Tao Xingzhi's rural educational sentiments, exemplified by the group of teachers dedicated to rural reconstruction with Tao Xingzhi at the center, represent a spiritual characteristic that encompasses political ambitions of rescuing the nation through education, educational convictions in rural reconstruction, and a sense of connection with peasants. The feelings are nurtured and sublimated in a practical logic of "cognition-emotion-behavior". Cognition refers to embracing rural transformation as one's own responsibility; emotion involves feeling emotionally connected to giving back to the countryside; and action represents the holistic advancement of rural education practices. The rural educational sentiments are the driving force and a profound emotion that Tao Xingzhi and others nurtured in the process of rural education reconstruction. The practice is not only the action of Tao Xingzhi and others with feelings, but also the way for them to continuously sublimate such emotions. This paper analyzes the connotation and practical value of Tao Xingzhi's rural educational sentiments and explores the practical significance of cultivating such sentiments among teachers in the new era. This paper aims to foster teachers' sentiments towards rural education, with political concern as the foundation, local emotion as the support, and the integration of knowledge and action as the approach.

Objective Exploration of recombinant Newcastle disease virus with stable expression of rabies virus glycoprotein(rL-RVG) to inhibit the proliferation of human lung adenocarcinoma cell lines A549 and PC9. Methods Human lung adenocarcinoma cell lines A549 and PC9 were cultured in vitro, and cells in logarithmic proliferation phase were collected and divided into control group, Newcastle disease virus infection group (NDV), recombinant Newcastle disease virus infection group(rL-RVG),ferroptosis group (Erastin) and recombinant Newcastle disease virus combined with ferroptosis inducer group (rL-RVG+Erastin). After 24 h of incubation, cell morphology was observed by microscope and cell proliferation was detected by CCK-8 method, cell migration was detected by scratch test, lactate dehydrogenase (LDH) release was measured by cytotoxicity assay kit, intracellular reactive oxygen species (ROS) content was detected by flow cytometry, and the expression of ferroptosis related proteins (p53, SLC7A11, GPX4) was detected by Western blot. Results Compared with the control group, the cell counting, cell proliferation and distance of migration were significantly reduced in the NDV group, rL-RVG group and Erastin group(P＜0.001). LDH release and total ROS level were significantly increased (P＜0.01), p53 protein expression was significantly increased(P＜0.001), while SLC7A11 and GPX4 protein expression were significantly decreased (P＜0.001). rL-RVG group had a more pronounced effect as compared with NDV group. rL-RVG+Erastin group had significantly decreased cell number, cell proliferation capacity and distance of migration compared with rl-RVG and Erastin groups(P＜0.05), and LDH release and total ROS level were significantly increased(P＜0.05), p53 protein expression was significantly increased (P＜0.001), while SLC7A11 and GPX4 protein expression were significantly decreased (P＜0.001). Conclusions rL-RVG can exert a similar effect to Erastin to inhibit tumor cell proliferation, and its effect is much stronger than that of NDV. This provides new insights into rL-RVG-induced cell death and highlights the critical role of oncolytic viruses in the treatment of tumors.

In order to further analyze the consumption problem caused by regional photovoltaic power generation, the actual output data of typical photovoltaic power plants in Gansu province was used as an analysis sample to study the output characteristics of regional photovoltaic power generation. The output characteristics of a single photovoltaic power station was considered, and the seasonal and weather characteristics of photovoltaic daytime output was analyzed. Considering the cluster characteristics of photovoltaic power generation output, the volatility, correlation and simultaneity rate among different photovoltaic power plants were analyzed. The research results show that the overall photovoltaic output level in Gansu province is relatively high, but there are certain regional differences. Daily output is cyclical, while fluctuations are random. The output of regional photovoltaic power plants is correlated. Compared with the output of a single photovoltaic power station, the output of regional photovoltaic power plants has been reduced at the same time, and the impact on the grid when connected to the grid is reduced, which can improve the occurrence of “photovoltaic abandonment phenomenon”. The proposed quantitative analysis index for photovoltaic power generation output characteristics provides ideas and methods for regional photovoltaic power consumption analysis, and provides a reference for the formulation of photovoltaic power generation development plans.

Objective To investigate the association between the osteogenic differentiation in the soft tissue lump and the clinicopathological characteristics of osteosarcoma patients. Methods We retrospectively reviewed the clinical data of conventional osteosarcoma patients with soft tissue lumps, including Enneking stages, chemotherapy sensitivity, overall survival and post-metastatic survival time. The ossification level in soft tissue lumps was assessed by imaging and the proportion of osteoid matrix was assessed by pathological examination. Results A total of 189 cases were included in this study. In patients with Enneking IIIB, non-osteoblastic, partially osteoblastic and osteoblastic types accounted for 30.2%, 9.6% and 6.3%, respectively. Non-osteoblastic osteosarcoma patients had a higher rate of initial metastasis (P < 0.05); Chemotherapy efficiency of non-osteoblastic, partially osteoblastic and osteoblastic types were 60.5%, 59.6% and 31.3%, respectively. The osteoblastic osteosarcoma held the worst rate of chemotherapy sensitivity (P < 0.05). The overall survival of non-osteoblastic osteosarcoma was shorter than those of partially osteoblastic and osteoblastic types (P < 0.05). Post-metastatic survival time of osteoblastic osteosarcoma was longer than that of non-osteoblastic osteosarcoma (P=0.078). Conclusion For conventional osteosarcoma, the osteogenesis level in soft tissue lumps is related to the surgical stage, chemotherapy sensitivity and prognosis of tumors, which may provide guidance for the individual decision regarding chemotherapy and surgery timing on patients.

RNA-Puzzles is a collective endeavor dedicated to the advancement and improvement of RNA three-dimensional structure prediction. With agreement from structural biologists, RNA structures are predicted by modeling groups before publication of the experimental structures. We report a large-scale set of predictions by 18 groups for 23 RNA-Puzzles: 4 RNA elements, 2 Aptamers, 4 Viral elements, 5 Ribozymes and 8 Riboswitches. We describe automatic assessment protocols for comparisons between prediction and experiment. Our analyses reveal some critical steps to be overcome to achieve good accuracy in modeling RNA structures: identification of helix-forming pairs and of non-Watson-Crick modules, correct coaxial stacking between helices and avoidance of entanglements. Three of the top four modeling groups in this round also ranked among the top four in the CASP15 contest.

Objective To develop a novel decellularized extracellular matrix (ECM) bioscaffold derived from human cirrhotic liver tissues and construct a 3-dimensional cell culture system using this bioscaffold material. Methods Clinical samples of cirrhotic liver tissues were collected from patients undergoing liver transplantation for hepatic cirrhosis. The cirrhotic liver tissues were cleansed, cut into small tissue blocks (5 mm×5 mm×5 mm), and agitated in deionized water and detergents to obtain decellularized cirrhotic liver scaffold. Human primary human hepatocytes (PHHs) were seeded into the prepared ECM scaffold and cultured continuously for 9 d. Histological studies of the seeded cells were performed to assess the cell engraftment, and qPCR was used to detect the expression of the liver-specific genes in the cultured cells. Results The cirrhotic liver tissue became transparent after decellularization, and HE staining and scanning electron microscopy showed no residual nuclei in the bioscaffold. Residual DNA identification showed that the scaffold material had a residue DNA level less than 10 ng/mg of the tissue. After 9 d of continuous culture in the ECM scaffold, the PHHs showed good engraftment as shown by HE staining; the results of qPCR showed that the cells cultured in this 3D culture system expressed higher levels of liver-specific genes as compared with the cells in conventional 2D culture. Conclusion We successfully construct decellularized ECM bioscaffold derived from human cirrhotic liver tissues and confirmed the feasibility of 3D cell culture using this new bioscaffold material.

In view of problem of autonomous location of inspection robot without GPS in underground coal mine, a method of simultaneous localization and mapping based on lidar was studied. Firstly, the observation model of lidar and prediction model of odometer are established, and the actual problems of robot localization and mapping are transformed into the logical reasoning problems of probabilistic mathematical model. At the same time, the adaptive Monte Carlo localization algorithm is used to estimate the real-time pose of the robot,the resampling method based on particle weight(maps matching degree) is proposed to remove particles with small weight, accurate representation of posterior probability distribution of robot posture with fewer and better particles is realized, requirement of using sensors to realize the real-time positioning of robots on raster maps is met. Fast-SLAM algorithm is optimized to reduce the number of particles, and mitigate particle dissipation,so as to improve accuracy of mapping. The experimental results show that the method effectively solves the problem of real-time pose estimation and environment mapping of inspection robot, and improves the self-adaptability of robot localization and accuracy of mapping combining with adaptive Monte Carlo localization algorithm and optimized Fast-SLAM algorithm.

BACKGROUND: Cancer health research relies on large-scale cohorts to derive generalizable results for different populations. While traditional epidemiological cohorts often use costly random sampling or self-motivated, preselected groups, a shift toward health system-based cohorts has emerged. However, such cohorts depend on participants remaining within a single system. Recent consumer engagement models using smartphone-based communication, driving projects, and social media have begun to upend these paradigms. METHODS: We initiated the Healthy Oregon Project (HOP) to support basic and clinical cancer research. HOP study employs a novel, cost-effective remote recruitment approach to effectively establish a large-scale cohort for population-based studies. The recruitment leverages the unique email account, the HOP website, and social media platforms to direct smartphone users to the study app, which facilitates saliva sample collection and survey administration. Monthly newsletters further facilitate engagement and outreach to broader communities. RESULTS: By the end of 2022, the HOP has enrolled approximately 35,000 participants aged 18-100 years (median = 44.2 years), comprising more than 1% of the Oregon adult population. Among those who have app access, ∼87% provided consent to genetic screening. The HOP monthly email newsletters have an average open rate of 38%. Efforts continue to be made to improve survey response rates. CONCLUSION: This study underscores the efficacy of remote recruitment approaches in establishing large-scale cohorts for population-based cancer studies. The implementation of the study facilitates the collection of extensive survey and biological data into a repository that can be broadly shared and supports collaborative clinical and translational research.

The recent discovery of SPINDLY (SPY)-catalyzed protein O-fucosylation revealed a novel mechanism for regulating nucleocytoplasmic protein functions in plants. Genetic evidence indicates the important roles of SPY in diverse developmental and physiological processes. However, the upstream signal controlling SPY activity and the downstream substrate proteins O-fucosylated by SPY remain largely unknown. Here, we demonstrated that SPY mediates sugar-dependent growth in Arabidopsis (Arabidopsis thaliana). We further identified hundreds of O-fucosylated proteins using lectin affinity chromatography followed by mass spectrometry. All the O-fucosylation events quantified in our proteomic analyses were undetectable or dramatically decreased in the spy mutants, and thus likely catalyzed by SPY. The O-fucosylome includes mostly nuclear and cytosolic proteins. Many O-fucosylated proteins function in essential cellular processes, phytohormone signaling, and developmental programs, consistent with the genetic functions of SPY. The O-fucosylome also includes many proteins modified by O-linked N-acetylglucosamine (O-GlcNAc) and by phosphorylation downstream of the target of rapamycin (TOR) kinase, revealing the convergence of these nutrient signaling pathways on key regulatory functions such as post-transcriptional/translational regulation and phytohormone responses. Our study identified numerous targets of SPY/O-fucosylation and potential nodes of crosstalk among sugar/nutrient signaling pathways, enabling future dissection of the signaling network that mediates sugar regulation of plant growth and development.
