Abstract

This paper presents an investigation of the process factors affecting the surface roughness in ultra-precision diamond turning with ultrasonic vibration. Stainless steel was turned by diamond tools with ultrasonic vibration applied in the feed direction with an auto-resonant control system. Surface roughness was measured and compared along with the change of the cutting parameters. The relation curves between the cutting parameters and surface roughness were achieved by comparing the experimental results with different cutting speeds, feed rates, cutting depths. Experimental results indicate that cutting parameters have an obvious effect on the surface roughness. The conclusions are draw in given conditions, the smaller amplitude of the vibration, the worse the surface quality and the higher vibrating frequency, the better surface quality, and the deeper the cutting depth and the more the feed rate, the worse the surface quality. Among these parameters, the feed rate was the most important factor on surface quality.

Abstract

SETD2, the histone H3 lysine 36 methyltransferase, previously identified by us, plays an important role in the pathogenesis of hematologic malignancies, but its role in myelodysplastic syndromes (MDSs) has been unclear. In this study, low expression of SETD2correlated with shortened survival in patients with MDS, and the SETD2levels in CD34+bone marrow cells of those patients were increased by decitabine. We knocked out Setd2in NUP98-HOXD13(NHD13) transgenic mice, which phenocopies human MDS, and found that loss of Setd2accelerated the transformation of MDS into acute myeloid leukemia (AML). Loss of Setd2enhanced the ability of NHD13+hematopoietic stem and progenitor cells (HSPCs) to self-renew, with increased symmetric self-renewal division and decreased differentiation and cell death. The growth of MDS-associated leukemia cells was inhibited though increasing the H3K36me3 level by using epigenetic modifying drugs. Furthermore, Setd2deficiency upregulated hematopoietic stem cell signaling and downregulated myeloid differentiation pathways in the NHD13+HSPCs. Our RNA-seq and chromatin immunoprecipitation–seq analysis indicated that S100a9, the S100 calcium-binding protein, is a target gene of Setd2and that the addition of recombinant S100a9 weakens the effect of Setd2deficiency in the NHD13+HSPCs. In contrast, downregulation of S100a9leads to decreases of its downstream targets, including Ikbaand Jnk, which influence the self-renewal and differentiation of HSPCs. Therefore, our results demonstrated that SETD2deficiency predicts poor prognosis in MDS and promotes the transformation of MDS into AML, which provides a potential therapeutic target for MDS-associated acute leukemia.

Abstract

When using diamond tool to cut the special stainless steel, the influence of cutting parameters on machining surface roughness is so great. Experiments of cutting special stainless steel by ultrasonic vibration were taken to develop the law that cutting parameters influence on surface roughness of the work piece. The relation curves between cutting parameters and the work piece surface roughness were achieved by contrast experiments with different cutting speed, tool feeds, cutting depth and vibration parameters. The result shows that cutting parameters have an obvious effect on surface roughness. Under given Conditions, the smaller amplitude of the vibration, the worse the surface roughness, and the higher vibrating frequency, the better surface roughness, and the more cutting depth and feeds, the worse surface roughness. Among these parameters, tool feeds was main effect on the surface roughness.

Abstract

To meet the accuracy and efficiency requirements in motion estimation, an adaptive diversity rapid search algorithm based on variable size block is proposed in this paper. Two kinds of search algorithms, diamond search and three-step search, are applied to different sub-blocks of the same macro block. The disadvantages of single search strategy are overcome by the algorithm diversity and the similarity of the sub-blocks, which reduce search points obviously. This paper also analyzes and researches XVID, which is one of the open source software of MPEG-4, and gives the optimization design of transporting XVID into ADSP-BF533 processor to achieve real time MPEG-4 encoding. The experimental results show that the MPEG-4 coder implemented in this paper can reach the requirement of real time coding.

Abstract

Interference will possibly happen between the tools and the surfaces to be machined while cutting the unitary wave-front correctors with single linear blade diamond tools. According to the mathematic model of the work piece surface and the tool path, the mathematical model is established to show the minimum setting angle while there is no interference between the tools and the surfaces to be machined. Also, the maximum setting angle meeting the ordered surface roughness is deduced based on the relationship between the microcosmic unevenness height of optical surfaces and the parameters of cutting edges and the setting angle. Practical cutting experiments are performed on an ultra-precision lathe. The results of experiments indicate that it is feasible to machine unitary wave-front correctors by linear blade nature diamond tool. The mathematics model of the minimum setting angle between the linear blade diamond tool and work piece's surface is practical when machining unitary wave-front correctors.

33. Setd2deficiency impairs hematopoietic stem cell self-renewal and causes malignant transformation

Abstract

The histone H3 lysine 36 methyltransferase SETD2 is frequently mutated in various cancers, including leukemia. However, there has not been any functional model to show the contribution of SETD2 in hematopoiesis or the causal role of SETD2 mutation in tumorigenesis. In this study, using a conditional Setd2knockout mouse model, we show that Setd2deficiency skews hematopoietic differentiation and reduces the number of multipotent progenitors; although the number of phenotypic hematopoietic stem cells (HSCs) in Setd2-deleted mice is unchanged, functional assays, including serial BM transplantation, reveal that the self-renewal and competitiveness of HSCs are impaired. Intriguingly, Setd2-deleted HSCs, through a latency period, can acquire abilities to overcome the growth disadvantage and eventually give rise to hematopoietic malignancy characteristic of myelodysplastic syndrome. Gene expression profile of Setd2-deleted hematopoietic stem/progenitor cells (HSPCs) partially resembles that of Dnmt3a/Tet2double knockout HSPCs, showing activation of the erythroid transcription factor Klf1-related pathway, which plays an important role in hematopoietic malignant transformation. Setd2deficiency also induces DNA replication stress in HSCs, as reflected by an activated E2F gene regulatory network and repressed expression of the ribonucleotide reductase subunit Rrm2b, which results in proliferation and cell cycle abnormalities and genomic instability, allowing accumulation of secondary mutation(s) that synergistically contributes to tumorigenesis. Thus, our results demonstrate that Setd2 is required for HSC self-renewal, and provide evidence supporting the causal role of Setd2deficiency in tumorigenesis. The underlying mechanism shall advance our understanding of epigenetic regulation of cancer and provide potential new therapeutic targets.
