Abstract

A strategy for the synthesis of iodo-substituted allylic pyrazoles through NIS-mediated regioselective addition of allenamides with pyrazoles is presented. This reaction proceeds rapidly, is scalable up to a gram scale, and tolerates a broad scope of substrates. Moreover, using 1,2,4-triazole as the nucleophile, the corresponding 1,2-addition product was obtained in good yield. Significantly, the products of this reaction, iodo-substituted allylic pyrazoles, could undergo coupling with phenylacetylene under Sonogashira cross-coupling conditions. [ABSTRACT FROM AUTHOR]

23. Microneedle-mediated polydeoxyribonucleotide nanoliposomes for skin penetration performance and anti-aging efficacy.

Abstract

The skin penet rat ion proper t ies and skin ant i-aging eff icacy of microneedle-mediated t ransdermal delivery of polydeoxyribonucleotide nanoliposomes (PDRN-NLPs) were investigated. The particle size of PDRN-NLPs was (50.1 ± 0.2) nm, the PDI was 0.267 ± 0.008, and the Zeta potential was (-30.2 ± 0.8) mV. The nanoliposomes, mediated by the microneedles, could effectively facilitate the rapid penetration of active ingredients into the skin. Cellular anti-aging efficacy studies showed that PDRN-NLPs significantly increased SOD activity, significantly increased GSH content and significantly decreased MDA level compared with the free component solution (P < 0.05 or P < 0.01). 31. Association between the Genetic Polymorphisms of CCL2, CCL5, CCL8, CCR2, and CCR5 with Chronic Hepatitis C Virus Infection in the Chinese Han Population.

Abstract

Hepatitis C virus (HCV) infection is a global public health burden. Chronic HCV infection leads to the development of fibrosis, cirrhosis, liver cancer, and liver failure over time. A total of 250 patients with chronic HCV infection and 299 healthy blood donors were recruited. Sixteen candidate single nucleotide polymorphisms (SNPs) in chemokine (C-C motif) ligand 2 (CCL2), CCL5, CCL8, C-C chemokine receptor 2 (CCR2), and CCR5 were genotyped in all participants. The rs1024610 AA genotype was significantly associated with decreased susceptibility to chronic HCV infection. Aspartate aminotransferase (AST) levels, AST/platelet ratio index, and the fibrosis 4 score were significantly lower in the CCL2 rs1024610 T allele and haplotype ATGC carriers. Moreover, expression levels of collagen IV (C–IV) and laminin (LN) were significantly higher in patients with the CCL5 rs2280788 C allele compared to the non-carriers. Similarly, the expression levels of C–IV, LN, and hyaluronic acid were significantly higher in patients with the CCL5 haplotype, TGCT. No significant differences were identified between the SNPs/haplotypes and plasma levels of CCL2, CCL5, CCL8, CCR2, and CCR5 in the healthy controls, and the rs1024610 allele alteration had no effect on CCL2 promoter activity. This is the first study to report an association between CCL2 rs1024610 and the risk of chronic HCV infection in the Chinese Han population. rs1024610 and ATGC haplotype in CCL2 were reasonable candidate markers of liver abnormalities. rs2280788 and TGCT haplotype in CCL5 are likely to play a significant role in liver fibrosis during chronic HCV infection. Abstract

Ischemic stroke caused by arterial occlusion is the most common type of stroke, which is among the most frequent causes of disability and death worldwide. Current treatment approaches involve achieving rapid reperfusion either pharmacologically or surgically, both of which are time‐sensitive; moreover, blood flow recanalization often causes ischemia/reperfusion injury. However, even though neuroprotective intervention is urgently needed in the event of stroke, the exact mechanisms of neuronal death during ischemic stroke are still unclear, and consequently, the capacity for drug development has remained limited. Multiple cell death pathways are implicated in the pathogenesis of ischemic stroke. Here, we have reviewed these potential neuronal death pathways, including intrinsic and extrinsic apoptosis, necroptosis, autophagy, ferroptosis, parthanatos, phagoptosis, and pyroptosis. We have also reviewed the latest results of pharmacological studies on ischemic stroke and summarized emerging drug targets with a focus on clinical trials. These observations may help to further understand the pathological events in ischemic stroke and bridge the gap between basic and translational research to reveal novel neuroprotective interventions. 46. Transition-metal-free three-component acetalation-pyridylation of alkenes viaphotoredox catalysis

Abstract

A general transition-metal-free photoinduced acetalation-pyridylation of alkenes using diethoxyacetic acid and cyanopyridine was developed under mild conditions. By employing 4CzIPN as the photocatalyst and Cs2CO3as the base, a diverse range of styrene derivatives and cyanopyridines worked well to give the desired products. The versatility of this method is highlighted by its application in the construction of various functional groups and the late-stage modification of drugs. Importantly, some of the synthesized compounds showed good in vitroantitumor activity, indicating that this protocol is of significance and potential for antitumor drug development.

50. Genome-wide identification and expression analysis of Argonautegene family from longan embryogenic callus

Abstract

Argonaute (AGO) proteins are the core of the RNA-induced gene silencing complex which regulate a wide variety of processes in plants, from organ development to abiotic stress responses. They have been identified in many plants, but little is known in longan (Dimocarpus longanLour.), and how AGOfunctions in the signaling pathways in plant embryos in response to changing environmental stimuli remains unclear. In the present research, a genome-wide analysis of the AGOgene family members and their roles in somatic embryogenesis (SE), zygotic embryogenesis (ZE), tissue developmental processes, and responses to hormones, light and abiotic stress in longan were conducted. Ten longan AGOgenes were identified genome-wide and divided into four clades. They were distributed on chromosomes 1, 4, 8, 10, 12, 13, 14, and 15, and had 2–23 introns. The expression profiling implied that DIAGOsregulated early and middle embryogenesis, as well as developmental processes of seed, flower, and stem in longan. In addition, the transcript levels of DIAGOsin response to exogenous hormones, light and abiotic stress showed differences in expression patterns. These results provide the useful information for further elucidation of RNAi-mediated gene silencing in longan embryogenic callus (EC).
