Objective To investigate the effects of JSH⁃23 combined with Stattic targeting inhibition of nuclear factor⁃κB (NF⁃κB) and signal transducer and activator of transcription factor 3 (STAT3) signaling pathways on the proliferation and migration ability of mesenchymal glioblastoma cells and the expressions of NF⁃κB pathway and STAT3 pathway⁃related proteins. Methods The mRNA⁃seq results of 529 glioma patients were downloaded from The Cancer Genome Atlas (TCGA). Bioinformatics was used to analyze the correlation between RelA/P65 and STAT3 and the markers of mesenchymal glioblastoma, as well as the expression of NF⁃κB pathway and STAT3 pathway⁃related proteins in mesenchymal, classical and proneural glioblastoma. The human glioma cell lines U87MG and U251MG in vitro were treated with JSH⁃23, Stattic, JSH⁃23 and Stattic, respectively. The half⁃inhibitory concentration (IC50) of the two drugs was calculated by cytotoxicity assay, and the synergistic effect of the two drugs was observed by drug synergistic assay. CCK⁃8 assay and colony⁃forming assay were used to detect the proliferation activity of tumor cells. Wound healing assay and Transwell assay were used to detect the migration ability of tumor cells. Western blotting was used to detect the relative expressions of NF⁃κB pathway and STAT3 pathway⁃related proteins P65, phosphorylated P65 (p⁃P65), STAT3, phosphorylated STAT3 (p⁃STAT3) and CD44. Results 1) Bioinformatics analysis showed that RelA/P65 mRNA and STAT3 mRNA were positively correlated with mesenchymal glioblastoma markers CD44, CXCR4, CHI3L1, IL⁃4R and TRADD (r=0.206-0.605; P＜0.01, for all); the expressions of NF⁃κB pathway and STAT3 pathway⁃related proteins in mesenchymal glioblastoma were the highest, followed by classical glioblastoma, and the lowest in proneural glioblastoma. 2) The cytotoxicity assay showed the IC50 of JSH⁃23 on U87MG and U251MG cells were 59.39 and 56.21 μmol/L, and Stattic were 0.96 and 1.08 μmol/L. The drug synergistic assay showed that the synergistic effect of JSH⁃23 at 40-80 μmol/L and Stattic at 0.50-1 μmol/L on U87MG was the highest, and the synergistic effect of JSH⁃23 at 40 μmol/L and Stattic at 1 μmol/L on U251MG was the highest. The final concentration of JSH⁃23 was 60 μmol/L and Stattic was 1 μmol/L. 3) After the combined treatment of JSH⁃23, Stattic, JSH⁃23 and Stattic, the proliferation activity of U87MG and U251MG cells decreased (P=0.000, for all), the colony forming efficiency decreased (P=0.000, for all), the cell mobility decreased (P＜0.05, for all), and the positive cell numbers of crystal violet staining decreased (P=0.000, for all). The relative expressions of P65 (P=0.000, for all), p⁃P65 (P=0.000, for all), STAT3 (P=0.000, for all), p⁃STAT3 (P=0.000, for all) and CD44 (P=0.000, for all) were decreased, especially after JSH⁃23 combined with Stattic treatment (P＜0.01, for all). Conclusions JSH⁃23 combined with Stattic targeting inhibition of NF⁃κB and STAT3 signaling pathways can effectively inhibit the proliferation and migration ability of mesenchymal glioblastoma cells, and down⁃regulate the relative expressions of NF⁃κB pathway and STAT3 pathway⁃related proteins.

Objective To investigate the anti⁃tumoral effect of ACT001, a novel antitumor compound, on the nuclear factor⁃κB (NF⁃κB) pathway and programmed cell death protein ligand 1 (PDL1) expression in U87 glioblastoma cell line. Methods We analyzed the correlation between the expression of P65 and PDL1, pathological grade, and prognosis by introducing The Cancer Genome Atlas (TCGA) and Chinese Glioma Genome Atlas (CGGA) databases. CCK⁃8 was used to observe the effect of ACT001 on the proliferation of U87 cells, and calculate the median inhibition concentration (IC50). Immunofluorescence staining was used to detect the effect of ACT001 on nuclear translocation of the transcription factor P65. U87 cells were transfected with small interference RNA (siRNA), and P65 knockdown efficiency was determined by quantitative real⁃time polymerase chain reaction (qRT⁃PCR). The relative expression of P65, phospho⁃P65 (p⁃P65) and PDL1 in different transfected groups and drug treatment groups were determined by Western blotting. Results 1) Bioinformatics analysis showed that gliomas with higher pathological grade hourbored higher P65 and PDL1 expression (P＜0.05, for all). Moreover, patients with high expression of P65 and PDL1 mRNA predicted poor prognosis (P＜0.05, for all). 2) CCK⁃8 assay showed that treatment with different concentrations of ACT001 (5, 10, 20, 40, 80, 160 and 320 μmol/L), the inhibition rates of U87 cells were (9.01±4.75)%, (17.03±2.91)%, (28.50±4.85)%, (45.50±5.15)%, (67.67±8.46)% , (83.02±5.79)% and (94.33±1.59)% , respectively, and the IC50 was about 42.98 μmol/L. 3) Immunofluorescence staining showed that ACT001 inhibited nuclear translocation of P65. 4) qRT⁃PCR showed that the P65 mRNA expression of U87 cells in different siRNA transfected groups was statistically significant (F=164.200, P=0.000), and the mRNA expression of P65 in si⁃P65⁃1 group (t=13.290, P=0.000) and si⁃P65⁃2 group (t=12.730, P=0.000) was lower than that of si⁃NC group. 5) Western blotting showed that the relative expression of P65 (F=681.300, P=0.000), p⁃P65 (F=128.800, P=0.000) and PDL1 (F=20.470, P=0.002) in U87 cells of different siRNA transfection groups were statistically significant different. Among them, P65 (t=59.630, P=0.000; t=29.380, P=0.000), p⁃P65 (t=24.370, P=0.000; t=13.460, P=0.000) and PDL1 (t=6.025, P=0.004; t=6.728, P=0.003) in si⁃P65⁃1 group and si⁃P65⁃2 group were lower than those in si⁃NC group. 6) There were statistically significant differences in the relative expression of p⁃P65 (F=269.700, P=0.000) and PDL1 (F=128.800, P=0.000) in U87 cells treated with different concentrations of drugs. P⁃P65 (t=19.750, P=0.000; t=24.640, P=0.000) and PDL1 (t=12.690, P=0.000; t=19.230, P=0.000) in ACT001 25 μmol/L group and 50 μmol/L group were lower than those in normal control group, ACT001 50 μmol/L group were also lower than those of 25 μmol/L group (t=5.288, P=0.006; t=2.868, P=0.046). Conclusions ACT001 can inhibit the proliferation of U87 glioma cells. By inhibiting the phosphorylation and nuclear translocation of P65 in U87 cells, ACT001 decreased the expression of PDL1 to improve the immunosuppressive environment of glioblastoma and thus inhibit the progression of glioblastoma.

The periodic synthetic jet emerging from a circular orifice actively controls the turbulent boundary layer (TBL). A time-resolved particle image velocimetry (TR-PIV) system was designed to capture the velocity field database and based on the single-pixel ensemble correlation (SPEC) algorithm, an average drag reduction rate of 6.2% was obtained. The results show that the synthetic jet causes a wide range of low momentum zones and a low-speed streak in the downstream flow field. And the places where the disturbance intensity is strong are often accompanied by a larger velocity deficit. The instantaneous flow fields are visualized with the Finite-Time Lyapunov Exponent (FTLE), and the hairpin vortex packet composed of five hairpin vortices and the generation of new hairpin vortices are observed when there is no control. Under the action of the synthetic jet, the hairpin vortices are continuously generated from the jet orifice. The synthetic jet mainly achieves the drag reduction effect mainly by modulating the mean convection term cC and the spatial development term cD. The drag reduction effect appears in the region of x/δ0 > 0.38, and the maximum drag reduction rate is 12.2% at x/δ0 = 0.75, and then gradually decreased. Using proper orthogonal decomposition (POD), it is found that the synthetic jet reduces the energy proportion of the large-scale energetic structures. After the conditional average, the synthetic jet limits the influence range of bursting events at various scales in the near-wall region, and weakens the normal transport of momentum and energy brought about by large-scale ejection events (Q2 events) and the wall friction resistance caused by large-scale sweep events (Q4 events). Glioma is the most common malignant tumor of the central nervous system, and it is difficult to cure. The growth of malignant glioma is invasive. In recent decades, the strategy of surgical resection combined with concurrent radiotherapy and chemotheraphy, and immunocheckpoint inhibitors have been made progress. However, the therapeutic effect is still not ideal, which may be related to the low immunogenicity of tumor cells and the immunosuppression of tumor microenvironment (TME). In the TME, immune cells compete with glioma cells and other proliferating cells for nutrients, while metabolites in the TME affect the differentiation and functions of immune cells. This article summarizes the research progress of glioma metabolism regulation and tumor immunosuppressive microenvironment in recent years, which provides new ideas for the study of the mechanism of tumor progression and glioma of treatment. As an important ecological barrier for the North China Plain, research on the spatial variations of soil nutrients in the southern foot of the Taihang Mountains is of great significance for the forestry ecological construction in this rocky mountainous area. With the typical slopes (the artificial forestland and the natural wild slope) of the southern foot of the Taihang Mountains as the research objects, we used the grid method to arrange sampling points, and combined classical statistics, geostatistics and constrained sorting methods to analyze the spatial variations of soil nutrients. The results showed that: 1) soil total carbon (TC) contents were 6.80-57.05 g·kg -1 , and the total nitrogen (TN) contents were 0.74-3.93 g·kg -1 . The coefficients of variation of both soil TC and TN were 25.0%-52.8%, belonging to the moderate degree of variation, which were caused by the combination of random and structural factors. The spatial aggregation of soil nutrients decreased with increasing lag distances. 2) The contents of soil nutrients had increasing trends from the top to the bottom of the slopes, with high nutrient values appearing at the bottom of the slopes. 3) The soil bulk density, gravel content, vegetation coverage, and soil water content were the main factors affecting the spatial variability of soil TC and TN on the southern foot of the Taihang Mountains. 4) Soil water content was the main factor affecting soil nutrients at the natural wild slope, but not at the artificial forestland.

Objective: To study the accuracy of endoscopic polyp size measurement by disposable graduated biopsy forceps (DGBF)., Methods: Accurate gradation of 1 mm was made in the wire of disposable graduated biopsy forceps, which was used to measure the size of tumors under endoscopy. Fifty-eight polyps from 43 patients underwent endoscopy in our department from May to June 2013 were enrolled. Size of polyp was measured and compared among DGBF, routine estimation and direct measurement after resection. The accuracy of polyp size measurement was investigated by four colonoscopists who had finished at least 2000 procedures of colonoscopy., Results: The mean diameter of post-polypectomy measurement was (1.02±0.84) cm. Diameter was less than 1 cm in 36 polyps, 1 to 2 cm in 15, and over 2 cm in 7. The mean diameter of visual estimation was (1.29±1.07) cm, and the difference was significant as compared with actual size (P=0.000). The mean diameter measured by DGBF was (1.02±0.82) cm, and the difference was not significant as compared with actual size (P=0.775). The ratio of visual estimation to actual size was 1.29±0.31, and DGBF estimation to actual size was 1.02±0.11 with significant difference (P=0.000). The accurate rate of DGBF in estimating polyp size was 77.6% (45/58), which was obviously higher as compared to visual estimation [19.0% (11/58), P=0.000]., Conclusion: The accuracy of DGBF as a scale in the estimation of poly size increases as compared to visual estimation.

Objective: To investigate the safety and feasibility of laparoscopic extraperitoneal sigmoid colostomy., Methods: Thirty-six patients with low rectal cancer undergoing laproscopic abdominoperineal resection from July 2011 to July 2012 were prospectively enrolled in the study and randomly divided into extraperitoneal colostomy group(EPC, n=18) and internal peritoneal colostomy group(IPC, n=18). Follow-up period was 4-16 (median, 7) months and postoperative complications were compared between two groups., Results: One case in EPC group was converted to IPC because of poor blood supply of the proximal sigmoid, who was eliminated from the subsequent analysis. Compared with the IPC group, the surgery time was longer in EPC group [(25.3±8.5) min vs. (14.7±6.4) min], while the difference was not statistically significant(P>0.05). Each group had 1 case of stoma ischemia, who both received the colostomy reconstructive surgery. The incidence of stoma edema was significantly higher in EPC group[35.3%(6/17) vs. 0, P<0.05). The early postoperative complications rate did not significantly different between the two groups[58.8%(10/17) vs. 27.8%(5/18), P>0.05]. The late postoperative complications rate was 22.2%(4/18) in IPC group, including 1 case of stoma prolapse, 1 case of stoma stenosis and 2 cases of parastomal hernia. No later postoperative complication occurred in EPC group., Conclusion: Extraperitoneal sigmoid colostomy is an easy and safe procedure with lower late complications as compared to internal peritoneal sigmoid colostomy.

Objective: To evaluate the effects of polyamidoamine dendrimer (PAMAM) liposome as gene carriers on the cellular uptake and its cytotoxicity in colonic cancer cell., Methods: The liposome modified PAMAM was synthesized with liposome and polyamidoamine dendrimer. Plasmid PEGFP-N1 was mixed with the liposome-modified PAMAM or unmodified PAMAM to form nanoparticle complexes. The shape and size of the nanoparticle complexes were observed by transmission electron microscope and the zeta potential was measured by analytical tool. The encapsulating efficiency was determined by ultraviolet spectrophotometer in centrifuging method. After the cell lines SW620 (colonic cancer cell), MCF-7 (breast cancer cell), ECV304 (vascular endothelial cell) were transfected by the two kinds of PAMAM nanoparticle complexes, the flow cytometry was used to determine the uptake of enhanced green fluorescent protein (EGFP) gene. The cytotoxicity of PAMAM liposome nanoparticles and PAMAM nanoparticles was evaluated by MTT assay., Results: The diameter of liposome modified PAMAM complex was (192 ± 16) nm, and that of PAMAM complex was (189 ± 19) nm (P > 0.05); and the zeta potential of liposome modified PAMAM complex was higher than that of PAMAM complex [(42 ± 7) mV vs. (32 ± 7) mV, P < 0.05]. There was no significant difference in envelopment rate between the two groups [(82 ± 7)% vs. (84 ± 6)%, P > 0.05]. After the colonic cancer cell line SW620 was transfected with the two kinds of PAMAM nanoparticle complexes, the cellular uptake of the cells with the liposome-modified PAMAM complex was significantly higher than that of the cell with PAMAM complex (P < 0.05). The cellular survival rate of the cell lines with liposome-modified PAMAM complex was significantly higher than that of cell lines with PAMAM complex (P < 0.05)., Conclusion: The liposome modified PAMAM can improve gene transfection efficiency and suppress its cytotoxicity.

Objective: To study the distribution characteristics of colorectal neoplasm and evaluate the implication for colorectal cancer screening., Methods: A total of 17,939 colonoscopies were performed in the National Center of Colorectal Surgery between October 2004 and June 2009. Characteristics of colorectal neoplasm including anatomical distribution, sex, and age were investigated., Results: Colorectal neoplasm was found in 24.8% (4450/17,939) of the patients during colonoscopy, including adenomatous polyp (n=3410, 19.0%) and adenocarcinoma (n=1040, 5.8%). The prevalence of colorectal neoplasm was higher in male and significantly increased in patients older than 40 years. 63.3% of the lesions located at the distal colon (sigmoid colon and rectum) and 36.7% at the proximal colon (36.7%). In patients with adenomatous polyp, 52.8% (1802/3410) of the lesions were at the distal colon, 30.8% (1049/3410) at the proximal colon, and 16.4% (559/3410) at both distal and proximal colon. In patients with carcinoma (n=1040), 921 (88.6%) lesions located at the distal colon, 118 (11.3%) at the proximal colon, and 1 (0.1%) at both segments., Conclusion: Sigmoidoscopy is inadequate for colorectal cancer screening as compared to colonoscopy.

Objective: To explore the indications for colonoscopy examination and the distribution of diagnostic diseases., Method: From Jan. 2000 to Dec. 2004, 5960 patients received colonoscopy examination in our colorectal center. The indications for colonoscopy examination and the distribution of its diagnostic diseases were analyzed., Results: There were 3096 males and 2594 females,and the mean age was (52+/-15) years. The reasons for colonoscopy included hemafecia (26.9%), atypical abdominal pain (25.8%), diarrhea or increased frequency of stool (11.1%), anal tenesmus or discomfort (7.6%), constipation (7.0%),mucous or bloody purulent stool (3.0%), intra-rectal mass or abdominal mass on physical examination (0.9%), re- examination after colonoscopic polypectomy (10.9%), re-examination after operation for colorectal cancer(1.5%), simple health examination (2.2%). Colonoscope reached the cecum in 97.7% of the cases,and at least one disease was found in 2283 cases (40.1%). Among them,colorectal cancer accounted for 10.3%, colorectal polyps 19.6%, ulcerative colitis 4.3%, and Crohn's disease 0.5% respectively., Conclusion: The indications for colonoscopy are too strict to screen the early stage colorectal cancer. Colonoscopy should be performed in the cases with symptoms such as bloody stool, diarrhea, abdominal pain, constipation, or with colorectal polyps, after operation for colorectal cancer,or as members of hereditary colorectal cancer family.
