Objective To evaluate the effect of miR-140 on the sensitivity of imatinib(IM)-resistant chronic myeloid leukemia (CML) cell strain KBM5R to IM and potential underlying mechanism. Methods The expression level of miR-140 and Bcl-2 in CML cells KBM5 and IM resistant CML cells KBM5R were detected by RT-qPCR and Western blot, respectively. KBM5R cells were transfected with miR-NC or miR-140 mimic, and then incubated with 25-100 nmol/L IM for 24 h. Cell viability was examined by CCK-8 assay. KBM5R cells transfected with miR-NC or miR-140 mimic were examined with 100 nmol/L IM for 24 h then cell apoptosis was detected by flow cytometry; Mitochondrial membrane potential was detected by JC-1 staining, and cleaved caspase-3 expression was detected by Western blot.The targeting relation between miR-140 and Bcl-2 was verified by bioinformatics and fluorescence activity analysis. Results Compared with KBM5 cells, the expression of miR-140 in KBM5R cells was significantly decreased (P＜0.01) and the expression of Bcl-2 was significantly increased(P＜0.001). In the presence of IM, compared with the miR-NC group, the cell viability in miR-140 mimic group was significantly decreased (P＜0.01 or P＜0.001); Cell apoptosis was significantly increased(P＜0.001); Mitochondrial membrane potential was significantly decreased (P＜0.001), and cleaved caspase-3 expression was significantly increased(P＜0.001). Bcl-2 was a target of miR-140. Conclusions miR-140 may increase the sensitivity of KBM5R cells to IM by inhibiting the target gene Bcl-2.

The mitochondrial proteins translocation and degradation coordinate to maintain the proteostasis network of mitochondria. Their dysfunction is one of the pathogenesis mechanims of metabolic diseases. The dysfunction of transportation and degradation prevents the nuclear-encoded mitochondrial protein from importing and cells from clearing abnormal proteins in time, which ultimately leads to mitochondrial damage. In the diabetic cardiomyocyte, decreased activity of electron transport chain (ETC), increased production of reactive oxygen species (ROS) and decreased energy productivity caused by disorders of mitochondrial transportation and degradation are main reasons for the development of diabetic cardiomyopathy (DCM).

This review reported lipid mediators associated with inflammatory responses in adipose tissue, pro-inflammatory mediators: eicosane compounds derived from cyclooxy-genase (e.g. prostaglandins); Pro-resolving mediators: lipoxins produced by omega-6 polyunsaturated fatty acids, and resolvins produced by omega-3 polyunsaturated fatty acids. The treatment of inflammatory responses should not be limited to the use of acute cascade antagonists or inhibitors, but should be expanded to take into account the great therapeutic potential of inducers or agonists in the inflammatory resolution phase.

Due to rapid urbanization, population growth, industrialization, and agricultural activities, there is an increasing demand for freshwater resources, leading to heightened pressure on watershed ecosystems. This study focused on the Nishan Reservoir, Qufu Sihe River, and Qufu Yihe River, conducting field investigations on these water bodies during the spring of 2021 and 2022. Water samples were collected and analyzed for key water quality indicators, including chemical oxygen demand (COD), biochemical oxygen demand (BOD), total nitrogen (TN), total phosphorus (TP), ammonia nitrogen (AN), chlorophyll A, and algal cell density. This study unveiled notable correlations among various water quality parameters, including positive associations between BOD and COD, chlorophyll A and algal cell density, and inverse relationships with total phosphorus. Moreover, significant positive correlations were identified between total nitrogen and ammonia nitrogen, as well as between ammonia nitrogen and chlorophyll A. The study highlighted that the TP concentration surpassed the threshold of 0.20 mg/L in 2021, potentially exacerbating the proliferation of algae, leading to algal blooms and adversely affecting the aquatic ecosystem. This study emphasizes the significance of broadening the geographical scope and utilizing long-term datasets to discern trends, determinants, and management approaches pertinent to water quality. Furthermore, this study underscores the imperative of investigating the influence of nitrogen-to-phosphorus nutrient ratios on the composition and proliferation of algal populations, while also taking into account the potential impact of additional factors like light availability, temperature, and water flow on the dynamics of algal communities. Objective: Obesity can affect periodontal tissues and exacerbate periodontitis. Pyroptosis, a newly identified type of inflammatory cell death, is involved in the development of periodontal inflammation. The saturated fatty acid palmitic acid (PA) is elevated in obese patients. The effect of PA on pyroptosis in periodontal ligament cells (PDLCs) and its underlying mechanisms remain unknown. Materials and Methods: Human PDLCs were isolated from healthy individuals and cultured for experiments. The effects of PA on PDLC pyroptosis and the underlying mechanisms were examined by transmission electron microscopy, quantitative real‐time PCR and western blotting. Results: The morphology of PDLCs in the PA group indicated pyroptotic characteristics, including swollen cells, plasma membrane rupture and changes in subcellular organelles. PA induced inflammatory responses in PDLCs, as indicated by an increase in IL‐1β in the cell culture supernatant. Furthermore, we found that the pyroptosis‐related proteins caspase‐1, caspase‐4 and GSDMD were involved in PA‐induced cell death. GSDMD and caspase‐4 inhibitors alleviated pyroptotic death of PDLCs. Moreover, PA promoted NF‐κB P65 phosphorylation. A NF‐κB inhibitor decreased IL‐1β expression and partly rescued cell death induced by PA. Conclusion: PA activated the NF‐κB pathway and induced the inflammatory response in PDLCs. Caspase‐4/GSDMD mediated PDLC pyroptosis induced by PA. [ABSTRACT FROM AUTHOR]

The major cause of morbidity and mortality of diabetes is cardiovascular disease, which is exacerbated by hypertension. Diabetes and hypertension are closely interlinked because of similar risk factors, and there is also substantial overlap in the cardiovascular complications of diabetes and hypertension. Controlling comorbidities, especially hypertension, is important in reducing the vascular complications of diabetes.

The collaborative recommendation algorithm based on the Variational Autoencoder (VAE) can help solve the sparsity problem in the recommendation algorithm, but the VAE model's prior is a single Gaussian distribution, which makes the expression tends to be simple and average, and suffers from the problem of underfitting. The Gaussian Mixture Variational Autoencoder (GMVAE) model has a more complex prior, which is more adaptable and effective for nonlinear tasks compared to the original VAE model, and has been widely used for unsupervised clustering and semi-supervised learning. Inspired by this, this paper investigates a collaborative filtering algorithm based on the GMVAE model. In this paper, the authors design experiments based on the Cornac recommender system comparison framework, and use the improved GMVAE for the collaborative recommendation task, the user-item matrix regenerated by the generative model is used for recommendation task. Deep features are extracted with one hidden layer in the inference model and one layer in the generation model to increase model robustness, and an early stop strategy is used to reduce overfitting. In this paper, experiments are conducted on multiple public datasets to compare with other recommendation algorithms in terms of NDCG and recall metrics. The experiments demonstrate that the improved collaborative filtering algorithm based on a GMVAE model performs well in the recommendation task. [ABSTRACT FROM AUTHOR]

When Inductively Coupled Plasma-Optical Emission Spectrometry (ICP-OES) is used to analyze the sulfur content of high-sulfur (S≤8%) bauxite, commonly used acid dissolving and alkali fusion methods cannot oxidize sulfur completely, leading to a false (lower) result of sulfur due to multiple valences of sulfur (-2, +4, +6). In this study, high-sulfur bauxite samples were fused with sodium hydroxide and leached out by hot water and hydrochloric acid, and the calibration curve was plotted through a matrix matching method to compensate for the spectral interferences of aluminum and sodium on sulfur. S182.034 nm (184 nm) was selected as the analytical line, and sulfur was analyzed by ICP-OES. Results show that sulfur in high-sulfur bauxite samples is completely oxidized with 3 g sodium peroxide at 700℃ for 10 min. The linear correlation coefficient of the calibration curve was 0.9999, the detection limit was 0.025 μg/mL, and the relative standard deviation (RSD, n=6) was less than 5%. Compared with the results of carbon sulfur analysis, there are no significant differences between the two methods. This method has the advantage of completely decomposing the samples without any loss, which will lay the foundations for the simultaneous determination of sulfur and other elements in high-sulfur bauxite by ICP-OES.
