Abstract Background Core stability exercise (CSE) is a globally acknowledged intervention for managing chronic low back pain. However, the sustained adherence of patients with chronic low back pain to CSE can be challenging, mainly due to the absence of supervision and guidance from physical therapists during home-based exercise sessions. Consequently, exercise compliance tends to decline, resulting in suboptimal long-term effectiveness of the intervention. In this trial, our primary aim is to evaluate the potential therapeutic equivalence between home-based rehabilitation training employing key point identification technology and exercise guidance administered in a hospital setting. Methods In this trial, we will randomly assign 104 adults with chronic low back pain (CLBP) to either an intervention or control group, with 52 participants in each group. Both interventions will consist of three weekly 0.5-h sessions of core stability exercise (CSE). The intervention group will engage in home rehabilitation training utilizing key identification technology for movement, while the control group will perform supervised exercises in a hospital setting. Outcome assessments will be conducted at 4 weeks and 16 weeks after randomization. The primary outcome measure will be the change in pain intensity based on numeric rating scale (NRS scores) from baseline to 4 weeks. Secondary outcomes will include changes in physical function (measured by the Oswestry Disability Index (ODI)) and lumbar spine mobility as well as activity participation and treatment satisfaction. Discussion If home-based rehabilitation method is demonstrated to be non-inferior or even superior to traditional face-to-face exercise guidance, it could significantly advance the adoption of digital medical care and contribute to improving the overall health of the population. The Leaf Area Index (LAI) is a critical parameter that sheds light on the composition and function of forest ecosystems. Its efficient and rapid measurement is essential for simulating and estimating ecological activities such as vegetation productivity, water cycle, and carbon balance. In this study, we propose to combine high-resolution GF-6 2 m satellite images with the LESS three-dimensional RTM and employ different machine learning algorithms, including Random Forest, BP Neural Network, and XGBoost, to achieve LAI inversion for forest stands. By reconstructing real forest stand scenarios in the LESS model, we simulated reflectance data in blue, green, red, and near-infrared bands, as well as LAI data, and fused some real data as inputs to train the machine learning models. Subsequently, we used the remaining measured LAI data for validation and prediction to achieve LAI inversion. Among the three machine learning algorithms, Random Forest gave the highest performance, with an R2 of 0.6164 and an RMSE of 0.4109, while the BP Neural Network performed inefficiently (R2 = 0.4022, RMSE = 0.5407). Therefore, we ultimately employed the Random Forest algorithm to perform LAI inversion and generated LAI inversion spatial distribution maps, achieving an innovative, efficient, and reliable method for forest stand LAI inversion. The transcribed ultraconserved regions (T-UCRs) encode long non-coding RNAs implicated in human carcinogenesis. Their mechanisms of action and the factors regulating their expression in cancers are poorly understood. Here we show that high expression of uc.339 correlates with lower survival in 210 non-small cell lung cancer (NSCLC) patients. We provide evidence from cell lines and primary samples that TP53 directly regulates uc.339. We find that transcribed uc.339 is upregulated in archival NSCLC samples, functioning as a decoy RNA for miR-339-3p, -663b-3p, and -95-5p. As a result, Cyclin E2, a direct target of all these microRNAs is upregulated, promoting cancer growth and migration. Finally, we find that modulation of uc.339 affects microRNA expression. However, overexpression or downregulation of these microRNAs causes no significant variations in uc.339 levels, suggesting a type of interaction for uc.339 that we call "entrapping". Ovarian cancer presents as an aggressive, advanced stage cancer with widespread metastases that depend primarily on multicellular spheroids in the peritoneal fluid. To identify new druggable pathways related to metastatic progression and spheroid formation, we integrated microRNA and mRNA sequencing data from 293 tumors from The Cancer Genome Atlas (TCGA) ovarian cancer cohort. We identified miR-509-3p as a clinically significant microRNA that is more abundant in patients with favorable survival in both the TCGA cohort (P = 2.3E-3), and, by in situ hybridization (ISH), in an independent cohort of 157 tumors (P < 1.0E-3). We found that miR-509-3p attenuated migration and disrupted multi-cellular spheroids in HEYA8, OVCAR8, SKOV3, OVCAR3, OVCAR4 and OVCAR5 cell lines. Consistent with disrupted spheroid formation, in TCGA data miR-509-3p's most strongly anti-correlated predicted targets were enriched in components of the extracellular matrix (ECM). We validated the Hippo pathway effector YAP1 as a direct miR-509-3p target. We showed that siRNA to YAP1 replicated 90% of miR-509-3p-mediated migration attenuation in OVCAR8, which contained high levels of YAP1 protein, but not in the other cell lines, in which levels of this protein were moderate to low. Our data suggest that the miR-509-3p/YAP1 axis may be a new druggable target in cancers with high YAP1, and we propose that therapeutically targeting the miR-509-3p/YAP1/ECM axis may disrupt early steps in multi-cellular spheroid formation, and so inhibit metastasis in epithelial ovarian cancer and potentially in other cancers.

Despite growing recognition of nature-based solutions (NBS), there remains a research gap in understanding their implementation in urban areas, which poses a significant challenge for urban forest development. Therefore, our paper aims to explore the intersection of NBS with urban forests (UF), identify current barriers, propose strategies to maximize the potential of urban forests as nature-based solutions (UF-NBS) in effectively improving the resilience of urban forests, and enhance the service capacity of urban forest ecosystems. To achieve our objective, we conducted a comprehensive analysis that included a bibliometric review to summarize the evolution of the UF-NBS literature and classify UF-NBS types for the first time. Subsequently, we identified and organized current challenges faced by UF-NBS. Additionally, we proposed an original technological framework system for urban forest development based on NBS principles. The results show the significance of UF-NBS for enhancing urban resilience and human wellbeing, with multiple successful implementations in both China and Europe, validating their effectiveness. However, the implementation of UF-NBS faces several challenges, including inadequate financing, the gap between scientific knowledge and practical implementation, the absence of region-specific information, and the need for interdisciplinary collaboration. This study contributes to establishing a scientific theoretical basis for integrating UF and NBS and provides a systematic approach for decision-makers in urban forest management. Future research should focus on exploring the integration of UF within the NBS framework and prioritize knowledge sharing, international cooperation, and education initiatives to promote the global adoption of UF-NBS and address pressing urban challenges. Dioecious plants are often used in landscaping because of sex differences in individual appearance, resistance and ornamental value. Although a large number of studies have investigated the overall differences in resource allocation between different sexes in dioecious plants, the effects of environmental factors such as temperature and precipitation on resource allocation in sex-dimorphic plants are not fully understood. In order to explore the influencing factors, 39 works of literature on dioecious plant resource allocation published in English databases from 1992 to 2023 were selected, and the biomass data of dioecious plant stems, leaves, flowers and fruits from these pieces of literature were extracted. A total of 545 independent experimental groups were obtained, which were divided into four groups for analysis, and the data were analyzed using METAWIN 2.1 software. Four sets of data were used to quantitatively study the effects of different temperatures, precipitations and life forms on the resource allocation of dioecious plants of different sexes in large-scale space. The results showed that female plants invested more resources in reproductive growth and less resources in vegetative growth. In terms of total biomass, the average biomass of female plants was 3.09% higher than that of male plants, indicating that female plants reduced nutrient investment to compensate for reproductive investment in the process of resource allocation. Temperature and precipitation significantly affect the adaptability of male and female plants to environmental changes and the cooperative relationships among the stressed components. The vegetative biomass investment of female plants showed a positive correlation with the increase in temperature, while the reproductive biomass showed a negative correlation with the decrease. The average annual precipitation had little effect on the vegetative biomass of dioecious plants, but had a significant effect on reproductive biomass. The study of this trade-off relationship is helpful in revealing the relationship between vegetative growth and reproductive growth of plants, exploring the countermeasures of plant life history, and providing a scientific basis for urban landscaping and urban forest management. A synthetic lethality-based strategy has been developed to identify therapeutic targets in cancer harboring tumor-suppressor gene mutations, as exemplified by the effectiveness of poly ADP-ribose polymerase (PARP) inhibitors in BRCA1/2-mutated tumors. However, many synthetic lethal interactors are less reliable due to the fact that such genes usually do not perform fundamental or indispensable functions in the cell. Here, we developed an approach to identifying the 'essential lethality' arising from these mutated/deleted essential genes, which are largely tolerated in cancer cells due to genetic redundancy. We uncovered the cohesion subunit SA1 as a putative synthetic-essential target in cancers carrying inactivating mutations of its paralog, SA2. In SA2-deficient Ewing sarcoma and bladder cancer, further depletion of SA1 profoundly and specifically suppressed cancer cell proliferation, survival, and tumorigenic potential. Mechanistically, inhibition of SA1 in the SA2-mutated cells led to premature chromatid separation, dramatic extension of mitotic duration, and consequently, lethal failure of cell division. More importantly, depletion of SA1 rendered those SA2-mutated cells more susceptible to DNA damage, especially double-strand breaks (DSBs), due to reduced functionality of DNA repair.
