Background Cardiovascular diseases seriously threaten the health of the elderly in our country and bring a huge economic burden to our country. Accurate screening of risk assessment tools for early cardiovascular diseases can specifically prevent and delay the development of cardiovascular diseases, studies have suggested that commonly used cardiovascular risk scoring tools are not effective in predicting risk in the elderly, but there is a lack of relevant external validation experimental studies, and there is no well-recognized effective risk prediction tool for elderly patients. Objective To independently validate and compare the efficacy of five commonly used cardiovascular risk assessment tools in China and abroad in predicting 10-year cardiovascular disease risk in our community elderly population, to explore the cardiovascular disease risk assessment tools suitable for our community elderly population, and to provide theoretical basis and support for the prevention and treatment of cardiovascular disease at the grass-roots level. Methods From January 2012 to December 2013, more than 20 000 people in the Liuyuan Community Health Service Centre and Runda Community Health Service Centre in Suzhou Province took part in community medical examinations. According to the inclusion and exclusion criteria, three hundred and forty-four people were identified, data on age, sex, region of residence, systolic blood pressure, diastolic blood pressure, BMI, waist circumference, total cholesterol, high-density lipoprotein cholesterol, triglyceride, atrial fibrillation, Left ventricular hypertrophy, cardiovascular disease, hypertension, diabetes, and smoking were collected from the start-up medical examination system In the follow-up survey of chronic diseases, the current smoking status and smoking age, whether taking antihypertensive drugs, family history of cardiovascular disease, prevalence of hypertension and drug use of hypertension, prevalence of diabetes mellitus, and whether and when cardiovascular disease occurred from January 2014 to December 2022 were improved. The outcome events were stroke and coronary heart disease, and were divided into positive group and negative group, all subjects were assessed with the Framingham Heart Risk Score (FRS-CVD) , the modified Framingham Stroke Scale (R-FSRS) , the European systemic coronary risk assessment scale 2019 (SCORE) , Chinese 10-year ischemic cardiovascular disease risk (ICVD) , and the China-PAR cardiovascular disease risk assessment, concordance Index (C-index) , Hosmer-Lemeshow χ2 and calibration chart were used to evaluate the relationship between the results and actual cardiovascular disease incidence. Results Of the 344 enrolled investigators, with a mean age of 62 (56, 68) years, as of December 2022, 169 had developed cardiovascular disease and 175 had not. Two groups of people were carried out 5 kinds of cardiovascular disease risk score, cardiovascular disease risk assessment tool forecast and the actual incidence of the comparative analysis results are as follows, discrimination test: C-index of FRS-CVD was 0.711 (95%CI=0.658-0.764) , C-index of R-FSRS was 0.728 (95%CI=0.675-0.781) , SCORE: C-index was 0.724 (95%CI=0.671-0.777) , ICVD: C-index was 0.727 (95%CI=0.674-0.779) , China-PAR: C-index was 0.735 (95%CI=0.682-0.788) ; Hosmer-Lemeshow test calibration status: FRS-CVD: χ2 =16.789 (P=0.032) , R-FSRS: χ2=11.019 (P=0.201) , SCORE: χ2=20.396 (P=0.002) , ICVD: χ2=24.311 (P=0.001) , China-PAR: χ2=15.149 (P=0.056) ; R-FSRS is the best calibration. In men, model discrimination: FRS-CVD: C-index was 0.642 (95%CI=0.577-0.707) , R-FSRS: C-index was 0.646 (95%CI=0.581-0.710) , SCORE: C-index was 0.646 (95%CI=0.581-0.711) , ICVD: C-index was 0.628 (95%CI=0.563-0.693) , China-PAR: C-index was 0.636 (95%CI=0.571-0.700) ; Hosmer-Lemeshow test calibration status: FRS-CVD: χ2=7.371 (P=0.288) , R-FSRS: χ2=8.470 (P=0.293) , SCORE: χ2=5.146 (P=0.525) , ICVD: χ2=6.103 (P=0.412) , China-PAR: χ2=9.555 (P=0.298) , SCORE was calibrated best in the calibration diagram. Model discrimination among women: FRS-CVD: C-index was 0.698 (95%CI=0.633-0.762) , R-FSRS: C-index was 0.731 (95%CI=0.666-0.795) , SCORE: C-index was 0.733 (95%CI=0.668-0.798) , ICVD: C-index was 0.747 (95%CI=0.682-0.811) , China-PAR: C-index was 0.754 (95%CI=0.689-0.818) ; Hosmer-Lemeshow test calibration status: FRS-CVD: χ2=14.515 (P=0.069) , R-FSRS: χ2=12.175 (P=0.032) , SCORE: χ2=9.611 (P=0.022) , ICVD: χ2=19.349 (P=0.007) , China-PAR: χ2=12.372 (P=0.135) , China-PAR calibration is the best in calibration chart. Conclusion R-FSRS model has a good performance in predicting the risk of cardiovascular disease in the elderly, especially in the elderly population. SCORE model did better in predicting cardiovascular disease risk in older men, while China-PAR model did better in predicting cardiovascular disease risk in older women. Therefore, the application of these assessment tools will help doctors to accurately predict the risk of cardiovascular disease in the elderly, and formulate corresponding prevention and treatment strategies.

Objective To explore the protective effects and the underlying mechanisms of bone marrow mesenchymal stem cells (BMSCs) on the brain in microgravity environment. Methods The physiological effects of microgravity were simulated with the hindlimb unloading (HU) mouse model. The animals were divided into 3 groups: the control group (wild-type mice), the hindlimb unloading (HU) group, and the BMSCs treatment group, with six in each. RT-qPCR was adopted to determine the expression levels of pro-inflammatory cytokines (Il-6, Il-1β and Tnf-α) in the cerebral cortex; immunohistochemistry was performed to evaluate the proportions of microglia (IBA-positive) and astrocytes (GFAP-positive); Western blot was used to measure the expression levels of apoptosis-and senescence-related molecules (BAX, BCL-2, p21, and p53). Results Compared with wild-type mice, the expression levels of Il-6, Il-1β and Tnf-α in the cerebral cortex of HU mice were significantly increased (P＜0.05), the expression level of BAX, p21 and p53 was also significantly increased(P＜0.05). However, the expression level of BCL-2 protein were significantly decreased (P＜0.05). The proportion of microglia (IBA1 positive) and astrocytes (GFAP positive) was increased (P＜0.05); After BMSCs treatment, the expression level of Il-1β in the cerebral cortex of HU mice was significantly decreased (P＜0.05), the expression levels of BAX, p21 and p53 were significantly decreased (P＜0.05), and the expression level of BCL-2 protein was significantly increased (P＜0.05). The proportion of microglia (IBA1 positive) and astrocytes (GFAP positive) decreased (P＜0.05). Conclusions BMSCs potentially relieve the detrimental effects of simulated microgravity on mouse brain by inhibiting inflammation, apoptosis and cellular senescence.

Background and purpose: In 2016 the National Cancer Institute (NCI) decided stopping to use NCI-60 cell lines for drug screening, suggesting that tumor cell lines were losing their value as a tool for drug discovery and basic research. The reason for NCI-60 cells 'retirement' was that the preclinical studies based on traditional cellular and animal models did not obtain the corresponding expected efficacy in clinical trials. Since the major cancer behaviors, such as proliferation and metastasis, are fundamentally altered with long-term culture, the tumor cell lines are not representative of the characteristics of cancer in patients. Currently, scientists hope to create a new cancer model that are derived from fresh patient samples and tagged with details about their clinical past. Our purpose was to create patient-derived breast cancer primary cell lines as new cancer model for drug screening and basic research. Methods: Breast cancer tissues were collected in the Department of Breast Surgery, Affiliated Hospital of Guizhou Medical University. The collection of tumor tissue samples was approved by the Ethics Committee of the Affiliated Hospital of Guizhou Medical University (approval number: 2022 ethics No. 313), and the collection and use of tumor tissues complied with the Declaration of Helsinki. The primary breast cancer cell lines were isolated from the patient's breast cancer tissues and cultured in BCMI medium. After the cells proliferated, the media were replaced with DEME medium. Cell line STR genotyping was done to determine cell-specific genetic markers and identification. Clone formation assay and transplantation assay were done to analyze the ability of breast cancer primary cell lines to form tumors. Results: We created 6 primary breast cancer cell lines. The 6 primary breast cancer cell lines from the patients were tagged with the definitively clinicopathological features, clinical diagnosis, therapeutic regimens, clinical effectiveness and prognostic outcomes. The STR genotyping assays identified the genetic markers and determined the identities of the 6 primary breast cancer cell lines. Clone formation assays and transplantation assay showed that the proliferative capacities of the patient-derived primary breast cancer cell lines were significantly greater compared with the conventional breast cancer cell lines. Conclusion: We created a panel of 6 patient-derived primary breast cancer cell lines as new cancer model for drug screening and basic research in breast cancer.

Objective To verify the effect of bone marrow mesenchymal stem cells (BM-MSCs) in alleviating immune senescence, and to explore the main immune cell population improved by BM-MSCs. Methods Mouse spleen lymphocytes were isolated and stimulated to proliferate for 7 days for constructing a replicative aging model. Flow cytometry was used to detect the p16ink4a(p16) and p21cip1(p21) expression by T cell subpopulation in the young control group, the replicative senescence control group and the BM-MSCs co-cultured group. Results In the replicative senescence model of T lymphocytes, it was observed that CD8+T cells senescent significantly as compared with CD4+T cells after continuous proliferation. Among the naive cells and effector cell subsets of CD8+T cells, effector cell senescence was the most significant. BM-MSCs co-culture had no significant effect on senescent effector cells, and mainly alleviated the senescence of CD8+T cells by delaying the senescence of naive T cells(P＜0.01,P＜0.001). Conclusions BM-MSCs co-culture can alleviate the replicative senescence phenotype of T cells and has a more significant anti-senescence effect on CD8+T cells by inhibiting the initial senescence of T cells as a major mechcanism.

Objective Modifying 3D tubular DNA origami nanostructures (DONs) onto the mesenchymal stem cells (MSCs) membrane surface to obtain DONs@MSCs. Utilizing the characteristics of MSCs homing lung tissue to achieve targeted delivery of DONs to the lungs of mice. Methods The computer-aided tools for DNA sequence were used to design and obtain obtain pre-designed DONs. The morphology of assembled DONs was characterized by atomic force microscopy(AFM), and the upload of functional groups was verified by agarose gel electrophoresis (AGE). Fluorescent labeled single-strand DNA(ssDNA) was modified on the surface of MSCs using biological orthometabolic sugar engineering. According to the principle of complementary base pairing, DONs were modified on the MSCs surface. Using confocal microscopy and flow cytometry to detect the ssDNA and DONs, and using in vivo imaging to observe the distribution of DONs in lung tissue. Results DONs had been successfully assembled. MSCs membrane surface was successfully modified with ssDNA, and DONs were successfully hybridized to obtain DONs@MSCs. Compared to DONs, the enrichment of DONs@MSCs in the lungs of mice was significantly enhanced. Conclusions DONs@MSCs can be used for DONs targeted delivery to lung tissue of mice. This method provides a new strategy for drug delivery of DONs in lung tissues.

Objective To explore the effect of aryl hydrocarbon receptor(AHR) on the immunomodulatory function of human adipose derived mesenchymal stem cells (MSCs). Methods RT-qPCR and immunofluorescence staining were used to detect the expression levels of indoleamine 2,3-dioxygenase 1 (IDO1), interleukin-4-inducible gene 1 (IL4I1) and AHR in MSCs before and after inflammatory cytokine stimulation; siCtrl, siIDO1, siIL4I1 and siAHR were transfected into MSCs and blocked the AHR signaling pathway; conditioned medium from MSCs was co-incubated with macrophages, RT-qPCR and ELISA were performed to detect the level of IL-6, IL-1β, TNF-α. Results MSCs without cytokine stimulation highly expressed AHR and hardly expressed IDO1 and IL4I1 proteins; IDO1, IL4I1 and AHR were up-regulated after different cytokine stimulation(P＜0.05), the expression of IDO1 and IL4I1 was most significantly up-regulated after stimulation by IFN-γ(P＜0.001), AHR entering the nucleus was increased; down-regulation of AHR expression could inhibit the immunomodulatory effect of MSCs(P＜0.05). Conclusions AHR regulates the immune function of MSCs, downregulates expression of AHR could inhibit the immunomodulatory effects of MSCs.

Irritable bowel syndrome (IBS) is a common digestive psychosomatic diseases, mainly manifested as recurrent abdominal pain and abnormal bowel movements. The intestinal dysfunction is often comorbidized with anxiety, depression and other emotional problems, resulting in increased intestinal burden and even a decrease in the quality of life. The etiology and pathogenesis of IBS are unclear, but it is currently believed that IBS involves abnormal gut-brain interaction caused by a combination of various factors, including visceral hypersensitivity, intestinal immune activation, intestinal infection, intestinal dysbacteriosis, and psychosocial stress. Recent studies have observed the disorders of gut microbiota-regulated kynurinaine metabolism in IBS patients, and found that the metabolite kynurenic acid (KA) has important correlations with inflammatory responses, pain stimuli and mood changes. KA may play a protective role in IBS such as anti-inflammatory and pain relief and thus has the potential to become a new approach for the diagnosis and treatment of IBS. In this work, studies related to KA and IBS are reviewed.

This paper summarized the treatment effect and nursing cooperation of using five-element music therapy to relieve the anxiety and pain of a patient undergoing arteriovenous fistula surgery. Key issues of nursing included preoperative nursing, intraoperative coordination, operation methods and music selection of five-element music therapy, postoperative observation and guidance, etc. The intervention of five-element music effectively alleviated the anxiety and depression of the patient, reduced the pain symptom of the patient, promoted the stability of intraoperative blood pressure and heart rate, improved the patients compliacne with surgery, and played a positive role in reducing complications, accelerating postoperative rehabilitation and improving the quality of life. (本文总结1例运用五行音乐改善动静脉瘘手术患者焦虑及疼痛的护理经验, 包括术前护理、术中配合、五行音乐治疗的操作方法及选曲、术后观察及指导等。五行音乐疗法能有效缓解动静脉瘘手术患者焦虑、抑郁状态, 减轻疼痛, 促进术中血压、心率平稳, 提高患者手术配合度。)

Aging related diseases seriously affect the health of the elderly. Studies on the roles and mechanisms of mesenchymal stem cells (MSCs) in aging related diseases have attracted more and more attention. Besides, a number of relative clinical trials have been carried out in the domestic and overseas. This review summarizes the roles, molecular mechanisms, clinical trials of MSCs and MSCs-derived exosomes in several typical age-related diseases such as osteoporosis, Alzheimer's disease, Parkinson's disease, type 2 diabetes, atherosclerosis, chronic obstruc-tive pulmonary disease and tumor. This review may provide some theoretical reference and advices for future studies of MSCs in aging related diseases.

Mesenchymal stem cells (MSCs) are pluripotent stem cells with self-renewal, multi-directional differentiation and immunoregulatory capabilities. More and more studies have demonstrated the beneficial role of MSCs in the treatment of immune disorders. However, there is heterogeneity in the immunoregulatory capacity of MSCs obtained in vivo or cultured in vitro, and the immunoregulatory capacity of MSCs is also regulated by the microenvironment. This review provides an overview of the mechanisms of the immunosuppressive and immune supportive abilities of MSCs, introduces recent studies on the immunoregulatory mechanisms of MSCs and the plasticity hypothesis of their immunoregulatory abilities, and discusses strategies to enhance the immunoregulatory abilities of MSCs in the cell production stage. This review aims to provide a reference for the clinical translational research of MSCs.

Kidney and triple energizers are inseparable in structure and function. Therefore, it is proposed that “kidney-triple energizers” might be the essence of mesenchymal stem cells (MSCs) system. As a cardinal, this article aims to interpret the theoretical connotation of the mesenchymal stem cell system from the perspective of “the kidney essence is the body and the triple energizers is the function”. It is reflected in the kidney-three energizers system, and guided by the theory of traditional Chinese medicine, the modern technology of MSCs has achieved good results in the application of refractory diseases such as COVID-19, and a preliminary exploration of the integration of Chinese and Western medicine research has been carried out.

Many neurodevelopmental disorders (NDDs) share common learning and behavioural impairments, as well as features such as dysregulation of the oxytocin hormone. Here, we examined DNA methylation (DNAm) in the 1st intron of the oxytocin receptor gene, "OXTR," in patients with autism spectrum (ASD), attention deficit and hyperactivity (ADHD) and obsessive compulsive (OCD) disorders. DNAm of "OXTR" was assessed for cohorts of ASD (blood), ADHD (saliva), OCD (saliva), which uncovered sex-specific DNAm differences compared to neurotypical, tissue-matched controls. Individuals with ASD or ADHD exhibiting extreme DNAm values had lower IQ and more social problems, respectively, than those with DNAm within normative ranges. This suggests that "OXTR" DNAm patterns are altered across NDDs and may be correlated with common clinical outcomes.

Objective To investigate the effect of high-quality nursing for COVID-19 patients in makeshift hospitals. Methods A medical team of doctors and nurses was established to participate in the nursing of 528 COVID-19 patients in makeshift hospitals. A high-quality nursing program including environment evaluation, psychological care, sleep quality improvement, symptomatic care, safety management and diet care. Results There was no death or nosocomial infection case reported. No serious adverse event was reported throughout the study. Conclusion High-quality nursing plays an important role in relief of negative emotion, prevention of cross-infection and promotion of rehabilitation among COVID-19 patients in makeshift hospitals. (目的 探讨优质护理在方舱医院内新型冠状病毒肺炎患者中的应用效果。方法 医疗队援沪期间共参与方舱医院内528例新型冠状病毒肺炎确诊患者的护理工作, 护理内容包括环境评估、心理护理、睡眠护理、症状护理、安全及饮食护理等方面实施优质护理。结果 援沪期间, 舱内患者无死亡, 无院内感染和不良事件发生, 均痊愈出舱。结论 优质护理体现了以人文本的护理理念, 在缓解患者负性情绪、预防交叉感染、促进患者康复方面有着重要作用。)

Ectopic epigenetic mechanisms play important roles in facilitating tumorigenesis. Here, we first demonstrated that ANKDD1A is a functional tumor suppressor gene, especially in the hypoxia microenvironment. ANKDD1A directly interacts with FIH1 and inhibits the transcriptional activity of HIF1α by upregulating FIH1. In addition, ANKDD1A decreases the half-life of HIF1α by upregulating FIH1, decreases glucose uptake and lactate production, inhibits glioblastoma multiforme (GBM) autophagy, and induces apoptosis in GBM cells under hypoxia. Moreover, ANKDD1A is highly frequently methylated in GBM. The tumor-specific methylation of ANKDD1A indicates that it could be used as a potential epigenetic biomarker as well as a possible therapeutic target.

Objective To investigate the effect of psychological nursing measures on mental state of COVID-19 patients in the mobile cabin hospital. Methods A Survey of psychological assessment was conducted to 400 COVID-19 patients who were admitted to the mobile cabin hospital between April 15th, 2022 and May 20th, 2022. The individualized psychological nursing interventions were carried out. Results There were 391(97. 75%) cases of mild anxiety and 9(2. 25%) cases of moderate anxiety. After implementation of psychological nursing interventions, all patients had good compliance with treatment, and no case of adverse events was reported. Conclusion Implementation of early psychological assessment and targeted psychological nursing interventions maybe helpful to improve the treatment compliance and recovery of COVID-19 patients. (目的 探讨心理护理措施在方舱医院内新型冠状病毒肺炎患者中的应用效果。方法 选取2022年4月15日—5月20日方舱医院收治的400例新型冠状病毒肺炎患者, 开展心理评估, 并针对性实施个体化心理护理措施。结果 400例患者中391(97. 75%)例存在轻微焦虑, 9(2. 25%)例患者存在中度焦虑。通过实施个性化心理护理措施, 患者均能保持情绪稳定, 配合治疗, 无不良事件发生。结论 早期评估新型冠状病毒肺炎患者的心理状态, 制定相应的心理护理措施, 有助于提高患者治疗依从性, 促进患者早日康复。)

The aim of this paper is to evaluate the application of gastrointestinal decompression device instead of Vacuum Sealing Drainage (VSD) in the home-based nursing of patients with pressure injury. Totally 30 patients with stage Ⅲ or above pressure injury who received treatment using the gastrointestinal decompression device instead of VSD were selected as the research objects. A home-based nursing plan was carried out to enhance the observation and maintenance of decompression device, drainage liquid and tube. The use of disposable gastrointestinal decompression device instead of VSD can be more convenient in the treatment of stage Ⅲ and above pressure injury. The use of disposable gastrointestinal decompression device is potentially effective to improve wound healing and reduce the nursing operation and economic burden of family members. (本文探讨胃肠减压器在压力性损伤患者居家护理中的应用效果。选取30例Ⅲ期及以上压力性损伤患者为研究对象, 均不具备负压封闭引流条件。医护人员使用一次性胃肠减压器代替负压封闭引流, 在居家换药护理过程中加强封闭持续负压装置、引流液和管路的观察护理。一次性胃肠减压器代替负压封闭引流技术引流治疗Ⅲ期及以上压力性损伤有效, 能够促进伤口愈合, 减轻家属的护理操作及经济负担。)

BackgroundDespite the overwhelming number of investigations on AKT, little is known about lncRNA on AKT regulation, especially in GBM cells.MethodsRNA-binding protein immunoprecipitation assay (RIP) and RNA pulldown were used to confirm the binding of LINC00470 and fused in sarcoma (FUS). Confocal imaging, co-immunoprecipitation (Co-IP) and GST pulldown assays were used to detect the interaction between FUS and AKT. EdU assay, CCK-8 assay, and intracranial xenograft assays were performed to demonstrate the effect of LINC00470 on the malignant phenotype of GBM cells. RT-qPCR and Western blotting were performed to test the effect of LINC00470 on AKT and pAKT.ResultsIn this study, we demonstrated that LINC00470 was a positive regulator for AKT activation in GBM. LINC00470 bound to FUS and AKT to form a ternary complex, anchoring FUS in the cytoplasm to increase AKT activity. Higher pAKT activated by LINC00470 inhibited ubiquitination of HK1, which affected glycolysis, and inhibited cell autophagy. Furthermore, higher LINC00470 expression was associated with GBM tumorigenesis and poor patient prognosis.ConclusionsOur findings revealed a noncanonical AKT activation signaling pathway, i.e., LINC00470 directly interacts with FUS, serving as an AKT activator to promote GBM progression. LINC00470 has an important referential significance to evaluate the prognosis of patients.

With the help of theoretical analysis and numerical simulation, the modal parameters of the two-dimensionally low-order beam elements and high-order beam elements based on the absolute nodal coordinate formulation were studied. The analytical solution of the first-order thickness natural frequency of low-order and high-order beam elements considering the Poisson effect was derived. Based on the generalized characteristic equation, four natural frequencies and natural modes of bending, axial, shear and thickness of the shear beam element based on the absolute nodal coordinate formulation were obtained. Taking a simply supported beam structure as an example, the effects of the polynomial displacement field of the shear beam element on the four natural frequencies and natural modes of bending, axial, shear and thickness were analyzed. The results show that the polynomial displacement field of the shear beam element needs to introduce both the horizontal and vertical high-order terms x~2 and y~2. They mainly affect the bending and axial modal parameters, but have little effect on the shear and thickness modal parameters, and first-order thickness natural frequencies of the lower-order and higher-order beam elements is relatively close to the derived analytical solutions of first-order thickness natural frequencies.
