Abstract

Glycosylation is a topic of intense current interest in the development of biopharmaceuticals because it is related to drug safety and efficacy. This work describes results of an interlaboratory study on the glycosylation of the Primary Sample (PS) of NISTmAb, a monoclonal antibody reference material. Seventy-six laboratories from industry, university, research, government, and hospital sectors in Europe, North America, Asia, and Australia submitted a total of 103 reports on glycan distributions. The principal objective of this study was to report and compare results for the full range of analytical methods presently used in the glycosylation analysis of mAbs. Therefore, participation was unrestricted, with laboratories choosing their own measurement techniques. Protein glycosylation was determined in various ways, including at the level of intact mAb, protein fragments, glycopeptides, or released glycans, using a wide variety of methods for derivatization, separation, identification, and quantification. Consequently, the diversity of results was enormous, with the number of glycan compositions identified by each laboratory ranging from 4 to 48. In total, one hundred sixteen glycan compositions were reported, of which 57 compositions could be assigned consensus abundance values. These consensus medians provide community-derived values for NISTmAb PS. Agreement with the consensus medians did not depend on the specific method or laboratory type. The study provides a view of the current state-of-the-art for biologic glycosylation measurement and suggests a clear need for harmonization of glycosylation analysis methods.

Abstract

Large-mode-area anti-resonant fibers (LMA ARF) with high beam quality have the potential to revolutionize mid-infrared high-power laser delivery by mitigating mode instabilities and nonlinear degradation. However, their tens-of-microns mode field diameters and small numerical aperture significantly increase the mode field mismatch and coupling complexity, respectively. In this work, an LMA all-solid anti-resonant fiber-microlens with a robust operation of high beam quality in the mid-infrared region was designed and prepared. It can significantly stabilize the efficiencies of coupling and transmission at a large radial offset. Consequently, this AS-ARF demonstrates substantial potential for advancing mid-infrared laser power delivery via fiber, ensuring exceptional beam quality throughout the process. Abstract

In order to address the 'capacity crisis' caused by the narrow bandwidth of the current C band and the demand for wide-spectrum sensing sources and tunable fiber lasers, a broadband luminescence covering the C + L bands using Er3+/Yb3+ co-doped fluorotellurite glass fiber is investigated in this paper. The optimal doping concentrations in the glass host were determined based on the intensity, lifetime, and full width at half maximum (FWHM) of the fluorescence centered at 1.5 µm, which were found to be 1.5 mol% Er2O3 and 3 mol% Yb2O3. We also systematically investigated this in terms of optical absorption spectra, absorption and emission cross-sections, gain coefficients, Judd–Ofelt parameters, and up-conversion fluorescence. The energy transfer (ET) mechanism between the high concentrations of Er3+ and Yb3+ was summarized. In addition, a step-indexed fiber was prepared based on these fluorotellurite glasses, and a wide bandwidth of ~112.5 nm (covering the C + L bands from 1505.1 to 1617.6 nm) at 3 dB for the amplified spontaneous emission (ASE) spectra has been observed at a fiber length of 0.57 m, which is the widest bandwidth among all the reports based on tellurite glass. Therefore, this kind of Er3+/Yb3+ co-doped fluorotellurite glass fiber has great potential for developing broadband C + L band amplifiers, ultra-wide fiber sources for sensing, and tunable fiber lasers. 43. Simple and Efficient Laser-Induced Evaporation Assembly Method to Fabricate SERS Optical Fiber Probe for Detection of PAHs

Abstract

Surface-enhanced Raman scattering (SERS) is a powerful tool for trace substance detection and analysis. However, traditional SERS detection based on substrates usually separates sampling and detection, which are time consuming and lack portability for practical applications. In this study, we developed a SERS fiber optic probe using a simple and efficient laser-induced evaporation assembly method. By direct immersion of the fiber in a microliter colloidal solution of silver nanocubes (Ag NCs) under laser induction, a monolayer of uniform and dense Ag NCs was modified on the end face of the optical fiber. The fabricated SERS fiber optic probe demonstrated excellent reproducibility and stability with a detection limit of less than 10–8 M for the 4-mercaptobenzoic acid (4-MBA), with the intensity of its two characteristic Raman peaks showing a high linear correlation with the logarithm of the concentration. Furthermore, by combining Ag NCs with bromide ions, the Ag-Br NCs fiber optic probe we fabricated and exhibited outstanding performance in detecting organic pollutants polycyclic aromatic hydrocarbons (PAHs). This work may have potential significance in broadening the application field of SERS fiber-optic probes.
