Simple Summary: Chiggers are common ectoparasites on rodents and other small mammals, and they transmit scrub typhus, a zoonotic disease. Dehong in Yunnan Province of southwest China is located on the China–Myanmar border, and it is a focus of scrub typhus. The present paper reports the infestation and distribution of chiggers on small mammals in Dehong for the first time. From 1760 rodents and other sympatric small mammals, a total of 9309 chiggers were identified, representing 117 species. Most chigger species had low host specificity. Leptotrombidium deliense, a major vector of scrub typhus in China, was the dominant chigger species in Dehong, and it was mainly distributed in flatland areas and indoors. The infestation and community indexes of chiggers in mountainous areas and outdoors were higher than those in flatland areas and indoors. The species abundance distribution of the chigger community conformed to log-normal distribution, and the total number of chigger species was roughly estimated to be 147. The species diversity of the chigger community is high in Dehong, with an obvious environmental heterogeneity. The low host specificity of chiggers and the occurrence of a large number of L. deliense in Dehong would increase the transmission risk of scrub typhus on the China–Myanmar border. Dehong Prefecture in Yunnan Province of southwest China is located on the China–Myanmar border and is an important focus of scrub typhus. Based on the field surveys in Dehong between 2008 and 2022, the present paper reports the infestation and ecological distribution of chiggers on the body surface of rodents and other sympatric small mammals (shrews, tree shrews, etc.) in the region for the first time. The constituent ratio (Cr), prevalence (PM), mean abundance (MA), and mean intensity (MI) were routinely calculated to reflect the infestation of small-mammal hosts with chiggers. Additionally, the species richness (S), Shannon–Wiener diversity index (H), Simpson dominance index (D), and Pielou's evenness index (E) were calculated to illustrate the chigger community structure. Preston's log-normal model was used to fit the theoretical curve of species abundance distribution, and the Chao 1 formula was used to roughly estimate the expected total species. The "corrplot" package in R software (Version 4.3.1) was used to analyze interspecific relationships, and the online drawing software was used to create a chord diagram to visualize the host–chigger associations. From 1760 small-mammal hosts, a total of 9309 chiggers were identified as belonging to 1 family, 16 genera, and 117 species, with high species diversity. The dominant chigger species were Leptotrombidium deliense, Walchia ewingi, and Gahrliepia longipedalis, with a total Cr = 47.65% (4436/9309), among which L. deliense is the most important vector of Ot in China. The overall infestation indexes (PM, MA, and MI) and community parameters (S, H, and E) of chiggers in the mountainous areas and outdoors were higher than those in the flatland areas and indoors, with an obvious environmental heterogeneity. Leptotrombidium deliense was the dominant species in the flatland and indoors, while G. longipedalis was the prevalent species in the mountainous and outdoor areas. The species abundance distribution of the chigger community conformed to log-normal distribution with the theoretical curve equation: S (R) ′ = 28 e − [ 0.23 (R − 0) ] 2 , indicating the existence of many rare species and only a few dominant species in the community. The expected total number of chigger species was roughly estimated to be 147 species, 30 more than the 117 species actually collected, suggesting that some uncommon species may have been missed in the sampling survey. The host–parasite association analysis revealed that one host species can harbor different chigger species, and one chigger species can parasitize different host species with low host specificity. A positive or negative correlation existed among different chigger species, indicating a cooperative or competitive interspecific relationship. The species diversity of chiggers is high in Dehong on the China–Myanmar border, and a large host sample is recommended to find more uncommon species. There is an obvious environmental heterogeneity of the chigger community, with different species diversity and dominant species in different environments. The low host specificity of chiggers and the occurrence of a large number of L. deliense in Dehong, especially in flatland areas and indoors, would increase the risk of persistent transmission of scrub typhus in the region. Functionally graded materials with porosity have received increased application in satellites, space vehicles, aircraft, and other transportation systems. The multi-physics coupled modeling technique for structural response analysis remains a big challenge. This paper develops a finite element formulation based on the first-order shear deformation (FOSD) hypothesis for evaluation of the static and dynamic behavior of functionally graded magneto-electro-elastic porous (FG-MEEP) cylindrical shells under thermal loads. Four types of thermal loads such as uniform, linear, sinusoidal, and heat conduction temperature rise are included in the FE model. Furthermore, FG-U, FG-V, FG-O, and FG-X distributions of porosity is considered. The material parameters of FG-MEEP are obtained by modified power law. Two forms of material gradation in the framework of 'B'-rich bottom and 'F'-rich bottom are used. The correctness of the present model is verified by comparing with the results of literature. Finally, parametric studies are adopted to analyze the static and dynamic response of FG-MEEP cylindrical shell by varying functionally graded pattern, gradient index, porosity volume, temperature, porosity distribution, etc. These numerical results can serve as benchmarks for the future study of porous structures in thermal environment. The 17 host species of L. imphalum crossed 13 genera and 5 families in 3 orders (Rodentia, Eulipotyphla, and Scandentia), indicating the low host specificity of the mite. The Asian house rat (Rattus tanezumi) was the dominant host species in the 21 sites where L. imphalum were collected, and 49.38% of mites were found on R. tanezumi. Different small mammals had different susceptibility to the infestation of L. imphalum. The prevalence (PM = 27.66%), infestation mean abundance (MA = 6 mites/per examined host), and mean intensity (MI = 21.69 mites/per infested host) for L. imphalum on the shrew gymnure (Neotetracus sinensis) were much higher than those on other host species (p < 0.05), indicating N. sinensis had a high susceptibility to the infestation of L. imphalum. The infestation indices for L. imphalum on small mammal hosts varied along different altitude and latitude gradients (p < 0.05), indicating the environmental heterogeneity of the mite infestation. This article deals with large deflection analysis of functionally graded carbon nanotube-reinforced magneto-electro-elastic (MEE) cylindrical shells using fully geometrically nonlinear finite element methods. The first-order shear deformation theory and strain–displacement relations of large rotations are employed to obtain the governing equations. The nonlinear dynamic models are derived from Hamilton's principle. Four distribution patterns of carbon nanotubes embedded in the piezoelectric matrix are considered. The credibility of the proposed fully geometrically nonlinear model is firstly verified by comparing the displacements of MEE laminated plates and shells in the published literature. Further, the influence of various parameters on the static and dynamic performance of the structure has been studied. The goal of our study was to evaluate the potential role of sTNF-RI as a biomarker of renal involvement in SLE patients and active SLE. The study sample consisted of two cohorts. The discovery cohort included 16 SLE patients without renal involvement (non-LN), 60 lupus nephritis (LN) patients and 21 healthy controls (HCs) and the replication cohort included 18 SLE non-LN patients, 116 LN patients and 36 HCs. The sTNF-RI levels differed significantly in the discovery cohort. The plasma sTNF-RI levels were higher in LN patients than in non-LN patients (p =.009) and HCs (p = 4 × 10−6). Plasma sTNF-RI levels were significantly higher in non-LN patients than in HCs (p =.03). The finding was confirmed in independent replication cohort (LNs vs. non-LN, p = 4.053 × 10−7; LNs vs. HCs, p = 2.395 × 10−18; non-LN vs. HCs, p = 2.51 × 10−4). The plasma sTNF-RI levels were associated with disease activity, renal function in SLE patients and urine protein in LN patients. The multivariate analysis revealed that high sTNF-RI was an independent risk factor for renal involvement. The multivariate logistic regression results suggested that high TNF-RI, high systolic blood pressure, high serum creatinine, low C4 and positive anti-dsDNA were independent risks of active SLE patients. A nomogram was constructed based on the results of multivariate logistic regression analysis and it was practical in predicting the risk of the active SLE patients. Immunohistochemistry suggested that the expression of TNF-RI in the kidney was increased. Plasma sTNF-RI might be a good biomarker of renal involvement and disease activity in SLE patients. A recent genome-wide association study (GWAS) of Asian ancestry reported that single nucleotide polymorphism (SNP) in TERT (telomerase reverse transcriptase) was associated with systemic lupus erythematosus (SLE). TERT has a critical role in maintaining the chromosomal stability and the length of telomere. Given that only a small portion of the genetic heritability of SLE has been explained so far, we aimed to identify novel loci in telomere-related genes responsible for SLE susceptibility in Chinese populations. We performed a comprehensive genetic association analysis of SLE with telomere-related genes. To identify functional significance, we analyzed the publicly available HaploReg v4.1 and RegulomeDB databases. Differential gene expression analysis was also performed using ArrayExpress. A novel signal of PINX1 rs6984094 was identified (Pdiscovery = 4.13 × 10-2, OR = 0.58, 95% CI 0.35-0.98) and successfully replicated (Preplication = 5.73 × 10-3, OR = 0.45, 95% CI 0.26-0.81). Multiple layers of functional analysis suggested that the PINX1 rs6984094 risk T allele exhibited increased nuclear protein binding. We also observed an increased expression of PINX1 mRNA in peripheral blood mononuclear cells from SLE patients compared with healthy controls. Overall, we observed a novel genetic association between PINX1 (encodes the PinX1 protein, an inhibitory telomerase enzyme that lengthens telomeres) and SLE susceptibility in Chinese populations. Background: Systemic lupus erythematosus (SLE) is a complex autoimmune disease with strong genetic disposition with more than 100 susceptibility genes identified until now. However, our knowledge on SLE genetic background is still limited. The present study was aimed at evaluating the role of single nucleotide polymorphisms (SNPs) in SCUBE3, a TGF-β signaling activator, with SLE susceptibility in Chinese populations.Methods: A total of 2801 individuals (490 cases and 493 controls from GWAS cohort and 1003 cases and 815 controls from our cohort) were enrolled, and SNPs located 10 kb up- and downstream of SCUBE3 (chr6:35182190-35218609) were included in the genetic association study. Multiple layers of bioinformatics were conducted, and the levels of SCUBE3 expression were confirmed.Results: Of the 31 SNPs in SCUBE3 tested, 24 SNPs were significantly associated with SLE at p ≤ 0.05. The top locus was rs1888822 with p = 8.74∗10-6 in the discovery cohort and was confirmed by the replication cohort with p = 0.012. Additionally, the levels of SCUBE3 mRNA expression were significantly lower in patients with SLE comparing with healthy controls (p = 4.28∗10-4). Further expression data from ArrayExpress showed that the expression of SCUBE3 was also lower in CD3+ T cells and B cells from patients with SLE.Conclusions: Our research revealed that variants in SCUBE3, which encode SCUBE3 as a TGF-β signaling activator, can be considered as a new genetic susceptibility factor for systemic lupus erythematosus. Background: Systemic lupus erythematosus (SLE) is a multifactorial autoimmune disease characterized by apoptotic clearance deficiency provoking autoimmune responses and leading to multiple organ damage. PPAR-δ, encoded by the PPARD gene, was induced in macrophages promoting the timely disposal of apoptotic cells. Biological studies had provided solid foundation of PPARD involvement in SLE; it is worthwhile to further explore the genetic contribution of PPARD to SLE.Methods: We performed a discovery-replication genetic association study. The discovery study was based on previous reported GWAS data. And the replication study was conducted in 1003 SLE patients and 815 healthy controls from Henan, Middle East of China. Further, we analyzed the eQTL effect to identify possible functional significance.Results: In the genetic association analysis, we observed significant association between the risk C allele of rs4713853 (p = 0.03, OR 1.167, 95% CI 1.015-1.341) and increased SLE susceptibility. Moreover, individuals with the risk C allele were associated with lower expression of PPARD and DEF6. Our clinical analysis showed that SLE patients with the risk C allele of rs4713853 were more likely to present a higher proportion of anti-Sm antibody presence (CC+CT vs. TT, 20.0% vs. 14.2%, p = 0.039) and higher level of Scr (median inter quarter range CC+CT vs. TT, 56 48-71 vs. 54 46-64 μmol/L, p = 0.002).Conclusions: In conclusion, our study identified a novel association between PPARD rs4713853 and SLE susceptibility in Chinese populations.
