8. Effects of different conditioning regimens on HLA-mismatched microtransplantation and changes in fine immune indices in acute myeloid leukaemia

Abstract

Abstract The aim of this study was to observe the effects of different conditioning regimens on fine immune indices after microtransplantation (MST) in patients with acute myeloid leukaemia (AML). This article discusses the possible immune mechanism of microtransplantation and describes a more optimized conditioning regimen. A total of 55 AML patients who received MST treatment at the Second Hospital of Lanzhou University from August 2015 to October 2023 were included in this study, and 13 AML patients who did not receive MST but directly received the maintenance therapy were included as the control group (C). The MST patients were divided into a conditioning regimen with venetoclax group (A) and a conditioning regimen without venetoclax group (B). The fine immune indices were detected by flow cytometry and cytometric bead array analysis. Changes in the immune indices before and after treatment were observed, and the progression-free survival (PFS) and overall survival (OS) of patients in the MST group were analysed. Compared with those in Group B, patients in Group A had better PFS and OS. 31. Research on a Single-Vibrator Multimodal Inertial Impact Piezoelectric Motor With Self-Clamping Backsliding Suppression

Abstract

Traditional inertial impact piezoelectric motors can only operate under quasi-static conditions, resulting in low output force, torque, and speed, and there is a backsliding distance at each cycle that affects their working stability. Recently, many research works have changed their quasi-static working state to a resonance state. In this study, a novel inertial impact piezoelectric motor is proposed. The multimodal and multifrequency band driving method can enable the driver to achieve bidirectional driving and wide resolution adjustment only using a single vibrator, and the unique self-clamping mechanism can effectively suppress the backsliding phenomenon. The dynamics model and FEM model are used for dynamic analysis and modal analysis of the proposed actuator. Manufacturing a prototype and building a test bench to test the performance of the motor. The experimental results show that in the resonant state, the maximum forward output speed is 4.401 mm/s and the displacement resolution is 66 μm in first-order vibration mode. The maximum reverse output speed is 5.369 mm/s and the displacement resolution is 12 μm in second-order vibration mode. Meanwhile, the maximum load capacity of the motor is 180 g. Under quasi-static conditions, the minimum displacement resolution of the motor can reach 4 μm. Under 1 N preload, the backslidings of forward and reverse motor operation are 7.83% and 2.89%, respectively. Under 2 N preload, they are 3.98% and 1.53%, respectively. When the preload exceeds 3 N, the motor does not experience any rebound phenomenon in any state.

32. Sub-Micrometer Resolution Self-Displacement Sensing for Electromagnetic Actuators

Abstract

Recently, soft electromagnetic actuators (SEMAs) have emerged with promising features for haptic feedback, human-machine interfaces and soft robotics. It is challenging to integrate sensors into an SEMA-based system due to the incompatibility of conventional sensors and space restrictions. Here, we present a self-displacement sensing solution for SEMAs, which exploits the eddy-current effect coupling between the driving coil and the metal surface of the permanent magnets for displacement sensing. A self-displacement sensing and driving circuit (SDSDC) was developed and evaluated, in which low-frequency driving and high-frequency sensing signals can function simultaneously without interfering with each other. Experimental results reveal that an SEMA prototype with the SDSDC can produce a motion over 200 μm with a sensing resolution of 0.038 μm (0.02% motion range). It has a response time of 17 ms and can operate from 0.1 to 100 Hz. Moreover, the SDSDC can operate with the same characteristics when the driving current varies in amplitude and in the presence of external stimuli. Finally, the SDSDC was demonstrated on a conventional voice coil motor. The SDSDC represents a promising solution for designing interactive and responsive soft machines and interfaces, and it can also be implemented in various electromagnetic actuators.

Abstract

DARS, encoding for aspartyl-tRNA synthetase, is implicated in the pathogenesis of various cancers, including renal cell carcinoma, glioblastoma, colon cancer, and gastric cancer. Its role in BCR/ABL1-negative myeloproliferative neoplasms (MPNs), however, remains unexplored. This study aimed to elucidate the expression of DARS in patients with MPNs (PV 23, ET 19, PMF 16) through immunohistochemical analysis and to examine the profiles of circulating immune cells and cytokines using flow cytometry. Our findings indicate a significant overexpression of DARS in all MPNs subtypes at the protein level compared to controls (P < 0.05). Notably, elevated DARS expression was linked to splenomegaly in MPNs patients. The expression of DARS showed a negative correlation with CD4+ T cells (R = − 0.451, P = 0.0004) and CD4+ T/CD8+ T cell ratio (R = − 0.3758, P = 0.0040), as well as with CD68+ tumor-associated macrophages (R = 0.4037, P = 0.0017). Conversely, it was positively correlated with IL-2 (R = 0.5419, P < 0.001), IL-5 (R = 0.3161, P = 0.0166), IL-6 (R = 0.2992, P = 0.0238), and IFN-γ (R = 0.3873, P = 0.0029). These findings underscore a significant association between DARS expression in MPNs patients and specific clinical characteristics, as well as immune cell composition. Further investigation into the interplay between DARS and the immune microenvironment in MPNs could shed light on the underlying mechanisms of MPNs pathogenesis and immune dysregulation. Abstract

Multiple myeloma bone disease (MBD) is a common clinical complication in patients with multiple myeloma (MM), and the occurrence of bone-related events seriously affects the quality of survival and prognosis of patients. The pathogenesis of MBD involves an imbalance in physiologic bone remodeling: overactivation of osteoclasts, suppression of osteoblasts, and multiple cell-cytokine interactions in the microenvironment, including osteoblasts, bone marrow stromal cells, and immune cells. The current treatment of MBD is based on standard antimyeloma therapy to control tumor progression, along with the use of bone-related drugs acting on bone remodeling within the indications. To prevent bone destruction. In order to prevent bone destruction, it is essential to induce new bone formation to repair the existing lesions while resisting bone resorption, and a variety of novel bone-targeting drugs are currently demonstrating anti-osteopathic effects in preclinical and clinical trials. 48. Analysis of the clinical efficacy and voice outcomes of CO2 laser resection versus laryngeal microsurgery for vocal cord polyps.

Abstract

To compare CO2 laser resection and laryngeal microsurgery for vocal cord polyps and provide evidence for the optimal surgical method. This was a retrospective cohort study that included 74 patients with vocal cord polyps who underwent either CO2 laser resection or laryngeal microsurgery in our hospital from August 2018 to December 2021. According to their preference, 77 patients were divided into two groups: a CO2 laser resection group (n = 35) and a laryngeal microsurgery group (n = 39). Patients were evaluated two days before surgery, and follow-ups were conducted one, two and four weeks after surgery. The voice handicap index (VHI-10) score, voice acoustic analysis results and electronic laryngoscopy results were collected for each patient, and the differences between the two groups were evaluated. The basic demographic characteristics of the 74 patients were comparable, and all patients completed postoperative follow-up observations. A total of 30 (85.71%) patients in the CO2 laser resection group and 22 (56.41%) patients in the laryngeal microsurgery group were healed. The total effectiveness rate of the CO2 laser resection group (94.29%) was significantly higher than that of the laryngeal microsurgery group (82.05%), and the difference between the two groups was statistically significant (p =.037). Both surgical methods had a positive effect on reducing VHI-10 scores with the effect of CO2 laser resection being more obvious. The difference between the two groups in this regard was statistically significant (p <.001). The effects of each surgical method on the average fundamental frequency perturbation (jitter), amplitude perturbation (shimmer), maximum phonation time and dysphonia severity index were not statistically significant (p >.05). CO2 laser resection and laryngeal microsurgery have similar effects on voice quality, but CO2 laser resection has higher clinical efficacy. [ABSTRACT FROM AUTHOR]
