During storage, winter jujube often experiences physiological disorders that can result in postharvest quality deterioration. To address this concern, a combination of 1% chitosan (CTS) and varying concentrations (0.02, 0.2, and 2 mg mL-1) of ε-polylysine (ε-PL) was applied. The findings indicated that the treatment involving the combination of 1% CTS with 0.2 mg mL-1ε-PL exhibited superior efficacy in preserving the hardness and titratable acid content of winter jujube. Additionally, the jujube fruits of this treatment exhibited a deceleration in the rise of decay incidence, red index, and weight loss rate. Simultaneously, the application of CTS combined with ε-PL treatment improved the levels of antioxidant enzyme activities (SOD, CAT, POD, and APX) and enhanced contents of antioxidant compounds (AsA and GSH). Moreover, the generation of reactive oxygen species (H2O2 and O2-.) was suppressed. Alternaria alternata is the main pathogenic fungus of postharvest black spots in fruits and vegetables. This study aimed to explore the antifungal activity of methionine on A. alternata in vitro and to reveal related antifungal mechanisms through a metabolomics analysis. The results showed that the inhibitory effects of L-methionine (Met) treatment on mycelium growth, spore germination, and the germ tube elongation of A. alternata were enhanced with an increase in the Met concentration, but the inhibitory effects decreased when the Met concentration was higher than 50 mmolL−1. The results of propidium iodide staining and scanning electron microscopy showed that the Met treatment damaged the plasma membrane integrity of the A. alternata spores and caused an irreversible deformation of mycelium. In addition, after the Met treatment, the leakage of electrolytes, nucleic acid, and proteins in the A. alternata cells was significantly higher than that in the control group, indicating that the Met treatment increased the permeability of the cell membranes. Eighty-one different metabolites, divided into seven categories, were identified through the metabolomics analysis, including forty-three downregulated metabolites and thirty-eight upregulated metabolites. Among them, these differential metabolites were mainly involved in amino acid synthesis and metabolism, the pentose phosphate pathway, and the TCA cycle. Therefore, the antifungal effect of the Met treatment on A. alternata was mainly to damage the integrity of the cell membranes, make nucleic acid and protein contents leak, and affect the TCA cycle, carbohydrate metabolism, amino acid synthesis metabolism, and the metabolic pathways associated with cell membrane biosynthesis. Thus, the growth and development of A. alternata were inhibited. The research enriched the investigation of the effect of the antifungal mechanism of Met treatment on A. alternata and provided a theoretical basis for the application of Met to prevent and treat postharvest black spots in fruits and vegetables. In this article, I examine the moral review councils (MRCs) established in China's rural areas since the early 1980s. I show that MRCs create a liminal plebeian public sphere in the context of a civilising offensive that deals with the uncivil behaviours of individuals and disputes between neighbours. In this plebeian public sphere, the MRC incorporates techniques of the Maoist mass meeting, the democratic election, traditional mediation and a pedagogy of exemplars, all of which are depoliticised into purely technical instruments. Their institutional legitimacy comes from organised virtues based on councillors' male seniority and the democratic method of their selection. MRCs, as an instrument of a civilising offensive, are a kind of paternalistic technology, which involves a complex strategy of a hybridity of acts, relationships, thoughts, desires and temptations of village residents in the context of the reform era. The people targeted in this civilising offensive often experience two levels of stigmatisation and their participation determines the effectiveness of the operation of MRCs. [ABSTRACT FROM AUTHOR]

In this article, we advocate the adoption of 'more temporal and processual characters' to understand contemporary community governance in China. We show that communities in China are seen both as producing moral problems and as being the solutions to these problems. Furthermore, we argue that the establishment of the moral clinic provides an alternative to neoliberal ways of self-governance. In the article, we present moral clinics as a new form of community self-governance whose aim is to achieve a complex balance between various conflicts in the context of China's unprecedented urbanisation in the name of governing for and through community harmony. Through examining the establishment of moral clinics, we expose how the relationship between the moral 'hospitalisation' of society and the socialisation of individuals can be understood in new ways. We argue that the institutionalisation of this 'moral work' is a strategy based on old techniques of Chinese traditional medicine that are being enhanced by modern organisational settings. Summary: Ethylene continues to be a major factor influencing quality of European pears during storage. Although research has been done on the effect of pre‐harvest 1‐methylcyclopropene (1‐MCP) on physiological characteristics in pears, a full understanding of cultivar response and antioxidant metabolism remains elusive. Spraying 1‐MCP on 'Bartlett' and 'd'Anjou' pears was studied with respect to physiology, storage and eating quality, disorders, and antioxidant properties at two harvest date (H1 and H2) during storage and ripening. Treatment with 1‐MCP extended the harvest window of 'Bartlett' and 'd'Anjou' pears 3 and 4 days, respectively, without reducing storage or eating quality. Treatment with 1‐MCP reduced ethylene production (EPR) and respiration rates (RR), maintained fruit firmness and green colour during storage and retarded development of desirable melting texture in both cultivars. Additionally, 1‐MCP lowered the incidence of disorders by alleviating membrane lipid peroxidation, retaining high total flavonoids (TF) and antioxidant capacity, and enhancing superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activity in both cultivars. Overall, pre‐harvest 1‐MCP applications could extend storage time of 'Bartlett' and 'd'Anjou' pears to 5 and 6 months, respectively, at −1.1 ± 0.5 °C, by reducing ethylene synthesis and enhancing antioxidant metabolism. Thrombocytopenia is a common hematological abnormality in patients with cirrhotic hypersplenism. Splenectomy with paraesophagogastric devascularization (SPD) is a conventional surgical therapy which can reverse pancytopenia in these patients. Platelets are traditionally recognized for their central role in hemostasis. However, the status of platelet aggregation in chronic hepatitis B patients with cirrhotic hypersplenism before and after SPD has not been reported yet. A total of 41 cirrhotic patients and 31 healthy controls were included in this study. Platelet aggregation was detected by AggRAM® Advanced Modular System (Helena Laboratories, USA). ELISA was used to detect the cytokines closely related to platelet aggregation. Expressions of platelet membrane glycoproteins (GPs) were evaluated by flow cytometric analysis. Platelet aggregation was found to be decreased distinctly in the cirrhotic patients, and to be restored to normal level after SPD. The cirrhotic patients showed higher plasma levels of the cytokines HMGB1, PEDF, vWF, cAMP and cGMP, which also improved partially after SPD. Moreover, the cirrhotic patients had much lower expression of GPIIb/IIIa, GPIbα and P-selectin than either the healthy controls or SPD patients at basal or activated level. Generally, SPD benefits cirrhotic patients with bleeding tendencies by improving platelet counts and aggregation. This paper examines the emergence of the role of "moral doctors" who volunteer in what are called "moral clinics" in Huzhou city. In these moral clinics, the characteristics, experiences, and attributes of older women, in particular, are highly valued and viewed as being essential to the role of the moral doctor. These moral doctors act as moral exemplars and conflict mediators in their local communities. Their moral capital and professionalism, combined with their gender, age, familial and neighborhood attributes, contribute to the accumulation of an affective feminized labor which employs the techniques of care, reason, and moral fortitude to govern the self and others. We unpack these ethical virtues exemplified by moral doctors and nurses in order to show how a female‐centric "ethic of care" can become a set of techniques in governing others. In this paper, we elaborate on the role that these moral doctors perform to support the aims of the moral clinics in terms of fostering pro‐social behavior and moral obligation in local communities. We argue that the performance of this type of "moral work" is both a mechanism of discipline and a process of self‐actualization. We contribute to the current literature on "therapeutic governance" in China by showing how the non‐expert medicalization of social ills by moral doctors is incorporated into the reproduction of social control. In this article, I reflect on the ethical implications of my research visit to China's prisons. I argue that satisfying instituted ethical protocols in this type of research is the beginning of the ethical process, not the end. "Ethical research" involves constant reflection on how the researcher's self is constituted in relation to others, not only the primary research group but also fellow researchers, gatekeepers, facilitators, prison officials, and the representatives of the many organizations involved in such a program. Interpretive or critical inquiries are not solely about deciding "whose side we are on," but more about what kind of relations between the self and various stakeholders existed before, during, and after fieldwork. Due to the existence of viral reservoirs, the rebound of human immunodeficiency virus type 1 (HIV-1) viremia can occur within weeks after discontinuing combined antiretroviral therapy. Immunotherapy could potentially be applied to eradicate reactivated HIV-1 in latently infected CD4+ T lymphocytes. Although the existence of HIV-1-specific CD8+ T memory stem cells (TSCMs) is well established, there are currently no reports regarding methods using CD8+ TSCMs to treat HIV-1 infection. In this study, we quantified peripheral blood antigen-specific CD8+ TSCMs and then expanded HIV-1-specific TSCMs that targeted optimal antigen epitopes (SL9, IL9, and TL9) in the presence of interleukin- (IL-) 21 or IL-15. The suppressive capacity of the expanded CD8+ TSCMs on HIV-1 production was measured by assessing cell-associated viral RNA and performing viral outgrowth assays. We found that the number of unmutated TL9-specific CD8+ TSCMs positively correlated with the abundance of CD4+ T cells and that the expression of IFN-γ was higher in TL9-specific CD8+ TSCMs than that in non-TL9-specific CD8+ TSCMs. Moreover, the antiviral capacities of IL-21-stimulated CD8+ TSCMs exceeded those of conventional CD8+ memory T cells and IL-15-stimulated CD8+ TSCMs. Thus, we demonstrated that IL-21 could efficiently expand HIV-1-specific CD8+ TSCMs to suppress HIV-1 replication. The progress in sample environment at the China Spallation Neutron Source (CSNS) is presented. During the past two years, the Sample Environment group (SE Group) has made substantial efforts towards the procurement, design, development and maintenance of a series of sample environments for the three commissioning instruments. The equipment includes cryostats, an 3He insert, magnets, furnaces, continuously-loaded pressure cells, clamp cells, a gas handling panel, automatic sample changers, etc. The SE Group also constructs and maintains three user laboratories for sample preparation, characterization and complimentary analysis. This paper examines the daily practices of caring for COVID-19 patients in Wuhan, China, in early 2020 and the challenges that nurses faced. The paper shows that the affective contagion, especially among patients, introduced unexpected challenges for nurses in caring for COVID-19-infected patients. Nurses had to contend with the challenges of treating both physical and psychological problems in patients simultaneously. As a result, it was necessary for nurses to adapt to the different rhythm of COVID-19 wards to address these challenges and do so through taking on a range of general and specific nursing tasks and playing a diverse range of roles on the wards, from garbage collector to "psychological counselor." Thus, the paper brings attention to the experiences and demands of providing nursing care in an emergency pandemic context, in particular the necessity of responding to both the physical and the psychological needs of patients. The utilization of natural preservatives presents a promising avenue for mitigating the spoilage of fresh-cut fruits and vegetables induced by microorganisms, enzymatic browning, and water loss. We have developed an innovative method for preserving fresh-cut green peppers using the combined effects of ε-polylysine (ε-PL) and p-Coumaric acid (p-CA). Through concentration screening experiments, we determined that the optimal concentrations of ε-PL and p-CA were 25 mg/L and 10 mg/L, respectively (ε-p-CA). Treatment with ε-p-CA significantly improved the quality of fresh-cut green peppers. It effectively reduced hardness and weight loss, preserving the texture and appearance of the peppers. Furthermore, ε-p-CA treatment delayed the increase in respiratory rate, electrolyte leakage, and ethylene production, thereby maintaining the structural integrity. Meanwhile, ε-p-CA treatment effectively inhibited the malondialdehyde (MDA) content increase and maintained DPPH radical scavenging activity. The microbial analysis demonstrated the ε-p-CA-treated peppers also showed lower total bacterial, mold, and yeast counts, which prolonged the freshness of fresh-cut peppers. In addition, ε-p-CA treatment improved the retention of phenolics and vitamin C without significantly affecting the color and soluble sugar content of green peppers. Irregular ripening is a major deterrent for the purchase of European pears. Cell wall polyuronides and related- modifying enzymes were investigated in two European winter pears, ‘Comice’ and ‘d’Anjou’, during ripening at 20 °C following 5 and 8 months of storage at −1.1 °C. Both pear cultivars developed a melting (buttery-juicy) texture and higher water-soluble polyuronides (WSP) in flesh tissue after 5 months, but the pears’ texture quality decreased and a mealy (coarse-dry) texture presented in ‘Comice’ pear after long-term storage. Activities of pectin methylesterase (PME) and α-arabinofuranosidase (α-ARF) in ‘Comice’ pears was positively correlated with texture development, whereas in ‘d’Anjou’ pears polygalacturonase (PG), PME, cellulase (CEL), and β-galactosidase (β-GAL) were more highly correlated with texture. 1-MCP reduced pear ripening by inhibiting ethylene production and the activities of cell wall-modifying enzymes, maintaining higher antioxidants, and alleviating membrane lipid peroxidation. The controversial duty to promote British values in Schools and Further Education Colleges in England, Scotland and Wales was introduced in the Counter-Terrorism and Security Act 2015. This duty is described by some as the introduction of securitising mechanism into British Schools, for others it is a symptom of the UK Government’s further retreat from Multiculturalism. In this article, we present the duty as the most recent reconstructive intervention designed to remedy alleged failings in the education system to produce liberal British citizens and to counter the growth of extremism. In the article, we will explore how local institutions such as schools and colleges have implemented the duty in a way that defuses some of the securitising aspects of the Statutory Duty through celebrating the UK as a multi-racial, multi-faith and multi-cultural society and through ensuring that their promotion of British values is consistent with their existing value system and ethos which includes the promotion of pupils’ Spiritual, Moral, Social and Cultural Development. Myeloid-derived suppressor cells (MDSCs) have been known to be a key factor in the regulation of the immune system under numerous conditions such as tumors, infections, autoimmune diseases, and transplantations. In contrast to the proposed deleterious role of MDSCs in tumors and infections, MDSCs with their suppressive function are now proved to have the beneficial potential of suppressing the autoimmune response and promoting tolerance to transplantation. Therefore, the expansion of MDSCs could be a promising therapeutic strategy for many diseases. In this study, we aimed to identify FDA-approved drugs that could aid in the expansion of functional MDSCs. We performed a high-throughput screening (HTS) of FDA-approved drugs based on the in vitro human MDSC-differentiation system and identified finasteride (FIN) to have the best potency to aid the generation of human MDSCs. The FIN-induced MDSCs were quite similar to monocytic MDSCs with regard to their surface phenotype, morphology, immunosuppressive function, and related gene expression. Next, we aimed to determine the mechanism of action of FIN and found that FIN induced the expansion of MDSCs through up-regulation of the COX2/PGE2 pathway by enhancing the activity of COX2 promoter. In addition, the administration of indomethacin (IND), a COX2 inhibitor, abrogated the effect of FIN. Based on these results, we suggested that FIN could find applications in the future in the expansion of MDSCs. Abstract: The effect of ultraviolet irradiation combined with chitosan coating on fresh jujube under ambient temperature was investigated. In the first period, the jujubes were irradiated for 4, 6, 8, 10 min at 253.7 nm ultraviolet, or coated with 1%, 1.5%, 2% and 2.5% of chitosan, respectively. In the second period, the jujubes were firstly irradiated 6 min, and then coated with 1.5% of chitosan. The result showed that combined treatment could reduce decay incidence of jujubes and restrain increase of respiration rate, weight loss, malonaldehyde content and electrolyte leakage of jujubes. Furthermore, the activities of superoxide dismutase, peroxidase, and catalase were maintained at higher level, and the decrease of ascorbic acid and chlorophyll was restricted. The present data indicated that treating post-harvest jujubes with ultraviolet irradiation combined with chitosan coating was an effective method, and might be considered in commercial application for preservation of postharvest fruit. Background To effectively extend the shelf life of fresh jujube, carbon monoxide (CO), as a small molecular gas, was applied to fumigate fresh jujube. The quality and antioxidant activity of jujubes fumigated with carbon monoxide at concentrations of 0 (control), 5, 10, 20 and 40 µmol L−1 for 1 h under ambient temperature were investigated. Results The jujube fumigated with 10 µmol L−1 CO showed the best preserving effect amongst all samples. At 30 days, the decay incidence of jujube fumigated with 10 µmol L−1 CO is only two-thirds of that of control sample; its red index and weight loss rate were 22.8% and 19.4% lower, and its firmness, soluble solids content (SSC) and acidity were 18.7%, 5.4% and 12.2% higher than that of control samples, respectively. Its vitamin C and total flavonoid contents were also the highest. However, no significant difference in total polyphenol content was found. The jujubes treated with 10 µmol L−1 CO exhibited the highest antioxidant activity in terms of reducing power and 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl radical scavenging activity. However, the jujubes fumigated with 40 µmol L−1 CO showed inferior characteristics compared with the control sample. Abstract: The effect of carbon monoxide (CO) on enzymatic browning of fresh-cut lotus root slices in relation to phenolic metabolism was investigated. Fresh-cut lotus root slices were fumigated with CO gas at 75, 125, 175 or 225 μL/L for 10, 20, 30 or 40 min, and then stored for 8 days at 5 °C. Changes in color, total phenol, malonaldehyde (MDA), and activities of phenolic-associated enzymes including polyphenol oxidase (PPO), peroxidase (POD) and phenylalanine ammonialyase (PAL) in lotus root slices were measured. The results indicated that fumigating fresh-cut lotus root slices with 175 μL/L of CO for 20 min delayed browning and inhibited increase of MDA. Besides, total phenol also showed higher level from 0 to 4 days. Furthermore, the activities of PPO and POD were inhibited, but PAL activity was increased. The present data suggests that CO has the potential to inhibit browning and maintain quality of fresh-cut lotus root slices. [Copyright &y& Elsevier]

Theeffect of carboxymethyl cellulose and alginate coating combinedwith brewer yeast on postharvest grape preservation was investigated. Thepostharvest grapeswere coatedwith2%of alginate and3%of carboxymethyl cellulose combined with 1.5 × 109 CFU/mL of brewer yeast.The combined treatment samples showed good sensory character on day 13 compared with control samples or only coated samples. The increase of weight loss and decrease of total soluble solids of combined treatment grapes were restrained. Furthermore, the protective enzymes including superoxide dismutase, peroxidase, and catalase of combined treatment sample showed higher activities.Accordingly, the increase ofmalonaldehyde contentwas also restrained andmore vitamin Cwas preserved in combined treatment samples. At day 13, the weight loss rate and the total soluble solids of grape treated with coating + yeast were 23.6% lower and 20.6% higher than those of control samples, respectively. Abstract: The effect of 1% chitosan film with 0.04% nano-silicon dioxide on the qualitative properties of harvested jujube under ambient temperature was investigated. After 32d, the red index, decay incidence, weight loss, and respiration rate of the coated jujubes were lower compared with those of the control. The lower phenylalnine ammonialyase activity and higher activities of scavenger antioxidant enzymes (i.e., superoxide dismutase, peroxidase, and catalase) of the coated jujubes can be attributed to the compound coating. Increased malonaldehyde in the coated jujubes was restrained. Composite coating had shown to be superior in preserving total flavonoid than chitosan coating alone. But no differences were observed in terms of vitamin C loss and total polyphenol content between composite coating and control. Coating jujubes with chitosan+nano-silicon dioxide will be a promising alternative if nano-silicon dioxide is allowed for food use. [Copyright &y& Elsevier]

CO is a gas signal molecule involved in various physiological and metabolic regulation processes in plants. In the present study, we evaluated the effects of CO fumigation on the control of Alternaria rot in jujube fruit and explored the mechanism of action. Results showed that CO at 10 μmol L−1 significantly reduced lesion diameter in the fruit inoculated with A. alternata without showing any antifungal activity in vitro. CO treatment enhanced the activities of four representative resistance-related enzymes (phenylalanine ammonia-lyase, polyphenol oxidase, chitinase, and β-1,3-glucanase). Furthermore, CO also led to increase in the content of phenolics, flavonoids, lignin, and H 2 O 2. These results indicate that CO induces resistance to Alternaria rot primarily by activating disease resistance-related enzymes and enhancing accumulation of antifungal substances, and that CO can be a promising elicitor of plant defense responses to resist Alternaria rot in postharvest jujube fruit. Northern China faces water scarcity, restricting water usage in place across Inner Mongolia's western region. The integrated irrigation and fertilization model for sugar beet is undergoing rapid development and application in production. However, there is a concerning trend in production where the frequency of irrigation and fertilization is being increased blindly, resulting in the wastage of valuable water and fertilizer resources. Limiting water and fertilizer usage is an effective approach to improve sugar beet production efficiency. Sugar beets are a significant sugar crop in China. A split-plot design was employed to examine the impact of reducing water and fertilizer use on sucrose metabolism in sugar beet root. Our study was performed at the Ulanqab Institute of Agricultural and Forestry Sciences in Inner Mongolia from 2022 to 2023. Three levels of fertilization and irrigation were utilized. We investigated the interactions between irrigation and fertilization on sucrose accumulation in sugar beet root. We examined key enzyme activities involved in sucrose metabolism alongside their gene expression levels. The findings suggested that reducing irrigation by 15%, fertilization by 10%, or both irrigation by 15% and fertilization by 10%, increased sucrose concentrations of sugar beets compared to the control group administered conventional water and fertilizer. Over the two-year period, the average sucrose concentration increased by 0.45, 0.57, and 0.65 degrees, respectively, under each treatment. Subsequent research verified that appropriately reducing water and fertilizer can regulate the expression of enzyme genes, thus influencing enzyme activity. Moreover, due to the higher efficiency of enzyme synthesis compared to decomposition, it contributed to an increase in net enzyme activity. These findings suggest that an appropriate reduction of water and fertilizer can improve sucrose synthesis rates and increase the sucrose concentration in sugar beets, providing a theoretical basis for environmentally friendly generation and enhanced efficiency in sugar beet growth. The relationship between active oxygen metabolism and resistance to late blight (Phytophthora infestans) in potato (Solanum tuberosum L.) was studied for 72 h post-inoculation by comparing three resistant cultivars (low disease index) with three susceptible ones (high disease index). Activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), and the content of ascorbic acid (ASA), were higher in the resistant cultivars than in the susceptible ones. The production rate of the superoxide anion radical (O2−) was lower in the resistant cultivars than in the susceptible ones. These changes, which were associated with the potato plant’s response to infection with P. infestans, provide some insight into the physiological basis of resistance and may also provide a screening tool for resistance to late blight. Sugar beet is a characteristic cash crop in Inner Mongolia, and water and fertilizer is an important management measure in sugar beet planting. The interaction between water and nitrogen is of practical significance in regulating the growth of sugar beet. The coordinated relationship between water and fertilizer can achieve the purpose of water saving, fertilizer saving, cost saving and efficiency increasing. The cultivation method of drip irrigation under plastic mulch was adopted in this study. The effects of water nitrogen supply on water metabolism and photosynthetic performance of sugar beet were studied through two years of field experiments in order to provide theoretical basis for improving the utilization efficiency of water and fertilizer and rational fertilization and irrigation. The results showed that the water nitrogen supply level and its proportion had influence with the water potential, Transpiration rate (Tr), Photosynthetic rate (Pn) and stomatal Conductance (Cond) of sugar beet leaves. And they were significantly positively correlated with the yield of sugar beet. When the water potential of sugar beet leaves was −0.38 ∼ −0.23 MPa in the period of leaf fast growth, root and sugar growth, and −0.45 ∼ −0.38 MPa in the period of sugar accumulation, the yield and sugar content were the highest. Under the coupling condition of water and nitrogen, the irrigation amount of sugar beet was 1350 ∼ 1427 m3·ha−1, and the nitrogen application amount was 150 ∼ 179.22 kg·ha−1. Melatonin (MT), as a putative pleiotropic signal molecule, could participate in inducing postharvest disease resistance and inhibiting postharvest pathogen infection in fruit. In this study, we investigate the mechanism of induced resistance by MT treatment against Alternaria rot of jujube. The results showed that 2 and 20 μmol/L MT did not have direct antimicrobial activity, 200 μmol/L MT had little influence on antifungal activity against Alternaria alternata in vitro. MT significantly reduced the lesion area of fruit inoculated with A. alternata. MT decreased the relative electrolytic leakage, malondialdehyde content, hydrogen peroxide, and the rate of superoxide anion production, while enhanced the ascorbic acid and reduced glutathione contents. Furthermore, MT observably increased the defensive enzyme activities of superoxide dismutase, the catalase, ascorbate peroxidase, peroxidase. Accordingly, these results indicated that MT treatment increased the disease resistance of jujube fruit by regulating reactive oxygen species metabolism. Novelty impact statement: The results showed that MT treatment could inhibit the expansion of lesion diameter, reduce the decay rate of winter jujube, and reduce the accumulation of reactive oxygen species by activating the oxidase system, thus enhancing the resistance of winter Jujube fruit to Alternaria alternata infection by adjusting the ROS mechanism. Xyloglucan endotransglucosylase/hydrolases (XTHs) play a crucial role in plant growth and development. However, their functional response to phytohormone in sugar beet still remains obscure. In this study, we identified 30 putative BvXTH genes in the sugar beet genome. Phylogenetic and evolutionary relationship analysis revealed that they were clustered into three groups and have gone through eigh t tandem duplication events under purifying selection. Gene structure and motif composition analysis demonstrated that they were highly conserved and all contained one conserved glycoside hydrolase family 16 domain (Glyco_hydro_16) and one xyloglucan endotransglycosylase C-terminus (XET_C) domain. Transcriptional expression analysis exhibited that all BvXTHs were ubiquitously expressed in leaves, root hairs and tuberous roots, and most of them were up-regulated by brassinolide (BR), jasmonic acid (JA), abscisic acid (ABA) and gibberellic acid (GA 3). Further mutant complementary experiment demonstrated that expression of BvXTH17 rescued the retarded growth phenotype of xth22 , an Arabidopsis knock out mutant of AtXTH22. The methods to leaching juice from dried jujube using six treatments and some factors related with juice yield such as total soluble solids (TSS), pectinase activity, pectin contents, galacturonic acid, and the microstructure morphology of cell were investigated. Six treatments including natural leaching, ultrasonic, microwave, ultrasonic before microwave, ultrasonic after microwave, and pressing directly were applied to extract juice. The group which treating with ultrasonic before microwave displayed its total soluble solids (TSS) was 16 °Brix, which was 6.67% higher than that of natural leaching. And its total soluble solids (TSS) reached to equilibrium in 2 hr, which was faster than that of natural leaching. The mechanism of improving the production efficiency of juice yield using ultrasonic combined with microwave was explored accordingly. The content of pectin and galacturonic acid increased by 58.52% and 59.01%, respectively, which were the highest among all samples. The activity of pectinase was 9.71 μg/(h·g), which was significantly decreased 40.23% as compared to natural leaching. And the treated cells became shriveled and pitted, which led to the leakage of the contents of cell. Thus, the result showed that treating with ultrasonic before microwave displayed the best juice yield. Ultrasonic cooperate with microwave was an efficient method to leaching juice from dried jujube. The K+ transporter/high-affinity K+/K+ uptake (KT/HAK/KUP) transporters dominate K+ uptake, transport, and allocation that play a pivotal role in mineral homeostasis and plant adaptation to adverse abiotic stresses. However, molecular mechanisms towards K+ nutrition in forest trees are extremely rare, especially in willow. In this study, we identified 22 KT/HAK/KUP transporter genes in purple osier willow (designated as SpuHAK1 to SpuHAK22) and examined their expression under K+ deficiency, drought, and salt stress conditions. Both transcriptomic and quantitative real-time PCR (qRT-PCR) analyses demonstrated that SpuHAKs were predominantly expressed in stems, and the expression levels of SpuHAK1, SpuHAK2, SpuHAK3, SpuHAK7, and SpuHAK8 were higher at the whole plant level, whereas SpuHAK9, SpuHAK11, SpuHAK20, and SpuHAK22 were hardly detected in tested tissues. In addition, both K+ deficiency and salt stress decreased the tissue K+ content, while drought increased the tissue K+ content in purple osier plant. Moreover, SpuHAK genes were differentially responsive to K+ deficiency, drought, and salt stresses in roots. K+ deficiency and salt stress mainly enhanced the expression level of responsive SpuHAK genes. Fifteen putative cis-acting regulatory elements, including the stress response, hormone response, circadian regulation, and nutrition and development, were identified in the promoter region of SpuHAK genes. Our findings provide a foundation for further functional characterization of KT/HAK/KUP transporters in forest trees and may be useful for breeding willow rootstocks that utilize potassium more efficiently. BACKGROUND: Although ancient physicians had rich discussions on rheumatoid arthritis, few have discussed based on the theory of collaterals diseases. OBJECTIVE: To summarize the drug treatment classification, etiology analysis, and "collateral-interest accumulation." METHODS: CNKI, El, SCI, and ISTP databases were searched for articles published after 1900. The keywords were "The bone erosion of meumatoid arthritis, TCM collaterals theory." Data from more than 10 domestic and foreign articles were selected to analyze the drug treatment classification, etiology analysis, and "collateral-interest accumulation". RESULTS AND CONCLUSION: (1) One of the causes of meumatoid arthritis bone disease is the collaterals disease of traditional Chinese medicine. The development of intractable diseases should follow the law of the development of diseases from the meridians to the collaterals, from the gas to the blood, and from the functional lesions to the organic diseases. (2) The pathogenesis of rheumatoid arthritis lies in the accumulation of collaterals, which causes joint swelling, joint deformity and damage. The effect of chitosan (0.1 mol/L) combined with sodium silicate (100 mmol/L) treatment on Alternaria rot caused by Alternaria alternata in postharvest jujube fruit (Ziziphus jujuba Mill. cv. Dongzao) was studied. The results showed that chitosan combined with sodium silicate treatment significantly reduced the lesion diameter, decay incidence, red index and weight loss of jujube fruit compared with control samples. Combining treatment increased the ascorbic acid, flavonoids, total phenolic compounds and lignin content. The level of superoxide anion (O2·-) and hydrogen peroxide of treated samples was also increased compared with the control samples. Meanwhile, the activities of phenylalanine ammonia lyase, polyphenol oxidase, superoxide dismutase, peroxidase, chitinase and β‐1,3‐glucanase were also accumulated in treated jujube samples, while the activity of catalase markedly decreased. These results indicated that chitosan combined with sodium silicate treatment could induce the disease resistance of postharvest jujube. For undisturbed loess with water sensitivity and dynamic vulnerability, which is often in a state of damage accumulation due to the influence of natural factors (the fluctuation of groundwater level, rainfall infiltration/evaporation, and seismic load), the impact of relevant engineering problems is significant, and the disaster-causing process is complex. Cyclic simple tests were used to illustrate the behaviour of the dynamic shear modulus of loess exposed to various factors of dry-wet(D-W) cycles and stress conditions (D-W cycles times N , lower limit water content w 1 , and consolidation stress σ v). Using scanning electron microscopy (SEM), we assessed the inherent damage mechanism under D-W cycles. The findings show that the D-W cycles considerably affect the dynamic shear modulus of undisturbed loess, with the strongest effect occurring during the first D-W cycles. Based on the Hardin hyperbolic model, the stress exponent n is a specific parameter of w 1. In addition, the relationship between N and G 0 / P a /(σ v / P a) n is a power function, and G 0 / P a /(σ v / P a) n drops as w 1 and N increase. The fitting parameters a of the dynamic shear modulus ratio normalization model are not sensitive to σ v / P a , D-W cycles times N , and lower limit water content w 1. Based on the dynamic shear modulus, a damage degree prediction method considering N , w 1 , and σ v is established. Our study aimed to investigate the histologic and immunological changes of portal hypertension (PH) pre- and postsplenectomy in hepatitis B virus (HBV)-related cirrhosis. Peripheral blood samples were obtained from 30 patients with HBV-related cirrhosis and PH at pre- and postsplenectomy time points and from 15 healthy subjects. Spleen tissue specimens were collected from 15 of the patients with HBV-related cirrhosis and from 8 control patients who had undergone splenectomy due to trauma. Immunohistochemical staining was performed to evaluate the immune effector cells and the expression of negative immune regulators. Flow cytometry was used to investigate the immunophenotypes and percentages. The spleen of cirrhotic patients with PH showed extensive depletion of splenic CD4, CD8, and human leukocyte antigen DR cells along with overexpression of the inhibitory receptors programmed death-1 (PD-1) and T cell immunoglobulin domain and mucin domain-3 and their ligands (PD-L2 and galectin-9). Peripheral blood of patients with PH showed remarkable decrease in proportions of CD8 T cell and natural killer (NK) cells and increase in regulatory T (Treg) cells, as well as high expression of PD-1 in CD4/8 T cells. Compared with presplenectomy patients, cirrhotic patients with PH showed increased proportions of CD8 and NK cells, decreased proportion of Treg cells, and decreased expression of PD-1 in peripheral blood CD4/8 T cells after splenectomy. Surface-enhanced Raman Spectroscopy (SERS) is supposed to be a promising detection technology for arsenic species owing to the unique fingerprints. However, further application of SERS is hampered by its poor repeatability. Herein, the role of surface silver ions on colloidal Ag was revealed in SERS analysis of arsenic species. Arsenic species were adsorbed on Ag nanoparticles (Ag NPs) driven by surface silver ions and were simultaneously sensed by the SERS "hot spots" generated from the aggregation of Ag NPs. So, the inconsistent SERS activities of Ag NPs synthesized from different batches can be significantly improved by modifying external silver ions onto Ag NPs (AgNPs@Ag+), Specific binding affinity of surface silver ions to arsenic species generated higher sensitivity (detection limit, 4.0 × 10-11 mol L-1 for arsenite, 8.0 × 10-11 mol L-1 for arsenate), wider linear range, faster response, cleaner spectra background and better reproducibility. Batch-to-batch reproducibility was significantly improved with a variation below 3.1%. The method was also demonstrated with drinking and environmental water with adequate recovery and high interference resistance. Abstract: In this paper, a mathematical model of the spherical involute tooth surface of a spiral bevel gear whose generating curve is an arc of a circle is built. The parameters on the base cone of the gear are derived from those on the pitch cone of the gear. The algorithm derived can be used for designing the spherical involute surface of straight and spiral bevel gears. The curve of action, the paths of contact and the contact patterns of spiral bevel gears are obtained by the tooth contact analysis (TCA). The TCA results show that: 1) the curve of action is on a spherical surface; 2) the paths of contact do not change with the variation of the radii of the generating arcs of circles of the tooth surfaces of the pinion and the gear; 3) the length of the major semi axis of their contact ellipse is determined by the radii of the generating arcs of circles of the tooth surfaces of the pinion and the gear and it does not change during the transmission process. The role of the spleen in sepsis is still controversial. Therefore, we investigated the effect of the spleen on sepsis-induced immune dysfunction in C57BL/6 mice subjected to caecal ligation and puncture (CLP). Changes in different immune cells and apoptotic cells in the spleen and peripheral blood were observed 4, 24 and 48 h after CLP. Then, we determined that 48 h following CLP was the most significant period of immunosuppression. Next, we divided the mice into four groups: control, CLP, CLP + spx (splenectomy 48 h after CLP) and spx + CLP (splenectomy surgery two weeks before CLP). Compared with the CLP mice, the CLP + spx and spx + CLP mice had improved survival rates and organ injuries, increased expression of inflammatory factors, a decreased proportion of regulatory T cells (Tregs), and reduced expression of the genes involved in the programmed cell death 1 and its ligand 1 (PD1-PDL1) pathway in immune cells and T-cell immunoglobulin-mucin domain 3 (Tim 3) and Galectin9 in the liver and lungs after 72 h in late-phase sepsis. In addition, the expression of PD-1 was significantly reduced in T cells in spx + CLP mice, and the expression of PD-L1 in myeloid-derived suppressor cells (MDSCs) was reduced in the CLP + spx group, especially in macrophages. These findings suggested that splenectomy could protect septic mice from exhaustion of immune cells by reducing the proliferation of Treg cells and expression of the PD-1/PD-L1 axis in immune cells during the immunosuppressive stage of sepsis. Splenectomy could also reduce liver and lung injuries possibly via the Tim 3 and/or Galectin-9 axis. • Apatite high Sr content and (F/Cl) melt ratios were caused by melt fractionation. • Au-fertile porphyry with the highest apatite δEu and lowest δCe value was oxidized. • High Sr/Y and δEu values reflect an adakitic affinity of apatite-bearing host rock. • Porphyries in different tectonic settings show distinguishable Cl and F/Cl ratios. Tethyan orogenesis and Indian–Eurasia collision facilitate the development of abundant Late Mesozoic and Cenozoic porphyries and associated Cu ± Mo ± Au deposits in the SE Tibetan plateau. Typical deposits including Beiya with the largest Au reserve of 350 t, Tongchanggou with the largest Mo reserve of 142.5 Mt and Pulang with the largest Cu reserve of 804 Mt were mostly concerned by economic geologists. Porphyries occurred in these three typical deposits were classified as Au-fertile (Wandongshan monzogranite porphyry in Beiya), Mo-fertile (granodiorite porphyry in Tongchanggou), Cu-fertile (quartz monzogranite porphyry in Pulang) and ore-barren (Baliancun biotite monzogranite porphyry in Beiya) systems. In this study, apatite geochemistry from four porphyry systems with distinctive metal endowments was investigated to further constrain magmatic fertility indicators and granitoid petrogenesis. All the studied apatite grains belong to magmatic fluorapatite and are generally wrapped within plagioclase and biotite phenocrysts, suggesting that apatite occurs as early-crystallization phase during crystal fractionation. Apatite grains with porous texture, irregular patchy zoning and secondary rare earth mineral inclusions were eliminated to make the minimal effect of hydrothermal alteration. The F and Cl contents in melt are also estimated based on analyzed F–Cl contents and corresponding F–Cl partition coefficients between apatite and melt under different P–T conditions in this study. Apatite from the Wandongshan (WDS) Au-fertile monzogranite porphyry (MP) has the highest Sr content (average of 1530 ppm) and (F/Cl) melt ratios (44 to 125), indicating that the WDS MP experienced the highest degree of melt fractionation. Additionally, the WDS Au-fertile porphyry is more oxidized than ore-barren, Mo-fertile and Cu-fertile porphyries, evidenced by its highest apatite δEu (0.56–0.83) and lowest δCe (0.98–1.05) values. Rare earth elements (REEs), Y and Sr in apatite were found to be most effective for discriminating magma types. All the apatite grains from typical Late Mesozoic and Cenozoic porphyries in SE Tibet show relatively high Sr/Y (0.5–23.6) and δEu (0.41–0.83) values, pointing to an adakitic affinity of apatite-bearing rock, which is consistent with the whole-rock geochemistry. These porphyries formed under distinctive tectonic settings show distinguishable Cl melt and (F/Cl) melt ratios. The Pulang Cu-fertile quartz monzonite porphyry formed under a subduction setting has relatively high Cl melt contents of 100–1000 ppm and low (F/Cl) melt ratios of 2–22, suggesting derivation of the parental magma from a metasomatized hydrous mantle wedge. Whereas, the WDS Au-fertile MP and Bailiancun ore-barren biotite monzonite porphyry generated under intracontinental setting show low Cl melt contents of 23–180 ppm and variable (F/Cl) melt ratios of 12–125, because F-bearing minerals were decomposing at greater depths under higher pressures during the collision. Background and aim: Splenectomy has been reported to attenuate liver fibrosis. In addition, phenotype transitions of infiltrating macrophages, including Ly6Chigh and Ly6Clow, play an essential role in the liver fibrosis. However, whether the spleen can regulate the phenotype switch of macrophages and the underlying mechanism still remain unclear. Methods: Chronic liver fibrosis in mice was induced by intraperitoneal injection with carbon tetrachloride. Splenectomy or sham operation was performed with or without depletion of macrophages during liver fibrosis. Liver fibrosis and the proportion of Ly6Chigh and Ly6Clow macrophages were analyzed. Western blotting of ERK1/2 signals was performed in isolated macrophages to investigate the underlying mechanism of phenotype transition. RAW264.7 cells were stimulated by liver total cells conditioned medium with or without preincubation of SCH772984, the ERK1/2 inhibitor, and the phenotype switch of RAW264.7 cells was examined. In vivo , intraperitoneal injection of SCH772984 was performed on the splenectomy mice and the phenotype switch of liver infiltrating macrophages was tested. Results: Splenectomy alleviated the liver inflammation and fibrosis and also promoted the phenotypic switch of infiltrating macrophages to a Ly6Clow phenotype in fibrotic liver. The p-ERK1/2 expression was upregulated in macrophages at the same time. Furthermore, splenectomy increased the percentage of Ly6Clow macrophages and decreased the percentage of Ly6Chigh macrophages both in vivo and in vitro , which was reversed by SCH772984. Conclusions: Splenectomy attenuates both the liver fibrosis and inflammation, and promotes the phenotype switch of infiltrating macrophages to an anti-inflammatory Ly6Clow phenotype by activating the ERK1/2 pathway during liver fibrosis.
