Abstract The multifaceted nature of sensor data has long been a hurdle for those seeking to harness its full potential in the field of 3D object detection. Although the utilisation of point clouds as input has yielded exceptional results, the challenge of effectively combining the complementary properties of multi‐sensor data looms large. This work presents a new approach to multi‐model 3D object detection, called adaptive voxel‐image feature fusion (AVIFF). Adaptive voxel‐image feature fusion is an end‐to‐end single‐shot framework that can dynamically and adaptively fuse point cloud and image features, resulting in a more comprehensive and integrated analysis of the camera sensor and the LiDar sensor data. With the aid of the adaptive feature fusion module, spatialised image features can be adroitly fused with voxel‐based point cloud features, while the Dense Fusion module ensures the preservation of the distinctive characteristics of 3D point cloud data through the use of a heterogeneous architecture. Notably, the authors’ framework features a novel generalised intersection over union loss function that enhances the perceptibility of object localsation and rotation in 3D space. Comprehensive experimentation has validated the efficacy of the authors’ proposed modules, firmly establishing AVIFF as a novel framework in the field of 3D object detection.

Abstract Backgrounds To our knowledge, there is no available nationwide data on omicron symptom patterns in China mainland. We aim to determine the acute and long COVID-19 symptoms in the omicron-dominant period and to evaluate its association with risk factors. Methods We designed a cross-sectional nationwide study and data about self-reported symptoms were collected by an online platform named Wenjuanxing. Eligible participants were aged 25–65 years and were symptomatic. In this study, the ratios of the number of people of different ages and genders were weighted by the data from the Seventh National Census (2020 years), and validated by a published nationwide representative study through comparing smoking rates. Descriptive indicators were calculated for demographic characteristics, diagnosis ways, and duration time, acute symptoms, hospitalization, severity and long COVID-19 symptoms. And, the associations between risk factors and acute and long COVID-19 symptoms were analyzed by multivariable logistic regression models. Results A total of 32,528 individuals diagnosed as COVID-19 infection from October 1, 2022 to February 21, 2023 were included. The first three acute symptoms of COVID-19 infection were fever (69.90%), headache (62.63%), and sore throat (54.29%), respectively. The hospitalization rate within 7 days was 3.07% and symptoms disappearance rate within 21 days was 68.84%, respectively. Among 3983 COVID-19 patients with 3 months or more time difference between first infection and participation into the study, the long COVID-19 rate was 19.68% and the primary symptoms were muscle weakness (19.39%), headache (17.98%) and smell/taste disorder (15.18%). Age groups, smoking, marriage status and vaccination were risk factors for numbers of acute phase symptoms and long COVID-19 symptoms. Lastly, female and current smokers also showed more numbers of symptoms during acute infection period. Conclusions In Chinese mainland, our respondent indicated that current smokers and women were associated with acute COVID-19 symptoms, which should be treated with caution due to the lack of representative.

Objective: To investigate the effectiveness of the original oblique conformal anastomosis presented in this research in reducing the incidence of cervical anastomotic leak after performing totally minimally invasive esophagectomy (TMIE). Methods: The esophagus and stomach of 27 fresh pigs, termed the esophagogastric model, were used to simulate human esophagogastric organs for this study's in vitro experimental objectives. Nine esophagogastric models of similar weight were divided into three groups. Esophagogastrostomy with circular-stapled end-to-side anastomosis was performed. A tension gauge was used to pull the anastomosis, and the tension at which anastomotic leakage occurred was recorded. Furthermore, a retrospective assessment of 539 patients who underwent TMIE was conducted to analyze the influencing factors of cervical anastomotic leakage. Abstract Objective The purpose of this study is to compare radiological and clinical outcomes between alternate levels (C4 and C6) and all levels mini-plate fixation in C3-6 unilateral open-door laminoplasty. Methods Ninety-six patients who underwent C3-6 unilateral open-door laminoplasty with alternate levels mini-plate fixation (54 patients in group A) or all levels mini-plate fixation (42 patients in group B) between September 2014 and September 2019 were reviewed in this study. Radiologic and clinical outcomes were assessed. Clinical results included Visual Analogue Scale (VAS) of axial neck pain and Japanese Orthopedic Association (JOA) score. Radiographic results included cervical range of motion (ROM), cervical curvature index (CCI), and the spinal canal expansive parameters including open angle, anteroposterior diameter (APD), and Pavlov`s ratio. Results There was no significant difference in VAS, JOA score, ROM, and CCI between two groups. There was no significant difference in canal expansion postoperatively between two groups. However, open angle, APD, and Pavlov`s ratio in group A decreased significantly during the follow-up. In group B, APD, Pavlov`s ratio, and open angle were maintained until the final follow-up. There was no hardware failure or lamina reclosure occurred in both groups during the follow-up. The mean cost of group B was higher than that of group A. Conclusions Despite the differences in the maintenance of canal expansion, alternate levels mini-plate fixation can achieve similar clinical outcomes as all levels mini-plate fixation in C3-6 unilateral open-door laminoplasty. As evidenced in this study, we believe C3-6 laminoplasty with alternate levels (C4 and C6) mini-plate fixation is an economical, effective, and safe treatment method.

Developing highly active oxygen evolution reaction (OER) electrocatalysts with robust durability is essential in producing high-purity hydrogen through water electrolysis. Layered double hydroxide (LDH) based catalysts have demonstrated efficient catalytic performance toward the relatively sluggish OER. By considering the promotion effect of phosphate (Pi) on proton transfer, herein, a facile phosphate acid (PA) surface-neutralization strategy is developed to in-situ construct NiCo-LDH/NiCoPi hetero-sheets toward OER catalysis. OER activity of NiCo-LDH is significantly boosted due to the proton promotion effect and the electronic modulation effect of NiCoPi. As a result, the facilely prepared NiCo-LDH/NiCoPi catalyst displays superior OER catalytic activity with a low overpotential of 300 mV to deliver 100 mA cm−2 OER and a Tafel slope of 73 mV dec−1. Furthermore, no visible activity decay is detected after a 200-h continuous OER operation. The present work, therefore, provides a promising strategy to exploit robust OER electrocatalysts for commercial water electrolysers.

Abstract Invasive candidiasis (IC) is a notable healthcare-associated fungal infection, characterized by high morbidity, mortality, and substantial treatment costs. Candida albicans emerges as a principal pathogen in this context. Recent academic advancements have shed light on the critical role of exosomes in key biological processes, such as immune responses and antigen presentation. This burgeoning body of research underscores the potential of exosomes in the realm of medical diagnostics and therapeutics, particularly in relation to fungal infections like IC. The exploration of exosomal functions in the pathophysiology of IC not only enhances our understanding of the disease but also opens new avenues for innovative therapeutic interventions. In this investigation, we focus on exosomes (Exos) secreted by macrophages, both uninfected and those infected with C. albicans. Our objective is to extract and analyze these exosomes, delving into the nuances of their protein compositions and subgroups. To achieve this, we employ an innovative technique known as Proximity Barcoding Assay (PBA). This methodology is pivotal in our quest to identify novel biological targets, which could significantly enhance the diagnostic and therapeutic approaches for C. albicans infection. The comparative analysis of exosomal contents from these two distinct cellular states promises to yield insightful data, potentially leading to breakthroughs in understanding and treating this invasive fungal infection. In our study, we analyzed differentially expressed proteins in exosomes from macrophages and C. albicans -infected macrophages, focusing on proteins such as ACE2, CD36, CAV1, LAMP2, CD27, and MPO. We also examined exosome subpopulations, finding a dominant expression of MPO in the most prevalent subgroup, and a distinct expression of CD36 in cluster14. These findings are crucial for understanding the host response to C. albicans and may inform targeted diagnostic and therapeutic approaches. Our study leads us to infer that MPO and CD36 proteins may play roles in the immune escape mechanisms of C. albicans. Additionally, the CD36 exosome subpopulations, identified through our analysis, could serve as potential biomarkers and therapeutic targets for C. albicans infection. This insight opens new avenues for understanding the infection's pathology and developing targeted treatments.

Abstract A novel method based on machine learning is proposed to estimate the electromagnetic radiation level at the ground plane near fifth‐generation (5G) base stations. The machine learning model was trained using data from various 5G base stations, enabling it to estimate the electric field intensity at any arbitrary radiation point when the base station provides service to different numbers of 5G terminals which are in different service modes. The inputs required for the model include the transmit power of the antenna, the antenna gain, the distance between the 5G base station and 5G terminals, terminal service modes, the number of 5G terminals and the environmental complexity around the 5G base station. Experimental results demonstrate the feasibility and effectiveness of the estimation method, with the mean absolute percentage error of the machine learning model being approximately 5.98%. This level of accuracy showcases the reliability of the approach. Moreover, the proposed method offers low costs when compared with on‐site measurements. The estimated results can be utilised to reduce test costs and provide valuable guidance for optimal site selection, thereby facilitating radio wave coverage or electromagnetic radiation regulation of 5G base stations.

Objectives Multi‐planar external fixation has been used for the management of segmental tibial fractures with severe soft tissue injuries. However, fewer specialized studies have been reported. The primary aim of this study was to describe our experience of treating fractures of this type using the Taylor Spatial Frame and Ilizarov external fixation methods. Methods We retrospectively analyzed 33 patients with segmental tibial fracture treated at our institution between January 2016 and December 2020. The patients were divided into double Taylor Spatial Frame (D‐TSF) and Ilizarov groups based on the external fixation structure. Baseline demographic data included sex, age, injury side and cause, open or closed fracture, time from injury to surgery, complications, and external frame removal and fracture healing time. The hip–knee–ankle angle (HKA) was measured from preoperative, immediate postoperative, and final follow‐up full‐length X‐rays of bilateral lower limbs. We determined the degree of deviation in the HKA by calculating the difference between the measured angle and the ideal value of 180°; the absolute value was used to assess recovery of the lower limb force line. At the final follow‐up, Johner–Wruhs tibial fracture outcome criteria (J‐W TFOC) were used to classify the postoperative function of the affected limb as excellent, good, moderate, or poor. Count data were analyzed with the chi‐square test or Fisher's exact test; the Mann–Whitney U test was used for rank data. Results No statistically significant differences were observed between the two groups in terms of sex, age, side of injury, cause of injury, closed or open fracture, or time between injury and surgery, which indicates that the groups were comparable (p > 0.05). A statistically significant difference was observed in external frame removal and fracture healing time between the D‐TSF and Ilizarov groups (36.24 ± 8.34 vs 45.42 ± 10.21 weeks, p = 0.009; 33.33 ± 8.21 vs 42.00 ± 9.78 weeks, p = 0.011). The Johner–Wruhs criteria were used to assess the function of the affected limb, the D‐TSF group performed better in correcting the lower limb force line than the Ilizarov group. A statistically significant difference in terms of excellent ratings was observed between the two groups (18/2/1/0 vs 5/5/1/1, p = 0.010). Postoperative follow‐up X‐rays demonstrated a significant improvement in the HKA in both groups immediately after surgery and at the final follow‐up compared to the angle before surgery. At the final follow‐up, a statistically significant difference was observed in the degree of deviation in the HKA between the two groups (1.58° ± 0.84° vs 2.37° ± 1.00°, p = 0.023). Conclusion The D‐TSF treatment is associated with minimal secondary damage to soft tissue, a straightforward and minimally invasive procedure, multiplanar stable fracture fixation, and optimization of fracture alignment and lower limb force lines, therefore, it is highly effective therapeutic option for segmental tibial fracture.

Abstract Ideal hydrogel fibers with high toughness and environmental tolerance are indispensable for their long-term application in flexible electronics as actuating and sensing elements. However, current hydrogel fibers exhibit poor mechanical properties and environmental instability due to their intrinsically weak molecular (chain) interactions. Inspired by the multilevel adjustment of spider silk network structure by ions, bionic hydrogel fibers with elaborated ionic crosslinking and crystalline domains are constructed. Bionic hydrogel fibers show a toughness of 162.25 ± 21.99 megajoules per cubic meter, comparable to that of spider silks. The demonstrated bionic structural engineering strategy can be generalized to other polymers and inorganic salts for fabricating hydrogel fibers with broadly tunable mechanical properties. In addition, the introduction of inorganic salt/glycerol/water ternary solvent during constructing bionic structures endows hydrogel fibers with anti-freezing, water retention, and self-regeneration properties. This work provides ideas to fabricate hydrogel fibers with high mechanical properties and stability for flexible electronics.

Abstract Zinc oxide nanoparticles (ZnO NPs) stand as among the most significant metal oxide nanoparticles in trigger the formation of reactive oxygen species (ROS) and induce apoptosis. Nevertheless, the utilization of ZnO NPs has been limited by the shallowness of short-wavelength light and the constrained production of ROS. To overcome these limitations, a strategy involves achieving a red shift towards the near-infrared (NIR) light spectrum, promoting the separation and restraining the recombination of electron-hole (e−-h+) pairs. Herein, the hybrid plasmonic system Au@ZnO (AZ) with graphene quantum dots (GQDs) doping (AZG) nano heterostructures is rationally designed for optimal NIR-driven cancer treatment. Significantly, a multifold increase in ROS generation can be achieved through the following creative initiatives: (i) plasmonic Au nanorods expands the photocatalytic capabilities of AZG into the NIR domain, offering a foundation for NIR-induced ROS generation for clinical utilization; (ii) elaborate design of mesoporous core-shell AZ structures facilitates the redistribution of electron-hole pairs; (iii) the incorporation GQDs in mesoporous structure could efficiently restrain the recombination of the e−-h+ pairs; (iv) Modification of hyaluronic acid (HA) can enhance CD44 receptor mediated targeted triple-negative breast cancer (TNBC). In addition, the introduced Au NRs present as catalysts for enhancing photothermal therapy (PTT), effectively inducing apoptosis in tumor cells. The resulting HA-modified AZG (AZGH) exhibits efficient hot electron injection and e−-h+ separation, affording unparalleled convenience for ROS production and enabling NIR-induced PDT for the cancer treanment. As a result, our well-designed mesoporous core-shell AZGH hybrid as photosensitizers can exhibit excellent PDT efficacy.

Abstract As a byproduct of manufacturing soybeans, okara is high in dietary fiber, protein, and fats, and it contains all of the essential amino acids. Wheat, the primary ingredient in noodles, will lose nutrients during manufacturing, creating an imbalance in nutrients. This experiment is for the purpose of studying the effects of okara on quality, antioxidant properties, amino acid content, resistant starch (RS) content, and microstructure of noodles. The results indicate that the addition of 9% okara noodles increased hardness and adhesiveness by 107.19% and 132.14%, respectively, and improved ABTS free radical scavenging activity by 60.78%. The addition of 12% okara noodles increased the DPPH free radical scavenging ability by 23.66%, reduced the rapidly digestible starch (RDS) content of the noodles to 21.21%, and the resistant starch content increased to 44.85% (p

Abstract Background There is still limited research on the prognostic value of Presepsin as a biomarker for predicting the outcome of COVID-19 patients. Additionally, research on the combined predictive value of Presepsin with clinical scoring systems and inflammation markers for disease prognosis is lacking. Methods A total of 226 COVID-19 patients admitted to Beijing Youan Hospital’s emergency department from May to November 2022 were screened. Demographic information, laboratory measurements, and blood samples for Presepsin levels were collected upon admission. The predictive value of Presepsin, clinical scoring systems, and inflammation markers for 28-day mortality was analyzed. Results A total of 190 patients were analyzed, 83 (43.7%) were mild, 61 (32.1%) were moderate, and 46 (24.2%) were severe/critically ill. 23 (12.1%) patients died within 28 days. Identification of the S genotype of Malus plants will greatly promote the discovery of new genes, the cultivation and production of apple, the breeding of new varieties, and the origin and evolution of self-incompatibility in Malus plants. In this experiment, 88 Malus germplasm resources, such as Aihuahong, Xishuhaitang, and Reguanzi, were used as materials. Seven gene-specific primer combinations were used in the genotype identification. PCR amplification using leaf DNA produced a single S-RNase gene fragment in all materials. The results revealed that 70 of the identified materials obtained a complete S-RNase genotype, while only one S-RNase gene was found in 18 of them. Through homology comparison and analysis, 13 S-RNase genotypes were obtained: S1S2 (Aihuahong, etc.), S1S28 (Xixian Haitang, etc.), S1S51 (Hebei Pingdinghaitang), S1S3 (Xiangyangcun Daguo, etc.), S2S3 (Zhaiyehaitang, etc.), S3S51 (Xishan 1), S3S28 (Huangselihaerde, etc.), S2S28 (Honghaitang, etc.), S4S28 (Bo 11), S7S28 (Jiuquan Shaguo), S10Se (Dongchengguan 13), S10S21 (Dongxiangjiao) and SeS51 (Xiongyue Haitang). Simultaneously, the frequency of the S gene in the tested materials was analyzed. The findings revealed that different S genes had varying frequencies in Malus resources, as well as varying frequencies between intraspecific and interspecific. S3 had the highest frequency of 68.18%, followed by S1 (42.04%). In addition, the phylogenetic tree and origin evolution analysis revealed that the S gene differentiation was completed prior to the formation of various apple species, that cultivated species also evolved new S genes, and that the S50 gene is the oldest S allele in Malus plants. The S1, S29, and S33 genes in apple-cultivated species, on the other hand, may have originated in M. sieversii, M. hupehensis, and M. kansuensis, respectively. In addition to M. sieversii, M. kansuensis and M. sikkimensis may have also played a role in the origin and evolution of some Chinese apples.

BackgroundThe real-time prognostic data of patients with poorly differentiated thyroid carcinoma (PDTC) after surviving for several years was unclear. This study aimed to employ a novel method to dynamically estimate survival for PDTC patients.MethodsA total of 913 patients diagnosed with PDTC between 2014 and 2015 from the Surveillance, Epidemiology, and End Results (SEER) database, was recruited in our study. Kaplan–Meier method was used to estimate the overall survival (OS). The conditional survival (CS) outcomes of PDTC were analyzed and CS rates were calculated using the formula CS(y/x) = OS(y+x)/OS(x), whereby CS(y/x) denotes the probability of a patient enduring an additional y years subsequent to surviving x years following the diagnosis of PDTC. The least absolute shrinkage and selection operator (LASSO) regression was employed to identify prognostic predicters and multivariate Cox regression was utilized to develop a CS-nomogram. Finally, the performance of this model was evaluated and validated.ResultsKaplan–Meier survival analysis unveiled patient outcomes demonstrating an OS rate of 83%, 75%, and 60% respectively at the end of 3, 5, and 10 years. The novel CS analysis highlighted a progressive enhancement in survival over time, with the 10-year cumulative survival rate progressively augmenting from its initiation of 60% to 66%, 69%, 73%, 77%, 81%, 83%, 88%, 93%, and finally 97% (after surviving for 1-9 years, respectively) each year. And then 11 (11/15) predictors including age at diagnosis, sex, histology type, SEER stage, T stage, N stage, M stage, tumor size, coexistence with other malignancy, radiotherapy and marital status, were selected by LASSO analysis under the condition of lambda.min. Multivariate Cox regression analysis further highlighted the significant impact of all these predictors on the OS of PDTC and we successfully established and validated a novel CS-nomogram for real-time and dynamic survival prediction.ConclusionsThis was the first study to analyze the CS pattern and demonstrate a gradual improvement in CS over time in long-term PDTC survivors. We then successfully developed and validated a novel CS-nomogram for individualized, dynamic, and real-time survival forecasting, empowering clinicians to adapt and refine the patient-tailored treatment strategy promptly with consideration of evolving risks.

Abstract The evolutionary patterns of the mitochondrial genome are influenced by both adaptive and nonadaptive forces, with their contributions varying among taxa. There appears to be a correlation linking mutagenesis and latitude, which could be due to differences in metabolic rates. These discrepancies in metabolic rates exhibit a positive connection with mutation pressure. On this basis, we hypothesise that nonadaptive forces play a role in the differences in mutation rates observed along latitudinal gradients. In this study, we selected widely distributed carnivores as representatives of mammals to test our hypothesis. We examined the correlations between the dN/dS ratio (ω), as well as the substitution rates (dS and dN), of 13 PCGs in the mtDNA of 122 carnivores, and the latitude and climatic factors. We found that taxa distributed in higher latitudes tend to have higher substitution rates, but not ω values indicating selective pressure. Notably, dN shows a strong positive correlation with dS, although dS is primarily influenced by mutation pressure, while dN is also influenced by effective population size (Ne). Phylogenetic generalised least squares (PGLS) regression analyses showed that both substitution rates were correlated with climatic factors representing the temperature, precipitation and variability of climate. Based on our findings, we propose that the mutations are primarily influenced by nonadaptive forces (mutation pressure). This forms the fundamental premise for natural selection and speciation. Moreover, the correlation between substitution rates and latitudinal distribution and climate, which are outcomes of nonadaptive factors, can aid in comprehending the global distribution of species diversity.

Effectively utilizing large-scale Private Electric Vehicles (PREVs) in load restoration can enhance the capability of Distribution System Operators (DSOs) to respond to accidental power outages. However, the challenge remains in incentivizing and guiding PREVs to participate in load restoration. This paper proposes a Stackelberg game-based incentive mechanism, with a V2G Station Operator (VSO) discharge guidance model based on the Huff attraction model embedded to address this challenge. The VSO is introduced to manage the issue of PREVs having small individual capacities, and not meeting the criteria for direct participation in electricity markets. Firstly, a framework for involving PREVs in load restoration is introduced, clarifying the responsibilities of PREVs, VSO, DSO, and Distribution Market Operator (DMO), as well as the interrelation of incentive mechanism and discharge guidance strategy. Secondly, models for each game participant are established. The VSO encapsulates the discharge guidance strategy into an optimization model using the Huff attraction-based model that captures the destination choice probabilities of PREVs, and the DSO develops a load restoration model. Thirdly, a Stackelberg game model is established with the DMO as the leader and VSO and DSO as followers. The existence and uniqueness of the proposed Stackelberg game’s equilibrium solution are proven, and a distributed iterative algorithm is employed for the solution. Finally, numerical simulations are performed with the IEEE 33-bus distribution system and a simplified road network to validate the proposed strategy’s effectiveness in guiding PREVs’ participation in load restoration, enhancing the restoration capability of the DSO, and improving the economic efficiency of each participant.

Deep Reinforcement Learning (DRL) is effective in solving complex, non-linear optimization problems, which is particularly relevant in energy management within Integrated Energy Systems (IESs). However, DRL approaches conventionally focus on single-objective policy learning, which is inadequate for the multi-objective optimization tasks commonly encountered in IESs energy management. To improve this, these approaches typically combine multi-objectives, such as operating cost objective and safety objective into a single reward function using scalarization techniques. This reduces the fidelity and interpretability of the objective space and limits its applicability to a wide range of IESs energy management. To address these challenges, this paper presents a novel framework called Multi-Agent and Multi-Objective DRL (MAMODRL). This framework combines value function decomposition and policy gradient methods to achieve a Pareto-optimal solution. The IESs energy management is initially formulated as a multi-objective Markov decision process. Then, an advanced MAMODRL architecture is developed, which includes objective value function networks to facilitate policy optimization. Finally, based on the definition of dominance, Pareto frontier is approximated of IESs energy management. A case study suggests that the proposed approach is effective in solving the Pareto frontier for IESs energy management. To ensure the safe operation of the system, safety threshold is set at the Pareto frontier forming a Pareto optimization with safety conditions. Compared to traditional DRL approaches, the proposed approach is more flexible, interpretable, and capable of making multi-dimensional decisions.

Background: Integrating immune checkpoint inhibitors with multi-target tyrosine kinase inhibitors presents an innovative and hopeful strategy in liver cancer treatment. Nonetheless, a degree of resistance to this treatment is noticeable in certain patients. Alternative splicing (AS) represents a common biological process that controls the variety of life functions via isoforms. Purpose: Investigating how gene AS affects the effectiveness of combined immunotherapy in treating hepatocellular carcinoma (HCC). Methods: Our retrospective examination focused on AS's effect on immune therapy effectiveness, utilizing accessible tissue sequencing and clinical records for HCC. For corroborating our results, we gathered samples of drug-resistant HCC tissue, nearby tissues, HCC tissue with high drug responsiveness, and healthy liver tissue from clinical studies. Results: The study revealed a link between the frequency of AS occurrences, the expression levels of programmed cell death 1 ligand 1, and the resistance to tumor medications. Our study detailed the AS occurrences in HCC, leading to the creation of a risk-assessment function and a predictive model using AS data. The results of our study revealed that the risk score effectively distinguished between various immune subtypes and the effectiveness of immune therapy. Additional examination of the chosen AS occurrences uncovered their effects on both the immune microenvironment and cellular immunity. Our investigation also delved into the regulatory framework of AS, uncovering the role of stringently controlled splicing factors in the emergence of tumors and the modulation of the body's immune response. Conclusions: Increased AS in HCC diminishes the efficacy of immunotherapy; conversely, more AS in peritumoral tissue elevates the likelihood of tumor immune evasion.

Abstract Accurately estimating large-area crop yields, especially for soybeans, is essential for addressing global food security challenges. This study introduces a deep learning framework that focuses on precise county-level soybean yield estimation in the United States. It utilizes a wide range of multi-variable remote sensing data. The model used in this study is a state-of-the-art CNN-BiGRU model, which is enhanced by the GOA and a novel attention mechanism (GCBA). This model excels in handling intricate time series and diverse remote sensing datasets. Compared to five leading machine learning and deep learning models, our GCBA model demonstrates superior performance, particularly in the 2019 and 2020 evaluations, achieving remarkable R2, RMSE, MAE and MAPE values. This sets a new benchmark in yield estimation accuracy. Importantly, the study highlights the significance of integrating multi-source remote sensing data. It reveals that synthesizing information from various sensors and incorporating photosynthesis-related parameters significantly enhances yield estimation precision. These advancements not only provide transformative insights for precision agricultural management but also establish a solid scientific foundation for informed decision-making in global agricultural production and food security.

Objective To investigate the experience in the clinical management of pregnancy and delivery after split liver transplantation (SLT). Methods Related clinical data were collected from a patient who became pregnant and delivered successfully after SLT in the Organ Transplantation Center of our hospital, and a literature review was also performed. Results The patient underwent allogeneic orthotopic SLT due to decompensated liver cirrhosis after autoimmune hepatitis and developed mild acute rejection after surgery, which was improved after the adjustment of immunosuppressive drugs. The patient became pregnant spontaneously at 2 years after surgery, and due to the medication history of mycophenolate sodium enteric-coated tablets, she decided to terminate pregnancy and withdraw from this drug. The patient became pregnant again at 3 years after surgery and expe-rienced fluctuations in graft function, and a series of treatments were given, such as adjustment of the dose of anti-rejection drugs, liver-protecting symptomatic treatment, and tocolytic therapy for threatened preterm labor. Finally under the guidance of liver transplantation, obstetrics, and neonatology, the patient gave birth to a healthy girl by cesarean section at 35 weeks of gestation. Conclusion Although SLT recipients may experience the situations such as fluctuations in graft function and preterm birth, they can achieve a satisfactory prognosis under multidisciplinary collaboration, scientific medication guidance, and close follow-up monitoring.

Abstract Venous thromboembolism, which includes deep venous thrombosis (DVT) and pulmonary embolism, is the third most common vascular disease in the world and seriously threatens the lives of patients. Currently, the effect of conventional treatments on DVT is limited. Endothelial progenitor cells (EPCs) play an important role in the resolution and recanalization of DVT, but an unfavorable microenvironment reduces EPC function. Non-coding RNAs, especially long non-coding RNAs and microRNAs, play a crucial role in improving the biological function of EPCs. Non-coding RNAs have become clinical biomarkers of diseases and are expected to serve as new targets for disease intervention. A theoretical and experimental basis for the development of new methods for preventing and treating DVT in the clinic will be provided by studies on the role and molecular mechanism of non-coding RNAs regulating EPC function in the occurrence and development of DVT. To summarize, the characteristics of venous thrombosis, the regulatory role of EPCs in venous thrombosis, and the effect of non-coding RNAs regulating EPCs on venous thrombosis are reviewed. This summary serves as a useful reference and theoretical basis for research into the diagnosis, prevention, treatment, and prognosis of venous thrombosis.

AbstractThe formulation of nationwide goals, strategic plans, and specifically zoned areas for typhoon hazard mitigation and prevention is paramount for addressing the threat in a spatially explicit manner. Utilizing historical rainfall and wind speed data associated with typhoon disasters in the Northwest Pacific and South China Sea from 1949 to 2017, we proposed a comprehensive typhoon hazard zoning delineated based on the assessment of typhoon disaster risks, considering hazard-causing factors, hazard-formative environments, and hazard-bearing bodies through an Analytic Hierarchy Process. High-risk areas were primarily located in the coastal regions of South and Southeast China. Medium-risk areas were situated in the Yangtze River Delta and towns adjacent to Bohai Bay. Low-risk areas were primarily located in the central region of Guangxi, the Guangdong-Hong Kong-Macao Greater Bay Area, the vicinity of Fuzhou, the majority of southern Jiangsu and northern Zhejiang, all of Shanghai, the northern and southern parts of the Shandong Peninsula, the area around Bohai Bay in Tianjin, a minor portion of Hebei, and the three northeastern provinces. Appropriate mitigation measures were proposed to address the threats posed by typhoons in these regions. This information is instrumental in providing reliable support for China in addressing the challenges posed by typhoons.

Background: DIP2B is related to cancer progression. This study investigated the roles and pathways of DIP2B in lung adenocarcinoma (LUAD). Methods: DIP2B expression and the relationship between survival time of cancer patients and DIP2B expression were analyzed. The relationship between DIP2B expression and survival time in LUAD patients was evaluated by a meta-analysis. Cox and survival analyses were used to evaluate the prognostic factors and construct a prognostic nomogram. The mechanisms and effects of DIP2B and the relationship between DIP2B expression and the immune microenvironment were investigated using bioinformatics, CCK-8, western blotting, and transwell experiments. Results: DIP2B was overexpressed in LUAD tissues. DIP2B overexpression was associated with shorter prognosis and was an unfavorable risk factor for prognosis in LUAD patients. DIP2B co-expressed genes were involved in cell division, DNA repair, cell cycle, and others. Inhibition of DIP2B expression could downregulate the proliferation, migration, and invasion of LUAD A549 and H1299 cells, which was related to the decrease in CCND1 and MMP2 protein expression. BRCA1 overexpression was associated with short prognosis, and the nomogram formed by DIP2B and BRCA1 was associated with a poor prognosis in LUAD patients. DIP2B expression correlated with immune cells (such as CD8 T cells, Tcm, and iDCs) and cell markers. Conclusion: DIP2B is a potential biomarker of poor prognosis and the immune microenvironment in LUAD. Inhibition of DIP2B expression downregulated cancer cell proliferation, migration, and invasion, which might be related to the decrease in CCND1 and MMP2 protein expression. DIP2B-related nomograms might be useful tools for predicting the prognosis of LUAD patients.

Este trabajo de investigación compara dos versiones completas de la traducción del chino al español de Las Analectas, una perteneciente a Chang Shiru, un académico chino, y la otra, a Joaquín Pérez Arroyo, un traductor español. A partir de un corpus de construcción propia se han elegido varios parámetros a nivel léxico, oracional y discursivo para desarrollar un estudio comparativo. Además, como estudio de caso, se han seleccionado el pronombre “yo”, la palabra clave “superior” y la partícula “que”. Según los resultados, en términos generales, Chang utiliza un inventario de vocabulario más rico y oraciones más simples en comparación con Pérez Arroyo, quien, a su vez, emplea un inventario menos variado y oraciones más complejas, y tiende a ser más explícito en su traducción. Sin embargo, la preferencia de los traductores hacia las técnicas de explicitación y simplificación varía según el público destinatario. Factores como las diferencias lingüísticas y culturales chino-español, las preferencias de los traductores, el propósito de la traducción y los antecedentes personales del traductor contribuyen posiblemente a configurar el estilo único de un traductor.

The utilization of antibiotics is prevalent among lactating mothers. Hence, the rapid determination of trace amounts of antibiotics in human milk is crucial for ensuring the healthy development of infants. In this study, we constructed a human milk system containing residual doxycycline (DXC) and/or tetracycline (TC). Machine learning models and clustering algorithms were applied to classify and predict deficient concentrations of single and mixed antibiotics via label-free SERS spectra. The experimental results demonstrate that the CNN model has a recognition accuracy of 98.85% across optimal hyperparameter combinations. Furthermore, we employed Independent Component Analysis (ICA) and the pseudo-Siamese Convolutional Neural Network (pSCNN) to quantify the ratios of individual antibiotics in mixed human milk samples. Integrating the SERS technique with machine learning algorithms shows significant potential for rapid discrimination and precise quantification of single and mixed antibiotics at deficient concentrations in human milk.

The first wall (FW) of a blanket or divertor faces high temperature plasma. Thus, the FW needs to withstand extremely high heat fluxes. Periodic plasma pulses bring some reliability challenges to FW structures. The FW is composed of tiles, heat sink and a bolt with different connection between the tile and the heat sink. In this study, the performances of two connection forms for the tile and the heat sink were investigated. The FLUENT software was used to evaluate the heat transfer performances for the two types structure. Elastic-plastic analysis was subsequently adopted to estimate the structural properties based on the thermal hydraulic results. At the same time, the thermal and structural results under a transient high thermal load (2 MW/m2) were also analyzed. Finally, structural fatigue analysis under a cyclic thermal load with 0.5 MW/m2 was also performed. The results indicated that there was a large discrepancy in the tile results for the two connections, while the differences in the heat sink result were not significant. The advantages and disadvantages of two connections also be discussed at last.

BackgroundTo explore the clinical characteristics, etiological factors, and clinical-related genetic variant of children with acute necrotizing encephalopathy (ANE) related to the Omicron BF.7.14 novel coronavirus.MethodsGenomic variations were detected through whole exome sequencing. Additionally, we summarized the clinical data to explore the inheritance patterns associated with novel coronavirus-related ANE.ResultsThis study included four patients (2 males and 2 females) with an average age of 2.78 ± 1.93 years. All the patients had prodromal symptoms of Omicron BF.7.14 virus infection, and exhibited symptoms such as altered consciousness, seizures and cognitive/language disturbances. Cranial MRI scans revealed damage to the thalamus, basal ganglia and brainstem. The cerebrospinal fluid (CSF) cell counts were nearly normal, but protein level in CSF increased significantly. Genetic analysis revealed a novel truncated variant of CRMP2 gene in one patient who suffered more severe coma score and prognosis and dead in the later stages. All children exhibited a decrease in the absolute count of T lymphocytes, helper T cells, suppressor T cells, and NK cells to varying degrees. Furthermore, levels of cytokines, including IL-1 β, IL-5, IL-6 and IL-8 were significantly elevated in the CSF, especially in patient with truncated variant of CRMP2 gene.ConclusionThe Omicron BF.7.14 type novel coronavirus can lead to ANE, characterized by T cell immunosuppression and a significant increase in cytokine levels in the CSF. The truncated variation of CRMP2 gene may affect the prognosis of ANE by affecting the migration of cerebral T cells.

It furnishes a detailed survey of leading knowledge institutions and critical focal points in this area, utilizing bibliometric analysis.MethodsVOSviewer and CiteSpace software platforms were employed to systematically analyze and graphically depict the relevant literature indexed in the WoSCC database over the preceding ten years.ResultsIn the interval between October 1, 2013, and October 1, 2023, a corpus of 795 publications was amassed. The primary research institutions involved in this study include Duke University, the University of Minnesota, and the University of Sydney. The leading trio of nations, in terms of publication volume, comprises the United States, China, and Brazil. Superconducting electrides are characterized by the emergence of electrons occupying the interstitial region of lattice and exhibit coexistence of distinctive electride state and superconductivity, which have attracted wide attention. However, the underlying crucial factors governing the superconductivity and influencing the structural stability by electride states remain elusive. Here, we performed structure searches in conjunction with first-principles calculations to identify a dynamically stable electride Be6C at 40 GPa, which exhibits p-orbital type electride states and demonstrates superconductivity with a Tc of 22.4 K. Furthermore, it remains dynamically stable even down to ambient conditions while maintaining a Tc of 18.9 K. Further analyses unveiled that the dual p-hybridized electrons within the orbitals of p-orbital electride states and Be-2p mainly composed of a van Hove singularity near the Fermi level and participate in electron–phonon interaction to form Cooper pairs leading to the high-Tc. The preservation of dynamic stability for Be6C at low pressure is primarily attributed to the presence of electride states that effectively form both covalent and ionic bonding properties to bind neighbor Be cations to lower enthalpy of system and subsequently, in turn, lowering the required pressure. Our findings not only explained the underlying factors affecting superconductivity but also revealed the crucial role of electride states in determining the dynamic stability of structure, providing valuable insights for subsequent research on superconducting electrides at low pressures.

Abstract Background Sutureless scleral fixed intraocular lens implantation (SF-IOL) has become one of the mainstream schemes in clinical treatment of aphakic eyes because of its advantages, such as avoiding dislocation of intraocular lens or subluxation caused by suture degradation or fracture and significant improvement of postoperative visual acuity. However, a consensus on the relative effectiveness and safety of this operation and other methods is still lacking. This study aimed to compare the efficacy and safety of sutureless SF-IOL with other methods. Aphakia means that the lens leaves the normal position and loses its original function, including absence or complete dislocation and subluxation of the lens which could cause anisometropic amblyopia, strabismus, and loss of binocular function in children and adolescents. For adults, the loss of the lens could lead to high hyperopia and affect vision. Above all this disease can seriously affect the quality of life of patients. Methods Literature about sutureless SF-IOL in PubMed, Cochrane Library, Embase, Web of Science, China National Knowledge Infrastructure, China Technical Journal VIP database, and Wanfang database published from 2000 to 2022 was reviewed. The weighted average difference was calculated by RevMan5.3 software for analysis. Two researchers independently selected the study and used the Cochrane collaboration tool to assess the risk of errors. Cochrane bias risk tool was used to evaluate the quality of evidence. This study is registered on PROSPERO (CRD42022363282). Results The postoperative IOL-related astigmatism of sutureless SF-IOL was lower than that of suture SF-IOL, and there was statistical difference when we compared the absolute postoperative spherical equivalent after sutureless SF-IOL and suture SF-IOL. Indicating that the degree of refractive error after sutureless SF-IOL was lower. Meanwhile, the operation time of sutureless SF-IOL was shorter than that of suture SF-IOL. The subgroup analysis showed that the absolute postoperative spherical equivalent and astigmatism values in Yamane technique were lower than those in suture SF-IOL. Conclusion Sutureless SF-IOL has the advantages of stable refraction, short operation time, and less postoperative complications. However, high-quality literature to compare these technologies is lacking. Some long-term follow-up longitudinal prospective studies are needed to confirm the findings.

Abstract Cities play a fundamental role in policy decision-making processes, necessitating the availability of city-level population projections to better understand future population dynamics and facilitate research across various domains, including urban planning, shrinking cities, GHG emission projections, GDP projections, disaster risk mitigation, and public health risk assessment. However, the current absence of city-level population projections for China is a significant gap in knowledge. Moreover, aggregating grid-level projections to the city level introduces substantial errors of approximately 30%, leading to discrepancies with actual population trends. The unique circumstances of China, characterized by comprehensive poverty reduction, compulsory education policies, and carbon neutrality goals, render scenarios like SSP4(Shared Socioeconomic Pathways) and SSP5 less applicable. To address the aforementioned limitations, this study made three key enhancements, which significantly refines and augments our previous investigation. Firstly, we refined the model, incorporating granular demographic data at the city level. Secondly, we redesigned the migration module to consider both regional and city-level population attractiveness. Lastly, we explored diverse fertility and migration scenarios.

Abstract Kidney fibrosis, characterized by the activation and expansion of the matrix-producing fibroblasts, is the common outcome of chronic kidney disease (CKD). While fibroblast proliferation is well studied in CKD, little is known about the regulation and mechanism of fibroblast depletion. Here, we show that exosomes derived from stressed/injured tubules play a pivotal role in dictating fibroblast apoptosis and fate. When human kidney tubular cells (HK-2) were stimulated with TGF-β1, they produced and released increased amounts of exosomes (TGFβ-Exo), which prevented renal interstitial fibroblasts from apoptosis. In vivo, injections of TGFβ-Exo promoted renal fibroblast survival, whereas blockade of exosome secretion accelerated fibroblast apoptosis in obstructive nephropathy. Proteomics profiling identified the tumor necrosis factor-α-induced protein 8 (TNFAIP8) as a key component enriched in TGFβ-Exo. TNFAIP8 was induced in renal tubular epithelium and enriched in the exosomes from fibrotic kidneys. Knockdown of TNFAIP8 in tubular cells abolished the ability of TGFβ-Exo to prevent fibroblast apoptosis. In vivo, gain- or loss- of TNFAIP8 prevented or aggravated renal fibroblast apoptosis after obstructive injury. Mechanistically, exosomal-TNFAIP8 promoted p53 ubiquitination leading to its degradation, thereby inhibiting fibroblasts apoptosis and inducing their proliferation. Collectively, these results indicate that tubule-derived exosomes play a critical role in controlling the size of fibroblast population during renal fibrogenesis through shuttling TNFAIP8 to block p53 signaling. Strategies to target exosomes may be effective strategies for the therapy of fibrotic CKD.

Soil nitrogen (N) mineralization typically governs the availability and movement of soil N. Understanding how factors, especially functional genes, affect N transformations is essential for the protection and restoration of forest ecosystems. To uncover the underlying mechanisms driving soil N mineralization, this study investigated the effects of edaphic environments, substrates, and soil microbial assemblages on net soil N mineralization in boreal forests. Field studies were conducted in five representative forests: Larix principis-rupprechtii forest (LF), Betula platyphylla forest (BF), mixed forest of Larix principis-rupprechtii and Betula platyphylla (MF), Picea asperata forest (SF), and Pinus sylvestris var. mongolica forest (MPF). Results showed that soil N mineralization rates (Rmin) differed significantly among forests, with the highest rate in BF (p < 0.05). Soil properties and microbial assemblages accounted for over 50% of the variability in N mineralization. This study indicated that soil environmental factors influenced N mineralization through their regulatory impact on microbial assemblages. Compared with microbial community assemblages (α-diversity, Shannon and Richness), functional genes assemblages were the most important indexes to regulate N mineralization. It was thus determined that microbial functional genes controlled N mineralization in boreal forests. This study clarified the mechanisms of N mineralization and provided a mechanistic understanding to enhance biogeochemical models for forecasting soil N availability, alongside aiding species diversity conservation and fragile ecosystem revitalization in boreal forests.

Due to the difficulty of accurately predicting system reliability for many engineering structures, bounds on system reliability have received increasing attention. By dealing with structural uncertain parameters with an ellipsoid model, a linear programming-based non-probabilistic reliability bounds method is proposed in this paper for series systems. In this research, a linear programming model is first established, and then several strategies are proposed to simplify the model by removing zero design variables. Three numerical examples are presented to demonstrate the feasibility and validity of the proposed method.

This study investigates lightweight and efficient candidates for sound absorption to address the growing demand for sustainable and eco-friendly materials in noise attenuation. Juncus effusus (JE) is a natural fiber known for its unique three-dimensional network, providing a viable and sustainable filler for enhanced sound absorption in honeycomb panels. Microperforated-panel (MPP) honeycomb absorbers incorporating JE fillers were fabricated and designed, focusing on optimizing the absorber designs by varying JE filler densities, geometrical arrangements, and MPP parameters. At optimal filling densities, the MPP-type honeycomb structures filled with JE fibers achieved high noise reduction coefficients (NRC) of 0.5 and 0.7 at 20 mm and 50 mm thicknesses, respectively. Using an analytical model and an artificial neural network (ANN) model, the sound absorption characteristics of these absorbers were successfully predicted. This study demonstrates the potential of JE fibers in improving noise mitigation strategies across different industries, offering more sustainable and efficient solutions for construction and transportation.

Background: One-third of intractable epilepsy patients have no visually identifiable focus for neurosurgery based on imaging tests [magnetic resonance imaging (MRI)-negative cases]. Stereo-electroencephalography-guided radio-frequency thermocoagulation (SEEG-guided RF-TC) is utilized in the clinical treatment of epilepsy to lower the incidence of complications post-open surgery. Objective: This study aimed to identify prognostic factors and long-term seizure outcomes in SEEG-guided RF-TC for patients with MRI-negative epilepsy. Design: This was a single-center retrospective cohort study. Methods: We included 30 patients who had undergone SEEG-guided RF-TC at Sanbo Brain Hospital, Capital Medical University, from April 2015 to December 2019. The probability of remaining seizure-free and the plotted survival curves were analyzed. Prognostic factors were analyzed using log-rank tests in univariate analysis and the Cox regression model in multivariate analysis. Results: With a mean time of 31.07 ± 2.64 months (median 30.00, interquartile range: 18.00–40.00 months), 11 out of 30 patients (36.7%) were classified as International League Against Epilepsy class 1 in the last follow-up. The mean time of remaining seizure-free was 21.33 ± 4.55 months [95% confidence interval (CI) 12.41–30.25], and the median time was 3.00 ± 0.54 months (95% CI 1.94–4.06). Despite falling in the initial year, the probability of remaining seizure-free gradually stabilizes in the subsequent years. The patients were more likely to obtain seizure freedom when the epileptogenic zone was located in the insular lobe or with one focus on the limbic system ( p = 0.034, hazard ratio 5.019, 95% CI 1.125–22.387). Conclusion: Our findings may be applied to guide individualized surgical interventions and help clinicians make better decisions.

There is a large amount of oil and gas loss in traditional conventional core samples. Revealing the rules of oil and gas loss is of great significance for restore the pristine oil content and oil component in the shale. In this study, four preserved shale cores with different thermal maturity (Ro = 1.01–1.53%) and different total organic carbon content (TOC = 1.69–5.48 wt.%) were selected. The samples are obtained from the first member of the Qingshankou Formation in the Gulong Sag. Nuclear magnetic resonance (NMR) T1–T2 mapping and thermal desorption gas chromatography (TD–GC, at a constant temperature of 300°C for 3 min) were performed on the preserved cores and their replicas that were exposed in open air for different times, to study dynamic loss process and the molecular composition changes of shale oil. The results show that during exposure, shale experiences a large amount of oil loss, with a loss ratio of about 42%–78%, and the higher the maturity, the greater the loss ratio. The oil loss is mainly contributed by free oil, with a loss ratio as high as 88%. The adsorbed oil content, however, remains basically unchanged and has a good positive correlation with the TOC of shale. Once the cores were crushed, the gaseous hydrocarbon in oil was basically evaporated in just 5 min. After long-term storage, 90% of the C14- light hydrocarbon is lost, while the C14+ heavy hydrocarbon experiences basically no loss. Therefore, effective and timely analysis of preserved shales is extremely important. The oil content of uncrushed shale cores characterized by NMR T1–T2 mapping is much greater than that of the crushed sample measured by TD-GC, which means that NMR T1–T2 mapping can be important method to evaluate the original fluid saturation of shale.

Wicking geotextiles have proven effective in reducing water content in road bases under both saturated and unsaturated conditions, thereby increasing granular base strength and mitigating moisture-related damage to the pavement. Despite their effectiveness in paved roads, the use of wicking geotextiles in permeable road and sidewalk, particularly in areas requiring robust drainage such as sponge cities, is not well explored. In “sponge city” roads, moisture content fluctuations and subsequent damage to the structure often cause concern, and the wicking geotextile’s drainage could be a potential solution. Therefore, this study aims to investigate and quantify the effectiveness of wicking geotextiles in reducing the moisture content and improving the resilient modulus of permeable sidewalk base layers. The moisture contents of unstabilized and one-directional wicking geotextile- and two-directional wicking geotextile-stabilized bases under permeable paving bricks were monitored with an interval of 7–10 days for over a year. An analytical approach to reconstruct daily moisture content in the base layer was proposed based on the simulated rainfall saturation test. This approach further assesses the enhancements in resilient modulus due to the drainage capabilities of wicking geotextiles. The experimental results indicated that the two-directional wicking geotextile outperforms its one-directional counterpart and both wicking geotextiles outperformed the control condition in terms of drainage efficiency. By reconstructing the daily moisture content and utilizing the relative damage model, the two-directional wicking geotextile significantly improved the annual equivalent resilient modulus of the base layer under permeable paving bricks.

Abstract BackgroundKnowledge construction in the context of children’s science education is an important part of fostering the development of early scientific literacy. Nevertheless, children sometimes struggle to comprehend scientific knowledge due to the presence of abstract notions. ObjectiveThis study aimed to evaluate the efficacy of augmented reality (AR) games as a teaching tool for enhancing children’s understanding of optical science education. MethodsA total of 36 healthy Chinese children aged 6-8 years were included in this study. The children were randomly divided into an intervention group (n=18, 50%) and a control group (n=18, 50%). The intervention group received 20 minutes of AR science education using 3 game-based learning modules, whereas the control group was asked to learn the same knowledge for 20 minutes with a non-AR science learning app. Predict observe explain tests for 3 topics (animal vision, light transmission, and color-light mixing) were conducted for all participants before and after the experiment. BackgroundGap junction proteins (GJPs) are a class of channel proteins that are closely related to cell communication and tumor development. The objective of this study was to screen out GJPs related prognostic signatures (GRPS) associated with clear cell renal cell carcinoma (ccRCC).Materials and MethodsGJPs microarray data for ccRCC patients were obtained from The Gene Expression Omnibus (GEO) database, along with RNA sequencing data for tumor and paired normal tissues from The Cancer Genome Atlas (TCGA) database. In the TCGA database, least absolute shrinkage and selection Operator (LASSO) and Cox regression models were used to identify GJPs with independent prognostic effects as GRPS in ccRCC patients. According to the GRPS expression and regression coefficient from the multivariate Cox regression model, the risk score (RS) of each ccRCC patient was calculated, to construct the RS prognostic model to predict survival. Overall survival (OS) and progression-free survival (PFS) analyses; gene pan-cancer analysis; single gene survival analysis; gene joint effect analysis; functional enrichment analysis; tumor microenvironment (TME) analysis; tumor mutational burden (TMB) analysis; and drug sensitivity analysis were used to explore the biological function, mechanism of action and clinical significance of GRPS in ccRCC. Further verification of the genetic signature was performed with data from the GEO database. Finally, the cytofunctional experiments were used to verify the biological significance of GRPS associated GJPs in ccRCC cell lines.ResultsGJA5 and GJB1, which are GRPS markers of ccRCC patients, were identified through LASSO and Cox regression models. Low expression of GJA5 and GJB1 is associated with poor patient prognosis. Patients with high-RS had significantly shorter OS and PFS than patients with low-RS (p< 0.001). The risk of death for individuals with high-RS was 1.695 times greater than that for those with low-RS (HR = 1.695, 95%CI= 1.439-1.996, p< 0.001). Receiver Operating Characteristic (ROC) curve showed the great predictive power of the RS prognostic model for the survival rate of patients. The area under curve (AUC) values for predicting 1-year, 3-year and 5-year survival rates were 0.740, 0.781 and 0.771, respectively. The clinical column chart was also reliable for predicting the survival rate of patients, with AUC values of 0.859, 0.846 and 0.796 for predicting 1-year, 3-year and 5-year survival, respectively. The GRPS was associated with immune cell infiltration, the TME, the TMB, and sensitivity to chemotherapy drugs. Further in vitro experiments showed that knockdown of GJA5 or GJB1 could promote the proliferation, migration and epithelial-mesenchymal transition (EMT) and inhibit apoptosis of ccRCC cells.ConclusionGJA5 and GJB1 could be potential biological markers for predicting survival in patients with ccRCC.

BackgroundRecently, serum sialic acid (SA) has emerged as a distinct prognostic marker for prostate cancer (PCa) and bone metastases, warranting differential treatment and prognosis for low-volume (LVD) and high-volume disease (HVD). In clinical settings, evaluating bone metastases can prove advantageous.ObjectivesWe aimed to establish the correlation between SA and both bone metastasis and HVD in newly diagnosed PCa patients.MethodsWe conducted a retrospective analysis of 1202 patients who received a new diagnosis of PCa between November 2014 and February 2021. We compared pretreatment SA levels across multiple groups and investigated the associations between SA levels and the clinical parameters of patients. Additionally, we compared the differences between HVD and LVD. We utilized several statistical methods, including the non-parametric Mann-Whitney U test, Spearman correlation, receiver operating characteristic (ROC) curve analysis, and logistic regression.ResultsThe results indicate that SA may serve as a predictor of bone metastasis in patients with HVD. ROC curve analysis revealed a cut-off value of 56.15 mg/dL with an area under the curve of 0.767 (95% CI: 0.703-0.832, P < 0.001) for bone metastasis versus without bone metastasis and a cut-off value of 65.80 mg/dL with an area under the curve of 0.766 (95% CI: 0.644-0.888, P = 0.003) for HVD versus LVD. Notably, PCa patients with bone metastases exhibited significantly higher SA levels than those without bone metastases, and HVD patients had higher SA levels than LVD patients. In comparison to the non-metastatic and LVD cohorts, the cohort with HVD exhibited higher levels of alkaline phosphatase (AKP) (median, 122.00 U/L), fibrinogen (FIB) (median, 3.63 g/L), and prostate-specific antigen (PSA) (median, 215.70 ng/mL), as well as higher Gleason scores (> 7). Multivariate logistic regression analysis demonstrated that an SA level of > 56.15 mg/dL was independently associated with the presence of bone metastases in PCa patients (OR = 2.966, P = 0.018), while an SA level of > 65.80 mg/dL was independently associated with HVD (OR = 1.194, P = 0.048).ConclusionThe pretreatment serum SA level is positively correlated with the presence of bone metastases.

We theoretically investigate coherent controlling single photon transport in one-dimensional waveguide coupled to a giant atom interacting with a resonator. The giant atom is considered as a Λ-type system. One transition of the atom couples to the waveguide and the other one is driven by the resonator. Single photon scattering amplitudes are calculated for three kinds of coupling schematics, respectively. Both the analytical and numerical results show that single photon scattering properties can be manipulated by adjusting the size of the giant atom, the coupling strengths of the waveguide-resonator and resonator-giant atom. Specifically, the transmission spectra exhibit two Rabi-splitting dips and the locations of the dips are strongly dependent on the size of the giant atom (GA). It also shows that the resonant single photon can be tuned from being transmitted to being reflected by controlling the coupling strength. Our results may find applications in designing quantum devices based on giant atom at single-photon level.

Objective: To investigate the metabolic trajectory of kidney aging and the effects of Polygonatum sibiricum polysaccharides (PSP) against kidney aging in D-galactose (D-gal)-induced aging mice, based on ultra-performance liquid chromatography/Q-Exactive Orbitrap mass spectrometry (UPLC-Q-Exactive MS/MS). Methods: A total of 36 C57 BL/6J mice were randomly allocated to six groups: control (CON), model (MOD), PSP low-dose (PSP-L), PSP medium-dose (PSP-M), PSP high-dose (PSP-H), and positive drug ascorbic acid (VC) groups. To create models of aging mice, D-gal was intraperitoneally administered to all other groups of mice except the CON group. After modeling, the appropriate Chinese medicine [PSP-L: 150 mg/(kg·d), PSP-M: 300 mg/(kg·d), PSP-H: 600 mg/(kg·d)] or positive drug [ascorbic acid, 300 mg/(kg·d)] was administered for intervention. Key markers of renal function in urine and serum of mice in each group, such as creatinine (Crea), urea nitrogen (BUN), and uric acid (UA) levels, as well as key indicators of oxidative stress in serum and kidney, including superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), and glutathione peroxidase (GSH-Px) were determined to validate the successful establishment of kidney aging models and to estimate the effects of PSP. Hematoxylin and eosin (HE), periodic acid Schiff (PAS), and β-galactosidase staining were used to assess the renal pathological changes. The metabolic profiles of serum, kidney, and urine samples from CON, MOD, and PSP-H groups were analyzed by UPLC-Q-Exactive MS/MS, and pattern recognition methods were used to outline the metabolic trajectory of kidney aging and to identify the characteristic metabolites. Results: Age-related alterations in renal histopathology and impaired renal function in mice were also associated with oxidative stress indicators. Following the injection of PSP [PSP-H: 600 mg/(kg·d)], the pathological indices associated with aging were adjusted to normal levels, renal function and oxidative stress were improved in aging mice, and renal pathological damage was markedly improved. Meanwhile, the potential biomarkers were identified by UPLC-Q-Exactive MS/MS analysis and were further analyzed to form related metabolic pathways, with P < 0.05 as a threshold. The results showed that purine, sphingolipid, glycerophospholipid, tryptophan, and riboflavin metabolisms were the main metabolic pathways associated with aging. After administration of PSP, these pathological indices returned to normal levels, and biomarkers related to the aging process, such as adenosine monophosphate (AMP), tryptophan, and 5-hydroxytryptophan, also demonstrated, to some degree, reverse regulation (promoting synthesis). Conclusion: Metabolomics methods based on UPLC-Q-Exactive MS/MS and multivariate statistical analysis can be adopted to establish metabolic profiles in aging mice. PSP has been shown to protect against kidney aging by interfering with the purine, sphingolipid, glycerophospholipid, tryptophan, and riboflavin metabolisms in the kidney.

AIM: To determine the 15-year outcomes of laser dacryoplasty (LDP) in patients with lacrimal duct obstruction; and to evaluate LDP combined with intubation using a new silicone tube to treat complicated cases. METHODS: Patients with lacrimal duct obstruction and treated with LDP between April 2000 and April 2005 were investigated retrospectively. Totally 116 eyes with completed 15-year follow-up records were included in this study. For complicated cases (52 eyes of 52 patients), both LDP and intubation using a self-made silicon tube were performed. For patients with uncomplicated obstruction (64 eyes of 61 patients), only LDP was performed. Outcomes were assessed based on results of lacrimal irrigation and degree of symptoms during follow-up. RESULTS: At the follow-up time of 15y, 81 eyes achieved full success (69.8%); 21 eyes got improved (18.1%); and 14 eyes were considered failure (12.1%). The success rate was 71.2% (37/52 eyes) for complicated cases; and 68.8% (44/64 eyes) for uncomplicated cases. No statistically significant difference between two groups was observed (P=0.961). No postoperative complication was observed. CONCLUSION: LDP is a well-tolerated, simple, and effective procedure with satisfactory long-term outcomes in selected patients, which make it a good alternative to conventional dacryocystorhinostomy. In addition, intubation with the self-made mono-canalicular silicone tube facilitates the management of complicated cases with few complications.

AIM: To analyze the causality between type 2 diabetes mellitus(T2DM)and age-related macular degeneration(ARMD)based on two-sample Mendelian randomization(MR).METHODS: T2DM and ARMD samples were extracted from the FinnGen database. Inverse variance weighted(IVW)was used as the main analysis method, MR-Egger and weighted median(WM)as supplementary methods to analyze the potential relationship between them. In addition, Cochran Q test and MR-Egger intercept were also used to analyze the sensitivity, and the P-value was used as the index of research results.RESULTS: IVW showed that T2DM was associated with the incidence of exudative ARMD(OR=1.14, 95%CI 1.01～1.28, P=0.021), but it was not significantly associated with the incidence of atrophic ARMD(OR=0.96, 95%CI 0.86～1.07, P=0.554). The results of sensitivity analysis confirmed that there was no heterogeneity and pleiotropy in this study, and the results were reliable.CONCLUSION: There is a causal relationship between T2DM and exudative ARMD. Considering the high rate of blindness caused by ARMD, it is of great significance to recognize and control the risk factors of ARMD to reduce its prevalence rate and early diagnosis and treatment.

Aiming at the problem that the unknown variation rule of molten pool and the interlayer effect on the forming efficiency and precision of products in the process of selective laser melting(SLM) of high-strength aluminum alloy A17075, the influence of different process parameters (laser power and scanning speed) on the morphology of the molten pool and the temperature field of the various forming layers was investigated. The finite element analysis software ANSYS was used to establish a multi-layer and multi-channel temperature field finite element model for SLM forming of thin-walled metal parts, and at the same time, the loading of the laser heat source, the laser power and the scanning speed were simulated by using the programming of the language APDL (ansys parametric design language), and the "element birth and death" technique was adopted to describe the dynamic growth process of the metal powder material and the distribution of the transient temperature field was derived. The results show that the laser power and scanning speed affect different temperature field factors, and the SLM process parameters suitable for Al7075 powder are power of 250~300 W and speed of 800~1 000 mm/s. The reasonable range of leser power and scanning speed is obtained, which provides theoretical reference and support for high-strength aluminum alloy in SLM practical experiments.

Abstract Background Lignin is a major restriction factor for the industrial production of biomass resources, such as pulp and bioenergy. Eucalyptus is one of the most important sources of pulp and bioenergy. After polyploidization, the lignin content of forest trees is generally reduced, which is considered a beneficial genetic improvement. However, the differences in the lignin content between triploid and diploid Eucalyptus and the underlying regulatory mechanism are still unclear. Results We conducted a comprehensive analysis at the phenotypic, transcriptional and metabolite levels between Eucalyptus urophylla triploids and diploids to reveal the effects of polyploidization on the lignin content and lignin metabolic pathway. The results showed that the lignin content of Eucalyptus urophylla triploid stems was significantly lower than that of diploids. Lignin-related metabolites were differentially accumulated between triploids and diploids, among which coniferaldehyde, p-coumaryl alcohol, sinapaldehyde and coniferyl alcohol had significant positive correlations with lignin content, indicating that they might be primarily contributing metabolites. Most lignin biosynthetic genes were significantly downregulated, among which 11 genes were significantly positively correlated with the lignin content and above metabolites. Furthermore, we constructed a co-expression network between lignin biosynthetic genes and transcription factors based on weighted gene co-expression network analysis. The network identified some putative orthologues of secondary cell wall (SCW)-related transcription factors, among which MYB52, MYB42, NAC076, and LBD15 were significantly downregulated in Eucalyptus urophylla triploids. In addition, potential important transcription factors, including HSL1, BEE3, HHO 3, and NAC046, also had high degrees of connectivity and high edge weights with lignin biosynthetic genes, indicating that they might also be involved in the variation of lignin accumulation between triploid and diploid Eucalyptus urophylla. Conclusions The results demonstrated that some lignin-related metabolites, lignin biosynthetic genes and transcription factors in Eucalyptus urophylla triploids may be relatively sensitive in response to the polyploidization effect, significantly changing their expression levels, which ultimately correlated with the varied lignin content. The analysis of the underlying formation mechanism could provide beneficial information for the development and utilization of polyploid biomass resources, which will be also valuable for genetic improvement in other bioenergy plants.

We used 256 apple germplasm resources for a thorough examination of the genetic diversity associated with 26 phenotypic traits (i.e., genetic diversity analysis, cluster analysis, correlation analysis, principal component analysis, and membership function). The average coefficient of variation for 12 morphological traits was 66.39% (21.10–201.5%). The coefficient of variation was highest and lowest for the fruit arris and the width of the eye basin, respectively. Additionally, the diversity index ranged from 0.54 to 1.33. Moreover, the coefficient of variation for 14 numerical traits varied from 5.37% to 50%. The titratable acid content had the highest coefficient of variation, with a diversity index ranging from 2.01 to 2.08 (average of 2.045). A cluster analysis categorized 256 germplasms into four groups, among which Group I included germplasms with large fruits and the best comprehensive performance. Of the top 10 principal components revealed by the principal component analysis, principal component 1 was mainly related to fruit size and flavor. The top 10 germplasms were selected on the basis of comprehensive scores using the membership function method. Furthermore, a stepwise regression analysis identified 15 key traits for identifying apple germplasms, including the vegetative growth day, fruit weight, and the firmness of the fruit without skin. These results can serve as the foundation for future analyses of the phenotypic diversity of apple germplasms, while also providing a theoretical basis for screening, characterizing, and further improving excellent apple germplasms.
