BackgroundThis study reports a case of severe cutaneous toxicity in a patient with metastatic gastric cancer induced by disitamab vedotin, emphasizing the need for careful monitoring and management in such treatments.Case presentationA 71-year-old female was admitted to hospital complaining of serious rashes following the first cycle of disitamab vedotin regimen for metastatic gastric cancer. The doctor diagnosedtoxic epidermal necrolysis (TEN) induced by the drug. The patient received high-dose methylprednisolone due to the side effects. This resulted in a gradual improvement of symptoms.ConclusionDuring the use of disitamab vedotin, patients need to be monitored for severe skin toxicity.

BackgroundMutations in PIEZO1 (Piezo type mechanosensitive ion channel component 1) cause human lymphatic malformations. We have previously uncovered an ORAI1 (ORAI calcium release-activated calcium modulator 1)-mediated mechanotransduction pathway that triggers lymphatic sprouting through Notch downregulation in response to fluid flow. However, the identity of its upstream mechanosensor remains unknown. This study aimed to identify and characterize the molecular sensor that translates the flow-mediated external signal to the Orai1-regulated lymphatic expansion.MethodsVarious mutant mouse models, cellular, biochemical, and molecular biology tools, and a mouse tail lymphedema model were employed to elucidate the role of Piezo1 in flow-induced lymphatic growth and regeneration.ResultsPiezo1 was found to be abundantly expressed in lymphatic endothelial cells. Piezo1 knockdown in cultured lymphatic endothelial cells inhibited the laminar flow-induced calcium influx and abrogated the flow-mediated regulation of the Orai1 downstream genes, such as KLF2 (Krüppel-like factor 2), DTX1 (Deltex E3 ubiquitin ligase 1), DTX3L (Deltex E3 ubiquitin ligase 3L,) and NOTCH1 (Notch receptor 1), which are involved in lymphatic sprouting. Conversely, stimulation of Piezo1 activated the Orai1-regulated mechanotransduction in the absence of fluid flow. Piezo1-mediated mechanotransduction was significantly blocked by Orai1 inhibition, establishing the epistatic relationship between Piezo1 and Orai1. Lymphatic-specific conditional Piezo1 knockout largely phenocopied sprouting defects shown in Orai1- or Klf2- knockout lymphatics during embryo development. Postnatal deletion of Piezo1 induced lymphatic regression in adults. Ectopic Dtx3L expression rescued the lymphatic defects caused by Piezo1 knockout, affirming that the Piezo1 promotes lymphatic sprouting through Notch downregulation. Consistently, transgenic Piezo1 expression or pharmacological Piezo1 activation enhanced lymphatic sprouting. Finally, we assessed a potential therapeutic value of Piezo1 activation in lymphatic regeneration and found that a Piezo1 agonist, Yoda1, effectively suppressed postsurgical lymphedema development.ConclusionsPiezo1 is an upstream mechanosensor for the lymphatic mechanotransduction pathway and regulates lymphatic growth in response to external physical stimuli. Piezo1 activation presents a novel therapeutic opportunity for preventing postsurgical lymphedema. The Piezo1-regulated lymphangiogenesis mechanism offers a molecular basis for Piezo1-associated lymphatic malformation in humans.

为了对比不同油体富集物理化性质及功能性质的差异，采用水萃离心法从大豆、花生、葵花籽3种大宗油料种子中提取油体富集物，对比研究了油体富集物的得率、基本成分、粒径、Zeta电位、蛋白质组成、黏度、起泡性、乳化性、分层稳定性以及冻融稳定性。结果表明：大豆油体富集物的得率和固形物含量都显著低于葵花籽和花生油体富集物的（p＜0.05），且具有不易掩盖的豆腥味；3种油体富集物的基本成分之间都存在显著性差异（p＜0.05），蛋白质的分子质量分布也不尽相同；3种油体富集物的粒径和Zeta电位绝对值由小到大顺序均为大豆油体富集物

为了探究葵花籽油体富集物在冰淇淋中应用的可行性，以葵花籽油体富集物作为脂肪来源制备低脂冰淇淋，在无均质条件下研究葵花籽油体富集物添加量对低脂冰淇淋浆料和产品品质的影响。结果表明：冰淇淋浆料的黏度、储能模量和损耗模量都随葵花籽油体富集物添加量的增加而增大，当葵花籽油体富集物添加量为3.5%～4.0%时黏度与对照组相当；浆料中不易流动水和结合水的比例随着葵花籽油体富集物添加量的增加逐渐增大,在4.0%时达到最大；冰淇淋的膨胀率和抗融率随着葵花籽油体富集物添加量的增加先增大后减小；添加葵花籽油体富集物的冰淇淋色泽较差；冰淇淋的硬度和咀嚼性变化规律一致，都是随着葵花籽油体富集物添加量的增大呈现先减小后增大的趋势，弹性变化则相反;冰淇淋黏附性随着葵花籽油体富集物添加量的增加而升高；葵花籽油体富集物添加量为2.5%时冰淇淋感官评分最高。合适的葵花籽油体富集物添加量可以起到与棕榈油相似的效果，因此葵花籽油体富集物可以应用在冰淇淋中。In order to investigate the feasibility of using sunflower seed oil body enrichment in ice cream,low fat ice cream was prepared with sunflower seed oil body enrichment as the fat under the condition of non-homogenization. The effect of the dosage of the oil body enrichment on the quality of low fat ice cream slurry and product was explored. The results showed that the viscosity, storage modulus and loss modulus of ice cream slurry increased with the increase of the oil body enrichment dosage. When the dosage of oil body enrichment was 3.5%- 4.0%, the viscosity was equivalent to that of the control group. The proportion of non-flowing water and bound water in the slurry increased gradually with the increase of the dosage of oil body enrichment, reaching the maximum at 4.0%. The expansion rate and anti-melting rate of ice cream products first increased and then decreased as the increase of the oil body enrichment dosage. The color of ice cream with sunflower seed oil body enrichment was poor. The hardness and chewability of ice cream showed the same change law, which first decreased and then increased with the increase of the oil body enrichment dosage,while the elasticity changed in the opposite direction. The adhesion of ice cream increased with the increase of the oil body enrichment dosage. The sensory score of ice cream was the highest when the dosage of oil body enrichment was 2.5%. The appropriate dosage of sunflower seed oil body enrichment can produce a similar effect as palm oil, and sunflower seed oil body enrichment can be applied in ice cream.

A PLS-based quality prediction method is proposed for batch processes using two-dimensional extended windows. To realize the adoption of information in the directions of sampling time and batch, a newly defined region of support (ROS), called the k-i-back-extended region of support (KIBROS), is proposed; it establishes an extended window by adding two regions of data to the traditional ROS to include all possible important data for quality prediction. Based on the new ROS, extended windows are established, and different models are proposed using the extended windows for batch process quality prediction. Furthermore, using the typical injection molding batch process as an example, the proposed quality prediction method is experimentally verified, proving that the proposed methods have higher prediction accuracy than the traditional method and that the prediction stability is also improved. Summary: The use of two emulsifiers to fabricate a composite emulsifier is a useful strategy used to strengthen the emulsifying capacity. To investigate the feasibility of using soy protein isolate (SPI) to stabilise Pickering emulsions stabilised by soy peptide nanoparticles (SPNs), the SPI was introduced into the SPN system to fabricate SPI‐SPN complex emulsifiers, and the effects of the SPI on the properties of the SPNs and SPN‐stabilised emulsions were investigated with various oil fractions. The addition of SPI significantly reduced the particle sizes and surface hydrophobicities. Furthermore, the SPI increased the structural rearrangements of the SPNs at the oil–water interface. Compared with emulsions stabilised with the SPN alone, the droplet sizes of the SPI‐SPN emulsions were smaller, and the interface protein adsorption percentages were lower for all oil fractions. The viscosity of the SPI‐SPN emulsion was higher than that of the SPNs emulsion at Φ ≤ 0.60. Decreases in G′ and G″ were observed when the SPI was added at all Φ values tested. More importantly, the added SPI markedly enhanced the creaming stabilities of the SPN emulsions at low Φ (≤0.40). These results confirmed that SPI improved the emulsifying activity of the SPNs and enhanced the stabilities of Pickering emulsions stabilised with SPNs at low Φ. [ABSTRACT FROM AUTHOR]

Heat treatment was used to extend the shelf life of postharvest Chaenomeles speciosa (Sweet) Nakai (C. speciosa), its effects on the quality and physiological properties of C. speciosa were studied. The results showed that heat treatment reduced the weight loss, inhibited the decline of titratable acids (TA), maintained a higher activity of peroxidase (POD) and catalase (CAT), compared to untreated C. speciosa. Treatment in hot water dipping at 50℃ for 2-4 min maintained the better quality of postharvest C. speciosa stored at 0℃ for 120 days, which indicated that heat treatment is an effective preservation technology to prolong the shelf life of C. speciosa.

Most industrial processes have nonlinear and time-delay characteristics leading to difficulty in prediction modeling. In addition, the working conditions of most industrial processes are complex, which results in multiple modes. Testing and modeling for each mode is a waste of time and resources. Therefore, it is urgent to complete model migration between different modes. In this work, a new prediction model, a nonlinear autoregressive model with exogenous inputs and back propagation neural network (NARX-BP), is proposed for the nonlinear and time-delay processes, where the input data order of the model is determined by the feedforward neural network (FNN) method, and the nonlinear relation is realized by the BP neural network. For the multi-mode characteristic, a new migration optimization algorithm, input–output slope/bias correction and differential evolution (IOSBC-DE), is provided for using a small amount of data under a new mode to correct the slope and bias of the relationship between the input and output variables through DE. The modeling and migration methods are applied to a wind tunnel system, and the simulation result shows the effectiveness of the proposed method. [ABSTRACT FROM AUTHOR]
