhibited the mRNA expression of α-SMA, Col1a1, Col1a2 and Tgfβ1, and reduced the content of HYP in liver tissues of mice with cholestasis(P < 0.05). In addition, ART obviously inhibited TGF-β-induced activation of PI3K/Akt/mTOR pathway in LX2 cells (P < 0.05). PI3K/Akt/mTOR signaling pathway inhibitors enhanced the inhibitory effect of HSC activation induced by TGF-β(P < 0.05). Conclusion ART observably improves DDC-induced cholestatic liver fibrosis, and it can inhibit HSC activation by regulating PI3K/Akt/mTOR signaling pathway, thus exerting anti-fibrosis effect.

In recent years, enhancing the overall instructional quality in higher vocational colleges and universities through robust performance management has emerged as a pivotal concern in educational discourse. This study delves into the challenges inherent in the existing personnel performance appraisal systems within these institutions, employing both questionnaires and interviews for data collection. To address issues such as the vague delineation of appraisal objectives and the diffuse focus of evaluations, we propose the development of a Balanced Scorecard (BSC) model tailored for assessing faculty performance. This model integrates the four dimensions of finance, customer satisfaction, internal processes, and learning and growth, establishing causal linkages among them. The appropriateness of performance indicators for faculty evaluation is subsequently quantified using the fuzzy comprehensive evaluation method. Experimental validation reveals that the prevalent appraisal systems suffer from indistinct objectives and a lack of clear focus, with 41.1% of respondents indicating that the performance appraisal index system for counselors is suboptimal. Consequently, this research underscores the significant role that the construction of KPI-based performance appraisal models plays in refining personnel management practices within higher vocational colleges and universities.

To accurately describe operational electric vehicle (EV) drivers' irrational charging decision-making behaviors, a decision-making model for charging of operational EVs based on adapted cumulated prospect theory (ACPT) is established. Firstly, considering factors including arriving time, battery power remaining and psychological safety electricity, the charging demand models according to different mixes of peak and valley electricity tariffs is constructed. Then, a variable heterogeneous reference point model for the risk-preference-based ACTP is proposed with consideration of drivers' differences in psychological perception of arriving time and battery power remaining. Finally, the charging decision is evaluated based on the cumulative prospect values of the operational EV. Simulations are implemented and the results show that under the premise of a psychologically safe power level, owners with weaker psychologically perceived value will charge in advance instead of delaying charging. By combining risk preference and psychologically perceived value, the charging decision-making model proposed by this paper can explain irrational decision-making behaviours of operational EV drivers more accurately.

Engineered cementitious composites (ECC) exhibit behaviors of multiple cracking and strain-hardening compared to ordinary concrete. However, the use of ultrafine silica sand (S) to produce ECC results in increased cost and environmental overload. This study was to develop an eco-friendly and cheaper ECC by incorporating different replacement ratios of industrial byproduct molybdenum tailings (MT). The mechanical properties of ECC with MT were evaluated. Based on industrial computer technology, nuclear magnetic resonance, and scanning electron microscope techniques, the three-dimensional spatial distribution and fluid distribution of pores and the microstructure of the ECC were investigated, respectively. The mechanical properties of stirrup-confined ECC stub columns were also investigated. Results indicated that the ECC incorporating 25% MT exhibited the best mechanical properties, in which the tensile strength (f t) increased by 32.16% and exhibited well strain-hardening behaviors. Under a 25% MT replacement ratio, ECC had the lowest porosity and good interfacial transition zone, and bonding interface between polyvinyl alcohol (PVA) fiber (PF) and matrix. Moreover, the stirrup-confined ECC stub column with 25% MT can ensure better both the peak stress (f cc) and peak strain (ε cc). The assessment of environmental impact and cost further indicated that the incorporation of MT to prepare ECC is a promising method.

In the present study, AA7075/Al2O3 composites have been fabricated via liquid metallurgy process. AA7075 alloy and Al2O3 particles were taken as the base matrix and reinforcements, respectively. Then, contents of 3 and 6 wt. % of Al2O3 subdivisions were added into the AA7075 matrix. To improve wettability and distribution, reinforcement particles were pre-heated to a temperature of 550°C for each composite sample. A hardened EN32 steel disc as the counter face was used to evaluate the wear rate pin-on-disc. The results showed that the wear rate of the AA/Al2O3 composites was smaller than that of the monolithic AA7075 samples. Finally, the worn surfaces of samples were investigated by SEM.

Sedimentary characterization of modern floods helps to reconstruct the hydrologic flood process and spot the source area of sediment erosion. It is of great referential value both to the design of flood-control works and the identification of paleo-flood layers. In mid-August 2018, two typhoons battered the Dan River Basin in northern Shandong, leading to flooding disasters. Based on the research of the flooded area, the boreholeDH1 of 21 cm long was obtained from the downstream with well-preserved flood sediments. The analyses on its grain size, loss on ignition, magnetic susceptibility, and sporo-pollen features, show that the sedimentary cycle under flooding is characterized by fine grains in the lower part and coarse grains in the upper part. Modern soil is developed at a depth of 11.5~21.0 cm; a typical flood silt layer is developed at a depth of 0~9.0 cm, which is relatively coarse in grain size, dominated by medium-to-coarse-grained silt, and with a sand content of 14.7% on average; and a fine-grained sedimentary layer is developed at a depth of 9.0~11.5 cm during the initial period of flooding. According to the various traits of sensitive component contents, the flooding process can be divided into two stages: the relatively weak hydrodynamic force at the earlier stage and the significantly accelerated flow velocity at the later stage; The former is characterized by severe vicious flow and deposition of clays under the combined action of natural conditions and human activities, which can be further divided into two sub-sections of the rising water level and the significant acceleration of flow velocity; The latter result in typical silty deposits from flooding. Carbonate, organic matter content, and sporo-pollen abundance all are negatively correlated with grain size, indicating that the weak hydrodynamic environment is conducive to its deposition and accumulation. The sporo-pollen assemblage of the surface soil can better indicate the distribution of vegetation in the study area, and the sporo-pollen assemblage of the silty layer from flooding can better reflect the overall condition of vegetation in the basin, which has revealed that the river flood is much greater than wind in transporting sporo-pollen. In addition, the sporo-pollen assemblage features in the clayey layer from flooding are in good agreement with the distribution of vegetation in the study area, and obviously different from those of the silty layer from flooding, according to which it is speculated that the primary cause of the water level rise during the initial period of flooding lies in precipitation incorporating with local surface runoff; therefore, the sediment and sporo-pollen are derived from within the study area, and meanwhile the influx of tourists in the later period has brought in more sporo-pollen from mountainous vegetation. The sporo-pollen deposition records from borehole DH1 show that the sporo-pollen assemblage features are in good response to the propagation process and hydrodynamic force, and have the potential to identify the source area of sediment erosion. The magnetic susceptibility mainly reflects the intensity of pedogenesis, and its value for both the clayey and silty layers from flooding is steadily low, significantly lower than that for the soil contact layer. Therefore, it can be used as a reliable indicator for identifying flood deposits, but the geospatial scope of their use needs to be further discussed.

Objective To investigate the underlying molecular mechanism by which melatonin suppresses bile acids (BAs)-initiated expression of chemokine C-X-C motif chemokine ligand 2 (Cxcl2). Methods Mice were fed a 1% cholic acid (CA)-supplemented diet for 2 weeks to establish a model of cholestatic disease, and were given an intraperitoneal injection of melatonin (100 mg/kg) once a day for 2 weeks. Then, the liver tissues were collected for HE staining, and the liver histology was scored using the Scheuer scoring system. Effects of melatonin on the expression of Cxcl2, Cxcr2, myeloperoxidase (Mpo) and c-Jun in the liver tissues at mRNA and protein levels were detected by real-time qPCR and Western blotting. The expression of c-Jun in the liver tissues was measured by immunohistochemical assay. Primary mouse hepatocytes were isolated and cultured, and then treated with taurine-conjugated bile acid, taurocholic acid (TCA) and melatonin in order to explore the mechanism of melatonin reducing BAs-initiated Cxcl2 expression. Furthermore, these primarily cultured cells were treated with anisomycin (JNK agonist) and melatonin to further detect the expression of Cxcl2. Results Melatonin treatment significantly ameliorated cholestasis-induced inflammatory liver injury (P < 0.05), and reduced the elevated expression of Cxcl2, Cxcr2, Mpo(a biomarker of neutrophils) and the transcription factor c-Jun in the livers of cholestatic disease mice (P

【Objective】 The ratio of transpiration (T) to evapotranspiration (ET) varies with plants and their growth stages. The aim of this paper is to experimentally study T and ET, as well as their temporal variations. 【Method】 The experiment was conducted at a grapevine vineyard in an arid oasis in northwestern China. The transpiration and evapotranspiration of the grapevines (Vitis vinifera L.) were measured using sap-flow sensor, microclimatic station and eddy-covariance methods, respectively, at different growth stages. 【Result】 During the whole growth season, the transpiration increased from the initial 0.20 mm/d to 8.13 mm/d at the middle of growth season; it then dropped gradually to 3.32 mm/d when the leaves started falling. In comparison, the associated evapotranspiration increased from initial 0.44 mm/d to 9.97 mm/d before gradually decreasing to 4.91 mm/d. The average T/ET ratio was approximately 63.5%. 【Conclusion】 Transpiration is the dominant soil water loss during the growth season of the grapevines (Vitis vinifera L.) in arid oasis in northwestern China.

Beijing Administration of Traditional Chinese Medicine had set up “Special Funding Program Supporting the development of Traditional Chinese Medicine-Improving the quality and efficacy of Traditional Chinese Medicine”, and two trainer studios for the training of Traditional Chinese Medicine nursing techniques were established: The Trainer Studio for Traditional Chinese Medicine Five-element Music therapy located in Xiyuan Hospital of China Academy of Chinese Medical Sciences, and Yanjing Trainer Studio for Traditional Chinese Medicine External Application located in Beijing Hospital of Traditional Chinese Medicine Affiliated to Capital Medical University. The establishment of two trainer studios has achieved outstanding results in the following aspects: innovation on training model of Traditional Chinese Medicine nursing techniques, enriched cultivating mode for Traditional Chinese Medicine nursing staff, standardization and promotion of Traditional Chinese Medicine, establishment of academic platform for Traditional Chinese Medicine. This paper summarized the nursing experience of holographic meridian scraping in a patient with refractory hypertension. A comprehensive evaluation based on Traditional Chinese Medicine diagnosis of face, hand and tongue inspection and scraping should be carried out before implementation of holographic meridian scraping therapy. Therapeutic principles and scraping area should be determined before nursing management based on syndrome differentiation, and a standard scraping procedure is required to control blood pressure and improve the symptoms. [Objectives] In order to realize lightweight design of hull structure, the steel sandwich structure is used to replace the traditional stiffened structure to reduce the weight of hull structure on the premise of guaranteeing the load-bearing capacity of the original structure.[Methods] In view of the load-bearing capacity of type V and type I steel corrugated sandwich panels under uniaxial compression, the buckling analysis and nonlinear finite element analysis were carried out by using ABAQUS,and the load-bearing capacity and failure modes of steel sandwich panels were compared with those of traditional stiffened panels.[Results] The load-bearing capacity and failure modes of steel sandwich panels were obtained. The results show that the buckling failure modes of steel sandwich panels are different because of different structure types. Under the same weight,the designed type I steel corrugated sandwich panel has the highest bearing capacity,followed by stiffened panel and type V steel corrugated sandwich panel.[Conclusions] The lightweight efficiency is different for different structural types,the lightweight law of the steel corrugated sandwich panel can provide reference for the selection and application of similar panels in hull structure design.

[Purpose/significance] Based on the review of half-life indexes in periodical evaluation at home and abroad, this paper finds that half-life indexes are mixed and misused in the process of being used. so, we analyze and explore these problems. [Method/process] Using the core journals’ index data of library and information science and economics as a sample, which come from the Statistical Database of International and Domestic Influences in China Academic Journals, we discussed the application rationality of half-life indexes in periodical evaluation though the visualization tools, correlation analysis and other statistical methods. [Results/conclusion] It has been found that half-life has some limitations in periodical evaluation. There is a great difference between historical and contemporary methods in reflecting the aging rate of literature. There are also some misunderstandings about the use of half-life indexes, According to these situations, this paper proposes some recommendations for half-life use.

BACKGROUND: Disruptions in vascular formation attributable to chemical insults is a pivotal risk factor or potential etiology of developmental defects and various disease settings. Among the thousands of chemicals threatening human health, the highly concerning groups prevalent in the environment and detected in biological monitoring in the general population ought to be prioritized because of their high exposure risks. However, the impacts of a large number of environmental chemicals on vasculature are far from understood. The angioarchitecture complexity and technical limitations make it challenging to analyze the entire vasculature efficiently and identify subtle changes through a high-throughput in vivo assay. OBJECTIVES: We aimed to develop an automated morphometric approach for the vascular profile and assess the vascular morphology of healthconcerning environmental chemicals. METHODS: High-resolution images of the entire vasculature in Tg(fli1a:eGFP) zebrafish were collected using a high-content imaging platform. We established a deep learning-based quantitative framework, ECA-ResXUnet, combined with MATLAB to segment the vascular networks and extract features. Vessel scores based on the rates of morphological changes were calculated to rank vascular toxicity. Potential biomarkers were identified by vessel--endothelium-gene--disease integrative analysis. RESULTS: Whole-trunk blood vessels and the cerebral vasculature in larvae exposed to 150 representative chemicals were automatically segmented as comparable to human-level accuracy, with sensitivity and specificity of 95.56% and 95.81%, respectively. Chemical treatments led to heterogeneous vascular patterns manifested by 31 architecture indexes, and the common cardinal vein (CCV) was the most affected vessel. The antipsychotic medicine haloperidol, flame retardant 2,2-bis(chloromethyl)trimethylenebis[bis(2-chloroethyl) phosphate], and tert-butylphenyl diphenyl phosphate ranked as the top three in vessel scores. Pesticides accounted for the largest group, with a vessel score of [greater than or equal to]1, characterized by a remarkable inhibition of subintestinal venous plexus and delayed development of CCV. Multiple-concentration evaluation of nine per- and polyfluoroalkyl substances (PFAS) indicated a low-concentration effect on vascular impairment and a positive association between carbon chain length and benchmark concentration. Target vessel-directed single-cell RNA sequencing of fli1a (+) cells from larvae treated with k-cyhalothrin, perfluorohexanesulfonic acid, or benzylbutyl phthalate, along with vessel--endothelium-gene--disease integrative analysis, uncovered potential associations with vascular disorders and identified biomarker candidates. DISCUSSION: This study provides a novel paradigm for phenotype-driven screenings of vascular-disrupting chemicals by converging morphological and transcriptomic profiles at a high-resolution level, serving as a powerful tool for large-scale toxicity tests. Mesenchymal stem cells (MSCs) are novel therapeutics for the treatment of Crohn's disease. However, their mechanism of action is unclear, especially in disease-relevant chronic models of inflammation. Thus, we used SAMP-1/YitFc (SAMP), a chronic and spontaneous murine model of small intestinal inflammation, to study the therapeutic effects and mechanism of action of human bone marrow-derived MSCs (hMSC). hMSC dose-dependently inhibited naïve T lymphocyte proliferation via prostaglandin E2 (PGE2) secretion and reprogrammed macrophages to an anti-inflammatory phenotype. We found that the hMSCs promoted mucosal healing and immunologic response early after administration in SAMP when live hMSCs are present (until day 9) and resulted in a complete response characterized by mucosal, histological, immunologic, and radiological healing by day 28 when no live hMSCs are present. hMSCs mediate their effect via modulation of T cells and macrophages in the mesentery and mesenteric lymph nodes (mLN). Sc-RNAseq confirmed the anti-inflammatory phenotype of macrophages and identified macrophage efferocytosis of apoptotic hMSCs as a mechanism that explains their long-term efficacy. Taken together, our findings show that hMSCs result in healing and tissue regeneration in a chronic model of small intestinal inflammation and despite being short-lived, exert long-term effects via sustained anti-inflammatory programming of macrophages via efferocytosis.
